Macro- and microalgae-based foods are becoming popular due to their high nutritious value. The algal biomass is enriched with polysaccharides, protein, polyunsaturated fatty acids, carotenoids, vitamins and minerals. However, the most promising fraction is polysaccharides (PS) or their derivatives (as dietary fibers) which are not entirely fermented by colonic bacteria hence act as potential prebiotic. Primarily, algae become famous as prominent protein sources. Recently, these are widely adopted as functional food (e.g., desserts, dairy products, oil-derivatives, pastas etc.) or animal feed (for poultry, cattle, fish etc.). Besides prebiotic and balanced amino acids source, algae derived compounds implied as therapeutics due to comprising bioactive properties to elicit immunomodulatory, antioxidative, anticancerous, anticoagulant, hepato-protective, and antihypertensive responses. Despite the above potentials, broader research determinations are inevitable to explore these algal compounds until microalgae become a business reality for broader and specific applications in all health domains. However, scale up of algal bioprocess remains a major challenge until commercial affordability is accomplished which can be possible by discovering their hidden potentials and increasing their value and application prospects. This review provides an overview of the significance of algae consumption for several health benefits in humans and animals mainly as prebiotics, however their functional food and animal feed potential are briefly covered. Moreover, their potential to develop an algal-based food industry to meet the people's requirements not only as a sustainable food solution with several health benefits but also as therapeutics is inevitable.

This study compares the antioxidant capacity of non-sterilized lemon product (NLFP) and sterilized lemon fermented product (LFP). After gastrointestinal simulation reaction, the antioxidant capacity decreased by 50% in lemon juice (LJ), 30%–40% in NLFP, and 40% in LFP respectively. For the bioactive ingredients, sterilized LFP had a polyphenol of 25.8 mg GAE/mL, ascorbic acid of 55.2 mg/mL, and flavonoids of 75.2 mg/L, which were higher than LJ. NLFP and LFP effectively increased the activity of Glutathione Peroxidase (GPx), superoxide dismutase (SOD), catalase (CAT) and mitochondrial membrane potential, reduced the reactive oxygen species (ROS) content. The results in this study indicate that LJ with lactic acid fermentation can increase biologically active ingredients and antioxidant capacity which is important for future development.

To assess the potential of by-products of the black bean fermented soybean sauce manufacturing process as new functional food materials, we prepared black bean steamed liquid lyophilized product (BBSLP) and analysed its antioxidant effects in vitro. RAW264.7 macrophages were cultured and treated with BBSLP for 24 h, and 1 μg/mL lipopolysaccharide (LPS) was then used for another 24 h to induce inflammation. The cellular antioxidant capacity and inflammatory response were then analysed. Activation of nuclear factor kappa B (NF-κB) signaling in RAW264.7 macrophages was also analysed. Results showed BBSLP had 2,2-diphenyl-1-(2,4,6-trinitrophenyl)hydrazyl (DPPH) and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium (ABTS+) radical-scavenging abilities and reducing power in vitro. The levels of both reactive oxygen species (ROS) and thiobarbituric acid reactive substances (TBARS) were reduced after RAW264.7 macrophages were treated with BBSLP after LPS induction. After RAW264.7 macrophage treatment with BBSLP and induction by LPS, the levels of inflammatory molecules, including nitric oxide (NO), prostaglandin E2 (PGE2), IL-1α, IL-6 and TNF-α, decreased. NF-κB signaling activity was inhibited by reductions in IκB phosphorylation and NF-κB DNA-binding activity after RAW264.7 macrophages were treated with BBSLP after LPS induction. In conclusion, BBSLP has antioxidant and anti-inflammatory capabilities and can be a supplement material for functional food.

Fish bones (FBs) are aquatic by-products that are sources of antioxidant-active peptides, calcium dietary supplements, and biomedical materials. Usually, fermentation of these by-products via microorganisms brings desirable changes, enhancing their value. This study investigates the value addition of FB when fermented with Monascus purpureus (MP) for different time intervals, such as 3 days (F3) and 6 days (F6). The results indicate that the soluble protein, peptide, amino acid and total phenol content, as well as the antioxidant capacity (DPPH, ABTS+ radical scavenging activity, and relative reducing power), of F3 and F6 were significantly increased after fermentation. Furthermore, the ROS contents of F3 and F6 were reduced to a greater extent than that of hydrogen peroxide (H2O2) in Clone-9 cells. The MMP integrity, as well as the SOD, CAT, and GPx activity, of F3 and F6 were also increased significantly compared to the H2O2 in Clone-9 cells. Notably, F3 and F6 displayed significant reductions in ROS content, as well as elevate, SOD activity and MMP integrity in Clone-9 cells, when compared with the native FB. These results indicate that the FBs fermented with MP for 3 days (F3), and 6 days (F6) have antioxidant capacity, with possible applications as natural food supplements.

Gynura bicolor (Roxb. and Willd.) DC (G. bicolor) is generally used as a dietary vegetable and traditional herb in Taiwan and the Far East. G. bicolor exerts antioxidant and anti-inflammatory effects and regulates blood lipids and cholesterol. However, the effects of G. bicolor on endothelial transmigration and atherosclerosis are not clear. The present study investigated the effects of G. bicolor on endothelial permeability and transmigration in human endothelial cells. We prepared G. bicolor ether extract (GBEE) for use as the experimental material. Under TNF-α stimulation, HL-60 cell adherence to EA.hy926 cells, the shape of EA.hy926 cells, and the expression of adhesion molecules and transmigration-related regulatory molecules were analysed after pretreatment with GBEE for 24 h. GBEE inhibited leukocyte adhesion to endothelial cells, reduced intercellular adhesion molecule-1 (ICAM-1) and platelet endothelial cell adhesion molecule-1 (PECAM-1) expressions, and decreased endothelial monolayer permeability. GBEE also reduced paracellular transmigration by reducing the levels of reactive oxygen species (ROS), Src phosphorylation, and vascular endothelial-cadherin (VE-cadherin) phosphorylation. GBEE reduced transcellular migration via inhibition of Ras homolog family member A (RhoA) and Rho-associated protein kinase (ROCK) expression and phosphorylation of the ezrin-radixin-moesin (ERM) protein. Incubation of EA.hy926 cells with GBEE for 8 h and stimulation with TNF-α for 3 h reduced the phosphorylation of the inhibitor of kappa B (IĸB) and DNA-binding activity of nuclear factor-ĸB (NF-ĸB). These results suggest that GBEE has a protective effect against endothelial dysfunction via suppression of leukocyte-endothelium adhesion and transmigration.

Gynura bicolor(Roxb. and Willd.) DC (G. bicolor) is a dietary vegetable in the Far East. The aims of the present study were to investigate the antioxidant effects of theG. bicolorwater extract (GBWE) and its ability to regulate the blood lipid and lipoprotein profiles. In this study, the pigment composition and antioxidant ability of the GBWE were analyzed. Syrian hamsters were fed a high-fat diet (HFD) and the GBWE for 12 weeks, and the blood lipid levels, lipoprotein profiles, and cholesterol metabolism-related enzyme levels were then examined. The results showed that the GBWE exhibited excellent 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging activity and ferrous-ion-chelating ability. The hepatic glutathione levels in the hamsters were increased after the administration of low (0.4 g/kg BW, GBWE-L) or high (0.8 g/kg BW, GBWE-H) levels of the GBWE. The GBWE-H-treated hamsters exhibited significantly decreased serum levels of total cholesterols (TC) and low-density lipoprotein-cholesterol (LDL-C) and significantly increased levels of lectin-cholesterol acetyltransferase (LCAT). These results showed that GBWE-H can reduce the total cholesterol and LDL-C levels in HFD-fed hamsters, and this reduction might be involved in the regulation of LCAT expression.

Lemon (Citrus limon) has antioxidant, immunoregulatory, and blood lipid-lowering properties. This study aimed to determine the effect of the lemon fermented product (LFP) which is lemon fermented with Lactobacillus OPC1 to prevent obesity. The inhibition of lipid accumulation in 3T3-L1 adipocytes is examined using a Wistar rat model fed a high-fat diet to verify the anti-obesity efficacy and mechanism of LFP. Here, it was observed that LFP reduced cell proliferation and inhibited the lipid accumulation (8.3%) of 3T3-L1 adipocytes. Additionally, LFP reduced body weight (9.7%) and fat tissue weight (25.7%) of rats, reduced serum TG (17.0%), FFA (17.9%), glucose (29.3%) and ketone body (6.8%), and increased serum HDL-C (17.6%) and lipase activity (17.8%). LFP regulated the mRNA expression of genes related to lipid metabolism (PPARγ, C/EBPα, SREBP-1c, HSL, ATGL, FAS, and AMPK). Therefore, LFP reduces body weight and lipid accumulation by regulating the mRNA expression of genes related to lipid metabolism. Overall, our results implicate LFP as a potential dietary supplement for the prevention of obesity.

2,3,5,4′-Tetrahydroxystilbene-2-O-β-D-glucoside (THSG) has been shown to have antioxidative and anti-inflammatory effects. Oxidative and inflammatory reactions are related to the development of colorectal carcinoma (CRC). In the present study, we characterized the preventive activities of THSG on colon carcinogenesis using the azoxymethane- (AOM-) mediated rat colon carcinogenesis model. F344 male rats were randomly divided into 5 groups (untreated and AOM model rats treated with or without THSG at 30, 150, or 250 mg/kg) after which the numbers of aberrant crypt foci (ACF) were assessed in the colon tissues of all rats. The expressions of nuclear factor-κB (NF-κB), cyclooxygenase-2 (COX-2), matrix metalloproteinase proteins (MMPs), and carcinoembryonic antigen (CEA) were measured as effective early predictors of CRC using western blot analysis. Treatment with THSG (150 or 250 mg/kg) induced a 50% reduction in total colonic ACF formation (P<0.05). Furthermore, our results revealed a downregulation of CEA and NF-κB protein levels in the reduced number of ACF elicited by treatment with THSG, whereas levels of COX-2 and MMPs proteins were not changed. Collectively, THSG may be a promising natural lead compound or drug candidate for treating early phases of CRC.

Panax notoginseng (P. notoginseng) and its components are used as traditional Chinese medicine for cardiovascular disease, although studies concerning the anti-metastatic properties of these compounds are limited. The goal of this study was to investigate the effects of notoginsenoside R1 (NGR1), an important compound derived from P. notoginseng, on the metastasis of human colorectal cancer (CRC). The migratory, invasive, and adhesive abilities of cultured human CRC cells (HCT-116) treated with NGR1 and expression of metastasis‑associated regulatory molecules were assessed. The migratory and invasive abilities of the HCT-116 cells were reduced after treatment with 75, 150 or 300 µM NGR1 for 24 h. When HCT-116 cells were incubated with 150 or 300 µM NGR1 for 24 h, matrix metalloproteinase (MMP)-9 expression was reduced compared with that of the control group. In the adhesion reaction assays, treatment with 150 or 300 µM NGR1 led to significantly decreased adhesion of the HCT-116 cells to endothelial cells (EA.hy926 cells). Levels of integrin-1 protein were significantly decreased in the HCT-116 cells following treatment with 75, 150 or 300 µM NGR1, and levels of E-selectin and intercellular adhesion molecule 1 (ICAM-1) proteins were significantly decreased in the EA.hy926 cells treated with 75, 150 or 300 µM NGR1. Scanning electron microscopy examination indicated that HCT-116 cells treated with lipopolysaccharide (LPS) combined with 300 µM NGR1 exhibited a less flattened and retracted shape compared with cells treated with LPS alone, and this change in shape is characteristic of extravasation. Additionally, the transepithelial electrical resistance of the EA.hy926 endothelial cell monolayer increased after incubation with 150 or 300 µM NGR1 for 24 h. Overall, these results demonstrated the anti-metastatic properties of 150 or 300 µM NGR1, a compound that affects CRC metastasis by inhibiting cell migration, invasion, and adhesion and by regulating expression of metastasis-associated signalling molecules.

Watermelon rind, comprising 35% of the fruit’s total mass, usually discarded due to its unappealing flavor and lack of sweetness. In this study, candied watermelon rind was prepared through osmotic dehydration at vacuum (VOD) and atmospheric pressure (OD). The effects of independent variables (sucrose concentration, immersion time and temperature) on the dependent variables, including solid gain (SG) and water loss (WL) of watermelon rind, were evaluated using multiple response surface methodology. The SG and WL increased with the increasing sucrose concentration, immersion time and temperature for both VOD and OD, but VOD greatly increased the mass transfer rate. The kinetic constants were obtained by fitting the data of SG and WL to the Fick’s diffusion model. The VOD had higher kinetic constants and dehydration efficiency indices than that of OD. Besides, the mass transfer model and RSM model were statistically compared by the coefficient of determination, root mean square error and absolute average deviation, based on the validation data set. The RSM and mass transfer models provide information about the osmotic dehydration for predicting the sugar content of candied product, and helping the designers and researchers find the best design parameters and the most effective process conditions.

Gynura bicolor (Roxb. & Willd.) DC., a perennial plant belonging to the Asteraceae family, is originated from the tropical area of Asia. The total hemocyte count (THC), phenoloxidase (PO) activity, respiratory bursts (RBs), superoxide dismutase (SOD) activity, and lysozyme activity were examined after white shrimp Litopenaeus vannamei had been fed diets containing the water extract of G. bicolor at 0 (control), 0.5, 1.0, and 2.0 g (kg diet) −1 for 7–28 days. The results indicated that these parameters increased accordingly with the amount of extract and time. THCs of the shrimp fed the G. bicolor diets at 1.0 and 2.0 g (kg diet) −1 were significantly higher than that fed the control diet for 14–28 days. For the shrimp fed the G. bicolor diets at 0.5, 1.0, and 2.0 g (kg diet) −1 , the PO, RBs, and lysozyme activities reached the highest levels after 7 days, whereas SOD activity reached the highest levels after 14 days. In a separate experiment, white shrimp L. vannamei fed the diets containing the G . bicolor extract for 28 days were challenged with Vibrio alginolyticus at 3 × 10 6 cfu shrimp −1 and white spot syndrome virus (WSSV) at 1 × 10 3 copies shrimp −1 . The survival rate of the shrimp fed the G . bicolor diets was significantly higher than that of the shrimp fed the control diet at 48–144 h post challenge V. alginolyticus and WSSV. For the shrimp fed the G. bicolor diets at 0.5, 1 and 2 g (kg diet) −1 under challenges of V. alginolyticus and WSSV, their LPS- and β-1,3-glucan-binding protein (LGBP) and peroxinectin (PE) mRNA expressions were significantly higher than those of the challenged control shrimp at 12–96 and 24–144 h post-challenge, respectively. We concluded that dietary administration of a G . bicolor extract could enhance the innate immunity within 28 days as evidenced by the increases in immune parameters (PO, RBs, and lysozyme) and antioxidant enzyme (SOD) activities of shrimp to against V. alginolyticus and WSSV infections.

Copper oxide nanoparticles (CuO NPs) are known to exhibit toxic effects on a variety of cell types and organs. To determine the oxidative impact of CuO NPs on hepatocellular carcinoma (HCC) cells, well-differentiated (HepG2) and poorly differentiated (SK-Hep-1) cells were exposed to CuO NPs. Cell viability assay showed that the median inhibition concentration (IC50) for SK-Hep-1 and HepG2 cells was 25 μg ml(-1) and 85 μg ml(-1), respectively. Cellular fluorescence intensity using DCFH-DA staining analysis revealed significant intracellular reactive oxygen species (ROS) generation of up to 242% in SK-Hep-1 cells, compared with 86% in HepG2 cells. HPLC analysis demonstrated that a CuO NP treatment caused cellular GSH depletion of 58% and a GSH/GSSG ratio decrease to ∼0.1 in SK-Hep-1 cells. The oxidative stress caused by enhanced superoxide anion production was observed in both HepG2 (146%) and SK-Hep-1 (192%) cells. The Griess assay verified that CuO NPs induced NO production (170%) in SK-Hep-1 cells. Comet assay and western blot further demonstrated that CuO NPs induced severe DNA strand breakage (70%) in SK-Hep-1 cells and caused DNA damage via increased γ-H2AX levels. These results suggest that well-differentiated HepG2 cells possess a robust antioxidant defense system against CuO NP-induced ROS stress and exhibit more tolerance to oxidative stress. Conversely, poorly differentiated SK-Hep-1 cells exhibited a deregulated antioxidant defense system that allowed accumulation of CuO NP-induced ROS and resulted in severe cytotoxicity.

Antrodia camphorata is a fungus that is endemic to Taiwan, and its fruiting body has been used as a folk medicine for the prevention or treatment of diverse diseases. The present study is aimed at investigating the antimelanogenesis and antioxidation effect of the ethanolic extract of Antrodia camphorata fruiting body (EE-AC), as well as its antiproliferation effects in B16-F0 melanoma cells. Regarding antimelanogenic effects, EE-AC had effective cupric ions reducing capacity and expressed more potent inhibitory effect than kojic acid on mushroom tyrosinase activity. Moreover, EE-AC significantly inhibited cellular tyrosinase activity and the melanin content in B16-F0 cells at 12.5 μg/mL concentration without cell toxicities. Regarding antioxidant effects, EE-AC exhibited potent DPPH radical- and SOD-like-scavenging activities. Regarding antiproliferative effects, EE-AC exhibited a selective cytotoxic effect and markedly inhibited the migration ability of B16-F0 cells. EE-AC increased the population of B16-F0 cells at sub-G1 phase of the cell cycle. EE-AC also caused the increase of early apoptotic cells and chromatin condensation, which indicated the apoptotic effects in B16-F0 cells. We demonstrated that EE-AC possessed antimelanogenic, antioxidant and anti-skin cancer actions. The results would contribute to the development and application of cosmetics, healthy food and pharmaceuticals.

α-Phellandrene (α-PA) is a component of dietary spices and herbs. The effect of α-PA on anticancer is unclear. This study aims to investigate the effects of α-PA on liver tumor cell death fate. Human liver tumor (J5) cells were incubated with α-PA and analyzed for cell cycle distribution, expression of Bax, Bcl-2, poly (ADP-ribose) polymerase (PARP) protein, and caspase-3 activity of J5 cells, and levels of nitric oxide (NO) production, lactate dehydrogenase (LDH) leakage, and ATP depletion were also analyzed in this study. Results found that α-PA significantly (P < 0.05) decreased the cell viability of J5 cells after 24-h treatment. The cell cycle distribution, Bax, Bcl-2, PARP protein levels, and caspase-3 activity of J5 cells did not change for 24 h after treatment with 30 μM α-PA. Reactive oxygen species levels significantly increased, mitochondrial membrane potential levels significantly decreased when J5 cells were treated with 30 μM α-PA for 24 h (P < 0.05). Thirty μM α-PA significantly (P < 0.05) increased the necrotic cell number, NO production, LDH leakage, and ATP depletion after 24 h of incubation. These results suggest that α-PA induced J5 cell necrosis but not apoptosis, and α-PA-induced necrosis possibly involved ATP depletion.

The increasing prevalence of hospital-acquired infection and the evolution and increasing resistance of pathogens toward antibiotics can cause serious health problems and disease-related mortality. In this study, we introduce a simple process and inexpensive method to synthesize CuO nanoparticles and silver-functionalized copper oxide (Ag@CuO) nanocomposites as well as to validate their potential antibacterial efficiency against the following three common nosocomial infection-associated bacterial pathogens: E. coli, S. enterica and S. aureus. We show that Ag@CuO significantly disturbs pathogen growth and viability compared with CuO. Further, we find that Gram-positive S. aureus is susceptible to CuO-induced cell structure damage, while Ag@CuO can induce more extensive architectural destruction and ROS generation in both Gram-positive and Gram-negative bacterial pathogens. This study indicates that Ag@CuO nanoparticles can act as a disinfection system and can be used in antibacterial applications for the future prevention of nosocomial infection in medical and/or health institutions.

Cells reorganize their membrane biomechanical dynamics in response to environmental stimuli or inhibitors associated with their physiological/pathological processes, and disease therapeutics. To validate the biophysical dynamics during cell exposure to anti-cancer drugs, we investigate the nanoscale biological characterization in melanoma cells undergoing cisplatin treatment. Using atomic force microscopy, we demonstrate that the cellular morphology and membrane ultrastructure are altered after exposure to cisplatin. In contrast to their normal spindle-like shape, cisplatin causes cell deformation rendering cells flat and enlarged, which increases the cell area by 3-4 fold. Additionally, cisplatin decreases the topography height values for both the cytoplasmic and nuclear regions (by 40-80% and 60%, respectively). Furthermore, cisplatin increases the cytoplasmic root mean square roughness by 110-240% in correlation with the drug concentration and attenuates the nuclear RMS by 60%. Moreover, the cellular adhesion force was enhanced, while the Young's modulus elasticity was attenuated by ∼2 and ∼2.3 fold, respectively. F-actin phalloidin staining revealed that cisplatin enlarges the cell size through enhanced stress fiber formation and promotes cytoskeletal reorganization. Immunoblot analyses further revealed that the activities of focal adhesion proteins, such as FAK and c-Src, are upregulated by cisplatin through phosphorylation at tyrosine 397 and 530, respectively. Collectively, these results show that cisplatin-treated melanoma cells not only exhibit the upregulation of FAK-mediated signaling to enhance the cytoskeleton mechanical stretch, but also promote the cytoskeletal rearrangement resulting in 43% decrease in the cell modulus. These mechanisms thus promote the malignancy and invasiveness of the melanoma cells.

Carcinoma cell differentiation stage is an important indicator of cell behavior. For example, cell mobility is much higher for poorly-differentiated hepatocellular carcinoma SK-Hep-1 cells than for well-differentiated HepG2 cells. In this study, we have cultured HepG2 and SK-Hep-1 cells on chemically patterned polydimethylsiloxane (PDMS) substrates to observe differences in the adhesion properties and cell structure that occur due to the patterns. Both cell lines showed a preference for the hydrophobic regions on the patterned PDMS surface with SK-Hep-1 cells achieving a higher density than HepG2 for the same cell-count solutions. Further, SK-Hep-1 cells adopted the square or hexagonal shape of the surface patterns while HepG2 cells maintained their more rounded shape. AFM force measurement arrays were also performed on the cell surfaces to measure and map adhesion values between the tip and cell surface membrane. These results demonstrate that, in addition to cell shape and size, adhesion expression in hepatocellular carcinoma cells is differentiation stage dependent. Further, the ability of the SK-Hep-1 cells to adopt the shape of the substrate pattern indicates they are more structurally labile, which may contribute to their higher mobility.

Zingerone, one of the active components of ginger, is a phenolic alkanone with antioxidant and anti-inflammatory properties. The effects of zingerone supplementation on the growth, immunity, and disease resistance of Pacific white shrimp ( Litopenaeus vannamei ) juveniles were studied. Four experimental diets, including a control diet (without zingerone enrichment) and 1, 2.5, and 5 mg zingerone (kg diet) −1 were used. After 56 days of culture, shrimp fed diets supplemented with 1, 2.5, and 5 mg zingerone (kg diet) −1 had significantly greater weight gain and feed efficiency than the controls. Furthermore, after 56 days of culture, shrimp fed all doses of the zingerone diet had higher survival rates compared to the controls after 24–72 h of challenge by the pathogen, Vibrio alginolyticus . Significantly increased phenoloxidase levels were found in shrimp fed the zingerone diets at all doses, and respiratory bursts, lysozyme and phagocytic activities of shrimp fed 2.5 and 5 mg zingerone (kg diet) −1 also significantly increased. Neither the total hemocyte count nor superoxide dismutase activity of the experimental and control groups revealed significant differences at any dose. The results indicate that zingerone can be recommended as a supplement to shrimp feed to increase growth, immunity, and disease resistance against the pathogen, V. alginolyticus . Use of zingerone as appetizer and immunostimulant in shrimp is promising.

Fish gelatin hydrolysates have been shown to possess various biological activities due to their unique Gly-Pro-Y and Gly-X-Hyp sequences. In the current study, fish gelatin was extracted from non-extruded milkfish scale (FSG1) or extrusion-pretreated milkfish scale (FSG2), extracted gelatins were hydrolyzed with different combinations of Flavourzyme and Alcalase to give four different hydrolysates, namely: FSGH1 (FSG1 hydrolyzed with Flavourzyme), FSGH2 (FSG1 hydrolyzed with Alcalase + Flavourzyme), FSGH3 (FSG2 hydrolyzed with Flavourzyme), and FSGH4 (FSG2 hydrolyzed with Alcalase + Flavourzyme). The extrusion-pretreatment process enhanced the extraction yield of gelatin from fish scale. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and Fourier transform infrared (FTIR) analyses showed the extracts FSG1 and FSG2 possessed characteristics of gelatin. Moreover, the physicochemical characteristics of FSGH1&ndash, FSGH4 were examined by analyses of their degree of hydrolysis, amino acid composition, UV spectrum, FTIR spectrum, molecular weight, and RP-HPLC profile. Additional biological functional analyses showed that all of the studied gelatin hydrolysates FSGH1&ndash, FSGH4 possessed antioxidant activity dose-dependently as revealed by DPPH scavenging, ABTS scavenging, and reducing power analyses. In addition, FSGH2 and FSGH4 showed higher angiotensin-I-converting enzyme (ACE)-inhibitory activity as compared to FSGH1 and FSGH3. Taken together, FSGH2 and FSGH4 showed high antioxidant activity and potent anti-ACE activity. Due to the potential antioxidant and antihypertensive properties of FSGH2 and FSGH4, further research is needed to explore their possible use as natural supplementary raw materials in food and nutraceutical products.

Introduction Colorectal cancer is the third most commonly diagnosed cancer in the word. Carnosine is an endogenous dipeptide found in muscle and brain tissues. The aims of this study are to investigate the regulation of carnosine on adhesion and extravasation of metastasis in human colorectal cancer. Material and methods The human colorectal cancer cells (HCT-116) and human umbilical vein cell line (EA. hy926) cultured were as experimental models. Those adhesive and extravasate abilities and regulate molecular expression were analysed in this study. Results and discussions In adhesion ability assay results shown carnosine were significantly decreased HCT-116 cells adhesion to endothelial cells (EA. hy926 cells). From adhesion-regulating proteins expression analysis result show that carnosine was decreased the levels of integrin in HCT-116 cells and ICAM-1 and E-selectin in EA hy926 cells after 24 hour incubation (p Conclusion The results indicate that carnosine can inhibit the adhesion and extravasation through suppressing of carnosine on NF-κB transcript activation and its regulator adhesion and extravasation related molecular expression in HCT-116 cells and EA hy926 cells.

Here we analyzed carboxymethylcellulase (CMCase; EC 3, 2, 1, 4), one of the key enzymes in the early symbiotic process, in Rhizobium. Specific immunogold labeling of electron microscopy was confirmed in Sinorhizobium fredii BCRC15769, ATCC35423, Sinorhizobium meliloti ATCC9930, and barely detected in Bradyrhizobium japonicum BCRC13528, ATCC10324 and Rhizobium rhizogenes ATCC11325. Non-specific labeling was detected in Rhizobium leguminosarum bv. viceae ATCC10004, Rhizobium leguminosarum bv. trifolii ATCC10328, and Mesorhizobium loti ATCC33669. Treatment of S. fredii BCRC15769 in the early log phase with the flavonoid genistein caused relocalization of CMCase. Together our data suggests a role for CMCase in early symbiosis.

The ferula oyster mushroom (Pleurotus eryngii var. ferulae) is medicinal, edible, and delicious. Response surface methodology was used to determine the optimal conditions for the pro- duction of P. eryngii var. ferulae biomass in liquid submerged culture. Lyophilized mycelium was subjected to two water extracts and two elutes through column chromatography using methanol-ethyl acetate-dichloromethane (referred to as EAM and CHE, respectively). In addition, the crude protein and water-soluble polysaccharides were extracted from the cultured broth. Six samples were tested to explore their antigenotoxicity with the Ames test and a rec-assay, as well as antioxidant activities in in vivo and in vitro tests. EAM had the highest 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical- scavenging activity and ferric-reducing ability. Moreover, hamster serum showed the highest activities of glutathione peroxidase and superoxide dismutase after EAM was administered at a dose of 0.3 g kg -1 body weight daily among all tested samples. Meanwhile, the EAM group displayed signifi cantly higher activities of the two enzymes than did the control group. In the antimutagenicity test, EAM exhibited the highest inhibition rate against every tested mutagen at a low concentration of 0.08 mg mL -1 and a high concentration of 0.3 mg mL -1 in each plate, and it also had the highest anti-DNA- damaging activity among all test samples. EAM displayed the highest antigenotoxicity effect, followed by CHE, when using water-insoluble extracts, and CP was the highest one, followed by water extract (100°C, 30 minutes), water extract (60°C, 30 minutes), and SPPF, when using water-soluble extracts. The major constituents of EAM and CHE were identifi ed as ergosterol and 2-pentanol, respectively. Some extracts from P. eryngii var. ferulae have significant antioxidant activity and are strong anti- genotoxic agents.

To improve hybrid tilapia (Oreochromis niloticusxO. aureus) survival under cold shock, the influence of diets containing various dietary lipids was investigated. Four different diets were used which consisted of 12% fish oil, 12% palmitoleic oil 12% coconut oil, and a mixture of fish oil (7%) and corn oil (5%). Our results showed that during cold shock, the proportion of saturated fatty acids in the fish steadily and significantly decreased for all of the diets, but the proportion of monounsaturated fatty acids increased. Proportions of polyenoic fatty acids initially increased then stabilized for the mixed, fish, and coconut oil diets, but did not significantly increase until day 4 for the palmitoleic oil diet. The stearoyl-CoA desaturase (SCD) activity was the lowest on day 0 and then gradually increased for all diets. At any point, the enzymatic activity of SCD was the highest for fish on the mixed and the coconut oil diet, followed by the palmitoleic oil diet, and was lowest for the fish oil diet. The expression of SCD mRNA steadily increased for all diets, but increased more substantially for the mixed diet. On day 6, the expression was the highest for fish on the mixed diet, followed by the coconut oil diet, with the lowest levels for those on the palmitoleic and fish oil diets. These results show that dietary lipids strongly affect the fatty acid composition and SCD expression in tilapia under cold shock, and cold tolerance of this species is also affected.

α-Phellandrene (α-PA) is a cyclic monoterpene. To investigate the induction of autophagy by α-PA and its mechanism, human liver tumor cells (J5) were incubated with α-PA and analyzed for cell viability and the molecular regulation of pre-autophagosome origination and autophagosome formation. According to the results, PI3K-I, mTOR, and Akt protein levels were decreased after α-PA treatment compared to those of the control group (p < 0.05). The phosphorylation of Bcl-2, and PI3K-III, LC3-II and Beclin-1 protein levels in J5 cells were increased after α-PA treatment (p < 0.05). In addition, α-PA up-regulated nuclear p53 and down-regulated cytoplasmic p53 expression in J5 cells. The NF-κB pathway was activated, as indicated by increase in cytosolic phosphorylated IκB, nuclear NF-κB levels, and the DNA-binding activity of NF-κB after α-PA treatment in J5 cells (p < 0.05). These results suggest that α-PA can induce J5 cell autophagy by regulating mTOR and LC-3II expression, p53 signaling, and NF-κB activation in J5 cells.

A receptor for activated C-kinase (RACK) cDNA in the Mozambique tilapia (Oreochromis mossambicus) was cloned by reverse transcriptase polymerase chain reaction (RT-PCR) and rapid amplification of cDNA ends (RACE), and it was compared with other vertebrates. Nucleotide sequence analysis revealed that the full length of cDNA (1109 bp) cloned encompasses 954 bp open reading frame encoding 317 amino acid residues. The deduced amino acid sequence shares 95–99% identity with the teleosts and 77–95% compared with other vertebrates. Like most RACK sequences in other vertebrates, seven repeats of the WD-40 motif are well-conserved in the sequence of the tilapia. Further, the effectiveness of gonadotropin-releasing hormone (GnRH) regulation of RACK LH, and FSH gene expression was further investigated by a single injection of salmon GnRH (sGnRH) and chicken GnRH-II (cGnRH-II). The results showed that injection of tilapia brain with either sGnRH or cGnRH-II significantly increased mRNA levels of RACK, which then stimulate LH and FSH. This result suggests that RACK and protein kinase C (PKC) are involved in GnRH-mediated gonadotropin hormone (GtH) release. In addition, cGnRH-II enhanced the FSH and LH mRNA as well as sGnRH expression levels in tilapia; these results further demonstrate that both sGnRH and cGnRH regulate GtH through PKC.

Oocyte maturation in teleosts is characterized by a complex series of cytoplasmic and nuclear changes. The purpose of this study is to present sequential changes in the oocyte constituents and regulatory mechanisms involved in the hydration and osmotic regulation of oocytes during ¢nal maturation of ayu, Plecoglossus altivelis Temminck & Schlegel. Oocyte volumes and water contents in ayu (P. altivelis )m arkedly increased from 0.372 mm 3 and 60.5% at the mature stage to 0.712 mm 3 and 79.4% at the time of oviposition. In the same period, net increases in total free amino acids and electrolytes were noted, and involvements of these constituents in the water uptake by oocytes are discussed. Moderate oocyte hydration in ayu is accompanied by steady increases in Na 1 , but not K 1 , and these observations are in contrast to results for all other marine teleosts reported elsewhere. In the present study, a steady increase in oocyte Na 1 during maturation until ovulation predominantly results from diiusional leakage due to concentration gradients, while at the time of oviposition, both active transport and diiusional pathways play important roles in osmotic regulation in ayu oocytes.

Colorectal cancer is the third most commonly diagnosed cancer in the word. Carnosine is an endogenous dipeptide found in vertebrate skeletal muscles. It is known to have anti-fatigue, antioxidative, antihypertensive, antidiabetic, and cancer inhibiting effects. However, little research has been done regarding its influence on the metastasis of colon cancer. This study cultivated HCT-116 human colon cancer cells as a test model in order to investigate the impact of carnosine on the migration and invasion of human colon cancer cells. The results showed that 48-hour treatments of HCT-116 cells with 0.5, 1, or 5 mM carnosine each significantly inhibited the migration ability of the cells (P < 0.05). The 48-hour treatments with 0.5, 1, or 5 mM carnosine were also found to significantly reduce MMP-9 activity (P < 0.05), but not MMP-2 expression. Furthermore, when HCT-116 cells treated with 1 or 5 mM carnosine, invasion ability are significantly decreased and significantly increased E-cadherin expression (P < 0.05). Antrodia camphorata is a fungus native to Taiwan, and it is considered a precious medicinal agent. We analyzed triterpenoids, polysaccharides and 1,3-β-D-glucan, three major effective components in A. camphorata extracts (ACE). ACE exhibited a selective cytotoxic effect on BxPC-3 human pancreatic cancer cells. ACE markedly inhibited the migration ability of BxPC-3 cells. Treatment of BxPC-3 cells with ACE resulted in the increase of cells in the sub-G1 phase and G2/M phase arrest. Apoptosis was confirmed by validating phosphatidylserine externalization, the observation of characteristic chromatin condensation, and nuclear DNA fragmentation. ACE induced apoptosis in BxPC-3 cells through a mitochondria-dependent pathway by triggering an appropriate balance of bax/bcl-2, cytochrome c release, activation of caspase-9 and -3, and poly(ADP-ribose) polymerase cleavage. ACE shows great therapeutic potential due to its cytotoxic effects against BxPC-3 cells which include inhibiting cell migration and inducing mitochondria-mediated apoptosis.

Gynura bicolor (Roxb. & Willd.) DC. is widely distributed in certain areas of Asia and is very popular in vegetarian cuisine in Taiwan. This study investigates the effects of G. bicolor extracts with different polarities of 80 mg/kg body weight (BW) G. bicolor alcohol extract, 80 mg/kg BW G. bicolor water extract, and 80 mg/kg BW G. bicolor ether extract on Fe bioavailability using the hemoglobin repletion efficiency assay. Wistar rats were assigned to five groups: a group receiving an iron-deficient (ID) diet; a group receiving an ID diet supplemented with ferrous sulfate (20 mg Fe/kg BW); and three groups receiving ID diets supplemented with ferrous sulfate and one of G. bicolor alcohol extract, G. bicolor water extract, or G. bicolor water extract. The results indicated that the levels of hemoglobin, serum iron, serum ferritin, liver ferritin, hemoglobin regeneration efficiency, relative biological value, and hepcidin all were significantly higher than those of the ID diet group. Besides, the iron transporter divalent metal transporter-1 was significantly reduced, but iron release protein expression of ferroportin was significantly increased. It was concluded that G. bicolor extracts may promote iron bioavailability and regulate the expressions of divalent metal transporter-1 and ferroportin.

SUMMARY: Diets containing 40% of either starch or glucose were fed to triplicate groups of tilapia Oreochromis niloticus x O. aureus (initial weight: 0.80 ± 0.01 g) for 8 weeks, followed by a fasting of 1, 2 and 3 weeks. In starvation, fish previously fed the diet containing glucose showed significant (P < 0.05) weight loss compared with fish fed the diet containing starch. Liver glycogen was higher in fish fed a starch diet compared with fish fed a glucose diet during both feeding and fasting periods. Starvation also resulted in decreased liver glycogen in fish as the fasting period increased. Plasma glucose was higher in starch-fed fish during feeding and 1 week after fasting compared with glucose-fed fish. Plasma glucose of both dietary groups was lower in starvation than in the feeding period. Elevated plasma glucose was found 3 weeks and 2 weeks after fasting in fish prefed with starch and glucose diets, respectively. Plasma triglyceride increased as the fasting period increased in both groups. In starvation, plasma triglyceride were higher in fish prefed the starch diet than fish fed the glucose diet. Relative to the glucose diet, body lipid content was higher in fish fed the starch diet prior to fasting and 1 week after fasting. These data suggest that the physiological responses of tilapia during starvation are affected by prefasting diets containing different carbohydrates.

This study investigated the effects of alkylsulfonated chitosan of different molecular weights on intestinal lipid absorption, blood lipid profiles and circulating adhesion molecules. Syrian hamsters were fed an AIN-93G-based high‑fat diet (HFD) and were orally administered 5 or 10 mg/kg BW of oligomer (6 kDa) chitosan (OC), low‑molecular-weight (70 kDa) chitosan (LMC) or high‑molecular-weight (200 kDa) chitosan (HMC) four times per week for 12 weeks. Animals receiving 2.5 mg/kg BW lovastatin (LOVA) served as a positive control. The blood lipid profiles of these control animals revealed that all chitosans and LOVA significantly decreased total triglyceride, total cholesterol, low‑density lipoprotein (LDL)-cholesterol and very‑low‑density lipoprotein (VLDL)-cholesterol levels in a dose‑dependent manner compared to the HFD-fed controls (P0.05). The blood lipid lowering effectiveness of the three chitosans followed the order of LMCOCHMC. Hamsters receiving 5 and 10 mg/kg LMC (P0.05) exhibited an increase in fecal fat content. Immunoblot assay revealed that acyl‑coenzyme A:cholesterol acyltransferase-2 (ACAT-2) expression was suppressed in all chitosan-fed animals compared to the HFD-fed controls (P0.05). These results suggest that chitosan effectively decreases blood lipid content, and its effectiveness depends on the molecular size of chitosan. The hypolipidemic effect of chitosan is partly attributed to its suppression of intestinal lipid absorption and hepatic ACAT-2 expression.

The effects of inorganic selenium (Se) (sodium selenate, SSe) and organic selenium (seleno-l-methionine, MSe) supplementation on the immune response, antioxidant status, and disease resistance of the giant freshwater prawn, Macrobrachium rosenbergii, were studied. Five experimental diets, including a control diet (without Se enrichment), 0.5 mg (kg diet)(-1) of MSe, 1 mg (kg diet)(-1) of MSe, 0.5 mg (kg diet)(-1) of SSe, and 1 mg (kg diet)(-1) of SSe, were used. After 75 days of culture, prawn fed the Se-enriched diets had lower mortality compared to that of prawn fed the control diet after being challenged by the pathogen, Debaryomyces hansenii. No significant differences in the total hemocyte count, superoxide dismutase activity, or clearance efficiency of prawn were recorded among the control and treated groups. Significantly increased phenoloxidase and phagocytic activities in prawn fed the Se-enriched diets were found compared to the controls. Respiratory bursts of prawn fed both forms of 1 mg Se (kg diet)(-1) significantly increased compared to control prawns. For the antioxidant status analysis, glutathione peroxidase, glutathione reductase, and glutathione s-transferase of prawn fed the SSe-enriched diet at 1 mg (kg diet)(-1) were significantly increased. The results indicated that the cheaper selenium, SSe is recommended to be added in prawn feed at the concentration of 0.5 mg resulting in 1.5 mg SSe (kg diet)(-1) increased prawn immunity and disease resistance against the pathogen, D. hansenii.

Fucose-containing sulfated polysaccharides, also termed “fucoidans”, which are known to possess antioxidant, anticoagulant, anticancer, antiviral, and immunomodulating properties, are normally isolated from brown algae via various extraction techniques. In the present study, two methods (SC1 and SC2) for isolation of fucoidan from Sargassum cristaefolium were compared, with regard to the extraction yields, antioxidant activity, and inhibition of growth of human colon cancer cells exhibited by the respective extracts. SC1 and SC2 differ in the number of extraction steps and concentration of ethanol used, as well as the obtained sulfated polysaccharide extracts, namely, crude fucoidan preparation (CFP) and purified fucoidan preparation (PFP), respectively. Thin layer chromatography, Fourier transform infrared analysis, and measurements of fucose and sulfate contents revealed that the extracts were fucoidan. There was a higher extraction yield for CFP, which contained less fucose and sulfate but more uronic acid, and had weaker antioxidant activity and inhibition of growth in human colon cancer cells. In contrast, there was a lower extraction yield for PFP, which contained more fucose and sulfate but less uronic acid, and had stronger antioxidant activity and inhibition of growth in human colon cancer cells. Thus, since the difference in bioactive activities between CFP and PFP was not remarkable, the high extraction yield of SC1 might be favored as a method in industrial usage for extracting fucoidan.

Gynura bicolor (Roxb & Willd.) DC. is widely distributed in Asia and is very popular for vegetarian cuisine in Taiwan. This study used G. bicolor as the main ingredient to develop a new vegetable juice. Its chemical composition and antioxidant activity were analyzed, and consumer preferences of G. bicolor vegetable juice (GBVJ) evaluated. The results showed that the major compounds in GBVJ were rutin, β-carotene, anthocyanin, and chlorophyll. The antioxidant activity of GBVJ including 1,1-diphenyl-2-picrylhydrazyl free radical scavenging activity, 2,2ʹ-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid), and superoxide anion were 85.1%, 95.9%, and 80.1%, respectively. The reducing power was about 60% of that of vitamin C (1 g/L). The consumer preferences for each investigated item were all above 5 points (total score 7 points). These results suggest that GBVJ may have positive effects on the antioxidant activity, and the high consumer acceptance may be used as the preliminary information to support that G. bicolor would be commercially viable in the food industry.
