We evaluated growth-related responses to ractopamine in steers and heifers. Sixteen Angus steers (512 kg) and 16 Angus heifers (473 kg) housed in individual pens were used in a complete block design. At 90 to 97 d before the experiment, steers were implanted with 120 mg of trenbolone acetate and 24 mg of estradiol-17beta (Component TE-S) and heifers were implanted with 140 mg of trenbolone acetate and 14 mg of estradiol-17beta (Component TE-H). Treatments were arranged as a 2 x 2 factorial and included sex (steer vs. heifer) and ractopamine-HCl (0 or 200 mg/d). Cattle were fed a diet based on steam-flaked corn once daily. Blood and LM and biceps femoris (BF) biopsy samples were collected on d 0 (before ractopamine feeding) and after 14 and 28 d of ractopamine feeding. Serum insulin concentrations were not affected by ractopamine or sex. Serum IGF-I concentrations were greater in steers than heifers (P0.001), and steers demonstrated greater IGF-I mRNA expression in BF than heifers (P = 0.05). Ractopamine decreased serum IGF-I concentrations in heifers on d 14, but increased serum IGF-I concentrations in steers on d 28 (sex x ractopamine x day interaction; P = 0.03). Ractopamine did not affect (Por= 0.19) mRNA expression of IGF-I, IGFBP-3, or calpastatin in BF or LM. However, ractopamine led to increases in LM expression of IGFBP-5 in heifers, but to decreases in expression in steers (ractopamine x sex interaction; P = 0.04). Ractopamine decreased myosin heavy chain IIA mRNA expression in BF (P = 0.04) but not in LM (P = 0.99). Ractopamine decreased beta(2)-receptor mRNA expression in LM of steers on d 14, but not on d 28; in contrast, expression of beta(2)-receptor mRNA in LM of heifers was not affected by ractopamine (sex x ractopamine x day interaction; P = 0.03). Although there were a few criteria for which ractopamine led to differences in response between steers and heifers, there were no striking disparities to suggest that the effectiveness of ractopamine would markedly differ between sexes.

Nutrient digestion and kinetic characteristics of alfalfa hay, brewers grains, beet pulp, and cottonseed hulls were studied in two experiments. In the first experiment, these feedstuffs were fed with a mineral/vitamin supplement in a 4 x 4 Latin square to eight ewes to determine total tract digestibilities by 8-d total collection and rates of passage using Yb as a marker. In the second experiment samples of the same feedstuffs were incubated in situ in four ruminally fistulated steers in a 4 x 4 Latin square design to quantify ruminal disappearance of NDF and ADF. Three Dacron bags were placed in the rumen for each of six time periods: 6, 12, 24, 48, 72, and 96 h. Total tract digestibilities of DM ranged from 33% for cottonseed hulls to 78% for beet pulp. Digestibilities of NDF ranged from 32% for cottonseed hulls to 81% for beet pulp. Total tract retention times ranged from 74 h for beet pulp to 44 h for alfalfa. Retention time for alfalfa was lower (P < .05) than those for beet pulp or cottonseed hulls. Calculation of TDN values yielded values similar to those of NRC other than for cottonseed hulls, for which NRC values were 18 to 29% higher. Alfalfa hay and beet pulp were the feedstuffs with the most rapidly disappearing NDF, .124/h for alfalfa and .116/h for beet pulp, which were faster (P < .05) than those found in brewers grains and cottonseed hulls, .035 and .043/h, respectively. Potentially digestible NDF ranged from 55% for alfalfa hay to 94% for beet pulp.(ABSTRACT TRUNCATED AT 250 WORDS)

To determine the effects of feeding zilpaterol hydrochloride (ZH) for 0, 20, 30, or 40 d (ZH0, ZH20, ZH30, ZH40) on semimembranosus (SM) steak color and color stability in 3 packaging systems, SM subprimals were removed from 60 calf-fed Holstein steers 24 h postmortem. A 7.62-cm-thick portion was removed from each subprimal and stored (2 degrees C) for 21 d; then two 2.54-cm-thick steaks were cut, overwrapped with polyvinyl chloride (PVC) film, and assigned to 0 or 3 d of display. Remaining portions of the subprimals were vacuum packaged for 10 d and then enhanced (10% with a solution containing 0.3% sodium chloride, 0.35% phosphate, and 0.05% rosemary extract), cut into steaks, packaged in high-oxygen (HO-MAP) or carbon monoxide (CO-MAP) modified atmosphere packaging (MAP), and assigned to 0, 3, or 5 d (HO-MAP) or 0 or 9 d (CO-MAP) of display. Panelists evaluated the deep and superficial portions of SM steaks for initial color, display color, discoloration, pH, L*, a*, b*, hue angle, and saturation indices. Feeding duration did not affect (P0.05) initial color scores of steaks in PVC. Steaks displayed in PVC from ZH20 or ZH30 diets were slightly brighter and less discolored than the ZH40 treatment. For enhanced steaks in HO-MAP, ZH20 steaks were darker on d 5 (P0.05) and more discolored (P0.05) on d 3 through 5 than all other diet treatments. For enhanced steaks from steers fed ZH40 and in CO-MAP, the deep and superficial SM tended (P0.05) to have improved display color compared with other dietary regimens; however, steaks in CO-MAP from all feeding durations had less than 20% metmyoglobin through d 9 of display. Overall, feeding ZH20 might result in steaks with slightly less color stability when packaged in HO-MAP; however, feeding ZH20 or ZH30 to calf-fed Holstein steers will yield steaks that have equal to or more desirable color traits when packaged in PVC or CO-MAP. Regardless of ZH feeding regimen, HO-MAP and CO-MAP extended the color life of the SM. The CO-MAP system minimized color differences between the superficial and deep portions of the SM muscle and extended total case life compared with traditional and HO-MAP packaging.

The deep (DSM) and superficial (SSM) portions of steaks were evaluated for initial color, display color, discoloration, pH, L*, a*, b*, hue angle, and saturation indices. For steaks in PVC, no differences (P0.05) occurred in initial or discoloration color scores because of ZH feeding duration. The enhanced SSM steaks from ZH20 in PVC were brighter red (P0.05) than SSM steaks from ZH40 in PVC. The DSM in PVC had less (P0.05) pH and paler (P0.05) color than the SSM. Display color scores for the DSM of PVC steaks were brighter red (P0.05) than the SSM initially (d 0 and 1), but the DSM discolored faster (P0.05) than the SSM on d 1 to 3. The SM steaks from steers fed ZH20 or ZH30 were slightly brighter and less discolored during display in PVC than the ZH40 diet. For enhanced steaks in HO-MAP, the DSM of ZH20 and ZH30 diets displayed 4 d and the DSM of ZH20 displayed 5 d was a brighter (P0.05) red than the DSM from ZH40. At display d 1 and 5, the SSM of ZH20 steaks in HO-MAP was a brighter (P0.05) red than SSM steaks from ZH40. The SSM of ZH40 HO-MAP steaks was darker (P0.05) red on d 3 than the SSM from other diets. For enhanced steaks in CO-MAP, ZH30 steaks were brighter (P0.05) red than ZH0 or ZH40 steaks on d 0 and 9 of display. Steaks in CO-MAP from all feeding durations were less than 20% discolored through d 9. The DSM was lighter (P0.05) than the SSM on d 0 for steaks packaged in HO-MAP and CO-MAP. Feeding cattle ZH for 20 or 30 d will yield steaks with color characteristics equal to or better than steaks from control cattle, whereas feeding ZH for 40 d will likely produce less desirable meat color traits.

Effects of glycine (Gly) and B-vitamins on sulphur amino acid (AA) utilization were studied in growing steers maintained under conditions where methionine (Met) was first limiting. Conditions were generated by limit feeding a diet low in ruminally non-degraded protein and abomasally infusing an AA mixture limiting in Met. Retained N tended (p = 0.07) to improve when steers received 10 mg folate, 10 mg vitamin B6, and 0.10 mg vitamin B12 daily. Hepatic vitamin B12 (p = 0.08) and folate (p = 0.05) concentrations increased with vitamin supplementation. In another trial, factorial treatments were 2 or 5 g/day L-Met and 0 or 50 g/day Gly infused abomasally. Retained N increased (p < 0.05) in response to Met, and responses were numerically larger in the presence of supplemental Gly. In a different trial, factorial treatments were 0 or 2.4 g/day L-cysteine (Cys) and 0 or 40 g/day Gly. Retained N was not affected by Cys in the absence of Gly, but was increased by Cys when Gly was supplemented (interaction, p = 0.01). B-vitamin status may affect sparing of Met by Cys. Supplemental Gly improved responses to supplemental Met and Cys.

The effect of altering supplementation frequency on host N balance and key N transactions in the ruminal ecosystem were monitored. Four ruminally fistulated beef steers (BW = 513 kg; SEM = 6.5) were used in a 2 x 2 crossover design with two periods and two supplementation frequency treatments. Supplementation frequencies were 2 and 7 d/wk. Steers were fed tallgrass prairie hay (73.1% NDF, 5.3% CP) ad libitum. Supplement (42% CP; DM basis) was fed at 0.36% BW/d to steers supplemented 7 d/wk. Steers supplemented 2 d/wk received the same amount of supplement per week, but it was equally split among the two supplementation events. Steers supplemented 7 d/wk had higher forage (P0.02) and total digestible OM intake (P0.06), total N intake, fecal N excretion, and N retention. Although both supplementation frequencies were characterized by positive N balance, the decrease in N retention in the steers supplemented 2 d/wk was due to higher (P0.01) urinary N loss. Ruminal fluid was sampled at 0, 2, 4, 6, 12, 24, 48, and 72 h after supplementation beginning on a day when both treatments were supplemented. Frequency x hour interactions (P0.02) were observed for ruminal N metabolism criteria. Counts of peptide- and AA-fermenting bacteria peaked at 2 h and returned to nadir by 12 h for steers supplemented 7 d/wk. Steers supplemented 2 d/wk peaked at 6 h with a greater population and returned to nadir at 48 h. Ruminal ammonia concentrations followed a similar trend. Specific activity of ammonia production was lower (Por = 0.05) immediately after supplementation for steers supplemented 2 d/wk, but by 12 h was the same as for 7 d/wk steers. Ruminal peptides and free AA peaked at 2 h for steers supplemented 2 d/wk and were generally higher (Por = 0.05) during the first 6 h compared with steers supplemented 7 d/wk. Total VFA concentration was not different (P = 0.35) due to supplementation frequency. Frequency x hour interactions (P0.01) were observed for all molar proportions of VFA. The molar proportion of acetate and acetate:propionate ratio were lower (P0.01) and the molar proportions of propionate and butyrate were higher for steers supplemented 2 d/wk from 4 h to 24 h. In conclusion, forage use and N balance improved with supplementation 7 d/wk, but supplementation 2 d/wk was associated with some desirable shifts in select ruminal events that may contribute to moderating potential negative impacts of supplementing infrequently.

To measure prostatic and blood fatty acid composition in a large group of patients undergoing prostatectomy for benign or malignant prostate disease, as there is evidence linking arachidonic acid metabolism and prostate cancer through its role as an eicosanoid precursor, and earlier studies showed lower prostatic arachidonic acid content in a few patients.Prostatic phospholipid fatty acid composition was determined in prostate tissue from 173 patients undergoing prostate surgery, i.e. radical prostatectomy, cystoprostatectomy or transurethral resection (TURP). Blood fatty acid composition was determined in 99 of these patients and in 85 undergoing prostatic needle biopsy.There was a significantly lower percentage of arachidonic acid in malignant than in benign portions of the prostate (15.2% vs 17%) in all patients assessed. The changes were greatest in those undergoing TURP for known prostate cancer (13.4% vs 17.2%), these patients having the greatest proportion of malignancy in the specimens. There were no consistent changes in blood fatty acid composition.This is the first prospective study of arachidonic acids levels involving many consecutive patients undergoing prostate surgery for either benign or malignant disease. The lower prostatic arachidonic acid level is probably a result of the increased use of arachidonic acid for producing prostaglandins and/or leukotrienes. Further understanding of the cause and/or consequence of this finding might lead to a better understanding of prostate cancer.

Experiments were conducted to determine the effects of novel anti-neoplastic isochalcones (DJ compounds), on cyclooxyegenase 1 and 2 (COX-1 and COX-2) enzyme expression in androgen receptor dependent human prostate cancer cell line LNCaP. Results from Western blot analysis and cell flow cytometry showed that DJ52 and DJ53 decreased the steady state levels of COX-1 and COX-2 protein levels in a dose dependent manner. In addition, DJ52 and DJ53 decreased the levels of epidermal growth factor (EGF) in LNCaP cells. In this study, we report that novel isochalcones decreased COX-1, COX-2 and EGF levels as well as LNCaP cellular growth in a dose responsive manner. Our findings indicate that relative decreases in COX-1, COX-2 and EGF expressions might serve as indicators of tumor growth inhibition in prostate neoplasms.

In this report we describe a prenatally diagnosed case with four-limb ectrodactyly and cleft lip/palate. The family history reveals three-generation oligodontia. The difficulties in counseling of the families with EEC syndrome are discussed.

We compare the efficacy of 4 versus 8 mg. doxazosin for benign prostatic hyperplasia.A total of 82 patients with benign prostatic hyperplasia successfully treated with 4 mg. doxazosin were randomized in a double-blind fashion to take 4 or 8 mg. Patients were evaluated with American Urological Association (AUA) symptom score, Boyarsky score, uroflowmetry and side effect profile before, and 1 and 3 months following randomization.Of the patients 42 and 40 were randomized to receive 4 and 8 mg. doxazosin, respectively. Both groups were similar with respect to patient age, baseline Boyarsky and AUA symptom scores, and baseline maximum urinary flow rate. At 3 months mean improvement from baseline plus or minus standard deviation in Boyarsky score was 0.6 +/- 6.5 and 4.9 +/- 6.6 in the 4 and 8 mg. groups (p0.05), respectively, mean improvement in AUA symptom score was 1.6 +/- 5.3 and 5.3 +/- 8.0 (p0.05), and mean maximum flow rate difference was -0.6 +/- 6.4 and +1.4 +/- 7.9 (p0.05). Of the patients 7 and 8 in the 4 and 8 mg. groups dropped out of the study, and there were no statistical differences in side effects between dosages.A dose of 8 mg. doxazosin was more efficacious than 4 mg. and the side effects associated with both dosages appeared to be similar. The 8 mg. dose should be tried in patients who have not achieved an adequate therapeutic response to 4 mg. and are tolerating the medication. Consideration should be given to increasing the dosage to 8 mg. in patients who are clinically improved at lower dosages.

Aberrant regulation of apoptosis may contribute to tumorigenesis. Relative levels of apoptosis regulatory proteins, such as Bcl-2 and Bax as well as interactions of these proteins with other gene products, may contribute to the rate of apoptosis in neoplasia. We examined Bcl-2 expression in 104 squamous cell carcinomas of the head and neck, as well as histologically normal mucosa several centimeters away from the tumor, and in control normal mucosa from patients without cancer. Immunohistochemistry and immunoblotting demonstrated Bcl-2 expression in 30% (31 of 104) of squamous cell carcinoma, with an increase in Bcl-2 protein levels compared with control normal mucosa from noncancer patients. Bcl-2-positive tumors demonstrated a 5-fold decrease in the number of apoptotic cells compared with Bcl-2-negative tumors. Bcl-2 protein expression was associated with poorly differentiated tumor grade but was not correlated with Bax expression or patient survival. These findings demonstrate that Bcl-2 contributes to apoptosis in normal and transformed squamous epithelium.

Two groups of patients with adenocarcinoma of the pancreas treated with either preoperative chemoradiation (preop CTRT) or postoperative chemoradiation (postop CTRT) were retrospectively analyzed for various treatment-related parameters. Between November 1986 and October 1996, a total of 70 patients with pancreatic adenocarcinoma were enrolled into preop CTRT protocols at our institution. Twenty-five patients with adenocarcinoma of the head of the pancreas underwent pancreaticoduodenectomy with curative intent. After the closure of the preop CTRT protocols, we had the opportunity to perform 23 pancreatic resections without preop CTRT. After surgery, these patients were advised to undergo CTRT. These two groups of patients were therefore selected consecutively, dependent only on the time of referral and no other bias. These two cohorts of patients are compared for various intraoperative parameters, length of hospital stay, pathologic findings, time to recurrence, and survival. Mean age was 65 and 66 years in the preop and postop CTRT groups, respectively. Sex distribution was almost equal. Treatment breaks resulting in greater than 1 week delay in the radiotherapy occurred in 2 (8%) of 25 patients in the preop CTRT group (myelotoxicity in 1 case and biliary sepsis in 1 case), whereas no treatment breaks1 week occurred in those receiving postop CTRT. Eleven patients in preop CTRT had grade 3 or 4 toxicity, whereas none was noted in those with postop CTRT. There was one postoperative death in the preop CTRT group and none in the postop CTRT group. Mean time to the start of CTRT was 45 days (range, 20-66 days) after pancreaticoduodenectomy. Delay of60 days to the onset of CTRT occurred in 2 (22%) patients and was attributable to patient delays in time to recover from surgery or patient noncompliance. Furthermore, 5 of 23 patients (22%) in the postop CTRT group did not receive treatment for various reasons. Average estimated operative blood loss was 1933 mL (median 1550) and 1060 mL (median 1000) for preop and postop CTRT groups, respectively. Mean length of operation was 488 minutes (median 480) and 486 minutes (median 480). Median length of postoperative stay was 22 and 20 days (ranges, 9-144 and 10-38). Pathological findings in the resected specimens showed significantly fewer involved nodes in the preop CTRT group (28 vs 87%; P = 0.0006), whereas similar numbers of nodes/patient were counted in each group (14 vs 22, P = 0.11). More negative resection margins were observed in the preop CTRT group (28 vs 56%; P = not significant). A significantly greater amount of fibrosis replacing the tumor was observed in the preop CTRT group (70 vs 40%; P = 0.0001). There were no significant survival differences observed (median 20 months vs 25 months; P = 0.48), in follow-up that ranged from 4 to 76 months (median 44 months for surviving patients) for the preop group and 4 to 40 months (median 16 months for surviving patients) for those with postop CTRT. Local failure either alone or as a component of distant failure occurred in 16 per cent (4 of 25 patients) with preop CTRT and 16.6 per cent (3 of 18) with postop CTRT. Analysis of differences between those treated with preoperative and postoperative CTRT demonstrates similarity in toxicity and effects. However, 22 per cent of patients intended for postoperative therapy did not receive treatment.

Six sets of four genetically identical Brangus steers (n = 24; X BW 409 kg) were used to determine the effect of different anabolic implants on visceral organ mass, chemical body composition, estimated tissue deposition, and energetic efficiency. Steers within a clone set were randomly assigned to one of the following implant treatments: C, no implant; E, estrogenic; A, androgenic, or AE, androgenic + estrogenic. Steers were slaughtered 112 d after implanting; visceral organs were weighed and final body composition determined by mechanical grinding and chemical analysis of the empty body. Mass of the empty gastrointestinal tract (GIT) was reduced approximately 9% (P.10) in steers implanted with estrogen alone or in combination with an androgen. Liver mass was increased (P.10) from 6 to 14% by implants. Steers implanted with the AE combination had greater (P.10) daily protein accretion (163.4 g/d) than either E (128.8 g/d) or A (137.1 g/d), and, because the combination improved gain above C (101.1 g/d), this demonstrates the additive effects of a combination implant on protein deposition. Anabolic implants did not alter (P.10) the efficiency of ME utilization. In general, estrogenic implants decreased GIT, androgenic implants increased liver, and all implants increased hide mass. Steers implanted with an AE combination had additive effects on protein deposition compared with either implant alone. The NEg requirements for body gain are estimated to be reduced 19% by estrogenic or combination implants.

Children born in the countries of the former Soviet Union and Eastern Europe are now a main source of international adoptions in the United States, but often little information is available about these children prior to adoption.To analyze the preadoptive medical reports of children adopted from Eastern Europe and the former Soviet Union and to compare these reports with their evaluations after arrival in the United States.Case series.A total of 56 children adopted from Eastern Europe and the former Soviet Union were evaluated in 2 international adoption clinics. Preadoptive medical records were available for 47 of these children.There were 43 (91%) of 47 medical reports available from the children's birth countries that included multiple unfamiliar neurologic diagnoses. Evaluations in the International Adoption Clinics frequently revealed growth delays (zscoreor = -1 for weight in 44% of children, height in 68%, and head circumference in 43%). Children had 1 month of linear growth lag for every 5 months in an orphanage (r=-0.48, P.001). Developmental delays were also common (gross motor delays in 70% of children, fine motor in 82%, language in 59%, and social-emotional in 53%). While serious medical problems were found or corroborated in 11 (20%) of the 56 children evaluated in our clinics, neurologic diagnoses cited in preadoptive medical reports were not confirmed.Preadoptive medical records from these international adoptees included multiple diagnoses suggesting severe neurologic impairment. Although these diagnoses were not confirmed when the children were evaluated in the United States, substantial growth and developmental delays were identified.

Although the popularity of visual laser ablation of the prostate (VLAP) as a treatment for symptomatic, benign prostatic hyperplasia (BPH) is increasing, the perceived advantages of VLAP over conventional transurethral electroresection of the prostate (TURP) is being debated because optimal technique and dosimetry for surgical lasers are still being refined. At this time, the 1.06 neodymium:yttrium-aluminum-garnet (Nd:YAG) laser and a laterally deflecting delivery system is the hardware combination most widely used for VLAP.Reported here is a study of an alternate system, a 805-nm diode laser (Diomed 25 Diomedics, The Woodlands, TX) with a cylindrically diffusing fiber (Surgimedics Inc., The Woodlands, TX). Eight mongrel dogs were prostatectomized by transurethral irradiation of the prostate with 15,000 J of diode laser energy delivered via a fiber that diffuses the energy in a 1.5-cm-long cylindrical pattern. The dogs were sacrificed and prostates harvested at 3 hours and 1,4,7,14,21,35, and 49 days after the procedure, fixed with 10% buffered formalin, and examined histologically.It was found that this laser/fiber combination created volumes of tissue coagulation similar to those encountered in our previous work with the Nd:YAG laser in combination with both laterally deflecting and diffuser fibers, while offering the distinct advantages of simplified technique, lower cost hardware, and fewer postoperative complications in the dog model.

Aberrant expression of the tumor suppressor gene RB1 is associated with a variety of solid tumors and hematopoietic neoplasms. Certain cancer cell lines in which the protein encoded by RB1 (p110RB) is absent have been reported to show decreased growth rate, clonogenicity, or tumorigenicity following insertion of a transcriptionally active RB1 gene. We asked whether these RB-deficient cells could be growth inhibited by direct exposure to purified p110RB. We report a decrease in uptake of tritiated thymidine by 5637 bladder carcinoma cells (RB-negative) when purified recombinant p110RB is added to culture media. Internalization of the protein by cells and translocation to the nucleus are demonstrated by immunohistochemistry, FACS, and detection of radiolabeled protein in subcellular fractions. Next, we chose a well-described leukemia cell culture model to investigate the potential effect of recombinant p110RB in clinical disease. We observed dose-related decreases in cell number of colony formation in vitro in 8 of 20 acute myelogenous leukemia samples, 7 of which did show endogenous p110RB detectable by immunohistochemistry. Histological appearance following exposure to p110RB shows cytoplasmic vacuolization and nuclear lobulation of degenerating cells. We conclude that purified p110RB added to culture media is internalized by cells, translocated to the nucleus, and exerts a growth-inhibitory effect on certain cancer cell types.

The daily percentage of cells proliferating and dying were determined for normal, premalignant, and cancerous prostatic cells within the prostate as well as for prostatic cancer cells in lymph node, soft tissue, and bone metastases from untreated and hormonally failing patients. These data demonstrate that normal prostatic glandular cells have an extremely low but balanced rate of cell proliferation and death (i.e., both0.20%/day). This results in a steady-state, self-renewing condition in which there is no net growth, although the glandular cells are continuously being replaced (i.e., turnover) every 500 +/- 79 days. Transformation of these cells into high-grade prostatic intraepithelial neoplastic cells initially involves an unbalanced increase in the daily percentage of cells proliferating versus dying, such that net continuous growth occurs (i.e., mean doubling time, 154 +/- 22 days). As these early proliferation lesions continue to grow into late stage high-grade prostatic intraepithelial neoplastic cells, the daily percentage of cells dying increases further to a point equaling the daily percentage of proliferation. This results in cessation of net growth while inducing a 6-fold increase in the turnover time of these cells (i.e., 56 +/- 12 days), increasing their risk of further genetic changes. The transition of late stage high-grade prostatic intraepithelial neoplastic cells into localized prostatic cancer cells involves no further increase in proliferation but a decrease in death resulting in net continuous growth of localized prostatic cancers with a mean doubling time of/=475 days. As compared to localized prostatic cancer cells, metastatic prostatic cancer cells within lymph nodes or bones of untreated patients have an increase in daily rate of proliferation coupled with a reduction in their daily percentage of cell death, producing net growth rates with a mean doubling time of 33 +/- 4 days and 54 +/- 5 days, respectively. Remarkably, there is no further increase in proliferation in hormonally failing patients, but instead an increase in the daily percentage of androgen-independent prostatic cancer cells dying within soft tissue or bone metastases. These changes result in doubling times which are two to three times longer (i.e., 126 +/- 21 and 94 +/- 15 days) in these lymph node and bone metastatic sites, respectively, compared to similar sites in hormonally untreated patients. These data demonstrate that the daily percentage of proliferation for either androgen-dependent or -independent metastatic prostatic cancer cells is remarkably low (i.e.,3. 0%/day), consistent with why antiproliferative chemotherapy has been of such limited value against such metastatic cells. These results also suggest that prostatic carcinogenesis starts in the second to third decade of life and may require over 50 years for progression to pathologically detectable metastatic disease.

Three experiments evaluated whether effects of altered frequency of supplementation on forage use and cow performance depended on supplement CP concentration and (or) grain type when fed to cattle eating low-quality forages. All experiments included supplementation frequency (daily = 7x; three times weekly = 3x) as one factor in a factorialized arrangement of treatments. In Exp. 1 and 2, the second factor was supplement CP concentration (10, 20, 30, and 40% CP), altered by changing the ratio of soybean meal to sorghum grain in supplements. Supplements were fed at 13.9 and 14.1 kg of DM/wk for Exp. 1 and 2, respectively. In Exp. 3, the second factor was supplement grain type (sorghum grain or corn) and supplements containing 21% CP were fed at 14.8 kg of DM/wk. In Exp. 1, eight ruminally fistulated steers (456 kg) consumed wheat straw ad libitum. Pregnant beef cows grazing dormant tallgrass prairie were used in Exp. 2 (475 kg; n = 128) and 3 (504 kg; n = 120). In Exp. 1, reducing supplementation frequency decreased (P < .01) straw intake but increased (P < or = .03) DM and NDF digestion. As CP concentration in supplements increased, straw DMI (P = .06) increased quadratically, whereas DM and NDF digestion increased linearly (P < .01). In Exp. 2, increasing CP concentration in supplements enhanced cows' ability to maintain BW and condition up to calving, with decreasing magnitude of difference between treatments at higher CP concentrations (P < .01). Increasing CP in supplements fed to the dams linearly (P = .05) increased calf weaning weight in Exp. 2. In Exp. 2 and 3, reducing supplementation frequency increased (P < or = .02) winter weight loss through calving. Grain type did not significantly affect most performance variables. In summary, response to supplementation frequency was not dependent on supplement CP concentration or grain type. Daily supplementation maximized forage intake and cow performance, although the magnitude of performance differences was not large.

Histologic analysis of atherectomy samples from400 patients who received directional coronary atherectomy at 3 separate institutions disclosed 2 major categories of tissue: atherosclerotic plaque (with or without thrombus) and intimal proliferation (hyperplasia, with or without thrombus). The predominant tissue type in atherectomy samples from native, primary, or de novo coronary artery stenoses was atherosclerotic plaque. The predominant tissue type in atherectomy samples from restenosis lesions (prior balloon angioplasty, atherectomy, or both) was intimal proliferation with variable amounts of atherosclerotic plaques (with or without thrombus). Deep vessel wall components (media, adventitia) were identified at varying frequencies. The clinical relevance of atherectomy tissue is reviewed.

Five castrated male llamas (mean body weight, 94 kg) were studied in an energy balance trial to determine maintenance energy requirement of llamas. Llamas were fed a 50% oat hay-50% pelleted concentrate diet (2.43 Mcal of metabolizable energy/kg of diet dry matter) at approximately 1.6% of body weight (BW). An 8-day total collection digestion trial was used to determine fecal and urine energy losses. Heat production and methane emissions were determined via indirect respiration calorimetry measurements on each llama fed at the same level of intake as during the digestion trial and subsequently on days 3 and 4 of a period of nonfeeding. Fecal, urine, and methane energy losses of the llamas fed near-maintenance intake were 32.5, 3.5, and 7.1% of gross energy intake, respectively. The postabsorptive metabolic rate, commonly called nonfed (fasting) heat production, was 59.3 kcal/BW0.75. Using a linear relation between postabsorptive and maintenance energy requirement and efficiency of energy use below maintenance of 0.702, metabolizable energy requirement at maintenance was determined to be 84.5 kcal/BW0.75.

Sarcomatoid carcinoma of the prostate is a rare tumor that can be difficult to distinguish from a true sarcoma. The authors report 12 patients in whom the typical light microscopic appearance of prostatic adenocarcinoma was accompanied by the appearance of spindled or pleomorphic sarcomatoid areas within the same specimen or in subsequent accessions. Immunostaining or electron microscopic study demonstrated epithelial differentiation within the sarcomatoid area(s) in 6 of the 11 patients in whom special studies were performed. All nine patients for whom follow-up data were available died of disease within 3 to 48 months (median time until death, 12.0 months) after the appearance of the sarcomatoid carcinoma, and the clinical course in each instance was characterized by aggressive local recurrence. Our experience confirms that sarcomatoid carcinoma of the prostate is an aggressive variant of prostatic adenocarcinoma.

Zoometric measurements and bioelectrical impedance analysis were evaluated as methods of body composition determination in healthy cats. Zoometric and impedance measurements were taken on 22 anesthetized adult cats of various ages, genders, breeds, and body weights. The cats were then euthanatized. The bodies were processed through a tissue homogenizer and free-catch specimens were taken, freeze-dried, and analyzed for total body water, protein, fat, potassium, and ash content. Stepwise regression analysis was implemented to identify statistically significant relationships between the chemically determined dependent variables (total body water, protein, potassium, fat-free mass, fat mass, and percent body fat) and the zoometric measurements, with or without bioelectrical impedance analysis. Statistical analysis revealed high correlations between the dependent variables and the corresponding predicted values of those variables. Body weight alone was a poor predictor of body composition in these cats. On the basis of these findings, we suggest that zoometric and bioelectrical impedance measurements may serve as practical, noninvasive, simple, and accurate methods for estimating body composition in domestic cats.

Small cell carcinoma of the urinary bladder is an uncommon tumor. The authors report the clinicopathologic findings in a series of 22 cases. Fifteen men and 7 women were studied; their ages ranged from 51 to 87 years (mean, 62.4 years). The most frequent presentation was hematuria (94.4%). At diagnosis, three patients had Stage B disease, six had Stage C, and ten had Stage D (unknown stage in three). Histologically, 6 were oat cell type tumors, 11 were of intermediate cell type, and 5 were of combined cell type. Immunohistochemical studies demonstrated positivity for neuron-specific enolase in ten of ten cases, cytokeratin in seven of ten cases, chromogranin in eight of nine cases, serotonin in seven of nine cases, and S-100 protein in four of ten cases. Neuroendocrine differentiation was seen in five of seven cases examined by electron microscopy. Treatment and follow-up data were available for 19 patients: 10 (52.6%) were dead of disease, 5 (26.3%) were alive and well, 3 (15.8%) were alive with disease, and 1 (5.3%) died of an unrelated cause. The 2-year survival rate was 50% for patients with Stage B, 25% for patients with Stage C, and 33% for patients with Stage D disease. Although overall survival was poor, some cases responded well to therapy. Based on the authors' experience, radical cystectomy with adjuvant chemotherapy appears to be the treatment of choice.

Seventy-one patients with pathologic Stage B (P2/3a/N0) transitional cell carcinoma (TCC) of the bladder underwent radical cystectomy alone without preoperative radiotherapy or perioperative chemotherapy between 1983 and 1987 and have been followed a median of fifty months. The five-year actuarial survival and disease-free survival rates were 82 percent and 77 percent, respectively, and only 13 patients (18%) have relapsed. Histologic parameters were evaluated as to prognostic impact; none correlated with disease-free survival rates although the presence of vessel involvement portended a worse disease-free survival rate (68% versus 80%). During this same period, an additional 15 patients underwent radical cystectomy for pathologic Stage B disease but received adjuvant chemotherapy on the basis of vessel invasion. Their disease-free survival rate at five years was 80 percent, comparable to the disease-free survival rate for patients with vessel invasion treated by surgery alone (68%). Although the role of systemic chemotherapy in the management of invasive bladder cancer remains under investigation, it would appear that patients with Stage B TCC are best treated with radical cystectomy alone. Continued analysis of modern surgical results grouped by current pathologic staging criteria is needed to identify patients who have a relatively low risk of relapse and thus little need for additional therapeutic intervention. These results demonstrate that Stage P2/3a/N0 TCC of the bladder is highly curable by surgery.

Normal hematological and blood chemistry parameters were measured in 45 American bison (Bison bison) that were divided into three age groups for comparison. There was a statistically significant (P less than 0.05) increase with advancing age in mean corpuscular volume, mean corpuscular hemoglobin, absolute neutrophil and eosinophil counts, total protein, globulin, creatinine, and blood urea nitrogen. There was a statistically significant (P less than 0.05) decrease with advancing age in levels of sorbital dehydrogenase, alkaline phosphatase, glucose, sodium, calcium and phosphorus.

Binding of the hypoglycemic sulfonylurea, [3H]glyburide, to crude membrane fractions from brain, heart and smooth (intestinal) muscle was saturable, linear with protein concentration and reversible. Saturation analysis revealed high affinity sites (KH values, 7 x 10(-11) M, 5 x 10(-11) M and 6 x 10(-11) M), with Bmax-H values 209, 36 and 23 fmol/mg protein in the brain, heart and smooth muscle, respectively. High affinity [3H]glyburide binding was pharmacologically specific, insensitive to a variety of receptor-active ligands, but sensitive to a series of sulfonylureas, and good, essentially 1:1, correlations were obtained between binding affinities and literature-derived pharmacologic activities. The K+ channel activators, cromakalim, nicorandil, pinacidil and minoxidil were not effective as inhibitors of [3H]glyburide binding. However, diazoxide was a modestly effective inhibitor. Putative low affinity sites (KL values, 3 x 10(-7) M, 1 x 10(-7) M and 2 x 10(-9) M) with Bmax-L values 4956, 336 and 53 fmol/mg protein in brain, heart and smooth muscle, respectively, were identified. Their significance remains to be established. Except for ATP gamma S, the ability of nucleotide triphosphates to inhibit high affinity [3H] glyburide binding was dependent on the presence of Mg++. ADP, in the presence of Mg++, inhibited binding with an IC50 value of 6.3 x 10(-4) M. Nucleotide monophosphates did not inhibit [3H] glyburide binding in the presence or absence of Mg++, whereas in the presence of Mg++, nucleotide triphosphates were equally potent inhibitors of binding. The rank order potency for nucleotide diphosphate inhibition of binding, in the presence of Mg++, is ADP greater than GDP greater than IDP = UDP. In the absence of Mg++, [3H]glyburide binding shows a biphasic response to ADP, and the inhibition of binding by ADP was prevented by ATP. It is suggested that this biphasic response is the result of a second nucleotide binding site.

Necropsy examination of a control Sprague-Dawley male rat from a carcinogenicity study disclosed the presence of a neoplastic mass in the trachea. Microscopic examination disclosed that the tumor consisted of closely packed cells arranged in lobules and cords separated by thin connective tissue stroma. The cells had distinct cell borders and abundant pale cytoplasm. Electron microscopic examination further disclosed that the cytoplasm contained numerous argyrophilic neurosecretory granules measuring 298 nm in diameter. These granules were positive for Grimelius silver stain. The histopathologic and electron microscopic features of this tumor are consistent with a diagnosis of clear-cell carcinoma.

Adenocarcinomas account for approximately 2% of primary epithelial malignancies of the urinary bladder. The clinicopathologic features of 72 cases treated at one institution are reported; 22 cases were evaluated immunohistochemically. Twenty-four tumors were urachal and 48 nonurachal. The cases were analyzed according to their stage at presentation, histologic type, and mucin staining; they were tested immunohistochemically to determine their reaction to carcinoembryonic antigen, Leu-M1, prostate-specific antigen, and prostatic acid phosphatase. Tumor stage was a highly significant predictor of outcome (P = 0.001). Nonurachal tumors tended to have a worse outcome than urachal, but the difference was not statistically significant (P = 0.07). Histologic type was not a significant predictor of outcome (P = 0.10). For adenocarcinoma of the urinary bladder, stage was the most significant predictive factor; separating urachal from nonurachal tumors was important, but mucin histochemistry and immunohistochemistry did not help in this distinction. On occasion, a few tumors may react with some polyclonal antibodies to prostate-specific antigen; thus these results must be interpreted with caution. In these instances, the possibility of using highly sensitive and specific monoclonal antibodies such as the one employed in this study should be considered.

Deficiency of the pectoralis major is a relatively common muscle anomaly. There is little documentation, however, of what, if any, functional deficit the anomaly imparts. Therefore, two highly successful amateur wrestlers with unilateral congenital absence of the pectoralis minor and sternocostal head of the pectoralis major are reported. Cybex testing revealed no appreciable decrease in shoulder internal rotation strength but did show a 20%-30% decrease in shoulder horizontal adduction strength. Other patients similarly affected can be advised that their shoulder strength will be slightly decreased, but significant functional limitations are unlikely.

The genetic variation in energy expenditures of cattle at fasting @TIP) and maintenance (ME&) was determined by using 12 pairs of monozygous twins at 20 mo of age. The pairs were of two breed types, eight Angus x Hereford (three steers, five heifers) and four Barzona x Hereford (three steers, one heifer). The heifers were 132 f 13 d pregnant at the time of measurement. The pairs were fed at 1.15 x maintenance energy requirements for a minimum of 30 d prior to heat production (Hp) measurements in dual indirect respiration calorimetry chambers. The diet fed was cracked c0m:alfalfa hay (4555) with a determined ME of 2.47 f .02 McaVkg DM. This diet was fed individually for 7 d prior to and during two consecutive 22-h HP measurements. The animals then were fasted for 2 d and fasting heat production measurements (FHP) were made on d 3 and 4 of the fast. Metabolizable energy required for ME, was calculated iteratively by assuming a semi-log relationship between HP and metabolizable energy intake. There were no differences (P > .lo) in measured energy expenditures due to different breed type. The FHP and efficiency of ME use for ME, (Kd were similar between sexes, although heifers had lower (P c .025) ME, than steers. Twin pair effects were detected for FHP (P < .005) and ME, (P c .05) but not for K,. Broad sense heritability estimates were calculated as the intraclass correlation between members of monozygous twin pairs. Heritability estimates for %, FHP, and Km were .52 f .22, .75 f .13, and .34 f .27, respectively. These results demonstrate the presence of a genetic component to maintenance energy metabolism measured as FHP and m, selection to change these traits should prove effective.

Pasteurella haemolytica pneumonia of the right caudal lung lobe was experimentally induced in 2-week-old Holstein calves (n = 11) by endobronchial inoculation of 7.9 x 10(10) colony-forming units of 6-hour log-phase bacteria. Calves were studied for 72 hours after inoculation. The challenge procedure consistently induced a lesion in the right caudal lung lobe, which was consistent radiographically with results of pathologic examination and a similar volume of bronchography contrast medium. Clinically, the calves developed a significant increase in rectal temperature within 24 hours after inoculation. Seventy-two hours after inoculation, the total WBC counts, absolute band neutrophil counts, monocyte counts, and blood fibrinogen concentrations were significantly higher than normal and albumin concentration was significantly decreased. Necropsy revealed a circular to oblong lesion that was congested, edematous, and firm and occupied 20 to 40% of the right caudal lung lobe. Histologic examination revealed a severe acute inflammatory reaction characterized by cellular exudate and proteinaceous fluid in the alveoli, interlobular septa, and pleura.

Experimental pneumonia caused by Pasteurella haemolytica was induced in 2-week-old gnotobiotic (n = 4) and conventional (n = 6) calves by endobronchial inoculation into the right caudal lung lobe of 7.9 x 10(10) +/- 0.6 x 10(10) (mean +/- SD) colony-forming units of P haemolytica in the 6-hour log phase of growth. The calves were studied for 24 hours or less. Regression lines for the relationship between clinical index and time for the gnotobiotic group and conventional group of calves were compared, and the clinical index was found to be significantly (P less than or equal to 0.005) more rapid in the gnotobiotic group. There was also a significant difference in the preinoculation, absolute segmented neutrophil count (P less than or equal to 0.05), and in the total serum protein, albumin, and globulin values (P less than or equal to 0.05). Comparison of the preinoculation and post inoculation blood cell and blood chemical values revealed a significant increase (P less than or equal to 0.05) in the numbers of band neutrophils and fibrinogen in conventional calves, and a significant decrease (P less than or equal to 0.05) in the total WBC count in gnotobiotic calves. Necropsy of both groups of calves revealed a circular to oblong lesion that was congested, edematous, and firm, and which occupied 20% to 100% of the right caudal lung lobe and involved the remaining lung lobes to a more minor degree. When mean lesion scores of the 2 groups of calves were compared, no significant difference (P less than or equal to 0.05) was found.(ABSTRACT TRUNCATED AT 250 WORDS)

Two experiments were conducted to quantify the impact on forage use and performance of varying supplementation frequency of cattle consuming forage diets across a range of frequencies. In both experiments, a common supplement was used that contained a relatively high concentration of CP (43%) and was fed at the following frequencies: 1) 2 d/wk; 2) 3 d/wk; 3) 5 d/wk; and 4) 7 d/wk. In Exp. 1, 120 Hereford x Angus cows (BW = 537 kg) grazing winter tallgrass-prairie range were supplemented at the various frequencies from December 7 until calving (average calving date = 3/7/99). All treatments provided the same quantity of supplement on a weekly basis (12.74 kg, as-fed) but divided the amount delivered on a given day equally among the number of supplementation events for that treatment. Less BW was lost from December 7 through calving (linear effect, P = 0.02) as frequency of supplementation increased, but the magnitude of difference in weight change was relatively small. Body condition responded similarly through early February (linear effect, P = 0.02), although treatment effects were not as distinct at calving (cubic effect, P = 0.11). In Exp. 2, 16 ruminally fistulated Hereford x Angus steers (BW = 257 kg) were blocked by weight and assigned to one of the four frequencies of supplementation. Steers were offered tallgrass prairie hay (73.5% NDF, 4.8% CP) ad libitum and were supplemented at a rate (relative to BW) similar to that of the cows in Exp. 1. Increasing frequency of supplementation increased (linear effect, P < or = 0.02) forage OM intake, OM and NDF digestion, and digestible OM intake. However, the most prominent differences in forage OM intake tended (cubic effect, P = 0.07) to occur with the two extreme frequencies of supplementation. In conclusion, forage use was improved with an increased frequency of supplementation, but the impact on performance is not likely to be large unless extreme differences in frequency occur.

Ruminally fistulated steers (n = 13; 263 kg) were used in an incomplete Latin square with 13 treatments and four periods to evaluate the effects of level and source of supplemental carbohydrate (CHO) and level of degradable intake protein (DIP) on the utilization of low-quality, tallgrass-prairie hay. Steers were given ad libitum access to forage (5.7% CP, 2.6% DIP, and 74.9% NDF). The supplementation treatments were fashioned as a 2x3x2 factorial arrangement plus a negative control (NC; no supplement). The factors included two DIP levels (.031 and .122% BW) and three CHO sources (starch, glucose, and fiber) fed at two levels (.15 and .30% BW) within each level of DIP supplementation. Hereford x Angus steers were used in a 13-treatment, four-period, incomplete Latin square design to examine the effects of starch and degradable intake protein (DIP) supplements on forage utilization and ruminal function. Steers were given ad libitum access to low-quality hay (4.9% CP) and were not supplemented (NS) or received different amounts of starch (cornstarch grits; 0, .15, and .3% of initial BW) and DIP (Na-caseinate; .03, .06, .09, and .12% of initial BW) administered via ruminal fistulae in a 3 x 4 factorial arrangement of treatments. Supplemented steers consumed more (P < .01) forage OM, total OM, NDF, and digestible OM (DOM) than NS steers. Forage OM, total OM, NDF, and DOM intakes increased linearly (P < .01) as the amount of supplemental DIP increased. The addition of starch to supplements linearly decreased ( P < .01) the intake of forage OM, NDF, and DOM. The digestion of DM, OM, and NDF increased linearly (P < .01) with supplemental DIP and decreased linearly (P < or = .06) with supplemental starch. Particulate and liquid passages generally increased with DIP; however, starch level influenced the nature of the response (P = .03 and .06, respectively). Similarly, ruminal acid detergent-insoluble ash content generally decreased as starch increased, but the effect was dependent on DIP level (P < .01). Supplementation increased (P < .01) ruminal NH3 and total VFA and decreased (P < .01) ruminal pH relative to NS. All treatments supported average pH values in a range (6.3 to 6.7) unlikely to inhibit fibrolytic bacteria. Ruminal NH3 concentration increased quadratically (P = .03) with DIP and decreased linearly (P = .02) with starch. As DIP increased, total VFA concentration increased linearly (P = .02). Providing supplemental DIP to steers fed low-quality forage increased OM intake and digestion, whereas addition of starch to supplements decreased forage intake and digestion.

Two experiments were conducted to examine changes in usage of energy and visceral organ mass in Targhee lambs selected on the basis of improved rate and efficiency of growth. In Exp. 1, Targhee ram lambs from a breeding line selected for improved growth rate and feed efficiency and a control line that was maintained without selection for 20 yr were provided with either a high (ad libitum) or low (maintenance) plane of nutrition. Rams were slaughtered and weights of the visceral organs were recorded. Analysis of variance, using a factorial model with BW as a covariant, was used to examine effects of lines of breeding, plane of nutrition, interaction between line of breeding and plane of nutrition, and days on feed. Weights of liver, kidneys, rumen, abomasum, and small and large intestines from lambs receiving a high plane of nutrition were, respectively, 39, 25, 12, 28, 40, and 31% greater than weights of those tissues in lambs receiving a low plane of nutrition. Ruminal weights were 13% greater for rams from the select line of breeding than for those from the control line. In Exp. 2, seven Targhee ewe lambs from the select line and eight from the control line were examined for differences in heat production and energy usage by indirect open circuit respiration calorimetry using a completely randomized design. Fasting heat production of lambs from the select line was 7.8% greater than that of lambs from the control line. Partial efficiencies of ME used for maintenance and tissue accretion were not different between lines of breeding.(ABSTRACT TRUNCATED AT 250 WORDS)

Eight Hereford steers with an initial weight of 344 kg and a final weight of 476 kg were allotted randomly to two groups that were alternated between low (120 x maintenance) and high (2.27 x maintenance) intakes during five consecutive seasons in order to measure effects of season (cold stress) and intake on thermoneutral energy requirements. The steers were housed outdoors with neither shelter nor bedding and adapted to the specified intake levels for 21 to 31 d before each season's measurement of maintenance. Maintenance energy requirements were estimated as fasting heat production (FHP) and maintenance metabolized energy (MEm) by respiration calorimetry at thermoneutral temperatures. The high plane of nutrition caused a consistent increase of about 7% in FHP and 14% in MEm averaged across seasons (P less than .05). Within plane of nutrition, season had no effect on FHP or MEm, with the exception that values recorded in fall were lower than those recorded during the first summer (P less than .05). Nevertheless, daily gain during the winter acclimatization period was lower (P less than .05). We conclude that season had no effect on thermoneutral maintenance energy requirements (FHP, MEm) even though cold increased requirements for weight maintenance or gain. No interactions were found between season and plane of nutrition for the parameters measured. Acute cold stress, rather than chronically elevated metabolic rate, likely was responsible for poorer feedlot performance during the cold months of the year.

Eight Holstein steers (four at 300 kg, four at 406 kg) fitted with an elevated carotid artery, hepatic portal and mesenteric venous catheters, and abomasal and ileal cannulas were used in several 4 x 4 Latin square experiments to evaluate small intestinal starch digestion. They were fed alfalfa hay at 1.5% of BW and abomasally infused with water or glucose, corn starch or corn dextrin (one carbohydrate per Latin square) at 20, 40 or 60 g/h, with subsequent determination of small intestinal disappearance and net portal glucose absorption. Increasing the amount of all three carbohydrates infused abomasally increased the amount of carbohydrate disappearing in the small intestine. Increased infusion of glucose caused a continual increase (linear, P less than .01) in net glucose absorption, whereas net glucose absorption for starch and dextrin was maximal at the 20 g/h infusion (quadratic, P less than .05). With the 60 g/h infusion, 94% of the glucose but only 38% of starch and 29% of small intestinal dextrin disappearance could be accounted for as net glucose absorption, leaving a large portion of starch and dextrin disappearance unaccounted for. Of the infused starch and dextrin passing the ileum, 5.8 and 7.3%, respectively, was unpolymerized glucose, indicating that, at least in the distal small intestine, complete starch hydrolysis exceeded the capacity for glucose disappearance. It is concluded that only about 35% of the raw corn starch disappearing in the steer's small intestine resulted in net portal glucose absorption.

Nine steers (300 to 350 kg body weight) were divided into three groups and received either no ionophore, monensin or lasalocid in a 70% cracked corn basal diet for the entire experiment. Within each group, a 3 X 3 Latin-square design was used to evaluate the effect of a high cation concentration added to the diets on in vivo methanogenesis. The three cation treatments consisted of NaCl or KCl addition (Na or K increased to 2.5% of diet dry matter) or no cation addition. Sodium addition decreased methane production in the monensin group 19% when compared with control (P less than .05). Sodium also tended to decrease methane in the lasalocid group (P less than .1) but had no effect on the no-ionophore group. There was no significant effect of K addition on methane production when compared to no added cation. However, K tended to increase methane in both the lasalocid and no-ionophore groups and decrease methane in the monensin group. Results of this experiment support in vitro evidence that cation levels may modulate ionophore effects on methane production.

Six ruminally cannulated steers, five Holsteins and one Hereford (250 to 295 kg), were fed 84% forage sorghum silage plus 16% supplement or 50% forage sorghum silage plus concentrate and supplement diets containing either no addition (controls), 1% sodium bicarbonate (NaHCO3) or 2% sodium bentonite in a 2 X 3 factorial arrangement of treatments in a 6 X 6 Latin-square experiment with 3-wk periods. Sodium bicarbonate increased dry matter (DM) intake when concentrate was included, but neither compound affected intake of the 84% silage diet. Bentonite lowered DM, neutral detergent fiber (NDF), and acid detergent fiber (ADF) digestibilities, but NDF disappearance from nylon bags was unchanged. Ruminal pH, osmolality and L(+) and D(-) lactate were not affected by treatment. Both NaHCO3 and bentonite tended to lower ruminal NH3-N concentrations. Bentonite lowered the molar proportion of isobutyrate in ruminal fluid relative to controls, but proportions of other volatile fatty acids (VFA) and total VFA concentrations were unchanged. Neither NaHCO3 nor bentonite affected ruminal liquid or solid volumes, dilution rate constants or ruminal outflow rates. Markers overestimated volumes, but correction with measured volumes did not change interpretation of treatment effects. It was concluded that control diets had sufficiently high baseline values of pH, dilution rate and acetate proportion to preclude changes induced by either compound, especially at 1 or 2% of DM intake. An effect on palatibility through neutralization of silage acids may have been responsible for the intake response to NaHCO3.

The effect of feeding pattern on energy balance and activity of rats was investigated with four treatment groups. Two groups were fed two meals per day at one of two levels (ad libitum or 1.25 times maintenance) and the other two groups were allowed to nibble either unrestricted or in 12 small meals per day. Four-day total collection digestion trials were conducted on individual rats. Pairs of rats were used for simultaneous activity and respiration calorimetry measurements. Meal-eaters gained less weight, digested and metabolized an approximately 4% greater fraction of their gross energy intake and had a 21% lower maintenance energy requirement. Ad libitum nibblers were about 23% more active during the average 24-hour period than high level meal-eaters, although this difference was not statistically significant. The energetic cost of activity as a percent of maintenance energy ranged from 60% to 18%. These estimates are higher than others in the literature and may have been affected by the activity measurements method, but indicate that differences in activity contributed to observed differences in energetic efficiency between meal-fed and nibbler rats.

Results of an epidemiology study of populations living near a freeway and in the high desert area of Southern California for levels of platinum, palladium and lead are presented. Three age groups (children, young adults, and mature adults) were sampled twice for blood, urine, hair, and feces. Air, water, and soil samples were collected in the areas of the residences of the two populations. The primary objective of this study was to obtain baseline levels of these metals prior to the introduction of the catalytic muffler. The samples were collected in September 1974.

An epidemiological survey was conducted in Houston, Texas on five trace metals in policemen, parking garage attendants, women living near freeways and three control groups of subjects. The controls were matched with the exposed groups for covariate information such as age, sex, smoking habits, ethnic background, socioeconomic status, hair color, and education. Each subject was sampled four times for blood, urine, hair, and feces, and these samples were analyzed for lead, cadmium, zinc, manganese, and copper. Lead and cadmium were correlated with airborne exposures but zinc, manganese and copper were not. The second part of this paper deals with a market study of platinum and palladium markets and a design of an epidemiology survey of individuals occupationally and nonoccupationally exposed to these two metals. The market survey shows that although the catalytic muffler will have a major impact on the market, it is predicted that producers can meet these demands.

Primary tumors arising in the retrovesical area are rare. We recently treated by primary excision a large retrovesical leiomyoma in a forty-five-year-old man. The smooth muscle nature of the tumor was confirmed by electron microscopy. Recognizing this entity is important, since treatment consists only of surgical removal, and the prognosis should be excellent.

Eight steers (327 kg average) were fed diets containing either nontreated or anhydrous NH3-treated wheat straw in a two-period crossover design. Intake and digestibilities of dry matter, gross energy, neutral detergent fiber, acid detergent fiber and crude protein were measured by total collection. Energy losses in urine and methane were determined. Heat production was determined by indirect respiration calorimetry on steers fed both diets and after they were fasted. Ammoniation of the straw raised its N content from .49 to 1.59% (P less than .001). Dry matter (DM) intake was increased (P less than .01) by the process from 1.0 to 1.3% of body weight. Digestibility coefficients of dry matter and energy, as well as those for fiber, were increased (P less than .001) by four percentage units or more. Crude protein digestibility, however, was depressed (P less than .001) from 67.8% to 53.5% by ammoniation. Urinary energy as a percentage of gross energy intake (GEI) was reduced (P less than .05) by NH3 treatment from 4.10 to 3.74%. Methane energy was not different (P greater than .10). Metabolizable energy was improved (P less than .001) by ammoniation, increasing from 45.2 to 50.0% of GEI. Daily heat production was higher (P less than .01) for steers consuming the ammoniated straw diet, increasing from 113.7 to 125.3 kcal/wt.75), and was due to higher metabolizable energy intake (MEI) since partial efficiency of MEI used for maintenance did not differ (P greater than .10) between diets. The higher net energy value of the ammoniated wheat straw diet (1.45 vs 1.26 kcal/g DM) was due mainly to decreased fecal loss and a slight decline in urinary loss.

Â? ABSTRACT The metabolic control of lipogenesis was investigated in growing and fattening lambs by fasting and refeeding studies. Fasting state was monitored by indirect calorimetry measurements of respiratory quotient and methane production. At various time periods, rump adipose tissue was obtained by biopsy and used for determi nation of lipogenic capacity and the activities of relevant enzymes. The activity of acetyl CoA carboxylase very closely paralleled the progressive decrease in fatty acid synthesis by adipose tissue slices during starvation and the increase upon refeeding. Acetyl CoA synthetase and fatty acid synthetase activities also decreased substantially during fasting, but in fattening lambs, this decrease did not occur until after 48 hours of fasting. Additionally, the activities of acetyl CoA synthetase and fatty acid synthetase did not increase significantly with refeeding. In both groups of lambs the activities of glucose-6-phosphate dehydrogenase, 6-phosphogluconate dehydrogenase, and NADP- isocitrate dehydrogenase decreased only moderately during starvation and had not increased significantly at the time of biopsy after refeeding. The results of these investi gations indicate that acetyl CoA carboxylase is the rate limiting enzyme in fatty acid synthesis by ruminant adipose tissue. The correlation between in vitro lipogenic capacity and acetyl CoA carboxylase activity during fasting and refeeding was 0.95 and 0.93 with growing and fattening lambs, respectively. J.
