Human adenoviruses cause disease at multiple mucosal sites, including the respiratory, gastrointestinal, and genitourinary tracts, and are common agents of conjunctivitis. One site of infection that has received sparse attention is the cornea, a transparent tissue and the window of the eye. While most adenovirus infections are self-limited, corneal inflammation (keratitis) due to adenovirus can persist or recur for months to years after infection, leading to reduced vision, discomfort, and light sensitivity. Topical corticosteroids effectively suppress late adenovirus keratitis but are associated with vision-threatening side effects. In this short review, we summarize current knowledge on infection of the cornea by adenoviruses, including corneal epithelial cell receptors and determinants of corneal tropism. We briefly discuss mechanisms of stromal keratitis due to adenovirus infection, and review an emerging therapy to mitigate adenovirus corneal infections based on evolving knowledge of corneal epithelial receptor usage.

Chikungunya virus has emerged as one of the most important global arboviral threats over the last decade. Inspite of large scale morbidity, with long lasting polyarthralgia, so far no licensed vaccine or antiviral is available. CHIKV nsP2 protease is crucial for processing of viral nonstructural polypeptide precursor to release enzymes required for viral replication, thus making it a promising drug target. In this study, high cell density cultivation (HCDC) of Escherichia coli in batch process was carried out to produce rCHIKV nsP2pro in a cost-effective manner. Invasion of host cell by pathogens induce various intracellular signalling pathways. The host cell through the initiation of these signalling circuits desperately wants to get rid of the pathogen, whereas the pathogen tries to subvert these defence strategies to create an environment for their successful survival. Leishmania spp. is not an exception. Leishmania have to evolve a range of strategic mechanisms to neutralize macrophage defensive arsenals which enable the parasite to replicate within the phagolysosome of infected host. Understanding these signalling mechanisms in detail will not only improve our basic knowledge of host-pathogen interaction but will also help us to develop effective drug targets not only against leishmaniasis but also for many other macrophage associated diseases. © 2018 IUBMB Life, 70(7):593-601, 2018.

Herein, we have reported a low cost simple aminoquinoline based pH sensor HL [HL = 2-methoxy-6-((quinolin-3-ylimino)methyl)phenol]. HL acts as a potential low pH fluorometric as well as colourimetric pH probe. In acidic media it is colourless whereas, in basic media, it is yellow in colour. HL is thoroughly characterized by different spectroscopic techniques viz. UV-visible, fluorescence, 1H and 13C NMR spectroscopy along with ESI-MS and elemental analysis. We are successful in elucidating the X-ray crystal structure of HL. Theoretical calculations such as DFT and TDDFT are also performed to support our experimental findings. The ability of this biocompatible pH sensor (HL) to differentiate normal and cancer cells by alteration of pH will open new vistas for various biomedical applications.

Small heterocyclic molecules such as piperazine are potential pharmacotherapeutic agents and binding of these molecules to the hydrophobic pocket of capsid protein (CP) offers a new perspective for therapeutic intervention. Here, we report the crystal structure of CP from Aura virus (AVCP) in complex with piperazine at 2.2 Å resolution. Piperazine binds to the conserved hydrophobic pocket of CP where dioxane based antivirals bind. Comparative structural studies of the piperazine-bound AVCP structure with the apo, active and dioxane-bound AVCP structures provide insights into the conformational variations in the pocket. Additionally, the molecular docking studies showed that piperazine binds into the hydrophobic pocket of Chikungunya virus CP (CVCP) with more affinity than with AVCP. Furthermore, the antiviral activity of piperazine against Chikungunya virus (CHIKV) was investigated by plaque reduction and immunofluorescence assays. The AVCP-piperazine complex may serve as a lead scaffold for structure-based design of piperazine derivatives as alphaviral inhibitors. The antiviral properties of piperazine provide its usefulness for further investigations towards the development of piperazine based anti-alphaviral drugs.

Human adenovirus infection of the ocular surface is associated with severe keratoconjunctivitis and the formation of subepithelial corneal infiltrates, which may persist and impair vision for months to years following infection. Long term pathology persists well beyond the resolution of viral replication, indicating that the prolonged immune response is not virus-mediated. However, it is not clear how these responses are sustained or even initiated following infection. This review discusses recent work from our laboratory and others which demonstrates different entry pathways specific to both adenovirus and cell type. These findings suggest that adenoviruses may stimulate specific pattern recognition receptors in an entry/trafficking-dependent manner, leading to distinct immune responses dependent on the virus/cell type combination. Additional work is needed to understand the specific connections between adenoviral entry and the stimulation of innate immune responses by the various cell types present on the ocular surface.

[RuIII(EDTA)(H2O)]− (EDTA4− = ethylenediaminetetraacetate) catalyzes the oxidation of biological thiols, RSH (RSH = cysteine, glutathione, N-acetylcysteine, penicillamine) using H2O2 as precursor oxidant. The kinetics of the oxidation process were studied spectrophotometrically as a function of [RuIII(EDTA)(H2O)]−, [H2O2], [RSH], and pH (4–8). Spectral analyses and kinetic data are suggestive of a catalytic pathway in which the RSH reacts with [RuIII(EDTA)] catalyst complex to form [RuIII((EDTA)(SR)]2− intermediate species. In the subsequent reaction step the oxidant, H2O2, reacts directly with the coordinated S of the [RuIII((EDTA)(SR)]2− intermediate leading to formation of the disulfido (RSSR) oxidation product (identified by HPLC and ESI-MS studies) of thiols (RSH). Based on the experimental results, a working mechanism involving oxo-transfer from H2O2 to the coordinated thiols is proposed for the catalytic oxidation.

Highlights • Development of a SYBR Green I based quantitative RT-PCR assay for emerging RRV. • Oral infection of Ae. aegypti with RRV revealed 100% infection and dissemination rate. • A transmission rate of 75% was observed in Ae. aegypti. • Assay was validated by screening potential antivirals against RRV. • qRT-PCR can be a remarkable tool for virus surveillance and antiviral screening., Ross River virus (RRV) is an emerging Alphavirus and is presently endemic in many parts of Oceania. Keeping in mind its emergence, we developed a molecular detection system and utilized it to study vector competence and evaluate activity of antiviral compounds against RRV. A SYBR Green I-based quantitative RT-PCR for detection of RRV was developed targeting the E2 gene, with a detection limit of 100 RNA copies/reaction. The specificity was confirmed with closely related Alphaviruses and Flaviviruses. The assay was applied to study the vector competence of Indian Aedes aegypti for RRV, which revealed 100% infection and dissemination rate with 75% transmission rate. Viral RNA was found in saliva as early as 3 day post infection (dpi). Further application of the assay in antiviral drug evaluation revealed the superior in vitro activity of ribavirin compared to chloroquine in Vero cells. Successful demonstration of this assay to detect RRV in low titre mosquito samples makes it a sensitive tool in vector surveillance. This study also showed that Indian Ae. aegypti are well competent to transmit RRV highlighting the risk of its introduction to naïve territories across continents. Further validation of this assay, revealed its utility in screening of potential antivirals against RRV.

Four complexes based on N,N′-bis(2-pyridylmethylene)-1,3-propanediamine (L) and different Cd(II) salts have been synthesized and characterized by single-crystal X-ray diffraction analysis. The complexes are [Cd2(L)2(μ-Cl)2](ClO4)2 (1), [Cd2(L)2(μ-Br)2](ClO4)2 (2), [Cd(L)I](ClO4) (3), and [Cd(L)(NCS)2] (4). L exhibits the same coordination mode in 1–4. The radius of each anion plays a role in affecting the structures and luminescent intensities of the final products. CdCl2, CdBr2, and CdI2 react with L to produce chloride or bromido-bridged binuclear complexes and mononuclear iodido complex, respectively, whereas an unusual mononuclear trigonal prismatic (TP) 4 is obtained when thiocyanate was used as a coligand. Photoluminescence properties of all the complexes show that the trend of fluorescence intensity is 1 > 4 > 2 > 3. All four complexes exhibit different supramolecular interactions such as C–H/σ, π–π, and C–H/π and hydrogen bonding interactions. The experimental findings are complemented by density functional theory (DFT) calculations.

Based on the experimental data, a working mechanism is proposed for the [RuIII(EDTA)] catalyzed oxidation of thiols.

Background. The framework currently used for living kidney donor selection is based on estimation of acceptable donor risk, under the premise that benefits are only experienced by the recipient. However, some interdependent donors might experience tangible benefits from donation that cannot be considered in the current framework (ie, benefits experienced directly by the donor that improve their daily life, well-being, or livelihood). Methods. We conducted semistructured interviews with 56 living kidney donors regarding benefits experienced from donation. Using a qualitative descriptive and constant comparative approach, themes were derived inductively from interview transcripts by 2 independent coders; differences in coding were reconciled by consensus. Results. Of 56 participants, 30 were in interdependent relationships with their recipients (shared household and/or significant caregiving responsibilities). Tangible benefits identified by participants fell into 3 major categories: health and wellness benefits, time and financial benefits, and interpersonal benefits. Participants described motivations to donate a kidney based on a more nuanced understanding of the benefits of donation than accounted for by the current “acceptable risk” paradigm. Discussion. Tangible benefits for interdependent donors may shift the “acceptable risk” paradigm (where no benefit is assumed) of kidney donor evaluation to a risk/benefit paradigm more consistent with other surgical decision-making.

Purpose Ocular infection by human adenovirus species D type 37 (HAdV-D37) causes epidemic keratoconjunctivitis, a severe, hyperacute condition. The corneal component of epidemic keratoconjunctivitis begins upon infection of corneal epithelium, and the mechanism of viral entry dictates subsequent proinflammatory gene expression. Therefore, it is important to understand the specific pathways of adenoviral entry in these cells. Methods Transmission electron microscopy of primary and tert-immortalized human corneal epithelial cells infected with HAdV-D37 was performed to identify the means of viral entry. Confocal microscopy was used to determine intracellular trafficking. The results of targeted small interfering RNA and specific chemical inhibitors were analyzed by quantitative PCR, and Western blot. Results By transmission electron microscopy, HAdV-D37 was seen to enter by both clathrin-coated pits and macropinocytosis; however, entry was both pH and dynamin 2 independent. Small interfering RNA against clathrin, AP2A1, and lysosome-associated membrane protein 1, but not early endosome antigen 1, decreased early viral gene expression. Ethyl-isopropyl amiloride, which blocks micropinocytosis, did not affect HAdV-D37 entry, but IPA, an inhibitor of p21-activated kinase, and important to actin polymerization, decreased viral entry in a dose-dependent manner. Conclusions HAdV-D37 enters human corneal epithelial cells by a noncanonical clathrin-mediated pathway involving lysosome-associated membrane protein 1 and PAK1, independent of pH, dynamin, and early endosome antigen 1. We showed earlier that HAdV-D37 enters human keratocytes through caveolae. Therefore, epidemic keratoconjunctivitis-associated viruses enter different corneal cell types via disparate pathways, which could account for a relative paucity of proinflammatory gene expression upon infection of corneal epithelial cells compared with keratocytes, as seen in prior studies.

Intramacrophage protozoan parasite Leishmania donovani, causative agent of visceral leishmaniasis, escapes Toll-like receptor (TLR) dependent early host immune response by inducing the deubiquitinating enzyme A20, which is sustained up to 6 h postinfection only. Therefore, Leishmania must apply other means to deactivate late host responses. Here, we elucidated the role of IL-1 receptor-associated kinase M (IRAK-M), a negative regulator of TLR signaling, in downregulating macrophage proinflammatory response during late hours of in vitro infection. Our data reveal a sharp decline in IRAK1 and IRAK4 phosphorylation at 24 h postinfection along with markedly reduced association of IRAK1-TNF receptor associated factor 6, which is mandatory for TLR activation. In contrast, IRAK-M was induced after A20 levels decreased and reached a maximum at 24 h postinfection. IRAK-M induction coincided with increased stimulation of TGF-β, a hallmark cytokine of visceral infection. TGF-β-dependent signaling-mediated induction of SMAD family of proteins, 2, 3, and 4 plays important roles in transcriptional upregulation of IRAK-M. In infected macrophages, siRNA-mediated silencing of IRAK-M displayed enhanced IRAK1 and IRAK4 phosphorylation with a concomitant increase in downstream NF-κB activity and reduced parasite survival. Taken together, the results suggest that IRAK-M may be targeted by L. donovani to inhibit TLR-mediated proinflammatory response late during in vitro infection.

Candida albicans is an important opportunistic fungal pathogen capable of causing both mucosal and disseminated disease. Infections are often treated with fluconazole, a front-line antifungal drug that targets the biosynthesis of ergosterol, a major component of the fungal cell membrane. Resistance to fluconazole can arise through a variety of mechanisms, including gain-of-function mutations, loss of heterozygosity events and aneuploidy. The clinical isolate P60002 was found to be highly resistant to azole-class drugs, yet lacked mutations or chromosomal rearrangements known to be associated with azole resistance. Transcription profiling suggested that increased expression of two putative drug efflux pumps, CDR11 and QDR1, might confer azole resistance. However, ectopic expression of the P60002 alleles of these genes in a drug-susceptible strain did not increase fluconazole resistance. We next examined whether the presence of three copies of chromosome 4 (Chr4) or chromosome 6 (Chr6) contributed to azole resistance in P60002. We established that Chr4 trisomy contributes significantly to fluconazole resistance, whereas Chr6 trisomy has no discernible effect on resistance. In contrast, a Chr4 trisomy did not increase fluconazole resistance when present in the standard SC5314 strain background. These results establish a link between Chr4 trisomy and elevated fluconazole resistance, and demonstrate the impact of genetic background on drug resistance phenotypes in C. albicans.

Chikungunya virus (CHIKV) has emerged as a global health concern due to its recent spread in both old and new world. So far, no CHIKV specific drug or vaccine is licensed for human use. In this study, we report production of Chikungunya virus like particles (CHIK-VLPs) using novel yeast expression system (Pichia pastoris) and its evaluation as vaccine candidate. The gene encoding structural polyprotein of CHIKV from a recent epidemic strain was cloned into yeast expression system. The multicopy integrants were processed for expression of CHIK-VLPs. The VLPs were purified and confirmed through electron microscopic analysis for their morphological identity with CHIKV. The in vitro and in vivo evaluation of CHIK-VLPs as vaccine candidate was determined in Balb/c mice. Induction of both humoral and cellular immune response was observed with different doses of CHIK-VLPs. The humoral immune response was studied through different techniques like enzyme linked immunosorbent assay, IgG Isotyping and plaque reduction neutralization test. CHIK-VLPs were found to elicit high titer of antibodies that are able to recognize native CHIKV. Higher level of IgG2a and IgG1 subtypes was identified suggestive of balanced Th1/Th2 response. Both in vitro and in vivo neutralization activity of CHIK-VLPs antibodies was observed even with low concentration, which shows its high specificity and neutralizing activity against two different CHIKV strains. Neonatal mice receiving anti-CHIK-VLPs antibodies were protected from CHIKV challenge. Induction of cellular immune response was confirmed through higher level of TNF-α, IL-10 and substantial level of IL-2, IL-4 and IFN-γ indicating a balanced response. This is the first report, where CHIK-VLPs has been expressed by Pichia pastoris and evaluated for neutralizing activity against CHIKV. These promising results indicate the utility of CHIK-VLPs as a promising vaccine candidate against emerging CHIKV., Author Summary Chikungunya virus (CHIKV) has emerged in many parts of tropics in last decade. The absence of an approved vaccine or antiviral drug for CHIKV makes it one of the important public health challenges. Though attempt to develop a CHIKV vaccine was initiated in 1980s, however it has not succeeded so far. The Virus like particles (VLPs) are now explored as promising vaccine candidate against many viruses viz. HBV, HPV etc. In this study, we report the production of CHIK-VLPs using novel yeast expression system (Pichia pastoris) and its evaluation as vaccine candidate. These CHIK-VLPs share morphological identity to native CHIKV. The results indicate that CHIK-VLPs induced both cell mediated as well as humoral response in a balanced manner, which fulfils its criteria as a potent immunogen. Further, antibodies generated against CHIK-VLPs demonstrated efficient in vitro and in vivo neutralization activity, as evaluated through plaque reduction in Vero cells and protection in CHIKV infected neonatal mice respectively using two different CHIKV strains, which makes it a promising vaccine candidate. The yeast expressed CHIK-VLPs has high potential for development of an effective vaccine candidate against CHIKV during epidemic situations.

Chikungunya virus (CHIKV) has emerged as one of the most significant arboviral threats in many parts of the world. In spite of large scale morbidity, and long lasting polyarthralgia, no licensed vaccine or antivirals are available for the clinical management of CHIKV infection. In this study, a novel RNA interference based strategy has been adopted for effective inhibition of CHIKV. Four artificial microRNAs (amiRNAs) were designed to target different regions of CHIKV genome. These amiRNAs significantly inhibited CHIKV replication in Vero cells at both RNA and protein levels as assessed by qRT-PCR, immunoblotting and immunofluorescence techniques. Further inhibition of the infectious CHIKV up to 99.8% was demonstrated by plaque reduction assay. Concatemerization of amiRNA resulted in higher inhibition of CHIKV than individual amiRNAs. In addition, we studied the effect of combination of RNAi based therapy with other classical antivirals like chloroquine, ribavirin and mycophenolic acid, that helped in understanding the rational selection of RNAi based combination therapy. These findings provide a promising avenue for the development of novel amiRNA or combination based therapeutics against emerging CHIKV., Highlights • amiRNAs targeting different ORF of CHIKV was designed. • Significant Inhibition of CHIKV replication through amiRNA was demonstrated. • Concatenated amiRNAs results in higher viral inhibition. • Combination of RNAi with classical drugs may obliterate failure of monotherapy.

A total of 10 samples were collected from waste dump sites of Dhapa and Barrackpore Municipality and 9 isolates of bacteria were isolated using Nutrient Agar medium. The optimal cultural conditions, microbiological characteristics, biochemical characteristics, antagonistic and synergistic activities within the strains, tolerance to 5 heavy metals (arsenic, zinc, lead, mercury, and cadmium), their sensitivity towards four different antibiotics (gentamycin, oxytetracycline, penicillin, streptomycin) and production of extracellular enzyme of the bacterial strains were documented. Among the 9 isolates 6 exhibited protease production and 1 was selected for protease production with high titer value. All of the 6 strains capable of producing protease were also applied for waste degradation efficiency test. These results have increased the scope of finding industrially important bacteria from municipal waste dump sites and these isolates could be vital source for the discovery of industrially useful

The draft genome sequence of an amylase-, protease-, lipase-, oxidase-, and catalase-producing Gram-negative bacillus ( Aeromonas sp. MDS8 strain MCC2167) with the ability to produce ammonia during 16 h of growth at 37°C, isolated from dairy sludge, with a size of 4,841,753 bp and a G+C content of 63.1%, is reported here.

The aim of this study was to evaluate and characterize the therapeutic potential of curdlan, a naturally occurring β-glucan immunomodulator, against visceral leishmaniasis, a fatal parasitic disease. Curdlan eliminated the liver and spleen parasite burden in a 45-day BALB/c mouse model of visceral leishmaniasis at a dosage of 10 mg/kg/day as determined by Giemsa-stained organ impression smears. Curdlan was associated with production of the disease-resolving T-helper (Th) 1 and Th17-inducing cytokines interleukin (IL)-6, IL-1β, and IL-23, as well as with production of Th17 cytokines IL-17 and IL-22, as determined by enzyme-linked immunosorbent assay (ELISA) and real time polymerase chain reaction (RT-PCR). Reversal of curdlan-mediated protection by anti-IL-17 and anti-IL-23 monoclonal antibodies showed the importance of Th17 cytokines. Significantly decreased production of both IL-17 and IL-22 by mice that received anti-IL-23 antibody suggested the essential role of IL-23 in Th17 differentiation. Although administration of recombinant IL-17 or IL-23 caused significant suppression of the organ parasite burden, with marked generation of interferon γ and nitric oxide (NO), effects were much faster for IL-17. These results documented that although both IL-23 and IL-17 play major roles in the antileishmanial effect of curdlan, the effect of IL-23 may occur indirectly, through the induction of IL-17 production.
