Body composition refers to body weight in terms of the absolute and relative amounts of muscle, bone, and fat tissues. Under graduate health science students are required to undertake various activities while pursuing their course so they are required to be fit. As a result, the students should be well aware of their body composition and how aerobic capacity varies on this basis. To study the effect of body composition on aerobic capacity and blood pressure in under graduate health science students. The objectives include estimation of Body composition by calculating body fat %, VO2 max after completion of Queen’s College Step Test, evaluating the blood pressure, resting & post QCST test protocol in under graduate health science students. 280 under graduate health science students able to complete QCST test were recruited. Documentation of demographic data and baseline parameters were done. Skin fold measurements at various sites were taken to estimate the body fat percentage. The QCST was done and Post-test recovery pulse rate and blood pressure were recorded. The post-test parameters were used to calculate the Vo2 max. The result of the study shows that there is an effect of body composition on aerobic capacity and blood pressure in under graduate health science students thereby supporting the aim of the study. It is clear that body composition does affect the aerobic capacity and blood pressure in undergraduate health science students. Furthermore it is to note that the fitness level of health science students are better if their body fat percentage is lower because they are exhibiting higher aerobic capacity and also the blood pressure values have found to be comparatively lower.

In recent years, air ventilation garments (AVG) have been reported effective to improve thermal comfort. In this study, an AVG incorporated with small fans was investigated on torso thermal comfort in moderate environment (Ta=25 °C, RH=50 %). Eight female subjects walked on the treadmill at a speed of 4 km·h−1 for 30 min and then rested for another 30 min. During the whole test protocol, the AVG was worn in three conditions of flow rates to examine which flow rate was the best choice to keep thermal comfort: fans off with no air ventilation (a controlled condition, CON), low flow rate (12 l/s, LOW) and high flow rate (20 l/s, HIGH). Objectives/hypothesis To develop a method for threshold estimation of the laryngeal adductor reflex (LAR) response using Cheung-Bearelly monofilaments. Study design Cross-sectional. Methods Twenty-two healthy adults (12 men, 10 women) were tested for LAR response outcome using 30 mm 5-0 and 4-0 nylon monofilaments. Tactile stimuli were delivered to the aryepiglottic (AE) fold and medial pyriform sinus (MPS). Dichotomous classifier features and performance (area under the curve (AUC)), the LAR response outcome agreement and disagreement matrix, and test-retest reliability were examined. From those data, a test protocol that would minimize patient burden to estimate the LAR triggering threshold was formulated. Results Classifier performance of 5-0 monofilament stimulation of the AE fold (sensitivity = 0.63, specificity = 0.63, PPV = 0.74, NPV = 0.25) and MPS (sensitivity = 0.45, specificity = 0.77, PPV = 0.74, NPV = 0.23,) was slightly below that of 4-0 monofilament stimulation of the AE fold (sensitivity = 0.82, specificity = 0.50, PPV = 0.82, NPV = 0.50) and MPS (sensitivity = 0.84, specificity = 0.64, PPV = 0.90, NPV = 0.56), based on AUC. LAR response outcome agreement for 5-0 and 4-0 stimulations was high (93%) for 5-0 positive response, but low (29%) for 5-0 negative response. Aesthesiometer test-retest reliability for LAR response outcome was excellent (Cronbach's alpha = 0.97). Conclusions Threshold estimation of the LAR response may be operationalized by adopting a decision tree protocol. For negative LAR response to initial 5-0 monofilament stimulation and positive response to subsequent 4-0 monofilament stimulation, the higher threshold is confirmed. Positive LAR response to 5-0 or 4-0 monofilament stimulation is expected in over 90% of asymptomatic adults. Negative LAR response to 4-0 monofilament stimulation identifies patients at risk for laryngopharyngeal hyposensitivity. Level of evidence 3b Laryngoscope, 2021.

Surgical treatment of fragility sacrum fractures with percutaneous sacroiliac (SI) screw fixation is associated with high failure rates. Turn-out is detected in up to 20% of the patients. The aim of this study was to evaluate a new screw-in-screw implant prototype for fragility sacrum fracture fixation. Twenty-seven artificial hemipelvises were assigned to three groups (n = 9) for instrumentation of an SI screw, the new screw-in-screw implant prototype, ora transsacral screw. Before implantation, a vertical osteotomy was set in zone 1 after Denis. All specimens were biomechanically tested to failure in upright position. Validated setup and test protocol were used for complex axial and torsional loading applied through the S1 vertebral body to promote turn-out of the implants. Interfragmentary movements were captured via optical motion tracking. Screw motions were evaluated by means of triggered anteroposterior X-rays. Interfragmentary movements and implant motions were significantly higher for SI screw fixation compared to both transsacral and screw-in-screw fixations. In addition, transsacral screw and screw-in-screw fixations revealed similar construct stability. Moreover, screw-in-screw fixation successfully prevented turn-out of the implant that remained during testing at 0° rotation for all specimens. From biomechanical perspective, fragility sacrum fracture fixation with the new screw-in-screw implant prototype provides higher stability than an SI screw, being able to successfully prevent turn-out. Moreover, it combines the higher stability of transsacral screw fixation with the less risky operational procedure of SI screw fixation and can be considered as their alternative treatment option.

Background and aims Walking is the core physical activity of older persons. The assessment of walking capacity is increasingly important for clinical purposes and clinical research. Differences between assessment tools and protocols for short walks to obtain gait characteristics can be responsible for changes, e.g., in gait speed from 0.1 to 0.2 m/s. The purpose of this study was to generate further knowledge for the harmonization and/or standardization of short walk-test protocols for assessing gait characteristics under supervised conditions. Methods For this cross-sectional study, 150 community-dwelling older adults (mean age 80.5 ± 4.5 years) were recruited. Participants performed eight walks differing in the distance (8-versus 4-m), static versus dynamic trials and comparing different test speed instructions (usual versus maximal) on an electronic walkway. Results A meaningful significant difference in mean usual gait speed was documented comparing the 4-m dynamic and static test protocol (0.12 m/s; p = 0.001). For the same comparison over an 8-m distance (dynamic versus static) and for the comparison between usual gait speed over 4-and 8-m, the differences in gait speed were smaller, but still statistically significant (p = 0.001). Conclusions Gait speed was faster, if the test protocol did not include a static start or stop. The differences were greater for a shorter walking distance. This aspect should be considered for the comparison of study results and is particularly relevant for systematic reviews and meta-analyses.

Objective. To develop a protocol for a normobaric hypoxic test and criteria of resistance assessment — normobaric hypoxia sensitivity in children. Characteristics of children and research methods . 56 healthy children aged from 8 to 12 years underwent a hypoxic test inhaling gas mixture with 10—11% of O 2 through mask to create hypoxemia conditions (SpO 2 = 80%). The authors used a normobaric hypoxic device (certified in the Russian Federation) to produce hypoxic and hyperoxic gas mixtures — ReOxy Cardio (S.A. Aimediq, Luxembourg) with an integrated pulse oximetry sensor (Masimo Rad-5, USA). During hypoxic test they monitored the arterial blood oxygen saturation (SpO 2 ) and heart rate (HR, bpm), and there were automatically recorded the individual minimum values of SpO 2 level (SpO 2 min) and maximum heart rate (HR max). A “hypoxic phase” was terminated in case of decrease in SpO2 to 80% or lower, or breathing with a hypoxic gas mixture during 9 minutes. An additional criterion for termination was an increase in HR by 30% and higher over the initial level. After reaching one of the described criteria, the teenagers began to breathe ambient air and the authors determined the recovery time of SpO 2 to 97%. In addition, the authors calculated the hypoxic index as desaturation/reoxygenation time ratio. Results. The authors developed a protocol for conductance and assessment of hypoxic resistance in children and adolescents. There were determined three types (phenotypes) of response to dosed normobaric hypoxia in adolescents: low, medium and high levels of dosed hypoxic stimulus resistance. Conclusion . The proposed hypoxic test protocol reflects the individual hypoxia tolerance and can be used to individualize hypoxic therapy courses, as well as additional diagnostic tool in assessing the adaptive capabilities and non-specific reactivity in children.

La investigación se centró en evaluar el impacto de un sistema de entrenamiento combinado para desarrollar la fuerza explosiva de los miembros inferiores de los taekwondocas, viene a reforzar el enfoque del desarrollo de la capacidad mediante una lógica metodológica que parte de la creación de las condiciones morfológicas y funcionales para asegurar la potenciación de una hipertrofia sarcoplasmática, como base del desarrollo de la hipertrofia sarcomérica y finalmente favorecer la conversión a potencia y el desarrollo de la fuerza explosiva, en concordancia se transitó por un diagnóstico, elaboración y validación, utilizando convenientemente métodos y técnicas como: entrevista, encuesta, observación, la prueba, la medición, el criterios de experto, la estadística descriptiva e inferencial y el pre-experimento, este último con tres momentos (pre-test/ intervención de 16 semanas/ post-test), participaron 20 taekwondocas universitarios Categoría Sénior y dos entrenadores. Se consideró la evaluación de la fuerza máxima (1RM) de los planos musculares que intervienen directamente en las técnicas de pateo, por estar asociada con la calidad del reclutamiento de la fibras motoras; la evaluación de la fuerza explosiva se realizó mediante el test de salto vertical: Squat Jump (SJ) extraído del protocolo del test de Bosco, para ello fue necesario emplear una plataforma de contacto digital. Los datos se compararon mediante una prueba T de diferencias de medias, cuyos resultados mostraron cambios significativos (p= .000) entre el pre y post tratamiento, verificándose Hi, concluyendo que el sistema de entrenamiento combinado mejoró el desarrollo de la fuerza explosiva de los miembros inferiores de los taekwondocas. Summary: The research focused on assessing the impact of a combined training system to develop the explosive strength of the lower limbs of the taekwondoins. It wants to reinforce the approach of capacity development through a methodological logic that starts from the creation of morphological and functional conditions to ensure the enhancement of a sarcoplasmic hypertrophy, as a basis for the development of sarcomeric hypertrophy, and finally, favor the conversion to power and the development of the explosive force. At the same time, it went through a diagnosis, elaboration and data validation, using propper methods and techniques such as: interview, survey, observation, testing, measurement, expert criteria, descriptive and inferential statistics, and the pre-experiment. The last one with three stages (pre-test / 16-week intervention / post-test) Twenty senior category university taekwondoins and two coaches participated. The evaluation of the maximum force (1RM) of the muscular planes that directly influence in the kicking techniques was considered. As it was associated to the quality of motor fiber recruitment; The evaluation of the explosive force was performed using the vertical jump test: Squat Jump (SJ) extracted from the Bosco test protocol. For this, it was necessary to use a digital contact platform. The data were compared using a T-test, the results of which showed significant changes (p = .000) between the pre and post treatment, verifying Hi. Concluding that the combined training system improved the development of the explosive strength of the limbs lower of the taekwondoinst.

Do haemoglobin mass and red blood cell volume increase in elite cyclists training in a hot environment compared to a control group training at normal temperature? What is the main finding and its importance? Five weeks of heat training increases haemoglobin mass in elite cyclists. There are small to intermediate effect sizes for exercise parameters favouring heat training. ABSTRACT In this study we tested the hypothesis that performing 1 h of regular light exercise in a heat chamber (HEAT; 37.8 ± 0.5°C; 65.4 ± 1.8% humidity) 5 times week-1 for a total of 5 weeks increases haemoglobin mass (Hbmass ) and exercise performance in elite cyclists ( VO2max = 76.2 ± 7.6 ml min-1 kg-1 ). Objective: To characterize the lingual frenulum of full-term newborns using two different protocols and to assess the association of the lingual frenulum with breastfeeding. Methods: This non-probabilistic sample consisted of 449 mother/baby binomials. For the anatomo-functional evaluation of the frenulum, the Neonatal Tongue Screening Test and the Bristol Tongue Assessment Tool were used for the evaluation of the lingual frenulum. Breastfeeding was evaluated using the protocol proposed by UNICEF. Scores were created (good, fair, poor) to evaluate every aspect of the breastfeeding to be observed. The results were analyzed through descriptive and inferential statistics and association tests (Pearson's chi-squared and Fisher's exact test). Results: The study showed that 14 babies had a lingual frenulum alteration, of whom three had difficulties during suction, requiring frenotomy in the first week of life, whereas 11 had no difficulties during breastfeeding. Regarding the breastfeeding evaluation, 410 mother/baby binomials had good, 36 regular, and three had bad scores. There was a statistically significant association between the tongue-tie test protocol and breastfeeding (p = 0.028) and between the Bristol Tongue Assessment Tool protocol and breastfeeding (p = 0.028). Conclusion: Alterations in the lingual frenulum are associated with interferences in the quality of breastfeeding and thus, evaluation of the lingual frenulum in newborns is important. Resumo Objetivo: Caracterizar o frênulo lingual de recém-nascidos a termo, utilizando dois protocolos diferentes e verificar a relação do frênulo lingual com o aleitamento materno. Método: A amostra não probabilística foi constituída por 449 binômios mãe/bebê. Para a avaliação anatomofuncional do frênulo, utilizou-se o protocolo de avaliação do frênulo lingual para bebês “Teste da Linguinha” e o Bristol Tongue Assessment Tool. A mamada foi avaliada com o protocolo proposto pelo UNICEF. Foram criados escores (bom, regular, ruim) para avaliar cada aspecto da mamada a ser observada. Os resultados foram analisados através de estatísticas descritivas e inferenciais e testes de associação (Qui-quadrado de Pearson e Exato de Fisher). Resultados: O estudo mostrou que 14 bebês apresentaram alteração de frênulo lingual, nos quais três com dificuldade durante a sucção, necessitando de frenotomia na primeira semana de vida e 11 sem dificuldades durante a amamentação. Quanto à avaliação da mamada, 410 binômios mãe/bebê apresentaram o escore bom, 36 regular e 3 ruim. Houve associação estatisticamente significativa entre o protocolo “Teste da Linguinha” e amamentação (p = 0,028) e entre o protocolo Bristol Tongue Assessment Tool e amamentação (p = 0,028). Conclusão: Alterações no frênulo lingual estão associadas a interferências na qualidade da amamentação, sendo importante a avaliação do frênulo lingual em recém-nascidos.

Background: Sahrmann five-level core stability test protocol has been used to evaluate the ability of the core muscles to stabilize the spine. However, validation studies on the Sahrmann protocol are limited. Objective: The purpose of this study was to compare the different levels of Sahrmann five-level core stability (levels 1–5) on the muscle activity of rectus abdominis (RA), external oblique (EO), and transverse abdominis/internal oblique (TrA/IO). Methods: Twenty-two asymptomatic male participants aged 21.3[Formula: see text].59 years were recruited. Participants were instructed to perform maximum voluntary contraction (MVC) and five levels of Sahrmann five-level core stability test guided with a pressure biofeedback unit (PBU). The surface electromyography (EMG) data of each muscle during five levels of Sahrmann five-level core stability test were normalized as a percentage of MVC. Results: Results showed significant differences in the normalized EMGs of RA [[Formula: see text](4) = 64.80, [Formula: see text].001], EO [[Formula: see text](4) = 58.11, [Formula: see text].001], and TrA/IO [[Formula: see text](4) = 56.00, [Formula: see text].001] between the five levels of Sahrmann five-level core stability test. Post-hoc analysis revealed Sahrmann levels 5 and 3 have significantly higher abdominal EMG signals than levels 4, 2, and 1 ([Formula: see text].001). Conclusion: In conclusion, the Sahrmann five-level core stability test differs according to the level of Sahrmann tests. Significantly higher abdominal muscle activities were observed during levels 3 and 5. Therefore, the classification exchange in levels 3 and 4 of the Sahrmann five-level core stability test should be reconsidered in the future.

The electromyographic threshold (EMGTh) has been suggested to indicate the onset of accelerated higher-threshold (type-II) MU recruitment. Previous research has demonstrated that boys’ EMGTh occurs at higher relative exercise intensities than men’s in both cycling- and isometric-based testing. Girls‒women EMGTh differences were demonstrated only in cycling, but findings were clouded by low EMGTh-detection rates in women (68%) and particularly in girls (45%) To examine the EMGTh, in girls and women, using the same males-employed isometric-based test protocol, and compare the females’ findings with those previously obtained in the males. Seventeen girls and 17 women had their EMGTh determined as well as their one-repetition-maximum isometric knee-extension strength (1RM). Vastus-lateralis sEMG root mean square was recorded and the EMGTh was defined as the exercise intensity (%1RM) at the bi-segmental point of the least sum of squares. EMGTh was detected in 88.2% of girls and 94.1% of women and occurred at higher relative intensities in the girls than in women (56.0 ± 11.1 vs. 47.7 ± 8.0% 1RM). The girls’ 1RM (normalized to lean body mass) was only 69.1% that of the women. Girls’ EMGTh values are higher compared with women’s, possibly reflecting lower ability to activate higher-threshold (type-II) motor units. The females’ EMGTh and detection-rate values were similar to the corresponding values previously observed in males. The females’ age-related difference in the recruitment of higher-threshold motor units, as reflected by the EMGTh, appears to be on par with the males.

Inbred and genetically modified mice are frequently used to investigate the molecular mechanisms responsible for the beneficial adaptations to exercise training. However, published paradigms for exercise training in mice are variable, making comparisons across studies for training efficacy difficult. The purpose of this systematic review and meta-analysis was to characterize the diversity across published treadmill-based endurance exercise training protocols for mice and to identify training protocol parameters that moderate the adaptations to endurance exercise training in mice. Published studies were retrieved from PubMed and EMBASE and reviewed for the following inclusion criteria: inbred mice; inclusion of a sedentary group; and exercise training using a motorized treadmill. Fifty-eight articles met those inclusion criteria and also included a “classical” marker of training efficacy. Outcome measures included changes in exercise performance, V˙O2max, skeletal muscle oxidative enzyme activity, blood lactate levels, or exercise-induced cardiac hypertrophy. The majority of studies were conducted using male mice. Approximately 48% of studies included all information regarding exercise training protocol parameters. Meta-analysis was performed using 105 distinct training groups (i.e., EX-SED pairs). Exercise training had a significant effect on training outcomes, but with high heterogeneity (Hedges’ g=1.70, 95% CI=1.47–1.94, Tau2=1.14, I2=80.4%, prediction interval=−0.43–3.84). Heterogeneity was partially explained by subgroup differences in treadmill incline, training duration, exercise performance test type, and outcome variable. Subsequent analyses were performed on subsets of studies based on training outcome, exercise performance, or biochemical markers. Exercise training significantly improved performance outcomes (Hedges’ g=1.85, 95% CI=1.55–2.15). Subgroup differences were observed for treadmill incline, training duration, and exercise performance test protocol on improvements in performance. Biochemical markers also changed significantly with training (Hedges’ g=1.62, 95% CI=1.14–2.11). Subgroup differences were observed for strain, sex, exercise session time, and training duration. These results demonstrate there is a high degree of heterogeneity across exercise training studies in mice. Training duration had the most significant impact on training outcome. However, the magnitude of the effect of exercise training varies based on the marker used to assess training efficacy.

Most of the Indian diets are typically based on cereals and lack micronutrient-rich vegetables, fruits, and flesh foods. The incorporation of zinc biofortified rice in daily diets can help overcome zinc deficiency. The present study aimed to analyze the consumers’ acceptance of zinc biofortified rice as the willingness of consumers to accept it is important for the success of biofortification strategy to combat micronutrient malnutrition. The consumers’ acceptance of zinc biofortified rice in terms of its hand feel mouth feel texture, taste, and overall acceptability was determined using the Five-point Hedonic scale. Hand-pounded samples of zinc biofortified rice and control were provided to sixty farm women to evaluate by the Home Use Test protocol. The index of acceptability (IA) was worked out for the acceptance of zinc biofortified rice and was found to be greater than 70% for all the parameters. The consumers’ socio-economic characteristics did not show a significant relationship with the acceptability of zinc biofortified rice except for hand feel texture. Based on the hedonic categorization suggested by Belmes (2019), the overall acceptability of both the zinc biofortified rice and control are in the acceptable category of hedonic rating. Therefore it can be concluded that the zinc biofortified rice variety DRR Dhan 45 is equally acceptable to the consumers as the control rice. The consumers’ socio-economic characteristics did not show a significant relationship with the acceptability of zinc biofortified rice. The zinc biofortified rice can be promoted for use in daily diets to help meet the zinc needs of the family and included in the nutrition intervention programs of the country for overcoming micronutrient malnutrition.

Summary The coronavirus disease 2019 (COVID‐19) is a global pandemic caused by a novel coronavirus called severe acute respiratory syndrome coronavirus 2 (SARS‐CoV‐2). To date, the virus has been detected in 219 countries of the world. Therefore, managing the disease becomes the priority, in which detecting the presence of the virus is a crucial step. Presently, real‐time RT polymerase chain reaction (RT‐qPCR) is considered a gold standard nucleic acid amplification test (NAAT). The test protocol of RT‐qPCR is complicated, places high demands on equipment, testing reagents, research personnel skills and is expensive. Therefore, simpler point‐of‐care (POC) tests are needed to accelerate clinical decision‐making and take some of the workload from centralized test laboratories. Various isothermal amplification‐based assays have been developed for the sensitive detection of different microorganisms, and recently some of them have been applied for detection of SARS‐CoV‐2. These do not require any programable thermocycler, can produce the results in a single temperature, and therefore, are considered simple. Unlike RT‐qPCR, these methods are highly sensitive, specific, less time‐consuming, simple and affordable, and can be used as POC diagnostic kit for COVID‐19. In this review, we have discussed the potential of isothermal amplification‐based assays as an alternative to RT‐qPCR for the detection of SARS‐CoV‐2.

Background Given the challenges and costs associated with implementing HIV-1 incidence assay testing, there is great interest in evaluating the use of commercial HIV diagnostic tests for determining recent HIV infection. A diagnostic test with the capability of providing reliable data for the determination of recent HIV infection without substantial modifications to the test protocol would have a significant impact on HIV surveillance. The Abbott ARCHITECT HIV Ag/Ab Combo Assay is an antigen/antibody immunoassay, which meets the criteria as the first screening test in the recommended HIV laboratory diagnostic algorithm for the United States. Methods In this study, we evaluated the performance characteristics of the ARCHITECT HIV Ag/Ab Combo signal-to-cutoff ratio (S/Co) for determining recent infection, including estimation of the mean duration of recent infection (MDRI) and false recent rate (FRR), and selection of recency cutoffs. Results The MDRI estimates for the S/Co recency cutoff of 400 is within the 4 to 12 months range recommended for HIV incidence assays, and the FRR rate for this cutoff was 1.5%. Additionally, ARCHITECT Combo S/Co values were compared relative to diagnostic test results from two prior prospective HIV-1 diagnostic studies in order to validate the use of the S/Co for both diagnostic and recency determination. Conclusion Dual-use of the ARCHITECT Combo assay data for diagnostic and incidence purposes would reduce the need for separate HIV incidence testing and allow for monitoring of recent infection for incidence estimation and other public health applications.

Background: The 6-min walk test (6MWT) is a validated tool, of submaximal intensity, used to objectively measure functional exercise capacity. In 2002, the American Thoracic Society (ATS) developed guidelines on standardising the implementation of the 6MWT. Despite the relative ease of conducting the 6MWT as per these guidelines, adaptations are implemented. Objectives: Identify (1) what 6MWT adaptations to the ATS guidelines have been described in low-resource settings (LRS), (2) the purpose of the adapted 6MWT and (3) the reported argumentation for making these adaptations in relation to the specific context. Methods: Five databases were searched from inception until February 2021. Studies that adapted and conducted the 6MWT in LRS were included. Data concerning the study source, participants, 6MWT: purpose, variations, outcome and rationale were extracted. Results: A total of 24 studies were included. The majority of studies ( n = 18; 75%) were conducted in lower-middle income countries. The most common adaptation implemented was variation to course length. Eight studies provided a rationale for adapting the 6MWT. Space constraint was the most common reason for adaptation. Conclusion: The most common reason (space constraints) for adapting the 6MWT in LRS was addressed through adaptations in course length and/or configuration. The results of this review suggest that the value of the ATS-guided 6MWT in LRS may need to be re-evaluated. Clinical implications: Using adapted forms of the 6MWT may lead to an underestimation of a patient’s abilities, misinformed discharge and developing inappropriate exercise programmes. Additionally, diverting from ATS guidelines may affect the continuity of care.

Background and objectives Extubation readiness testing (ERT) in the Neonatal Intensive Care Unit (NICU) is highly variable and lacking standardized criteria. To address this gap, an evidence-based, inter-professionally developed ERT protocol was implemented to assess effectiveness on extubation failure within 72 h and on duration of intubation (DOI). Methods A longitudinal retrospective chart review in a level III, fully outborn NICU, of intubated infants admitted 1-year prior (Group 1), and 1 year after implementation (Group 2). Patients were extubated if they passed a 2-stage ERT protocol (3 min continuous positive airway pressure (CPAP) followed by 7 min CPAP + pressure support). Descriptive, comparative statistics, and univariate and multiple logistic regression were completed on all patients and a ≤32 6/7 weeks subgroup (intubated at day-of-life 1); p < 0.05 is considered significant. Results All patients (n = 589 (n = 294 Group 1, n = 295 Group 2)) were included (preterm, intubated day of life one subgroup: n = 42 Group 1, n = 38 Group 2). For all patients, extubation failure decreased significantly from 9.9% to 4.1% (p = 0.006); Group 1 patients were 2.42 times more likely to experience extubation failure compared with Group 2. Extubation failure in the preterm subgroup decreased from 21.7% to 2.6% (p = 0.01); Group 1 patients were 10.71 times more likely to experience extubation failure. Median DOI was similar in both groups for all patients and in the preterm subgroup. Conclusions A unique two-stage ERT protocol was effective at reducing extubation failure rate, without increasing DOI, largely in preterm infants. The evidence-based, interprofessionally developed ERT protocol and its integration into the NICU culture largely contributed to its success.

Background/Aims: The available episodic memory tests are not specifically constructed to examine older subjects. Their use in outpatient memory clinics may result in aborted test administration. We used a strict adherence to the test protocol in cognitively healthy, amnestic mild cognitive impairment (aMCI), and Alzheimer’s disease dementia subjects to assess the possibility of preventing this. Methods: This is a cross-sectional study in memory outpatient subjects with a mean age of 74.5 years. Primary study outcomes were: number of missing values and test results in the Visual Association Test (VAT) and the 15 Word Test (15WT). Results: A strict adherence to the test protocol resulted in a 10-fold decrease in the number of missing values in the VAT. For the 15WT this could not be realized mostly because the test was deemed too demanding for 1 in 6 patients. Conclusions: This study is one of the few examining the applicability of well-known episodic memory tests in older subjects. A strict adherence to the test protocol reduced the number of missing values. Floor effects were stronger for the 15WT than for the VAT. Results favor the use of the VAT in senior subjects and show the unsuitability of the 15WT in this group.

Background and aims Conditioned pain modulation (CPM) is of considerable interest within pain research. Often CPM testing is conducted in experimental settings using complicated instrumentation, thus challenging the implementation in clinical settings. Being able to assess CPM in a fast and reliable way in clinical settings could lead to a new diagnostic tool allowing improved profiling of pain patients. Methods A test-retest reliability study and a methodological development study were conducted based on different populations. The reliability study included 22 healthy subjects, mean age 23.6 years (SD: 2.4) and the methodological study included 29 healthy subjects, mean age 21.5 years (SD: 1.6). As painful phasic test stimulus, a 6–10 kg handheld, spring-based pressure algometer was applied perpendicularly to the muscle belly of the tibialis anterior muscle for 10 s and as painful tonic conditioning stimulus, 1–2 standard clamps, inducing a force of 1.3 kg, were applied extra-segmentally at the ipsilateral earlobe for 60–120 s. Four different test protocols were evaluated, of which one protocol was investigated for reliability. Test protocol 1 used a 6 kg pressure algometer as painful phasic test stimulus and a single clamp applied for 60 s as painful tonic conditioning stimulus. Test protocol 2 used a 10 kg pressure algometer as painful phasic test stimulus, and two clamps applied for 60 s as painful tonic conditioning stimulus. Test protocol 3 used a 10 kg pressure algometer as painful phasic test stimulus and a single clamp applied for 120 s as painful tonic conditioning stimulus. Test protocol 4 used a 6 kg pressure algometer as painful phasic test stimulus and a single clamp applied for 120 s as painful tonic conditioning stimulus. Results None of the stimuli caused any adverse events, e.g. bruises. In the reliability study (test protocol (1), non-significant CPM effects of 0.3 (SD: 1.6) and 0.2 (SD: 1.0) were observed in session 1 and 2, respectively. The intra-class correlations were 0.67 and 0.72 (p = Conclusions The bedside test developed for investigating CPM was feasible and easy to use in healthy volunteers. No significant CPM effects were measured and a large variation in CPM effect ranging from −0.14 to 0.32 was observed. Intra-class correlation (ICC) values for the pressure algometer were interpreted as “good relative reliability” (test protocol 1), and LoA revealed a somewhat low absolute reliability. Implications The pressure algometer provided reproducible measurements and was useful for inducing phasic test stimuli. Since no significant CPM effects were detected, no recommendations for the bedside test can yet be made. Further examinations will have to establish if the “one size fits all” application of both test and conditioning stimuli is useful. Future bedside studies involving patient populations are warranted to determine the usefulness of the method.

To evaluate the ability to recognise different types of developmental defects of enamel (DDE) by a group of general dental staff, trained prior to the comprehensive prevalence study (the BITA study), and to compare their skills to that of an untrained group. To evaluate the validity and reliability of the DDE registrations and MIH diagnoses, 3 years after the BITA study ended. To evaluate the reliability of an MIH diagnose, 3 years after the study ended. The dental staff from five clinics (n = 43) was given instructions in identifying different types of DDE, according to the mDDE index (FDI in Int Dent J 42:411–426 1992). Motor skills are considered important for children's physical, social, and psychological development. Pre-school age seems to be decisive for the development of motor skills. Our study's aim was to investigate the effect of 12 weeks game education on the motor development of pre-school children aged 4 to 6 years. In the research, experimental method pre-test / post-test and control group design was used. In this study, a total of 30 boys and 30 girls were enrolled. The research consisted of a total of 60 children who were studying in pre-school and were selected via random sampling model. In order to collect data, 12 weeks game education was conducted on participants. Motor Performance test protocol, developed by Morris, Atwater Williams and Willmore in 1980, was used on both experiment and control group children, in order to measure their motor performance before and after 12-weeks game education. Average and standard deviation values were calculated for the data collected. Children's throwing, catching, long jump, vertical jump, running, and stopping skills were measured. Mean and standard deviations of the data obtained in the study were taken. A statistically significant difference was found between the experimental and control groups in terms of post-test. A statistically significant difference was found in the experimental group pre-test and post-test comparisons. In these comparisons, the post-test values were found to be higher than the pre-test values. According to the results of the research, it is necessary to apply more long-term applications to increase the positive effect of 12-Weeks basic game education at the levels of motor development in children aged 4 to 6 years old.

Background: Various field tests can be used to evaluate children's motor skills. Because of the complexity of their motor skills, which at their age are still in a very intensive stage of development, it is inadequate to use the same test protocol as in the adult population, as children receive and process information in a different way. Some evidence showed that familiarization with test protocols is important in children's motor skills evaluation process. Objective: The purpose of this study was to examine the influence of familiarization on motor test results for the assessment of motor skills in preschool children population. A controlled research study design was conducted. Methods: The sample included 40 children, 20 boys and 20 girls (age 5.89 ± 0.42 years). Subjects underwent two testing sessions with two weeks between the trials, performed 6 motor skills: leg power, flexibility, abdominal muscles endurance, shoulder muscles endurance, agility, and running speed. Before the second testing session, subjects underwent a period of familiarization with motor tasks in 3 sessions, with 3 trials every 3 days. Results: Significantly better results in the second testing session were observed in countermovement jump (with both fixed and free arms), curl-ups, speed and agility test. The present study indicates that children significantly improved the results in the motor test of explosive strength, muscle endurance, speed and agility, being influenced by familiarization. Conclusions: The results of this study indicate that familiarization with the test procedures prior to testing affect the results of the motor test, and represent an important part of the evaluation of motor skills of preschool children.

Medicinal plants have bioactive compounds which are used for curing of various human diseases and also play an important role in healing. In Indian Ayurvedic system, Lagerstroemia Indica (Lythraceae) are well-known plants used for major and minor ailments. The aim of the present study is to examine leaf of Lagerstroemia Indica for phytochemical profile, antioxidant potential, antimicrobial including anti acne activity. Qualitative analysis of various phytochemical constituents and quantitative analysis of total phenolics and flavonoids were determined by the well-known test protocol available in the literature. Quantitative analysis of phenolic and flavonoids was carried out by FolinsCiocalteau reagent method and aluminium chloride method respectively. Phytochemical analysis revealed the presence of phenols, flavonoids, tannins, saponins, alkaloids, fixed oil and fats. The present study concluded that the crude extract of Lagerstroemia Indica is a rich source of secondary phytoconstituents which impart significant antioxidant potential. The extract of Lagerstroemia Indica possess antimicrobial effectiveness againt P. acne and other microbes, In Vitro anti acne activity. Introduction The Oral Minimal Model (OMM), a differential-equations based mathematical model of glucose-insulin dynamics, utilizes data from a frequently sampled oral glucose tolerance test (OGTT) to quantify insulin sensitivity ( S I ). OMM-based estimates of S I can detect differences in insulin resistance (IR) across population groups and quantify effects of clinical or behavioral interventions. These estimates of S I have been validated in healthy adults using data from OGTTs with durations from 2 to 7 h. However, data demonstrating how protocol duration affects S I estimates in highly IR populations such as adolescents with obesity are limited. Methods A 6-h frequently sampled OGTT was performed in adolescent females with obesity. Two, 3-, and 4- hour implementations of OMM assuming an exponentially-decaying rate of glucose appearance beyond measured glucose concentrations were compared to the 6-h implementation. A 4- hour OMM implementation with truncated data (4h Tr) was also considered. Results Data from 68 participants were included (age 15.8 ± 1.2 years, BMI 35.4 ± 5.6 kg/m2). Although S I values were highly correlated for all implementations, they varied with protocol duration (2h: 2.86 ± 3.31, 3h: 2.55 ± 2.62, 4h: 2.81 ± 2.59, 4h tr: 3.13 ± 3.14, 6h: 3.06 ± 2.85 x 10-4 dl/kg/min per U/ml). S I estimates based on 2 or 3 h of data underestimated S I values, whereas 4-h S I estimates more closely approximated 6-h S I values. Discussion These results suggest that OGTT protocol duration should be considered when implementing OMM to estimate S I in adolescents with obesity and other IR populations.

Novel methods for advancing reliability testing of photovoltaic (PV) modules and materials have recently been developed. Combined-accelerated stress testing (C-AST) is one such method which has demonstrated reliable reproduction of some field-failures which were not reproducible by standard certification tests. To increase confidence and assist in the development of C-AST, and other new testing protocols, it is important to validate that the failure modes observed and mechanisms induced are representative of those observed in the field, and not the product of unrealistic stress conditions. Here we outline a method using appropriate materials characterization and modelling to validate the failure mechanisms induced in C-AST such that we can increase confidence in the test protocol. The method is demonstrated by applying it to a known cracking failure of a specific polyamide (PA)-based backsheet material. We found that the failure of the PA-based backsheet was a result of a combination of stress factors. Photo-oxidation from ultra-violet (UV) radiation exposure caused a reduction in fracture toughness, which ultimately lead to the cracking failure. We show that the chemical and structural changes observed in the backsheet following C-AST aging were also observed in field-aged samples. These results increase confidence that the conditions applied in C-AST are representative of the field and demonstrates our approach to validating the failure mechanisms induced.

BACKGROUND AND OBJECTIVES Reduced laser-evoked potential (LEP) habituation indicates abnormal central pain processing. But the paradigm (four stimulation blocks a 25 stimuli) is time consuming and potentially omits important information on the exact habituation time course. This study examined whether a high temporal resolution (HTR) analysis (dividing the four stimulation blocks into 12 analysis blocks) can answer the following questions: (a) After how many stimuli does LEP habituation occur? (b) Is there a difference in LEP habituation in younger versus older subjects? (c) Is HTR applicable on radiculopathy patients? METHODS EEG data of 129 subjects were included. Thirty-four young healthy and 28 advanced-aged healthy subjects were tested with LEPs on the hand dorsum. Thirty-seven radiculopathy patients and 30 controls were tested with LEPs on the L3 dermatome. The EEG data of the hand dorsa have been analysed conventionally and with HTR analysis. The applicability of HTR has been tested on radiculopathy patients and respective controls. RESULTS HTR was well feasible in young healthy subjects and revealed a strong habituation effect during the first 25 stimuli (i.e. within the first 5 min). After approximately 48 stimuli, no further significant habituation was detectable. LEP amplitudes were higher in young subjects. HTR was unsuitable for elderly subjects and middle-aged radiculopathy patients. CONCLUSIONS In young healthy subjects, HTR allows a shortening of the test protocol while providing a detailed information on the time course of LEP habituation. A shorter protocol might be useful for the applicability of the LEP paradigm for clinical and experimental settings as well as pharmacological studies. SIGNIFICANCE The usage of high temporal resolution (HTR) analysis in young healthy subjects enables a short test protocol and provides the exact time course of laser-evoked potential habituation. This can be useful for the examination of neurological conditions affecting younger patients and for pharmacological studies. HTR was inapplicable in advanced-aged subjects and patients with radiculopathy.

Purpose: Rating of perceived exertion (RPE) is a widely used tool to assess subjective perception of effort during exercise. The authors investigated between-subject variation and effect of exercise mode and sex on Borg RPE (6–20) in relation to heart rate (HR), oxygen uptake (VO2), and capillary blood lactate concentrations. Methods: A total of 160 elite endurance athletes performed a submaximal and maximal test protocol either during cycling (n = 84, 37 women) or running (n = 76, 32 women). The submaximal test consisted of 4 to 7 progressive 5-minute steps within ∼50% to 85% of maximal VO2. For each step, steady-state HR, VO2, and capillary blood lactate concentrations were assessed and RPE reported. An incremental protocol to exhaustion was used to determine maximal VO2 and peak HR to provide relative (%) HR and VO2 values at submaximal work rates. Results: A strong relationship was found between RPE and %HR, %VO2, and capillary blood lactate concentrations (r = .80–.82, all Ps 2 decreased linearly with increased RPE, from ∼10% to 15% at RPE 8 to ∼5% at RPE 17. Compared with cycling, running induced a systematically higher %HR and %VO2 (∼2% and 5%, respectively, P 2 than men (P Conclusions: Among elite endurance athletes, exercise mode influenced RPE at a given %HR and %VO2, with greater differences at lower exercise intensities. Athletes should manage different tools to evaluate training based on intensity and duration of workouts.

Until now the reproducibility of the short latency stretch reflex of the internal rotator muscles of the glenohumeral joint has not been identified. Twenty-three healthy male participants performed three sets of external shoulder rotation stretches with various pre-activation levels on two different dates of measurement to assess test-retest reliability. All stretches were applied with a dynamometer acceleration of 104°/s2 and a velocity of 150°/s. Electromyographical response was measured via surface EMG. Reflex latencies showed a pre-activation effect (ƞ2 = 0,355). ICC ranged from 0,735 to 0,909 indicating an overall “good” relative reliability. SRD 95% lay between ±7,0 to ±12,3 ms.. The reflex gain showed overall poor test-retest reproducibility. The chosen methodological approach presented a suitable test protocol for shoulder muscles stretch reflex latency evaluation. A proof-of-concept study to validate the presented methodical approach in shoulder involvement including subjects with clinically relevant conditions is recommended .

The incidence of presyncopal events is high soon after a long-duration spaceflight;>60% of returning astronauts could not complete a 10-min 80° head-up tilt test on landing day (R+0) after ~6 months of spaceflight. The objective of this study was to demonstrate the ability of a lower body gradient compression garment (GCG) to protect against an excessive increase in heart rate and a decrease in blood pressure during standing after long-duration spaceflight. Methods: Eleven astronauts (9 M, 2 F) volunteered to participate. The stand test protocol consisted of 2 min of prone rest followed by 3.5 min of standing. Subjects completed one familiarization session, two preflight data collection sessions in standard clothing, and three tests on landing day while wearing GCG. Postflight tests were conducted 1–4 h (R+0A), ~12 h (R+0B), and ~28 h after landing (R+0C). Results: All astronauts completed the stand test preflight. Three astronauts were unable to attempt the stand test at R+0A, and one of these was unable to start the test at R+0B. One astronaut was unable to complete 3.5 min of standing at R+0B (test ended at 3.3 min). Review of the individual’s blood pressure data revealed no hypotension but the astronaut reported significant motion sickness. Of the astronauts who participated in testing on landing day, the heart rate and mean arterial pressure responses to standing (stand-prone) were not different than preflight at any of the postflight sessions. Conclusion: Wearing the GCG after spaceflight prevented the tachycardia that normally occurs while standing after spaceflight without compression garments and protected against a decrease in blood pressure during a short stand test.

This study aimed to explore a valid test protocol for measuring VO2max in healthy untrained male Wistar rats of different ages and quantifying the exercise intensity (%VO2max) of running under different treadmill grades and speeds. The test protocols and %VO2max will provide a reference for the design of exercise intensity. We tested male Wistar rats aged 4 weeks, 10 weeks, 10 months and 16 months old with three test protocols (Procedure 1 [P1], 2 [P2] and 3 [P3]) for each age group to quantify VO2max. We analysed VO2max, respiratory exchange ratio and test duration to determine an optimal test protocol of VO2max for different age groups. We used the optimal test protocol to explore the changes in age-related VO2max. Finally, %VO2max of running under different treadmill speeds and grades was quantified. VO2max of Wistar rats decreased significantly after the age of 4 weeks (p 2max can be induced by personalised protocols for different ages. In 4-week-old Wistar rats, the highest VO2max values were attained by P1 (104.4 ± 6.9 mL · kg−1 · min−1, p = 0.032). The highest VO2max value (84.7 ± 3.7 mL · kg−1 · min−1, p = 0.037) of 8-week-old Wistar rats was attained in P2. In 10-month-old Wistar rats, the highest VO2max value was obtained in P3 (63.3 ± 1.7 mL · kg−1 · min−1). This work could be used as a reference for assessing aerobic capacity in studies on exercise intervention with untrained male Wistar rats. However, the %VO2max measurements at various treadmill speeds and grades only apply to untrained male Wistar rats.

Introduction This study aims to develop a standard number of repetition at angular velocities of 60°/sec and 180°/sec and define the difference of number of repetitions between genders. Method One hundred ninety-six athletes (judo, volleyball, Greco-Roman wrestling, runners, and gymnastics) were involved. Concentric-concentric isokinetic muscle strength of the knee flexor-extensor muscles of the athletes were performed with an ISOMED 2000 isokinetic dynamometer. The evaluation test protocol consisted of 10 repetition of knee flexion-extension at 60°/sec and 30 repetitions at 180°/sec. Results Athletes performed with maximal knee flexion strength at their second and third trial at 60°/sec and third-forth trial at 180°/sec. It was found that 2 repetitions and 2–3 repetitions were needed for knee extension peak torque at 60°/sec and 180°/sec respectively. The difference between the genders was significant at 60°/sec for knee flexion and extension on the dominant side; at 180°/sec for knee extension on the dominant and non-dominant side. It was determined that 3 repetitions for 60°/sec, 4 repetitions for 180°/sec were enough to determine peak torque values. Discussion Optimal number of repetitions is crucial so that athletes could have the opportunity to reach maximal strength without unnecessary workload or waste of time. Conclusions Protocols involving 3 repetitions for 60°/sec, 4 repetitions for 180°/sec were enough to determine peak torque values. Gender of athletes should be considered when establishing isokinetic knee evaluation protocols.

The aim of this study was to investigate if the order of submaximal lactate threshold and maximal oxygen uptake testing would influence test outcomes. Twelve well-trained male cross-country skiers (mean age 19.6 years) performed two test sessions within a week in a within-subjects repeated measures with cross-over design study. A maximal oxygen uptake test (VO2max) followed by a lactate threshold (LT) test and vice versa, were performed. The test data included VO2, blood lactate (La-b), heart rate (HR), performance speed, Borg scale (RPE) at all stages and lactate accumulation throughout the whole test protocol including the breaks. No significant effect of testing order was found for: VO2max (74.23 vs. 73.91 mL∙min&minus, 1∙kg&minus, 1), maximal HR (190.7 vs. 189.9 bpm) and speed at LT during uphill running. Three out of four common definitions of LT resulted in the same La-b at the last two steps, 11 and 12 km/h respectively, in the two protocols. It is worth noting that VO2, HR and La-b were higher in the first two stages of the LT test when VO2max was tested first in the protocol. Well-trained cross-country skiers conclusively attained a similar VO2max and LT in both protocols, and the two tests did not seem to influence each other in terms of the degree of exhaustion that occurs in a single VO2max or an incremental LT test. However, when using a curvilinear function to define the LT, it is important to know that the VO2max test can influence levels of VO2, HR and La-b at the first two stages of the LT test.

Far side has been identified in the literature as a potential cause of numerous injuries and fatalities. Euro NCAP developed a far side test protocol to be performed to assess adult protection. A monitoring phase was undertaken between January 2018 and December 2019, and the far side assessment will become part of the rating for all vehicles launched in 2020 onward. A test buck was developed and 6 paired WorldSID / Post Mortem Human Subjects (PMHS) were subjected to the test protocol proposed by Euro NCAP to contribute to the development of limits. The buck consisted of a rigid seat and a rigid central console covered with 50 mm of Ethafoam TM 180 with a density of 16 kg/m3. The buck was mounted on the sled with an angle of 75° between the X axis of the vehicle and the X axis of the sled. The peak head excursion was compared between PMHS and the WorldSID dummy. It was found reasonably similar. However, the dummy repeatability was found to be poor. Out of 6 tests conducted on 6 PMHS, 2 specimens sustained AIS3 and, 3 specimens AIS2 cervical spine injuries, 3 specimens sustained AIS3, 1 AIS2 and 1 AIS1 thoracic injuries, and 2 specimens sustained AIS2 abdominal injuries. The peak values recorded on the dummy according to the Euro NCAP protocol were compared with the injury assessments of the PMHS tests. In the configuration used, which includes a central console, the hard thorax injury prediction was found to be excellent. For the neck injury prediction, the data were merged with similar results available in the literature and an Injury Risk Curve was proposed as a derivative from the curve published by Mertz et al. (2003) for neck extension.

Background High-intensity interval training (HIIT) is an important training component to improve aerobic and anaerobic exercise capacity. Higher HIIT workloads in general may generate additional effects on the improvement of exercise capacity, while missing adherence to more strenuous training regimes may affect training success. This study investigated if higher training workload generated by progressive HIIT (proHIIT) is superior to HIIT when used in an uncontrolled setting. Methods Thirty-four moderately trained females and males performed a 4-week training intervention with three exercise sessions per week. Participants were randomized into two HIIT groups using the individual lactate threshold at baseline: Group 1 (N.=17), HIIT, four runs at maximal speed (all-out) with 30 s active recovery (total: 48 runs), Group 2 (N.=17), proHIIT, 4 runs at maximal speed (all-out) with 30-second active recovery with one extra repetition every week (up to seven runs, for a total of 66 runs). An incremental field test protocol with standard blood lactate (LA) diagnostic and heart rate monitoring was used to access changes in exercise capacity. Results Overall, power output (running speed) at LA threshold (baseline LA+1.5 mmol/L) increased by +3.6% (P=0.004, effect size [ES] 0.38) after 4 weeks of HIIT. However, no significant between-group differences pre- vs post-intervention were detected. Conclusions Our data suggest that proHIIT does not provide additional improvement of running speed at individual lactate threshold over HIIT in an uncontrolled setting.

The fathead minnow fish embryo toxicity (FET) test has been proposed as a more humane alternative to current toxicity testing methods as younger organisms are thought to experience less distress during toxicant exposure. However, the FET test protocol does not include endpoints that allow for the prediction of sublethal adverse outcomes, limiting its utility relative to other test types. Researchers have proposed the development of sublethal endpoints for the FET test to increase its utility. The present study 1) developed methods for previously unmeasured sublethal metrics in fathead minnows (i.e., spontaneous contraction frequency and heart rate) and 2) investigated the responsiveness of several sublethal endpoints related to growth (wet wt, length, and growth-related gene expression), neurodevelopment (spontaneous contraction frequency, eye size, and neurodevelopmental gene expression), and cardiovascular function and development (pericardial area, heart rate, and cardiovascular system-related gene expression) as additional FET test metrics using the model toxicant 3,4-dichloroaniline. Of the growth, neurological, and cardiovascular endpoints measured, length, eye size, and pericardial area were found to be more responsive than the other endpoints evaluated. Future studies linking alterations in these endpoints to longer-term adverse impacts are needed to fully evaluate the predictive power of these metrics in chemical and whole-effluent toxicity testing. BACKGROUND A network comprising 11 laboratories aimed to consolidate PNH testing by developing and validating an assay guided by previous guidelines and studies. Network analyses of >20 native samples yielded key findings that were used to create and reshape the final protocol. METHODS Twenty-seven native samples were distributed to all participating laboratories for blind testing, local analysis, and subsequent re-analysis by a central laboratory. Inter-laboratory clone size precision (coefficient of variation [CV]) was monitored for each sample, and the findings used to refine the test protocol. Linearity and precision tests were performed, assay limits of blank and detection were calculated, and limits of quantification were determined. RESULTS When using the final protocol, enumeration of cells with a PNH phenotype by all participating laboratories was comparable, with no clinically significant discrepancies or false-positive or false-negative results reported. Of note, the biological characteristics of the sample affected precision. For example, CVs were higher when PNH and normal cells showed contiguous expression of GPI-linked antigens. A red cell gating strategy that eliminated non-specific type II PNH phenotypic events was devised, enabling reliable reporting of events in the type II red cell gate. The final protocol provided an assay with a Limit of Quantification of 0.01% for neutrophils and red cells, and 0.1% for monocytes. CONCLUSIONS We describe a robust network-validated PNH assay that may aid other laboratories to set up and validate high resolution PNH analysis. © 2018 International Clinical Cytometry Society.

Inconsistent findings between laboratories are hampering scientific progress and are of increasing public concern. Differences in laboratory environment is a known factor contributing to poor reproducibility of findings between research sites, and well-controlled multisite efforts are an important next step to identify the relevant factors needed to reduce variation in study outcome between laboratories. Through harmonization of apparatus, test protocol, and aligned and non-aligned environmental variables, the present study shows that behavioral pharmacological responses in Shank2 knockout (KO) rats, a model of synaptic dysfunction relevant to autism spectrum disorders, were highly replicable across three research centers. All three sites reliably observed a hyperactive and repetitive behavioral phenotype in KO rats compared to their wild-type littermates as well as a dose-dependent phenotype attenuation following acute injections of a selective mGluR1 antagonist. These results show that reproducibility in preclinical studies can be obtained and emphasizes the need for high quality and rigorous methodologies in scientific research. Considering the observed external validity, the present study also suggests mGluR1 as potential target for the treatment of autism spectrum disorders.

ZusammenfassungEine Ruptur des vorderen Kreuzbandes (VKB) stellt eine schwerwiegende Verletzung im Spielsport dar, die sowohl für den Athleten als auch für den Verein zeit- und kostenintensiv ist. Aufgrund der vielseitigen und anspruchsvollen Belastung der unteren Extremitäten sind vor allem Spielsportler häufig von der schweren Verletzung mit langem Rehabilitationszeitraum betroffen. Bezogen auf den Wiedereinstieg in den Sport variieren die Zeiträume stark. Zur Bestimmung des Zeitpunkts der Rückkehr in den Sport werden bereits verschiedene Ansätze diskutiert, diesen mittels funktioneller Testverfahren zu quantifizieren. Ein Konsens hinsichtlich relevanter Parameter, der verfügbaren Testverfahren und der Interpretation der Testwerte existiert jedoch nicht. Auf der Basis des Anforderungsprofils der Spielsportart Fußball berücksichtigt die vorliegende Arbeit insbesondere funktionelle Tests für die Anforderungen Kraft, Schnelligkeit, Agility und Bewegungsqualität als koordinative Eigenschaft. Neben den Testverfahren für einzelne Beanspruchungsformen werden auch kombinierte Testprotokolle diskutiert. Es werden sowohl die Durchführungsvarianten und wissenschaftlichen Gütekriterien als auch die Anwendbarkeit in der Praxis beschrieben. Während beidbeinige Sprünge Seitenunterschiede nur durch zusätzliches Equipment wie Kraftmessplatten oder Marker ermitteln können, eignen sich vor allem Hop Tests zur Aufdeckung von Seitendifferenzen anhand des Symmetrie-Index. Eine Vielzahl an kombinierten Testprotokollen ist bereits vorhanden, jedoch decken diese bisher nur einen Teilbereich des Anforderungsprofils eines Spielsportlers ab. Für die Praxis sollte eine geeignete Testbatterie mit einer Auswahl der beschriebenen funktionellen Tests zusammengestellt werden, die wissenschaftlichen und praktikablen Richtwerten unterliegen und zudem als Pre-Injury Screening vor und während der Saison eingesetzt werden können.

Methadone has a neurophysiological effect in humans that can be analyzed quantitatively using an electroencephalogram (EEG). This study considers the effects of neurophysiological for delta, theta, alpha, and beta frequency band in all brain regions for two dosage ranges categorized as low dose ( 100 mg). The EEG signals of methadone patients are compared to the healthy subject to analyze the brain alteration after one-hour methadone administration using resting condition (open-eyes and closed-eyes) test protocol. This study uses 17 channels with 10-20 international standard systems for the electrode caps. This study uses 4th order Butterworth bandpass (0.3-70 Hz) filter and STFT Hanning-window to determine the power spectral density. The results indicate that for low dose methadone patient, all the power spectrum of frequency band increase relative to healthy subjects in all brain regions and still follow the trend of healthy subjects’ EEG oscillation. Nevertheless, for high dose methadone patients, only the delta and theta that follow the trend of healthy subject’s EEG oscillation, but the value of power spectrum decreases relative to healthy subjects in all brain regions. Further, the power spectrum of alpha and beta for high dose methadone patients has different values for different brain regions.

Trail running places specific physiological demands on the human body due to its uphill and downhill running sections. We developed and investigated a more sport-specific trail exercise test protocol (inclination and speed incremental protocol), and compared it to two standard exercise test protocols (horizontal step and ramp protocol) in thirteen highly trained trail runners (age 31±6 years, height 179±6.4 cm, weight 69.2±7.9 kg, BMI 21.6±2.1 kg/m2). The maximum oxygen uptake (VO2max) measured during the trail test (62.5±5.9 ml·kg−1·min−1, [95% CI: 59.0-66.1]) was significantly higher compared to both the step test (60.1±5.3 ml·kg−1·min−1, [95% CI: 56.8-63.3], p=0.024) and the ramp test (59.7±5.5 ml·kg−1·min−1, [95% CI: 56.4-63.0], p=0.028). Background Despite data supporting the use of simulation training in procedural specialties and accreditation requirements, few options exist for electrophysiology (EP) training. We aimed to develop a low-cost, accessible simulator for training in EP mapping, and to test the simulator in a group of novice users. Methods Our mapping simulator is composed of three components: an acrylic case representing torso and thigh; three-dimensional (3D) printed cardiac models; and a commercially available mapping system. Using a proprietary flexible material that mimics the consistency of human heart tissue, we created an anatomically accurate model of a normal right atrium (RA) from computed tomography data. We developed a test protocol consisting of two activities: creation of a RA shell and timed navigation to specific locations within the RA shell. Seventeen participants were randomized to either practice versus no practice on the simulator, and repeated simulator and self-assessment tests were performed after 1 week. We measured volume of the RA map and time taken and distance from the target sites for each target location. Results Both groups showed improvement in generation of geometry, volume, time to target, and self-assessed comfort level after initial exposure to the simulator. Compared with no-practice, the practice group demonstrated an improved sense of confidence in mapping. Conclusions Focused training in EP mapping using a novel simulator created with 3D printed heart models and a standard mapping system is feasible for use in the training environment. Exposure to the simulator is associated with improved mapping skills and trainee comfort level.

The intervertebral disc is a complex joint that acts to support and transfer large multidirectional loads, including combinations of compression, tension, bending, and torsion. Direct comparison of disc torsion mechanics across studies has been difficult, due to differences in loading protocols. In particular, the lack of information on the combined effect of multiple parameters, including axial compressive preload and rotation angle, makes it difficult to discern whether disc torsion mechanics are sensitive to the variables used in the test protocol. Thus, the objective of this study was to evaluate compression-torsion mechanical behavior of healthy discs under a wide range of rotation angles. Bovine caudal discs were tested under a range of compressive preloads (150, 300, 600, and 900N) and rotation angles (± 1, 2, 3, 4, or 5°) applied at a rate of 0.5°/s. Torque-rotation data were used to characterize shape changes in the hysteresis loop and to calculate disc torsion mechanics. Torsional mechanical properties were described using multivariate regression models. The rate of change in torsional mechanical properties with compression depended on the maximum rotation angle applied, indicating a strong interaction between compressive stress and maximum rotation angle. The regression models reported here can be used to predict disc torsion mechanics under axial compression for a given disc geometry, compressive preload, and rotation angle.

Preventing healthcare worker exposure to dangerous pathogens is a crucial factor in sustaining the fight against Ebola, Coronaviruses, and other microorganisms that may cause infectious diseases. It is critical that personal protective equipment (PPE) used to protect healthcare workers be safe and effective in meeting labeled performance claims for barrier resistance to deadly viruses such as Ebola (Zaire ebolavirus) and severe acute respiratory syndrome coronavirus 2. The aim of this study is to develop a readily reproducible and standardized simulated use test for gown and drape materials using a blood test soil to evaluate the ability of the materials to prevent penetration of clinically relevant test soils at clinically relevant pressures. We designed and constructed a simulated use "elbow lean"apparatus based on the apparatus in ASTM F1819, Standard Test Method for Resistance of Materials Used in Protective Clothing to Penetration by Synthetic Blood Using a Mechanical Pressure Technique, that applies a physiologically relevant load to PPE material. We developed a test protocol for our proposed method and used the apparatus on a variety of PPE materials to assess penetration of a blood test soil. The testing demonstrated that the new apparatus can be used to assess penetration of a blood test soil through PPE materials under conditions that are more clinically relevant than those in ASTM F1670, Standard Test Method for Resistance of Materials Used in Protective Clothing to Penetration by Synthetic Blood, and ASTM F1671, Standard Test Method for Resistance of Materials Used in Protective Clothing to Penetration by Blood-Borne Pathogens Using Phi-X174 Bacteriophage Penetration as a Test System. The apparatus and procedure detailed in this study will be proposed for use in the development of a new standard test method for assessing mechanical penetration of PPE to test soils that better simulate clinical use conditions. Mechanomyography (MMG) is a non-invasive technique that records muscle contraction using sensors positioned on the skin's surface. Therefore, it can have its signal attenuated due to the adipose tissue, directly influencing the results. This study evaluates the influence of different mass added to a sensor's assembly and the adipose tissue on MMG signals of elbow flexor muscles. Test protocol consisted of skinfold thickness measurement of 22 volunteers, followed by applying 2–3 s electrical stimulation for muscle contraction during the acquisition of MMG signals. MMG signals were processed in the time domain, using the average of the absolute amplitude, and expressed in gravity values (G), termed here as MMG(G). Tests occurred four times with different sensor masses. MMG data were processed and analyzed statistically using Friedman and Kruskal-Wallis tests to determine the differences between the MMG signals measured with different sensor masses. The Mann-Whitney analysis indicated differences in the MMG signals between groups with different skinfold thickness. MMG(G) signals suffered attenuation with increasing sensor mass (0.4416 G to 0.94 g; 0.3902 G to 2.64 g; 0.3762 G to 5.44 g; 0.3762 G to 7.14 g) and adipose tissue.

Although ice hockey helmet standards mitigate the risk of catastrophic head injuries, the risk of concussion remains high. To improve protection, helmets need to be evaluated using impact conditions reflecting how concussions occur in ice hockey. The purpose of this research was to evaluate how three helmet impact tests represent three common concussive events in ice hockey. An ice drop test (representing head-to-ice impacts), 30° and 45° anvil boards drop tests (representing head-to-boards impacts), and medium and high shoulder compliance pneumatic ram tests (representing shoulder-to-head impacts) were performed on a hybrid III headform. Finite element analysis using the University College Dublin Brain Trauma Model was conducted to calculate maximum principal strain (MPS). The mean dynamic response and MPS from each helmet test were compared to a dataset of concussive injury reconstructions. Stepwise forward multiple linear regressions identified the dynamic response variables producing the strongest relationship with MPS for each helmet test and concussion reconstructions. The results indicated that the ice and boards drop test and shoulder ram test had magnitudes and relationships between variables similar to the concussion reconstructions. The proposed testing methodologies in this study closely approximated concussion mechanics in ice hockey and inform improved helmet test standards and design.

The Japan New Car Assessment Program (J-NCAP) evaluates the performance of cars in terms of protection against whiplash injuries in rear-end collisions. In the test protocol, a simplified triangular acceleration is applied to the sled. This study clarifies whether biofidelic rear-impact dummy II (BioRID II) measurements obtained for simplified triangular acceleration reflect car-to-car rear-end impacts in real-world accidents in Japan. We conducted a car-to-car rear-end impact experiment and a simplified-triangular-acceleration sled test. Our results indicate that the time series of dummy responses were approximately consistent in the two test conditions. The neck injury criterion (NIC) and maximum acceleration of the head and T1 measured using the BioRID II dummy were similar in the car-to-car and sled experiments. This revealed that the J-NCAP test protocol using simplified triangular acceleration reflects the car-to-car rear-end impact experiment using Japanese cars, in terms of the NIC and maximum acceleration of the head and T1.
