Glacier and ice sheet surfaces host diverse communities of microorganisms whose activity (or inactivity) influences biogeochemical cycles and ice melting. Supraglacial microbes endure various environmental extremes including resource scarcity, frequent temperature fluctuations above and below the freezing point of water, and high UV irradiance during summer followed by months of total darkness during winter. One strategy that enables microbial life to persist through environmental extremes is dormancy, which despite being prevalent among microbial communities in natural settings, has not been directly measured and quantified in glacier surface ecosystems. Here, we use a combination of metabarcoding and metatranscriptomic analyses, as well as cell‐specific activity (BONCAT) incubations to assess the diversity and activity of microbial communities from glacial surfaces in Iceland and Greenland. We also present a new ecological model for glacier microorganisms and simulate physiological state‐changes in the glacial microbial community under idealized (i) freezing, (ii) thawing, and (iii) freeze–thaw conditions. We show that a high proportion (>50%) of bacterial cells are translationally active in‐situ on snow and ice surfaces, with Actinomycetota, Pseudomonadota, and Planctomycetota dominating the total and active community compositions, and that glacier microorganisms, even when frozen, could resume translational activity within 24 h after thawing. Our data suggest that glacial microorganisms respond rapidly to dynamic and changing conditions typical of their natural environment. We deduce that the biology and biogeochemistry of glacier surfaces are shaped by processes occurring over short (i.e., daily) timescales, and thus are susceptible to change following the expected alterations to the melt‐regime of glaciers driven by climate change. A better understanding of the activity of microorganisms on glacier surfaces is critical in addressing the growing concern of climate change in Polar regions, as well as for their use as analogues to life in potentially habitable icy worlds. Cryoconite holes are miniature freshwater aquatic ecosystems that harbor a relatively diverse microbial community. This microbial community can withstand the extreme conditions of the supraglacial environment, including fluctuating temperatures, extreme and varying geochemical conditions and limited nutrients. We analyzed the physiological capabilities of microbial isolates from cryoconite holes from Antarctica, Greenland, and Svalbard in selected environmental conditions: extreme pH, salinity, freeze-thaw and limited carbon sources, to identify their physiological limits. The results suggest that heterotrophic microorganisms in cryoconite holes are well adapted to fast-changing environmental conditions, by surviving multiple freeze-thaw cycles, a wide range of salinity and pH conditions and scavenging a variety of organic substrates. Under oxic and anoxic conditions, the communities grew well in temperatures up to 30°C, although in anoxic conditions the community was more successful at colder temperatures (0.2°C). The most abundant cultivable microorganisms were facultative anaerobic bacteria and yeasts. They grew in salinities up to 10% and in pH ranging from 4 to 10.5 (Antarctica), 2.5 to 10 (Svalbard), and 3 to 10 (Greenland). Their growth was sustained on at least 58 single carbon sources and there was no decrease in viability for some isolates after up to 100 consecutive freeze-thaw cycles. The elevated viability of the anaerobic community in the lowest temperatures indicates they might be key players in winter conditions or in early melt seasons, when the oxygen is potentially depleted due to limited flow of meltwater. Consequently, facultative anaerobic heterotrophs are likely important players in the reactivation of the community after the polar night. This detailed physiological investigation shows that despite inhabiting a freshwater environment, cryoconite microorganisms are able to withstand conditions not typically encountered in freshwater environments (namely high salinities or extreme pH), making them physiologically more similar to arid soil communities. Advances in microbial ecology in the cryosphere continue to be driven by empirical approaches including field sampling and laboratory-based analyses. Although mathematical models are commonly used to investigate the physical dynamics of Polar and Alpine regions, they are rarely applied in microbial studies. Yet integrating modelling approaches with ongoing observational and laboratory-based work is ideally suited to Polar and Alpine microbial ecosystems given their harsh environmental and biogeochemical characteristics, simple trophic structures, distinct seasonality, often difficult accessibility, geographical expansiveness and susceptibility to accelerated climate changes. In this opinion paper, we explain how mathematical modelling ideally complements field and laboratory-based analyses. We thus argue that mathematical modelling is a powerful tool for the investigation of these extreme environments and that fully integrated, interdisciplinary model-data approaches could help the Polar and Alpine microbiology community address some of the great research challenges of the 21st century (e.g. assessing global significance and response to climate change). However, a better integration of field and laboratory work with model design and calibration/validation, as well as a stronger focus on quantitative information is required to advance models that can be used to make predictions and upscale processes and fluxes beyond what can be captured by observations alone. There is now compelling evidence that microbially mediated reactions impart a significant effect upon the dynamics, composition, and abundance of nutrients in glacial melt water. Consequently, we must now consider ice masses as ecosystem habitats in their own right and address their diversity, functional potential, and activity as part of alpine and polar environments. Although such research is already underway, its fragmentary nature provides little basis for developing modern concepts of glacier ecology. This paper therefore provides a much-needed framework for development by reviewing the physical, biogeochemical, and microbiological characteristics of microbial habitats that have been identified within glaciers and ice sheets. Two key glacial ecosystems emerge, one inhabiting the glacier surface (the supraglacial ecosystem) and one at the ice-bed interface (the subglacial ecosystem). The supraglacial ecosystem is characterized by a diverse consortium of microbes (usually bacteria, algae, phytoflagellates, fungi, viruses and occasional rotifers, tardigrades, and diatoms) within the snowpack, supraglacial streams, and melt pools (cryoconite holes). The subglacial system is dominated by aerobic/anaerobic bacteria and most probably viruses in basal ice/till mixtures and subglacial lakes. A third, so-called englacial ecosystem is also described, but it is demonstrated that conditions within glacier ice are sufficient to make metabolic activity and its impact upon nutrient dynamics negligible at the glacier scale. Consideration of the surface and internal heat balances of the glacier show that all glacial ecosystems are sensitive to climate change, although at different timescales. Thus, while rapid, melt-driven habitat changes lead to melt-out, resuscitation, and redistribution of microorganisms in many supraglacial ecosystems, much slower climatic and glacial mass-balance processes effect such changes in the subglacial ecosystem. Paradoxically, it is shown that these forces have brought about net refreezing and the onset of cryostasis in the subglacial ecosystems of many Arctic glaciers subject to thinning in recent decades. The frequency of visibly phage-infected bacterial cells (FVIB) and the average number of phages per cell [i.e. burst size (BS)] were determined in Antarctic and Arctic ultra-oligotrophic freshwater environments. Water samples were collected from two Antarctic freshwater lakes and cryoconite holes from a glacier in the Arctic. Data from this bipolar study show the highest FVIB (average 26.1%, range 5.1% to 66.7%) and the lowest BS (average 4, range 2–15) ever reported in the literature. The bacterial density is low in these ultra-oligotrophic freshwater environments but a large proportion of the bacteria are visibly infected. Our results suggest that a constant virioplankton population can be maintained in these extreme environments even though host density is low and often slow growing. [ABSTRACT FROM AUTHOR]

Microbial communities and soil carbon (C) have been shown to vary in response to increasing vegetation cover during soil development after deglaciation. However, little is known about the ability of microorganisms to utilize various C sources in glacier forefield soils. We supplied ecologically relevant 13 C-labeled C sources ( Chlorella , Penicillium and Festuca ) to three distinct environments (supraglacial sediments, barren soils and vegetated soils) of the Damma glacier area to monitor 13 CO 2 production. We identified prokaryotic and fungal populations able to utilize these sources by using DNA-stable isotope probing coupled with Illumina MiSeq sequencing of ribosomal markers. A high initial 13 CO 2 pulse indicated that 13 C-labeled microbial and plant material were consumed. The 13 C-enriched DNA results indicated that betaproteobacterial taxa affiliated to the families Oxalobacteraceae and Comamonadaceae were important players in C utilization from different sources and present in all environments. In contrast, different fungal taxa played different roles in C degradation depending on the soil environment. Overall, our findings reveal that C utilization is driven by similar prokaryotic populations along a glacier forefield, while the distribution of active fungal populations are more influenced by environmental factors. Abstract: The contribution of biofilm and artificial feeds for the growth of the pink shrimp Farfantepenaeus paulensis reared in tanks and net cages were evaluated by using stable isotope (δ 13C and δ 15N) analysis. In the net cage experiment, mass balance and mixing equations showed that the biofilm contribution to shrimp growth was over 49% of carbon and 70% of nitrogen. These data suggest that the artificial feed offered in this experiment, although ingested, was not properly incorporated in the shrimp tissue. In contrast, biofilm growing in tanks contributed little to the nitrogen incorporated by the shrimp (maximum of 29%). In this experiment, shrimp presented an unusually large isotope fractionation. These results imply that the shrimp are selectively ingesting some items from the biofilm, or differentially assimilate specific items of the artificial feed, due to differential digestibility. The hypothesis of selective feeding of biofilm microorganisms was corroborated in the experiment with net cages, where shrimp preferentially consumed centric diatoms. This study also demonstrated that shrimp biofilm consumption and the presence of artificial feed largely influenced biofilm formation. The consumption of centric diatoms by shrimp seem to stimulate biofilm chlorophyll-a, while biofilm δ 15N values measured in tanks indicate that microorganisms present in the biofilm absorbs nitrogen released by artificial feed after its decomposition.
