The testis morphology, spermatogenesis, and the spermatozoon structure in two palaeonemerteans, Callinera sp. and Parahubrechtia sp., have been examined by light, laser confocal, and electron microscopy. Early spermatogenesis is quite similar between the two species. The similarities include the formation of the initial proacrosomal vesicle at the primary spermatocyte stage, the presence of electron-dense non-membranous substance around the centrioles in spermatogonia and spermatocytes, and the occurrence of lipid droplets at those stages. However, some differences in the process of acrosome formation during spermiogenesis were also found. In Callinera sp., a spherical acrosomal vesicle (rounded in the sections) transforms into a convex disk structure (arc-shaped in the longitudinal sections) in the developing posterior part of the spermatid and then migrates anteriorly; in Parahubrechtia sp., a rounded acrosomal vesicle migrates towards the apex of the cell where it is transformed into the arc-shaped structure. Both species have spermatozoa of a primitive type, i.e., with a small acrosomal complex, a rounded nucleus, a short mid-piece, and a posteriorly oriented flagellum. Nevertheless, some modifications in the sperm organization such as the asymmetric location of the acrosomal complex and mid-piece were recorded. It is possible that such modifications are synapomorphic for palaeonemerteans. In Callinera sp., the mid-piece contains 2–4 mitochondria, while Parahubrechtia sp. has only one ring-shaped but unclosed mitochondrion. The presence of several mitochondria in Callinera sp. may be regarded as a result of the so-called ‘progenetic spermatogenesis’.

The process of spermatogenesis and spermatozoon morphology was characterized from a deep-sea bivalve, Calyptogena pacifica (Vesicomyidae, Pliocardiinae), a member of the superfamily Glossoidea, using light and electron microscopy. Spermatogenesis in C. pacifica is generally similar to that in shallow-water bivalves but, the development of spermatogenic cells in this species has also some distinguishing features. First proacrosomal vesicles are observed in early spermatocytes I. Although, early appearance of proacrosomal vesicles is well known for bivalves, in C. pacifica, these vesicles are associated with electron-dense material, which is located outside the limiting membrane of the proacrosomal vesicles and disappears in late spermatids. Another feature of spermatogenesis in C. pacifica is the localization of the axoneme and flagellum development. Early spermatogenic cells lack typical flagellum, while in spermatogonia, spermatocytes, and early spermatids, the axoneme is observed in the cytoplasm. In late spermatids, the axoneme is located along the nucleus, and the flagellum is oriented anteriorly. During sperm maturation, the bent flagellum is transformed into the typical posteriorly oriented tail. Spermatozoa of C. pacifica are of ect-aqua sperm type with a bullet-like head of about 5.8 μm in length and 1.8 μm in width, consisting of a well-developed dome-shaped acrosomal complex, an elongated barrel-shaped nucleus filled with granular chromatin, and a midpiece with mainly four rounded mitochondria. A comparative analysis has shown a number of common traits in C. pacifica and Neotrapezium sublaevigatum.

Activated charcoal (AC) can be taken orally as enterosorbent for treatment of pathological states related to exogenous and endogenous intoxications. Synthesized granulated AC with a highly developed active surface (SBET ~2700 m2/g) was used as a medical countermeasure (MCM) to acute radiation sickness (ARS) in rats after total body X-ray irradiation. AC demonstrates positive results in ARS treatment, as expressed in, (i) a decrease in body weight loss, (ii) a protection of bone marrow (BM) cells colony formation capacity, (iii) a reduction of BM chromosomal aberrations and small intestine and spleen tissue damage, (iv) an amelioration of white blood cell count, and (v) a mitigation of superoxide ion generation rate in the liver. Successful gametogenesis in invertebrates is tightly associated with functioning of specific nutrient-storing cells. In oysters, cells of vesicular connective tissue (VCT-cells), also referred to as storage cells, which form a meshwork around gonadal acini, are the major population of cells that accumulate and provide nutrients for developing gametes. During the annual reproductive cycle, populations of developing germ cells and VCT-cells demonstrate the inversely proportional size dynamics: the larger the acini, the smaller the VCT-cells. In the present study, the morphology of VCT-cells in the Pacific oyster, Crassostrea gigas, at the active gametogenesis stage of reproductive cycle has been studied using light and transmission electron microscopy. At this stage, VCT-cells are big, irregularly shaped cells containing large nucleus with a single large nucleolus. The cytoplasm contains weakly developed endoplasmic reticulum, mitochondria in the perinuclear area and at periphery of the cell, numerous lipid droplets, and glycogen particles. Ultrastructure of VCT-cells is similar to the organization of brown adipocytes in mammals. The surface of cells has numerous cytoplasmic processes that are presumably associated with the transport function and provide close interaction with adjacent cells. The spatial relationship between VCT-cells and myoepithelial elements of the gonad area is demonstrated and discussed.

The aim of this work was a study of the functional activity of neutrophils and peripheral blood monocytes in rats with the transplanted Walker carcinosarcoma as potential predictors of the sensitivity of this tumor to doxorubicin treatment. This study provides an evidence that such indices of the functional activity of circulating phagocytes of the tumor-bearing rats as the quantity and the phagocytosis intensity of monocytes, as well as the intensity of ROS production by monocytes and neutrophils, may reflect the degree of sensitivity of the tumor to doxorubicin. So it was shown that the growth of the resistant tumor caused a significant increase of the number of circulating phagocytic cells and the intensity of phagocytosis by more than 100% (p < 0.001) compared with the corresponding indices of intact rats and rats with the parental variant of the tumor. The ability of blood mono-cytes and neutrophils of rats with a resistant tumor to produce ROS was also significantly different from that in intact rats and animals with the parental carcinosarcoma variant. The predictive value of these indices is especially important in the dynamic monitoring of the development of tumor drug resistance during long-term cancer chemotherapy. Considering the standard 2 - 3 week interval between the courses of cancer therapy and the short lifetime of circulating phagocytes, an assessment of the indices of their functional activity before each subsequent course can be considered as a pretreatment assessment. Meanwhile, further studies are needed to determine the spectrum of malignant neoplasms for which the degree of tumor drug resistance correlates with the functional activity of circulating phagocytes.

Recent findings regarding early lophotrochozoan development have altered the conventional model of neurogenesis and revealed that peripheral sensory elements play a key role in the initial organization of the larval nervous system. Here, we describe the main neurogenetic events in bivalve mollusks in comparison with other Lophotrochozoa, emphasizing a novel role for early neurons in establishing larval nervous systems and speculating about the morphogenetic function of the apical organ. We demonstrate that during bivalve development, peripheral sensory neurons utilizing various transmitters differentiate before the apical organ emerges. The first neurons and their neurites serve as a scaffold for the development of the nervous system. During veliger stage, cerebral, pleural, and visceral ganglia form along the lateral (visceral) nerve cords in anterior-to-posterior axis. The pedal ganglia and corresponding ventral (pedal) nerve cords develop much later, after larval settlement and metamorphosis. Pharmacological abolishment of the serotonin gradient within the larval body disrupts the navigation of “pioneer” axons resulting in malformation of the whole nervous system architecture. Comparative morphological data on neurogenetic events in bivalve mollusks shed new light on the origin of the nervous system, mechanisms of early axon navigation, and sequence of the tetraneurous nervous system formation. Furthermore, this information improves our understanding of the basic nervous system architecture in larval Bivalvia and Mollusca.

It is known that the arsenal of chemotherapeutic agents for the treatment of malignant brain tumors is quite limited, which causes the high relevance of research aimed at finding new effective antitumor regimens, including the use of energy metabolism modifiers. AIM To investigate the anti-glioma activity of sodium dichloroacetate (DCA) and metformin (MTF) used in combination in vitro and in vivo. MATERIALS AND METHODS Cell survival, cell cycle, apoptosis, mitochondrial membrane potential (Δψm), ATP level, the glucose consumption rate, and lactate production rate were determined in vitro in cultured glioma C6 cells. The antitumor action of agents in vivo was evaluated routinely by the prolongation of the life span of rats with transplanted intracerebral glioma C6 and was confirmed by histological examination of tumor tissue. RESULTS The half maximal inhibitory concentration (IC50) for DCA and MTF used separately was 79.2 ± 2.1 mM and 78.4 ± 4.0 mM, respectively, whereas IC50 for DCA used in combination with 7.8 mM MTF was 3.3 fold lower (24.0 ± 1.2 mM, p < 0.05). The 1-day incubation of cells with DCA at a concentration close to IC50 (25 mM), in combination with MTF at a concentration by order lower than IC50 (7.8 mM), in contrast to their separate use, resulted in a decrease in the number of viable cells by 40% (p < 0.05); redistribution of the cells by the cell cycle phases toward decreased proportion of cells in the S-phase by 46% (p < 0.05) and an increased percentage of cells in the G0/G1 phase by 24% (p < 0.05) compared to similar indices in the control. High proapoptotic activity of DCA in combination with MTF was supported by a significantly higher percentage of apoptotic cells in vitro than in the control (18.9 ± 4.4% vs 5.7 ± 1.3%, p < 0.05) and a high number of tumor cells with signs of apoptosis revealed during the histological examination of tumor pathomorphosis. The combined effect of DCA and MTF resulted in almost 4-fold decrease of the glucose consumption rate by glioma C6 cells (0.23 ± 0.05 μmol/106 cells/h vs 0.91 ± 0.12 μmol/106 cells/h, p < 0.05) compared to the corresponding parameters in the control, and 2-fold increased rate of lactate production (1.06 ± 0.03 μmol/106 cells/h vs 0.53 ± 0.03 μmol/106 cells/h, p < 0.05). At the same time, both Δψm and the level of intracellular ATP in the glioma C6 cells treated with DCA and MTF, both separately and in combination, did not differ significantly from those indices in the control. In in vivo studies, the average life span of rats with intracranial transplanted glioma C6, treated with DCA in combination with MTF in a total dose of 1.1 and 2.6 g/kg body weight, respectively, was 50% higher (p < 0.001) than in the control group. In contrast, in the case of single-use (at a dose of 2.6 g/kg), MTF increased the life span of tumor-bearing animals just by 19% (p < 0.01), whereas DCA alone (at a dose of 1.1 g/kg) did not significantly change the survival time of rats. CONCLUSIONS The obtained data indicate synergism of anti-glioma action of DCA and MTF in a case of their combined use both in vitro and in vivo and may be considered a starting point for the development of effective treatment regimens for malignant brain tumors based on the combined use of DCA and MTF.

Spiophanes uschakowi is a common polychaete living in tubes in sandy sediments in shallow waters of the Sea of Japan. Females and males release their gametes into the water where fertilization and holopelagic, planktotrophic larval development occur. In females, oogenesis is intraovarian: vitellogenesis occurs when the oocytes grow in paired ovaries attached to genital blood vessels in fertile segments. The developed oocytes are accumulated in the coelomic cavity prior to spawning. The newly released oocytes are lentiform, 185–200 μm in diameter, with honey-combed envelopes 5–7 μm thick. Each oocyte has 41–49 cortical alveoli regularly arranged in a peripheral circle, a nucleus 80–83 μm in diameter, and a single nucleolus about 30 μm in diameter. In males, spermatogonia proliferate in testes and the rest of spermatogenesis occurs in the coelomic cavity. During spermiogenesis, the acrosomal vesicle migrates from the posterior to the anterior part of the spermatid. The spermatozoa are ect-aquasperm with a plate-like acrosome 0.58 ± 0.06 μm thick and 2.14 ± 0.13 μm in diameter, barrel-shaped nucleus 2.23 ± 0.13 μm long and 3.18 ± 0.13 μm in diameter, short midpiece 0.93 ± 0.09 μm long with five spherical mitochondria, two centrioles and one small lipid droplet, and a flagellum 62–63 μm long with 9 × 2 + 2 organization of microtubules. The acrosome is a complex heterogeneous structure with 4–6 subspherical apical bodies, and numerous small branched basal cisternae. The anterior end of the nucleus is truncate, while its posterior end has wide shallow depressions accommodating the mitochondria. The centrioles are situated in the center of the midpiece between mitochondria and oriented obliquely to each other. The structure of the gametes of broadcast-spawning spionids is reviewed and the roles of surface granules in species-specific attraction of sperm toward eggs by releasing chemical signals (sperm chemotaxis), and cortical alveoli as a place of penetration of spermatozoa into oocytes (micropyle) are suggested. The lentiform oocytes of Spiophanes spp. are unique among Spionidae by their shape, while spermatozoa are unique by their plate-like acrosomes.

Background Bivalves comprise a large, highly diverse taxon of invertebrate species. Developmental studies of neurogenesis among species of Bivalvia are limited. Due to a lack of neurogenesis information, it is difficult to infer a ground pattern for Bivalvia. To provide more comprehensive morphogenetic data on bivalve molluscs and relationships among molluscan clades, we investigated neurogenesis in the Pacific oyster, Crassostrea gigas, from the appearance of the first sensory cells to the formation of the larval ganglionic nervous system by co-immunocytochemistry of the neuronal markers FMRFamide or 5-HT and vesicular acetylcholine transporter (VAChT). Results Neurogenesis begins with the emergence of the apical serotonin-immunoreactive (5-HT-ir) sensory cells and paired sensory posttrochal dorsal and ventral FMRFamide-immunoreactive (FMRFamide-ir) cells at the early trochophore stage. Later, at the early veliger stage, the apical organ (AO) includes 5-HT-ir, FMRFamide-ir, and VAChT-ir cells. At the same stage, VAChT-ir cells appear in the posterior region of larvae and send axons towards the AO. Thus, FMRFamide-ir neurites and VAChT-ir processes form scaffolds for longitudinal neurite bundles develop into the paired ventral nerve cords (VNC). Later-appearing axons from the AO/CG neurons join the neurite bundles comprising the VNC. All larval ganglia appear along the VNC as paired or fused (epiathroid) clusters in late veliger and pediveliger larvae. We observed the transformation of the AO into the cerebral ganglia, which abundantly innervated the velum, and the transformation of ventral neurons into the pedal ganglia, innervating the foot, gills, and anterior adductor muscle. The visceral ganglia appear last in the pediveliger oyster and innervate the visceral mass and posterior adductor of premetamorphic larvae. In addition, a local FMRFamide-ir network was detected in the digestive system of pediveliger larvae. We identified VAChT-ir nervous elements in oyster larvae, which have not been observed previously in molluscs. Finally, we performed a morphology-based comparative analysis of neuronal structures among bivalve, conchiferan, and aculiferan species. Conclusions We described the development of the nervous system during the larval development in Crassostrea gigas. These data greatly advance the currently limited understanding of neurodevelopment in bivalves and mollusks, which has hampered the generation of a ground pattern reconstruction of the last common ancestor of Mollusca. Our morphological data support phylogenomic data indicating a closer Bivalvia-Gastropoda sister group relationship than the Bivalvia-Scaphopoda (Diasoma) group relationship. Commercial importance and ability to live in a wide range of salinities have made the common mussel, Mytilus trossulus, a relevant model to study modulation of larval growth and development. We investigated the effects of various salinities combined with neomycin and ampicillin application on Mytilus larvae survival and growth. Both neomycin and ampicillin enhanced trochophore and veliger survival under condition of low salinity. The average veliger size was increasing in accordance with the increase of salinity. In case of neomycin treatment 3.6% of the larvae reached the pediveliger stage. No abnormalities of larval morphology of the FMRFamide and 5-HT systems occurred after 7 days of culturing with both antibiotics.

Adults of Aonides oxycephala, common inhabitants of shallow boreal waters in the Atlantic and Pacific Oceans, release gametes into the water where fertilization and lecithotrophic larval development occur. During spermiogenesis, the acrosomal vesicle migrates from the posterior to the anterior end of the spermatid and the number of mitochondria reduces from six in early spermatids to four in mature spermatozoa. Each spermatozoon has an ovoid head with the acrosome 1.4 ± 0.1 µm long and 1.6 ± 0.1 µm wide and the nucleus 1.7 ± 0.1 µm long and 2.3 ± 0.1 µm in diameter, four spherical mitochondria, two centrioles oriented perpendicular to each other, putative glycogen in the shape of dense granules in the midpiece, and a flagellum with 9 × 2 + 2 organization of microtubules. Chemoresistance is one of the major hurdles to overcome for the successful treatment of breast cancer. At present, there are several mechanisms proposed to explain drug resistance to chemotherapeutic agents, including decreased intracellular drug concentrations, mediated by drug transporters and metabolic enzymes; impaired cellular responses that affect cell cycle arrest, apoptosis, and DNA repair; the induction of signaling pathways that promote the progression of cancer cell populations; perturbations in DNA methylation and histone modifications; and alterations in the availability of drug targets. Both genetic and epigenetic theories have been put forward to explain the mechanisms of drug resistance. Recently, a small non-coding class of RNAs, known as microRNAs, has been identified as master regulators of key genes implicated in mechanisms of chemoresistance. This article reviews the role of microRNAs in regulating chemoresistance and highlights potential therapeutic targets for reversing miRNA-mediated drug resistance. In the future, microRNA-based treatments, in combination with traditional chemotherapy, may be a new strategy for the clinical management of drug-resistant breast cancers.

Spermatogenesis in Boccardiella hamata (Polychaeta: Spionidae) from the Sea of Japan: sperm formation mechanisms as characteristics for future taxonomic revision. —Acta Zoologica (Stockholm) 91: 477–456. To characterize novel features that will be useful in the discussion and validation of the spionid polychaete Boccardiella hamata from the Sea of Japan, the successive stages of spermatogenesis were described and illustrated. Spermatogonia, spermatocytes and early spermatids are aflagellar cells that develop synchronously in clusters united by a cytophore. At the middle spermatid stage, the clusters undergo disintegration and spermatids produce flagella and float separately in coelomic fluid as they transform into sperm. Spermatozoa are filiform. The ring-shaped storage platelets are located along the anterior nuclear area. The nucleus is cupped by a conical acrosome. A nuclear plate is present between the acrosome and nucleus. The nucleus is a cylinder with the implantation fossa throughout its length and with the anterior part of the flagellum inside the fossa. There is only one centriole, serving as a basal body of the flagellum, situated in close vicinity of the acrosomal area. A collar of four mitochondria is located under the nuclear base. The ultrastructure of B. hamata spermatozoa from the Sea of Japan appears to be close to that of B. hamata from Florida described by Rice (Microscopic Anatomy of Invertebrates, Wiley-Liss, Inc., New York, 1992), suggesting species identity of the samples from the two regions. However, more detailed study of Florida’s B. hamata sperm is required for a reliable conclusion concerning the similarity of these two polychaetes. In addition to sperm structure, features such as the cytophore-assigned pattern of spermatogenic cell development, the synchronous pattern of cell divisions, the non-flagellate early spermatogenic stages, and the vesicle amalgamation that drives meiotic cell cytokinesis and spermatid diorthosis will likely be useful in future testing of the validity of B. hamata and sibling species throughout the world.

Some pairs of oyster species in the genus Crassostrea can produce viable hybrids, and heterospecific fertilization may be symmetric, asymmetric, or very low. For example, with C. angulata and C. gigas, fertilization is equally successful whether the mature oocytes come from the first or the second species. Fertilization is asymmetric between C. ariakensis and C. virginica, i.e., it is higher when C. ariakensis is the source of the oocytes. Fertilization success is low between C. ariakensis and C. gigas. These differences probably relate chiefly to molecular mechanisms of gamete recognition, but the present study examines whether the size and certain aspects of gamete morphology may also play a role. Individuals of four species of the genus Crassostrea (Ostreidae: Bivalvia), collected from different geographic regions between 2006 and 2011, C. angulata (Penghu Island, Taiwan Strait), C. ariakensis (Nha Trang Bay, South China Sea), C. gigas (Peter the Great Bay, Sea of Japan), and C. virginica (Galveston Bay, Gulf of Mexico), were examined for conspecific relationships between the thickness of the vitelline envelope in mature oocytes and the size of the acrosome or subacrosomal spaces in spermatozoa. Thicker vitelline envelopes corresponded with bigger acrosomes and subacrosomal spaces in three of the four species. The exception was C. gigas, which has the thickest vitelline envelope, but the lytic compartments in its spermatozoa are not the largest. Moreover, sizes of acrosome, subacrosomal space, and vitelline envelope are similar between the species with symmetric heterospecific fertilization, while difference in sizes of these structures increases the possibility of asymmetric cross-fertilization or decreased fertilization rate.

The ultrastructural mechanism of postspawning sperm resorption in the testes of the sea urchin Strongylocentrotus nudus is described. Two types of phagosomes (containing sperm and containing residual bodies) are formed in the cytoplasm of nutritive phagocytes. The phagosomes fuse with electron-dense globules, and their contents are gradually destroyed. Afterward, phagosomes are transformed into electron-transparent vacuoles, which are finally compressed by the surrounding cytoplasm.

Phagocytic resorption during spermatogenesis was studied in the sea urchin Anthocidaris crassispina. Nutritive phagocytes in gonad absorbed both waste sperm cells and residual bodies discarded from maturing spermatids, and these materials were subsequently compartmented in heterophagosomes. Based on 180 heterophagosomes examined by transmission electron microscopy, over 99% of heterophagosomes contained either residual bodies or sperm cells only. Simultaneous resorption of sperm cells and residual bodies in a heterophagosome was uncommon, with only ∼0.56% occurrence, suggesting that heterophagosomes have a selective resorption ability in nutritive phagocytes.
