Ultraschalllevitation kombiniert die Vorteile von Mikrofluidik, wie beispielsweise die sehr geringe benötigte Probenmenge, mit einer wandlosen Probenhandhabung. Obwohl die Kopplung zwischen le- vitierten Tröpchen und verschiedenster analytischer Methoden wie optischer Spektroskopie und Röntgenbeugung sehr genau untersucht ist, fehlt es immer noch an einer etablierten Kopplung mit einer massenspektrometrischen Methode für die Analyse auf molekularer Ebene. Die vorliegende Arbeit beschreibt die Prinzipien, auf denen eine kontaktlose massenspektrometrische Analyse von levitierten flüssi- gen Proben beruht. Zuerst wurde der neu entworfene akustische Levitator bezüglich des Einflusses seiner Geometrie auf die Levi- tationseigenschaften experimentell und mittels numerischer Simul- tationen untersucht. Die anschließend durch geführten Experimen- te demonstrieren das Potential von Infrarot-Lasern als kombinierte Desorptions- und Ionisationsquelle für organische Substanzen aus einer Mischung aus Wasser und Glycerin als Cromophor. Um einen tieferen Einblick in die hierbei ablaufenden Ionisationsmechanismen zu erhalten, wurde als Modell ein “Sonic-Spray” Konus räumlich per Massenspektrometrie und Laser-induzierter Fluoreszenz untersucht. Levitator-Geometrie auf die Levitationseigenschaften stimmen sehr gut mit numerischen Simulationen überein. Als komplementäre Ionisationsmethode wurde eine Niedertemperatur-Plasmaquelle ein- gesetzt. Nach einer zeitaufgelösten Untersuchung der grundlegenden Ionisationsmechanismen wurde diese Quelle für die Untersuchung flüchtiger Spezies aus der levitierten Probe in deren direkten Umgebung ohne störende Interferenzen ge- nutzt. Ultrasonic levitation combines advantages of microfluidics like the required small sample volumes with a wall-less sample handling. While the coupling of analytical methods like optical spectroscopy as well as x-ray scattering are very well elaborated, an established mass spectrometric method to obtain molecular analytical information is still lacking. The herein presented work describes the fundamental processes for a contactless mass spectrometric analysis of levitated droplets. First, the influences of the specially designed levitator geometry on the levitation capabilities is described. During further experiments, the use of infrared lasers has proven useful as a combined desorption and ionization source for organic molecules from a mixture of water and glycerol as chromophore. Subsequently, sonic-spray ionization was used to gain a deeper understanding of the ionization processes occurring within the spray plume. Mass spectrometric mapping as well as laser-induced fluorescence were performed to investigate the ionization during an aerodynamic breakup of the micro droplets in the spray process. As a complementary sampling method, the ionization with a low- temperature plasma source is described. First, a time-resolved mass spectrometric investigation of the ionization process is shown. Sub- sequent to this fundamental study, the application of such a plasma source for the direct analysis of volatile compounds from within the droplets in the surrounding environment without interferences from the droplets bulk phase is described.

In dieser Arbeit konnte ein humaner IgG1-Antikörper mittels verschiedener massenspektrometrischer Methoden erfolgreich untersucht und charakterisiert werden. Neben der Bestimmung der molekularen Massen des betrachteten Anti-Interleukin-8-Antikörpers und dessen Fragmenten konnte auch die Bildung von Oligomeren untersucht werden. Es wurden verschiedene Oligomere, bis hin zum Pentamer, massenspektrometrisch gemessen und es konnten auch Erkenntnisse gewonnen werden, dass Konzentrationseffekte maßgeblich zur Oligomerbildung bei diesem Antikörper beitragen. Die Antikörperglykosylierung, eine sehr prominente und hoch diverse Modifikation, wurde bei diesem Antikörper ebenfalls gefunden und neben dem intakten Antikörper auch in dessen Fragmenten und im Dimer gemessen. Neben dieser natürlichen Modifikation wurden auch Derivatisierungen des Antikörpers analysiert, dabei wurden unterschiedliche Metallmarkierungen analysiert. Es konnten verschiedene Markierungsgrade bestimmt werden, auch wenn diese nebeneinander als Mischung vorlagen. Der Nachweis, dass der Antikörper auch weiterhin intakt und bindungsfähig ist, konnte erbracht werden. Die Bildung von Komplexen des Antikörpers mit verschiedenen Proteinen konnte ebenfalls mittels Massenspektrometrie gezeigt werden. Weiterhin konnte gezeigt werden, dass es möglich war, eine Quantifizierungsstrategie für Antikörper zu entwickeln. Mit dieser können absolute Quantifizierungen durchgeführt werden, während diese gleichzeitig unabhängig von Art und Struktur des einzusetzenden Standards sind. Damit ist es möglich, die Probleme des Bradford-Assays zu umgehen. Des Weiteren sind nur minimale Kenntnisse über den zu analysierenden Antikörper notwendig, sodass dieses Verfahren auch hervorragend auf bisher nicht charakterisierte Antikörper angewendet werden kann. The aim of this work is the characterization of an antibody. The used human IgG1-antibody was successfully investigated with various mass spectrometric methods. In addition to the determination of the molecular masses of the analyzed Anti-Interleukin-8 antibody itself and its fragments the formation of oligomers was examined. Different oligomers up to pentamers could be mass spectromatrically measured. Resulting from this, it was possible to find that concentration effects are the main factors for the oligomer formation of this particular antibody. The glycosylation of antibodies, a prominent and high diverse modification, was also found in this antibody and was measured in the intact molecule, in its fragments and also the dimer. Next to this native modification, also a derivatization of the antibody was investigated. Different metal containing modifications were analyzed. Thereby it was possible to determine various tagging degrees, even in a mixture of these metal containing modifications. The important question, whether the tagged antibody is still active and capable for binding, can be answered positively. Furthermore the formation of antibody-protein complexes could be shown via mass spectrometry. It was also successful to develop a quantification strategy for this antibody. With this an absolute quantification was possible, which was independent from the type and the structure of the used standard. With this method it is possible to overcome the problems of the Bradford assay. In addition to this only very little information about the antibody is needed for the quantification, which allows to use this method for non characterized antibodies.

Verunreinigungen von Fremdelementen können den Wirkungsgrad von Solarzellen schon im Spurenbereich beeinträchtigen. Die Kenntnis der Verunreinigungen in Si ist entscheidend für die Produkt- und Produktionskontrolle neuer Solarzellenmaterialien. In dieser Arbeit wurden Analysenmethoden mit unterschiedlichen Messverfahren unter besonderer Berücksichtigung der Ansprüche der Solarindustrie entwickelt, verbessert, charakterisiert und verglichen. Mit der Sektorfeld-Massenspektrometrie (SFMS) mit induktiv gekoppeltem Plasma (ICP) nach Matrixabtrennung konnten 22 Elemente mit Bestimmungsgrenzen bis zu 120 pg g−1, quantifiziert werden. Das neue Verfahren erlaubte die Bestimmung aller Elemente in einem Analysengang ohne Analytverlust. Dabei wurde ein bisher in der Literatur nicht beschriebener Mechanismus aufgeklärt, welcher die Retention von Bor im Matrixverdampfungsschritt ohne Zusatz von Komplexbildnern erlaubt. Mit der Glimmentladungs-(GD )MS wurden 32 Elemente bis in den sub ng g−1-Bereich bestimmt. Es gelang, relative Empfindlichkeitsfaktoren zur Quantifizierung von B, P, As, Ga, Ge und Fe zu errechnen. Methoden basierend auf der elektrothermischen Verdampfung (ETV), gekoppelt an ICP-MS und ICP-Emissionsspektroskopie (OES) sowie Gleichstrombogen-OES wurden zur Charakterisierung von metallurgischem Si-Pulver mit Gehalten im µg g−1-Bereich verwendet. Die Totalreflexion-Röntgenfluoreszenz wurde als Volumenmessmethode für die Si-Analytik eingesetzt. Komplettiert wird das Methodenspektrum durch Oberflächenanalytik von Wafern mittels Laserablation-(LA)-ICP-MS. Es wurden erstmalig für Silicium Konzepte zur quantitativen Bestimmung der Verunreinigungen auf Si-Wafern über eine Kalibrierung der LA mit eingetrockneten flüssigen Standards erarbeitet und gezeigt, dass sich das Verfahren zum Nachweis typischer metallischer Ausscheidungen eignet. Die Neutronenaktivierungsanalyse wurde als anerkannte Referenzmethode der Halbleiterindustrie zur Validierung der Methoden eingesetzt. Element impurities can affect the efficiency of solar cells already on the trace level. The knowledge of the impurities in Si is thus crucial for the product and production control of new solar cell materials. In this work, analysis methods based on different measurement principles have been developed, improved, characterized and compared with special consideration of the requirements of the solar industry. Sector field mass spectrometry (SFMS) with inductively coupled plasma (ICP) subsequent to matrix separation has been used to determine 22 elements with limits of determination down to 120 pg g−1 on sample basis. The new, optimized procedure allowed the determination of all analytes in one sweep without analyte loss during the evaporation step. A so-far unexplained mechanism for the retention of boron without use of additional complexing agents was elucidated. Glow discharge (GD)MS was used to measure 32 elements down to the sub-ng g−1 range. Relative sensitivity factors for the quantification of B, P, As, Ga, Ge and Fe have been calculated. Methods based on electrothermal vaporization (ETV) coupled to ICP-MS and ICP emission spectroscopy (OES) as well as direct current arc OES were used for the characterization of metallurgical grade Si powder with concentrations in the µg g−1 range. Total reflection X-ray fluorescence was used as a method for bulk impurity concentration analysis. The spectrum of methods is complemented by surface analysis of silicon wafers by laser ablation (LA)-ICP-MS. New concepts for quantitative analysis of silicon surfaces by the calibration of LA with dried liquid standards were elaborated. It has been demonstrated that this method is suitable for the detection of typical metallic precipitations in silicon like copper silicide. For validation of the methods, instrumental neutron activation analysis was used as the generally accepted reference method in the semiconductor industry.

Die von der caprotec bioanalytics GmbH entwickelte und vermarktete Capture Compounds Mass SpectrometryTM (CCMS-Technologie) ermöglicht die spezifische Isolierung und Identifizierung von Proteinen, basierend auf ihre gezielte Wechselwirkung mit kleinen trifunktionalen Moleküle (sog. Capture CompoundsTM). Dadurch wird sowohl eine Reduzierung der Komplexität zellulärer Proteingemische als auch eine Anreicherung niedrig abundanter Proteine erreicht. In der vorliegenden Arbeit erfolgte die Synthese von neuartigen Capture Compounds, die dazu dienen sollen, unterschiedliche biochemische Fragestellungen zu beantworten und das Anwendungsspektrum der CCMS-Technologie stark erweitern. The Capture Compound Mass SpectrometryTM (CCMS-Technology) is a novel technology developed and marketed by caprotec bioanalytics GmbH. CCMS allows the isolation of sub-proteomes based on specific interactions of target proteins with synthetic small molecules, called Capture Compounds (CCs). In this way, CCMS affords the functional reduction of complex protein mixtures that derived from e.g. cell lysates, and the enrichment and identification of low abundant proteins. In the present work, the synthesis of different novel Capture Compounds that helps studying different biological problems and overcoming present technological limitations of CCMS is described.

Die Phosphorylierung als posttranslationale Modifikation von Proteinen spielt bei Signalwegen in Zellen eine große Rolle. Für die Entschlüsselung des molekularen Mechanismus der circadianen Uhr ist es daher von Interesse, die Phosphorylierungsstellen beteiligter Proteine zu identifizieren. Im Rahmen dieser Arbeit wurden die Proteine Period I und II mittels Flüssigkeitschromatographie und Tandemmassenspektrometrie (LC-MS/MS) auf Phosphorylierungsstellen untersucht. Hierbei lag der Fokus auf der Verbesserung bestehender Methoden, um eine bessere und umfassendere Identifizierung der Phosphorylierungsstellen, insbesondere in Bezug auf mehrfach phosphorylierte Peptide, zu erreichen. Der Arbeitsablauf beinhaltete die Verwendung mehrerer Proteasen, um eine hohe Sequenzabdeckung des Proteins zu erreichen. Nach der Proteolyse wurden die Phosphopeptide mittels Titandioxid angereichert. Hierbei und bei der LC-MS/MS-Analyse wurde Citrat als Additiv verwendet, welches eine bessere Chromatographie multiphosphorylierter Peptide ermöglicht. Bei der MS/MS-Analyse wurden CID und ETD als Fragmentierungsmethoden eingesetzt. Es konnten durch diese Methodik 30 bzw. 42 Phosphorylierungsstellen an den Proteinen Period I und II identifiziert werden, von denen 26 bzw. 14 zuvor nicht beschrieben waren. Nach der qualitativen Identifizierung wurden quantitative Varianten der optimierten Analytik untersucht, um die biologische Funktion der gefundenen Phosphorylierungsstellen untersuchen zu können. Hierbei wurden das metabolische Labeling der Zellen mit 15N-stickstoffhaltigen Aminosäuren und die säurekatalysierte Isotopenmarkierung auf Peptidebene mit 18O-Sauerstoff untersucht. Mit einer optimierten Variante der säurekatalysierten Isotopenmarkierung mit 18O-Sauerstoff lassen sich die Carboxygruppen der Peptide in 5h 30min mit einer Rate von >97% 18O-Sauerstoff markieren. Mit dieser Methode können weitere funktionelle Untersuchungen der Phosphorylierung an den Period-Proteinen durchgeführt werden. Protein phosphorylation, a posttranslational modification, plays an important role in signal cascades in cells. In order to understand the molecular mechanism of the circadian clock, it is thus of interest to identify the phosphorylation sites on proteins contributing to the system. During the work for this thesis, the proteins Period I and II were analyzed for phosphorylation sites with liquid chromatography and tandem mass spectrometry (LC-MS/MS). Hereby the focus was on improving existing methods in order to better identify multi-phosphorylated peptides. In the workflow, the Period proteins were digested with several proteases in order to archive a high sequence coverage for analysis. After proteolysis the phosphopeptides were subsequently enriched with titanium dioxide. During phosphopeptide enrichment and reversed phase chromatography, citrate was used as an additive for a better chromatography and recovery of multiphosphorylated peptides. During LC-MS/MS analysis, CID and ETD were used as fragmentation mechanisms in the mass spectrometer. Using these methods, 30 and 42 phosphorylation sites could be identified on the proteins Period I and II, respectively, including 26 and 14 which were previously unpublished. In order to unravel the biological function of these phosphorylation sites, quantitative methods for the optimized LC-MS approach were investigated. This included the metabolic labeling of cells with amino acids containing 15N-nitrogen as well as acid catalyzed 18O-oxygen labeling on peptide level. The developed optimized variant of acid catalyzed 18O-oxygen labeling achieves an inclusion of 18O-oxygen at the peptide carboxy groups with a rate of >97% in 5h 30min. This method can be used for further investigation of the biological function of the phosphorylation on the Period proteins.

Bdellovibrionaceae sind außergewöhnliche Bakterien, da sie als die kleinsten bekannten räuberischen Organismen gelten. Seit ihrer Entdeckung in Bodenproben im Jahr 1962 durch Stolp und Starr konnte eine weite Verbreitung in der Natur nachgewiesen werden. Besonderes Interesse erlangten Bdellovibrionaceae durch ihre Fähigkeit bakterielle Erreger wie Escherichia coli, Salmonella und Yersinia zu attackieren. Der bestuntersuchte Vertreter der Familie Bdellovibrionaceae ist Bdellovibrio bacteriovorus. Er durchläuft einen komplexen Lebenszyklus, der nur partiell verstanden ist. In der Attack-Phase werden durch einen noch nicht aufgeklärten Mechanismus potentielle Beutebakterien erkannt und der interzelluläre Kontakt hergestellt. Nachdem eine Pore in der Zellwand der ausschließlich Gram-negativen Beutebakterien erzeugt wurde, dringt B. bacteriovorus in den periplasmatischen Raum ein. Nach etwa 30 Minuten ist der Invasionsprozess abgeschlossen und man kann die Ausbildung sphärischer Bdelloblasten beobachten. Im Beutebakterium verdaut B. bacteriovorus makromolekulare Bestandteile des Wirtes und wächst zu einem langen spiralförmigen Stäbchen aus. Es setzt schließlich die Zellteilung ein bei der 5 bis 30 Tochterzellen in Abhängigkeit zur Größe des Beutebakteriums freiwerden. Mit der Reproduktion ist der parasitäre Lebenszyklus nach etwa 3 Stunden beendet. Die komplexe Regulation der Expression von Proteinen konnte für die einzelnen Wachstumsphasen durch ein- sowie zweidimensionale Gelelektrophorese nachgewiesen werden. In der Vergangenheit haben verschiedene Studien die Komplexität der Wechselwirkung mit den Beutebakterien belegt. Hierbei wurde, neben dem offensichtlichen Abbau makromolekularer Bestandteile der Beutebakterien, ein Recycling und Einbau von Membranbestandteilen wie Lipopolysacchariden (LPS) und Outer Membrane Proteine (OmP) in das Membransystem von B. bacteriovorus diskutiert. Die biologische Interpretation dieses Phänomens ist eine erhöhte Effizienz in der Reproduktion von B. bacteriovorus wenn es Bestandteile des Wirtsbakteriums wiederverwertet im Gegensatz zur Eigensynthese. Trotz der morphologischen Ähnlichkeit und des ungewöhnlichen Lebensstils wurde festgestellt, dass die Familie der Bdellovibrionaceae phylogenetisch sehr heterogen ist. Es werden zur Zeit zwei Gattungen unterschieden Bdellovibrio und Bacteriovorax. Aufgrund von 16S rRNA Untersuchungen konnten auch innerhalb der Gattungen eine Vielzahl von phylogenetisch distinkten Arten nachgewiesen werden. In der Literatur wird eine regulierende Wirkung auf pathogene Gram-negative Bakterien für aquatische Systeme durch Bdellovibrionaceae beschrieben. Weiterhin konnten sie bei verschiedenen Nutztieren im Verdauungstrakt mikrobiologisch nachgewiesen werden. Ein positiver Einfluss auf die Gesundheit der Tiere wurde bei Vorkommen von B. bacteriovorus festgestellt. Für eine Charakterisierung von Umweltisolaten werden in der vorliegenden Arbeit genetische Methoden vorgestellt. Hierfür wurden Hybridisierungsmethoden und PCR-methoden entwickelt, die es ermöglichen aus der Umwelt isolierte Bdellovibrionaceae phylogenetisch einzuordnen. Es konnte gezeigt werden, das sowohl B. bacteriovorus als auch Bacteriovorax stolpii im Verdauungstrakt von Nutztieren vorkommen. Es ist weiterhin gelungen eine PCR-methode für Kotproben zu entwickeln, die einen direkten Nachweis ermöglicht ohne mikrobiologische Anzucht. B. bacteriovorus ist ein Gram-negatives Bakterium, allein dieser Sachverhalt spiegelt die Schwierigkeit wider, wenn der enzymattische Abbau von Zellwandbestandteilen der Beutebakterien Ziel der Untersuchung ist, da auch die Beutebakterien Gram-negativ sind. Es ergibt sich aufgrund eines ähnlichen Zellwandaufbaus ein immenses analytisches Problem die makromolekularen Bestandteile von Wirtsbakterium und Jäger zu trennen. Um dem Lebensstil angepasst Modifikationen von B. bacteriovorus zu untersuchen, wurde in dieser Arbeit LPS, charakteristischer Bestandteil der Äußeren Membran, von Wildtypstamm B. bacteriovorus HD100 und seiner wirtsunabhängigen Mutante HI100 isoliert und das Lipid A strukturell aufgeklärt. Die Isolation gelang durch die Ausnutzung unterschiedlicher Fällungseigenschaften des LPS von B. bacteriovorus HD100 gegenüber des E. coli K-12 LPS aus dem Extraktionsmittel. Weiterhin konnte nachgewiesen werden, das B. bacteriovorus S-Form LPS besitzt. Die außergewöhnliche Struktur des Lipid A von B. bacteriovorus wurde im Detail mit massenspektrometrischen Methoden, ein- und zweidimensionalen NMR Methoden sowie mikrochemischer Methoden in Kombination mit der GC/MS charakterisiert. Der Fettsäureanker besteht aus a-D-ManpII-(1(R)4)-ß-D-GlcpN3NII-(1(R)6)-ß-D-GlcpN3NI-(1(R)1)-a-D-ManpI. Dies stellt eine neuartige Struktur dar, da an allen bisher bekannten Lipid A´s sich Brönstedtsäuren am hydrophilen Fettsäureanker befinden, die in physiologischer Lösung durch Protonenabgabe negative Ladungen tragen. Im Lipid A von B. bacteriovorus sind diese Substituenten durch den Neutralzucker Mannose ersetzt. Weiterhin wurden als Fettsäuren nur 3-Hydroxyfettsäuren nachgewiesen, wobei sich auf die 6 gebunden Fettsäuren etwa 1,5 Doppelbindungen verteilen. Dieses Lipid A zeigt eine wesentlich reduzierte endotoxische Aktivität im Vergleich zu E. coli Lipid A und weist als biophysikalische Besonderheit eine erhöhte Fluidität über einen weiten Temperaturbereich auf. Die vorgestellte 16S rRNA Analyse und die Strukturanalyse des Lipid A von B. bacteriovorus belegen seine besondere Stellung in der Welt der Bakterien., Bdellovibrionaceae are extraordinary bacteria known as the smallest predatory organism so far. Since their discovery in soil samples by Stolp and Starr 1963 they have been detected in a wide range of other natural habitats. Bdellovibrionaceae became the focus of attention concerning their ability to attack pathogens like Escherichia coli, Salmonella and Yersinia. For Bdellovibrio bacteriovorus the most detailed studies are available. The up to now only partially understood lifecycle consists of several complex phases. In the attack phase B. bacteriovorus is motile possessing flagella and the preys are recognized by an unknown mechanism. After attachment on the cell wall within 15 to 30 minutes a pore is formed which is used as entrance to the periplasmatic space of the Gram-negative prey bacteria. The completion of the invasion process can be observed by the change of the prey s shape to spherical bdelloblasts. Inside of the prey B. bacteriovorus degrades macromolecular compounds and transforms into a long spirally shaped rod. At the end of the lifecycle the rod divides yielding 5 up to 30 daughter cells depending on the size of the prey bacteria. This reproduction phase is completed within 3 hours. The complex regulation of expression of a certain number of proteins was observed by one and two dimensional gelelectrophoresis. The complexity of the interaction between predator and prey were examined in several studies. Besides the obvious degradation of macromolecular compounds of the prey, reutilising of lipopolysaccharides (LPS) and Outer Membrane Proteins (OmP) into the membrane system of B. bacteriovorus was discussed. The biological interpretation of such behaviour was that it is more efficient for reproduction to recycle components of the prey than to perform de novo synthesis. Unexpectedly, despite the unique predatory lifecycle and common morphological features, Bdellovibrionaceae show a great phylogenetical diversity based on 16S rRNA analyses. Bdellovibrionaceae are divided into the three species Bdellovibrio bacteriovorus, Bacteriovorax stolpii, Bacteriovorax starrii and some strains yet to be assigned. In two studies Bdellovibrionaceae were found as part of regulation processes for decreasing the number of pathogenic Gram-negative bacteria in aquatic systems. Furthermore, B. bacteriovorus were found in the intestinal tract of several domestic animals showing a positive influence on the state of health. For the phylogenetic characterization of environmental isolates techniques were developed based on hybridisation methods and the PCR. In this work we detected B. bacteriovorus and B. stolpii strains in the gut of animals. Furthermore, a PCR method for direct detection of Bdellovibrionaceae in fecal samples was developed. B. bacteriovorus are Gram-negative bacteria. This fact complicates the study of the degradation of the prey s cell wall as it possesses the architecture of Gram-negative bacteria also. Furthermore, the search for important modifications of the cell wall of B. bacteriovorus concerning the predatory lifestyle becomes an analytical problem. In this study we isolated LPS of the wild type strain B. bacteriovorus HD100 and its host independent mutant strain HI100. For the isolation of pure B. bacteriovorus HD100 S-form LPS we took advantage of different precipitation properties in the extraction solvent of E. coli K-12 LPS and the predator s LPS. The structure of the lipid A was examined in detail by mass spectrometric methods, one- and two-dimensional NMR and chemical analytical techniques. The novel structure consists of backbone built of a-D-ManpII-(1(R)4)-ß-D-GlcpN3NII-(1(R)6)-ß-D-GlcpN3NI-(1(R)1)-a-D-ManpI. This is the first known natural lipid A without negatively charged substituents in physiological solution. The lipid A of B. bacteriovorus carries the neutral sugar mannose instead of Brönstedt acids. Furthermore, the lipid A exclusively consists of 3-hydroxy fatty acids with approximately 1.5 double bounds distributed on six bounded fatty acids.
