Although some evidence is consistent with the notion that distinct cortical systems support memory and perception, mounting evidence supports a representational-hierarchical view of cognition, which posits that distinctions lie in simple feature representations versus more complex conjunctive representations of many stimulus features simultaneously. Thus, typical memory tasks engage different regions from typical perception tasks because they inherently test information on opposing ends of this continuum. Memory deficits are reliably reported with age, but the tasks used to make these conclusions predominantly rely on conjunctive representations. To test the extent to which age-related deficits may be accounted for by perceptual processing, this study investigated discriminations involving conjunctive representations in older adults. Results show that adults aged 50 to 77 are impaired, relative to their younger counterparts, on discriminations requiring feature conjunctions, but not simple feature representations. These findings support recent data showing an age related decline in the ability to form conjunctive representations. Furthermore, these data suggest that some 'mnemonic' deficits associated with age may in fact be the result of deficits in perception rather than memory.

Background: Aging is typically accompanied by memory decline and changes in hippocampal function. Among these changes is a decline in the activity of the dentate gyrus (DG) during behavior. Lasting memory, however, is thought to also require recapitulation of recent memory traces during subsequent rest - a phenomenon, termed memory trace reactivation, which is compromised in hippocampal CA1 with progressive age. This process has yet to be assessed in the aged DG, despite its prominent role in age-related memory impairment. Using zif268 transcription to measure granule cell recruitment, DG activity in adult and aged animals was assessed both during spatial exploration and as animals remained at rest in the home cage in order to detect potential memory-related replay., Results: Consistent with the observation of memory trace reactivation in DG, the probability that an individual granule cell transcribes zif268 during rest in the animal's home cage is increased by recent experience in a novel environment. Surprisingly, a comparable increase was observed in the probability of granule cells in the aged DG expressing zif268 during rest. Moreover, no significant age-related difference was observed in the number of granule cells expressing zif268 during rest. Thus, the number and pattern of granule cell expression of zif268 during rest is preserved in aged animals, despite a significant decline in exploration-related zif268 expression., Conclusions: These data lead to the hypothesis that the input the aged DG receives from backprojections from CA3 (the region widely hypothesized to mediate reactivation) remains functionally intact despite loss of innervation from the perforant path.

The patterns of ensemble activity in the hippocampal formation during wakeful, attentive behavior are recapitulated during subsequent resting states. This replay of activity has also been found in several brain regions across many species, indicating a very general biological phenomenon. Concomitantly, transcription of immediate-early genes (IEGs) such as Arc also reoccurs in the same hippocampal neurons, suggesting that IEGs contribute to "off-line" consolidation. If continued IEG expression during rest reflects a correlate of ensemble replay, then the same generality should be observed in IEG transcription patterns. This hypothesis was tested by examining Arc in F344 rats engaging in spatial exploration alongside a rest episode. The probability that an individual neuron participates in "constitutive" Arc expression during rest is increased by recent experience in multiple cortical regions as well as across the septal and temporal poles of the hippocampus, consistent with memory trace reactivation. That is, neurons that were recently active during spatial exploration are preferentially recruited into further Arc expression during subsequent rest. The continued Arc expression, however, occurs in only a small fraction of the cells that were engaged in transcription during previous behavior. This fraction is greatest in CA3 and progressively decreases in CA1, superficial, and deep cortical layers and is consistent with the idea that consolidation occurs rapidly in the hippocampus (centering on the CA3 recurrent network) while changes are much more gradual in neocortical synaptic networks., (Copyright © 2012 Wiley Periodicals, Inc.)

Decades of research identify the hippocampal formation as central to memory storage and recall. Events are stored via distributed population codes, the parameters of which (e.g., sparsity and overlap) determine both storage capacity and fidelity. However, it remains unclear whether the parameters governing information storage are similar between species. Because episodic memories are rooted in the space in which they are experienced, the hippocampal response to navigation is often used as a proxy to study memory. Critically, recent studies in rodents that mimic the conditions typical of navigation studies in humans and nonhuman primates (i.e., virtual reality) show that reduced sensory input alters hippocampal representations of space. The goal of this study was to quantify this effect and determine whether there are commonalities in information storage across species. Using functional molecular imaging, we observe that navigation in virtual environments elicits activity in fewer CA1 neurons relative to real-world conditions. Conversely, comparable neuronal activity is observed in hippocampus region CA3 and the dentate gyrus under both conditions. Surprisingly, we also find evidence that the absolute number of neurons used to represent an experience is relatively stable between nonhuman primates and rodents. We propose that this convergence reflects an optimal ensemble size for episodic memories.SIGNIFICANCE STATEMENT One primary factor constraining memory capacity is the sparsity of the engram, the proportion of neurons that encode a single experience. Investigating sparsity in humans is hampered by the lack of single-cell resolution and differences in behavioral protocols. Sparsity can be quantified in freely moving rodents, but extrapolating these data to humans assumes that information storage is comparable across species and is robust to restraint-induced reduction in sensory input. Here, we test these assumptions and show that species differences in brain size build memory capacity without altering the structure of the data being stored. Furthermore, sparsity in most of the hippocampus is resilient to reduced sensory information. This information is vital to integrating animal data with human imaging navigation studies.

Spontaneous novelty preference is apparent in a wide array of animals, including mammals, birds, reptiles, and fish. This provides a powerful behavioral assay to assess whether an animal can recognize a diverse array of stimuli in a common paradigm. Surprisingly, no research has been conducted in birds using novelty approach under conditions comparable to the spontaneous object recognition (SOR) protocols that have become standard across other animals. To correct this, the current study adapts a number of SOR protocols commonly used in mammals to characterize novelty approach in Silver King pigeons and Japanese quail. We show that, in general, both quail and pigeons readily approach novel objects or locations when tested using SOR protocols, although pigeons show a neophilic response under some conditions in which quail do not. Neither quail nor pigeons readily approach objects in novel contexts or novel locations. These data show that SOR can be successfully adapted to birds, allowing for more direct comparison between mammals and birds in tasks of shared ecological relevance. [ABSTRACT FROM AUTHOR]

In mammals, episodic memory and spatial cognition involve context-specific recruitment of unique ensembles in the hippocampal formation (HF). Despite their capacity for sophisticated spatial (e.g., for migration) and episodic-like (e.g., for food-caching) memory, the mechanisms underlying contextual representation in birds is not well understood. Here we demonstrate environment-specific Egr1 expression as male brown-headed cowbirds (Molothrus ater) navigate environments for food reward, showing that the avian HF, like its mammalian counterpart, recruits distinct neuronal ensembles to represent different contexts. The nature of memory is a central issue in neuroscience. How does our representation of the world change with learning and experience? Here we use the transcription of Arc mRNA, which permits probing the neural representations of temporally separated events, to address this in a well characterized odor learning model. Rat pups readily associate odor with maternal care. In pups, the lateralized olfactory networks are independent, permitting separate training and within-subject control. We use multiday training to create an enduring memory of peppermint odor. Training stabilized rewarded, but not nonrewarded, odor representations in both mitral cells and associated granule cells of the olfactory bulb and in the pyramidal cells of the anterior piriform cortex. An enlarged core of stable, likely highly active neurons represent rewarded odor at both stages of the olfactory network. Odor representations in anterior piriform cortex were sparser than typical in adult rat and did not enlarge with learning. This sparser representation of odor is congruent with the maturation of lateral olfactory tract input in rat pups. Cortical representations elsewhere have been shown to be highly variable in electrophysiological experiments, suggesting brains operate normally using dynamic and network-modulated representations. The olfactory cortical representations here are consistent with the generalized associative model of sparse variable cortical representation, as normal responses to repeated odors were highly variable (~ 70% of the cells change as indexed by Arc). Abstract: The human hippocampus supports the formation of episodic memory without confusing new memories with old ones. To accomplish this, the brain must disambiguate memories (i.e., accentuate the differences between experiences). There is convergent evidence linking pattern separation to the dentate gyrus. Damage to the dentate gyrus reduces an organism''s ability to differentiate between similar objects. The dentate gyrus has tenfold more principle cells than its cortical input, allowing for a divergence in information flow. Dentate gyrus granule neurons also show a very different pattern of representing the environment than “classic” place cells in CA1 and CA3, or grid cells in the entorhinal cortex. More recently immediate early genes have been used to “timestamp” activity of individual cells throughout the dentate gyrus. These data indicate that the dentate gyrus robustly differentiates similar situations. The degree of differentiation is non-linear, with even small changes in input inducing a near maximal response in the dentate. Furthermore this differentiation occurs throughout the dentate gyrus longitudinal (dorsal–ventral) axis. Conversely, the data point to a divergence in information processing between the dentate gyrus suprapyramidal and infrapyramidal blades possibly related to differences in organization within these regions. The accumulated evidence from different approaches converges to support a role for the dentate gyrus in pattern separation. There are however inconsistencies that may require incorporation of neurogenesis and hippocampal microcircuits into the currents models. They also suggest different roles for the dentate gyrus suprapyramidal and infrapyramidal blades, and the responsiveness of CA3 to dentate input. [Copyright &y& Elsevier]

Immediate-early genes (IEGs) are tightly coupled to cellular activity and play a critical role in regulating synaptic plasticity. While encoding spatial experience, hippocampal principal cells express IEGs in a behaviorally dependent and cell-specific manner. This expression can be detected through the use of cellular compartment analysis of temporal activity by fluorescence in situ hybridization to generate estimates of cellular activity that match direct neuronal recording under comparable conditions. During rest, IEG expression continues to occur in a small number of cells, and the role of this basal expression is unknown. Imaging IEGs expressed during exploration and adjacent rest periods reveals that "constitutive" IEG expression during rest is not random. Rather, consistent with proposed memory consolidation mechanisms, it recapitulates a subset of the pattern generated by recent experience. [ABSTRACT FROM AUTHOR]

Abstract: In order to develop a profile of how individual synapses in the hippocampal formation alter their structure following learning experience, a meta-analysis synthesized the available literature on morphological change following hippocampal-dependent learning. Analysis of the 132 calculated effect sizes suggest a consistent profile of morphological change in the hippocampus following learning experience. Across the hippocampal formation, dendritic complexity, spine density, and the size of perforated postsynaptic densities showed consistent increases following training. Both the density of synapses in general and perforated synapses in particular showed unique responses to training, depending on the duration of training and/or different cell layers of the hippocampal formation. Most importantly, it seems that this profile, while consistent, is small and specific—only a select few of the morphological parameters typically measured in anatomical studies of plasticity showed significant change following training. Collectively, these data suggest that the distinct electrophysiological properties of neocortical versus hippocampal synapses may be at least partially mediated by distinct morphological cascades. That is, on the basis of theory, and with the support of the current data, it seems that synaptogenesis correlates with enduring neocortical plasticity, while structural changes correlate with more transient hippocampal plasticity. To be able to state these conclusions with conviction, however, more data are needed in several key areas for continued pursuit of the morphological correlates of hippocampal-dependent learning. Abstract: Describing the neural mechanisms underlying learning and memory continues to be an intensive area of study within neuroscience. Of specific interest are changes in synaptic number and structure in the neocortex, which may play a distinct role in learning and memory. As such, characterizing the structural correlates of neocortical learning and memory may be critical to understanding the link between synaptic structure and function. Towards this understanding, a meta-analysis was conducted on several well-researched paradigms of behavioral plasticity, categorized by those which enhance or deprive plasticity-inducing experience (PIE). Results revealed several distinct groups. Several variables (spine size, density of multisynaptic terminals, vesicular content) showed distinct dynamics under enhanced vs. deprived PIE, but changed consistently within these categories, regardless of the manipulation. A second set of variables (i.e., density of excitatory, inhibitory, excitatory spinuous, and inhibitory spinuous synapses) showed the same qualitative changes following both enhanced and impoverished PIE. A third group (total synapse density, total basilar branches, apical spine density, total postsynaptic density size, and total bouton size) showed significant heterogeneity that could not be accounted for by partitioning enhancement and deprivation of PIE. However, this variance was accounted for by the modality and duration of the manipulation, the delay between this manipulation and sacrifice, and the stereological/methodological rigor of the study. These data, along with suggestions for future investigation based on gaps in the literature may go far towards the goal of relating neural structure and function. [Copyright &y& Elsevier]

Molecular and cellular studies of the mechanisms underlying mammalian learning and memory have focused almost exclusively on postsynaptic function. We now reveal an experience-dependent presynaptic mechanism that modulates learning and synaptic plasticity in mice. Consistent with a presynaptic function for endogenous H-ras/extracellular signal-regulated kinase (ERK) signaling, we observed that, under normal physiologic conditions in wild-type mice, hippocampus-dependent learning stimulated the ERK-dependent phosphorylation of synapsin I, and MEK (MAP kinase kinase)/ERK inhibition selectively decreased the frequency of miniature EPSCs. By generating transgenic mice expressing a constitutively active form of H-ras (H-rasG12V), which is abundantly localized in axon terminals, we were able to increase the ERK-dependent phosphorylation of synapsin I. This resulted in several presynaptic changes, including a higher density of docked neurotransmitter vesicles in glutamatergic terminals, an increased frequency of miniature EPSCs, and increased paired-pulse facilitation. In addition, we observed facilitated neurotransmitter release selectively during high-frequency activity with consequent increases in long-term potentiation. Moreover, these mice showed dramatic enhancements in hippocampus-dependent learning. Importantly, deletion of synapsin I, an exclusively presynaptic protein, blocked the enhancements of learning, presynaptic plasticity, and long-term potentiation. Together with previous invertebrate studies, these results demonstrate that presynaptic plasticity represents an important evolutionarily conserved mechanism for modulating learning and memory. Abstract: It is well known that antidepressants both improve mood and increase the rate at which the dentate gyrus (DG) generates new neurons. In addition to the implications of neurogenesis for mood regulation, the production and survival of granule cells has also been implicated in learning and memory. Despite this evidence, the results of studies on the effect of antidepressants on memory have been mixed. A critical piece of data that may be missing from previous studies, however, is insight into (a) the location that newborn neurons migrate to following fluoxetine administration and (b) their ability to express normal patterns of activity-related genes. Here we demonstrate a finding that may resolve the discrepancy in the effects fluoxetine-induced neurogenesis on mood and memory: after 5 weeks delay, the net additional neurons generated in animals given the antidepressant fluoxetine during treatment are functionally normal, but preferentially accumulate (due to changes in migration and/or survival) in an area of the DG that is not recruited by spatial memory tasks.
