The catfishes are a group of economically important freshwater fish in China, which in recent years have suffered heavy losses as a result of bacterial outbreaks. In this study, we examined the diversity of the microbiome of infected skin mucus of yellow catfish (Pelteobagrus fulvidraco) and channel catfish (Ictalurus punctatus) and analyzed the bacterial pathogens. We found several common pathogenic bacteria, such as Aeromonas spp., Vibrio spp., Moraxella spp. and Proteus spp. present in both fish species, but with significantly different bacterial community structures. We isolated and cultured Aeromonas veronii and Proteus mirabilis and validated their infectivity in zebrafish (Danio rerio) and yellow catfish. Intraperitoneal injection of either bacteria into zebrafish, but not immersion, caused 100% mortality and ovary fragmentation. Yellow catfish was more sensitive than zebrafish with 100% mortality in the immersion challenges with A. veronii or P. mirabilis, and with a higher abundance of A. veronii in the P. mirabilis-challenged group compared to control. To our knowledge, this is the first study on the pathogenicity of P. mirabilis in yellow catfish, and the results will help to develop effective strategies for the disease prevention and control in catfish farming.

This study aimed to determine the bioavailability and pharmacokinetic parameters of sulfadiazine (SDZ) in channel catfish (Ictalurus punctatus) following oral gavage and intravenous injection. The healthy channel catfish were orally and intravenously administrated with SDZ solution at doses of 50 and 5 mg/kg, respectively. Plasma samples were determined by ultra-performance liquid chromatography with an ultraviolet detector. The results demonstrated that the concentration-time profile of SDZ after oral dosing was best described by a one-compartmental open model with first-order absorption. The absorption half-life (t1/2Kα ), the elimination half-life (t1/2Ke ), and the area under concentration-time profile (AUC0-∞ ) were estimated to be 0.87 h, 29.04 h, and 1311.72 mg.h/L, respectively. After intravenous administration, the concentration-time curve of SDZ conformed to a two-compartmental open model without absorption. The distribution half-life (t1/2α ), the elimination half-life (t1/2β ), the apparent distribution volume (Vss ), the total clearance (CL), and AUC0-∞ were calculated to be 0.19 h, 14.24 h, 0.36 L/kg, 0.018 L/h/kg, and 277.12 mg.h/L, respectively. Finally, the bioavailability was estimated to be 47.33%. This study will provide some useful information for the modification of the dosage form of SDZ in aquaculture, and is partly beneficial for appropriate use of SDZ in the future.

The channel catfish (Ictalurus punctatus, Rafinesque) ovary (CCO) cell line is the standard cell line used for channel catfish diagnostics. Next-gen sequencing studies of a virus cultured in the CCO cells revealed mitochondrial sequences matching those of brown bullhead (Ameiurus nebulosus, Lesueur). Therefore, we systematically performed partial cytochrome oxidase 1 gene sequencing of several sources of the CCO cell line and all matched the brown bullhead and not the channel catfish.

Cathelicidins are a class of antimicrobial peptides (AMPs) known to possess rapid and direct antimicrobial activities against a variety of microorganisms. Recently identified cathelicidins derived from alligator and sea snake were found to be more effective in inhibiting microbial growth than other AMPs previously characterized. The ability of these two cathelicidins along with the peptides, cecropin and pleurocidin, to protect channel catfish (Ictalurus punctatus, Rafinesque) and hybrid catfish (I. punctatus ♀ × blue catfish, Ictalurus furcatus, Valenciennes ♂) against Edwardsiella ictaluri, one of the most prevalent pathogens affecting commercial catfish industry, was investigated. Cathelicidin-injected fish (50 µg ml-1 fish-1 ) that were simultaneously challenged with E. ictaluri through bath immersion at a concentration of ~1 × 106 CFU/ml had increased survival rates compared with other peptide treatments and the infected control. Bacterial numbers were also reduced in the liver and kidney of channel catfish and hybrid catfish in the cathelicidin treatments 24 hr post-infection. After 8 days of challenge, serum was collected to determine immune-related parameters such as bactericidal activity, lysozyme, serum protein, albumin and globulin. These immune-related parameters were significantly elevated in fish injected with the two cathelicidins as compared to other peptide treatments. These results indicate that cathelicidins derived from alligator and sea snake can stimulate immunity and enhance the resistance to E. ictaluri infection in channel catfish and hybrid catfish.

The aim of this study was to evaluate the in vitro and in vivo efficacy of propolis against Ligictaluridus floridanus on Ictalurus punctatus gills. This study used an ethanol extract of propolis at a concentration of 300 mg mL−1, with a total polyphenol concentration of 2.77 mg mL−1 as determined by chromatography. In vitro trial used propolis concentrations at 5, 30 as well as 60 mg L–1 and a 70% ethanol control group to treat naturally infected catfish. The in vivo experiment was 16 weeks in duration, consisting of seventy-two fish infected by cohabitation and subjected to one-hour baths every 72 h during the trial. Catfish Ictalurus spp. are subjected to stressful conditions during harvest, which may be linked to fillet coloration and quality. Poor water quality in ponds, socks or hauling tanks, as well as handling stress, have been suggested to cause red fillets in catfish; however, chronic exposure has not resulted in red fillets. Short‐term occurrences of extreme poor water quality, particularly low dissolved oxygen, high carbon dioxide and high temperature, may occur in ponds or during harvest. Therefore, market‐sized Channel Catfish Ictalurus punctatus were acutely exposed (12 hr) to one of the three water quality treatments while confined during a simulated socking procedure and evaluated for stress responses by means of change in blood parameters and fillet quality. In fish subjected to the extreme treatment, hematocrit, plasma cortisol, glucose and lactate levels increased, with 22% mortality, indicating highly stressful conditions. In fish subjected to moderate and typical (control) treatments, cortisol increased but a lack of change or decrease in glucose and lactate indicated minimal anaerobic metabolism. Only one red fillet was produced by the extreme treatment and two by the typical treatment; therefore, the results suggest red fillets are not a product of poor water quality compounded by handling during harvest.

This study investigated the effects of dietary lysolecithin on growth performance, feed utilization and metabolic responses of channel catfish (Ictalurus punctatus). Fish (initial weight: 14.77 ± 0.45 g) were randomly fed one of five practical diets supplemented with graded levels of lysolecithin (0, 125, 250, 375 and 500 mg/kg) in quadruplicate groups for 10 weeks. Results showed that no significant differences in weight gain, condition factor and viscerosomatic index among all the treatments. Fish fed diet supplemented with 250 mg/kg lysolecithin had significantly decreased feed conversion ratio and significantly increased protein efficiency ratio compared to that fed the control diet. The group with 250 mg/kg of dietary lysolecithin had higher protein and lower lipid contents in the whole body, and lower lipid content in the liver than those in the control group. Groups supplemented with 250–500 mg/kg of dietary lysolecithin had significantly higher gastric and intestinal lipase activities than the control group. Dietary lysolecithin at the level of 250 mg/kg significantly increased the activities of Na⁺, K⁺‐ATPase, alkaline phosphatase, total antioxidative capacity, total superoxide dismutase, glutathione peroxidase and catalase and significantly decreased the content of malondialdehyde compared to the control diet. In conclusion, dietary supplementation of lysolecithin could confer benefits of feed utilization, body composition and antioxidative capacity of channel catfish.

A study was conducted in earthen ponds to investigate the effects of available lysine (AL) concentrations in 28 and 32% protein diets on production and processing characteristics, proximate composition, and lysine concentrations in the fillet of channel catfish, Ictalurus punctatus. Diets were formulated to contain 28% protein with 1.22 and 1.43% AL, and 32% protein with 1.43 and 1.63% AL, which were equivalent to 4.37, 5.1, 4.46, and 5.1% AL of protein, respectively. Fingerlings with a mean initial weight of 32 g/fish were stocked into 20 ponds (0.04 ha) at 19,760 fish/ha. Fish were fed once daily to apparent satiation for 181 days. No significant differences were observed for total diet fed, net yield, weight gain, survival, or fillet proximate composition among dietary treatments. However, the 28% protein with low AL diet (1.22% AL of diet or 4.37% AL of protein) resulted in significantly lower carcass and fillet yield and fillet lysine level compared with fish fed the 28% protein diet with 1.43% AL (5.1% AL of protein) and 32% protein diets with 1.43 and 1.63% AL (4.47 and 5.1% AL of protein). Results suggest 1.43% AL of diet is adequate for both 28 and 32% protein diets for optimum growth, processing yield, and lysine retention in fillets for pond‐raised channel catfish.

The Channel Catfish Ictalurus punctatus is an important ecological, recreational and aquaculture species. In both natural and pond culture systems, Channel Catfish are periodically exposed to acute increase in temperature, especially in the summer, which may induce a stress response. With the effects of global climate change, the impact of rising temperatures on aquatic systems is expected to be exacerbated in the future. Hence, there is the need to understand the physiological mechanisms controlling the thermal stress response in fish. Acid–base, haematological, osmotic and metabolic regulations were investigated following acute temperature increase from 28°C to either 32°C or 36°C at a rate of 2°C/hr to assess the physiological response of Channel Catfish to temperature increase. Blood was sampled 1 hr after treatment temperatures were reached. When temperature increased from 28 to 32°C, blood bicarbonate (HCO3-) decreased while cortisol, partial pressure of carbon dioxide (pCO₂) and glucose increased. When temperature increased from 28 to 36°C, blood pH, HCO3-, chloride, total proteins and mean corpuscular haemoglobin concentration decreased while cortisol, pCO₂, haematocrit, haemoglobin, glucose, lactate, mean corpuscular volume and mean corpuscular haemoglobin all increased. The changes observed are characteristic of acute stress responses in fish and suggest that acute temperature increases to both 32 and 36°C are physiologically stressful to Channel Catfish.

Cortisol is a glucocorticoid hormone which is an endocrine signaling molecule in all vertebrates and acts through intracellular glucocorticoid receptors (GRs). Cortisol affects many biological functions including immunity, stress, growth, and reproduction. The objective of this study was to investigate the ontogeny of the cortisol and GR stress response in channel catfish (Ictalurus punctatus) at several early life stages. To accomplish this, resting and stress-induced levels of tissue cortisol and the two catfish GRs (GR-1 and GR-2) expression were measured. Resting cortisol levels in newly fertilized eggs averaged 2.4 ± 0.2 ng/egg and decreased to 0.4 ± 0.01 ng/egg by day 5. Cortisol levels in newly fertilized eggs subjected to an acute stress (lowered dissolved oxygen from 6.5 mg/L to 1.8 mg/L) averaged 2.3 ± 0.1 ng/egg and decreased to 0.3 ± 0.03 ng/egg by day 5. At hatching, resting cortisol levels were 24 ± 1.0 ng/0.1 g tissue while levels increased to 83 ± 2.0 ng/0.1 g tissue in fry subjected to an acute stress (P < .05). Four days post-hatch, resting cortisol levels were 83 ± 1.0 ng/0.1 g tissue while levels increased to 149 ± 4.0 ng/0.1 g tissue in fry subjected to an acute stress (P < .01). There was no significant difference between GR-1 and GR-2 mRNA in stressed and unstressed newly hatched fry. Four days post-hatch, GR-1 mRNA increased 3-fold while GR-2 mRNA increased 2-fold in fry that were subjected to low dissolved oxygen conditions (P < .05). These results indicate that cortisol biosynthesis, integration and maturation of the hypothalamic-pituitary-interrenal (HPI) axis can be observed in channel catfish at hatching. The upregulation of GR-1 and -2 mRNA in stressed fry supports roles for both transcripts in integrating the channel catfish stress response.

Henneguya adiposa is one of ten known, closely related myxozoan species that parasitize a variety of tissue sites in the channel catfish, Ictalurus punctatus. Reported to specifically target the adipose fin, H. adiposa is not associated with morbidity or mortality, although detailed descriptions of its associated histologic pathology are lacking. The objective of this work was to confirm the presence of H. adiposa within fin lesions of affected channel catfish using DNA sequenced from histologic sections obtained by laser capture microdissection, as well as to describe pathologic changes induced by infection. The parasite formed large, white, elongate, nodular plasmodia that caused localized tissue damage and incited a granulomatous inflammatory response within a deep connective tissue layer at the base of the adipose fin. Myxospores released from ruptured plasmodia into adjacent tissue were observed to migrate superficially in tracts through the skin, indicating a portal of exit for environmental dispersal. Defects in the connective tissue layer created by ruptured plasmodia were infiltrated by granulomatous inflammation and fibroplasia, suggesting lesion resolution by scar formation over time. Sequencing of the 18S rRNA gene amplified from excised myxospores confirmed the myxozoan's identity as H. adiposa, with 100% similarity to the reference sequence from previous published work.

Aquaculture offers great potential for fish production in Lake Chapala, but reports of heavy metal contamination in fish have identified a main concern for this activity. In the present study, cultures of the species Cyprinus carpio and Ictalurus punctatus were grown in a net cage in Lake Chapala. The patterns of heavy metal accumulation (Cu, Zn, Cd, Hg, Pb, As) in muscle and liver were monitored in order to evaluate the level of metal incorporation in the fish. Estimates of weekly metal intake (EWI) were made based on the results of the concentrations in edible parts of fish of commercial size. The patterns of metal bioaccumulation between tissues and species showed that liver had a higher concentrating capacity for Zn, Cu, Cd, and Pb. In contrast, similar concentrations of Hg and As were found in the liver and muscle tissue. According to the EWI estimates, the heavy metals in these cultured fish do not represent a risk for human consumption.

One of the major goals in aquaculture is to protect fish against infectious diseases as disease outbreaks could lead to economic losses if not controlled. Antimicrobial peptides (AMPs), a class of highly conserved peptides known to possess direct antimicrobial activities against invading pathogens, were evaluated for their ability to protect Channel Catfish Ictalurus punctatus and hybrid catfish (female Channel Catfish × male Blue Catfish I. furcatus) against infection caused by the fish pathogen Aeromonas hydrophila ML09-119. To identify effective peptides, the minimum inhibitory concentrations against bacterial pathogens Edwardsiella ictaluri S97-773, Edwardsiella piscicida E22-10, A. hydrophila ML09-119, Aeromonas veronii 03X03876, and Flavobacterium columnare GL-001 were determined in vitro. In general and overall, cathelicidins derived from alligator and sea snake exhibited more potent and rapid antimicrobial activities against the tested catfish pathogens as compared to cecropin and pleurocidin AMPs and ampicillin, the antibiotic control. When the peptides (2.5 µg of peptide/g of fish) were injected into fish and simultaneously challenged with A. hydrophila through immersion, increased survival rates in Channel Catfish and hybrid catfish were observed in both cathelicidin (alligator and sea snake) treatments as compared to other peptides and the infected control (P < 0.001) with alligator cathelicidin being the overall best treatment. Bacterial numbers in the kidney and liver of Channel Catfish and hybrid catfish also decreased (P < 0.05) for cathelicidin-injected groups at 24 and 48 h after challenge infection. These results show the potential of cathelicidin to protect catfish against bacterial infections and suggest that an approach overexpressing the peptide in transgenic fish, which is the long-term goal of this research program, may provide a method of decreasing bacterial disease problems in catfish as delivering the peptides via individual injection or feeding would not be economically feasible.

Enteritis has resulted in large economic losses in channel catfish aquaculture. Yet only scarce information is available on Plesiomonas shigelloides as a causal agent for this disease. In this study, a virulent strain, temporarily named BD1, was isolated from diseased channel catfish suffering from enteritis, and was identified as P. shigelloides through molecular and phenotypic methods. A phylogenetic tree was also constructed to determine its taxonomic position. In addition, the BD1 isolate has developed multiple resistances to aminoglycosides, amphenicols, quinolones, tetracyclines and sulfonamides drugs for veterinary uses in aquaculture as revealed when screened against a range of common antibiotics. To the best of our knowledge, this is the first report of P. shigelloides as a potential pathogen of enteritis in channel catfish.

Some fishes rely on large regions of the dorsal (epaxial) and ventral (hypaxial) body muscles to power suction feeding. Epaxial and hypaxial muscles are known to act as motors, powering rapid mouth expansion by shortening to elevate the neurocranium and retract the pectoral girdle, respectively. However, some species, like catfishes, use little cranial elevation. Are these fishes instead using the epaxial muscles to forcefully anchor the head, and if so, are they limited to lower-power strikes? We used X-ray imaging to measure epaxial and hypaxial length dynamics (fluoromicrometry) and associated skeletal motions (XROMM) during 24 suction feeding strikes from three channel catfish (Ictalurus punctatus). We also estimated the power required for suction feeding from oral pressure and dynamic endocast volume measurements. Background: A large proportion of Nigerians consume fish as the source of protein in their meals. This may be attributed to health factors, preference and affordability for low income earners. The incidence of Aeromonas hydrophila in fresh catfish may constitute a significant health risk to the consumer if there is a horizontal transfer to man as it has been reported to be pathogenic. This study examined the possibility of fresh water catfish being a reservoir of pathogenic Aeromonas hydrophila. Method: Aeromonas hydrophila was isolated from the different organs of fresh water catfish (Clarias gariepinus and Ictalurus punctatus) obtained from Kporoko river in Lokoja. Aeromonas hydrophila was identified using both phenotypic and genotypic methods. The pathogenic traits of the Aeromonas species such as biofilm formation, production of haemolysin, enterotoxin and enzymes were determined. Results: Aeromonas hydrophila occurred in all the examined fish organs (fish, liver, kidney, skin and gut) of Clarias gariepinus but occurred only in the skin, intestine, kidney and gut of Ictalurus punctatus examined, but the incidence of Aeromonas hydrophila was prevalent in the gut of all the fishes analyzed. All the Aeromonas isolates analysed in this study produced biofilm, haemolysins and lipase enzymes. They also produced enterotoxins with values ranging between 0.069 - 1.138. Conclusion: The occurrence of Aeromonas in fresh catfish possessing these pathogenic traits is of great public health significance to man as it indicates the likelihood of man being predisposed to toxigenicity when the toxin concentration reaches a lethal value. It is therefore recommended that the internal organs of fresh catfish be thoroughly cleaned and cooked before consumption.

Affinity maturation of the antibody response, a process of antibody affinity increasing over response, is one of the key features of the mammalian immune system. However, the process is incompletely understood in teleost, including channel catfish (Ictalurus punctaus). In this study, IgM affinity maturation in channel catfish was investigated by estimating the kinetics of antibody affinity using ELISA and ELISPOT assays. Fish were immunized with a T-cell dependent antigen (TNP-KLH), and individual serum IgM antibody titers and affinities, and IgM+ antibody-secreting cells (ASCs) in peripheral blood were analyzed over a period of 14 weeks. A detectable serum anti-TNP response developed by 2-weeks post-immunization, and the maximal antibody production was observed by 6-weeks post-immunization. The average affinity of anti-TNP serum antibody increased consistently and reached the maximum by 10-weeks post-immunization. The increase of antibody affinity beyond the point of optimal antibody titer revealed that the affinity maturation of IgM antibody response occurred in channel catfish. Dissection of dynamics of individual affinity subpopulations indicated that a significant proportion of low affinity subpopulations appeared at early response, and high affinity subpopulations appeared predominantly at later, resulting in a 100-fold increase in affinity over response. Additional, TNP+ IgM+ ASCs was detected by 2-weeks post-immunization and achieved the maximal number by 6-weeks post-immunization. Using an inhibition ELISPOT assay, the findings of a consistent increase in the average affinity of secreted IgM antibody by peripheral blood ASCs, as the immune response progressed, confirmed the occurrence of the affinity maturation. Taken together, the results of this study indicated that affinity maturation occurred in channel catfish following immunization with a TD antigen TNP-KLH.

In this study, we evaluated different dietary fishmeal and protein levels on growth performance, intestinal structure and intestinal microbial community of juvenile channel catfish, Ictalurus punctatus. A total of 1800 fish distributed into 36 tanks were fed with nine different diets containing three protein levels (300, 330 and 360 g/kg) with three fishmeal (FM) levels (0, 30 and 60 g/kg) for 90 days. The results showed that significant interactions between the protein level and FM level were observed in final weight (FW), weight gain (WG), Na⁺, K⁺‐ATPase and alkaline phosphatase (AKP) activities. The significant lowest FW, WG, Na⁺, K⁺‐ATPase and AKP activities were observed in fish fed with no fishmeal and 300 g/kg protein dietary while the highest were shown in 60 g/kg fishmeal and 330 g/kg protein treatment. Additionally, the microvillar length of the mid‐intestine in catfish was significantly affected by the interaction between dietary protein level and fishmeal level. The intestinal samples were dominated by three major phyla, Firmicutes, Proteobacteria and Fusobacteria. Genera Romboutsia and Turicibacter accounted for probably 800 g/kg of the phylum Firmicutes; meanwhile, genus Cetobacterium represented more than 900 g/kg of the phylum Fusobacteria. In conclusion, this study indicated that channel catfish juveniles can be fed with a practical diet without fishmeal as long as the protein level increased to 360 g/kg; however, if the percentage of dietary protein was 300 g/kg, it seemed that fishmeal need to be supplied as a protein source.

Channel catfish is the leading aquaculture species in the US, and one of the reasons for its application in aquaculture is its relatively high tolerance against hypoxia. However, hypoxia can still cause huge economic losses to the catfish industry. Studies on hypoxia tolerance, therefore, are important for aquaculture. Fish swimbladder has been considered as an accessory respiration organ surrounded by a dense capillary countercurrent exchange system. In this regard, we conducted RNA-Seq analysis with swimbladder samples of catfish under hypoxic and normal conditions to determine if swimbladder was responsive to low oxygen treatment and to reveal genes, their expression patterns, and pathways involved in hypoxia responses in catfish. A total of 155 differentially expressed genes (DEGs) were identified from swimbladder of adult catfish, whereas a total of 2,127 DEGs were identified from swimbladder of fingerling catfish under hypoxic condition as compared with untreated controls. Subsequent pathway analysis revealed that many DEGs under hypoxia were involved in HIF signaling pathway ( nos2, eno2, camk2d2, prkcb, cdkn1a, eno1, and tfrc), MAPK signaling pathway (voltage-dependent calcium channel subunit genes), PI3K/Akt/mTOR signaling pathway ( itga6, g6pc, and cdkn1a), Ras signaling pathway ( efna3 and ksr2), and signaling by VEGF ( fn1, wasf3, and hspb1) in catfish swimbladder. This study provided insights into regulation of gene expression and their involved gene pathways in catfish swimbladder in response to low oxygen stresses.

There is a trend toward the increased incidence and prevalence of Edwardsiella piscicida septicemia in US catfish aquaculture, particularly in channel ♀, Ictalurus punctatus, × blue ♂, I. furcatus, hybrid catfish. From 2013 to 2017, a total of 3242 disease case submissions were made to the Aquatic Research and Diagnostic Laboratory (ARDL) at the Thad Cochran National Warmwater Aquaculture Center in Stoneville, MS. Of these, 1400 (43.2%) were hybrids. E. piscicida was suspected in 138 (4.3%) of cases, the majority of which (89.1%) were from hybrid catfish. A molecular survey of these isolates confirmed the majority (92.0%) to be E. piscicida. Furthermore, cases of E. piscicida from hybrids submitted to the ARDL and the Aquatic Diagnostic Laboratory of the Mississippi State University College of Veterinary Medicine in Starkville, MS, were documented for gross lesions and histological analysis. Grossly, E. piscicida presents with small dermal ulcerations, a raised fluid‐filled cranial midline lesion that is frequently ulcerated, hemorrhage in the gills, exophthalmia, and abdominal distension. Internally, lesions include splenomegaly, straw‐colored ascites, renomegaly, and occasionally hemorrhagic intestines. Histopathological examination is in agreement with gross observations, and infected fish repeatedly demonstrate a mononuclear meningoencephalitis, hemorrhagic branchitis, splenitis, ulcerative dermatitis, granulomatous interstitial nephritis, and hepatitis coupled with a hemorrhagic enteritis.

Repressible knockdown approaches were investigated to manipulate for transgenic sterilization in channel catfish, Ictalurus punctatus. Two primordial germ cell (PGC) marker genes, nanos and dead end, were targeted for knockdown and an off-target gene, vasa, was monitored. Two potentially copper-sensitive repressible promoters, yeast ctr3 (M) and ctr3-reduced (Mctr), were coupled with four knockdown strategies separately including: ds-sh RNA targeting the 5′ end (N1) or 3′ end (N2) of channel catfish nanos, full-length cDNA sequence of channel catfish nanos for overexpression (cDNA), and ds-sh RNA-targeting channel catfish dead end (DND). Each construct had an untreated group and treated group with copper sulfate as the repressor compound. Spawning rates of full-sibling P1 fish exposed or not exposed to the constructs as treated and untreated embryos were 85 and 54%, respectively, indicating potential sterilization of fish and repression of the constructs. In F1 fish, mRNA expressions of PGC marker genes for most constructs were downregulated in the untreated group and the knockdown was repressed in the treated group. Gonad development in transgenic, untreated F1 channel catfish was reduced compared to non-transgenic fish for MctrN2, MN1, MN2, and MDND. For 3-year-old adults, gonad size in the transgenic untreated group was 93.4% smaller than the non-transgenic group for females and 92.3% for males. However, mean body weight of transgenic females (781.8 g) and males (883.8 g) was smaller than of non-transgenic counterparts (984.2 and 1254.3 g) at 3 years of age, a 25.8 and 41.9% difference for females and males, respectively. The results indicate that repressible transgenic sterilization is feasible for reproductive control of fish, but negative pleiotropic effects can result.

As the ethanol industry matures and adopts new technologies, the co-products produced are also changing with their composition adjusted to meet feed industry needs. High protein distillers dried grain with yeast (HP40Y) is a relatively new product which is a variant of distillers dried grains that could be used as an improved protein source in catfish feed formulations. To evaluate the efficacy of HP40Y, a 10-weeks growth trial was conducted on the growth performance of juvenile catfish, Ictalurus punctatus (mean initial weight 1.80 ± 0.05 g). In the growth trial, graded levels of HP40Y (0.00, 3.10, 6.20 and 9.30%) were used to replace poultry meal (PM: 6.0, 4.0, 2.0 and 0.0%) and other series of diets were used with HP40Y (5.0, 10.0, 15.0, 20.0, 30.0 and 40.0%) to replace soybean meal (SBM: 51.00, 46.49, 41.90, 37.40, 28.20, 19.20%). Two channel catfish glucocorticoid receptor genes, ipGR1 (NR3C1_1) and ipGR2 (NR3C1_2) were partially characterized. Identification and analysis of the genomic organization of two channel catfish glucocorticoid (GC) receptors (GRs) revealed differences in the lengths of exons 1 and 2 and the addition of an extra 27-bp exon inserted after exon 2 in the GR1 gene, yielding a 9-aa insert in the receptor protein. Sequence of the 9-aa insert in ipGR1 (WRARQNTHG) is unique compared to other teleost fish GRs. Amino acid sequence alignment of the two channel catfish GRs, revealed 55% sequence identity between them, with a high degree of sequence conservation (82%) in the DNA binding and ligand binding domains. Real-time PCR indicated that ipGR1 and ipGR2 were expressed in all tissues evaluated. Channel catfish GR1 was predominantly expressed in the gills, nearly 25-fold higher than in the liver. GR1 expression was higher than GR2 expression in gills, intestine, head kidney and heart (P0.05). Channel catfish hepatic GR1 mRNA expression was significantly (P0.05) increased from pre-stress expression 30min following removal of the acute stressor. After 30min of stress and during the 2h recovery period, ipGR1 mRNA expression was higher relative to ipGR2 expression. Hepatic ipGR2 expression was not affected (P0.05) by the acute stress event. The present study adds to the growing body of information on GR evolution and function and further demonstrates the unique regulation of the GC/GR system in teleost fish.

An understanding of factors responsible for population structure including the origins of individuals from among habitats is fundamental to conservation and management of large-river fishes. The prevalence of population mixing of channel catfish Ictalurus punctatus was evaluated within a large-river tributary environment using information from recent environmental history and natal origin derived from otolith microchemistry. Trace elements in water and otoliths were assessed using univariate and multivariate statistical approaches. Water and otolith trace elements differed among river segments facilitating classification of channel catfish to the river segment of capture. Accuracy of the classification tree model for juvenile channel catfish ranged from 44% to 88%. Recent environmental and natal origin microchemistry signatures suggested the channel catfish population within a large-river tributary comprises individuals from multiple locations. Population demographics of channel catfish is likely influenced by mixing of individuals from across the riverine-network. Consideration of the importance of connectivity between main-stem and tributary systems may, therefore, benefit conservation and management of channel catfish and other large-river fishes displaying similar life-history strategies.

An emerging pathotype of Aeromonas hydrophila (vAh) has been responsible for widespread farm losses in the US catfish industry over the last decade. While our genetic and biochemical understanding of vAh has been greatly enhanced in this time frame, our ability to reliably induce the disease in the laboratory has remained limited. Taking cues from observed farm conditions associated with outbreaks, here we perturbed iron scavenging dynamics and catfish feeding status. Addition of a xenosiderophore, deferoxamine mesylate (DFO), to vAh cultures prior to immersion challenge significantly increased virulence in several vAh isolates but not in a non-epidemic strain. DFO addition did not impact vAh growth dynamics or perturb iron-sensitive gene pathways, but did significantly enhance hemolysis of catfish blood. Furthermore, hours between last feeding and immersion challenge (postprandial status), was observed to be a critical determinant of catfish susceptibility. Fish with a full gastrointestinal tract had significantly lower survival than those in a fasted state, and this effect was cumulative with that of DFO-enhanced vAh virulence. Taken together, our results not only provide a more robust challenge model, they offer actionable insights into pond level host-pathogen-environmental interactions potentially underlying vAh pathogenesis.

Water hardness above the optimal level can incite toxic effects in fish, which are often species specific. Hence, we aimed at obtaining insights on the potential effects of elevated water hardness as well as coping strategies in channel catfish (Ictalurus punctatus). First, a toxicity assay was performed where the 96 h-LC50 was calculated as 4939 mg/L CaCO3. Thereafter, to gain knowledge on the underlying adaptive strategies to high water hardness, fish were exposed to seven hardness levels (150, 600, 1000, 1500, 2000, 3000 and 4000 mg/L CaCO3 at pH 8.15) for 15 days. Results showed that branchial activities of Ca2+-ATPase and Na+/K+-ATPase, which facilitate Ca2+ uptake, reduced starting respectively from 1000 mg/L and 1500 mg/L CaCO3. Nevertheless, Ca2+ burden in plasma and tissue (gills, liver and intestine) remained elevated. Hardness exposure also disturbed cations (Na+, K+, Mg2+) and minerals (iron and phosphorus) homeostasis in a tissue-specific and dose-dependent manner. Both hemoglobin content and hematocrit dropped significantly at 3000–4000 mg/L CaCO3, with a parallel decline in iron content in plasma and gills. Muscle water content rose dramatically at 4000 mg/L CaCO3, indicating an osmo-regulation disruption. Higher hardness of 3000–4000 mg/L CaCO3 also incited a series of histopathological modifications in gills, liver and intestine; most likely due to excess Ca2+ accumulation. Overall, these data suggest that channel catfish can adapt to a wide range of elevated hardness by modulating Ca2+ regulatory pathways and histomorphological alterations, however, 1500 mg/L CaCO3 and above can impair the performance of this species.

Microcystins (MCs) are hepatotoxic cyanobacterial metabolites produced sporadically in aquatic environments under favorable environmental conditions. Affinity of these toxins to covalently bind with protein phosphatases poses a challenge in their detection. Lemieux oxidation to release 2-methyl-3-methoxy-4-phenylbutyric acid (MMPB), a common moiety to all MCs congeners, has been used in detection of these compounds, however a lack of sensitivity has limited the usefulness of the method. In this study, modifications of the oxidation and solid phase extraction procedures, combined with a sensitive LC/MS/MS (liquid chromatography/mass spectrometry) detection, have resulted in 25 ng/g method detection limits in both liver and plasma samples. Samples harvested from six fingerling channel catfish ( Ictalurus punctatus) dosed intraperitoneally with a sublethal MC-LR dose of 250 μg/kg were analyzed, and microcystin concentrations ranging from 370 to 670 ng/g in plasma and 566–1030 ng/g in liver were detected. Similarly, 250 μg/kg nodularin-dosed channel catfish fish were found to contain 835–1520 ng/g in plasma and 933–1140 ng/g in liver. Detection of the toxins in serum and liver combined with the presence of histopathological lesions consistent with these hepatocellular toxin in exposed fish and no positive findings in the control fish demonstrates the usefulness of this analytical procedure for the diagnosis of suspected algal toxicity cases.

The present study investigated the replacement of soybean meal with combinations of two or three alternative protein sources in diets for pond-raised hybrid catfish, ♀ Ictalurus punctatus × ♂ Ictalurus furcatus. Alternative protein sources evaluated included cottonseed meal, distillers dried grains with solubles (DDGS), peanut meal, and porcine meat and bone meal (PMBM). Hybrid catfish fingerlings with a mean initial weight of 35 g/fish were stocked into 25 earthen ponds (0.04 ha) at a density of 14,826 fish/ha. Fish were fed once daily to apparent satiation for 166 d. No significant differences were observed for total diet fed, net yield, weight gain, survival, carcass yield, fillet yield, or fillet proximate composition among dietary treatments. Results show soybean meal may be completely replaced by combinations of cottonseed meal and one or two other alternative protein sources including DDGS, peanut meal, and PMBM in the diet without markedly affecting production and processing characteristics and fillet proximate composition of pond-raised hybrid catfish. These alternative diets may be used during foodfish production when prices are favorable.

The present study was conducted to determine the minimum effective dose of salmon gonadotropin-releasing hormone analog (sGnRHa) to stimulate ovulation in Channel Catfish Ictalurus punctatus. Four doses of sGnRHa—0, 10, 15, and 25 µg/kg body weight (BW)—were compared with the commonly used dose of 100 µg/kg BW of mammalian luteinizing hormone releasing hormone analog (mLHRHa) to induce spawning in Channel Catfish. Hormone-induced catfish yielded significantly higher levels of plasma estradiol and ovulation success than saline-injected control fish. Ovulation ratio, relative fecundity, and number of fry per kilogram with 10 µg sGnRHa/kg were lower than those treated with 15 and 25 µg. A virulent clonal population of Aeromonas hydrophila (VAh) is recognized as the etiological agent in outbreaks of motile aeromonas septicemia (MAS) in catfish aquaculture in the southeastern United States since 2009. Genomic subtraction revealed three outer membrane proteins present in VAh strain ML09-119 but not in low virulence reference A. hydrophila strains: major outer membrane protein OmpA1, TonB-dependent receptor (Tdr), and transferrin-binding protein A (TbpA). Here, the genes encoding ompA1, tdr, and tbpA were cloned from A. hydrophila ML09-119 and expressed in Escherichia coli. The purified recombinant OmpA1, Tdr, and TbpA proteins had estimated molecular weights of 37.26, 78.55, and 41.67 kDa, respectively. Catfish fingerlings vaccinated with OmpA1, Tdr, and TbpA emulsified with non-mineral oil adjuvant were protected against subsequent VAh strain ML09-119 infection with 98.59%, 95.59%, and 47.89% relative percent survival (RPS), respectively. Furthermore, the mean liver, spleen, and anterior kidney bacterial concentrations were significantly lower in catfish vaccinated with the OmpA1 and Tdr than the sham-vaccinated control group. ELISA demonstrated that catfish immunized with OmpA1, Tdr, and TbpA produce significant antibody response by 21 days post-immunization. Therefore, OmpA1 and Tdr proteins could be used as potential candidates for vaccine development against virulent A. hydrophila infection. However, TbpA protein failed to provide strong protection.

Flavobacterium columnare, the causative agent of columnaris disease, causes substantial mortality worldwide in numerous freshwater finfish species. Due to its global significance and impact on the aquaculture industry continual efforts to better understand basic mechanisms that contribute to disease are urgently needed. The current work sought to evaluate the effect of L-rhamnose on the growth characteristics of F. columnare. While we initially did not observe any key changes during the total growth of F. columnare isolates tested when treated with L-rhamnose, it soon became apparent that the difference lies in the ability of this carbohydrate to facilitate the formation of biofilms. The addition of different concentrations of L-rhamnose consistently promoted the development of biofilms among different F. columnare isolates; however, it does not appear to be sufficient as a sole carbon source for biofilm growth. Our data also suggest that iron acquisition machinery is required for biofilm development. Finally, the addition of different concentrations of L-rhamnose to F. columnare prior to a laboratory challenge increased mortality rates in channel catfish (Ictalurus punctatus) as compared to controls. These results provide further evidence that biofilm formation is an integral virulence factor in the initiation of disease in fish.

AQUI-S 20E (10% eugenol) has been shown to be an effective fish sedative for freshwater finfish, but to be approved for use in the United States, the U.S. Food and Drug Administration must be provided with data demonstrating the product is safe to treated animals. Accordingly, we evaluated the safety of AQUI-S 20E when used to sedate Rainbow Trout Oncorhynchus mykiss, Yellow Perch Perca flavescens, and Channel Catfish Ictalurus punctatus. After identifying the highest effective concentrations likely to be used by fisheries biologists to sedate each species to handleable, replicate groups of fish were exposed to this intended concentration (1×) or a 50% higher eugenol concentration (1.5×) for periods of time adequate to effectively sedate 80% of fish to handleable (ET80) or for predetermined treatment periods exceeding the ET80. Taste sensation plays a pivotal role in nutrient identification and acquisition. This is particularly true for channel catfish (Ictalurus punctatus) that live in turbid waters with limited visibility. This biological process is mainly mediated by taste receptors expressed in taste buds that are distributed in several organs and tissues, including the barbels and skin. In the present study, we identified a complete repertoire of taste receptor and gustatory associated G protein genes in the channel catfish genome. A total of eight taste receptor genes were identified, including five type I and three type II taste receptor genes. Their genomic locations, phylogenetic relations, orthologies and expression were determined. Phylogenetic and collinear analyses provided understanding of the evolution dynamics of this gene family. Furthermore, the motif and dN/dS analyses indicated that selection pressures of different degrees were imposed on these receptors. Additionally, four genes of gustatory associated G proteins were also identified. It was indicated that expression patterns of catfish taste receptors and gustatory associated G proteins across organs mirror the distribution of taste buds across organs. Finally, the expression comparison between catfish and zebrafish organs provided evidence of potential roles of catfish skin and gill involved in taste sensation.

The genus Eustrongylides includes nematodes that infect fish species and fish-eating birds inhabiting freshwater ecosystems. Nematodes belonging to the genus Eustrongylides are potentially pathogenic for humans, infection occurs after the consumption of raw or undercooked fish. In the two-year period 2019&ndash, 2020, a total of 292 fish belonging to eight species were examined for the occurrence of Eustrongylides spp. from Lake San Michele, a small subalpine lake in northwest Italy. The prevalence of infestation was 18.3% in Lepomis gibbosus, 16.7% in Micropterus salmoides, and 10% in Perca fluviatilis. The other five fish species (Ameiurus melas, Ictalurus punctatus, Squalius cephalus, Carassius carassius, and Scardinius erythrophthalmus) were all negative for parasite presence. There were no significant differences in prevalence between the three fish species (Fisher&rsquo, s exact test, p = 0.744). The mean intensity of infestation ranged from 1 (M. salmoides and P. fluviatilis) to 1.15 (L. gibbosus), and the mean abundance ranged from 0.1 (P. fluviatilis) to 0.28 (L. gibbosus). There were significant differences in the infestation site between the four muscle quadrants (anterior ventral, anterior dorsal, posterior ventral, and posterior dorsal) and the visceral cavity (Kruskal&ndash, Wallis test, p = 0.0008). The study findings advance our knowledge about the distribution and host range of this parasite in Italy.

SynopsisMost predatory ray-finned fishes swallow their food whole, which can pose a significant challenge, given that prey items can be half as large as the predators themselves. How do fish transport captured food from the mouth to the stomach? Prior work indicates that, in general, fish use the pharyngeal jaws to manipulate food into the esophagus, where peristalsis is thought to take over. We used X-Ray Reconstruction of Moving Morphology to track prey transport in channel catfish (Ictalurus punctatus). By reconstructing the 3D motions of both the food and the catfish, we were able to track how the catfish move food through the head and into the stomach. Food enters the oral cavity at high velocities as a continuation of suction and stops in the approximate location of the branchial basket before moving in a much slower, more complex path toward the esophagus. This slow phase coincides with little motion in the head and no substantial mouth opening or hyoid depression. Once the prey is in the esophagus, however, its transport is surprisingly tightly correlated with gulping motions (hyoid depression, girdle retraction, hypaxial shortening, and mouth opening) of the head. Although the transport mechanism itself remains unknown, to our knowledge, this is the first description of synchrony between cranial expansion and esophageal transport in a fish. Our results provide direct evidence of prey transport within the esophagus and suggest that peristalsis may not be the sole mechanism of esophageal transport in catfish.

The distribution and further range expansion of non-native blue catfish Ictalurus furcatus in coastal waters throughout the United States Atlantic slope depend, in part, on the salinity tolerance of the fish. However, temperature-mediated sublethal effects of increased salinities on blue catfish biology are not yet known. We assessed the effects of salinity and temperature on growth, body condition, body composition and food consumption of juvenile blue catfish in a controlled laboratory experiment. Temperature and salinity had an interactive effect on blue catfish biology, although most fish survived 112 days in salinities up to 10 psu. At salinities ≤7 psu, mean growth rate, body condition and consumption rates were higher at 22°C than at 12°C. Mean consumption rates declined significantly with increasing salinities, yet, salinities ≤7 psu were conducive to rapid growth and high body condition, with highest growth and body condition at 4 psu. Fish at 10 psu exhibited low consumption rates, slow growth, low body condition and lower proportions of lipids. Habitats with hyperosmotic salinities (>9 psu) likely will not support the full lifecycle of blue catfish, but the fish may use salinities up to 10 psu for foraging, dispersal and even growth. Many oligohaline and mesohaline habitats in U.S. Atlantic slope drainages may thus be vulnerable to establishment of invasive blue catfish, particularly given the increasing temperatures as a result of climate warming.

The live transport of juvenile channel catfish (Ictalurus punctatus) is an important part of channel catfish breeding in China, but the anesthetic dose and physiological response of channel catfish after transport have not been studied yet. The purpose of this study was to assess the potential of eugenol and thymol in live transport and study their anesthetic effects of on juvenile channel catfish by analyzing a large number of parameters associated with stress and nonspecific immunity. First we tested eugenol and thymol concentrations of 10, 20, 30, 40, 50, and 100 mg/L. Eugenol successfully anesthetized the experimental fish at concentrations of 30–100 mg/L for 1258–82 s (resuscitation time was 147–300 s), while thymol was effective at 20–100 mg/L for 1612–81 s (recovery time was 333–475 s). The second experiment involved adding 10 and 20 mg/L of eugenol and thymol (4 groups) to the water, simulating transport (5 h), analyzing the stress and non-specific immune parameters of the serum, and determining gene expression changes in the liver and head kidney for 48 h. Anesthetics in the transport water inhibited the increase in serum cortisol levels (treatment group: 39.81–60.79 ng/mL, control group: 142.14 ng/mL), and Hsp90 in the liver was a more suitable biomarker for transport stress in channel catfish than Hsp70. Concentrations of serum total Ig, lysozyme, lectin, and ACH50, as well as the expression levels of cytokines (IL-1β, IL-8, and TNF-α) and pattern recognition receptors (NOD-1) in the head kidney increased. The expression of cytokines and NOD-1 in the head kidney returned to the pre-transport level after 48 h. Both anesthetics are suitable for channel catfish, but thymol is more suitable as a transport sedative. The results of this study could help maintain the health of juvenile channel catfish after live transport, decrease their infection and mortality, improve wellbeing, and reduce economic losses to the aquaculture industry.

Apolipoproteins are protein component of plasma lipoproteins. They exert crucial roles in lipoprotein metabolism and serve as enzyme cofactors, receptor ligands, and lipid transfer carriers in mammals. In teleosts, apolipoproteins are also involved in diverse processes including embryonic and ontogenic development, liver and digestive system organogenesis, and innate immunity. In this study, we identified a set of 19 apolipoprotein genes in channel catfish (Ictalurus punctatus). Phylogenetic analysis and syntenic analysis were conducted to determine their identities and evolutionary relationships. The expression signatures of apolipoproteins in channel catfish were determined in healthy tissues and after infections with two major bacterial pathogens, Edwardsiella ictaluri and Flavobacterium columnare. In healthy channel catfish, most apolipoprotein genes exhibited tissue-specific expression patterns in channel catfish. After ESC and columnaris infections, 5 and 7 apolipoprotein genes were differentially expressed respectively, which presented a pathogen-specific and time-dependent pattern of regulation. After ESC infection, three exchangeable apolipoproteins (apoA-IB, apoC-I, and apoE-B) were suppressed in catfish intestine, while two nonexchangeable apolipoproteins (apoB-A and apoB-B) were slightly up-regulated. After columnaris infection, apoB-B, apoD-B, and apoE-A were significantly down-regulated in catfish gill, while apoF, apoL-IV, apoO-like, and apo-14 kDa showed significantly up-regulation. Taken together, these results suggested that apolipoprotein genes may play significant roles in innate immune responses to bacterial pathogens in channel catfish.

Extruded and pelleted feeds were fed with different amounts (100, 90, 80, and 70% of satiation) to channel catfish (42 g initial weight) and the effects on growth, nutrient retention, and serum biochemistry were examined. The two feeds were designed with the same formula (containing 308 g kg−1 crude protein and 53 g kg−1 crude lipid), and were fed to eight treatments of fish (pelleted feed with a satiation of 100, 90, 80, 70% and extruded feed with a satiation of 100, 90, 80, 70%) with triplicate cage per treatment (20 fish per cage) for 10 weeks. The results indicated that weight gain (WG) of the eight groups were 371.1, 328.1, 302.1, 281.1 and 370.5, 334.0, 311.0, 285.3%, respectively. The fish fed extruded and pelleted feed at the same feeding rate showed no significant differences in growth performance, whole-body composition, and nutrient retention (P > 0.05). The North American channel catfish Ictalurus punctatus Rafinesque 1818 is cultivated in the United States, Asia and Brazilian fish farms, and also utilized as a model species in aquaculture and genetic studies. In this work, cytogenetic analysis of . punctatus from Brazilian aquaculture revealed for the first time the presence of extra elements (supernumerary or B chromosomes) in this species. These elements were characterized as dot-like micro B chromosomes and were found in three individuals (varying from 0 to 1) and in one individual with higher incidence per cell (varying from 0 to 5; mean number of Bs per cell = 2.01). More specific cytogenetic techniques in this individual revealed 58 A chromosomes (standard complement) containing heterochromatic bands in the centromeric regions, a single Ag-NOR in a subtelocentric pair (also positive for 18S rDNA using the FISH technique) and multiple 5S rDNA clusters in three different subtelocentric chromosomes. Four B chromosomes were entirely Ag-NOR positive (also fully heterochromatic) and three presented 18S rDNA clusters by FISH. The occurrence of Ag-NOR and 18S ribosomal genes in both A and B chromosome complements may indicate an intraspecific origin for these extra chromosomes. Additionally, the terminal location of 18S ribosomal clusters in the Ag-NOR-bearing chromosomes and the presence of active NOR in the B chromosomes suggested that breakage events may be related to a possible recent origin of these extra elements. We suggest this data may be useful as cytogenetic information for future elucidation of the composition, origin and evolution of extra chromosomes in fishes.

Use of peanut meal as an alternative protein source in diets for Channel Catfish Ictalurus punctatus was evaluated in a 9-week study under controlled laboratory conditions. Five practical diets (28% crude protein and 6% crude lipid) were formulated to contain 0, 10, 15, 20, and 25% peanut meal as a replacement for cottonseed meal and soybean meal in the control diet. The control diet was a plant-based diet containing principally soybean meal (25%), cottonseed meal (20%), corn (20%), and corn germ meal (20%). All diets met or exceeded all known nutrient requirements of Channel Catfish. Twenty Channel Catfish fingerlings (mean initial weight: 12.8 g/fish) were stocked into 25 water flow-through glass aquaria (110 L). The fish were fed once daily to apparent satiation. Putative spermatogonia A from a fresh-cell isolate or a density-gradient-centrifuged isolate from the testes of Blue Catfish Ictalurus furcatus were transplanted into the gonads of triploid Channel Catfish I. punctatus. The cells were introduced into gonads of the host via catheterization (2 × 104–1.43 × 106 cells) or by surgically inserting the cells directly into the gonad (7 × 104–1.25 × 105 cells). Ten months after implantation, DNA was analyzed from biopsies of the gonads and seven of eight males were found to be xenogenic, having Blue Catfish cells in their gonads. The xenogenic males successfully courted normal Channel Catfish that had been induced with hormones to ovulate, but none of the eggs hatched, indicating inadequate sperm production, an inability to ejaculate, and/or low sperm quality. Male xenogenic catfish treated with luteinizing hormone releasing hormone analog had well-developed testes, and sperm production was detected in three of seven xenogenic males examined 2 years after transplantation. Sperm were removed from a male that had been surgically transplanted with Blue Catfish cells and used to fertilize eggs from a hand-stripped Channel Catfish female. One percent of these eggs hatched. All seven surviving 6-month-old progeny of this male had the external morphology, swim bladder shape, nuclear DNA profile, and mitochondrial DNA profile of female Channel Catfish × male Blue Catfish F1 hybrids, indicating that this triploid Channel Catfish male produced Blue Catfish sperm. This is the first report of successful production of xenogenic catfish and the first report of producing 100% hybrid progeny using xenogenesis in fish. The present study was designed to determine whether female Channel Catfish Ictalurus punctatus exhibiting accelerated maturation display similar steroid (estradiol and testosterone) and gonadotropin (luteinizing hormone and follicle-stimulating hormone [FSH] messenger RNA) profiles within the first reproductive period compared with full-sibling females that reached maturity 1 year later. The accelerated maturation (AM) fish were maintained under a shortened annual temperature cycle, consisting of 4 months in 26°C water followed by 2 months at 13–14°C, repeated for three cycles. The study was conducted to examine the effects of different methionine (Met) sources on the growth performance of channel catfish (Ictalurus punctatus). Six diets with isonitrogen and isoenergy were formulated as positive control (PC) diet (0.57% Met), negative control (NC) diet (0.42% Met) without or with the addition of crystalline DL-Met (CM), Met hydroxy analogue (MHA), Met hydroxy analogue calcium salt (MHA-Ca) and microcapsulated DL-Met (MM) to obtain the same Met level as PC diet. After 8 weeks feeding trial, channel catfish (76.0 ± 0.3 g) fed MHA-Ca and MM diets, but not CM and MHA diets, had higher weight gain, lipid productive value, protein productive value and lower feed conversion ratio than those in NC group (P 0.05). Dual-energy X-ray absorptiometry (DXA) provides a noninvasive way to determine lean tissue mass (LTM), fat mass (FM), bone mineral content (BMC), and bone mineral density (BMD) in humans and small mammals. Live channel catfish (n=74, 78g - 1200 g) were anesthetized and scanned in both a lateral position and a dorsa-ventral position. Six individual fish (300g - 600g) were scanned five times each to determine precision by the coefficient of variation. Precision was good for LTM (0.75-1.06%) and for BMC and BMD (2-2.6%). Precision for FM was not good (27-34%), which was due to the very low FM (0-1g) recorded by the DXA. However, using the predicted values, FM precision improved to 5-5.5%. DXA values for LTM, FM, and BMC were significantly different from chemical analysis (P 0.05). DXA may also be a valuable tool for evaluating body condition longitudinally in commercial or in threatened or endangered fish species, where non-invasive procedures would be invaluable.

A pond study was initiated to evaluate effects of no feeding, maintenance feeding, and refeeding on production and processing characteristics of market-size hybrid catfish (female Channel Catfish Ictalurus punctatus × male Blue Catfish I. furcatus). Fish with an average weight of 644 g were stocked into 20 earthen ponds (0.04 ha) at 14,826 fish/ha at the end of May. They were either not fed (N2), fed once weekly (W2) to satiation for 2 months to simulate a long-term harvest delay, or fed daily (D2) to satiation. After 2 months fish in half the ponds in each of the two feed-restricted treatments were harvested, and all fish in the remaining ponds were fed daily for an additional month. Metabolic adjustments were studied in channel catfish Ictalurus punctatus exposed to 1.5 mg L-1 of phe nol (10% LC50) for four days and recovered for seven days. Lower triacylglycerol (TGA) stores and increased muscle fat free acids (FFA) suggest fat catabolism in muscle. Remarkable liver FFA decrease (-31%) suggests liver fat catabolism as well. Increased muscular ammonia levels and ASAT (aspartate aminotransferase) and decreased plasma aminoacids suggest higher muscular amino acid uptake. Constant levels of glucose and increased liver glycogen stores, associated with lower amino acids in plasma, indicate gluconeogenesis from amino acids. This is supported by higher hepatic ALAT and ASAT. Higher hepatic LDH followed by lower plasma lactate may indicate that plasma lactate was also used as gluconeogenic substrate. Biochemical alterations were exacerbated during the post-exposure recovery period. Reduction in muscle and plasma protein content indicate proteolysis. A higher rate of liver fat catabolism was resulted from a remarkable decrease in hepatic TGA (-58%). Catabolic preference for lipids was observed in order to supply such elevated energy demand. This study is the first insight about the metabolic profile of I. punctatus to cope with phenol plus its ability to recover, bringing attention to the biological consequences of environmental contamination. Os ajustes metabólicos foram estudados em bagre do canal Ictalurus punctatus exposto a 1.5 mg L-1 de fenol (10% da CL50) por quatro dias e após recuperação por sete dias. Redução da concentração de triacilglicerol e aumento de ácidos graxos livres no músculo são indicativos de catabolismo de lipídeos. Redução de ácidos graxos livres (-31%) no fígado também indica catabolismo de lipídeos. Aumento da concentração de amônia e da atividade da aspartato aminotransferase (ASAT) no músculo, e a redução na concentração plasmática de aminoácidos sugerem aumento na captação muscular de aminoácidos. Níveis constantes de glicose e o aumento da reserva hepática de glicogênio, associados a baixos níveis de aminoácidos no plasma, indicam gliconeogênese a partir de aminoácidos. Este fato é respaldado pelo aumento de ALAT e ASAT. A indução da LDH hepática acompanhada da redução de lactato plasmático podem indicar que o lactato plasmático foi também utilizado como substrato para a gliconeogênese. As alterações bioquímicas foram exacerbadas após o período de recuperação. Reduções no conteúdo de proteína muscular e plasmática indicam proteólise. O aumento do catabolismo lipídico no fígado foi resultado de uma expressiva redução da concentração hepática de TGA (-58%). Uma preferência catabólica por lipídeos foi observada na tentativa de suprir a elevada demanda energética. Este estudo é o primeiro a abordar o perfil metabólico de I. punctatus frente à exposição ao fenol e sua capacidade de recuperação, trazendo a atenção para as consequências biológicas da contaminação ambiental.

Recently, the detection of pharmaceuticals in surface waters has increased worldwide. Pharmaceuticals are typically found in the environment at concentrations well below therapeutic levels in humans; however, their mechanisms of action may be largely unknown in non-target organisms, such as teleost species. Thus, chronic exposure to these types of compounds warrants further investigation. The goal of this study was to examine the potential for diazepam, a model benzodiazepine drug, to bioconcentrate in tissues of channel catfish and to examine its ability to interact with the endocrine system through modulation of steroid hormones and/or steroidogenic genes. To investigate the bioconcentration potential of diazepam, channel catfish (Ictalurus punctatus) were exposed to 1 ng/mL diazepam for seven days, followed by clean water for another seven days, using an abbreviated OECD 305 Fish Bioconcentration Test study design. This concentration of diazepam is well below environmentally relevant concentrations of diazepam (ng/L). To evaluate steroidogenic effects, fish were exposed to 1 ng/mL diazepam for seven days only. Steroid hormone concentrations were analyzed for various tissues, as well as expression of selected steroidogenic genes. Calculated bioconcentration factors for diazepam were well below regulatory threshold values in all tissues analyzed. No changes in steroid hormone concentration were detected in any tissue analyzed; however, the steroidogenic gene cytochrome P450 side chain cleavage (P450scc) was significantly down-regulated at day 5 and 3β-hydroxy steroid dehydrogenase (3β-HSD) was significantly down-regulated at day 7 in the gonad. These results indicate that although diazepam does not significantly bioconcentrate, low-level chronic exposure to diazepam may have the potential to interact with endocrine function by altering gene expression.
