1. Clinoptilolite supported feeding reduces excessive iron in thalassemia rat model created with iron loading

Abstract

There is no regulatory mechanism for the removal of iron that accumulates in the body. Thalassemia patients are most affected by iron overload. The method often used in the treatment of these patients is iron chelation therapy, which involves removing excess iron from the bloodstream, but it is insufficient. Nutritional supplements and herbal remedies are complementary tools that may help improve the health of an individual with iron load. Here, we tried to develop a method that affecting intestinal absorption in an animal model with clinoptilolite feeding to reduce the iron load in the circulation. 32 rats were divided into 4 groups as control, Cli, Iron, Cli+Iron. Iron and Cli+Iron groups received iron at a dose of 250 mg/kg/day for 10 days, and Cli and Cli+Iron groups were fed a diet with 50% clinoptilolite for one month. Histological preparation and Fe2+, Cu2+, Zn2+ measurements were done in all tissues. Consequently, clinoptilolite significantly lowered the iron level in the stomach. On the contrary, iron absorption was increased in the small intestine, but iron transportation to the blood was decreased by clinoptilolite. The iron levels of clinoptilolite groups (Cli and Cli+Iron) were reduced in the tissues of heart, lung, liver, kidney, and spleen because of the different level of iron necessities of each tissue compared to the group with iron overload. The clinoptilolite could preserve organs against iron toxicity by enhancing the absorption of iron in the small intestine but lowering the iron level in blood. Even though there was no detailed information regarding the mechanism of reduction iron overload by the clinoptilolite, serum and chyme could help to make some helpful inferences to elucidate the mechanism of iron chelation. Abstract

Colorectal cancer is the third most common cancer worldwide. Cancer stem cells are known to play an important role in relapse, and metastases of the disease after chemotherapy. Investigation of new drugs, and their combinations targeting these cells and thus eliminating cancer is one of the most urgent needs of today’s chemotherapy. The aim of the present study was to evaluate the effects of Bryophytes like Abietinella abietina (AA), Homolothecium sericeum (HS), Tortella tortuosa (TT), Syntrichia ruralis (SR), and Bryoerythrophyllum rubrum (BR) species extracted with ethyl alcohol on 5-fluorouracil(5-FU) resistant colorectal cancer cell lines (HCT116 and HT29). After extraction, stock solutions of bryophytes were prepared, and IC50 values were detected in drug-resistant cells obtained with 5-FU application. CD24+, CD44+/CD133+ surface markers and P‐glycoprotein (P-gp) mediated efflux were isolated from both 5-FU treated cells and analyzed using the flow cytometry. In all bryophyte-treated groups, the binding Rho123low (low Rho fluorescence) and Rhohigh (high Rho fluorescence) were sorted from 5-FU resistant HCT116, and HT-29 cells. All types of bryophytes were found cytotoxic. Bryophyte extract reduced the percentage of Rholow cells in cultures incubated with 5-FU. In summary, the implementation of these bryophytes might be regarded as an effective approach for treatment of colorectal cancer due to their cytotoxic effect that decreases the recurrence of the disease. Abstract

In order to provide more effective treatment strategies for the rapid healing of diabetic wounds, novel therapeutic approaches need to be developed. The therapeutic potential of peroxisome proliferator-activated receptor-γ (PPAR-γ) agonist pioglitazone hydrochloride (PHR) in two different release kinetic scenarios, burst release and sustained release, was investigated and compared with in vitro and in vivo tests as potential wound healing dressings. PHR-loaded fibrous mats were successfully fabricated using polyvinyl-pyrrolidone and polycaprolactone by scalable pressurized gyration. The results indicated that PHR-loaded fibrous mats expedited diabetic wound healing in type-1 diabetic rats and did not show any cytotoxic effect on NIH/3T3 (mouse embryo fibroblast) cells, albeit with different release kinetics and efficacies. The wound healing effects of fibrous mats are presented with histological and biochemical evaluations. PHR-loaded fibrous mats improved neutrophil infiltration, oedema, and inflammation and increased epidermal regeneration and fibroblast proliferation, but the formation of hair follicles and completely improved oedema were observed only in the sustained release form. Thus, topical administration of PPAR-γ agonist in sustained release form has high potential for the treatment of diabetic wounds in inflammatory and proliferative phases of healing with high bioavailability and fewer systemic side effects.

Abstract

Şahin, Yeşim Müge (Arel Author), The use of natural compounds such as biocompatible and non-toxic plant extracts, without undesired side effects, in tissue engineering applications, is highly preferred compared to chemical drugs. In this study, the characterization and performance of electrospun Achillea lycaonica-loaded (0.125, 0.250 and 0.500, wt%) poly(lactic acid) (PLA) (8%, w/v) nanofibrous mats for skin tissue engineering were investigated. SEM, FTIR, DSC, and tensile strength test of the electrospun nanofibers have been investigated. Drug releasing test and cell culture study were also carried out. Achillea lycaonica-loaded nanofibrous mats in 0.250 (wt%) and 0.500 (wt%) demonstrated excellent cell compatibility and increased the viability of NIH/3T3 (mouse embryo fibroblast) cells within 72 h. According to the results, Achillea lycaonica-loaded PLA nanofibers have proper tensile strength and controlled release. The working temperature range enlarged for the composites having higher plant extract content. Consequently, Achillea lycaonica-loaded nanofibrous mats have a great potential in skin tissue engineering applications. © 2019 Elsevier Ltd, Dr. Muhammet E.
