SARS-CoV-2 is one of the main serious challenges of human societies, which emerged in December 2019 from China and quickly extends to all parts of the world. The virus was previously believed to only affect the lungs and respiratory system, but subsequent research has revealed that it affects a variety of organs. For this reason, this disease is known as a multiorgan disease. Current article aimed to highlight latest information and updates about molecular studies regarding pathogenesis of SARS-CoV-2 in kidney, liver, and cardiovascular and respiratory systems, as well as the mechanisms of interaction of these organs with each other to cause clinical manifestations in patients.

Numerous prescribed drugs and herbal and dietary supplements have been reported to cause drug-induced acute liver injury, which is a frequent cause of acute liver failure (ALF). It is a tremendous challenge with ever-increasing drug application in the medication system for huge populations. Drug-induced acute liver injury can lead to diverse pathologies similar to acute and chronic hepatitis, acute liver failure, biliary obstruction, fatty liver disease, and so on. Recently, extensive work demonstrated that isoflavones play an essential and protecting role in drug-induced liver injury (DILI). The isoflavones mediated hepatoprotection by modulating specific genes linked with control of cellular redox homeostasis and inflammatory responses. Isoflavones upregulate oxidative stress-responsive nuclear factor erythroid 2-like 2 (Nrf2), downregulate inflammatory nuclear factor-κB (NF-κB) signaling pathways, and modulate a balance between cell survival and death. Moreover, isoflavones actively inhibit the expression of cytochromes P450 (CYPs) enzyme during drug metabolism. Moreover, isoflavones are also linked with farnesoid X receptor (FXR) activation and signal transducer and activator of transcription factor 3 (STAT3) phosphorylation in hepatoprotection DILI. In vivo and in vitro studies clearly stated that isoflavones bear strong antioxidant potential and promising agents for hepatotoxicity prevention and stressed their potential role as therapeutic supplements in DILI. The current review will elaborate on isoflavones’ preventive and therapeutic potential concisely and highlight various molecular targets to exert a protective effect on DILI.

The biologically active phytochemicals are sourced from edible and medicinally important plants and are important molecules being used for the formulation of thousands of drugs. These phytochemicals have great benefits against many ailments particularly the inflammatory diseases or oxidative stress-mediated chronic diseases. Eugenol (EUG) is a versatile naturally occurring molecule as phenolic monoterpenoid and frequently found in essential oils in a wide range of plant species. EUG bears huge industrial applications particularly in pharmaceutics, dentistry, flavoring of foods, agriculture, and cosmeceutics. It is being focused recently due to its great potential in preventing several chronic conditions. The World Health Organization (WHO) has declared EUG as a nonmutant and generally recognized as safe (GRAS) molecule. The available literature about pharmacological activities of EUG shows remarkable anti-inflammatory, antioxidant, analgesic, and antimicrobial properties and has a significant effect on human health. The current manuscript summarizes the pharmacological characteristics of EUG and its potential health benefits.

Senescence is a cellular process that can be initiated by certain stressors such as UVA irradiation. The mechanism by which skin cells protect themselves from the UVA-induced senescence has not been fully investigated. Here, we demonstrate that Bach2 modulates the extent of UVA-induced photoaging through regulation of autophagy in skin fibroblasts. In fact chronic exposure of skin fibroblasts to UVA resulted in a significant decrease in Bach2 expression, both in vitro and in vivo. In addition, knockdown of Bach2 in skin fibroblasts led to an increased expression of cell senescence-related genes, which further enhanced the UVA irradiation-induced photoaging. On the other hand, the overexpression of Bach2 resulted in a decrease in the expression of cell senescence-related genes. We also demonstrate that the knockdown of Bach2 in skin fibroblasts can lead to a decreased expression of autophagy-related genes and vice versa, suggesting that autophagy is involved in Bach2-mediated regulation of senescence in skin fibroblasts. Additionally, inhibition of autophagy with autophagy inhibitor 3-MA suppressed the expression of autophagy-related proteins and promoted cell senescence. Furthermore, knockout of Atg5 or Atg7 in embryonic mouse fibroblasts led to a significant increase in the expression of cell senescence-related genes. Immunoprecipitation assays further demonstrated that Bach2 directly interacts with Beclin-1, Atg3, Atg7, and LC3 in fibroblasts. Taken together, these findings revealed a critical role for Bach2 in suppressing the UVA irradiation-induced cell senescence via autophagy in skin fibroblasts. Bach2 can therefore be a potential target for the therapy of UV-induced photoaging because of its ability to regulate the process of autophagy in the skin.

Purpose: To synthesize and characterize some new gabapentin Schiff base derivatives, and to assess their antibacterial, antioxidant and antiepileptic activities.Methods: Four Schiff base derivatives of gabapentin, termed G1, G2, G3 and G4, were synthesized by condensation with benzoin, vanillin, acetophenone, and benzophenone, respectively. Their chemical identities were established by FTIR, 1 H NMR and 13C NMR techniques. The new compounds were screened for antibacterial activity using agar well method, antioxidant activity by DPPH assay, and anticonvulsant activity against pentylenetetrazole (PTZ) induced seizures in mice.Results: All the compounds showed antibacterial activity against the test strains to variable degrees, while the parent drug did not exhibit antibacterial activity. The zones of inhibition of compound G2 against Micrococcus luteus (36.2 ± 1.0 mm) and Serratia marcescens (28.2 ± 1.0 mm), and of compound G4 against Stenotrophomonas maltophilia (36.8 ± 1.0 mm) were larger compared to thestandard drug, doxycycline, exhibiting zones of inhibition 28.2 ± 1.3, 28.2 ± 0.9 and 20.0 ± 0.9 mm, respectively. In addition, compounds G1 and G2 possessed significantly greater (p < 0.05) radical scavenging activity (82.3 ± 1.8 and 92.3 ± 2.2 %, respectively) than the precursor drug, gabapentin (63.2± 2.6 %). The seizure scores for compounds G1 (0.7 ± 0.06) and G2 (0.9 ± 0.07) were comparable(p ˃ 0.05) with gabapentin (0.8 ± 0.06), while compounds G3 and G4 were less active (p < 0.05) than gabapentin.Conclusion: Compounds G1 and G2 exhibit good antibacterial and antioxidant activities while retaining the anticonvulsant activity of the parent drug, gabapentin, thus making them suitable candidates for further development for the treatment of neurodegenerative pathologies associated with bacterial infections. Context: Patients with liver disease are always at high risk of post-operative complications. The prediction of postoperative outcome is very crucial in the management of these patients. Various prediction models are in use to serve this purpose. Childs-Turcotte-Pugh (CTP) and Model of End-Stage Liver Disease (MELD) scoring systems are globally used to predict post-operative mortality in end stage liver disease patients. Aims: To compare the results of CTP and MELD scoring systems for predicting outcome in patients of chronic liver disease and to conclude which model is better for risk stratification, so as to enable us in better management of these patients. Settings and Design: Observational study Methods and Material: This is an Observational study that was carried out at General Surgery Department of Nishtar Medical University and Hospital from October 2019 to March 2020. We collected data from 30 patients. All the surgeries were done by the same team of consultant surgeons. All the investigations were done by the same institution. Scores of both CTP and MELD models were calculated preoperatively and post-operative outcome was compared with them to find out which model was a better predictor of mortality. Statistical analysis used: All the continuous variables were reported as mean ± standard deviation. Kaplan Meyer Survival Analysis was conducted to compare post-op survival time among patients divided on the basis of MELD score and CTP Grade. Pooled log rank test was conducted to determine if there were differences in the survival distributions for the different categories in each group. Objective: To study the clinical outcomes and determine the antibiotic resistance in patients of enteric perforation in a tertiary care hospital. Study Design: Cross Sectional study. Setting: Department of General Surgery at Nishtar Hospital, Multan. Period: May 2017 to December 2019. Material & Methods: Sample size was calculated to be 97 with a confidence level of 95% and margin of error of 10%. We collected data from 100 patients. All the surgeries were done by the same team of consultant surgeons. The abdomen was closed by the same surgical team using the same surgical technique to avoid any bias. All the continuous variables were reported as mean ± standard deviation. The antibiotic resistance was compared with the number of patients with wound infections, burst abdomen, ICU admissions and number of in-hospital stay days. Categorical variables were compared using Chi square test for Independence. The number of in-hospital stay days were compared using Student t-test. The results were compiled using SPSS version 20. Results: Our result indicated that a patient had a greater chance of having a burst abdomen and wound infection if there was resistance to ciprofloxacin or ceftriaxone. Similar results were obtained for ICU admissions. There was a significantly longer in-hospital stay observed for patients who were resistant to the standard regimen of Ceftriaxone. Conclusion: It seems only rationale that the patients susceptible should be treated with Imipinem for 2 weeks as the empirical therapy rather than the standard empirical therapy of ceftriaxone and ciprofloxacin. The blood cultures can be sent before starting Imipinem and treatment management plan changed if the culture results are sensitive for the empiric antibiotics.

BACKGROUND: For augmentation and sustainability of Human Systems Management (HSM), it is significant to analyse and reposition journals, which is very well done by bibliometric analysis. OBJECTIVE: The study aims to provide the retrospective of HSM between 1980– 2019. METHOD: Scopus database has been employed to bibliographic indicators for the representation of available data for forty years. By utilising the VOSviewer software, bibliographic materials being drawn from keywords, graphical visuals, bibliographic coupling & co-citation analysis have been done. RESULTS: Results enabled to establish that the average number of publications remained almost the same throughout forty years, tinting the journal has been as vital in 2019 for researchers as it was in 1980. Citations originated drastic mounting with recently published documents in comparison with earlier production. CONCLUSIONS: Authors and researchers are suggested to cover knowledge and knowledge management related articles. This study suggests management to ensure the inculcation of more related areas in the scope by announcing special issues to broaden the journals’ scope as it remained the main reason for limiting HSM from gaining as much attention as it deserved.

Penicillium digitatum (green mold) is pathogenic fungi and causes citrus fruit postharvest rotting that leads to huge economic losses across the world. The current study was aimed to develop a new derivative of cinnamaldehyde (4-methoxycinnamaldehyde) through the cross-hydroxyaldehyde condensation method with benzaldehyde substituted by a benzene ring under the catalysis of alkaline reagent and, moreover, to test their antifungal potential against P. digitatum, the major citrus fruit rotting fungi. Multiple derivatives of cinnamaldehyde viz. 4-nitro CA, 4-chloro CA, 4-bromo CA, 4-methyl CA, 4-methoxy CA, and 2,4-dimethoxy CA were synthesized in the current study whereas the 4-methoxy CA showed highest antifungal actions for citrus fruit postharvest rotting fungi P. digitatum. Moreover, 4-methoxy CA was found to reduce the spore germination and growth by damaging the fungal cell membrane, as well as declined the levels of reducing sugars.

Objectives: Recognizing the risk factors affecting the in-hospital stay of the patients operated for peptic ulcer perforation. Study Design: Cross Sectional study. Setting: General Surgical Wards, Nishtar Hospital Multan. Period: From 15th November 2018 to 5th of March 2019. Material & Methods: A total of 100 patients of all ages who had a peptic ulcer perforation were included. Patients with a perforation that wasn't attributable to a peptic ulcer e.g. malignancy or gastrinoma were excluded. Patients were asked about their smoking and drinking habits. Data was collected about various risk factors of peptic ulcer, their co-morbidities and the medications. Results: Gender, smoking, alcohol intake, H-pylori infection, size & site of perforation did not have a significant effect on hospital stay days. In patient stay days differed in groups based on comorbidities and medication. A weak correlation was found among age and stay days. Conclusion: Patients with complain of COPD and other pulmonary complications have a longer hospital stay in the hospital. The association of arthritis and hypertension could not be demonstrated comprehensively.

Solar light has fundamental roles in vast chemical, biochemical, and physical process in biosphere and hence been declared as “source of life.” Solar light is further classified into a broad range of electromagnetic waves, and each region in the solar spectrum bears its unique actions in the universe or biosphere. Since centuries, solar light is believed as a potent source of killing pathogens causing postharvest losses on food products as well as human skin diseases. Citrus fruit crops are widely produced and consumed across the world, but due to their higher juicy contents, Penicillium italicum (blue mold) and Penicillium digitatum (green mold) make their entry to decay fruits and cause approximately 80% and 30% fruit losses, respectively. Agrochemicals or synthetic fungicides are highly efficient to control these postharvest fungal pathogens but have certain health concerns due to toxic environmental residues. Therefore, the scientific community is ever looking for some physical ways to eradicate such postharvest fungal pathogens and reduce the yield losses along with maintaining the public health concerns. This review article presents and discusses existing available information about the positive and negative impacts of different spectrums of solar light exposure on the postharvest storage of citrus fruits, especially to check citrus postharvest rotting caused by Penicillium italicum (blue mold) and Penicillium digitatum (green mold). Moreover, a special focus shall be paid to blue light (390–500 nm), which efficiently reduces the decay of fruits, while keeping the host tissues/cells healthy with no known cytotoxicity, killing the fungal pathogen probably by ferroptosis, but indepth knowledge is scanty. The study defines how to develop commercial applications of light in the postharvest citrus industry.

Introduction: There are two techniques for reconstruction of anterior cruciate ligament (ACL), open technique and arthroscopic assisted technique. Arthroscopic assisted technique has many advantages over open procedure but it needs more expertise and cost comparatively.The objective of this study is to identify the clinical outcomes on basis of lyshlomknee score (LKS) system and find out patients satisfaction after performing both procedures in two groups separately. Material and Methods: Retrospective analysis of 600 patients undergoing open ACL reconstruction and arthroscopic reconstruction from 2005 to 2018 was done, at the Department of Orthopaedics, Ghurki Hospital, Lahore. We included all those patients who were 18 to 45 years of age and had at least 1 year follow up.

The immune system is influenced by vitamin B complex: B1, B2, B3, B5, B6, B7, B9 and B12. The B complex insufficiency can cause significant impairment in cellular and immune function and trigger an inflammatory response. There is currently no certified SARS-CoV-2 treatment or a credible vaccine, but strengthening the immune system with B vitamins could go a long way in preventing and treating COVID-19 patients. Thus, a complete and nutritious diet must be followed before approved drugs and potential good vaccine research results are available to boost the normal functioning of the immune system. In order to activate adaptive and inborn immune responses, reduce cytokine levels such as proinflammatory cytokines, decrease oxidative stress, preserve endothelial homogeneity, improving pulmonary function, prevents hypercoagulable conditions and shortening the length of hospital stay; B-Complex vitamins play a significant role. Thus, the role of B complex in patients with COVID-19 needs to be evaluated and additional non-drug B vitamins can be used in existing treatments.

DNA barcodes are regarded as hereditary succession codes that serve as a recognition marker to address several queries relating to the identification, classification, community ecology, and evolution of certain functional traits in organisms. The mitochondrial cytochrome c oxidase 1 (CO1) gene as a DNA barcode is highly efficient for discriminating vertebrate and invertebrate animal species. Similarly, different specific markers are used for other organisms, including ribulose bisphosphate carboxylase (rbcL), maturase kinase (matK), transfer RNA-H and photosystem II D1-ApbsArabidopsis thaliana (trnH-psbA), and internal transcribed spacer (ITS) for plant species; 16S ribosomal RNA (16S rRNA), elongation factor Tu gene (Tuf gene), and chaperonin for bacterial strains; and nuclear ITS for fungal strains. Nevertheless, the taxon coverage of reference sequences is far from complete for genus or species-level identification. Applying the next-generation sequencing approach to the parallel acquisition of DNA barcode sequences could greatly expand the potential for library preparation or accurate identification in biodiversity research. Overall, this review articulates on the DNA barcoding technology as applied to different organisms, its universality, applicability, and innovative approach to handling DNA-based species identification.

Solar UVA irradiation-generated reactive oxygen species (ROS) induces the expression of matrix metalloproteinase 1 (MMP-1), leading to photoaging, however the molecular mechanism remains unclear. In the present study, we found that eriodictyol remarkably reduces UVA-mediated ROS generation and protects the skin cells from oxidative damage and the ensuing cell death. Moreover eriodictyol pretreatment significantly down-regulates the UVA-induced MMP-1 expression, and lowers the inflammatory responses within the skin cells. Pretreatment with eriodictyol upregulates the expression of tissue inhibitory metalloproteinase 1 (TIMP-1) and collagen-I (COL-1) at the transcriptional level in a dose-dependent manner. UVA-induced phosphorylation levels of c-Jun N-terminal kinase (JNK), extracellular signal-regulated kinase (ERK) and p38 leading to increased MMP-1 expression are significantly reduced in eriodictyol-treated skin cells. In addition, eriodictyol pretreatment significantly suppresses inflammatory cytokines and inhibits the activation of MAPK signaling cascades in skin cells. Taken together, our results demonstrate that eriodictyol has both potent anti-inflammatory and anti-photoaging effects.

Background. Solar ultraviolet radiation A (UVA, 320-400 nm) is a significant risk factor leading to various human skin conditions such as premature aging or photoaging. This condition is enhanced by UVA-mediated iron release from cellular iron proteins affecting huge populations across the globe. Purpose. Quercetin-loaded zinc oxide nanoparticles (quercetin@ZnO NPs) were prepared to examine its cellular iron sequestration ability to prevent the production of reactive oxygen species (ROS) and inflammatory responses in HaCaT cells. Methods. Quercetin@ZnO NPs were synthesized through a homogenous precipitation method, and the functional groups were characterized by Fourier transform infrared (FTIR) spectroscopy, whereas scanning electron microscopy (SEM) described the morphologies of NPs. MTT and qRT-PCR assays were used to examine cell viability and the expression levels of various inflammatory cytokines. The cyclic voltammetry (CV) was employed to evaluate the redox potential of quercetin-Fe3+/quercetin-Fe2+ complexes. Results. The material characterization results supported the loading of quercetin molecules on ZnO NPs. The CV and redox potential assays gave Fe-binding capability of quercetin at 0.15 mM and 0.3 mM of Fe(NO3)3. Cytotoxicity assays using quercetin@ZnO NPs with human HaCaT cells showed no cytotoxic effects and help regain cell viability loss following UVA (150 kJ/m2). Conclusion. Quercetin@ZnO NPs showed that efficient quercetin release action is UV-controlled, and the released quercetin molecules have excellent antioxidant, anti-inflammatory, and iron sequestration potential. Quercetin@ZnO NPs have superior biocompatibility to provide UVA protection and medication at once for antiphotoaging therapeutics.

To date, the SARS-CoV-2 spread has already become a global pandemic with a considerable death toll. The associated symptoms of the COVID-19 infection varied with increased inflammation as an everyday pathological basis. Among various other symptoms such as fever, cough, lethargy, gastrointestinal (GI) symptoms included diarrhea and IBD with colitis, have been reported. Currently, there is no sole cure for COVID-19, and researchers are actively engaged to search out appropriate treatment and develop a vaccine for its prevention. Antiviral for controlling viral load and corticosteroid therapy for reducing inflammation seems to be inadequate to control the fatality rate. Based on the available related literature, which documented GI symptoms with diarrhea, inflammatory bowel diseases (IBD) with colitis, and increased deaths in the intensive care unit (ICU), conclude that dysbiosis occurs during SARS−COV-2 infection as the gut-lung axis cannot be ignored. As probiotics play a therapeutic role for GI, IBD, colitis, and even in viral infection. So, we assume that the inclusion of studies to investigate gut microbiome and subsequent therapies such as probiotics might help decrease the inflammatory response of viral pathogenesis and respiratory symptoms by strengthening the host immune system, amelioration of gut microbiome, and improvement of gut barrier function.

We investigated the effect of various growth substances such as hydrogen peroxide, salicylic acid (SA), moringa leaf-extract (MLE) and ascorbic acid (ASA) on leaf physiology and seed cotton yield (SCY) of heat-stressed cotton. Cotton plants were exposed to elevated temperatures at three reproductive stages, either by staggering planting time in the field or by increasing growth cabinet temperatures (38/24°C and 45/30°C) in glasshouse. Elevated temperature at any reproductive phase significantly damaged cellular membrane and reduced SCY. Plants exposed to 38/24°C and 45/30°C in glasshouse produced 63% and 22% lower SCY, respectively, compared with plants under optimal temperature ((32/20°C). In response to high temperature, cotton plants up-regulated activities of anti-oxidative enzymes e.g. peroxidase and ascorbic acid. However, this defensive system could not protect cellular membrane of stressed plants from extreme temperature (38 and 45°C). In contrast, growth substances such as H2O2, ASA and MLE significantly increased anti-oxidative enzymes activity to an extent, which reduced heat-induced damage to cellular membrane. No significant effect of any regulator was observed on SCY under optimum temperatures; although H2O2, MLE and ASA significantly increased SCY of heat-stressed cotton. Hydrogen peroxide increased SCY of April and May thermal regimes crops by 16% (averaged across both sowing dates) under field, while it caused 14% and 20% increase in SCY of plants exposed to sub (38/24°C) and supra optimal (45/30°C) thermal regimes under glasshouse. We concluded that growth regulators, specifically, H2O2 can protect cotton crops from heat-induced cellular membrane damage by up-regulating antioxidant defense system.

Objective: To compare the efficacies of neurological physical examination, neurothesiometer and PainDETECT questionnaire in diagnosing diabetic neuropathy. Study Design: Prospective cross-sectional Place and Duration of Study: Department of General Medicine, Nishtar Hospital Multan, Pakistan from 1st December 2018 to 10th March 2019. Materials and Methods: One hundred and four patients of both type 1 and type 2 diabetes visiting the outdoor department were included in this study. They were assessed by lab results of glycosylated hemoglobin, fasting and random blood sugar levels and neurological physical examination. Results: The physical examination with Michigan Neuropathy Screening instrument showed that around 29 of the patients were having established neuropathy. The PainDETECT questionnaire on the other hand showed about 42 patients having a definitive neuropathy while the neurothesiometer showed that 79 of the total patients had varying degrees of neuropathy. Conclusion: The neurothesiometer is a better diagnostic tool for diagnosing diabetic neuropathy in patients.

Drug-induced liver injury (DILI) is a major cause of acute liver failure (ALF) as a result of accumulated drugs in the human body metabolized into toxic agents and helps generate heavy oxidative stress, inflammation, and apoptosis, which induces necrosis in hepatocytes and ultimately damages the liver. Sirtuin 1 (SIRT1) is said to have multiple vital roles in cell proliferation, aging, and antistress systems of the human body. The levels of SIRT1 and its activation precisely modulate its critical role in the interaction between multiple step procedures of DILI. The nuclear factor kappa-light-chain-enhancer of activated B cell- (NF-κB-) mediated inflammation signaling pathway, reactive oxygen species (ROS), DNA damage, mitochondrial membrane potential collapse, and endoplasmic reticulum (ER) stress also contribute to aggravate DILI. Apoptosis is regarded as the terminal reaction followed by multiple signaling cascades including caspases, p53, and mitochondrial dysfunction which have been said to contribute in DILI. The SIRT1 activator is regarded as a potential candidate for DILI, because the former could inhibit signaling of p53, NF-κB, and ER stress. On the other hand, overexpression of SIRT1 also enhances the activation of antioxidant responses via Kelch-like ECH-associated protein 1- (Keap1-) nuclear factor- (erythroid-derived 2-) like 2 (Nrf2) signaling. The current manuscript will highlight the mechanism of DILI and the interaction of SIRT1 with various cytoplasmic factors leading to DILI along with the summary of potent SIRT1 agonists.

Carvacrol has long been studied for its natural antifungal potential and food preservative. But the exact mode of its action remained highly complex as a general, but especially for Penicillium digitatum (P. digitatum) largely remained unexplored. Herein, a 1H-NMR-based metabolomic technique was used to investigate the antifungal mechanism of carvacrol. The metabolomic profiling data showed that alanine, aspartate, glutamate and glutathione metabolism were imbalanced in the fungal hyphae. A strong positive correlation was seen between aspartate, glutamate, alanine and glutamine, while negative correlation among glutathione and lactate. These metabolic changes revealed that carvacrol-induced oxidative stress had disturbed the energy production and amino acid metabolism of P. digitatum. Current study will improve the understanding of the metabolic changes posed by plant-based fungicides in order to control citrus fruit green mold caused by P. digitatum. Moreover, the study will provided certain experimental and theoretical basis for the development of novel citrus fruit preservatives.

Background: Exposure of human skin to solar ultraviolet A (UVA) irradiation causes severe oxidative stress with damage to various cellular components and concomitant inflammation and carcinogenesis. Objective: The aim of this study is to investigate the protective effect of acetyl-11-keto-β-boswellic acid (AKBA) against UVA radiation on human skin keratinocytes. Methods: HaCaT cells were pretreated with AKBA followed by UVA irradiation. Radiation effects on cell morphology, cell viability, intracellular reactive oxygen species (ROS) levels, and antioxidant enzymes were examined. Results: AKBA reduces UVA irradiation-induced cell viability loss, accompanied by a decreased production of UVA-induced ROS, decreased malondialdehyde, and increased superoxide dismutase expression. In addition, AKBA increased basal and UVA-induced levels of Nrf2 (NF-E2-related factor 2), the redox-sensitive factor, and its target genes NQO1 and heme oxygenase-1 (HO-1), whereas expression of the transcriptional repressor Bach1 (BTB and CNC homology 1) was reduced. Furthermore, the cytoprotective effects of AKBA against UVA-derived oxidative damage were accompanied by modulating expression of inflammatory mediators (i.e., cyclooxygenase-2 and nuclear factor-κB) and NOX1. Conclusions: AKBA protects skin cells from UVA-induced damage by modulating inflammatory mediators and/or ROS production. Therefore, AKBA has potential in the development of skin care products.

Objectives: To highlight an economical laparoscopic technique for rapidrecovery & early return to work especially for the 3rd world inguinal hernia patients. StudyDesign: Descriptive study. Setting: Surgical unit of Nishtar Hospital Multan. Period: 02 yearsi.e. from January 2014 to January 2016. Methods: There were 90 patients in the study. A prolenemesh of 15 x 15 cm used for the repair of inguinal hernia laparoscopiclly without the use ofspacer balloon & tacker (To make it cost effective). Results: Out of 90 patients only 3 patientsdevelop scrotal seroma. All the patients discharged on the 1st postoperative day & allowed toreturn to their normal routine work from the 3rd postoperative day onward. Conclusion: (e-TEP)without spacer balloon & tacker is an economical approach toward the inguinal hernia repair.

Ultraviolet A (UVA) irradiation is a potential environmental stressor, which contributes to inflammation, photoaging, and carcinogenesis. UVA causes endoplasmic reticulum stress, hence phosphorylates the α subunit of eIF2. Meanwhile, UVA also induces expression of haem oxygenase-1 (HO-1) and nuclear factor erythroid-derived two related factor 2 (Nrf2) in human skin cells. In mouse JB6 cell, we found high dose UVA could change cell morphology, cause cell viability loss. UVA irradiation activated phosphorylation of eIF2α and Nrf2-HO-1 pathway in a dose-dependent manner. Besides, modulation of eIF2α phosphorylation status could alter expression pattern of Nrf2-HO-1 signalling. Salubrinal, a selective inhibitor of eIF2α dephosphorylation, increased the S phase in cell cycle of JB6 cells after UVA irradiation, suggesting phosphorylation status of eIF2α may affect cellular homeostasis under UVA irradiation. The study directed to further acknowledge about the relationship of UVA-induced eIF2α phosphorylation and Nrf2-HO-1 pathway, which may play a role in phototherapy and photo protection.

Phytochemical investigation of fruits of Chaenomeles speciosa Lindley (Rosaceae) led to the isolation of 20 compounds including 5 flavonoids (1-5), 5 phenylpropanoids (6-10), 3 benzoic acid derivatives (11-13), 2 phloroglucinols (14 and 15), 2 purines (16 and 17), and 3 terpenoids (18–20). Their structures were elucidated by nuclear magnetic resonance analyses and from mass spectrometry data. These compounds were confirmed as catechin (1), epicatechin (2), catechin-5- O-β-d-glucoside (3), procyanidin B1 (4), quercetin-3- O-β-d-glucoside (5), p-coumaric acid (6), ferulic acid (7), caffeic alcohol (8), 1- O-p-coumaroyl-β-d-glucose (9), 1- O-cinnamoyl-β-d-glucose (10), p-hydroxybenzonic acid (11), protocatechuic acid (12), benzoic acid-β-d-gentiobioside (13), phloracetophenone 4′-glucoside (14), 3,5-dihydroxyphenyl β-d-glucopyranoside (15), adenine (16), adenosine (17), betulalbuside A (18), vomifoliol (19), and roseoside (20). Compounds 3-5, 8, 10, and 13-18 were isolated from the genus Chaenomeles and C. speciosa for the first time. Out of all these, compound 17 showed the best anti-inflammatory properties, comparable with those of the already known minocycline.

Polymorphous light eruption (PLE) is one of the acquired idiopathic photodermatosis mainly induced by immoderate UV radiation. In order to realize UV protection and medicine administration simultaneously for polymorphous light eruption protection and therapy, Acetyl-11-keto-β-boswellic acid (AKBA) loaded Zinc Oxide (ZnO) nanoparticles of which drug release behavior is UV-controlled has been successfully synthesized. Such nanoparticles can not only reflect UV but also transfer the energy to release AKBA which presents an excellent antioxidant and anti-inflammatory effects. In addition, they are biocompatible to HaCaT cells. As a result, they have a great potential in combining UV protection and medicine administration simultaneously for PLE protection and therapy.

Background Generation of reactive oxygen species (ROS), triggered by ultraviolet radiation (UVR), is associated with carcinogenesis of the skin. UV irradiation induced superoxide anion (O2•−) is the key ROS involved in the cellular damage. The cytoprotective efficacy of an unknown anti-oxidant compound can be evaluated by analyzing the production of O2•− from treated cells. Methods In this study, a glass carbon electrode functionalized with nanotube@DNA-Mn3(PO4)2 composite was applied to quantitative determination of generation of highly unstable O2•− from the melanoma A375 cell line following UVR(UV, UVA and UVB). In addition, the cytoprotective efficacy of anti-oxidant α-tocopherol was evaluated by quantifying the production of O2•−. Results The results showed that, UVR triggers generation of O2•− in melanoma A375 cells, and α-tocopherol is effective in diminishing the production of O2•− following UV irradiation. By comparing the conventional cell-survival assays results, we found that our simple and quick electrochemical sensing method can quantify O2•− generation through the biological activity of an anti-oxidant compound (α-tocopherol). Conclusion Our label-free electrochemical quantification method for ROS (O2•− major) in cells facing UVR stress demonstrates its potential application for high-throughput screening of anti-oxidation compounds.

The human Telomerase Reverse Transcriptase (hTERT) is one of key players in cancer metastasis and stemness, but its exact function in tongue squamous cell carcinoma remains unknown. Here, we aim to understand the role of hTERT by utilizing the CRISPR/Cas9 gene editing system to deplete hTERT in the SCC-15 cell line. Functional comparison of SCC-15 control and knockout cells (hTERT −/− ) showed that loss of hTERT suppressed cell proliferation and migration/invasion. Furthermore, hTERT depletion significantly decreased tumorigenesis, including alterations in cellular morphology that areindicative for epithelial-mesenchymal transition (EMT). Mechanistically we demonstrated that the hTERT knockout attenuates NF-κB signaling via a negative feedback regulation in tumorprogression. From these results we propose a novel molecular mechanism of hTERT to promote SCC-15 invasion and metastasis via NF-κB activation. We conclude that targeting hTERT may represent a new therapeutic strategy to improve therapy and survival of tongue squamous cell carcinoma patients.

Consequences of drought stress in crop production systems are perhaps more deleterious than other abiotic stresses under changing climatic scenarios. Regulations of physio-biochemical responses of plants under drought stress can be used as markers for drought stress tolerance in selection and breeding. The present study was conducted to appraise the performance of three different maize hybrids (Dong Dan 80, Wan Dan 13, and Run Nong 35) under well-watered, low, moderate and SD conditions maintained at 100, 80, 60, and 40% of field capacity, respectively. Compared with well-watered conditions, drought stress caused oxidative stress by excessive production of reactive oxygen species (ROS) which led to reduced growth and yield formation in all maize hybrids; nevertheless, negative effects of drought stress were more prominent in Run Nong 35. Drought-induced osmolyte accumulation and strong enzymatic and non-enzymatic defense systems prevented the severe damage in Dong Dan 80. Overall performance of all maize hybrids under drought stress was recorded as: Dong Dan 80 > Wan Dan 13 > Run Nong 35 with 6.39, 7.35, and 16.55% yield reductions. Consequently, these biochemical traits and differential physiological responses might be helpful to develop drought tolerance genotypes that can withstand water-deficit conditions with minimum yield losses.

Long wave UVA radiation (340-400 nm) causes detrimental as well as beneficial effects on human skin. Studies of human skin fibroblasts irradiated with UVA demonstrate increased expression of both antifibrotic heme oxygenase-1 (HO-1) and matrix metalloproteinase 1 (MMP-1). The use of UVA-induced MMP-1 is well-studied in treating skin fibrotic conditions such as localized scleroderma, now called morphea. However, the role that UVA-induced HO-1 plays in phototherapy of morphea has not been characterized. In the present manuscript, we have illustrated and reviewed the biological function of HO-1 and the use of UVA1 wavebands (340-400 nm) for phototherapy; the potential use of HO-1 induction in UVA therapy of morphea is also discussed.

UVA treatment of cultured human skin fibroblasts (FEK4) has been shown previously to reduce transcriptional activation of heme oxygenase 1 (HO-1) following a second dose of UVA radiation, a phenomenon known as refractoriness. This study demonstrates that the levels of HO-1 protein are also reduced after a second dose of UVA radiation as are Nrf2 levels, and there is less accumulation of Nrf2 in the nucleus where as Bach1 does accumulate in the nucleus. Cell viability is further reduced and cell membrane damage increased as compared with a single UVA treatment when an initial UVA treatment was followed by a second dose. Knockdown of Nrf2 by siRNA (siNrf2) targeting caused additional refractoriness of HO-1 protein induction to a second UVA or heme treatment and this treatment also further enhanced cell damage by a second dose of UVA radiation. However, transfection with Nrf2 caused less refractoriness of HO-1 to a second dose of UVA and reduced cell damage by a second dose of UVA radiation. These findings are consistent with the proposal that Nrf2 is involved in HO-1 refractoriness and could serve as a cytoprotective factor against cell damage caused by repeated exposure to moderate doses of UVA radiation. We propose that protection by the Nrf2-HO-1 pathway protection may have clinical relevance since human skin is exposed repeatedly to UVA radiation.

Objectives: To determine the effectiveness of interferential current on overactive bladder syndrome. Methodology: It is a quasi-experimental study. The duration of the study is 8 weeks. Total 40 patients were selected for this experimental study from Benazir Bhutto Hospital (BBH) Rawalpindi Pakistan. Sample was drawn through non-probability sampling technique called simple convenient sampling. Standard questionnaire was used for measurement through which results were calculated. Complete information about the patients including their biodata, symptomatology was entered in a Performa and then data was entered on the basis of Overactive Bladder Symptom Score (OABSS) scoring system. A total of 40 patients between the age of 5 to 12 years of age with no gender, religion, or ethnic background differentiation and with the diagnosis of overactive bladder were included. Results: it showed that baseline Mean pyuria was 1.75 ± 0.493 SD and after treatment 1.25 ± 0.439 SD and dysuria baseline mean is 1.78 ± 0.474 SD and after treatment Mean is 1.32 ± 0.423 SD. Hematuria baseline mean is 1.98 ± 0.423 SD and after treatment mean is 1.78 ± 0.158 SD mean frequency of urination is 2.0 ± 0.234 and after treatment mean is 1.2 ± 0.342 bed wetting mean is 1.62 ± 0.423 SD and after treatment mean is 1.22 ± 0.490 SD OABSS Score mean is 2.125 ± 0.334 SD and after treatment mean is 1.55 ± 0.597 SD all values showed significant difference. Conclusion: Interferential Current (IFC) Therapy is a very effective and beneficial tool to improve the symptoms and quality of life of the patients with Hinman syndrome.

Wound healing is a complex process which involves proliferation and migration of keratinocyte for closure of epidermal injuries. A member of microRNA family, let-7b, has been expressed in mammalian skin, but its exact role in keratinocyte migration is still not in knowledge. Here, we showed that let-7b regulates keratinocyte migration by targeting the insulin-like growth factor IGF2BP2. Overexpression of let-7b led to reduced HaCaT cell migration, while knockdown of let-7b resulted in enhanced migration. Furthermore, let-7b was decreased during wound healing in wild-type mice, which led us to construct the transgenic mice with overexpression of let-7b in skin. The re-epithelialization of epidermis of let-7b transgenic mice was reduced during wound healing. Using bioinformatics prediction software and a reporter gene assay, we found that IGF2BP2 was a target of let-7b, which contributes to keratinocyte migration. Introduction of an expression vector of IGF2BP2 also rescued let-7b-induced migration deficiency, which confirms that IGF2BP2 is an important target for let-7b regulation. Our findings suggest that let-7b significantly delayed the re-epithelialization possibly due to reduction of keratinocyte migration and restraints IGF2BP2 during skin wound healing.

Medicinal plants have served humans since prehistoric times to treat various ailments. Both developed and underdeveloped countries rely on traditional systems of medication using natural sources from plants. Phyllanthus is one of the largest genus in the family Phyllanthaceae, comprising over 700 well known species cosmopolitan in distribution mainly in the tropics and subtropics. Phyllanthus species are being in constant used in traditional medications to cure an array of human diseases (constipation, inhalation related, arthritis, loss of appetite, injuries, conjunctivitis, diarrhoea, running nose, common cold, malaria, blennorrhagia, colic, diabetes mellitus, dysentery, indigestion, fever, gout, gonorrheal diseases of males and females, skin itching, jaundice, hepatic disorders, leucorrhea, vaginitis, menstrual irregularities, obesity, stomach pains, and tumors), confectionaries, food industry, and in some pesticides. Phyllanthus species are rich in diversity of phytochemicals e.g., tannins, terpenes, alkaloids, glycosidic compounds, saponins, and flavones etc. More in depth studies are a direly needed to identify more compounds with specific cellular functions to treat various ailments.
