In Argentina, studies on Varroa destructor resistance to coumaphos are still unknown. At present, high infestation levels of V. destructor are being detected in colonies of Apis mellifera after treatment with this acaricide. The aim of the present study was to determine the LC50 of coumaphos in V. destructor from four apiaries with high mite density after treatment with coumaphos. The LC50's were 112, 319, 127 and 133 microg/Petri dish for mites from the four apiaries. Significant LC50 differences were detected between resistant and susceptible mites. LC50 increased 197-559-fold when compared to the corresponding baseline, suggesting the development of resistance. These results are the first report of resistance to coumaphos in V. destructor in Argentina.

This study aimed to detect mutations associated with pyrethroid resistance mechanism in different populations of Varroa destructor from Uruguay and Argentina with different degree of susceptibility to this family of acaricides. Bioassays were performed to evaluate the degree of susceptibility to flumethrin and tau-fuvalinate; then mites were studied in order to determine the presence or absence of the resistance-associated mutations by sequencing a region of the sodium channel gene. In the mites studied from Colonia, which showed resistance to flumethrin and susceptibility to tau-fluvalinate in the bioassays, L925V mutation was observed in heterozygous and homozygous state. The presence of the wild-type allele in homozygous state was identified in all mites from Treinta y Tres, in which acaricides have never been used. The mite population from Mar del Plata through bioassays were determined as susceptible to both pyrethroids, however, genotyping results showed the presence of heterozygous individuals (925 V/L) as well as homozygous wild type (925L), while homozygous resistant individuals (925V) were present at low frequency. This would suggest that this population presents a great potential to give a generalized resistance to pyrethroids. Phoretic mites associated with bumblebees are considered commensals that represent little or no risk for their hosts. The aim of the present study was to assess the potential role of the phoretic mites Pneumolaelaps longanalis and Parasitellus fucorum, as carriers of parasites that are known to affect bees. Mites were extracted from bumblebees dating between 1945 and 1981 which belong to museum collections, and from a freshly collected queen. The DNA was extracted and amplified, and the final products were sequenced and analyzed. Parasite's DNA was found in all samples. Ascosphaera spp., Crithidia bombi, Lotmaria passim, and Nosema ceranae were present in both mite species. Moreover, DNA from Apis mellifera filamentous virus (AmFV), Apicystis bombi, Ascosphaera apis, Nosema apis, and Nosema bombi was found in P. longanalis. This study represents the oldest report of parasite's DNA in bumblebee phoretic mites, highlighting the relevance of museum samples for historical studies in parasitology. [ABSTRACT FROM AUTHOR]

Varroa destructor (Anderson & Trueman, 2000) causes the most important parasitosis of beekeeping in the world. For this reason, prevention is needed to avoid colony death. The most typical treatments involve synthetic acaricides. However, the use of these acaricides results in the emergence of resistant populations of mites to these products and in the appearances of drug residues in products of the hives. Compounds of essential oils have emerged as an alternative to traditional acaricides; however the toxicity produced by these mixtures is currently poorly explored. The aim of this work was to assess, by means of in vitro tests with adult bees, how acaricidal action and toxic interactions in a binary mixture of essential oil compounds (Thymol, Phellandrene, Eucalyptol, Cinnamaldehyde, Myrcene, and Carvacrol) affect V. destructor. Calculations of LC50 's of the individual compounds on A. mellifera and V. destructor made clear that the toxic effect of each compound is different for both species. Thymol and Phellandrene turned out to be lethal for mites at lower concentrations than for bees. The binary mixture of these two substances presented a different toxicity than one produced by each pure compound, as it was highly selective for mites in bioassays at 24 hours through complete exposure to both A. mellifera and V. destructor. Biological activity was evaluated of the Tagetes minuta essential oil in different in vitro laboratory experiments on the mite (Varroa destructor), honeybees (Apis mellifera), the bacterium that causes the American Foulbrood (Paenibacillus larvae), and the fungus that produces chalkbrood (Ascosphaera apis). Two methods of complete exposure were used for mite lethality test: by spraying in Burgerjon's tower with 10 mg of active ingredient in solution with distilled water and emulsion, and in unmodified Petri dishes (60 x 20 mm) with oil (different concentrations) diluted in 1 mL of ethanol. Ratio selection was obtained as: LD50 of Apis mellifera/ LD50 of V. destructor. Determination of Minimal Inhibitory Concentrations (MIC): to P. larvae was tested at concentrations of 25, 50, 100, 150, 200, 250, 350, 450, 500, 600, 650, 700, 800 and 1000 ppm. An A. apis strain was grown on agar MY20 supplemented with variable concentrations (between 0-800 ppm) of T. minuta oil was evaluated. Results obtained in tests of total exposure showed that the concentration was able to kill 50% of mites in 24 h (DL50) and was estimated to be 4.37 mg/cage. The efficacy after spray treatment reached 56%. The ratio selection was 3:11. Against P. larvae, the oil showed MIC values ranging from 700-800 µL/L depending on the tested bacterial strains. Tagetes minuta oil in agar MY20 inhibited mycelial growth of A. apis above concentrations of 200 ppm (p = 0.0001). Oil concentrations of 700 and 800 ppm achieved maximum growth inhibition of A. apis (67% of growth inhibition on average). Tagetes minuta oil demonstrated in vitro antibacterial, antifungical and miticide activity, although this oil shows a moderate inhibitor effect compared with other essential oils of native plants from Argentina. Abstract Varroa destructor is one of the biggest threats to honey bee populations worldwide. Three classes of disparately acting acaricidal compounds are currently used to combat infestations: the pyrethroid insecticides, fluvalinate and flumethrin; the organophospate, coumaphos; and the formamidine, amitraz. Unfortunately, these compounds tend to persist in both honey and wax compromising honey bee health and contaminating hive products used by humans. In addition, overuse of synthetic compounds encourages the emergence of acaricide resistance in V. destructor. Current evidence suggests that resistance to pyrethroid insecticides in V. destructor initially arose in Italy during a four-year period of heavy pyrethroid application. After that, several cases of resistant populations have been reported around the world. More data is needed to track the origin and dispersal of mites resistant to the organophosphate coumaphos, and formamidine amitraz. Little is known about the genetic procedures leading to fixation of acaricide resistance genes, which can vary depending on mite genetics, environmental conditions, and beekeeping management. Lotmaria passim (Kinetoplastea) is considered the most prevalent as well as the most virulent trypanosomatid associated to the European honey bee Apis mellifera. We used qPCR to screen for the presence of this parasite in 57 samples from ten Argentinian provinces, and were able to detect its presence throughout most of the country with 41% of the samples testing positive. In a retrospective analysis, we detected L. passim in 73% of honey bee samples from 2006 showing that this flagellate has been widely present in Argentina for at least ~15 years. Additionally, three primer sets for L. passim detection were compared, with the pair that produced smallest PCR product having the best detection capability. Finally, we also found L. passim DNA in 100% (n = 6) of samples of the mite Varroa destructor. The role of this ectoparasite in the lifecycle of Lotmaria , if any, remains unrevealed.
