Purpose: to evaluate the effects of acute hypoxic hypoxia on eye tissue cells in adult rats and determining the effectiveness of 0.007 % solution of para-aminobenzoic acid (PABA) for apoptotic damage to the tissues of the ocular surface in rats.Materials and methods. In the experiment, 27 male Wistar rats (54 eyes) were used, divided into 5 groups. Group I — intact control, Group II — hypoxic control after 1 hour, Group III — hypoxic control after 3 hours, Group IV — with the injection of the drug 1 hour after hypoxia, Group V — injection of the drug 24 hours before hypoxia. The identification of apoptotic cells in the tissues of the eye was performed by the TUNEL method on frozen sections of the eye stained with Hoechst 33342 nuclear fluorescent dye. The localization and fluorescence intensity of the damaged cells was analyzed under a fluorescence microscope using the Image J. computer program.Results. Under the conditions of simulated acute hypoxic hypoxia, apoptotic lesions of the conjunctiva, corneal epithelium, choroid, and photoreceptor layer of the retina were observed. In group II, the number of damaged cells in the conjunctiva was 67 %, and in group III it was 120 % more compared to group I — the norm (p < 0.05). With the injection of PABA before hypoxia, there were no significant differences compared with the norm (group I) in the state of the affected cells in the conjunctiva. In the group of animals (group III) with the injection of PABA after hypoxia after 1 h, the number of damaged cells does not statistically differ from group II (hypoxic control after 1 h), but significantly lower than in group III (hypoxic control after 3 hours). The same pattern is observed in the corneal epithelium.Conclusion. Para-aminobenzoic acid (0.007 %) has a prophylactic and therapeutic effect, preventing and stabilizing the development of apoptosis of conjunctival cells and anterior corneal epithelium induced in experimental acute hypoxic hypoxia.

Retinal pigment epithelium (RPE) is a source of cells for retinal regeneration in amphibians in vivo and the development of retinal diseases in mammals and humans. Transdifferentiation of RPE cells into cells of other phenotypes is the basis for both processes: RPE cells transform into neural in the first case and into mesenchymal cells in the second case. The review describes the main stages of RPE cell transdifferentiation: initiation of the process, cell migration and proliferation, dedifferentiation, reprogramming, and specialization of cells into new directions. Information about the molecular and genetic mechanisms that ensure the passage of these stages by cells is given. Molecular participants of the regulation of transdifferentiation on the levels of the whole organism, the local cellular microenvironment (growth factors, signaling cascades), the expression of transcription factors, and the epigenome regulation are presented. Similarities and differences in the molecular and genetic mechanisms of implementation of different strategies for RPE transdifferentiation in amphibians and mammals are noted. The discovery of key molecular regulators of this choice serves both for the development of the theory of cellular reprogramming and approaches for the treatment of proliferative diseases of the human retina associated with RPE pathologies.

The review examines the role of oxidative stress (OS) in the primary response of the neural eye tissue cells to damage and degenerative processes. The imbalance (homeostasis) of the redox system towards oxidative processes underlies the development of OS. OS can be used as a part of the protective mechanism initiating the processes of healing and regeneration of damaged tissues. Violation of redox homeostasis and OS development triggers an inflammatory response and immune response in the retina. The main reactions to stress (release of ATP, calcium ions, and reactive oxygen species (ROS) into the extracellular space, attraction of exogenous immune cells, activation of endogenous macro- and microglia, apoptosis of neurons) are universal and conservative in all vertebrates. However, with the similarity of some parts of cellular and molecular processes, there are evolutionary fixed functional differences in the regenerative response of retinal cells, and the final result in different vertebrate species is not equivalent. This determines the choice of regeneration strategies: activation of endogenous stem/progenitor cells and/or reprogramming of differentiated cells (retinal pigment epithelium, Muller glia). The detection of key signaling pathways, through which the effect of OS on regenerative responses is realized after damage and pathology of the neural tissues of the vertebral eye, will contribute to the choice of optimal strategies of the cell and/or gene therapy to activate the endogenic regenerative potential of retinal neural tissue in humans.

The initial stages of apoptotic damage to the conjunctiva and corneal epithelium, in the renewing cell populations of the eye has been studied in adult rats exposed to acute hypobaric hypoxia in vivo, and the effect of para-aminobenzoic acid (PABA) administered as the Aktipol pharmaceutical preparation (0.007% PABA) has been assessed in this experimental setup. Apoptotic damage to cell DNA was aggravated after two instances of hypoxia, and this change was accompanied by an increase in the intensity of the regenerative process inferred from the expression levels of phosphorylated histone pH3, a mitotic cell marker, and phosphorylated histone γ-H2AX, a DNA repair marker. Parabulbar injection of Aktipol 24 h before hypoxia has been shown to block development of the apoptotic process and to maintain a physiologically normal level of apoptosis, and thus the preparation exerted a protective effect in hypoxia.

Nucleotide sequences were identified in the eye tissues (lens, retina, and retinal pigment epithelium) of the adult newt Pleurodeles waltl by the polymerase chain reaction with primers for the Ns gene. Sequencing showed that these nucleotide sequences belong to the Ns gene of the newt P. walt, which encodes the nucleolar protein nucleostemin. Structural analysis revealed a high homology of Ns nucleotide sequences of P. walt! with those of newts. Cynops pyrrhogaster and Notophthalmus viridescens. The expression of the Ns gene of P. walt, identified in the specialized eye cells of adult newts of the studied species, indicates that these differentiated cells retain some of the molecular characteristics inherent to the undifferentiated cells.

In this paper, we analyzed our own results and published data on the expression of regulatory genes encoding transcription factors Pax6/PAX6, Pitx2/PITX2, Fox1/FOXC1, Prox1/PROX1, Oct4/OCT4, Nanog/NANOG, and TGFβ2 signaling protein during morphogenesis of the cornea in vertebrates. We considered the results obtained for the cornea of model animals, primarily mice, and human fetal cornea. The main possibility of establishing common mechanisms of eye development in vertebrates in health and disease is comparative studies of eye morphogenesis of humans and animal models.

The effect of acute hypoxia on the occurrence of apoptosis in eye cells in rats placed in a pressure chamber was studied. Selective primary lesion of cells of the conjunctiva and the anterior corneal epithelium was found. A possible role of the simulated hypoxic conditions in the dry eye syndrome pathogenesis, which is accompanied by primary lesion of cells in the anterior eye surface tissues is discussed.

The FGF2 signaling pathway components in tissues of the posterior wall in the normal and regenerating eye of the adult Pleurodeles waltl newt were detected for the first time. The fgf2 gene expression was found in the retina, retinal pigment epithelium, and choroid using polymerase chain reaction (PCR). A high homology of the mRNA nucleotide sequence of the most conservative fgf2 gene region in the P. waltl with the fgf2 orthologs in other vertebrates was proved. The Fgf2 protein aminoacid sequence of the P. waltl newt demonstrates even more homology with this growth factor in other vertebrates. The Fgf2 protein with a molecular weight 35 kDa was found in the studied eye tissues using Western blot hybridization. Localization of the Fgf2 protein and its Fgfr receptors was immunohistochemically studied in the pigment epithelium, choroid, central and growth retina regions of the newt native eye, and in the connective cilium of photoreceptors. Using real-time PCR and immunohistochemistry methods, it was found that the fgf2 gene down-regulation and a decrease in the intensity of the immunochemical reaction of its protein product (Fgf2) occur in the early period after the retina removal (in 4-8 days) (as compared with those in the same department of the unoperated eye).

The spatiotemporal intracellular localization of the transcription factor PROXl in the human retina during prenatal development (fetal weeks 9.5 to 31) and in the adult human retina was studied for the first time. The PROXl protein was identified in the cell nuclei of the neuroblast retinal layers at the stage of active cell proliferation (fetal week 9.5) as well as in the nuclei of differentiating neurons of the inner nuclear retinal layer (horizontal, amacrine, and bipolar cells) from weeks 13 to 31 of prenatal development. The PROXl protein localization in the adult retina was the same as at the late stage of prenatal development. Our results indicate the involvement of the transcription factor PROXl in the regulation of proliferation of progenitor cells and differentiation of the inner nuclear layer cells of the human retina. These results confirm the conservative functions of Proxl/PROXl in the vertebrate retina.

Expression of genes and heat shock proteins in normal intact retina of the Spanish Ribbed Newt Pleurodeles waltl was studied using polymerase chain reaction, Western blot hybridization, and immunohis-tochemistry. It was shown that the proteins HSP70 and HSP90, as well as their encoding transcripts of relevant genes, are constitutively expressed in eye tissues. These proteins were distributed differentially, and they were characterized by expression of different levels in the retina: HSP70 dominated in the external retina, while HSP90 dominated in the internal one, in particular, in Muller glial cells and the optic nerve. Transcripts and heat shock proteins HSP70 and HSP90 were also found in the retinal pigment epithelium and eye growth zone.

The action of a current in the radio frequency range with a periodic impulse mode of modulation on the activation of recovery processes in the skin and skeletal muscles has been studied. The action of a radio frequency current with a power of 1 W, as opposed to that of the weaker action (0.1 W) and stronger (4 W) action, leads to the activation of recovery processes in the skin and skeletal muscles. Recovery processes are manifested in the increase in proliferation and activation of angiogenesis in the skin, and also in formation of new muscle fibers. Recovery processes in muscles are accompanied by activation and migration of satellite cells of muscle tissue in the zone of action of the radio frequency current.

For the first time immune-chemical analysis of transcriptional factor Pitx2 localization during triton retina regeneration after its removal and also in tissues of a nonoperated eye of an adult triton has been carried out. Protein Pitx2 has been found in the nucleus of the earliest neuroblasts that form the germ of the retina. At a later stage of retina regeneration, Pitx2 was found in the nucleus of differentiating cells of ganglionic layers that correspond to Pitx2 protein localization in the native retina. Protein Ptix2 has also been found in the nucleus of less differentiated cells of the peripheral region of regenerative and native retina. It was demonstrated that protein Pitx2 is expressed not only in retina but also in other tissues of the posterior sector of the eye (pigment epithelium, choroid) using immune-histochemical and Western blot hybridization. It is supposed that transcriptional factor Pitx2 has been involved in the control of subsequent stages of retina regeneration from pigment epithelium cells.

Molecular-genetic mechanisms of regeneration of adult newt (Pleurodeles waltl) retina were studied. For the first time, a comparative analysis of the expression of regulatory genes Pax6, Otx2, and Six3 and Fgf2 genes encoding signal molecules was performed in the native retinal pigment epithelium (RPE) and retina and at successive stages of retina regeneration. Cell differentiation types were determined using genetic markers of cell differentiation in the RPE (RPE65) and the retina (betaII-tubulin and Rho). Activation of the expression of neurospecific genes Pax6 and Six3 and the growth factor gene Fgf2 and suppression of activation of the regulatory gene Otx2 and the RPE65 were observed at the stage of multipotent neuroblast formation in the regenerating retina. The expression of genes Pax6, Six3, and Fgf2 was retained at a later stage of retina regeneration at which the expression of retinal differentiation markers, the genes encoding betaII-tubulin (betaII-tubulin) and rhodopsin (Rho), was also detected. We assume that the above regulatory genes are multifunctional and control not only transdifferentiation of RPE cells (the key stage of retina regeneration) but also differentiation of regenerating retina cells. The results of this study, demonstrating coexpression of Pax6, Six3, Fgf2, betaII-tubulin, and Rho genes, provide indirect evidence for the interaction of regulatory and specific genes during retina regeneration.

The expression of β-III tubulin, a marker protein of early neuronal cells, was studied by molecular genetic and immunochemical techniques. The study was performed with the eyes of human fetuses on weeks 8.5 to 27–28 of intrauterine development. Expression of β-III tubulin was detected immunochemically in the retina and lens fibers of fetuses at stages of 8.5 to 22–23 weeks. By means of PCR, a strong expression of the β-III tubulin gene was revealed in the retina of 9.5-week fetuses. Its level remained high until week 18, became slightly lower by week 24, and decreased to zero by week 27–28. The expression of this gene was also revealed in the lens of 9.5-week fetuses. Its level at stages of 15 to 24 weeks was very low, and no expression could be detected in 27-to 28-week fetuses. The results of PCR analysis were consistent with immunochemical data.

The pattern of the PITX2 gene expression was studied in the cornea, lens, retina, iridocorneal complex (ICC), and eye coats of human fetuses at weeks 9.5–22 of intrauterine development. Using the PCR method, PITX2 expression in all these tissues was revealed already at the earliest stage studied (9.5 weeks), being especially strong in the anterior eye complex (the cornea and lens) and weaker in the retina and sclera. The level of PITX2 expression in all eye tissues slightly decreased by week 15, increased to a high level in the ICC on week 18, and further decreased in all tissues by week 22. Using cDNA derived from the whole eyes of 8-, 9-, 10.5-, and 11-week fetuses, the expression of two PITX2 isoforms specific for eye tissues (A and B) was revealed. By means of in situ hybridization, the PITX2 mRNA was localized in the eye tissues of ectodermal and neuroectodermal origin.

Expression of transcription factors PAX6, PROX1, and PITX2 was evaluated in eye tissues after 9.5 and 22.0 weeks of human fetus development using polymerase chain reaction. Pax6, Prox1 and Pitx2 expression has been revealed in the cornea, lens, retina, and eye membranes (total preparation of the pigment epithelium, choroid, and sclera) after 9.5 weeks of prenatal development with the maximum expression of Pax6 gene in all studied tissues. After 22.0 weeks of development, Pax6 expression increased in the retina and lens but decreased in the cornea. Insignificant levels of Pax6 transcription have been detected in the eye membranes. Prox1 expression was apparent in the cornea, lens, retina, and eye membranes after 9.5 weeks. After 22.0 weeks, Prox1 expression increased in the lens and retina, decreased in the cornea, and was undetectable in the eye membranes. High level of Pitx2 expression has been revealed in all studied eye tissues after 9.5 weeks with the lowest transcription level observed in the retina. After 22.0 weeks, Pitx2 expression notably decreased in the lens and cornea and became undetectable in the retina and eye membranes. The differential pattern of Pax6, Prox1 and Pitx2 expression in developing human eye tissues after 9.5 and 22.0 weeks of development agrees with our histological data.

Expression of transforming growth factor-β2 was detected by PCR in the vitreous body, lens, retina, and ciliary-iris complex of human eye at early stages of fetal development. Immunochemical assay of the corresponding protein in eye tissues revealed a correlation between the localization of transforming growth factor-β2 and the development of intraocular hyaloid vascular network, its regression, formation of the vitreous body, and development of definite retinal vessels.
