To explore the role of adiponectin-resistin (AR) index as a better indicator of obesity-related hypertension.This study continued a case control study that had finished recruiting 153 subjects divided as four characteristic groups. Fasting serum resistin levels (FSR) and Fasting serum adiponectin levels (FSA) were tested by ELISA. And, other related anthropometric clinical and metabolic data were collected. Analyzation on correlations between research index and differences between groups were done by SPSS. AR index's performance was also validated by the receiver operating characteristic (ROC) curves, the net reclassification improvement (NRI), and the integrated discrimination improvement (IDI).The AR index was defined as 1+ logThe AR index was more strongly associated with increased risk of obesity-related hypertension than the solely index of FSR or FSA and was useful for early diagnosis of obesity-related hypertension.

Urban air quality issues are closely related to human health and economic development. In order to investigate street-scale flow and air quality, this study developed the atmospheric photolysis calculation framework (APFoam 1.0), an open-source computational fluid dynamics (CFD) code based on OpenFOAM, which can be used to examine microscale reactive pollutant formation and dispersion in an urban area. The chemistry module of APFoam has been modified by adding five new types of reactions, which can implement the atmospheric photochemical mechanism (full O 3 –NO x –volatile organic compound chemistry) coupled with a CFD model. Additionally, the model, including the photochemical mechanism (CS07A), air flow, and pollutant dispersion, has been validated and shows good agreement with SAPRC modeling and wind tunnel experimental data, indicating that APFoam has sufficient ability to study urban turbulence and pollutant dispersion characteristics. By applying APFoam, O 3 –NO x –volatile organic compound (VOC) formation processes and dispersion of the reactive pollutants were analyzed in an example of a typical street canyon (aspect ratio H / W = 1 ). The comparison of chemistry mechanisms shows that O 3 and NO 2 are underestimated, while NO is overestimated if the VOC reactions are not considered in the simulation. Moreover, model sensitivity cases reveal that 82 %–98 % and 75 %–90 % of NO and NO 2 , respectively, are related to the local vehicle emissions, which is verified as the dominant contributor to local reactive pollutant concentration in contrast to background conditions. In addition, a large amount of NO x emissions, especially NO, is beneficial to the reduction of O 3 concentrations since NO consumes O 3 . Background precursors (NO x /VOCs) from boundary conditions only contribute 2 %–16 % and 12 %–24 % of NO and NO 2 concentrations and raise O 3 concentrations by 5 %–9 %. Weaker ventilation conditions could lead to the accumulation of NO x and consequently a higher NO x concentration but lower O 3 concentration due to the stronger NO titration effect, which would consume O 3 . Furthermore, in order to reduce the reactive pollutant concentrations under the odd–even license plate policy (reduce 50 % of the total vehicle emissions), vehicle VOC emissions should be reduced by at least another 30 % to effectively lower O 3 , NO, and NO 2 concentrations at the same time. These results indicate that the examination of the precursors (NO x and VOCs) from both traffic emissions and background boundaries is the key point for understanding O 3 –NO x –VOCs chemistry mechanisms better in street canyons and providing effective guidelines for the control of local street air pollution.

Objective: To explore the role of resistin in the onset and development of obesity-related hypertension.Methods: Resistin serum levels were tested by ELISA in 153 adult subjects among four characteristic Chinese adult physical examination groups. Waist circumference (WC), body mass index (BMI), systolic blood pressure (SB), diastolic blood pressure (DB), and other clinical laboratory data were collected. Following, correlations between research index and differences between groups were analyzed using SPSS.Results: Serum resistin levels statistically significantly negatively correlated with SB, DB and BMI, but statistically significantly positively correlated with serum creatinine (SCR) and serum albumin (ALB), even after adjustment for age and/or gender. The serum level of resistin in the normal healthy subject group (NH) was higher than in other groups.Conclusions: Resistin's role in the onset of obesity-related hypertension may be more important than what has been previously assumed. Regulatory T cells (Tregs) are often enriched in tumors, where their immunosuppressive function has a key role in tumor persistence and progression. In colorectal cancer (CRC), however, Tregs are frequently associated with an improved clinical outcome. Tumor-infiltrating Tregs have been shown to exhibit a distinct signature comprising the co-stimulatory molecules (OX40, 4-1BB), cytokine receptors (IL1R2, IL21R, CCR8, CD30), and co-inhibitory molecules (PD-L1, TIGIT). Here, we showed by flow cytometry that circulating CD45RO+ Tregs from patients with CRC (n = 25) have elevated CD30 and OX40 expression compared to healthy subjects (n = 14). We identified co-expression of CD30 and OX40 on circulating CD45RO+ Tregs using single-cell images captured by the DEPArray™ system. The frequency of CD30+OX40+CD45RO+ Tregs was significantly higher in CRC patients than in healthy subjects (P +OX40+ Treg subset could strongly discriminate between CRC patients and healthy subjects with the highest accuracy of 92.3%, an AUC of 0.92, a sensitivity of 88%, a specificity of 100%, a positive predictive value of 100%, a negative predictive value of 82.35%, and a trade-off value of 3.44%, compared to other Treg subsets. Consistently, multiplex-IHC/IF of tumor-infiltrating Tregs revealed a significant association between high densities of CD30+OX40+ Tregs and improved overall survival; no such association was found for other subsets. These data suggest a potential role for CD30+OX40+ Tregs as a diagnostic or prognostic biomarker in CRC.

BACKGROUND: The role of fecal aerosols in the transmission of severe acute respiratory syndrome coronavirus 2 has been suspected. OBJECTIVE: To investigate the temporal and spatial distributions of 3 infected families in a high-rise apartment building and examine the associated environment variables to verify the role of fecal aerosols. DESIGN: Epidemiologic survey and quantitative reverse transcriptase polymerase chain reaction analyses on throat swabs from the participants; 237 surface and air samples from 11 of the 83 flats in the building, public areas, and building drainage systems; and tracer gas released into bathrooms as a surrogate for virus-laden aerosols in the drainage system. SETTING: A high-rise apartment building in Guangzhou, China. PARTICIPANTS: 9 infected patients, 193 other residents of the building, and 24 members of the building's management staff. MEASUREMENTS: Locations of infected flats and positive environmental samples, and spread of virus-laden aerosols. RESULTS: 9 infected patients in 3 families were identified. The first family had a history of travel to the coronavirus disease 2019 (COVID-19) epicenter Wuhan, whereas the other 2 families had no travel history and a later onset of symptoms. No evidence was found for transmission via the elevator or elsewhere. The families lived in 3 vertically aligned flats connected by drainage pipes in the master bathrooms. Both the observed infections and the locations of positive environmental samples are consistent with the vertical spread of virus-laden aerosols via these stacks and vents. LIMITATION: Inability to determine whether the water seals were dried out in the flats of the infected families. CONCLUSION: On the basis of circumstantial evidence, fecal aerosol transmission may have caused the community outbreak of COVID-19 in this high-rise building. Thyroid dysfunction plays a role in blood pressure (BP) regulation. However, the associations between thyroid function and BP and arterial stiffness in the general Chinese population without thyroid disease are unknown. This population‐based cross‐sectional study aimed to investigate the association between thyroid function and peripheral and central BP and arterial stiffness in Chinese individuals. After excluding those who had thyroid diseases or incomplete clinical measurements, this study included 691 participants. Of the participants, 444 (64.2%) were women and 215 (31.1%) had hypertension. After adjustment for covariates, serum FT3 was significantly associated with a higher pulse rate in both sexes. In men, each 2.72‐fold increase in serum FT4 levels was associated with higher peripheral systolic BP (+10.82 mmHg, p = .005) and pulse pressure (+5.71 mmHg, p = .03). Each 2.72‐fold increase in serum FT4 levels was associated with higher central systolic BP (+8.03 mmHg, p = .03) and pulse pressure (+3.89 mmHg, p = .05). In women, serum FT4 was only associated with a higher central pulse pressure (+2.96 mmHg, p = .04). After adjustment for covariates, serum FT4 was significantly associated with a faster cfPWV exclusively in men. Our study showed that serum FT4 is associated with higher peripheral and central BP and faster cfPWV in men, whereas serum FT3 is positively associated with a higher pulse rate in both sexes, indicating that the effects of thyroid function on BP and arterial stiffness are more significant in men than in women.

After spinal cord injury, dysregulated miRNAs appear and can participate in inflammatory responses, as well as the inhibition of apoptosis and axon regeneration through multiple pathways. However, the functions of miRNAs in spinal cord ischemia-reperfusion injury progression remain unclear. miRCURY LNATM Arrays were used to analyze miRNA expression profiles of rats after 90 minutes of ischemia followed by reperfusion for 24 and 48 hours. Furthermore, subsequent construction of aberrantly expressed miRNA regulatory patterns involved cell survival, proliferation, and apoptosis. Remarkably, the mitogen-activated protein kinase (MAPK) signaling pathway was the most significantly enriched pathway among 24- and 48-hour groups. Bioinformatics analysis and quantitative reverse transcription polymerase chain reaction confirmed the persistent overexpression of miR-22-3p in both groups. These results suggest that the aberrant miRNA regulatory network is possibly regulated MAPK signaling and continuously affects the physiological and biochemical status of cells, thus participating in the regulation of spinal cord ischemia-reperfusion injury. As such, miR-22-3p may play sustained regulatory roles in spinal cord ischemia-reperfusion injury. All experimental procedures were approved by the Animal Ethics Committee of Jilin University, China [approval No. 2020 (Research) 01].

Multiple successful vaccines against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) are urgently needed to address the ongoing coronavirus disease 2019 (Covid-19) pandemic. In the present work, we describe a subunit vaccine based on the SARS-CoV-2 spike protein coadministered with CpG adjuvant. To enhance the immunogenicity of our formulation, both antigen and adjuvant were encapsulated with our proprietary artificial cell membrane (ACM) polymersome technology. Structurally, ACM polymersomes are self-assembling nanoscale vesicles made up of an amphiphilic block copolymer comprising poly(butadiene)-b-poly(ethylene glycol) and a cationic lipid, 1,2-dioleoyl-3-trimethylammonium-propane. Functionally, ACM polymersomes serve as delivery vehicles that are efficiently taken up by dendritic cells (DC1 and DC2), which are key initiators of the adaptive immune response. Two doses of our formulation elicit robust neutralizing antibody titers in C57BL/6 mice that persist at least 40 days. Furthermore, we confirm the presence of functional memory CD4+ and CD8+ T cells that produce T helper type 1 cytokines. This study is an important step toward the development of an efficacious vaccine in humans.

A novel bacterial strain, designated NS28T, was isolated from interfacial sediment sampled at Taihu Lake, PR China. Cells were rod-shaped, Gram-negative, aerobic and non-motile on Reasoner's 2A medium. The results of phylogenetic analysis based on 16S rRNA gene sequences indicated that strain NS28T was most closely related to species from the genus Dyadobacter , with 98.4 and 96.0 % 16S rRNA gene sequence similarity to its closest phylogenetic neighbours Dyadobacter sediminis CGMCC 1.12895T and Dyadobacter luticola CCTCC AB 2017091T, respectively. MK-7 was the only cellular menaquinone. The major fatty acids were summed feature 3 (C16 : 1ω7c and/or C16 : 1ω6c), iso-C15 : 0 and C16 : 1ω5c. The major polar lipids were phosphatidylethanolamine, one phospholipid, one aminolipid, one lipid and two unidentified lipids. Genomic analysis of strain NS28T indicated that the total genome size was 6 477 094 bp with a G+C content of 44.8 mol%, 5380 protein-coding genes, 79 contigs and an N50 length of 299 584 bp. On the basis of the genomic DNA sequence, the average nucleotide identity values were 90.5 and 74.1 % with D. sediminis CGMCC 1.12895T and D. luticola CCTCC AB 2017091T, respectively. Digital DNA-DNA hybridization results of strain NS28T with D. sediminis CGMCC 1.12895T and D. luticola CCTCC AB 2017091T were 40.9 and 18.6 %, respectively. Based on the phenotypic, chemotaxonomic, phylogenetic and genome sequence data presented here, it is proposed that strain NS28T represents a novel species of the genus Dyadobacter for which the name Dyadobacter flavalbus is proposed . The type strain is NS28T (=NBRC 113854T=MCCC 1K03764T).

Certain mutations in mitochondrial DNA (mtDNA) are associated with Leber's hereditary optic neuropathy (LHON). In particular, the well‑known NADH dehydrogenase 4 (ND4) m.11778G>A mutation is one of the most common LHON‑associated primary mutations worldwide. However, how specific mtDNA mutations, or variants, affect LHON penetrance is not fully understood. The aim of the current study was to explore the relationship between mtDNA mutations and LHON, and to provide useful information for early detection and prevention of this disease. Following the molecular characterization of a Han Chinese family with maternally inherited LHON, four out of eight matrilineal relatives demonstrated varying degrees of both visual impairment and age of onset. Through PCR amplification of mitochondrial genomes and direct Sanger sequencing analysis, a homoplasmic mitochondrial‑encoded ND4 m.11778G>A mutation, alongside a set of genetic variations belonging to human mtDNA haplogroup B5b1 were identified. Among these sequence variants, alanine transfer RNA (tRNA)Ala m.5601C>T was of particular interest. This variant occurred at position 59 in the TψC loop and altered the base pairing, which led to mitochondrial RNA (mt‑RNA) metabolism failure and defects in mitochondrial protein synthesis. Bioinformatics analysis suggested that the m.5601C>T variant altered tRNAAla structure. Therefore, impaired mitochondrial functions caused by the ND4 m.11778G>A mutation may be enhanced by the mt‑tRNAAla m.5601C>T variant. These findings suggested that the tRNAAla m.5601C>T variant might modulate the clinical manifestation of the LHON‑associated primary mutation.

Mild behavioral impairment (MBI), which can include compulsive behavior, is an early sign of Alzheimer's disease (AD), but its underlying neural mechanisms remain unclear. Here, we show that 3–5-month-old APP/PS1 mice display obsessive-compulsive disorder (OCD)-like behavior. The number of parvalbumin-positive (PV) interneurons and level of high gamma (γhigh) oscillation are significantly decreased in the striatum of AD mice. This is accompanied by enhanced β-γhigh coupling and firing rates of putative striatal projection neurons (SPNs), indicating decorrelation between PV interneurons and SPNs. Local field potentials (LFPs) simultaneously recorded in prefrontal cortex (PFC) and striatum (Str) demonstrate a decrease in γhigh-band coherent activity and spike-field coherence in corticostriatal circuits of APP/PS1 mice. Furthermore, levels of GABAB receptor (GABABR), but not GABAA receptor (GABAAR), and glutamatergic receptors, were markedly reduced, in line with presymptomatic AD-related behavioral changes. These findings suggest that MBI occurs as early as 3–5 months in APP/PS1 mice and that altered corticostriatal synchronization may play a role in mediating the behavioral phenotypes observed.

Klotho, an important anti-aging protein, may be related to elevated blood pressure (BP) and arterial stiffness. We aimed to investigate associations between the serum klotho concentration and peripheral/central BP and arterial stiffness based on the carotid–femoral pulse wave velocity (cfPWV) in a Chinese population. We invited all inhabitants aged ≥ 18 years in two Dali communities for participation. The SphygmoCor system was used to record radial arterial waveforms. Aortic waveforms were derived using a generalized transfer function. The central BP was assessed by calibrating the brachial BP, which was measured using an oscillometric device. The serum klotho concentration was measured using an enzyme-linked immunosorbent assay and logarithmically transformed. Of the 716 participants (mean age: 51.9 ± 12.6 years), 467 (65.2%) were women. The median serum klotho concentration was 381.8 pg/mL. The serum klotho concentration did not significantly differ between patients with and without hypertension (P > 0.05) and between those with and without arterial stiffness (cfPWV ≥ 10 m/s) (P > 0.05). After adjusting for confounders, the serum klotho concentration was not significantly associated with the peripheral or central BP (P > 0.05) and cfPWV (P > 0.05). Our data indicated that the serum klotho concentration was not associated with BP or cfPWV in the general Chinese population.

Additionally, numerical model has been validated and shows the good agreement with wind tunnel experimental data, indicating that the APFoam has sufficient ability to study urban turbulence and pollutant dispersion characteristics. By applying the APFoam, O3–NOx–VOCs formation processes and dispersion of the reactive pollutants are analyzed in an example of typical street canyon (aspect ratio H / W = 1). Chemistry mechanism comparison shows that O3 and NO2 are underestimated while NO is overestimated if the VOCs reactions are not considered in the simulation. Polycystic ovary syndrome (PCOS) is a common endocrine disorder with unknown etiology and unsatisfactory clinical treatment. Considering the ethical limitations of studies involving humans, animal models that reflect features of PCOS and insulin resistance (IR) are crucial resources in investigating this syndrome. Our previous study showed that mitochondrial dysfunction resulted from pathogenic mutations of mitochondrial DNA (mtDNA), and that oxidative stress had an active role in the phenotypic manifestation of PCOS‑IR. Therefore, it was hypothesized that limiting oxidative stress and mitochondrial damage may be useful and effective for the clinical treatment of PCOS‑IR. For this purpose, the present study examined the therapeutic effects of the mitochondria‑targeted antioxidant MitoQ10 for PCOS‑IR. Furthermore, the histopathology was used to analysis the ovarian morphological changes. The endocrine and reproductive related parameters were analyzed by ELISA approach. A PCOS‑IR model was successfully established by subcutaneous injection of rats with testosterone propionate and feeding a high‑fat diet. The 30 female Sprague‑Dawley rats were then divided into three groups, comprising a control (n=10), animal model (PCOS‑IR, n=10) and MitoQ10 treatment (n=10) group. It was found that MitoQ10 significantly improved the IR condition and reversed the endocrine and reproductive conditions of PCOS. In addition, the impaired mitochondrial functions were improved following MitoQ10 administration. Notably, western blot results suggested that this antioxidant reduced the expression levels of apoptosis‑related proteins cytochrome c and B‑cell lymphoma‑2 (Bcl‑2)‑associated X protein, whereas the anti‑apoptotic protein Bcl‑extra large was increased following MitoQ10 treatment. Taken together, the data indicated that the MitoQ10 may have a beneficial favorable therapeutic effect on animals with PCOS‑IR, most likely via the protection of mitochondrial functions and regulation of programmed cell death‑related proteins.

We describe a batch-extraction with simulated digestive fluid (salivary fluid, gastric fluid and intestinal fluid) to estimate the bioaccessibility of inhaled trace metals (TMs) in particulate matter less than 10 and 2.5 μm in aerodynamic diameter (PM10 and PM2.5). Concentrations of the assayed TMs (As, Cd, Cr, Ni, Mn, Cu, Zn, Sb, Hg and Pb) were determined in PM10 and PM2.5 samples by inductively coupled plasma-mass spectrometry. The TMs with the largest soluble fractions for airborne PM collected from winter and summer in saliva were Mn and Sb, respectively; in seasons this became Co in gastric fluid and Cu in intestinal fluid. Clearly, bioaccessibility is strongly dependent on particle size, the component of simulated digestive fluids (e.g., pH, digestive enzymes pepsin and trypsin), and the chemical properties of metal ions. The particle size and seasonal variation affected the inhaled bioaccessible fraction of PM-bound TMs during mucociliary clearance, which transported PM from the tracheal and the bronchial region to the digestive system. This study provides direct evidence for TMs in airborne PM being bioaccessible TMs are likely to possess an enhanced digestive toxic potential due to airborne PM pollution.

This is one of a limited number of studies that investigates multimedia exposure to metal(loid)s and the related human health risks caused by atmospheric deposition in China and Asia. The aggregate multimedia human exposure included the accidental ingestion and dermal adsorption of soil and surface water, as well as the intake of grains, fruits, vegetables, meats and eggs, milk and dairy products, and freshwater fish. The values and variabilities of atmospheric deposition and exposure factors were identified as two major uncertainty sources. The annual variations in wet and bulk deposition were propagated from the variabilities of particulate matter (PM) × precipitation and the linear regressed coefficients of flux~PM × precipitation, and the annual variation in dry deposition was propagated by subtracting the wet deposition from the bulk deposition. The variabilities in the exposure factors were updated based on the latest published regional data. Long-term atmospheric deposition was identified as a vital source of Cd contamination in surface soil and freshwater bodies and As accumulation in freshwater fish. The noncarcinogenic (NC) risk probabilities caused by Cd were the highest, followed by As. The overall uncertainties related to the NC risk probabilities were much lower for both adults and 6- to 17-year-old children/adolescents, while those related to the carcinogenic risk (CR) probabilities were much lower for 2- to 5-year-old children and 6- to 17-year-old children/adolescents. Our results could help infer the cobenefits due to the current regional air pollution control policy. CD38 is involved in the adenosine pathway, which represents one of the immunosuppressive mechanisms in cancer. CD38 is broadly expressed across immune cell subsets, including human macrophages differentiated in vitro from monocytes, but expression by tissue-resident macrophages remains to be demonstrated. We analysed tissue samples obtained from 66 patients with hepatocellular carcinoma (HCC) from Singapore and analysed using immunohistochemistry. Tumor-infiltrating leukocytes (TILs) were further examined using DEPArrayTM., and the phenotype of freshly isolated TILs was determined using flow cytometry. It is found that CD38 was frequently co-expressed with the macrophage-specific marker CD68. CD38+CD68+ macrophage density was associated with improved prognosis after surgery, while total CD68+ macrophage density was associated with poor prognosis. DEPArrayTM. analysis revealed the presence of large (>10 µm), irregularly shaped CD45+CD14+ cells that resembled macrophages, with concurrent CD38+ expression. Flow cytometry also revealed that majority of CD14+HLA-DR+ cells expressed CD38. In conclusion, CD38 expression was clearly demonstrated on human macrophages in an in vivo setting. The positive association identified between CD38+ macrophage density and prognosis may have implications for routine diagnostic work.

Cancer is among the most serious diseases characterized by uncontrollable cell growth and spread of abnormal cells. Cancerous cells form tumors that negatively impact the functions of the body, inducing serious malfunctioning leading to fatalities in most cases. Up to now, the effective diagnosis and treatments of cancer have remained a big challenge. Nanotechnology is an emerging field encompassing science, engineering and medicine, which has attracted great attention for cancer therapy in recent years. Among the numerous nanomaterials, mesoporous silica nanoparticles (MSNs) have attracted great attention and are being considered as promising biomedical materials for the development of cancer therapies because of their size tunability, surface functionality, optically transparent properties, low toxicity and high drug loading efficiency. In this review, we first outline the properties and structure of different configurations of MSNs and their subsequent application in the field of cancer theranostics. Thereafter, the potential of MSNs as multifunctional delivery platforms for therapeutic agents and their significance in next generation cancer therapy is discussed.

People are exposed to multiple stimuli in urban environments, but most studies have investigated the unimodal effect of thermal and visual conditions on human comfort perception. It remains unclear whether the cross-modal effect found in indoor multisensory studies applies to outdoor environments. To understand the cross-modal effect of thermal and visual conditions on outdoor comfort perception, we conducted a thermal comfort survey (n = 4304) in Guangzhou and Zhuhai (September 2018). We used the Universal Thermal Climate Index (UTCI) heat stress classification and sky conditions to stratify our results. The thermal sensation vote was positively correlated with sun sensation vote. There was a significant interaction between UTCI heat stress conditions and sunlight preference vote on thermal comfort vote. The sun sensation (brightness) and sunlight preference vote had a cross-modal effect on thermal sensation and thermal comfort vote under various UTCI heat stress conditions. Under extreme heat stress, respondents’ thermal sensation did not differ significantly between different sun sensation and sunlight preference groups. Thermal sensation, preference and comfort vote had a cross-modal effect on sun sensation and sunlight preference under different sky conditions. Under partly cloudy conditions, sun sensation did not differ significantly between certain thermal sensation and preference groups. A theoretical framework is provided to explain the cross-modal effect between thermal and visual perception. Our findings suggest outdoor thermal discomfort can be alleviated by improving visual comfort and vice versa. Therefore, urban design should consider the combined effect of visual-thermal stimulants in optimizing overall pedestrian comfort and promoting urban liveability.

Background Previous research has reported the effects of long-term fine particulate matter (PM2.5) pollution on years of life lost (YLL), but these effects may not represent the full impact. This study aims to estimate potential benefits in life time from adhering to daily ambient PM2.5 concentration standards/guidelines. Methods This study evaluated the relationship between daily ambient PM2.5 level and YLL using a two-stage approach with generalized additive models and meta-analysis. Potential life expectancy gains were then estimated by presuming that daily PM2.5 levels were in compliance with the Chinese and WHO standards. In addition, the attributable fraction of YLL due to excess PM2.5 exposure was also calculated. Results During 2013–2016, 459,468 non-accidental deaths were recorded in the six cities of Pearl River Delta, China. Each 10 μg/m3 increment in four-day average (lag03) level of PM2.5 was related to an increment of 13.31 [95% confidence interval (CI): 5.74, 20.87] years of life lost. Implementation of the WHO guidelines might avoid 180,980.83 YLLs (95% CI: 78,116.07, 283,845.60), which corresponded to 0.39 (95% CI: 0.17, 0.62) years of increased life time per death. Additionally, an estimated 0.15% (95% CI: 0.06%, 0.23%) or 2.04% (95% CI: 0.88%, 3.20%) of YLLs could be attributed to PM2.5 exposures higher than the Chinese or WHO guidelines, respectively. Conclusions This study suggests that people might live longer by controlling daily PM2.5 concentration and highlights the need to adopt stricter standards in China.

The severity of urban air pollution problems in China increases with the city's size and population density. Outdoor air pollution not only affects indoor air quality but also increases building energy consumption. We analyzed the characteristics of urban air pollution in China based on long-period monitoring data with high temporal resolution (hourly) recorded between September 1, 2014 and August 31, 2019. We compared five key regions in detail: “2 + 26 cities,” the Yangtze River delta (YRD), the Pearl River delta (PRD), the Northeast China Plain (NECP), and the Sichuan Basin (SCB). The results show decreases in the annual mean concentrations of particulate matter, PM2.5 and PM10, SO2, and CO but an increase in the O3 concentration. The increasing O3 rate in the 2 + 26 cities region was nearly 14 times that of the global rate. In terms of diurnal variation, NO2 and CO reached maxima between 8:00 and 9:00 a.m. due to morning rush hour traffic, which was approximately 1 h before the PMs and SO2 reached their maxima. In the monthly variation profiles, the SO2 concentration magnitudes in the NECP were similar to those of the 2 + 26 cities during winter heating periods and the YRD region during the non-heating periods. Minima associated with “plum rain” and the rainy season were also observed in the O3 monthly variation profiles of the YRD and SCB regions, respectively. In the 2 + 26 cities region, each pollutant showed significantly large temporal variation relative to the spatial variation, which is indicative of the regional air pollution trend.

Background:Mutations in mitochondrial tRNA (mt-tRNA) genes have been found to be associated with both syndromic and non-syndromic hearing impairment. However, the pathophysiology underlying mt-tRNA mutations in clinical expression of hearing loss remains poorly understood.Objective:The aim of this study was to explore the potential association between mttRNA mutations and hearing loss.Methods and Results:We reported here the molecular features of a pedigree with maternally transmitted non-syndromic hearing loss. Among 12 matrilineal relatives, five of them suffered variable degree of hearing impairment, but none of them had any medical history of using aminoglycosides antibiotics (AmAn). Genetic screening of the complete mitochondrial genomes from the matrilineal relatives identified the coexistence of mt-tRNAHis G12192A and mt-tRNAThr G15927A mutations, together with a set of polymorphisms belonging to human mitochondrial haplogroup B5b1b. Interestingly, the G12192A mutation occurred 2-bp from the 3’ end of the TψC loop of mt-tRNAHis, which was evolutionarily conserved from various species. In addition, the well-known G15927A mutation, which disrupted the highly conserved C-G base-pairing at the anticodon stem of mt-tRNAThr, may lead to the failure in mt-tRNA metabolism.:Furthermore, a significant decreased in ATP production and an increased ROS generation were observed in polymononuclear leukocytes (PMNs) which were isolated from the deaf patients carrying these mt-tRNA mutations, suggested that the G12192A and G15927A mutations may cause mitochondrial dysfunction that was responsible for deafness. However, the absence of any functional mutations/variants in GJB2, GJB3, GJB6 and TRMU genes suggested that the nuclear genes may not play important roles in the clinical expression of non-syndromic hearing loss in this family.Conclusion:Our data indicated that mt-tRNAHis G12192A mutation may increase the penetrance and expressivity of deafness-associated m-tRNAThr G15927A mutation in this family.

A semi-empirical multilayer urban canopy model is developed to estimate the vertical dispersion of traffic emissions in high density urban areas. It is motivated by the heterogeneity of urban morphology in real urban cities and the need of quick urban design and planning. The urban canopy is divided into multiple layers, to include the impact of building height variance on pollutant dispersion. The model is derived by mass conservation within each layer through adopting a box model. To validate the model, results in several cases with uniform and non-uniform building height distributions are compared with CFD simulations. The validation study indicates that the assumption of zero pollutant concentration over the modeled canopy and no horizontal pollutant transfer has increasingly negligible influence with increasing urban densities. The new multilayer model performs well to model the vertical pollutant transport, and modelling results can mostly follow the trend of the CFD simulations. The present paper conducts two case studies in metropolitan areas in Singapore and Hong Kong to illustrate how to implement this multilayer urban canopy model in the planning practice. With an in-house GIS team using available data, the multilayer model provides planners a way to understand air pollutant dispersion in high-density urban areas.

Rationale: Despite growing use of engineered nanomaterials (ENM) in applications from electronics to medicine, the potential risk to human health remains a critical concern within clinical use. ENM exposure during pregnancy can potentially cause reproductive toxicity even at levels that produce no measurable harm to animals in normal conditions. Methods: Phospholipid micelle-encapsulated CdSe/CdS/ZnS semiconductor nanocrystals with an average hydrodynamic diameter of 60 nm were intravenously injected during pregnancy in both rodent and nonhuman primate animal models. Cadmium concentration levels and maternal haematological and biochemical markers were determined, along with histopathological examination of major organs. Results: Nanocrystals were found to have crossed the placenta from mother to fetus in both rodents and nonhuman primates. However, the animal models display different responses with respect to reproductive toxicity. In the rodent model, toxicity symptoms are absent in treated subjects, with no observed gestational or fetal abnormalities and complications. A significantly higher miscarriage rate of 60% is recorded for macaques after prenatal nanoparticle administration. There was a miscarriage rate of 15% in the general population despite only ~0.16% of the initial cadmium dose present in the fetus. Blood and biochemical markers of treated macaques indicate acute hepatocellular injury within a week after nanoparticle administration. Histology of major organs of the miscarried macaque fetuses show no abnormalities. Conclusion: The potential of nanomaterials to cross the placenta and impact fetal survival in primates suggest the necessity of precautionary measures to prevent gestational exposure of ENMs.

Despite 70 years of research that has intensified in the past decade, a safe and efficacious dengue vaccine has yet to be available. In addition to the expected challenges such as identifying immune correlates of protection, the dengue vaccine field has faced additional hurdles including the necessity to design a tetravalent formulation and the risk of antibody-mediated disease enhancement. Nevertheless, tetravalent live attenuated vaccine candidates have reached efficacy trials and demonstrated some benefit, despite imbalanced immunogenicity and incomplete protection against the four serotypes. Meanwhile, the development of sub-unit dengue vaccines has gained momentum. As the target of most of the neutralizing antibodies so far reported, the virus envelope E protein has been the focus of much effort and represents the leading dengue sub-unit vaccine candidate. However, its notorious poor immunogenicity has prompted the development of innovative approaches to make E-derived constructs part of the second generation dengue vaccines portfolio.

Substantial evidence has linked short-term exposure to ambient fine particulate matter (PM2.5) with increased cardiovascular mortality, however, the specific chemical constituent and emission source responsible for this effect remained largely unclear. A time series Poisson model was employed to quantify the association of cardiovascular mortality with two sets of shipping pollution emission: nickel (Ni), vanadium (V) (the indices of shipping emission) and estimated shipping emission using a source apportionment approach in Guangzhou, China in 2014. We observed that Ni, V, and estimated shipping emission in PM2.5 were associated with increased cardiovascular mortality, an inter-quartile range (IQR) increase in lag2 Ni was associated with 4.60% (95% CI: 0.14%, 9.26%) increase in overall cardiovascular mortality, and 13.35% (95% CI: 5.54%, 21.75%) increase in cerebrovascular mortality; each IQR increase of lag1 V was correlated with 6.01% (95% CI: 1.83%, 10.37%) increase in overall cardiovascular mortality, and 11.02% (95% CI: 3.15%, 19.49%) increase in cerebrovascular mortality; and each IQR increase in lag1 shipping emission was associated with 5.55% (95% CI: 0.78%, 10.54%) increase in overall cardiovascular mortality, and 10.39% (95% CI: 1.43%, 20.14%) increase in cerebrovascular mortality. The results remained robust to adjustment for PM2.5 mass and gaseous air pollutants. This study suggests that shipping emission is an important detrimental factor of cardiovascular mortality, and should be emphasized in air pollution control and management in order to protect the public health in Guangzhou, China.

Background The obesity-hypertension pathogenesis is complex. From the phenotype to molecular mechanism, there is a long way to clarify the mechanism. To explore the association between obesity and hypertension, we correlate the phenotypes such as the waist circumference (WC), body mass index (BMI), systolic blood pressure (SB), and diastolic blood pressure (DB) with the clinical laboratory data between four specific Chinese adult physical examination groups (newly diagnosed untreated just-obesity group, newly diagnosed untreated obesity-hypertension group, newly diagnosed untreated just-hypertension group, and normal healthy group), and the results may show something. Objective To explore the mechanisms from obesity to hypertension by analyzing the correlations and differences between WC, BMI, SB, DB, and other clinical laboratory data indices in four specific Chinese adult physical examination groups. Methods This cross-sectional study was conducted from September 2012 to July 2014, and 153 adult subjects, 34 women and 119 men, from 21 to 69 years, were taken from four characteristic Chinese adult physical examination groups (newly diagnosed untreated just-obesity group, newly diagnosed untreated obesity-hypertension group, newly diagnosed untreated just-hypertension group, and normal healthy group). The study was approved by the ethics committee of Hangzhou Center for Disease Control and Prevention. WC, BMI, SB, DB, and other clinical laboratory data were collected and analyzed by SPSS. Results Serum levels of albumin (ALB)，alanine aminotransferase (ALT), low density lipoprotein cholesterol (LDLC), triglyceride (TG), high density lipoprotein cholesterol (HDLC), alkaline phosphatase (ALP), uric acid (Ua), and TC/HDLC (odds ratio) were statistically significantly different between the four groups. WC statistically significantly positively correlated with BMI, ALT, Ua, and serum levels of glucose (GLU), and TC/HDLC, and negatively with ALB, HDLC, and serum levels of conjugated bilirubin (CB). BMI was statistically significantly positively related to ALT, Ua, LDLC, WC, and TC/HDLC, and negatively to ALB, HDLC, and CB. DB statistically significantly positively correlated with ALP, BMI, and WC. SB was statistically significantly positively related to LDLC, GLU, serum levels of fructosamine (FA), serum levels of the total protein (TC), BMI, and WC. Conclusion The negative body effects of obesity are comprehensive. Obesity may lead to hypertension through multiple ways by different percents. Background: The linkage between ambient fine particle pollution (PM2.5) and chronic obstructive pulmonary disease (COPD) and the attributable risk remained largely unknown. This study determined the cross-sectional association between ambient PM2.5 and prevalence of COPD among adults ≥50 years of age. Methods: We surveyed 29,290 participants aged 50 years and above in this study. The annual average concentrations of PM2.5 derived from satellite data were used as the exposure indicator. A mixed effect model was applied to determine the associations and the burden of COPD attributable to PM2.5. Results: Among the participants, 1872 (6.39%) were classified as COPD cases. Our analysis observed a threshold concentration of 30 μg/m3 in the PM2.5-COPD association, above which we found a linear positive exposure-response association between ambient PM2.5 and COPD. The odds ratio (OR) for each 10 μg/m3 increase in ambient PM2.5 was 1.21(95% CI: 1.13, 1.30). Stratified analyses suggested that males, older subjects (65 years and older) and those with lower education attainment might be the vulnerable subpopulations. We further estimated that about 13.79% (95% CI: 7.82%, 21.62%) of the COPD cases could be attributable to PM2.5 levels higher than 30 μg/m3 in the study population. Conclusion: Our analysis indicates that ambient PM2.5 exposure could increase the risk of COPD and accounts for a substantial fraction of COPD among the study population. A novel actinobacterial strain, designated X17T, was isolated from the sediment of Taihu Lake in China and its taxonomic position was investigated by using a polyphasic approach. The isolate formed milky-white colonies comprising aerobic, Gram-stain-positive, rod-shaped cells. Phylogenetic analysis based on the 16S rRNA gene sequences revealed that the organism belonged to the genus Nocardioides and consistently formed a distinct cluster with Nocardioides agariphilus JCM 16020T and Nocardioides islandensis MSL 26T, sharing 95.5 and 94.8 % 16S rRNA gene sequence similarities, respectively. The genomic DNA G+C content was 69.9 mol%. Chemotaxonomically, the isolate contained ll-diaminopimelic acid in the cell-wall peptidoglycan, MK-8 (H4) as the predominant menaquinone, and diphosphatidylglycerol, phosphatidylglycerol and phosphatidylinositol as the polar lipids found in the cell wall. The major fatty acids were 18 : 1 ω9c (38.6 %), 16 : 0 iso (20.3 %), 15 : 0 iso (6.8 %) and 18 : 0 (5.8 %). Based on its physiological, biochemical and chemotaxonomic characteristics, the strain represents a novel species of the genus Nocardioides , for which the name Nocardioides taihuensis sp. nov. (type strain X17T=CGMCC 4.7318T=NBRC 112321T) is proposed.

Home environment is suggested to be an essential influencing factor for asthma development or asthma exacerbation. However, the association between home environment and childhood asthma among preschool children in the hot humid subtropical region in South China is unclear. We investigated doctor-diagnosed asthma in preschool children due to the parental perception of odors in homes. A total of 1946 children aged from 3 to 6 years from 22 kindergartens in Foshan were randomly recruited for this study in May 2016. The associations among childhood asthma, indoor environment and parental perceptions of odors were examined by using logistic regression model in terms of odds ratio (OR) and 95% confidence interval (CI). Parental atopy had the highest risk for childhood asthma with odds ratio = 6.007 (95% CI: 2.369–15.231). Parental perception of dry air significantly increased asthma with odds ratio = 1.284 (95% CI: 1.027–1.606). The parental perceptions of odors were strongly related with new furniture at home. We conclude that parental perception of dry air is a risk factor for the increase of childhood asthma in the present study. Hence, our study indicates a need to control the household humidity level and reduce the usage of new furniture, in order to reduce the risk of childhood asthma in South China.

Uniformly sized synephrine molecularly imprinted polymer (MIP) microparticles were prepared via precipitation polymerization. The presence of the template, the amount of monomer, crosslinker, and porogenic solvent could affect the size distribution and morphology of the polymers. The pseudo-second-order kinetic model could provide a better correlation for the adsorption than the pseudo-first-order model. The intraparticle diffusion study showed that the sorption involved intraparticle diffusion, but that was not the only rate-controlling step. The equilibrium data better fit the Freundlich model than the Langmuir model. The Scatchard plot of MIPs revealed MIPs had two groups of sites with different affinities. Following central nervous system injury, axonal sprouts form distal to the injury site and extend into the denervated area, reconstructing neural circuits through neural plasticity. How to facilitate this plasticity has become the key to the success of central nervous system repair. It remains controversial whether fine motor skill training contributes to the recovery of neurological function after spinal cord injury. Therefore, we established a rat model of unilateral corticospinal tract injury using a pyramidal tract cutting method. Horizontal ladder crawling and food ball grasping training procedures were conducted 2 weeks before injury and 3 days after injury. The neurological function of rat forelimbs was assessed at 1, 2, 3, 4, and 6 weeks after injury. Axon growth was observed with biotinylated dextran amine anterograde tracing in the healthy corticospinal tract of the denervated area at different time periods. Our results demonstrate that compared with untrained rats, functional recovery was better in the forelimbs and forepaws of trained rats. The number of axons and the expression of growth associated protein 43 were increased at the injury site 3 weeks after corticospinal tract injury. These findings confirm that fine motor skill training promotes central nervous system plasticity in spinal cord injury rats.

Recently, an inverse role for Wnt signaling in the development of osteoclasts in the bone was demonstrated. In the present study, we examined whether there is a commonality in the mechanism of bone resorption and lysis that occur in a diverse set of bone metastatic lesions, as well as in primary bone lesions. Compared with control bone tissue and bone biopsies from patients with nonmetastatic primary tumors (i.e., breast carcinoma, lung adenocarcinoma, and prostate carcinoma), patients with bone metastatic lesions from the three aforementioned primary tumors, as well as osteolytic lesions obtained from the bone biopsies of patients with multiple myeloma, demonstrated an upregulated expression of the glycoprotein Dickkopf-1 at both the mRNA and protein levels. Additionally, by coimmunoprecipitation, Dickkopf-1 pulled-down low-density lipoprotein receptor-related protein 6 (Lrp6), which is a key downstream effector of the Wnt signaling pathway. The expression of Lrp6 was unaltered in the osteometastatic lesions. This negative regulation was associated with a lowered expression of osteoprotegerin in the osteometastatic lesions, an observation that was previously reported to promote osteoclastogenesis. These findings provide a common mechanism for the inverse relationship between the Wnt signaling pathway and the development of primary or metastatic bone lesions. Pharmacological modulation of the Wnt signaling pathway might benefit the clinical management of primary and metastatic bone lesions.

Plant growth regulating substances are involved in the physiological and metabolic processes of plants and enable them to cope with numerous environmental stresses. The effect of exogenously applied brassinolide (BR) with various concentrations (0.01, 0.1, and 1.0 mg L -1 ) was studied on morphological and physiological traits of Leymus chinensis (Trin.) Tzvelev under room and high temperatures in pots. The experimental results revealed that high temperature stress substantially perturbed growth, photosynthetic pigments, and root activity of L. chinensis; however, the deleterious effects of high temperature were partially ameliorated by the foliar application of BR. Compared to room temperature, high temperature stress decreased the plant height, leaf area, plant fresh and dry weight, chlorophyll a and b content, chlorophyll a/b ratio as well as root activity, while exacerbated the membrane damage as indicated by enhanced production of malondialdehyde (MDA). Accumulation of proline content, soluble protein and sugar content in L. chinensis improved by heat stress, compared with normal temperature; application of BR further improved their production thus aiding in the attainment of tolerance against heat stress. Elevated levels of antioxidant enzymes such as superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), ascorbate peroxidase (APX), and glutathione reductase (GR) were observed under heat stress compared to room temperature, however, application of BR further proved beneficial in this regard. Our results indicated that BR could improve the growth and development of L. chinensis by enhancing the biosynthesis of photosynthetic pigments, osmolytes and antioxidant enzymes system in plants under both room and high temperature.

The bacterial isolate Paracoccus sp. strain FLN-7 hydrolyzes amide pesticides such as diflubenzuron, propanil, chlorpropham, and dimethoate through amide bond cleavage. A gene, ampA , encoding a novel arylamidase that catalyzes the amide bond cleavage in the amide pesticides was cloned from the strain. ampA contains a 1,395-bp open reading frame that encodes a 465-amino-acid protein. AmpA was expressed in Escherichia coli BL21 and homogenously purified using Ni-nitrilotriacetic acid affinity chromatography. AmpA is a homodimer with an isoelectric point of 5.4. AmpA displays maximum enzymatic activity at 40°C and a pH of between 7.5 and 8.0, and it is very stable at pHs ranging from 5.5 to 10.0 and at temperatures up to 50°C. AmpA efficiently hydrolyzes a variety of secondary amine compounds such as propanil, 4-acetaminophenol, propham, chlorpropham, dimethoate, and omethoate. The most suitable substrate is propanil, with K m and k cat values of 29.5 μM and 49.2 s −1 , respectively. The benzoylurea insecticides (diflubenzuron and hexaflumuron) are also hydrolyzed but at low efficiencies. No cofactor is needed for the hydrolysis activity. AmpA shares low identities with reported arylamidases (less than 23%), forms a distinct lineage from closely related arylamidases in the phylogenetic tree, and has different biochemical characteristics and catalytic kinetics with related arylamidases. The results in the present study suggest that AmpA is a good candidate for the study of the mechanism for amide pesticide hydrolysis, genetic engineering of amide herbicide-resistant crops, and bioremediation of amide pesticide-contaminated environments.

De-esterification is an important degradation or detoxification mechanism of sulfonylurea herbicide in microbes and plants. However, the biochemical and molecular mechanisms of sulfonylurea herbicide de-esterification are still unknown. In this study, a novel esterase gene, sulE , responsible for sulfonylurea herbicide de-esterification, was cloned from Hansschlegelia zhihuaiae S113. The gene contained an open reading frame of 1,194 bp, and a putative signal peptide at the N terminal was identified with a predicted cleavage site between Ala37 and Glu38, resulting in a 361-residue mature protein. SulE minus the signal peptide was synthesized in Escherichia coli BL21 and purified to homogeneity. SulE catalyzed the de-esterification of a variety of sulfonylurea herbicides that gave rise to the corresponding herbicidally inactive parent acid and exhibited the highest catalytic efficiency toward thifensulfuron-methyl. SulE was a dimer without the requirement of a cofactor. The activity of the enzyme was completely inhibited by Ag + , Cd 2+ , Zn 2+ , methamidophos, and sodium dodecyl sulfate. A sulE -disrupted mutant strain, Δ sulE , was constructed by insertion mutation. Δ sulE lost the de-esterification ability and was more sensitive to the herbicides than the wild type of strain S113, suggesting that sulE played a vital role in the sulfonylurea herbicide resistance of the strain. The transfer of sulE into Saccharomyces cerevisiae BY4741 conferred on it the ability to de-esterify sulfonylurea herbicides and increased its resistance to the herbicides. This study has provided an excellent candidate for the mechanistic study of sulfonylurea herbicide metabolism and detoxification through de-esterification, construction of sulfonylurea herbicide-resistant transgenic crops, and bioremediation of sulfonylurea herbicide-contaminated environments.

By means of computational fluid dynamic (CFD) simulations, this paper investigated how rural winds transport relatively clean air into high-rise urban areas to dilute airborne pollutants. Two ventilation concepts for indoor environments were applied: the age of air to quantify the time taken by rural young air to reach a given place after it enters an urban area and the air exchange efficiency to evaluate the efficiency of rural winds flushing the entire urban canopy layer. Several square building arrays with street aspect ratios (building height/street width, H / W ) ranging from 1.5 to 5.3 and building area densities of 0.25 (medium) or 0.4 (compact) were considered as the approaching wind is parallel to the main street whose length varies from street scales to neighborhood scales (330–510 m to 1.03–1.65 km in full-scale). Results show that considerable young rural air enters windward entries but a major fraction of air is vertically driven out as flowing deeper into such high-rise building arrays. So air exchange efficiencies are less than 50% in street-scale arrays, and smaller in longer (neighborhood-scale) or narrower arrays. For the neighborhood-scale medium arrays, considering the power-law velocity profile in the upstream free flow, a taller array gains a larger inflow rate across its windward entry and experiences younger air and greater air exchange efficiency than a lower one. If all buildings are theoretically open-based in a neighborhood-scale compact array, air becomes much younger everywhere and the air exchange efficiency doubles. In arrays of buildings with different heights, the secondary streets in front of taller buildings get younger air due to the downward flows within them. Although further investigations are still required before providing a practical framework, this paper is one of the first attempts to find ways in improving the ventilation performance in high-rise cities like Hong Kong.

A yellow-coloured bacterium, T41T, was isolated from a soil sample of a subtropical rainforest in Nepal. Cells were Gram-reaction-positive, aerobic, non-motile, short rods. Phylogenetic analysis based on 16S rRNA gene sequences indicated that the strain formed a cluster with Terrimonas ferruginea, Terrimonas lutea, Niabella soli, Flavisolibacter ginsengiterrae, Flavisolibacter ginsengisoli, Niastella yeongjuensis and Niastella koreensis in the phylum Bacteroidetes. The strain showed the highest sequence similarity to the type strain of Terrimonas lutea (93.2 %). The major isoprenoid quinone was MK-7 and the predominant cellular fatty acids (>10 %) were iso-15 : 0 (33.8 %), iso-15 : 1 G (13.3 %) and iso-17 : 0 3-OH (12.9 %). The DNA G+C content was 48.1 mol%. On the basis of phenotypic and phylogenetic data and genomic distinctiveness, strain T41T represents a novel species in a new genus in the phylum Bacteroidetes, for which the name Flavihumibacter petaseus gen. nov., sp. nov. is proposed. The type strain of Flavihumibacter petaseus is strain T41T (=CGMCC 1.7723T =NBRC 106054T).

Phenol and nitrate are two major pollutants simultaneously occurring in several industrial wastewaters. In this study, a 110-day gradual enrichment of an anaerobic culture has been carried out at 25°C in an anaerobic bioreactor for continuously treating a synthetic wastewater containing 600 mg∕L phenol and 430 mg∕L NO3− –N . The results showed that the enriched culture can utilize phenol as a sole electron donor and nitrate as a sole electron acceptor. At the end of the enrichment (on Day 110), 93.3% of phenol and 98.0% of NO3− –N were simultaneously removed at a hydraulic retention time of 20.25 h in the anaerobic bioreactor. The removal of 1 g NO3− –N required about 3.19 g chemical oxygen demand as the electron donor. Batch tests further revealed that cresol, nitrophenol, and monochlorinated phenol (MCP) could exert detrimental influences on the treatment abilities of the enriched culture. In this paper, a three-dimensional model of pulsatile blood flow througth a vessel with an asymmetric stenosis is developed to numerically study the hemodynamics circumstance under EECP (Enhanced External Counter Pul- sation). The governing equations are the usual Navier-Stokes equations for unsteady elliptic flows, and are numerically solved by using finite volume method. The numerical computing is based on the animal experiment. The blood flow rate curves in a cadiac cycle pre-EECP and post-EECP are detected in a pig's carotid respectively, the counterpulsation pressure is 0.03MPa. The blood flow rate curves are transformed to blood velocity distributions along the arterial diameter by using the Womersley arithmetic. And the Womersley velocity profiles are used as inflow boundary conditions to simulate the whole hemodynamics circumstance inside the 3-dimensional artery model. The computational results show that when EECP is applied, the blood flow rate and the wall shear stress level of the artery are increased in a cadiac cycle, as well as the pulsatile character of blood flow and wall shear stress. Which are possibly the hemodynamic actors that lead to the improving of the endothelial function which is thought to be straightly relating to the atherosclerosis.

Hepatocytes are anchorage-dependent cells sensitive to microenvironment; the control of the physicochemical properties of the extra-cellular matrices may be useful to the maintenance of hepatocyte functions in vitro for various applications. In a microcapsule-based 3-D hepatocyte culture microenvironment, we could control the physical properties of the collagen nano-fibres by fine-tuning the complex-coacervation reaction between methylated collagen and terpolymer of hydroxylethyl methacrylate-methyl methacrylate-methylacrylic acid. The physical properties of the nano-fibres were quantitatively characterized using back-scattering confocal microscopy to help optimize the physical support for hepatocyte functions. We further enhanced the chemical properties of the collagen nano-fibres by incorporating galactose onto collagen, which can specifically interact with the asialoglycoprotein receptor on hepatocytes. By correlating a range of collagen nano-fibres of different physicochemical properties with hepatocyte functions, we have identified a specific combination of methylated and galactosylated collagen nano-fibres optimal for maintaining hepatocyte functions in vitro. A model of how the physical and chemical supports interplay to maintain hepatocyte functions is discussed.

The use of MEMS implantable drug delivery pump device enables one to program the desired drug delivery profile in the device for individualized medicine treatment to patients. In this study, a MEMS drug delivery device is prepared and employed for in vivo applications. 12 devices are implanted subcutaneously into Kunming mice for evaluating their long term biocompatibility and drug-delivery efficiency in vivo. All the mice survived after device implantation surgery procedures. Histological analysis result reveals a normal wound healing progression within the tissues-to-device contact areas. Serum analysis shows that all measured factors are within normal ranges and do not indicate any adverse responses associated with the implanted device. Phenylephrine formulation is chosen and delivered to the abdominal cavity of the mice by using either the implanted MEMS device (experimental group) or the syringe injection method (control group). Both groups show that they are able to precisely control and manipulate the increment rate of blood pressure in the small animals. Our result strongly suggests that the developed refillable implantable MEMS devices will serve as a viable option for future individualized medicine applications such as glaucoma, HIV-dementia and diabetes therapy.

Cephalexin was synthesized in aqueous two-phase systems (ATPSs) by using penicillin G acylase (PGA) (penicillin amidohydrolase, EC3.5.1.11) as a catalyst and 7-aminodeacetoxicephalosporanic acid (7-ADCA) and phenylglycine methyl ester (PGME) as substrates. In the ATPS composed of 20% (w/w) PEG 400 and 15% (w/w) magnesium sulfate, the partition coefficient of PGA (KE) was less than 0.01, PGA biased to the magnesium sulfate-rich bottom phase. The partition coefficient of cephalexin (KC) was 6.7, and the partition coefficients of PGME (KP) and 7-ADCA (KA) were 1.5 and 1.2, respectively. The stability of PGA in 20% (w/w) PEG 400 and 15% (w/w) magnesium sulfate ATPS was investigated. The activity of PGA was remained 80.6% after PGA was retained in the ATPS for 35 h and reserved 59% in the 20% magnesium sulfate solution at pH 7.0. When free enzyme was used for bioconversion, a high yield of cephalexin (60%) was achieved at the concentration of 100 mM 7-ADCA and 100 mM PGME in this ATPS than that of in an aqueous system (21%).

Essential hypertension (EH) is a frequent, chronic, age-related disorder, which remains a major modifiable risk factor for cardiovascular disease despite important advances in our understanding of its pathophysiology. Previous studies have noted a consistent maternal effect on blood pressure (BP). Consequently, mutations in mitochondrial DNA (mtDNA) have become an additional target of investigations on the missing BP heritability. Among these mutations, mt-transfer RNA (tRNA) is a hot mutational spot for pathogenic mutations associated with EH. Mutant mtDNA aggravates mitochondrial dysfunction, pivotally contributing to the clinical phenotype. Moreover, the damaged mitochondria, due to their inability to provide the high-energy requirements for cells, generate reactive oxygen species (ROS) and induce mitochondrial-mediated cell death pathways. Therefore, mitochondrial dysfunction plays a critical role in the pathogenesis of EH. This review summarizes the basic knowledge of mitochondrial genetics and EH-associated mtDNA mutations and further discusses the molecular mechanisms behind these mtDNA mutations in clinical manifestations of EH.

Individualized disease diagnosis and therapy has emerged as a new direction in the research of future medication. Over the past several years, innovative approaches based on microelectromechanical system (MEMS) technology have demonstrated promising potential in individualized therapy. In this contribution, a sustainable approach for the individualized treatment of chronic disease is presented using a compact, implantable and refillable MEMS drug delivery device with an electrolysis based actuator. As a demonstration, we utilized the device for programmable delivery of a chemotherapy drug for the treatment of pancreatic cancer with an in vitro configuration based on cancer cell colonies. After the delivery of drug using the device, the growth of the colonies has been greatly inhibited as compared with the control samples. These results show that our new approach has a great potential for future in vivo studies and opens up promising opportunities for future medication.
