Staphylococcus aureus is one of the main bacterial agents responsible for cases of mastitis in ruminants, playing an important role in the persistence and chronicity of diseases treated with antimicrobials. Using the multilocus sequence typing technique, network approaches and study of the population diversity of microorganisms, we performed analyzes of S. aureus (ES-GPM) isolated from goats with persistent mastitis (GPM). The most strains of ES-GPM were categorically different phylogenetically from the others and could be divided into two lineages: one with a majority belonging to ES-GPM and the other to varied strains. These two lineages were separated by 27 nuclear polymorphisms. The 43 strains comprised 22 clonal complexes (CCs), of which the ES-GPM strains were present in CC133, CC5 and a new complex formed by the sequence type 4966. The genetic diversity of some alleles showed be greater diversity and polymorphism than others, such as of the aroE and yqiL genes less than glpF gene. In addition, the sequences ES-GPM to the arc gene and glpF alleles showed the greatest number of mutations for ES-GPM in relation to non-ES-GPM. Therefore, this study identified genetic polymorphisms characteristic of S. aureus isolated from milk of goats diagnosed with persistent mastitis after the failed treatment with the antibiotic enrofloxacin. This study may help in the future to identify and discriminate this agent in cases of mastitis, and with that, the most appropriate antibiotic treatment can be performed in advance of the appearance of persistent mastitis caused by the agent, reducing the chances of premature culling and animal suffering.

Background Staphylococcus aureus is one of the main causative agents of mastitis in small ruminants. Antimicrobial use is the major treatment, but there are many flaws linked to resistance, tolerance or persistence. This study aimed to verify changes in resistance, virulence and clonal profiles of S. aureus isolated from persistent mastitis goat milk before and after enrofloxacin treatment. Results MIC increased to at least one antimicrobial in S. aureus isolates after enrofloxacin treatment compared to before. The most detected resistance genes before and after treatment were tetK, tetM, and blaZ, with more resistance genes detected after enrofloxacin treatment (p hla, fnbA, fnbB, eta, etb, sea, sec, seh, and sej) were detected at both times, and between these, the hla and eta genes were detected more in isolates after treatment. All isolates of S. aureus belonged to the same sequence type (ST) 133, except for two S. aureus isolates prior to enrofloxacin treatment which were classified as ST5 and the other as a new one, ST4966. Isolates of S. aureus 4, 8, and 100 from before and after treatment had identical pulse types, while others obtained from other animals before and after treatment were classified into distinct pulse types. Conclusion There were occasional changes in the studied profiles of S. aureus isolated before and after treatment of animals with enrofloxacin, which may have contributed to the permanence of bacteria in the mammary gland, even when using traditional treatment, resulting in persistent mastitis.

Antibiotic resistance has become a major concern for human and animal health. As fluoroquinolones have been extensively used in human and veterinary medicine, there has also been the rapid emergence and spread of antimicrobial resistance around the world. Here, we analysed the microbiome of goat milk using samples from healthy goats and those diagnosed with persistent mastitis and treated using the antibiotic enrofloxacin with 16S rRNA amplicon sequencing. We selected a group of 11 goats and 22 samples of milk that did not respond clinically to enrofloxacin treatment. Milk samples were evaluated before and after treatment to verify changes of the microbiota; the three first lactating goats were selected from the healthy control group. The milk samples from the healthy control animals presented a larger abundance of different species of bacteria of the Staphylococcus genus, but a smaller number of different genera, which indicated a more specific niche of resident bacteria. The Firmicutes phylum was predominantly different between the studied groups. Samples from before-treatment animals had a higher number of new species than those from the control group, and after being treated again. These microbiota received new bacteria, increasing the differences in bacteria even more in relation to the control group. Genotypes such as Trueperella and Mannheimia, between other genera, had a high abundance in the samples from animals with persistent mastitis. The dysbiosis in this study, with marked evidence of a complex microbiota in activity in cases of the failure of antimicrobial treatment for persistent chronic mastitis, demonstrates a need to improve the accuracy of pathogen identification and increases concern regarding antibiotic treatments in milk production herds.

A Zona da Mata de Minas Gerais possui uma cadeia especializada de produção de leite de cabra. O leite de cabra é superior em qualidade em comparação com o leite de outras espécies domésticas, e a demanda por leite e produtos lácteos do público tem aumentado. Os dados sobre o sistema de criação de cabras leiteiras em Minas Gerais são escassos e relativamente antigos, e essa falta de informação limita a implementação de medidas profiláticas, especialmente para a mastite, que representa o maior problema sanitário nos rebanhos leiteiros. Isso também causa problemas socioeconômicos e problemas de mercado para a criação de cabras leiteiras. O objetivo deste trabalho foi caracterizar a mastite e as bactérias associadas em cabras leiteiras na Zona da Mata de Minas Gerais. Um total de 539 cabras em lactação foi examinado e 268 amostras individuais (uma por teto) foram coletadas de animais positivos no teste da caneca de fundo escuro e/ou Califórnia Mastitis test (CMT). As culturas microbiológicas foram realizadas em meio Agar sangue e as bactérias foram submetidas a testes fenotípicos, genotípicos e testes de susceptibilidade antimicrobiana. A prevalência de mastite subclínica foi de 28,0% e a prevalência clínica foi de 2,8%. A multiplicação bacteriana foi obtida em 62,0% das amostras. Cento e oitenta e sete bactérias foram identificadas. As espécies mais identificadas foram: Staphylococcus aureus (60,4%), seguida de Staphylococcus epidermidis (9.1%,), Escherichia coli (6.9%), Staphylococcus saprophyticus (5.9%) e Staphylococcus caprae (4,3%) em ordem decrescente. As bactérias do gênero Staphylococcus apresentaram um perfil de resistência aos antimicrobianos pertencentes à classe de beta-lactâmicos - penicilina, ampicilina e oxacilina - além da tetraciclina, em contraste com os outros antimicrobianos testados. Doze por cento dos isolados apresentaram resistência múltipla a antibióticos (MDR) e foram encontrados em cinco microrregiões. Entre as bactérias com maior prevalência de MDR, 38,5% foram E. coli e 10,6% S. aureus. Os produtores da Zona da Mata de Minas Gerais são tecnificados, trabalham com raças leiteiras especializadas praticam e possuem bom manejo. No entanto, algumas medidas relacionadas à profilaxia e ao controle das doenças, como a vacinação, têm baixa adesão ou não são realizadas por falta de assistência veterinária. Este é o primeiro estudo com foco nesta região, que possui grande relevância na produção de leite de cabra no Brasil, fornecendo informações importantes que podem auxiliar na implementação de medidas de profilaxia e controle das doenças, e na manutenção de um fornecimento constante de produtos em quantidade e qualidade suficientemente adequada para a população consumidora. The Zona da Mata of Minas Gerais has a specialized goat milk production chain. Goat milk is superior in quality compared with milk of other domestic species, and the demand for milk and milk products for the public has increased. Data on dairy goat breeding in Minas Gerais are scarce and relatively old, and this lack of information has limited the implementation of prophylactic measures, especially for mastitis, which represents the biggest sanitary problem for dairy herds. The objective of this work was to characterize mastitis and bacteria associated with it in milking goats in the Zona da Mata of Minas Gerais. It also causes socioeconomic problems and market issues for dairy goat farming. A total of 539 lactating goats were examined and 268 individual samples (one for teat) were collected from animals positive for strip cup test and/or the California Mastitis Test (CMT). Microbiological cultures were carried out on blood agar medium and the bacteria were subjected to phenotypic, genotypic and antimicrobial susceptibility tests. The prevalence of subclinical mastitis was 28.0% and the clinical prevalence was 2.8%. Bacterial multiplication was obtained in 62% of samples. One hundred eighty seven total bacteria were identified. The most common species identified was Staphylococcus aureus (60%), followed Staphylococcus epidermidis (9.1%,), Escherichia coli (6.9%), Staphylococcus saprophyticus (5.9%) e Staphylococcus caprae (4.3%). Bacteria of the genus Staphylococcus presented a profile of resistance to antimicrobials belonging to the beta-lactam class (penicillin, ampicillin and oxacillin) in addition to tetracycline, in contrast to the other antimicrobials tested. Twelve percent of multidrug resistence (MDR) was found in five microregions. Among the bacteria with the highest prevalence of MDR, 38.5% were E. coli and 10.6% were S. aureus. The producers of the Zona da Mata of Minas Gerais are technicians who work with specialized dairy breeds and practise good management. However, some measures related to prophylaxis and control of diseases, such as vaccination, have low adherence or are not performed due to a lack of veterinary assistance. This is the first study focusing on this region, which is highly prominent in goat milk production in Brazil. It provides important information that can help in the implementation of measures for the prophylaxis and control of diseases, and for maintenance of a constant supply of products in sufficient quantities and of a quality suitable for the consumer population.

Mycobacterium avium subspecies paratuberculosis , the etiologic agent of Johne's disease or paratuberculosis, was identified by culture and/or polymerase chain reaction (PCR) in 50% and 30% of water samples for animal and human consumption, respectively, from ten dairy goat farms in Brazil. IS 1311 restriction fragment length polymorphism analysis identified the isolates as cattle type C.

Among the infectious diseases of the dairy herd, mastitis is the main cause of direct and indirect economic losses. Mastitis etiological agents are usually bacteria, but fungi and algae may be involved. Staphylococcus aureus is commonly isolated from mastitis and deserves special attention due to the numerous hospital infections related to methicillin resistant strains. Although the mammary gland presents different defense mechanisms, in many cases the etiologic agent can escape of the immune system and multiply, resulting in mastitis. Several nonspecific antimicrobial factors are found in the mammary gland, such as lactoperoxidase–thiocyanate¬–hydrogen peroxide, lysozyme, lactoferrin, and complement system. Adaptive or acquired immunity can lead to an immediate response in repetitive exposure to the same microorganism. Two types of adaptive responses are described: humoral and cellular immunities. Conventional diagnostic methods are widely considered for mastitis detection, like microbiological isolation and somatic cell count; however, alternative diagnosis methods are being increasingly used, like infrared thermography that measures the surface temperature of mammary gland skin and sphincters of the teats. Conventional therapy based on antibiotics is not always efficient, resulting in cases of persistent infections. Recent research using inhibitors of multidrug efflux systems and biofilms has been tested in vitro with reliable results. Studies have also demonstrated that bioactive substances extracted from plants with and without chemical modifications have shown synergistic associations with the traditional antimicrobials against bacteria causing mastitis. Other infectious diseases, such as brucellosis, tuberculosis, Johne's disease, leptospirosis, Salmonellosis, bovine viral diarrhea (BVD), and infectious bovine rhinotracheitis (IBR) can affect the dairy cattle herd, compromising the milk production chain in economic, social, and public health aspects.

In the last decade there was a significant increase dairy goat participation in the world and Brazilian agricultural scenario. Minas Gerais state is the third largest producer of goat milk in Brazil. Paratuberculosis is a chronic intestinal disease that mostly affects ruminants and can become a public health problem due to the possible association with Crohn's disease. It is transmitted by ingesting Mycobacterium avium subsp. paratuberculosis (MAP) contaminated food or water. In recent years, with the development of molecular techniques, the disease has been identified with greater precision and more quickly. MAP strains (S-Sheep, Cattle-C and B-Bison) have also been typed. The detection of a circulating MAP strain in a herd is important to develop epidemiological studies and thus establish better strategies to control paratuberculosis. This study aimed to identify and type MAP in dairy goat farms in the Zona da Mata, the main producing region of Minas Gerais state, Brazil. Feces and milk samples of 467 animals were collected, processed, inoculated in Herrold's Egg Yolk Agar (HEYM) and submitted to PCR and REA techniques. Eleven (2.36%) animals were positive for the presence of MAP in four properties and the isolates were characterized as type C strain. It was concluded that MAP is present in dairy goats from properties in Zona da Mata, MG and that strain type C circulates in the area. This is the first report of MAP typing isolated from dairy goat in Brazil.

Despite the known importance of Clostridium perfringens as an enteropathogen in small ruminants, little is known about the role of its additional virulence factors or the frequency of the various C. perfringens genotypes in healthy goats; this complicates the laboratory diagnosis of the infections caused by this microorganism. In light of this, the aim of the present study was to isolate and genotype C. perfringens from stool samples from healthy goats in Brazil. Stool samples from 250 apparently healthy adult goats from 17 different herds in Minas Gerais, Brazil were collected, and isolation and genotyping of C. perfringens was performed. C. perfringens type A was isolated from 189 (75.6%) goats, whereas C. perfringens types C and D were each detected in one goat (0.4%). All isolates were negative for enterotoxin-, NetB-, NetE-, and NetF-encoding genes. These results confirmed C. perfringens type A as part of the microbiota in these animals, and they suggested that C. perfringens type C and D are rarely isolated from healthy goats. RESUMO: Apesar da reconhecida importância de Clostridium perfringens como enteropatógeno de pequenos ruminantes, pouco se sabe sobre a frequência dos genótipos ou do papel de fatores de virulência adicionais de C. perfringens em cabras saudáveis, dificultando o diagnóstico laboratorial da infecção causada por esse micro-organismo. Dessa forma, o presente estudo teve como objetivo caracterizar C. perfringens de amostras de fezes de cabras adultas saudáveis. Amostras de fezes de 250 cabras saudáveis de 17 rebanhos diferentes em Minas Gerais, Brasil, foram submetidas ao isolamento e genotipagem de C. perfringens. C. perfringens tipo A foi isolado de 189 (75,6%) cabras, enquanto C. perfringens tipos C e D foram detectados em um animal (0.4%) cada. Todos os isolados foram negativos para os genes codificadores das toxinas NetB, NetE, NetF e enterotoxina. Os resultados apresentados confirmam C. perfringens tipo A como parte da microbiota de cabras saudáveis e sugere que C. perfringens tipos C e D são raramente encontrados em caprinos saudáveis.

Goat farming is a low-cost alternative to dairy production in developing countries. In Brazil, goat production has increased in recent years due in part to the implementation of programs encouraging this activity. Mycobacterium avium ssp. paratuberculosis (MAP) is the causative agent of paratuberculosis, a disease that causes chronic granulomatous enteritis in ruminants, but MAP transmission dynamics are still poorly understood in goats. In a previously published study of our research group, 10 dairy goat farms (467 animals) from Minas Gerais state were analyzed for MAP detection; 2 fecal cultures and 11 milk samples tested positive for MAP by conventional PCR and were confirmed by sequencing. Because no clinical signs were observed over 1 yr of monitoring, we hypothesized that these MAP-positive goats could be passive shedders. Thus, in the present study, 4 positive goats (4/13) from the previous study were purchased and feces and milk samples were collected for evaluation (twice, with an interval of 3 mo between tests) by culture of MAP, IS900 PCR, or both. All analyses were negative for MAP. At the last time point, blood samples were collected for ELISA, the animals were killed, and tissues collected for tissue culture and histopathology. At necropsy, no macroscopic lesions related to paratuberculosis were observed. Similarly, no histological changes were observed and MAP in samples stained by Ziehl-Neelsen was not detected. These animals were characterized as potential passive shedders with upward contamination of the teat canal by MAP. This is the first report of the passive shedding phenomenon in goats in Brazil and it highlights the importance of identifying these animals for control programs and to ensure the quality of dairy products.

Objetivou-se mensurar a movimentação das massas de gordura e proteína corporal, além de quantificar as massas dos órgãos e vísceras de cabras Alpinas ao longo das oito semanas de lactação. Foram utilizadas 51 cabras multíparas da raça Alpina, alocadas em baias metabólicas individuais. As cabras foram distribuídas em um delineamento inteiramente casualizado, com oito tratamentos e seis repetições, sendo os tratamentos constituídos pelas semanas de lactação. Foram realizados abates sequenciais de seis cabras por semanas de lactação para mensuração das massas de gordura e proteína, e também o peso dos órgãos e vísceras. Ocorreu redução significativa na massa dos tecidos proteico e adiposo, porém de forma adversa, sendo que o tecido proteico foi mobilizado com maior intensidade nas primeiras duas semanas pós-parto, já a o tecido adiposo uma queda de forma constante. Não houve efeito sobre peso do fígado, rúmen, omaso, abomaso e intestino grosso, mas ocorreu redução nos pesos do intestino delgado, útero e glândula mamária. As cabras Alpinas nas três semanas pós-parto mobilizam não apenas gordura, mas também proteína corporal para atender a demanda de produção de leite. Somente o intestino delgado, útero e glândula mamária tiveram seus pesos afetados com as oito semanas de lactação. This study aimed to measure the changes of masses of body fat and protein, and also quantify the masses of organs and viscera of Alpine goats throughout the eight weeks of lactation. We used 51 multiparous Alpine goats, allocated in individual metabolic pens. The goats were distributed in a completely randomized design with six replications and eight treatments, and the treatments by weeks of lactation. Slaughters were carried sequential six goats per week of lactation to measure the mass of fat and protein, as well as the weight of the organs and viscera. Significant reduction in mass and adipose tissue protein, but adversely, and the tissue protein was mobilized with greater intensity in the first two weeks postpartum, since the fat falling steadily. There was no effect on liver weight, rumen, omasum, abomasum and large intestine, but there was a reduction in the weights of the small intestine, uterus and mammary gland. The Alpine goats within three weeks postpartum mobilize not just fat, but also body protein to meet the demand of milk production. Only the small intestine, uterus and mammary gland shown weight affected with eight weeks of lactation.
