A quantitative spectral analysis of the ultraviolet (UV) broad excitation bands, which are located in the range 300–400 nm, for red emissions at around 610 nm in Pr-doped CaTiO3, SrTiO3:Al and BaTiO3:Mg phosphors has been carried out using a peak fitting technique. The obtained results demonstrate that the UV broad band of CaTiO3:Pr consists of four primary excitation bands centered around 330, 335, 365 and 380 nm and those of both SrTiO3:Al and BaTiO3:Mg consist of three primary bands centered around 310, 345 and 370 nm. Based on the behavior patterns and the values of the respective primary excitation bands’ parameters, i.e. center gravity (λtop), maximum height (Imax) and full-width at half-maximum (FWHM), the UV-to-red relaxation processes in these titanate phosphors can be explained to be essentially the same, except for the existence of an additional relaxation pathway via electron-trap states in CaTiO3:Pr, which gives a characteristic shape of its UV excitation spectrum in the wavelength range of >360 nm.

Various tRNA transcripts were constructed to study the identity elements of Escherichia coli tRNA(Asp). Base substitutions from G34 to U34 at the first position of the anticodon, and from U35 to A35 at the second, severely impaired the aspartate charging activity. The activity was also decreased, but in a more moderate fashion, by base changes at G2-C71, C36 and C38. Identity nucleotides of tRNA(Asp) are distributed in a different fashion between E. coli and yeast, which occur at the second base pair of the acceptor stem, G10-U25 base pair in the D-stem and 3' half of the anticodon loop.

To investigate the identity determinants of E. coli threonine tRNA, various transcripts were prepared by in vitro transcription system with T7 RNA polymerase. Substitutions of the anticodon second letter G35 and the third letter U36 to other nucleotides led to a remarkable decrease of threonine charging activity. Charging experiments with a series of anticodon-deletion transcripts also suggest the importance of the G35U36 sequence. A mutation at either the G1-C72 or C2-G71 base pair in the acceptor stem seriously affected the threonine charging activity. These results indicate that the second and third positions of the anticodon and the first and second base pairs in the acceptor stem are the recognition sites of E. coli tRNA(THR) for threonyl-tRNA synthetase. Discriminator base, A73, is not involved in threonine charging activity.

Various tRNA transcripts were constructed to study the identity elements of E.coli tRNA(Arg) and tRNA(Lys). Exchange of the anticodon of the major tRNA(Arg) from ACG to either CCG or CCU did not result in a significant loss of arginine acceptor activity, whereas not only that to UUU but also that to ACA or ACC decreased the activity. Base substitutions and deletion at A20 also impaired the arginine charging activity by over 50-fold. Arginine charging activity was introduced by either substitution of the anticodon from UAC to ACG in tRNA(Val) or from UUU to UCU in tRNA(Lys). Only a single base substitution at the third position of tRNA(Trp) anticodon (CCA) from A to G also gave rise to arginine charging activity, which was elevated to a comparable level to that of the tRNA(Arg) transcript by an additional A20 insertion. Base substitutions of the major tRNA(Arg) at the discriminator position into pyrimidines led to a decrease by factors of three to four. These data show that the third letter of the anticodon G36 or U36 besides the second letter C35 and the A20 in the variable pocket is responsible for the arginine acceptor identity, to which the discriminator base A73 or G73 contributes in an auxiliary fashion. In contrast to the arginine system, the transcript with the wild-type tRNA(Lys) sequence showed only 140-fold lower lysine charging activity than the native tRNA(Lys), suggesting the involvement of base modifications in recognition. Replacement of the anticodon UUU with not only UCU and UAC but also UUA and UUC seriously affected the lysine acceptor activity, and those with GUU and UUG also decreased by factors of 17 and 5, respectively. Introduction of UUU into the anticodons conferred lysine charging activity upon both tRNA(Val) and tRNA(Arg). Substitution of the discriminator base A73 by any of the other bases decreased the lysine acceptor activity by a factor of ten. These results indicate the involvements of all the three bases of the anticodon and A at the discriminator position in lysine specific aminoacylation.

In order to study the identity elements of valine tRNA, various transcripts of E. coli valine tRNA mutants were constructed. Both mutations at the second letter A35 of the anticodon and at the discriminator base A73 seriously damaged valine charging activity. Mutations at either the G3-C70 or U4-A69 base pairs in the acceptor stem also affected the activity. Only one nucleotide substitution of the second letter G35 of the anticodon with A35 brought an 18% valine charging activity into alanine tRNA, which acquired an almost full charging activity with valine by introducing an additional change at those two base pairs in the acceptor stem. These results indicate that the second letter A35 of the anticodon, discriminator base and acceptor stem are involved in recognition by valyl-tRNA synthetase.

To investigate the sequence requirements of Escherichia coli tRNA(Ser) for recognition by seryl-tRNA synthetase, various mutants of unmodified tRNA(Ser) were constructed. Substitution of G2.C71 by C2.G71, but not by A2.U71 or U2.A71, impaired the serine-accepting activity, indicating that this position is not involved in recognition by seryl-tRNA synthetase, but contributes to discrimination from other tRNAs processing C2.G71 such as tRNA(Leu). Other nucleotides characteristic of tRNA(Ser), including the discriminator base, were not involved in recognition by seryl-tRNA synthetase. The anticodon was not involved, as suggested by its sequence variety within the isoacceptors. The long variable arm composed of over ten nucleotides, which is a characteristic feature of tRNA(Ser) together with tRNA(Leu) and tRNA(Tyr), was stem-length-specifically, but not sequence-specifically, important for recognition. In order to introduce a sufficient serine-accepting activity to a tRNA(1LEU) transcript in vitro, besides the change from C2.G71 to G2.C71, the following elements had to be changed to those characteristic of tRNA(Ser): the sequence in the D-loop, the stem pairing pattern of the variable arm, the tertiary base-pair 15.48 and the nucleotide at position 59 in the T psi C-loop. None of the nucleotides at these changed positions was involved in base-specific recognition, indicating that seryl-tRNA synthetase selectively recognizes tRNA(Ser) on the basis of its characteristic tertiary structure rather than the nucleotides specific to tRNA(Ser).

In order to study how Escherichia coli leucyl-tRNA synthetase recognizes tRNA(Leu) and discriminates it from the other two class II tRNAs, tRNA(Ser) and tRNA(Tyr), various mutations were introduced into class II tRNA transcripts. The discriminator base A73, but not the anticodon sequence, was found to serve as a critical recognition element of tRNA(Leu). A base substitution at the invariant nucleotide A14, but not at any of the other nucleotides characteristic of the E. coli tRNA(Leu) isoacceptors among the three class II tRNAs, caused significantly damaged aminoacylation with leucine. A two base-pair deletion in the long variable arm also resulted in no significant decrease of activity. Transplanting the three tertiary elements characteristic of E. coli tRNA(Leu) (i.e. the location of the G18G19 sequence in the D-loop, the A15 U48 base-pair and the stem pairing pattern of the long variable arm) besides the discriminator base change introduced the leucine charging activity in terms of Vmax/Km, up to 0.1 of that for the normal sequence of tRNA(Leu) into both tRNA(Ser) and tRNA(Tyr). These results indicate that A73 and A14 (or its vicinity) are involved in recognition by leucyl-tRNA synthetase, and that several tertiary elements play a significant role in the discrimination of tRNA(Leu) from the other two class II tRNAs.

Studies using the T7 transcription system revealed that the discriminator base A73 and the G20 in the variable pocket play important roles in the Escherichia coli alanine tRNA identity. The C60 in the T-loop, which is unique to alanine tRNA, was not found to be crucial for alanine identity. Anticodon replacement into the valine anticodon UAC did not decrease alanine charging activity, and no alanine charging activity was detected in the mutant valine tRNA possessing the alanine anticodon UGC.

Enterotoxigenic Escherichia coli strains were isolated from the feces of 34.4% of 64 diarrheaic neonatal piglets on seven farms. Of a total of 518 isolates, 86 (16.6%) were enterotoxigenic and grouped into four phenotypes: K99+ST+ (K99 pilus antigen and heat-stable enterotoxin producing, 36 strains), ST+ (37 strains), K88+LT+ (K88 pilus antigen and heat-labile enterotoxin producing, 11 strains), and K88+ST+ (2 strains). K99+ST+ and ST+ isolates showed multiple drug resistance and most of those (58.3% and 56.8%, respectively) belonged to O serogroup 101. A K99+ST+ isolate proved to be capable of inducing diarrhea and death in hysterectomy-produced colostrum-deprived piglets when orally inoculated.

An Escherichia coli strain ZP118 of porcine origin was shown to harbor a 68-megadalton (Md) plasmid coding for a colonization factor K99 and heat-stable enterotoxin (STa), and a self-transmissible 51-Md plasmid coding for drug resistance. One of the transconjugants obtained by mating between ZP118 and E. coli C was found to harbor a 90-Md plasmid coding for K99, STa and drug resistance. Restriction endonuclease analysis suggested that the 90-Md plasmid could be a recombinant between the 68-Md plasmid and the 51-Md plasmid.

The role of "theoretical physics" in the elucidation of the origin of life has been advanced by Eigen's hypercycle theory on ancient protein synthesis system and by Dyson's random drift theory on the autocatalytic formation of enzymes, although these approaches from the informational side involve many problems to be resolved. On the other hand, our C4N model calls for a stronger role of physico-chemistry in the understanding of the origin of life. Plant virus and movable genes could be the fossil vestiges of the ancient protein synthetic system, where tRNA identical to mRNA identical to gene. It is pointed out that functional differentiation of proteins closely correlates with the structure of the primitive tRNAs. Possible initial forms of the primitive tRNAs used by the earliest organisms are explicitly shown, in which the above correlation is carefully taken into account. The system of the primitive genes and the primitive enzymes in the proto-cellular membrane, say the marigranule, might simulate the earliest organism.

There has not been elucidated the conclusive resistance mechanism of macrolide antibiotics (Mac) in Staphylococcus aureus of clinical sources. It was reported that its main mechanism may be dependent upon the impermeability of Mac into cells of S. aureus. It was also reported that the ribosomes from in vitro developed erythromycin-resistant mutants decrease their affinities for erythromycin in Bacillus subtilis and Esherichia coli. Whereas, it was reported that spiramycin (SP) -resistance did not affact the SP-binding to ribosomes in Bacillus subtilis. It has been also known that there are differences in biochemical mechanisms of drug-resistance between in vitro deqeloped resistant strains and those from clioical sources in either chloramphenicol or srreptomycin.Therefore, I have started studies of the resistance mechanism of Mac in staphylococci of clinical sources and concluded here that that the resistance mechanism of Mac depends upon decreaseing of the binding affinity of ribosomes (50S subunits) to Mac as followings : 1) There was elucidated the crrelation among the resistance level of SP, the decrease of SP-binding affinity of various levels of Mac resistance.2) It was indi/cated that the SP-binding affinity of ribosomes (50S subunits) are decreased not only by mutation but by a reversible induction in the inducible resistant strains. This reversible alteration seems to be interesting for one of regulatory mechanisms.3) It was indicated that in vitro developed resistance of SP mainly depends upon impermeability of SP SP into S. aureus cells differently from that of clinical souces.4) lt was isolated the mutant of which the Mac resistance is induced by high temperature (42°C). And it was indicated that this induced resistance also acc/ompanies the decrease of SP-binding affininity of ribosomes.

Some absorption features in the ultraviolet spectrum of Venus observed by the OAO-2 cannot be interpreted in terms of H2SO4. Carbon suboxide polymer has a yellow colour and absorption at 2000 A. Fine graphite grains have an absorption band at about 2175 A as is well known in the case of the interstellar extinction curves. A mixture of these substances which is inevitably formed in the Venus atmosphere by photochemical reactions is the best candidate for explaining the Venus absorption features in the ultraviolet.

Unmodified E. coli tRNA Asp was obtained by in vitro transcription with T7 RNA polymerase from a plasmid carrying a tRNA Asp sequence adjacent to the T7 promoter. The transcript exhibited almost the same level of amino acid acceptor activity as intact tRNA Asp , and the kinetic parameters were also similar. However, substitution of the discriminator base (the fourth base from the 3′-end) markedly affected the amino acid acceptor activity. These results suggest that the discriminator base in tRNA Asp plays an important role in the recognition of aspartyl-tRNA synthetase.

Escherichia coli isolates from calves with diarrhea (1 day to 8 weeks old, 140 individuals) were surveyed for the three immunologically distinct fimbrial adhesins FY, 31A, and K99. Of a total of 1,370 strains isolated, 96 (7.0%), 34 (2.5%), 75 (5.5%), and 13 (0.9%) were identified as FY+, 31A+, FY+.31A+, and K99+, respectively. The K99+ strains also manifested heat-stable enterotoxin production (ST+), while FY+, 31A+, and FY+.31A+ strains were ST-. Expression of FY and 31A was repressed at lower temperatures or poor aeration. The FY+ and 31A+ E. coli showed mannose-resistant hemagglutinating activity with bovine erythrocytes. Electron microscopy revealed that FY is a gently curled fimbria with a mean diameter of 4.2 nm, and 31A is a fimbria with a mean diameter of 5.1 nm. The molecular mass of protein subunits was found to be approximately 20 kilodaltons (Kd) and 19 Kd for FY and 31A, respectively. Lethal diarrhea of neonatal calves was induced by challenge with the combination of a K99+.ST+ E. coli strain and either a 31A+ E. coli strain or a 31A+ E. coli strain plus an FY+ E. coli strain under the experimental conditions in which lethal diarrhea was not induced by challenge with a K99+.ST+ E. coli strain alone.

Twelve weaned piglets (3-week-old) were divided into three groups according to the time of feed change and observed for diarrhea during the time they were 3 to 8 weeks of age. A total of 553 strains of Escherichia coli were isolated from rectal fecal samples and examined for heat-labile (LT) and heat-stable (ST) enterotoxins, pilus antigens (K88, K99, and 987P), hemolysin (Hly), raffinose utilization (Raf) and drug resistance. Enterotoxigenic and/or hemolytic E. coli strains appeared in the rectal feces of 5- to 6-week-old piglets with diarrhea in connection with feed change and changing temperatures. Most of the isolates showed multiple drug resistance to sulfonamides (Sa), streptomycin (Sm), chloramphenicol (Cp), tetracycline (Tc), kanamycin (Km), spectinomycin (Sp), ampicillin, and/or mercury. Enterotoxigenic E. coli isolates represented four phenotypes: K88+.LT+.Hly+.Raf+.(SaSmCpTcKmSp) (12 strains), K99+.ST+.Raf+.(TcKm) (7 strains), ST+.Raf+.(TcKm) (7 strains), and ST.+(SaSmKm) (25 strains). The drug resistance determinants were transferable concurrently and some of them mobilized the determinants for K88, LT, Hly, and Raf to an E. coli C strain.

A heat-inducible mutant, resistant to macrolide antibiotics (Mac), was isolated from Staphylococcus aureus MS537 in which Mac-resistance was induced by subinhibitory concentrations of erythromycin (EM). After induction at 42 C, this mutant acquired a high resistance to both Mac and lincomycin (LMC). Transduction and biochemical studies revealed that spiramycin (SP)-resistance in this mutant was induced by exposure to a high temperature (42 C) or by treatment with EM in broth but not in phosphate buffer. Induction did not take place when chloramphenicol (CM) was added to the induction mixture. Ribosomes from the mutant cultured at 42 C decreased their affinity for SP and consequently polypeptide synthesis on such ribosomes was not inhibited by SP, when compared with those cultured at 30 C. From these results, it was concluded that alteration of ribosomes took place after induction by exposure at high temperatures or by EM-treatment and that the mechanism of SP-resistance after induction was accounted for by a decrease in SP-binding to ribosomes.
