Shrub removal is a common management method in forest ecosystems, but comparatively little is known regarding the effects of shrub removal on soil microbial communities among primary forest, secondary forest, and plantation forests in temperate forests, which limits our accurate assessment of sustainable management of understory vegetation removal. Given this, we used a long-term operation experiment across a contrasting mixed broadleaved-Pinus koraiensis forest, Betula platyphylla forest, and Larix gmelinii plantation forest to explore the variations of soil properties and microbial community after 5 years of shrub removal on Changbai Mountain, as well as the contribution of the soil properties and understory plant diversity to the soil microbial community. The results demonstrated that shrub removal could significantly alter soil SWC and TN, TP, and AP contents of the L. gmelinii, as well as N/P of B. platyphylla. Moreover, shrub removal also clearly improved soil bacterial Pielou_e index and Simpson index of mixed broadleaved-P. koraiensis and soil bacterial Simpson index of L. gmelinii, and decreased soil fungal Pielou_e index and Shannon index of L. gmelinii and soil bacterial Pielou_e index and soil fungal Shannon index of B. platyphylla. Identically, shrub removal notably altered the soil bacterial community composition. Soil characteristics and understory plant diversity accounted for 48.02% and 26.88%, and 45.88% and 27.57% of the variance in the bacterial and fungal community composition, respectively. Pine wood nematode, Bursaphelenchus xylophilus, as one of the greatest threats to pine trees, is spreading all over the world. Plant microorganisms play an important role in the pathogenesis of nematodes. The phyllosphere and rhizosphere bacterial and fungal communities associated with healthy Pinus koraiensis (PKa) and P. koraiensis infected by B. xylophilus at the early (PKb) and last (PKc) stages were analyzed. Our results demonstrated that pine wood nematode (PWD) could increase the phyllosphere bacterial Pielou_e, Shannon, and Simpson index; phyllosphere fungal Chao 1 index, as well as rhizosphere bacterial Pielou_e, Shannon, and Simpson index; and rhizosphere fungal Pielou_e, Shannon, and Simpson index. What's more, slight shifts of the microbial diversity were observed at the early stage of infection, and the microbial diversity increased significantly as the symptoms of infection worsened. With the infection of B. xylophilus in P. koraiensis, Bradyrhizobium (rhizosphere bacteria), Massilia (phyllosphere bacteria), and Phaeosphaeriaceae (phyllosphere fungi) were the major contributors to the differences in community compositions among different treatments. With the infection of PWD, most of the bacterial groups tended to be co-excluding rather than co-occurring. These changes would correlate with microbial ability to suppress plant pathogen, enhancing the understanding of disease development and providing guidelines to pave the way for its possible management. Overexploitation of magnesium has caused serious environmental pollution, and the ecological restoration of magnesite mines has attracted increasing attention in China. The aim of this study was to assess the effects of phytoremediation with Robinia pseudoacacia (RP) on soil quality and microbial communities in magnesite mines. Here, soil samples were collected from restored areas (R. pseudoacacia), natural forest (NF), and unrestored areas (CK), and the soil microbial communities were assessed by Illumina high‐throughput sequencing. Phytoremediation with R. pseudoacacia could alleviate soil alkalization and contribute to the accumulation of soil nutrients. A total of 531,329 effective 16S rRNA and 596,344 valid Internal Transcribed Spacer rRNA gene sequences were obtained, which were classified into 30 bacterial and 12 fungal phyla. The predominant bacterial and fungal phyla were Proteobacteria and Ascomycota, respectively. Bacterial diversity indices of RP were higher than those of the CK, both of which were lower than those in the NF site. While, fungal Chao1 and ACE (abundance‐based coverage estimator) indices of RP were higher than those of the CK and even surpassed those in the NF site. Soil microbial communities significantly differed among different treatments, and the effects of soil factors on microbial communities were comprehensive and not determined by a single factor. Generally, these results indicated that phytoremediation with R. pseudoacacia could effectively improve the soil environment of the abandoned magnesium mine, which provide useful insights and practical references for the suitable species that would be planted on magnesite mine for restoration. Different revegetatiom types can affect the chemical composition of soil organic carbon (SOC), soil microbial community and the functional genes related to carbon cycle. However, the relationships between SOC chemical functional groups and soil microbial communities and the functional genes remains poorly unclear under different revegetation types. Using the solid-state 13C nuclear magnetic resonance (NMR) spectroscopy, we examined changes in the SOC chemical composition of five soils (0–10 cm depth) from Larix gmelinii Rupr. (LG), Pinus koraiensis Sieb. (PK), Quercus mongolica Fisch. (QM), Juglans mandshurica Maxim. (JM), and conifer-broadleaf forest (CB). And the soil microbial community genes related to metabolism of macro-molecular compounds were determined via whole genome shotgun based on Illumina HiSeq. Our results indicated that broadleaf forests (JM, QM) had increased the contents of soil total carbon (C), total nitrogen (N), dissolved organic carbon (DOC), and microbial biomass carbon (MBC), compared with coniferous forests (LG, PK) and the conifer-broadleaf forest (CB). While, the coniferous forests generated a lower O-alcoxyl C, a higher alkyl C, and the ratio of alkyl C/O-alkyl C than broadleaf forests. A total of four kingdoms were identified via whole metagenome shotgun sequencing, including eight archaea, 55 bacteria, 15 eukaryota, and two viruses, giving a total 80 phyla. The contents of alkyne C, phenolic C, methoxyl C, COO/NC=O, and alkyl C were strong related to the composition of soil microbial community and their contents illuminated a major part of the variation in soil microbial composition. We detected seven corresponding macro-molecular compounds of different organic carbon functional group, and 244 genes related to metabolism across all samples, and soil total C, total N, and DOC could be the main factors for microbial functional gene composition. Interestingly, the relative abundances of different SOC chemical functional groups, the phylogenetic distance for microbes, the genes of C cycling based on the KEGG database, and the relative abundance of genes related to metabolism of macro-molecular compounds of different SOC chemical functional groups under different revegetation types all could be divided into three groups, including PK plus LG, JM plus QM, and CB. Our results also illustrated that variations in SOC chemical functional groups were strongly associated with changes of soil microbial community taxa and functional genes, which might be affected by the changes of soil characteristics.
