Search

The search for new antimicrobials is imperative due to the emergent resistance of new microorganism strains. In this context, revisiting known classes like 8-hydroxyquinolines could be an interesting strategy to discover new agents. The 8-hydroxyquinoline derivatives nitroxoline and clioquinol are used to treat microbial infections; however, these drugs are underused, being available in few countries or limited to topical use. After years of few advances, in the last two decades, the potent activity of clioquinol and nitroxoline against several targets and the privileged structure of 8-hydroxyquinoline nucleus have prompted an increased interest in the design of novel antimicrobial, anticancer, and anti-Alzheimer agents based on this class. Herein, we discuss the current development and antimicrobial structure-activity relationships of this class in the perspective of using the 8-hydroxyquinoline nucleus for the search for novel antimicrobial agents. Furthermore, the most investigated molecular targets concerning 8-hydroxyquinoline derivatives are explored in the final section.

The oral administration of clioquinol – a potent 8-hydroxyquinoline antimicrobial drug – was forbidden due to suspicion of it being the cause of SMON (subacute myelo-optic-neuropathy) in Japan. However, this adverse effect was only observed in Japan, despite the fact that the drug was used worldwide. In addition, studies have shown that vitamin B12 deficiency could be involved in this process. Thus, the interest in the modification of this compound has recently emerged as a strategy to find new drug candidates. In the present work, we have designed and synthesized a novel series of clioquinol derivatives by the modification of the 7-position of this compound. Twenty-one derivatives were prepared from commercially available clioquinol in two or three steps: 8-OH-protection of clioquinol followed by the palladium-catalyzed cross-coupling reaction to introduce the arylamine group at position 7 resulting in derivatives 4a–4j. Finally, deprotection of the hydroxyl group gave derivatives 5a–5i and 10a–10b. The compounds prepared were tested against Candida spp. and dermatophyte isolates by the broth microdilution method. We have found a clear structure–activity relationship. The presence of free hydroxyl in the 8-position and the introduction of the hydrophilic heterocyclic substituent at the 7-position are important for the antifungal activity of this class. Moreover, the introduction of dimethoxy groups at the pyrimidinyl ring attached to the 7-position seems to be promising against dermatophytes and Candida albicans. Among all derivatives, compound 5h presented interesting activity against all fungal species tested (with MIC values of 4 μg mL−1), along with low toxicity in normal cells. This derivative is non-irritant with the absence of topical toxicity. In addition, 5h does not seem to act as an ionophore in fungal cells, similarly to clioquinol. These compounds appear to have a scavenger effect, which suggests their selectivity for fungal cells. Taken together, these findings indicate the potential of 5h to be developed as an antifungal agent.

Bis zum Ende des 20. Jh. erfolgte die Versorgung von Kindern und Jugendlichen durch „Kinderarzte“, in „Kinderarztpraxen“ und „Kinderkliniken“. Mit der Anderung der Weiterbildungsordnung der Bundesarztekammer vollzog sich die Fortentwicklung des Fachgebietes zum Facharzt fur „Kinder- und Jugendmedizin“. Bei einer Analyse der Situation ist festzustellen, dass die Strukturen zwar neu benannt wurden, dass aber die inhaltliche Ausrichtung nicht immer dem Titel der Institution entspricht. Dies gilt in der gesamten Breite der jugendmedizinischen Versorgung. Der vorliegende Beitrag stellt die Kurzfassung einer Bestandsaufnahme, einschlieslich vorgeschlagener Losungsstrategien, zur Jugendmedizin in Deutschland dar, die die Kommission Jugendmedizin der Deutschen Akademie fur Kinder- und Jugendmedizin (DAKJ) erstellt hat. Im Folgenden findet sich die ungekurzte Einfuhrung; Teil 1 (Jugendmedizin in deutschen Kinderkliniken), Teil 2 (Jugendmedizin in der padiatrischen Praxis), Teil 3 (Jugendmedizin in sozialpadiatrischen Zentren) und Teil 4 (Jugendmedizin im Offentlichen Gesundheitsdienst) werden auf die wesentlichen Aspekte gekurzt, aber ohne inhaltliche Veranderung dargestellt.

Since nutritional requirements are increased at the end of gestation to meet the demands of the pregnant uterus, pregnant beef cows are susceptible to mobilization of body reserves (mainly fat and amino acids (AAs)) and to alter the metabolism of nutrients in the liver and muscle to support such demands. The objective of this study was to evaluate the effect of CP supplementation on maternal nutrient metabolism in the late gestation of beef cows grazing a low-quality pasture. Forty-three pregnant Nellore cows gestating male fetuses (average age = 6 years; average weight = 544 kg) at 193 ± 30 (mean ± SD) days (d) of gestation were divided into eight groups (experimental units, with four to five cows each). Treatments were (1) control (CON, n = 4): pasture-based (PB) diet without CP supplementation and (2) supplemented (SUP, n = 4): PB diet daily supplemented with 2 g/kg of BW of a 43.5% CP supplement. Liver and skeletal muscle biopsies were performed at 265 days of gestation and samples were collected for mRNA expression. On day 280 of gestation, blood samples were collected to assess plasma levels of AA. The CON-fed cows tended to have greater (P = 0.057) total circulating AA than SUP-fed cows. The circulating glycogenic AA was greater (P = 0.035) in CON than in SUP cows. CON cows was greater for histidine (P = 0.015), methionine (P = 0.007) and alanine (P = 0.036) than SUP cows. The CON- and SUP-fed showed no differences for gluconeogenesis, fatty acid transport and signaling axis markers in the liver. The mRNA expression of markers for skeletal muscle synthesis, p7056k (P = 0.060) and GSK3B (P = 0.096), tended to be greater in cows from CON than SUP group. No differences were found for mRNA expression of markers for skeletal muscle degradation. We conclude that CP supplementation to CP-restricted late-pregnant beef cows reduces the maternal tissue mobilization and changes the profile of plasma circulating AA and the mRNA expression of markers for the synthesis of skeletal muscle tissue.

Crotalaria lanceolata E. Mey. and Crotalaria pallida Aiton. are leguminous plants of family Fabaceae found in most of the Brazilian territory. They were initially used as green manure and due their easy spread they are currently considered weeds in crops. Soybean and corn contamination can occur through the mechanical harvesting of these grains along with seeds of the Crotalaria species, which end up in the formulation of feed for production animals. Crotalaria spp. genus has toxic pyrrolizidine alkaloids (PA). Most plant species belonging to this genus can cause acute or chronic liver injury. In a first stage, one-day old broilers were divided into three groups: Group A (C. pallida seeds), Group B (C. lanceolata seeds), and Group C (Control). Groups A and B were divided into five subgroups, each with eight broilers, which received the following doses of the respective seeds in feed as of the 7th day of age: daily doses of 0.4%, 0.8% and 2.5%, and single doses of 15% and 25%. Four broilers in each study group were euthanized at 28 days of age - completing 21 days of seed consumption, and the four remaining broilers were euthanized at 42 days of age - completing 35 days of seed consumption. In a second stage, experiments were conducted using seeds of both the aforementioned plants with 28-day old broilers. These were divided into three groups of four animals each: Group D (C. pallida seeds) and Group E (C. lanceolata seeds), which received the respective seeds at daily doses of 1% and 2% in feed for 20 days, and Group F (Control). These broilers were euthanized when they were 80 days old. C. lanceolata seeds showed higher toxicity to broilers than C. pallida seeds, both supplied as of the 7th day of life. Clinical signs included inappetence, ruffled feathers, and brown diarrhea. The following gross lesions were observed: subcutaneous edema, ascites, hydropericardium, yellowish liver with hypertrophy or atrophy and enhanced lobular pattern, and distended gallbladder. Histologic lesions present in all birds in varying degrees were characterized by tumefaction and vacuolar degeneration of hepatocytes. The following clinical conditions and gross lesions were observed in the broilers: hepatocyte megalocytosis and karyomegaly, slight biliary epithelial hyperplasia, eosinophilic spheroids, and nuclear invagination with loss of hepatocyte cord architecture. O gênero Crotalaria spp. possui alcaloides pirrolizidínicos de ação tóxica. A maioria das espécies desse gênero causam lesões hepáticas, com evolução aguda, ou, crônica. Frangos de corte de um dia de vida foram divididos em três grupos: Grupo A (sementes de C. pallida - doses diárias de 0,4%, 0,8%, 2,5% e doses únicas de 15% e 25%), Grupo B (sementes de C. lanceolata - doses diárias de 0,4%, 0,8%, 2,5% e doses únicas de 15% e 25%) e Grupo C (Controle). Os Grupos A e B foram divididos em cinco subgrupos, com oito frangos cada, que a partir do sétimo dia de vida, receberam as doses estabelecidas. Quatro frangos de cada grupo foram sacrificados aos 28 dias, e os quatro restantes aos 42 dias de vida. Também foram conduzidos experimentos com as sementes dessas duas plantas com aves de 28 dias, as quais foram divididas em três grupos: Grupo D (C. pallida - doses de 1% e 2% diariamente, durante vinte dias), Grupo E (C. lanceolata - doses de 1% e 2% diariamente, durante vinte dias) e Grupo F (Controle). Cada grupo composto por quatro aves. Estas aves foram sacrificadas ao completarem 80 dias de vida. Sementes de C. lanceolata demonstraram maior toxicidade para frangos de corte do que sementes de C. pallida. Os sinais clínicos foram inapetência, penas arrepiadas e diarreia acastanhada. Na macroscopia observou-se edema subcutâneo, ascite, hidropericárdio, fígado de coloração amarelada com hipertrofia e, ou, atrofia, evidenciação do padrão lobular e vesícula biliar distendida. As lesões histológicas presentes em todas as aves, em diferentes graus caracterizaram-se por tumefação e degeneração vacuolar de hepatócitos. Nas aves que manifestaram alterações clínicas e lesões macroscópicas, havia megalocitose, cariomegalia, hiperplasia do epitélio biliar, leve, esferoides eosinofílicos e invaginação nuclear com perda da arquitetura dos cordões de hepatócitos.

The aim was to determine effects of maternal feed restriction in dairy goats at gestational different stages on skeletal muscle development and energy metabolism in kids at birth. Six pregnant goats were fed 50% of total digestible nutrients (TDN) and crude protein (CP) (NRC, 2007) recommendations in the first half of gestation and then fed to 100% of the recommendations in the second half of gestation (treatment R-M). In the other group, eight pregnant goats were fed 100% of TDN and CP in the first half of gestation and 50% of a restricted diet the second half of gestation (treatment M-R). Birth weight, blood glucose concentration, muscle fiber number, and size of kids at birth were not affected by maternal feed restriction. The mRNA and protein abundance of myogenic, adipogenic and fibrogenic markers were not affected (P > 0.05) by maternal diet. With regard to values for variables in kid energy metabolism, mRNA abundance of the glycolic enzyme HKII was less (P = 0.03) in the M-R group. In conclusion, maternal feed restriction in the first or second half of gestation had no affect mRNA abundance on myogenic, adipogenic, and fibrogenic markers nor were there changes in skeletal muscle mesenchymal stem cell population of kids at the time of birth. There, however, may be detrimental effects on energy metabolism by reducing HKII gene expression in skeletal muscle of dairy goat kids at the time of birth.

No. of bitstreams: 0 Previous issue date: 2021-02-01 Context: It is hypothesised that protein supplementation in pregnant Nellore cows during the dry season would improve reproductive performance in the next breeding season, as well as growth, myogenesis and intramuscular lipogenesis of the progeny until weaning. Aims: To evaluate the effect of maternal nutrition on cow reproductive performance, as well as on the growth, myogenesis and lipogenesis of the progeny until weaning. Methods: A total of 92 pregnant cows were fed on pasture, and half of the cows were also fed a mineral-protein supplement (36% crude protein) from 124 ± 21 days of pregnancy to calving. Therefore, two treatments were tested: non-supplemented or supplemented cows. Progeny were weighed after birth, 130 days after birth and at weaning. Six newborn calves from each treatment were slaughtered to collect muscle and jejunum samples to analyse histology and gene expression. In addition, Longissimus thoracis muscle biopsies were collected at 11 days after birth and weaning for gene expression analyses. Key results: Supplemented cows had greater bodyweight (P = 0.03) and body condition score (P = 0.05) during gestation, and the pregnancy rate in the subsequent breeding season had a tendency (P = 0.10) to be greater. The progeny from supplemented cows had greater bodyweight at birth (P = 0.05). However, no differences (P > 0.05) were found in bodyweight at weaning or in the average daily gain during this period. Non-supplemented calves had greater SLC27A4 (P = 0.04) expression and a tendency for greater expression of SLC5A1 (P = 0.08) in the jejunum. Muscle gene expression data showed that progeny from supplemented cows had greater expression of myogenic (WNT10B), adipogenic (PPARG, ZFP423, CD36) and fibrogenic (TGFβ1) markers at birth and at weaning (P ≤ 0.10). Conclusions: In conclusion, protein supplementation of pregnant Nellore cows leads to positive effects for subsequent reproductive performance and for muscle development of their progeny. In addition, the progeny from feed-restricted cows increases prenatal intestinal development for better nutrients absorption under a potentially impaired environmental condition. Implications: The use of protein supplementation in pregnant Nellore cows has a positive impact on the production system, increasing productivity in a cow/calf operation. Background: In efforts to reduce misuse of opioids, new abuse deterrent formulations of medications have been developed. Insurers increasingly give abuse-deterrent opioid formulations preferred formulary status, which can result in required formulation rotation for patients. Xtampza® (oxycodone myristate extended-release) is an abuse-deterrent opioid formulation that maintains its extended-release properties with any physical manipulation. Blood levels of oxycodone myristate extended-release (OxyM-ER) may vary with dietary caloric and fat intake. Case Description: A woman with metastatic breast carcinoma had severe myalgias and arthralgias well-managed with oxycodone hydrochloride extended-release (OxyHCl-ER) and hydrocodone/acetaminophen. A switch from OxyHCl-ER to OxyM-ER resulted in worsened pain management, decreased functional status, and a referral to palliative care (PC). Recognizing calorie-depending pharmacokinetic variability with OxyM-ER, the interdisciplinary PC team obtained a detailed dietary history from the patient, which revealed a low-fat, low-calorie healthy diet with inconsistent meals. After repeated education, the patient changed her diet and had improved pain and functional status without increasing her total daily opioid dose. Conclusion: The efficacy of OxyM-ER may be compromised in patients with cancer experiencing anorexia, decreased or inconsistent food intake, or low-fat/low-calorie diets. An interdisciplinary team approach can improve pain control in the setting of “forced” formulary switches to OxyM-ER.

The current study aimed to identify differentially expressed genes and their biological process in the skeletal muscle of newborn goats as a function of maternal feed restriction during different stages of gestation. A total of 14 pregnant dams were randomly divided into one of the follow dietary treatments: Animals fed at 50% of maintenance requirement from 8-84 d of gestation and then fed at 100% of maintenance requirement from day 85 of gestation to parturition (RM, n = 6), and animals fed at 100% of maintenance requirement from 8-84 d of gestation and then fed at 50% of maintenance requirement from day 85 of gestation to parturition (MR, n = 8). At birth, all male offspring were euthanized and a sample of Logissimus muscle was collected for total RNA extraction and sequencing. Cuffdiff tool (Cufflinks 2.2.1) was used to count reads, normalize transcript expression (FPKM), and identify differentially expressed genes (DEG) between treatments. Network analysis was performed with String 11.0 using the available genome from the closest specie (Ovis aries). A total of 66 DEG (q-value < 0.05) were identified, with 6 up-regulated genes in skeletal muscle of RM compared to MR newborn goats. These genes are related to tissue development, such as CYTL1, UGT8, and NPNT. The 60 down-regulated genes in skeletal muscle of RM compared with MR newborn goats are related to transcription factors (TF) complex. Among the TF, FOS and JUNB families forms heterodimers in response to several stimuli (cytokines,growth factors, stress), and play key roles controlling a number of cellular processes, including signal transduction, cell proliferation, differentiation and apoptosis. In conclusion, maternal feed restriction at different stages of gestation affects tissue development and changes the transcription pattern in the skeletal muscle of newborn goats, which may lead to negative consequences in animal growth and performance.

As theories about the causes of racial inequities in infant mortality evolve, they are becoming increasingly complex. Interventions to address these inequities must be similarly complex, incorporating both upstream and downstream approaches. The Best Babies Zone Initiative (BBZ) has been in operation since 2012 with an aim of reducing racial inequities in infant mortality. BBZ is designed to be flexible and responsive to the conditions creating toxic stress in communities of color. After seven years of operation in nine sites across the United States, and interventions implemented in housing, economic, and environmental justice, the Initiative has identified strategies to support the development and advancement of aligned programs. Lessons learned and opportunities were identified across the Initiative’s theoretical foundation (the life course perspective) and each of the four foundational strategies: place-based, community-driven, multi-sector work that contributes to broader social movements. Overarching lessons learned about advancing equity in MCH were identified including: the need to focus explicitly on racial equity, the imperative of shifting the time horizon for change, and the importance of identifying sustainable funding mechanisms. A complex initiative such as BBZ is relatively nascent in the field of Maternal and Child Health. However it represents an innovative approach to building community power and fostering strategic organizational partnerships in service of addressing root causes of racial inequities in birth outcomes. The lessons learned and opportunities identified by BBZ can serve as a foundation from which to build other programs and initiatives to advance racial justice.

Therefore, the objective was to analyze the expression of PPARA, SREBF1 and genes involved in lipid metabolism in longissimus thoracis (LT) muscle of cattle fed diets containing soybean or cottonseed with or without vitamin E. Twenty-eight bulls were used in a completely randomized design using a 2 × 2 factorial arrangement. The inclusion of soybean and cottonseed in the diets provided 64.8 g kg−1 and 65.6 g kg−1 of ether extract, respectively, and vitamin E supplementation was 2500 IU per day. Background A young adolescent female, aged 17 years and one month, presented seeking orthodontic treatment to address the congenital absence of her maxillary lateral incisors. Aims The therapeutic aims were to provide an adequate aesthetic and functional occlusion, coupled with sound adjunctive periodontal tissue relationships and a favourable long-term prognosis. Methods The malocclusion was managed by customised lingual orthodontic appliances and the adjunctive use of direct skeletal anchorage derived from two palatal mini-implants. Results The treatment objectives of good aesthetics, a functional occlusion, normal function, a healthy periodontium and a balanced profile were readily achieved. The retention records demonstrate the stability of the correction. The treatment duration was approximately 41 months, which encompassed the use of full fixed customised lingual appliances over a period of 24 months.

An increasing body of evidences has demonstrated the ability of the mature adipocyte to dedifferentiate into a population of proliferative-competent cells known as dedifferentiated fat (DFAT) cells. As early as the 1970s, in vitro studies showed that DFAT cells may be obtained by ceiling culture, which takes advantage of the buoyancy property of lipid-filled cells. It was documented that DFAT cells may acquire a phenotype similar to mesenchymal stem cells and yet may differentiate into multiple cell lineages, such as skeletal and smooth muscle cells, cardiomyocytes, osteoblasts, and adipocytes. Additionally, recent studies showed the ability of isolated mature adipocytes to dedifferentiate in vivo and the capacity of the progeny cells to redifferentiate into mature adipocytes, contributing to the increase of body fatness. These findings shed light on the potential for use of DFAT cells, not only for clinical purposes but also within the animal science field, because increasing intramuscular fat without excessive increase in other fat depots is a challenge in livestock production. Knowledge of the mechanisms underlying the dedifferentiation and redifferentiation of DFAT cells will allow the development of strategies for their use for clinical and animal science purposes. In this review, we highlight several aspects of DFAT cells, their potential for clinical purposes, and their contribution to adipose tissue mass in livestock.

We assessed the effect of maternal feed restriction at different stages of gestation on growth and development of organs and small intestine of newborn goats. A total of 14 pregnant dams were randomly divided into one of the follow dietary treatments: Animals fed at 50% of maintenance requirement from 8-84 d of gestation and then fed at 100% maintenance requirement from day 85 of gestation to parturition (R-M, n = 6), and animals fed at 100% of maintenance requirement from 8-84 d of gestation and then fed at 50% maintenance requirement from day 85 of gestation to parturition (M-R, n = 8). At birth, newborns were slaughtered for collection of corporal components. No differences were observed among treatments for birth weight (P = 0.46). The weight of the complex reticulum-rumen-omasum tended to be heavier (P = 0.057) in the M-R group, when expressed per kg of body weight. The small intestine (P = 0.038) and total intestine (P = 0.038) were heavier in the offspring of the M-R group when expressed in kg of body weight. The M-R newborns had greater length of the small intestine (P = 0.043) and total intestine (P = 0.026). Maternal feed restriction did not influence the intestinal villi height (P = 0.406). However, newborn from R-M group had lower villus height:crypt depth ratio (P = 0.016), due to the tendency of the greater of crypt depth in these animals (P = 0.081). No differences among treatments were observed in mRNA expression of the MGAM and GLP-2R in the jejunum of the offspring (P > 0.12). Newborns of the M-R group tended to present greater mRNA abundance of the SLC5A1 (P = 0.091), SLC2A2 (P = 0.091), and OCLN (P = 0.061). In summary, restriction in the first half of gestation may be more detrimental to the performance and health of offspring throughout life due to increased impairment of intestinal development.

Background Diffusion kurtosis imaging has been applied to evaluate white matter and basal ganglia microstructure in mixed Parkinson's disease (PD) groups with inconclusive results. Objectives To evaluate specific patterns of kurtosis changes in PD and to assess the utility of diffusion imaging in differentiating between healthy subjects and cognitively normal PD, and between PD with and without mild cognitive impairment. Methods Diffusion scans were obtained in 92 participants using 3T MRI. Differences in white matter were tested by tract-based spatial statistics. Gray matter was evaluated in basal ganglia, thalamus, hippocampus, and motor and premotor cortices. Brain atrophy was also assessed. Multivariate logistic regression was used to identify a combination of diffusion parameters with the highest discrimination power between groups. Results Diffusion kurtosis metrics showed a significant increase in substantia nigra (p = 0.037, Hedges' g = 0.89), premotor (p = 0.009, Hedges' g = 0.85) and motor (p = 0.033, Hedges' g = 0.87) cortices in PD with normal cognition compared to healthy participants. Combined diffusion markers in gray matter reached 81% accuracy in differentiating between both groups. Significant white matter microstructural changes, and kurtosis decreases in the cortex were present in cognitively impaired versus cognitively normal PD. Diffusion parameters from white and gray matter differentiated between both PD phenotypes with 78% accuracy. Conclusions Increased kurtosis in gray matter structures in cognitively normal PD reflects increased hindrance to water diffusion caused probably by alpha-synuclein-related microstructural changes. In cognitively impaired PD, the changes are mostly driven by decreased white matter integrity. Our results support the utility of diffusion kurtosis imaging for PD diagnostics.

Background Feed efficiency is one of the most important parameters that affect beef production costs. The energy metabolism of skeletal muscle greatly contributes to variations in feed efficiency. However, information regarding differences in proteins involved in the energy metabolism of the skeletal muscle in beef cattle divergently identified for feed efficiency is scarce. In this study, we aimed to investigate energy metabolism of skeletal muscle of Nellore beef cattle, identified for low and high residual feed intake using a proteomics approach. We further assessed the expression of candidate microRNAs as a one of the possible mechanisms controlling the biosynthesis of the proteins involved in energy metabolism that were differentially abundant between high and low residual feed intake animals. Results A greater abundance of 14–3-3 protein epsilon (P = 0.01) was observed in skeletal muscle of residual feed intake (RFI) high animals (RFI-High). The present study aimed to investigate metabolic and physiological changes induced by pregnancy, as well as the expression of key genes linked to the absorptive and proliferative activity of the ruminal epithelium towards the end of the gestational period in zebu beef heifers. Twelve ruminally cannulated Zebu beef heifers were randomly assigned into two experimental treatments: a pregnant group (n = 7) and a control group (non-pregnant; n = 5). All heifers received the same diet throughout the experiment. Respiratory and heart rates and plasma glucose levels were assessed just before and four hours after morning feeding at 110, 171, 206, 242, 266, and 286 days of pregnancy (DOP). Blood samples were collected for serum non-esterified fatty acids (NEFA), beta-hydroxybutyrate (BHB), and urea analysis prior to the morning feedings at 110, 206, 266, and 286 DOP. At 215 and 272 DOP the ruminal epithelium was biopsied to evaluate the mRNA expression of key genes involved in remodeling, inflammation, and transport. The respiratory rate was similar (P ≥ 0.79) between groups over days. However, the heart rate increased (P = 0.01) in late-pregnant heifers compared to controls. Blood concentrations of NEFA (P = 0.11) and BHB (P = 0.67) did not vary over the gestation period in pregnant heifers. Glucose concentrations, before morning feeding, were similar during all collection periods with an exception at 286 days (DOP × physiological status interaction; P = 0.05) when glucose was lower in pregnant (83 mg/dL) compared to non-pregnant (107 mg/dL) heifers. The mRNA expression of genes related to cellular remodeling (PCNA and CASP3), inflammation (KLK9 and KLK10), and transport of volatile fatty acids (SLC16A1 and SLC16A3), H+ (SLC9A1), HCO3− (SLC26A3 and SLC26A6), and glucose (SLC5A1) in the ruminal epithelium were downregulated at late gestation. These results suggest that the ruminal epithelium saves energy at late pregnancy to benefit fetal development. In addition, the increase in heart rate coupled with tissue mobilization can be considered homeorhetic mechanisms that help meet fetal nutrient requirements.

We investigated how feed restriction at 50% of maintenance requirements during different stages of gestation affects the transcriptome of newborn goats' skeletal muscle. Fourteen pregnant dams were randomly assigned into one of the following dietary treatments: animals fed at 50% of maintenance requirement from 8-84 d of gestation and then fed at 100% of maintenance requirement from day 85 of gestation to parturition (RM, n = 6), and animals fed at 100% of maintenance requirement from 8-84 d of gestation and then fed at 50% of maintenance requirement from day 85 of gestation to parturition (MR, n = 8). At birth, samples of offspring's Longissimus muscle were collected for total RNA extraction and sequencing. Our data showed 66 differentially expressed (DE) genes (FDR < 0.05). A total of 6 genes were upregulated and 60 downregulated (FDR < 0.05) in the skeletal muscle of the newborns resulting from treatment RM compared with MR. Our results suggest that the DE genes upregulated in newborn goats' skeletal muscle from the RM group compared to MR, included genes related to satellite cells, and genes that indicates impaired insulin sensitivity and changes in the composition of intramuscular fat. The DE genes upregulated in newborn goats' skeletal muscle from the MR group compared to RM, are also related to impaired insulin sensitivity, as well as a predominantly oxidative metabolism and cellular oxidative stress. However, protective mechanisms against insulin sensitivity and oxidative stress may have been augmented in the skeletal muscle of offspring from MR treatment compared to RM, in order to maintain cellular homeostasis.

We aimed to investigate the effects of maternal protein restriction during mid-gestation on the skeletal muscle composition of the offspring. In the restriction treatment (RES, n = 9), cows were fed a basal diet, while in the control (CON, n = 9) group cows received the same RES diet plus the protein supplement during mid-gestation (100–200d). Samples of Longissimus dorsi muscle were collected from the offspring at 30d and 450d postnatal. Muscle fiber number was found to be decreased as a result of maternal protein restriction and persisted throughout the offspring’s life (p &lt, 0.01). The collagen content was enhanced (p &lt, 0.05) due to maternal protein restriction at 30d. MHC2X mRNA expression tended to be higher (p = 0.08) in RES 30d offspring, however, no difference (p &gt, 0.05) was found among treatments at 450d. Taken together, our results suggest that maternal protein restriction during mid-gestation has major and persistent effects by reducing muscle fiber formation and may slightly increase collagen accumulation in the skeletal muscle of the offspring. Although maternal protein restriction may alter the muscle fiber metabolism by favoring the establishment of a predominant glycolytic metabolism, the postnatal environment may be a determinant factor that establishes the different proportion of muscle fiber types.

Objective To determine how an adolescent's risk of cesarean varies by maternal age and race/ethnicity, and evaluate the contribution of obstetric and sociodemographic factors to mode of delivery. Study Design This is a retrospective cohort study of 604,287 births to women aged 13 to 23 years. Regression techniques were used to determine maternal ages at lowest risk of primary cesarean in each major racial/ethnic group before and after adjustment for various cesarean risk factors. Results Adolescent age was associated with lower risk of cesarean compared with young adults (17.2% at age 13 years vs 24.8% at age 23 years, p Conclusion Young maternal age is protective against cesarean delivery in all racial/ethnic groups. Adolescents also experience racial/ethnic disparities in mode of delivery similar to those observed in adults, which were unexplained by either obstetric or sociodemographic factors.

Szasz famously declared mental illness a “myth” and a “metaphor,” arguing that psychiatry’s diagnostic categories are only temporary stops on the road to “real” and “legitimate” bodily diseases. He argued that conditions once regarded as “mental illnesses” would rightly be reclassified as “brain diseases,” insofar as scientific investigations would uncover their neuropathology. Based on a critique of six foundational claims in Szasz’s writings, the author of this chapter argues that psychopathology and neuropathology are complementary rather than contradictory or disjunctive. Just as some mental illnesses may be considered brain diseases, some brain diseases may manifest as “mental illness.” The locution, “mental illness,” remains useful, albeit imperfect, shorthand to describe a particular kind of suffering and incapacity, usually affecting cognition, emotion, reasoning, or behavior. Even if all mental illnesses were conclusively and causally linked to specific brain abnormalities, we would still need “mental language” in both ordinary discourse and the vocabulary of clinical work.

SUMMARY The objective of this study was to evaluate the effect of probiotic consortium on feed intake, feeding behavior, feed efficiency, fecal score, weight gain and carcass traits in finishing Nellore heifers fed high energy diets in feedlot. Twenty-four Nellore heifers were distributed in 12 pens with two animals each, being six pens per treatment. In the control treatment (CON) the animals were fed exclusively with the base diet, which contained 35% corn silage and 65% concentrate. In the treatment with probiotic consortium technology (TCP), the animals received the base diet and a single dose of 75 mL/animal/day of Bio Ciclo Completo (Global Saúde Brasil) top-dressed in the morning feeding. There was no period of adaptation to the diet. The experiment lasted 93 days. The use of TCP did not affect (P = 0.980) the dry matter intake. The average daily gain was 12.5% greater (P = 0.025) in the animals treated with TCP (0.941 kg/day for TCP versus 0.834 kg/day for CON). The animals that received TCP improved (P = 0.021) the feed efficiency by 12.6%. The animals that received TCP tended to have a better (P = 0.094) fecal score within the first 27 days of feedlot. There were no significant effects of TCP on the feeding behavior and carcass traits. Based on the results, there is a great potential of using TCP to improve productivity in beef cattle.

Current study evaluated the inclusion of protected fat and different levels of crude protein (CP) in the diet of crossbreed cows in early lactation. Twelve cows were distributed in a 4 x 4 factorial design, with four treatments (12 or 16% CP, with or without MEGALAC-E(r)). Statistical analyzes with ANOVA and Pearson's linear correlation procedures were performed. Protected fat did not affect intake of dry matter (DM) and organic matter (OM) (p > 0.05). Increased CP raised consumption of CP and EE and decreased the non-fiber carbohydrates (NFC; p 0.05) on milk yield and composition due to protected fat or CP rates. Since the inclusion of protected fat and higher content of CP (16%) in the diet did not benefit the production and composition of milk in crossbreed cows, diets with 12% CP without the inclusion of protected fat are recommended. RESUMO. O objetivo desse estudo foi avaliar a inclusão de gordura protegida e de diferentes níveis de PB, na dieta para vacas mestiças, no início da lactação. Foram utilizadas 12 vacas, distribuídas em quadrados latinos 4 x 4 e quatro tratamentos (12 ou 16% de PB, com ou sem MEGALAC-E(r)). Análises estatísticas foram realizadas utilizando Anova e correlações lineares de Pearson. A gordura protegida não influenciou o consumo de MS e MO (p > 0,05). O aumento de PB elevou o consumo de PB e EE e diminuiu o de carboidratos não fibrosos (CNF; P 0,05) na produção e composição do leite em função da gordura protegida ou do teor de PB. A inclusão de gordura protegida e o maior teor de PB (16%) na dieta não resultaram em benefícios na produção e composição do leite de vacas mestiças, portanto recomenda-se utilizar dietas contendo 12% de PB, sem a inclusão de gordura protegida.

The history of psychiatry is characterized by some deep ideological and conceptual divisions, as adumbrated in Professor Hannah Decker's essay. However, the schism between "biological" and "psychosocial" models of mental illness and its treatment represents extreme positions among some psychiatrists-not the model propounded by academic psychiatry or its affiliated professional organizations. Indeed, the "biopsycho-social model" (BPSM) developed by Dr. George L. Engel has been, and remains, the foundational model for academic psychiatry, notwithstanding malign market forces that have undermined the BPSM's use in clinical practice. The BPSM is integrally related to "centralizing" and integrative trends in American psychiatry that may be traced to Franz Alexander, Karl Jaspers, and Engel himself, among others. This "Alexandrian-Jaspersian-Engelian" tradition is explored in relation to Professor Decker's "cyclical swing" model of psychiatry's history.

The proto-oncogene recepteur d'origine nantais (RON, MST1R) and its alternatively spliced variants are involved in various tumor biological processes, such as cell motility, adhesion, proliferation, apoptosis and epithelial-to-mesenchymal transition (EMT). RON overexpression and the occurrence of specific alternatively spliced RON isoforms have been detected in ovarian cancer. In the present study, we evaluated the role and regulation of cancer-related RON splicing isoforms in primary ovarian cancer. Expression of RON variants (RONΔ165, RONΔ160) was determined in 45 primary ovarian cancer and 4 physiological ovarian tissue specimens by RT-PCR and western blot analysis. The results were correlated to clinicopathological parameters. Additionally, expression of splicing factors with known involvement in RON alternative splicing regulation was examined. Increased RON levels were detected in all tumor samples (p=0.001) without differences between the primary tumors and metastases. Alternative RON variants were present in the majority of tumor samples (39 of 45; 86.67%). Potential RONΔ165 occurred more often (82.22%) than potential RONΔ160 or RONΔ155 (24.40%). Several significant correlations of RON and splicing factor expression [e.g. ASF/SFRS1 (p=0.035)] were detected. Correlations of RON expression to clinicopathological parameters were not observed. The term "mental illness" has been criticized on a variety of grounds, most notably by those who have argued that the term is merely a "myth" or a "metaphor." Some have argued that if and when so-called mental illnesses are exhaustively explained by disturbed brain function or structure, we will no longer need the term "mental illness," on the supposition that neuropathology and psychopathology are mutually exclusive constructs. The author argues that, on the contrary, the locution "mental illness" is not rendered useless or unnecessary when neuropathology is discovered, nor is the term "mental illness" a metaphor. Rather, it is an instance of "ordinary language" that we apply quite literally to certain types of suffering and incapacity in the realm of thought, emotion, cognition, and behavior. Although its use carries the risk of perpetuating mind-body dualism and it may be misused as a pejorative label, "mental illness" is likely to remain a useful and meaningful descriptive term, even as we discover the neurobiological underpinnings of psychiatric illness.

To determine the effects of maternal nutrition on modifications of foetal development of the skeletal muscle and possible increase in the potential of skeletal muscle growth in cattle, gestating cows were either fed 190% NRC recommendations (overnourished; ON) or 100% NRC recommendation (control; CO). Interaction between maternal nutrition (MN) and the foetal sex (FS) was also investigated. Foetuses were necropsied at four different time points throughout gestation (139, 199, 241 and 268 days of gestation) to assess the mRNA expression of myogenic, adipogenic and fibrogenic markers in skeletal muscle. Phenotypic indicators of the development of skeletal muscle fibres, intramuscular lipogenesis and collagen development were also evaluated. Modifications in mRNA expression of skeletal muscle of foetuses were observed in function of MN and FS despite the lack of effect of MN and FS on foetal weight at necropsy. Maternal ON increased the mRNA expression of the myogenic marker Cadherin-associated protein, beta 1 (CTNNB1) and adipogenic markers Peroxissome proliferator-activated receptor gamma (PPARG) and Zinc finger protein 423 (ZNF423) at midgestation. However, no differences on foetal skeletal muscle development were observed between treatments at late gestation indicating that a compensatory development may have occurred on CO foetuses making the effect of MN on skeletal muscle development not significant at late gestation. Moreover, our data have shown an evidence of sexual dimorphism during foetal stage with a greater skeletal muscle development in male than in female foetuses. In conclusion, providing a higher nutritional level to pregnant cows changes the trajectory of the development of skeletal muscle during midgestation, but apparently does not change the potential of post-natal growth of muscle mass of the offspring, as no differences in skeletal muscle development were observed in late gestation.

The appropriate supply of nutrients in pregnant cows has been associated with the optimal development of foetal tissues, performance of their progeny and their meat quality. The aim of this study was to evaluate supplementation effects of grazing cows in different stages of gestation on skeletal muscle development and performance of the progeny. Thereby, 27 Nellore cows were divided into three groups (n=9 for each group) and their progeny as follows: UNS, unsupplemented during gestation; MID, supplemented from 30 to 180 days of gestation; LATE, supplemented from 181 to 281 days of gestation. The percentage composition of the supplement provided for the matrices was the following: ground corn (26.25%), wheat bran (26.25%) and soya bean meal (47.5%). The supplement was formulated to contain 30% CP. Supplemented matrices received 150 kg of supplement (1 and 1.5 kg/day for cows in the MID and LATE groups, respectively). After birth, a biopsy was performed to obtain samples of skeletal muscle tissue from calves to determine number and size of muscle fibres and for messenger RNA (mRNA) expression analysis. The percentage composition of the supplement provided for the progeny was the following: ground corn grain (30%), wheat bran (30%), soya bean meal (35%) and molasses (5%). The supplement was formulated to contain 25% CP and offered in an amount of 6 g/kg BW. Performance of the progeny was monitored throughout the suckling period. Means were submitted to ANOVA and regression, and UNS, MID and LATE periods of supplementation were compared. Differences were considered at P0.10). Similarly, no differences were observed between calves for nutrient intake (P>0.10). However, greater subcutaneous fat thickness (P=0.006) was observed in the calves of LATE group. The ribeye area (P=0.077) was greater in calves born from supplemented compared with UNS cows. The supplementation of pregnant cows did not affect the muscle fibre size of their progeny (P=0.208). On the other hand, calves born from dams supplemented at mid-gestation had greater muscle fibre number (P=0.093) compared with calves from UNS group. Greater mRNA expression of peroxysome proliferator-activated receptor α (P=0.073) and fibroblast growth factor 2 (P=0.003) was observed in the calves born from MID cows. Although strategic supplementation did not affect the BW of offspring, it did cause changes in carcass traits, number of myofibres, and mRNA expression of a muscle hypertrophy and lipid oxidation markers in skeletal muscle of the offspring.

Effects of replacement of soybean meal by wet or dry castor bean meal (CM) in diets of feedlot lambs on animal performance, carcass traits, intake, digestibility and hepatic function were assessed. Thirty-five mixed-breed woolless lambs averaging 20±0.26 kg body weight were randomly assigned into five treatments in a 2×2+1 factorial design with 7 animals per treatment. Control animals were fed a diet containing soybean meal while lambs from the remaining groups received diets with 50 or 100% level by dry (DCM) or wet (WCM) calcium hydroxide treated CM as a replacement of soybean meal. Lambs were fed for 70 days and slaughtered at the end of the trial for further carcass traits evaluation. Dry matter intake and digestibility, final body weight, average daily gain and carcass weights were not affected ( P >0.05) by the substitution of soybean meal by either CM source. Serum levels of aspartate aminotransferase and alanine aminotransferase enzymes did not differ among treatments ( P >0.05) indicating no effect of treated CM on hepatic function. These results suggest that castor bean meal treated with calcium hydroxide solution (60 g/kg) can totally replace the soybean meal (up to 18% of dry matter of diet) in the diet of finishing lambs without negative effects on weight gain, intake, digestibility and hepatic function. Castor bean meal treated with calcium hydroxide solution can be fed to animals after 18 h in room temperature, in wet form, without sun or oven-drying.

Twenty-four pregnant Nellore cows were randomly assigned into 2 feeding level groups (control [CTL]; fed 1.0 times the maintenance requirement; n = 12; and overnourished [ON]; fed at 1.5 times the maintenance requirement; n = 12) to evaluate effects of maternal overnutrition on fetal skeletal muscle development. Cows were slaughtered at 135, 190, and 240 d of gestation and samples of fetal LM were collected for analysis of mRNA expression analysis and for histological evaluation of collagen content and number of muscle cells. There was no interaction between gestational period and maternal nutrition for the variables evaluated (P > 0.05). The mRNA expression of Cadherin-associated protein, β 1 (β-catenin) tended to be greater in fetuses from ON cows (P = 0.08), while myogenic differentiation 1 (MyoD; P = 0.56), myogenin (MyoG; P = 0.70), and the number of muscle cells (P = 0.90) were not affected by maternal overnutrition. Gestational period did not affect the mRNA expression of β-catenin (P = 0.60) and MyoG (P = 0.21). The mRNA expression of MyoD tended to increase with days of gestation (P = 0.06). The mRNA expression of zinc finger protein 423 (Zfp423; P < 0.0001), C/EBPα (P = 0.01), and PPARγ (P < 0.0001) were enhanced in ON fetuses. No effects of days of gestation were observed for mRNA expression of Zfp423 (P = 0.75) and C/EBPα (P = 0.48). The mRNA expression of PPARγ in fetuses at 190 d of gestation tended to be greater than those at 135 and 240 d of gestation (P = 0.06). The mRNA expression of transforming growth factor β (TGF-β; P < 0.0001), collagen type III, α I (COL3A1; P < 0.0001), and collagen content (P = 0.01) were increased in ON fetuses. Gestational period did not affect the mRNA expression of collagen type I, α I (COL1A1; P = 0.65). The mRNA expression of COL3A1 (P = 0.09) in fetuses at 190 d of gestation tended to be greater than fetuses at 135 and 240 d of gestation. The mRNA expression of TGF-β in fetuses at 190 d of gestation was greater than in fetuses at 135 d of gestation (P = 0.03), and the values observed in fetuses at 240 d of gestation did not differ from the other gestational time points. The least value of collagen content (P = 0.01) was observed in fetuses at 135 d of gestation, and no differences were observed among the other gestational time points. These data shows that maternal overnutrition enhances fibrogenesis and likely adipogenesis without compromising myogenesis in fetal skeletal muscle of cattle.

In order to analyze the influence of vitamin E on the immune response of birds vaccinated and challenged with Infectious Bronchitis Virus in poultry (IBV), and which dose of vitamin E is the best to answer this, an experiment was conducted using 50 SPF birds housed with one day of life at Embrapa – Suínos e Aves. All birds were divided into 10 groups of five birds each, including positive and negative control groups, supplemented with 15, 50, and 200 IU of vitamin E/Kg added into the feed. On the 14th day of life the birds were vaccinated with a commercial vaccine for infectious bronchitis (H-120) and after 28 days, one group for each treatment was challenged with IBV, strain classical (M-41). Five days after challenge birds were necropsied and abdominal macrophages were collected for analysis of microbicidal activity and measurement of nitric oxide (NO). The lung was collected for cell count by flow cytometry and also for NO dosage. Organs of predilection were also collected for histological analysis and virus isolation to analyze the efficiency of the vaccine. The results were evaluated by ANOVA and Student t-test. An increased of microbicidal activity of abdominal macrophages was observed in the groups of birds with higher levels of vitamin E (200 IU/Kg) supplementation when compared with groups that did not receive or received minor amounts (0, 15, 50 IU/Kg), indicating an improvement in the innate immune response influenced by vitamin E supplementation in the diet. The supplementation of this vitamin in high doses also increases the ability of macrophages to produce NO. And, by flow cytometry, suggests that the macrophages are the primary cells recruited in the lung tissue to combat infectious bronchitis and this action was potentiated by the addition of vitamin E, independent of the dosage.

Isolates of Salmonella sp (104) from poultry samples were isolated and serotyped where eleven were identified as Salmonella Typhimurium (ST). ST isolates were phenotypically tested by disk diffusion and minimum inhibitory concentration (MIC). Four genes related to enrofloxacin (GyrA, GyrB, ParC and ParE), two to gentamicin (AadA and AadB) and two to ceftiofur (BlaCMY-2 and AmpC) resistance were searched by PCR. Our results showed ST resistance to all three antibiotics tested (18.1% for ceftiofur, 45.4% for gentamicin, and 18.1% for enrofloxacin) according to the diffusion test. In the MIC test, the ST isolates showed higher levels of resistance (27.2% for ceftiofur, 54.5% for gentamicin, and 18.2% for enrofloxacin). Three resistance genes out of four searched genes for enrofloxacin were found in the ST isolates. Regarding gentamicin and ceftiofur, resistance genes were found mainly in samples with resistant phenotypic profile. Interestingly, some phenotypically-resistant strains did not present the resistance gene, which suggests an alternative route of resistance. Also, sensitive strains had the presence of the gene. It is possible to conclude that the ST isolates evaluated in this study have a multidrug resistance profile to the antibiotics routinely used in poultry production, and potential of greater levels of resistance in the near future.

In recent years, maternal and child health professionals have been seeking approaches to integrating the Life Course Perspective and social determinants of health into their work. In this article, we describe how community input, staff feedback, and evidence from the field that the connection between wealth and health should be addressed compelled the Contra Costa Family, Maternal and Child Health (FMCH) Programs Life Course Initiative to launch Building Economic Security Today (BEST). BEST utilizes innovative strategies to reduce inequities in health outcomes for low-income Contra Costa families by improving their financial security and stability. FMCH Programs' Women, Infants, and Children Program (WIC) conducted BEST financial education classes, and its Medically Vulnerable Infant Program (MVIP) instituted BEST financial assessments during public health nurse home visits. Educational and referral resources were also developed and distributed to all clients. The classes at WIC increased clients' awareness of financial issues and confidence that they could improve their financial situations. WIC clients and staff also gained knowledge about financial resources in the community. MVIP's financial assessments offered clients a new and needed perspective on their financial situations, as well as support around the financial and psychological stresses of caring for a child with special health care needs. BEST offered FMCH Programs staff opportunities to engage in non-traditional, cross-sector partnerships, and gain new knowledge and skills to address a pressing social determinant of health. We learned the value of flexible timelines, maintaining a long view for creating change, and challenging the traditional paradigm of maternal and child health.

This study was developed aiming to evaluate the effects of maternal feed-restriction ond evelopment of gastrointestinal tract (GIT) of bovine fetus at different gestational stages. Feed-restricted cows were fed 1.2 times the maintenance level while the control group was fed ad libitum. Pregnant cows were slaughtered at 136, 189, 239, and 269 days of gestation and gastrointestinal tracts of the fetuses were evaluated. No effects of maternal nutrition on body weight (P 1⁄4 0.17) and body length (P 1⁄40.13) of the fetuses were observed. No major effects of feed restriction on GIT mass of the fetuses were observed (P 1⁄4 0.51). However, the weight of small intestine per unit of body weight was 11.24% greater (P 1⁄40.04) in fetuses from restricted dams. Additionally, the length of small intestine and its villi were 12.93% and 16.44% respectively greater (P o .001) in fetuses from restricted dams compared to those from non-restricted dams. These data indicates that maternal feed-restriction does not affect the development of most of fetal gastrointestinal parts besides small intestine which in turn increases its surface area as a response of maternal feed restriction.

IntroductionTo date, the proposition of recurrence as a subclinical bipolar disorder feature has not received adequate testing.Objectives/AimsWe used the Italian version of the bipolar spectrum diagnostic scale (BSDS), a self-rated questionnaire of bipolar risk, in a sample of patients with mood disorders to test its specificity and sensitivity in identifying cases and discriminating between high risk for bipolar disorder major depressive patients (HRU) and low risk (LRU) adopting as a high recurrence cut-off five or more lifetime major depressive episodes.MethodsWe included 115 patients with DSM-5 bipolar disorder (69 type I, 41 type II, and 5 NOS) and 58 with major depressive disorder (29 HRU and 29 LRU, based on the recurrence criterion). Patients filled-out the Italian version of the BSDS, which is currently undergoing a validation process.ResultsThe BSDS, adopting a threshold of 14, had 84% sensitivity and 76% specificity. HRU, as predicted, scored on the BSDS intermediate between LRU and bipolar disorder. Clinical characteristics of HRU were more similar to bipolar disorder than to LRU; HRU, like bipolar disorder patients, had more lifetime hospitalizations, higher suicidal ideation and attempt numbers, and higher rates of family history of suicide.ConclusionsThe BSDS showed satisfactory sensitivity and sensitivity. Splitting the unipolar sample into HRU and LRU, on the basis of the at least 5 lifetime major depressive episodes criterion, yielded distinct unipolar subpopulations that differ on outcome measures and BSDS scores.Disclosure of interestThe authors have not supplied their declaration of competing interest.

RESUMO: Descrevem-se o quadro clínico, patológico e o potencial tóxico das folhas verdes de Crotalaria pallida em ovinos. Para o experimento folhas verdes de C. pallida foram coletadas no município de São João do Sul em Santa Catarina e administradas por via oral para seis ovinos adultos, em doses únicas de 40, 20, 10, 5, 2,5 e 2,5g/kg, e para dois ovinos, em doses diárias de 0,63 e 1,25g/kg durante 30 dias. Reproduziu-se a intoxicação com doses letais a partir de 2,5g/kg. Um dos ovinos que recebeu a dose única de 2,5g/kg e os que receberam as doses diárias, não adoeceram. Todos os ovinos que apresentaram sinais clínicos morreram entre 13 e 22 horas após a administração das folhas. Sinais clínicos leves como fezes pastosas, inquietação e aumento das frequências cardíaca e respiratória tiveram início cerca em 11 horas após a ingestão da planta. Minutos antes da morte observaram-se dispneia intensa com respiração abdominal pronunciada. As principais lesões macroscópicas foram restritas à cavidade torácica, como hidrotórax e edema pulmonar acentuado. O exame microscópico revelou no pulmão, edema acentuado e difuso na superfície pleural, nos espaços interlobulares e ao redor das artérias peribronquiais e peribronquiolares. Edema multifocal foi observado no interior de alvéolos e ao redor de brônquios e bronquíolos, além de leve infiltrado neutrofílico multifocal. O quadro clínico-patológico da intoxicação experimental foi similar ao observado na intoxicação espontânea e caracterizou-se por edema iulmonar acentuado e morte logo após o início dos sinais clínicos. ABSTRACT: This paper describes the toxic potential of the green leaves of Crotalaria pallida and the clinical signs in experimentally poisoned sheep. Green leaves of C. pallida were collected in São João do Sul, Santa Catarina, and administered orally to six adult sheep, as single dose of 40, 20, 10, 5, 2.5 and 2.5g/kg, and for two sheep at doses of 0.63 and 1.25g/kg during 30 days. The poisoning was reproduced at lethal doses of 2.5g/kg. One of the sheep that received a single dose of 2.5g/kg and another that received daily doses did become ill. All sheep wich developed clinical signs died between 13 and 22 hours after administration of the plant. Slight signs started in about 11 hours after ingestion of the plant, and clinical signs included loose stool, restlessness and increased heart and respiratory rates. A few minutes preceding death, pronounced abdominal breathing and severe dyspnea was observed. The main gross lesions were restricted to the thoracic cavity, as severe pulmonary edema and hydrothorax. Lung microscopy showed marked edema in the diffuse pleural surface, the interlobular spaces and around the peribronchial arteries. Multifocal edema was observed within alveoli and around the bronchi and bronchioles, and mild multifocal infiltration of neutrophils. The clinical and pathological alerations of the experimental poisoning was similar to that observed in the spontaneous poisoning, characterized by marked pulmonary edema and death shortly after the onset of clinical signs.

Adhesion proteins from Mycoplasma gallisepticum (MG) encoded by cytadhesion genes mgc1 and mgc2 were cloned into plasmid vectors and transformed into E. coli. Seventeen groups of specific-pathogen free (SPF), birds at four weeks of age were used to inoculate these two proteins (MGC1 and MGC2) mixed into an oil emulsion creating a novel MG vaccine. Six different protein concentrations (50, 100, 200, 400, 800, and 1000µg/bird) were tested with two equal concentration doses at four and seven weeks of age. In addition, many control groups were needed such as bacterin, membrane, no vaccine or challenge, oil emulsion alone, and no vaccine but challenged. Three weeks following the second vaccination, 50% of the birds in each treatment group were challenged with MG strain S6. The remaining birds were left as contacts to verify protection against horizontal transmission. All birds were bled before vaccinations, challenge and euthanasia. Birds were negative for MG at the first vaccination, as shown by serum plate agglutination test. At necropsy, tissue samples (trachea, lungs, and air sacs) were collected for histopathological examination. Swabs from trachea were used for PCR analysis. ELISA results showed a strong immune response to both protein preparations and almost the same response level for different doses tested, proving the immunogenic features of MGC1 and MGC2. However, humoral responses failed to prevent MG infection and disease when challenged as demonstrated by PCR and histopathology. MGC1 contact birds showed some degree of infection by PCR analysis. In addition, histopathological and ELISA results suggest that contact birds did not have enough time to develop lesions and to mount an immune response. Os genes mgc1 e mgc2, codificadores de duas proteínas de adesão (MGC1 e MGC2) da bactéria Mycoplasma gallisepticum, foram clonados em E. coli. Dezessete grupos de aves livres de patógenos específicos (SPF), com quatro semanas de idade, foram inoculados com uma emulsão oleosa contendo as proteínas MGC1 e MGC2 purificadas. Seis concentrações (50, 100, 200, 400, 800, e 1000µg/ave) foram testadas com duas doses idênticas, às quatro e sete semanas de vida, respectivamente. Além disso, grupos controles foram avaliados com uma vacina comercial contra micoplasmose aviária, membrana de MG, grupo sem vacina/sem desafio, grupo vacina oleosa de MGC1 sem desafio, grupo com vacina oleosa de MGC2 sem desafio, grupo desafiado mas sem vacina. Três semanas após a segunda e a última vacinação, 50% dos animais dos grupos tratamentos foram desafiados com a cepa S6 de MG. O restante dos animais foi deixado como contato para averiguar proteção contra a transmissão horizontal da doença. Amostras de sangue de todas as aves foram coletadas antes das vacinações, do desafio e da eutanásia. As aves eram negativas para MG às quatro semanas de vida, conforme visto na aglutinação em placa. Na necropsia, tecidos (traqueia, pulmão e sacos aéreos) foram coletados para exame histopatológico. Suabes da traqueia foram utilizados para a PCR. Os resultados do ELISA demonstraram forte resposta imune contra as duas proteínas testadas e resposta similar independentemente do número de doses, provando a sua capacidade imunogênica. Porém, esta resposta humoral gerada foi incapaz de prevenir a infecção e a doença após desafio, conforme demonstrado pelos exames PCR e histopatológico. Aves-contato, inoculadas com MGC1, demonstraram estar infectadas nas análises de PCR. Além disso, os resultados do histopatológico e ELISA sugerem que os animais-contato não tiveram tempo suficiente para demonstrar lesões ou resposta imune.

The objective of this study was to evaluate low and high Brix diets, treated or not with 5 g of calcium oxide per kg of natural matter, and corn silage on intake, digestibility and performance of beef cattle. Forty cattle with initial body weight (BW) of 350 kg were used: five composed the control group, 30 were distributed into random blocks (control) and the other five were distributed in a 5 × 5 incomplete Latin square, with the objective of determining digestibility. The 30 animals evaluated for performance were slaughtered and empty body weight (EPW), carcass dressing and meat cuts were determined. The diet with corn silage (CS) presented the best intake of the other ingredients and the best weight gain, except for neutral detergent fiber intake in g/kg of BW. Only carcass dressing, in relation to BW and EBW, was not affected by the treatments, and the others were greater for animals fed diets with sugar cane silage. Animals fed diets with high brix sugar cane silage and treated high brix sugar cane silage presented lower intake of indigestible neutral detergent fiber and neutral detergent fiber corrected for ash and protein (g/kg of BW) in relation to diets with low and high brix sugar cane silage, respectively. Animals fed diets with corn silage presented higher digestibility, except for crude protein and non-fibrous carbohydrates. Animals subjected to diets with corn silage presented low excretion of nitrogen compounds and higher microbial crude protein synthesis. Animals fed sugar cane silage present greater intake, performance and digestibility. The use of lime during 15 or 20o Brix sugar cane ensilage does not alter intake, digestibility or performance of beef cattle.

BACKGROUND: Pre-DSM-III (where DSM is Diagnostic and Statistical Manual), a series of studies demonstrated that major depressive syndromes were common after bereavement and that these syndromes often were transient, not requiring treatment. Largely on the basis of these studies, a decision was made to exclude the diagnosis of a major depressive episode (MDE) if symptoms could be "better accounted for by bereavement than by MDE" unless symptoms were severe and very impairing. Thus, since the publication of DSM-III in 1980, the official position of American Psychiatry has been that recent bereavement may be an exclusion criterion for the diagnosis of an MDE. This review article attempts to answer the question, "Does the best available research favor continuing the 'bereavement exclusion' (BE) in DSM-5?" We have previously discussed the proposal by the DSM-5 Mood Disorders Work Group to remove the BE from DSM-5. METHODS: Prior reviews have evaluated the validity of the BE based on studies published through 2006. The current review adds research studies published since 2006 and critically examines arguments for and against retaining the BE in DSM-5. RESULTS: The preponderance of data suggests that bereavement-related depression is not different from MDE that presents in any other context; it is equally genetically influenced, most likely to occur in individuals with past personal and family histories of MDE, has similar personality characteristics and patterns of comorbidity, is as likely to be chronic and/or recurrent, and responds to antidepressant medications. Netherton syndrome is a rare autosomal recessive disease characterized by erythroderma, ichthyosis linearis circumflexa, atopy, failure to thrive and a specific hair shaft abnormality called trichorrhexis invaginata or bamboo hair, considered pathognomonic. We report the case of a 4-year-old boy with erythroderma since birth, growth deficit and chronic diarrhea. Trichoscopy was used to visualize typical bamboo and "golf tee" hair and of key importance to diagnose Netherton syndrome. We suggest the use of this procedure in all children diagnosed with erythroderma.

The second half of beef cows gestation in tropical areas usually occurs during the drought season, where pastures are low in CP. The objective was to evaluate the effects of CP supplementation during the last third of gestation on beef cows weight variation and indicators of nutrient uptake and skeletal muscle mobilization. Forty-three pregnant (gestating males) Nellore cows (543 ± 66 kg) were divided into 8 paddocks (5–6 cows per paddock) of Urochloa decumbens pasture. Four paddocks were supplemented daily with 2 g/kg of a 43.5% CP supplement (SUP, n = 4) and four received only a mineral supplement (CON, n = 4). Treatments started 90 days before the predicted calving date. Liver and Longissimus muscle biopsies for relative mRNA expression and circulating plasma levels of AA were taken at 270 days of gestation. Data were analyzed using MIXED procedure of SAS considering dietary treatment as fixed effect. Differences were declared at P < 0.10. Supplemented cows had greater weight at calving (P = 0.065) and lost less maternal tissues (without considering gestation-related compounds) (P = 0.052) but the calf birth weight was not affected (P = 0.649). Control cows had greater circulating levels of total AA, non-essential AA, glucogenic AA, His, Met, Ala Asn and Ser (P < 0.10) than supplemented cows, probably due to mobilization of skeletal muscle to support liver gluconeogenesis. The expression of mRNA markers for gluconeogenesis in liver was not affected by the supplementation (P > 0.10). Supplemented cows had greater expression of mRNA markers for protein synthesis in muscle (P < 0.10) but not for protein degradation (P > 0.10). In conclusion, CP supplementation for beef cows in late gestation reduces the maternal weight loss probably by giving more substrates for gluconeogenesis and thus reducing the mobilization of AA from maternal skeletal muscle.
