Self-assembling amphiphilic designer peptides have been successfully applied as nanomaterials in biomedical applications. Understanding molecular interactions at the peptide-membrane interface is crucial, since interactions at this site often determine (in)compatibility. The present study aims to elucidate how model membrane systems of different complexity (in particular single-component phospholipid bilayers and lipoproteins) respond to the presence of amphiphilic designer peptides. We focused on two short anionic peptides, V4WD2 and A6YD, which are structurally similar but showed a different self-assembly behavior. A6YD self-assembled into high aspect ratio nanofibers at low peptide concentrations, as evidenced by synchrotron small-angle X-ray scattering and electron microscopy. These supramolecular assemblies coexisted with membranes without remarkable interference. In contrast, V4WD2 formed only loosely associated assemblies over a large concentration regime, and the peptide promoted concentration-dependent disorder on the membrane arrangement. Perturbation effects were observed on both membrane systems although most likely induced by different modes of action. These results suggest that membrane activity critically depends on the peptide's inherent ability to form highly cohesive supramolecular structures. [ABSTRACT FROM AUTHOR]

Non-lamellar liquid crystalline aqueous nanodispersions, known also as ISAsomes (internally self-assembled ‘somes’ or nanoparticles), are gaining increasing interest in drug solubilisation and bio-imaging, but they often exhibit poor hemocompatibility and induce cytotoxicity. This limits their applications in intravenous drug delivery and targeting. Using a binary mixture of citrem and soy phosphatidylcholine (SPC) at different weight ratios, we describe a library of colloidally stable aqueous and hemocompatible nanodispersions of diverse nanoarchitectures (internal self-assembled nanostructures). This engineered library is structurally stable in human plasma as well as being hemocompatible (non-hemolytic, and poor activator of the complement system). By varying citrem to lipid weight ratio, the nanodispersion susceptibility to macrophage uptake could also be modulated. Finally, the formation of nanodispersions comprising internally V 2 (inverse bicontinuous cubic) and H 2 (inverse hexagonal) nanoarchitectures was achieved without the use of an organic solvent, a secondary emulsifier, or high-energy input. The self-assembly and self-organization behavior of uracil-conjugated enantiopure (R)- or (S)-1,T-binaphthyl-2,2'-diol (BINOL) and a hydrophobic oligo(p-phenylene ethynylene) (OPE) chromophore exposing 2,6-di(acetylamino)-pyridine termini are reported. Systematic spectroscopic (UV-vis, CD, fluorescence, NMR, and SAXS) and microscopic studies (TEM and AFM) showed that BINOL and OPE compounds undergo triple H-bonding recognition, generating different organic nanostructures in solution. Depending on the solvophobic properties of the liquid media (toluene, CHCl3, CHCl3/CHX, and CHX/THF), spherical, rod-like, fibrous, and helical morphologies were obtained, with the latter being the only nanostructures expressing chirality at the microscopic level. SAXS analysis combined with molecular modeling simulations showed that the helical superstructures are composed of dimeric double-cable tape-like structures that, in turn, are supercoiled at the microscale. This behavior is interpreted as a consequence of an interplay among the degree of association of the H-bonded recognition, the vapor pressure of the solvent, and the solvophobic/solvophilic character of the supramolecular adducts in the different solutions under static and dynamic conditions, namely solvent evaporation conditions at room temperature. Wedescribe the hydrothermal synthesis of zeolite Linde type A(LTA) submicrometer particles using a water-soluble amphiphilic blockcopolymer of poly(dimethylsiloxane)-b-poly(ethyleneoxide) as a template. The formation and growth of the intermediateaggregates in the presence of the diblock copolymer have been monitoredby small-angle X-ray scattering (SAXS) above the critical micellarconcentration at a constant temperature of 45 °C. The early stageof the growth process was characterized by the incorporation of thezeolite LTA components into the surface of the block copolymer micellaraggregates with the formation of primary units of 4.8 nm with a core–shellmorphology. During this period, restricted to an initial time of 1–3h, the core–shell structure of the particles does not showsignificant changes, while a subsequent aggregation process amongthese primary units takes place. A shape transition of the SAXS profileat the late stage of the synthesis has been connected with an aggregationprocess among primary units that leads to the formation of large clusterswith fractal characteristics. The formation of large supramolecularassemblies was finally verified by scanning electron microscopy, whichevidenced the presenceof submicrometer aggregates with size ranging between 100 and 300nm, while X-ray diffraction confirmed the presence of crystallinezeolite LTA. The main finding of our results gives novel insight intothe mechanism of formation of organic–inorganic mesoporousmaterials based on the use of a soft interacting nanotemplate as wellas stimulates the investigation of alternative protocols for the synthesisof novel hybrid materials with new characteristics and properties. A new mechanism for mesostructure formation of ordered mesoporous carbons (OMCs) was investigated with in situ small-angle X-ray scattering (SAXS) measurements: thermally induced self-assembly. Unlike the well-established evaporation-induced self-assembly (EISA), the structure formation for organic-organic self-assembly of an oligomenc resol precursor and the block-copolymer templates Pluronic P123 and F127 does not occur during evaporation but only by following a thermopolymerization step at temperatures above 100 °C. The systems investigated here were cubic (Im3m), orthorhombic Fmmm) and 2D-hexagonal (plane group p6mm) mesoporous carbon phases in confined environments, as thin films and within the pores of anodic alumina membranes (AAMs), respectively. The thin films were prepared by spin-coating mixtures of the resol precursor and the surfactants in ethanol followed by therinopolymerization of the precursor oligomers. The carbon phases within the pores of AAMs were made by imbibition of the latter solutions followed by solvent evaporation and thermopolymerization within the solid template. This thermopolymerization step was investigated in detail with in situ grazing incidence small-angle X-ray scattering (GISAXS, for films) and in situ SAXS (for AAMs). It was found that the structural evolution strongly depends on the chosen temperature, which controls both the rate of the mesostructure formation and the spatial dimensions of the resulting mesophase. Therefore the process of structure formation differs significantly from the known EISA process and may rather be viewed as thermally induced self-assembly. Abstract: A method for producing oriented periodic mesoporous organosilica filaments within the confined channels of anodic alumina membranes is presented. Deposition of the mesoporous filaments were performed under a variety of conditions, which favoured the evaporation induced self-assembly of the mesoporous material. The experimental conditions examined included different drying rates, over a range of humidity values, and sol compositions, e.g. varying the amount of the organosilica component. The deposition process was followed in situ by time resolved small angle X-ray scattering which was essential for evaluating the formation mechanism of the mesophase structures. Through careful control of the deposition environment, the structure and orientation of the mesoporous filaments could readily be varied. [Copyright &y& Elsevier]

Abstract: We report the variation of the molecular assembly and crystallinity of polyethylene-block-poly(ethylene oxide) (EEO, 1400gmol−1) in a non initiated liquid resin of unsaturated polyester (UP). We particularly focus on the driving force that governs the variation of the molecular assembly of the block copolymer in the UP resin upon heating. For this purpose, we performed a set of experiments combining time resolved in situ SAXS and WAXS measurements upon heating and cooling. Upon heating, SAXS shows that the inter-distance of the EEO domains decreases dramatically for temperature above 75°C (between 90 and 30nm for the initial system at room temperature vs 9nm at 110°C) suggesting that the initially homogeneously dispersed EEO domains undergo aggregation. This is consistent with the macroscopic phase separation observed in such temperature range. In situ WAXS shows that the onset of the aggregation coincides with the melting of the PE domains suggesting that the flexibility of the platelets plays an important role in their aggregation. Upon cooling, peculiar behaviour is observed for the systems with higher EEO content, with an irreversible structure formation leading to macroscopic EEO rich domains dispersed in continuous UP rich phase. Lectin−sugar recognition systems are of interest in the pharmaceutical field, especially for the development of drug carriers, tailored for selective delivery. This paper deals with the anhydrous and aqueous self-organization properties of a synthetic cholesteryl oligoethyleneglycol glycoside with the aim of their incorporation in liposomes. Successive phases (lamellar, R3m, Im3m, micelles) have been described depending on water content and temperature. As a result of the presence of sugar residues and their hydration ability, this glycolipid shows a large range of packing parameter with increasing water content. However, because of oligoethyleneglycol spacer, a slight dehydration has been observed with increasing temperature from 20 to 60 °C. [ABSTRACT FROM AUTHOR]

Abstract: A spiropyran molecule has been introduced via one-pot synthesis into mesostructured silica films, which have been obtained through a micelle templating self-assembling process. The mesostructure has been characterized by transmission electron microscopy and small-angle X-ray scattering performed with synchrotron light. The final material exhibited a p6mm 2d-hexagonal organization and the mesostructure has not been affected by the introduction of spiropyrans in the precursor solution. A comparative characterization of the optical properties of the spiropyrans dissolved in different solvents and after incorporation in the mesostructured films has been done by UV–Vis absorption spectroscopy, fluorescence spectroscopy and ellipsometric spectroscopy. The spiropyran-doped films appeared transparent before external stimuli had applied, a colour change to yellow (thermally induced) or to red (light induced) has been observed. Both the colour changes have been observed to be reversible under room temperature or visible light exposure; the colour transitions have been attributed to different equilibria among the various forms of merocyanine of spiropyrans.
