Microalgae biofilms have been demonstrated to recover nutrients from wastewater and serve as biomass feedstock for bioproducts. However, there is a need to develop a platform to quantitatively describe microalgae biofilm production, which can provide guidance and insights for improving biomass areal productivity and nutrient uptake efficiency. This paper proposes a unified experimental and theoretical framework to investigate algae biofilm growth on a rotating algae biofilm reactor (RABR). Experimental laboratory setups are used to conduct controlled experiments on testing environmental and operational factors for RABRs. We propose a differential-integral equation-based mathematical model for microalgae biofilm cultivation guided by laboratory experimental findings. The predictive mathematical model development is coordinated with laboratory experiments of biofilm areal productivity associated with ammonia and inorganic phosphorus uptake by RABRs. The unified experimental and theoretical tool is used to investigate the effects of RABR rotating velocity, duty cycle (DC), and light intensity on algae biofilm growth, areal productivity, nutrient uptake efficiency, and energy efficiency in wastewater treatment. Our framework indicates that maintaining a reasonable light intensity range improves biomass areal productivity and nutrient uptake efficiency. Our framework also indicates that faster RABR rotation benefits biomass areal productivity. However, maximizing the nutrient uptake efficiency requires a reasonably low RABR rotating speed. Struvite was observed within the microalgae biofilm matrix of an outdoor, pilot-scale rotating algal biofilm reactor (RABR) designed to remove nitrogen and phosphorus from municipal anaerobic digester filtrate. The bottom layer of cells (2.5-month growth) and two top layers of cells (1-week and 2.5-month growth) were evaluated on east- and west-facing sides of the RABR. Sun orientation and shading effects of upper biofilm layers impacted the species composition and microalgae content of the bottom biofilm layers. Struvite formed within the microalgae biofilm matrix, and a higher struvite content appeared to be correlated with a higher microalgae content. The highest struvite content (expressed as %wt. of total solids) was observed in the east- and west-facing bottom layers of growth and west-facing 1-week growth (5.0%, 4.3%, and 4.1%, respectively). The lowest struvite content was observed in east- and west-facing 2.5-month growth and east-facing 1-week growth (1.1%, 1.5%, and 1.1%, respectively). Despite RABR influent component ion molar ratios with potential for various magnesium and calcium precipitates, microalgae biofilm provided pH and nucleation sites favorable to struvite precipitation. This evaluation is the first in the refereed literature the authors are aware of that reports on the association of struvite formation in the presence of a microalgae biofilm.

Rotating algal biofilm reactor (RABR) technology was successfully employed in an effective strategy to couple the removal of wastewater nutrients with accumulation of valuable bioproducts by grown algae. A secondary stage municipal wastewater was fed to the developed system and the effects of the hydraulic retention time (HRT) parameter on both nutrient removal and bioproduct production were evaluated under fed-batch operation mode. Two sets of bench scale RABRs were designed and operated with HRTs of 2 and 6days in order to provide competitive environment for algal growth. The HRT significantly affected nitrogen and phosphorus uptakes along with lipid and starch accumulations by microalgae in harvested biofilms. Domination of nitrogen removal in 2-day HRT with higher lipid accumulation (20% on dried weight basis) and phosphorus removal in 6-day HRT with higher starch production (27% on dried weight basis) was observed by comparing the performances of the RABRs in duplicate runs., (Copyright © 2016 Elsevier Ltd. Methods to convert microalgal biomass to bio based fuels and chemicals are limited by several processing and economic hurdles. Research conducted in this study modified/optimized a previously published procedure capable of extracting transesterifiable lipids from wet algal biomass. This optimization resulted in the extraction of 77% of the total transesterifiable lipids, while reducing the amount of materials and temperature required in the procedure. In addition, characterization of side streams generated demonstrated that: (1) the C/N ratio of the residual biomass or lipid extracted (LE) biomass increased to 54.6 versus 10.1 for the original biomass, (2) the aqueous phase generated contains nitrogen, phosphorous, and carbon, and (3) the solid precipitate phase was composed of up to 11.2 wt% nitrogen (70% protein). Microalgal biofilms grown to evaluate potential nutrient removal options for wastewaters and feedstock for biofuels production were studied to determine the influence of bicarbonate amendment on their growth, nutrient uptake capacity, and lipid accumulation after nitrogen starvation. No significant differences in growth rates, nutrient removal, or lipid accumulation were observed in the algal biofilms with or without bicarbonate amendment. The biofilms possibly did not experience carbon-limited conditions because of the large reservoir of dissolved inorganic carbon in the medium. However, an increase in photosynthetic rates was observed in algal biofilms amended with bicarbonate. The influence of bicarbonate on photosynthetic and respiration rates was especially noticeable in biofilms that experienced nitrogen stress. Medium nitrogen depletion was not a suitable stimulant for lipid production in the algal biofilms and as such, focus should be directed toward optimizing growth and biomass productivities to compensate for the low lipid yields and increase nutrient uptake., (Copyright © 2015 Elsevier Ltd. Microalgae have gained considerable attention recently as a sustainable means to produce biofuels and bioproducts. It has previously been demonstrated that single strain microalgae can be harvested and processed through a wet lipid extraction procedure (WLEP). After WLEP processing, acetone, butanol, ethanol, and biodiesel can be produced, and growth of recombinant Escherichia coli can be achieved from the microalgae. This study demonstrates the application of different wastewater microalgae harvesting techniques and processing through WLEP on the production of polyhydroxybutyrate (PHB) by E. coli. The harvesting techniques include: cationic potato starch (CPS), cationic corn starch (CCS), aluminum sulfate, and centrifugation. The microalgae-based media were used to grow E. coli to ∼10(13)CFU/mL and produce approximately 7.8% of dry cell weight as PHB. Microalgal biofilm based technologies are of keen interest due to their high biomass concentrations and ability to utilize light and CO2. While photoautotrophic biofilms have long been used for wastewater remediation, biofuel production represents a relatively new and under-represented focus area. However, the direct measurement and characterization of fundamental parameters required for industrial control are challenging due to biofilm heterogeneity. This study evaluated oxygenic photosynthesis and respiration on two distinct microalgal biofilms cultured using a novel rotating algal biofilm reactor operated at field- and laboratory-scales. Clear differences in oxygenic photosynthesis and respiration were observed based on different culturing conditions, microalgal composition, light intensity and nitrogen availability. The cultures were also evaluated as potential biofuel synthesis strategies. Nitrogen depletion was not found to have the same effect on lipid accumulation compared to traditional planktonic microalgal studies. The potential of microalgae as a source of sustainable energy, nutritional supplements and specialized chemicals necessitates a thorough evaluation of the methods of harvesting microalgae with regards to the bioproduct(s) desired. This research assessed the effect of coagulation, flocculation, and centrifugation on the wet lipid extraction procedure, which fractionated microalgae into hydrolyzed biomass for fermentation into acetone, butanol, and ethanol, an aqueous phase as growth media for genetically engineered Escherichia coli, and a lipid fraction for the production of biodiesel. Biomass harvested by cationic starches, alum, and centrifugation produced 30, 19, and 22.5mg/g of dry wt. algae of total combined acetone, butanol, and ethanol, respectively. Higher biodiesel production was also observed for the cationic starches (9.6 mg/g of dry wt. algae) than alum (0.6 mg/g of dry wt. algae) harvested biomass. Harvesting microalgae presents a challenge in selecting the most economical method for low cost algal bioproducts. Previous studies have shown coagulation-flocculation to be the most efficient method for large scale microalgae harvesting. This study focused on modifying native potato starch with biogenic amines and optimizing the reaction parameters. Such modification rendered the starch cationic, with an ability to destabilize microalgae suspensions or colloids. The effect of time, temperature, and reactant concentrations on the zeta potential of the cationic amino starch was studied. Biogenic amines including putrescine, histamine, cadaverine, and tyramine were selected for study based on the number of nitrogen groups in their structure. Zeta potential for histamine cationic amino starch was significantly higher (+9.0±2.0 mV) at lower reaction temperatures, regardless of the amine to starch ratio and reaction time intervals. Putrescine, cadaverine, and tyramine cationic amino starches exhibited significantly higher zeta potential values (13.76±3.60, 6.81±1.64, and 5.68±1.60 mV, respectively) with amine to starch ratio higher than reaction stoichiometry, irrespective of reaction temperature or time intervals. Microalgae are a source of renewable oil for liquid fuels. However, costs for dewatering/drying, extraction, and processing have limited commercial scale production of biodiesel from algal biomass. A wet lipid extraction procedure was developed that was capable of extracting 79% of transesterifiable lipids from wet algal biomass (84% moisture) via acid and base hydrolysis (90 °C and ambient pressures), and 76% of those extracted lipids were isolated, by further processing, and converted to FAMEs. Furthermore, the procedure was capable of removing chlorophyll contamination of the algal lipid extract through precipitation. In addition, the procedure generated side streams that serve as feedstocks for microbial conversion to additional bioproducts. The capability of the procedure to extract lipids from wet algal biomass, to reduce/remove chlorophyll contamination, to potentially reduce organic solvent demand, and to generate feedstocks for high-value bioproducts presents opportunities to reduce costs of scaling up algal lipid extraction for biodiesel production., (Copyright © 2012 Elsevier Ltd. Acetone, butanol, and ethanol (ABE) fermentation by Clostridium saccharoperbutylacetonicum N1-4 using wastewater algae biomass as a carbon source was demonstrated. Algae from the Logan City Wastewater Lagoon system grow naturally at high rates providing an abundant source of renewable algal biomass. Batch fermentations were performed with 10% algae as feedstock. Fermentation of acid/base pretreated algae produced 2.74 g/L of total ABE, as compared with 7.27 g/L from pretreated algae supplemented with 1% glucose. Additionally, 9.74 g/L of total ABE was produced when xylanase and cellulase enzymes were supplemented to the pretreated algae media. The 1% glucose supplement increased total ABE production approximately 160%, while supplementing with enzymes resulted in a 250% increase in total ABE production when compared to production from pretreated algae with no supplementation of extraneous sugar and enzymes. Additionally, supplementation of enzymes produced the highest total ABE production yield of 0.311 g/g and volumetric productivity of 0.102 g/Lh. The use of non-pretreated algae produced 0.73 g/L of total ABE. The ability to engineer novel methods to produce these high value products from an abundant and renewable feedstock such as algae could have significant implications in stimulating domestic energy economies., (Copyright © 2012.
