Introduction: Preliminary research has indicated that numerous drivers perceive their risk of traffic crash to be less than other drivers, while perceiving their driving ability to be better. This phenomenon is referred to as ‘comparative optimism’ (CO) and may prove to inhibit the safe adoption of driving behaviors and/or dilute perceptions of negative outcomes. The objective of this study was to investigate comparative judgments regarding crash risk and driving ability, and how these judgments relate to self-reported speeding. Method: There were 760 Queensland motorists comprised of 51.6% males and 48.2% females, aged 16–85 (M = 39.60). Participants completed either a paper or online version of a survey. Judgments of crash risk and driving ability were compared to two referents: the average same-age, same-sex driver, and the average same-age, same-sex V8 supercar champion. Results: Drivers displayed greater optimism when comparing their crash risk and driving ability to the average same-age, same- sex driver (respectively, 72%, 72.4%), than when comparing to a V8 supercar champion (respectively, 60%, 32.9%). When comparing judgements of crash risk and driving ability to a similar driver, it appears that participants in the present study are just about as optimistic about their risk of crash (i.e. 72%) as they are optimistic about their driving ability (i.e. 74.2%).

Summary Background Staging and treatment of human African trypanosomiasis caused by Trypanosoma brucei gambiense (g-HAT) required lumbar puncture to assess cerebrospinal fluid (CSF) and intravenous drugs that cross the blood–brain barrier for late-stage infection. These procedures are inconvenient in rural health systems of disease-endemic countries. A pivotal study established fexinidazole as the first oral monotherapy to be effective against non-severe stage 2 g-HAT. We aimed to assess the safety and efficacy of fexinidazole in early g-HAT. Methods In this prospective, multicentre, open-label, single-arm cohort study, patients with stage 1 or early stage 2 g-HAT were recruited from eight treatment centres in the Democratic Republic of the Congo. Primary inclusion criteria included being older than 15 years, being able to ingest at least one complete meal per day (or at least one sachet of Plumpy'Nut®), a Karnofsky score higher than 50, evidence of trypanosomes in the blood or lymph but no evidence of trypanosomes in the CSF, willingness to be admitted to hospital to receive treatment, having a permanent address, and being able to comply with the follow-up visit schedule. Exclusion criteria included severe malnutrition, inability to take medication orally, pregnant or breastfeeding women, any clinically important medical condition that could jeopardise patient safety or participation in the study, severely deteriorated general status, any contraindication to imidazole drugs, HAT treatment in the past 2 years, previous enrolment in the study or previous intake of fexinidazole, abnormalities on electrocardiogram that did not return to normal in pretreatment repeated assessments or were considered clinically important, QT interval corrected using Fridericia's formula of at least 450 ms, and patients not tested for malaria or not having received appropriate treatment for malaria or for soil-transmitted helminthiasis. Patients were classified into stage 1 or early stage 2 g-HAT groups following evidence of trypanosomes in the blood, lymph, and absence in CSF, and using white-blood-cell count in CSF. Patients received 1800 mg fexinidazole once per day on days 1–4 then 1200 mg fexinidazole on days 5–10. Patients were observed for approximately 19 months in total. Study participants were followed up on day 5 and day 8 during treatment, at end of treatment on day 11, at end of hospitalisation on days 11–18, at week 9 for a subset of patients, and after 6 months, 12 months, and 18 months. The primary endpoint was treatment success at 12 months. Safety was assessed through routine monitoring. Analyses were done in the intention-to-treat population. The acceptable success rate was defined as treatment efficacy in more than 80% of patients. This study is completed and registered with ClinicalTrials.gov ( NCT02169557 ). Findings Patients were enrolled between April 30, 2014, and April 25, 2017. 238 patients were recruited: 195 (82%) patients with stage 1 g-HAT and 43 (18%) with early stage 2 g-HAT. 189 (97%) of 195 patients with stage 1 g-HAT and 41 (95%) of 43 patients with early stage 2 g-HAT were finally included and completed the 10 day treatment period. Three patients with stage 1 g-HAT died after the 10 day treatment period and before the 12 month primary follow-up visit, considered as treatment failure and were withdrawn from the study. Treatment was effective at 12 months for 227 (99%) of 230 patients (95% CI 96·2–99·7): 186 (98%) of 189 patients (95·4–99·7) with stage 1 and 41 (100%) of 41 patients (91·4–100·0) with early stage 2, indicating that the primary study endpoint was met. No new safety issues were observed. The most frequent adverse events were headache and vomiting. In total, 214 (93%) of 230 patients had treatment-emergent adverse events, mainly common-terminology criteria for adverse events grades 1 to 3. None led to treatment discontinuation. Interpretation Fexinidazole is a valuable first-line treatment option in the early stages of g-HAT. Introduction : In Brazil, driver aggressiveness in road traffic is a critical issue and could be an important contributing factor to the high number of traffic accidents . Because no instruments are available in Portuguese to register driving aggressiveness or driving anger in Brazil, we adapted English instruments into the Brazilian context. The aims of this study were to provide a Brazilian adaptation of the Driving Anger Expression Inventory (DAX) and to try to validate it by testing its psychometric properties and investigating its relationships with risky driving behaviors (DBQ), road accidents, driving sensation seeking, and hostility. Method: The Brazilian adaptations of the DAX, DBQ, the Driving Sensation Seeking Scale (DSSS) and the hostility Scale were administered to a sample of 512 undergraduate students (with a mean age of 23.7 years, 52.1% men). Results : Confirmatory factor analysis of the Brazilian DAX (DAX-BR) items yielded a four-factor solution with 43 items, which obtained the best goodness-of-fit to the data. Cronbach’s alpha for the DAX-BR factors ranged from 0.69 to 0.88. Other results on validity were a positive correlation (range 0.39–0.59) between the factors of the DAX-BR, DSSS, and DBQ. Conclusion : DAX-BR as the same structure as the original and is a reliable instrument for use with young drivers. Other studies should be conducted to further validate the DAX-BR in different types of populations such as older and more experienced drivers, professional drivers, and traffic regulation offenders whose driver's license has been taken away. Practical applications: This Brazilian version can be recommended for the assessment of driving anger expression in Brazil among young drivers in view of helping them driver more safely, and in particular to reduce traffic violations.

BACKGROUND: Recurrent mutations in the promoter of the telomerase reverse transcriptase (TERT) gene (C228T and C250T) detected in tumours and cells shed into urine of urothelial cancer (UC) patients are putative biomarkers for UC detection and monitoring. However, the possibility of detecting these mutations in cell-free circulating DNA (cfDNA) in blood and urine, or DNA from urinary exfoliated cells (cellDNA) with a single-gene sensitive assay has never been tested in a case-control setting. METHODS: We developed a single-plex assay (UroMuTERT) for the detection of low-abundance TERT promoter mutations. We tested 93 primary and recurrent UC cases and 94 controls recruited in France (blood, urine samples and tumours for the cases), and 50 primary UC cases and 50 controls recruited in Portugal (urinary exfoliated cell samples). We compared our assay with urine cytology. FINDINGS: In the French series, C228T or C250T were detected in urinary cfDNA or cellDNA in 81 cases (87·1%; 95% CI 78·6–93·2), and five controls (Specificity 94·7%; 95%CI 88·0–98·3), with 98·6% (95% CI 92·5–99·96) concordance in matched tumours. Detection rate in plasma cfDNA among cases was 7·1%. The UroMuTERT sensitivity was (i) highest for urinary cfDNA and cellDNA combined, (ii) consistent across primary and recurrent cases, tumour stages and grades, (iii) higher for low-risk non-muscle invasive UC (86·1%) than urine cytology (23·0%) (P

Introduction: The growth of the European market for road-freight transport has recently led to important changes. The growing number of foreign pavilion drivers transiting in France, which plays a bridging role among European countries, has influenced the lives of truck drivers by increasing competition, pressure on day-to-day activities, and constraints related to delivery deadlines. Adding this new pressure to those inherent in the road-freight transport sector has raised concerns, especially ones linked to levels of perceived stress by truck drivers. Method: With safety concerns in mind, we devised a questionnaire aimed at understanding how French truck drivers and non-French truck drivers, passing through four highway rest areas in France perceive stress, organizational factors, mental health, and risky driving behaviors. A sample of 515 truck drivers took part in the survey (260 French nationals), 97.9% of whom were male. Results: The results of a structural equation model indicated that perceived stress can increase self-reported risky driving behaviors among truck drivers. Furthermore, organizational factors and mental health were closely linked to perceived stress. Finally, some differences were found between French and non-French truck drivers with respect to mind-wandering and mental health, and to perceive driving difficulties to overcome and driving skills. Practical Applications: Several recommendations based on the findings are provided to policymakers and organizations.

The worldwide incidence of pulmonary carcinoids is increasing, but little is known about their molecular characteristics. Through machine learning and multi-omics factor analysis, we compare and contrast the genomic profiles of 116 pulmonary carcinoids (including 35 atypical), 75 large-cell neuroendocrine carcinomas (LCNEC), and 66 small-cell lung cancers. Here we report that the integrative analyses on 257 lung neuroendocrine neoplasms stratify atypical carcinoids into two prognostic groups with a 10-year overall survival of 88% and 27%, respectively. We identify therapeutically relevant molecular groups of pulmonary carcinoids, suggesting DLL3 and the immune system as candidate therapeutic targets; we confirm the value of OTP expression levels for the prognosis and diagnosis of these diseases, and we unveil the group of supra-carcinoids. This group comprises samples with carcinoid-like morphology yet the molecular and clinical features of the deadly LCNEC, further supporting the previously proposed molecular link between the low- and high-grade lung neuroendocrine neoplasms., The worldwide incidence of pulmonary carcinoids is increasing, but little is known about their molecular characteristics. Summary Background Few therapeutic options are available to treat the late-stage of human African trypanosomiasis, a neglected tropical disease, caused by Trypanosoma brucei gambiense (g-HAT). The firstline treatment is a combination therapy of oral nifurtimox and intravenous eflornithine that needs to be administered in a hospital setting by trained personnel, which is not optimal given that patients often live in remote areas with few health resources. Therefore, we aimed to assess the safety and efficacy of an oral regimen of fexinidazole (a 2-substituted 5-nitroimidazole with proven trypanocidal activity) versus nifurtimox eflornithine combination therapy in patients with late-stage g-HAT. Methods In this randomised, phase 2/3, open-label, non-inferiority trial, we recruited patients aged 15 years and older with late-stage g-HAT from g-HAT treatment centres in the Democratic Republic of the Congo (n=9) and the Central African Republic (n=1). Patients were randomly assigned (2:1) to receive either fexinidazole or nifurtimox eflornithine combination therapy according to a predefined randomisation list (block size six). The funder, data management personnel, and study statisticians were masked to treatment. Oral fexinidazole was given once a day (days 1–4: 1800 mg, days 5–10: 1200 mg). Oral nifurtimox was given three times a day (days 1–10: 15 mg/kg per day) with eflornithine twice a day as 2 h infusions (days 1–7: 400 mg/kg per day). The primary endpoint was success at 18 months (ie, deemed as patients being alive, having no evidence of trypanosomes in any body fluid, not requiring rescue medication, and having a cerebrospinal fluid white blood cell count ≤20 cells per μL). Safety was assessed through routine monitoring. Primary efficacy analysis was done in the modified intention-to-treat population and safety analyses in the intention-to-treat population. The acceptable margin for the difference in success rates was defined as 13%. This study has been completed and is registered with ClinicalTrials.gov, number NCT01685827. Findings Between October, 2012, and November, 2016, 419 patients were pre-screened. Of the 409 eligible patients, 14 were not included because they did not meet all inclusion criteria (n=12) or for another reason (n=2). Therefore, 394 patients were randomly assigned, 264 to receive fexinidazole and 130 to receive nifurtimox eflornithine combination therapy. Success at 18 months was recorded in 239 (91%) patients given fexinidazole and 124 (98%) patients given nifurtimox eflornithine combination therapy, within the margin of acceptable difference of −6·4% (97·06% CI −11·2 to −1·6; p=0·0029). We noted no difference in the proportion of patients who experienced treatment-related adverse events (215 [81%] in the fexinidazole group vs 102 [79%] in the nifurtimox eflornithine combination therapy group). Treatment discontinuations were unrelated to treatment (n=2 [1%] in the fexinidazole group). Temporary nifurtimox eflornithine combination therapy interruption occurred in three (2%) patients. 11 patients died during the study (nine [3%] in the fexinidazole group vs two [2%] in the nifurtimox eflornithine combination therapy group). Interpretation Our findings show that oral fexinidazole is effective and safe for the treatment of T b gambiense infection compared with nifurtimox eflornithine combination therapy in late-stage HAT patients. Introduction: Exploratory data reduction techniques, such as Factor Analysis (FA) and Principal Component Analysis (PCA), are widely used in questionnaire validation with ordinal data, such as Likert Scale data, even though both techniques are indicated to metric measures. In this context, this study presents an e-survey, conducted to obtain self-reported behaviors between Brazilian drivers (N = 1,354, 55.2% of males) and Portuguese drivers (N = 348, 46.6% of males) based on 20 items from the Driver Behavior Questionnaire (DBQ) on a five-point Likert Scale. This paper aimed to examine DBQ validation using FA and PCA compared to Categorical Principal Component Analysis (CATPCA) which is more indicative to use with Likert Scale data. Results: The results from all techniques confirmed the most replicated factor structure of DBQ, distinguishing behaviors as errors, ordinary violations, and aggressive violation. However, after Varimax rotation, CATPCA explained 11% more variance compared to FA and 2% more than PCA. We identified cross-loadings among the component of the techniques. An item changed its dimension in the CATPCA results but did not change the structural interpretability. Individual scores from dimension 1 of CATPCA were significantly different from FA and PCA. Individual scores from factor 1 of CATPCA were significantly different from FA and PCA. Practical applications: The CATPCA seems to be more advantageous in order to represent the original data and considering data constrains. In addition to finding an interpretable factorial structure, the representation of the original data is regarded as relevant since the factor scores could be used for crash prediction in future analyses.

To assess and explain finely drivers’ a priori acceptance of highly automated cars, this study used the Theory of Planned Behaviour (TPB) and the Unified Theory of Acceptance and Use of Technology (UTAUT). Further, the current study sought to extend upon previous research to assess if intentions to use highly automated cars in the future differed according to country (i.e., Australia, France, & Sweden). These three countries were selected to enable comparisons of a priori acceptance between countries of differing levels of exposure to highly automated cars. Participants (N = 1563; 62.1 % male) were recruited in Australia (n = 558), France (n = 625), and Sweden (n = 380) to complete a 20 min online questionnaire. The findings differed according to country of residence. Individuals residing in France reported significantly greater intentions to use highly automated cars when they become publicly available compared to individuals residing in Australia and in Sweden. Of the TPB constructs entered at step 1 in the hierarchical regression, attitudes, subjective norms, and perceived behavioural control (capability and controllability) were significant predictors of intentions to use highly automated cars for participants residing in Australia and France. For participants residing in Sweden, only attitudes and PBC-capability were significant predictors of intentions. Of the UTAUT constructs entered at step 2, performance expectancy and effort expectancy were significant predictors of intentions for participants residing in France and only performance expectancy a significant predictor of intentions for participants residing in both Australia and Sweden. Age and gender did not add to the prediction of intentions when entered at step 3. However, pre-existing knowledge was a significant negative predictor of intentions when entered at step 3 for participants residing in Australia. Overall, the findings found some support for applying the TPB and UTAUT to assess intentions to use highly automated cars in different countries. The findings also highlight differences in a priori acceptance across countries and the factors which predict such acceptance.

The utility of KRAS mutations in plasma circulating cell-free DNA (cfDNA) samples as non-invasive biomarkers for the detection of pancreatic cancer has never been evaluated in a large case-control series. We applied a KRAS amplicon-based deep sequencing strategy combined with analytical pipeline specifically designed for the detection of low-abundance mutations to screen plasma samples of 437 pancreatic cancer cases, 141 chronic pancreatitis subjects, and 394 healthy controls. We detected mutations in 21.1% (N=92) of cases, of whom 82 (89.1%) carried at least one mutation at hotspot codons 12, 13 or 61, with mutant allelic fractions from 0.08% to 79%. Advanced stages were associated with an increased proportion of detection, with KRAS cfDNA mutations detected in 10.3%, 17,5% and 33.3% of cases with local, regional and systemic stages, respectively. We also detected KRAS cfDNA mutations in 3.7% (N=14) of healthy controls and in 4.3% (N=6) of subjects with chronic pancreatitis, but at significantly lower allelic fractions than in cases. Combining cfDNA KRAS mutations and CA19-9 plasma levels on a limited set of case-control samples did not improve the overall performance of the biomarkers as compared to CA19-9 alone. Whether the limited sensitivity and specificity observed in our series of KRAS mutations in plasma cfDNA as biomarkers for pancreatic cancer detection are attributable to methodological limitations or to the biology of cfDNA should be further assessed in large case-control series.

Circulating tumor DNA (ctDNA) is emerging as a key potential biomarker for post-diagnosis surveillance but it may also play a crucial role in the detection of pre-clinical cancer. Small-cell lung cancer (SCLC) is an excellent candidate for early detection given there are no successful therapeutic options for late-stage disease, and it displays almost universal inactivation of TP53. We assessed the presence of TP53 mutations in the cell-free DNA (cfDNA) extracted from the plasma of 51 SCLC cases and 123 non-cancer controls. We identified mutations using a pipeline specifically designed to accurately detect variants at very low fractions. We detected TP53 mutations in the cfDNA of 49% SCLC patients and 11.4% of non-cancer controls. When stratifying the 51 initial SCLC cases by stage, TP53 mutations were detected in the cfDNA of 35.7% early-stage and 54.1% late-stage SCLC patients. The results in the controls were further replicated in 10.8% of an independent series of 102 non-cancer controls. The detection of TP53 mutations in 11% of the 225 non-cancer controls suggests that somatic mutations in cfDNA among individuals without any cancer diagnosis is a common occurrence, and poses serious challenges for the development of ctDNA screening tests., Highlights • CtDNA may play a crucial role in the detection of pre-clinical cancer. • TP53 mutations are detectable in the cfDNA of SCLC patients with early-stage tumors. • Detection of TP53 mutations in non-cancer controls poses serious challenges for the development of ctDNA screening tests. Cell-free DNA (cfDNA) has potential for monitoring response to treatment and relapse, but also for early detection. This is the first study reporting the detection of circulating-tumor DNA (ctDNA) in cases diagnosed with small-cell lung cancer (SCLC). Our results show that TP53 mutations are detectable in the cfDNA of SCLC patients including those with early-stage tumors. Importantly, we also provide evidence that cancer-like TP53 mutations are present in non-cancer controls, which poses serious challenges for the development of ctDNA screening tests.

Background: Detecting pre-clinical bladder cancer (BC) using urinary biomarkers may provide a valuable opportunity for screening and management. Telomerase reverse transcriptase (TERT) promoter mutations detectable in urine have emerged as promising BC biomarkers. Methods: We performed a nested case-control study within the population-based prospective Golestan Cohort Study (50,045 participants, followed up to 14 years) and assessed TERT promoter mutations in baseline urine samples from 38 asymptomatic individuals who subsequently developed primary BC and 152 matched controls using a Next-Generation Sequencing-based single-plex assay (UroMuTERT) and droplet digital PCR assays. Findings: Results were obtained for 30 cases and 101 controls. TERT promoter mutations were detected in 14 pre-clinical cases (sensitivity 46·67%) and none of the controls (specificity 100·00%). At an estimated BC cumulative incidence of 0·09% in the cohort, the positive and negative predictive values were 100·00% and 99·95% respectively. The mutant allelic fractions decreased with the time interval from urine collection until BC diagnosis (p = 0·033) but the mutations were detectable up to 10 years prior to clinical diagnosis. Interpretation: Our results provide the first evidence from a population-based prospective cohort study of the potential of urinary TERT promoter mutations as promising non-invasive biomarkers for early detection of BC. Further studies should validate this finding and assess their clinical utility in other longitudinal cohorts. Environmental perturbations induce transcriptional changes, some of which may be inherited even in the absence of the initial stimulus. Previous studies have focused on transfers through the germ-line although microbiota is also passed on to the offspring. Thus, we inspected the involvement of the gut microbiome in transgenerational inheritance of environmental exposures in Drosophila melanogaster. We grew flies in the cold versus control temperatures and compared their transcriptional patterns in both conditions as well as in their offspring. F2 flies grew in control temperature while we controlled their microbiota acquisition from either F1 sets. Transcriptional status of some genes was conserved transgenerationally, and a subset of these genes, mainly expressed in the gut, was transcriptionally dependent on the acquired microbiome. Vehicle-dynamics data, now more readily available thanks to moderate-cost, embedded data logging solutions, have been used to study drivers' behavior (acceleration, braking, and yaw rate) through naturalistic driving research aimed at detecting critical safety events. In addition, self-reported measures have been developed to describe these events and to assess various individual risk factors such as sensation seeking, lack of experience, anger expression while driving, and sensitivity to distraction. In the present study, we apply both of these methods of gathering driving data in order to assess risk proneness as accurately as possible. Data were obtained from 131 drivers, who filled in an introductory questionnaire pertaining to their driving habits. Their vehicles were equipped with an external, automatic data-capture device for approximately two months. During that period, the participants reported critical safety events that occurred behind the wheel by (a) pressing a button connected to the device and (b) describing the events in logbooks. They also filled in weekly questionnaires, and at the end of the participation period, a final questionnaire with various self-reported measures pertaining to their driving activity. We processed the data by (a) performing a multiple correspondence analysis of the characteristics assessed via the automatic data capture and self-reports, and (b) categorizing the participants via hierarchical clustering of their coordinates on the dimensions obtained from the correspondence analysis. This allowed us to identify a group of drivers (n=43) at risk, based on several self-reported measures, in particular, their recent crash involvement, and the frequency of critical acceleration/deceleration events as an objectivemeasure. However, the at-risk drivers did not themselves report more critical safety events than the other two groups.; Les recherches relatives au risque routier et ses antécédents se sont couramment appuyées sur l'analyse des accidents, des sanctions pour infractions et(ou) des comportements à risque. Peu d'entre elles se sont intéressées aux « presque accidents ». L'on entend par presque accident, un événement dangereux lequel, sans action corrective de la part d'autrui ou de soi-même, aboutirait à un accident. Dans le cadre du projet MEDOC, nous avons essayé d'apporter un éclairage sur les presque accidents se produisant au volant des automobilistes et d'identifier les automobilistes les plus exposés au risque. Croisant des mesures déclaratives et des mesures objectives, nous nous sommes appuyés sur un ensemble d'indicateurs 1) les variables liées à la dynamique du véhicule et aux actions de conduite et 2) les variables liées aux déclarations des participant(e)s pour décrire ces presque accidents et identifier les facteurs de risque individuels. Nous avons examiné ces résultats auprès d'un échantillon composé de 131 automobilistes (52,7 % d'hommes, en moyenne âgés de 39,7 ans, ayant le permis voiture depuis 19,2 ans et parcourant au volant 16 606 km par an) les presque accidents qu'ils ont rencontrés au cours de deux mois de conduite. Nous avons identifié trois groupes d'automobilistes en particulier un groupe d'automobilistes (n = 43) à risque (plutôt jeunes, masculins, impliqués dans des accidents de la route, ayant perdu beaucoup de points, chercheurs de sensations fortes et sensibles à la distraction, utilisateurs de leur véhicule pour exprimer leur colère, qui ont des occurrences plutôt élevées de presque accidents de type accélération).

Introduction The development of skills essential for avoiding crashes depends, in particular, on how drivers explain the causes of dangerous driving behaviors that resulted in a near crash. Here, we analyze causes attributed to such behaviors by car drivers in a self-report study. We explore the relationships between the dimensions of causal attribution, attribution of responsibility for the near crash, and drivers' comparative judgments. Method For approximately two months, drivers used logbooks to document the near crashes that occurred during their trips. The causes attributed in those reports to driving behaviors resulting in near crashes were then coded by two judges on the basis of several causal dimensions. Drivers also estimated their own and an average driver's skill levels, and their risk of being involved, as a driver, in a crash. Results We distinguished main types of causes of the near crashes reported. Drivers had a tendency to more often attribute external causes to their own behaviors resulting in near crashes than to those of others. The probability of attributing a controllable cause increased with overestimation of one's own skills and decreased with underestimation of one's own risk in comparison to other drivers. The probability of attributing a stable cause increased with underestimation of one's own risk. Conclusions When they explained their own behaviors resulting in near crashes, drivers mentioned different causes than when they explained those of others. Overestimation of one's own skills as compared to other drivers could be beneficial for developing crash-avoiding skills, insofar as it seems to foster attribution of controllable causes. By contrast, underestimation of one's own risk could have the opposite effect. Practical applications Vulnerability to road risks should be stressed in driver’s training and risk communication campaigns. However, self-confidence with respect to one's skills should not always be targeted as a safety problem.

Based on an extended Theory of Planned Behavior (TPB, Ajzen, 1985, 1991), we conducted surveys in order to explain and predict violations at a railroad crossing, among pedestrians (n=153) and car drivers (n=151). Measures were made with respect to three chronologically related railroad crossing situations that varied in risk level. The situations were described in scenarios and depicted on photographs. The participants were recruited in the suburbs of Paris, at two automated railroad crossings with four half-barriers. We found that the pedestrians had stronger crossing intentions than did car drivers, especially at the more congested crossing of the two under study. For both categories of road users, intentions and the amount of intention variance explained by the extended TPB factors decreased significantly with risk level. In the most dangerous situations, risk-taking was the most unlikely and the least predictable Self-reported past frequency of crossing against safety warning devices was the main predictor of the intention to commit this violation again, especially among males, followed by the attitude and the injunctive norm in favor the violation. Moreover, car drivers were influenced in their crossing intentions by the descriptive norm. The presence of another vehicle on the tracks when the safety warning devices were activated was perceived not as facilitating, but as an additional risk factor. The discussion addresses the importance of taking into account these determinants of violations in conceiving countermeasures. Our findings could be especially useful for conceiving risk-communication campaigns.

BioImpedance Spectroscopy (BIS) has been clinically used to determine the hydrational status of patients undergoing haemodialysis (HD). In the present project we are developing a calf-localised, integrated impedimetric device to periodically and conveniently measure and transmit information on the hydrational status of home-based patients to a remote clinic. Surprisingly, we have found that simple postural changes before or during measurement lead to significant fluid shifts in the lower leg that are as important and as long lasting as the effects of haemodialysis. These must be taken into account if potentially hazardous errors are not to be made in assessing a patient's hydrational status.

Objective: An experiment was performed in a driving simulator to investigate the impacts of practice, trust, and interaction on manual control recovery (MCR) when employing fully automated driving (FAD). Background: To increase the use of partially or highly automated driving efficiency and to improve safety, some studies have addressed trust in driving automation and training, but few studies have focused on FAD. FAD is an autonomous system that has full control of a vehicle without any need for intervention by the driver. Method: A total of 69 drivers with a valid license practiced with FAD. They were distributed evenly across two conditions: simple practice and elaborate practice. Results: When examining emergency MCR, a correlation was found between trust and reaction time in the simple practice group (i.e., higher trust meant a longer reaction time), but not in the elaborate practice group. This result indicated that to mitigate the negative impact of overtrust on reaction time, more appropriate practice may be needed. Conclusions: Drivers should be trained in how the automated device works so as to improve MCR performance in case of an emergency. Application: The practice format used in this study could be used for the first interaction with an FAD car when acquiring such a vehicle.

Drivers’ tend to overestimate their competences, which may result in risk taking behavior. Providing drivers with feedback has been suggested as one of the solutions to overcome drivers’ inaccurate self-evaluations. In practice, many tests and driving simulators provide drivers with non-evaluative feedback, which conveys information on the level of performance but not on what caused the performance. Is this type of feedback indeed effective in reducing self-enhancement biases? The current study aimed to investigate the effect of non-evaluative performance feedback on drivers’ self-evaluations using a computerized hazard perception test. A between-subjects design was used with one group receiving feedback on performance in the hazard perception test while the other group not receiving any feedback. The results indicated that drivers had a robust self-enhancement bias in their self-evaluations regardless of the presence of performance feedback and that they systematically estimated their performance to be higher than they actually achieved in the test. Furthermore, they devalued the credibility of the test instead of adjusting their self-evaluations in order to cope with the negative feelings following the failure feedback. We discuss the theoretical and practical implications of these counterproductive effects of non-evaluative feedback.

BNC2 is an extremely conserved zinc finger protein with important functions in the development of craniofacial bones and male germ cells. Because disruption of the Bnc2 gene in mice causes neonatal lethality, the function of the protein in adult animals has not been studied. Until now BNC2 was considered to have a wider tissue distribution than its paralog, BNC1, but the precise cell types expressing Bnc2 are largely unknown. We identify here the cell types containing BNC2 in the mouse and we show the unexpected presence of BNC1 in many BNC2-containing cells. BNC1 and BNC2 are colocalized in male and female germ cells, ovarian epithelial cells, sensory neurons, hair follicle keratinocytes and connective cells of organ capsules. In many cell lineages, the two basonuclins appear and disappear synchronously. Within the male germ cell lineage, BNC1 and BNC2 are found in prospermatogonia and undifferentiated spermatogonia, and disappear abruptly from differentiating spermatogonia. During oogenesis, the two basonuclins accumulate specifically in maturing oocytes. During the development of hair follicles, BNC1 and BNC2 concentrate in the primary hair germs. As follicle morphogenesis proceeds, cells possessing BNC1 and BNC2 invade the dermis and surround the papilla. During anagen, BNC1 and BNC2 are largely restricted to the basal layer of the outer root sheath and the matrix. During catagen, the compartment of cells possessing BNC1 and BNC2 regresses, and in telogen, the two basonuclins are confined to the secondary hair germ. During the next anagen, the BNC1/BNC2-containing cell population regenerates the hair follicle. By examining Bnc2−/− mice that have escaped the neonatal lethality usually associated with lack of BNC2, we demonstrate that BNC2 possesses important functions in many of the cell types where it resides. Hair follicles of postnatal Bnc2−/− mice do not fully develop during the first cycle and thereafter remain blocked in telogen. It is concluded that the presence of BNC2 in the secondary hair germ is required to regenerate the transient segment of the follicle. Postnatal Bnc2−/− mice also show severe dwarfism, defects in oogenesis and alterations of palatal rugae. Although the two basonuclins possess very similar zinc fingers and are largely coexpressed, BNC1 cannot substitute for BNC2. This is shown incontrovertibly in knockin mice expressing Bnc1 instead of Bnc2 as these mice invariably die at birth with craniofacial abnormalities undistinguishable from those of Bnc2−/− mice. The function of the basonuclins in the secondary hair germ is of particular interest.

Speeding is one of the main factors of car crash-risk, but it also contributes to increasing air-pollution. In two studies we attempted to lead drivers to abide by speed limits using "reducing air-pollution" as a new argument. We presented prevention messages that highlighted the role of speeding in increasing "crash-risk", "air-pollution", or both (Studies 1 and 2). The messages were also positively or negatively framed (Study 2). Given that women are more concerned with environmental issues than are men, we expected the following hypotheses to be validated for women. The message with the "air-pollution" argument was expected to be evaluated more positively than the "crash-risk" message (H1). The "air-pollution" and "crash-risk and air-pollution" messages were expected to be more effective than the "crash-risk" message on the behavioral intention to observe speed limits (H2a) and on the perceived efficacy of speed-limit observance in reducing air-pollution (H2b; Studies 1 and 2). Furthermore, positive framing was expected to be more effective than negative framing (H3), and presenting a message to be more effective than presenting no message (H4; Study 2). Broadly, our results argue in favor of our hypotheses. However in Study 2, the effects of message framing did not allow us to conclude that negative or positive framing was superior. All in all, messages with the "air-pollution" argument were more effective at leading drivers to observe speed limits. Thus, environmental protection may be a fruitful route to explore for increasing road safety.

This study was conducted to provide further evidence of reliability and validity for the Driving Anger Scale, French version. A sample of 202 drivers, ages 18–25 years, completed the scale and the general Trait Anger scale from NEO Personality Inventory-Revised. Factor analysis indicated five factors similar to those found in the original U.S. version. The ‘Traffic Obstructions’ and ‘Slow Driving' factors have been slightly modified, and ‘Discourtesy’ was abandoned. Positive correlations were observed between scores on the Trait-Anger scale from the NEO Personality Inventory-Revised and the Driving Anger Scale ( r = .41) and with its factors (from .24 to .38), except for ‘Illegal Driving’. The internal consistency of the Driving Anger Scale was acceptable for the total score (α = .82) and for factors ranged from .74 to .80. This French version can be preliminarily recommended for the assessment of driving anger in France among young drivers.

INTRODUCTION: This study investigated the relation between co-presence and bicyclists' riding behavior. We assumed that the presence of peer riders would either facilitate or inhibit risky behaviors depending on bicyclists' perceptions of three traffic contexts conducive to risk taking (i.e., red-light, go straight, and turn to left). METHOD: Young bicyclists (N=207) were randomly assigned to two experimental conditions (alone vs. with peers). They filled in a scenario-based questionnaire about their intentions to adopt risky behaviors in three specific traffic situations as well as their risk perception of these situations and their general self-perceived efficacy as a bicyclist. We hypothesized that the presence of peer riders engaged in a risky behavior will facilitate the intention to adopt risky behaviors in situations where group risk is evaluated as lower than individual risk. In opposition, the presence of peer riders engaged in a risky behavior will inhibit the intention to adopt risky behaviors in situations where group risk is evaluated as higher than individual risk. RESULTS: The results confirmed the hypotheses. PRACTICAL APPLICATIONS: The findings offer insights for developing new effective education and intervention programs in order to reduce the frequency of dangerous behavior among bicyclists. Crashes at level crossings are a major issue worldwide. In Australia, as well as in other countries, the number of crashes with vehicles has declined in the past years, while the number of crashes involving pedestrians seems to have remained unchanged. A systematic review of research related to pedestrian behaviour highlighted a number of important scientific gaps in current knowledge. The complexity of such intersections imposes particular constraints to the understanding of pedestrians' crossing behaviour. A new systems-based framework, called Pedestrian Unsafe Level Crossing framework (PULC) was developed. The PULC organises contributing factors to crossing behaviour on different system levels as per the hierarchical classification of Jens Rasmussen's Framework for Risk Management. In addition, the framework adapts James Reason's classification to distinguish between different types of unsafe behaviour. The framework was developed as a tool for collection of generalizable data that could be used to predict current or future system failures or to identify aspects of the system that require further safety improvement. To give it an initial support, the PULC was applied to the analysis of qualitative data from focus groups discussions. A total number of 12 pedestrians who regularly crossed the same level crossing were asked about their daily experience and their observations of others' behaviour which allowed the extraction and classification of factors associated with errors and violations. Two case studies using Rasmussen's AcciMap technique are presented as an example of potential application of the framework. A discussion on the identified multiple risk contributing factors and their interactions is provided, in light of the benefits of applying a systems approach to the understanding of the origins of individual's behaviour. Potential actions towards safety improvement are discussed.

BACKGROUND: Few data are available regarding the use of antithrombotic strategies in coronary artery disease patients with atrial fibrillation (AF) in everyday practice. We sought to describe the prevalence of AF and its antithrombotic management in a contemporary population of patients with stable coronary artery disease.METHODS AND FINDINGS: CLARIFY is an international, prospective, longitudinal registry of outpatients with stable coronary artery disease, defined as prior (≥12 months) myocardial infarction, revascularization procedure, coronary stenosis >50%, or chest pain associated with evidence of myocardial ischemia. Overall, 33,428 patients were screened, of whom 32,954 had data available for analysis at baseline; of these 2,229 (6.7%) had a history of AF. Median (interquartile range) CHA2DS2-VASc score was 4 (3, 5). Oral anticoagulation alone was used in 25.7%, antiplatelet therapy alone in 52.8% (single 41.8%, dual 11.0%), and both in 21.5%. OAC use was independently associated with permanent AF (pCONCLUSIONS: In this contemporary cohort of patients with stable coronary artery disease and AF, most of whom are theoretical candidates for anticoagulation, oral anticoagulants were used in only 47.2%. Half of the patients received antiplatelet therapy alone and one-fifth received both antiplatelets and oral anticoagulants. Absence of mitosis and meiosis are distinguishing properties of male germ cells during late fetal and early neonatal periods. Repressors of male germ cell meiosis have been identified, but mitotic repressors are largely unknown, and no protein repressing both meiosis and mitosis is known. We demonstrate here that the zinc-finger protein BNC2 is present in male but not in female germ cells. In testis, BNC2 exists as several spliced isoforms and presumably binds to DNA. Within the male germ cell lineage, BNC2 is restricted to prospermatogonia and undifferentiated spermatogonia. Fetal prospermatogonia that lack BNC2 multiply excessively on embryonic day (E)14.5 and reenter the cell cycle prematurely. Mutant prospermatogonia also engage in abnormal meiosis; on E17.5, Bnc2−/− prospermatogonia start synthesizing the synaptonemal protein SYCP3, and by the time of birth, many Bnc2−/− prospermatogonia have accumulated large amounts of nonfilamentous SYCP3, thus appearing to be blocked at leptonema. Bnc2−/− prospermatogonia do not undergo proper male differentiation, as they lack almost all the mRNA for the male-specific methylation protein DNMT3L and have increased levels of mRNAs that encode meiotic proteins, including STRA8. Bnc2−/− prospermatogonia can produce spermatogonia, but these enter meiosis prematurely and undergo massive apoptotic death during meiotic prophase. This study identifies BNC2 as a major regulator of male germ stem cells, which is required for repression of meiosis and mitosis in prospermatogonia, and for meiosis progression during spermatogenesis. In view of the extreme evolutionary conservation of BNC2, the findings described here are likely to apply to many species.

Ventricular premature beats are common in patients with mitral valve prolapse (MVP). The purpose of this study was to determine whether symptomatic patients with MVP had certain functional characteristics and if ventricular arrhythmia (VA) could be explained by functional extravalvular abnormalities. Single photon emission computed tomography equilibrium radionuclide angiography with Fourier phase analysis was preferred to the planar radionuclide method. Only patients without significant mitral regurgitation were studied. A total of 23 symptomatic patients with MVP (13 men, 10 women, mean age, 47 ± 14 years) without mitral regurgitation underwent single photon emission computed tomography equilibrium radionuclide angiography. Symptoms were present in 20 patients, and VA was present in 14 patients. Ejection fraction, regional wall motion, and Fourier phase analysis were examined in both ventricles and compared with results for normal subjects. Ventricular abnormalities were observed in 20 (87%) patients: decreased left ventricular and right ventricular ejection fractions, increased standard deviations of the mean phase and focal wall motion, and/or delayed phase abnormalities. Abnormalities were less frequent but more marked in the right ventricular free wall, the infundibulum, or the septum compared with left ventricular delayed abnormalities, which were more frequent but limited. In 12 of 14 patients with VA, phase-delayed areas were observed in the ventricle where the origin of ventricular premature beats was suspected on the basis of their electrocardiographic morphologic features. A relation was found between late potentials and delayed-phase areas (right ventricle or septum) and left bundle branch block morphologic features of VA. Symptomatic patients with MVP frequently have ventricular dysfunction in 1 or both ventricles, sometimes limited but more marked in the presence of severe VA even without significant mitral regurgitation, suggesting structural modification. The use of a sensitive, accurate, and 3-dimensional method such as single photon emission computed tomography equilibrium radionuclide angiography may be of interest for a noninvasive investigation, especially in young symptomatic patients with MVP and VA.

Complex motor skills require planning and programming before execution. The autonomic nervous system (ANS) is thought to transcribe these central operations at the peripheral level: a motor act is thought to be simultaneously programmed by central and autonomic nervous structures. The aim of this study was to verify that autonomic responses reflect the quality of central motor programming leading to successful or failed performance when subjects are required to perform a complex motor skill. The specificity of the ANS response has already been demonstrated through direct recording from sympathetic fibres. It has also been demonstrated through several mental tasks and closed motor skills such as shooting: ANS responses have been shown to be capable of distinguishing success from failure. The aim of this experiment was to test whether ANS responses are capable of distinguishing two levels of achievement during the performance of a skill involving uncertainty (open skill). The subjects had to intercept a ball on a volleyball court, using the forearm receive and pass technique, in order to pass it on to a moving human target. The results were displayed in terms of accuracy: accurate passes were successful and inaccurate passes missed the target. Six autonomic variables were recorded simultaneously during the task: skin resistance and potential, skin blood flow and temperature, instantaneous heart rate and respiratory frequency. Results showed that autonomic variables were capable of distinguishing success from failure in 22 subjects out of 24. This made it possible to build up autonomic patterns characterising subjects' performances, and to confirm that autonomic functioning may reveal information processing in the central nervous system. Thus, the study of autonomic responses may constitute an inferential model of central nervous system functioning. Such a method could be used as an index for the control of mental preparation.

It is now well established that mental imagery practice improves motor skills, but performance efficiency depends on many factors: the main one being individual differences. The aim of this study is to evaluate performance improvement with imagery quality estimated during ANS recording. Volleyball training (“receiving serve”) afforded us the experimental paradigm. Subjects were required to pass an opponent’s serve to a given team mate. The receiver’s performance was evaluated from the accuracy of his pass to the targeted team mate. From these first test results, subjects were divided into two equivalent groups: imagers and controls. After mental practice the two groups were submitted to a posttest similar to the first one. During the pretest, posttest actual practice as well as the last session of corresponding mental rehearsal, six autonomic parameters were continuously recorded. Furthermore, and for the first time, a grade obtained from four different aspects of this response permits qualitative evaluation of each subject’s mental imagery. This estimation, based on the well-established link between performance and autonomic response, is validated by the fact that good correlation was obtained between this grade and the performance improvement of each of the “imager” group subjects.

The aim of this study was to assess objectively the processes of mental rehearsing (imagery) by measuring variations of the autonomic nervous system (or ANS responses) during an open-ended complex motor skill in two actual experiments (volleyball) and during mental rehearsing taking place between them. Comparison between pre- and post-test (volleyball) scores related to imagining and non-imagining performances revealed significant improvement in the former (chi2 = 20.9, P0.00001) while in the latter chi2 = 27, P0.9, NS. The ANS parameters (skin potential and resistance, skin temperature and heat clearance, instantaneous heart rate and respiratory frequency) were quantified by original techniques and indices. Results from a principal component analysis showed a strong correlation between the responses in actual tasks (pre- and post-test volleyball) and during mental imagery, since the same preferential variables appeared on the main axis in 87% of cases. Thus the same autonomic channels seemed to be used during the actual activity and during the mental imagery of this activity. So far as phasic results were concerned, the main finding was a differing development of skill between imagining and non-imagining volleyball players. No clear difference was seen between pre- and post-tests in non-imaginers, except an increase in the median of the duration of the response observed in heat clearance, m1 and m2 respectively [m1 = 5.8 (SD 4.1) s, m2 = 7.6 (SD 3.9) s, P0.001]. Conversely, for other ANS parameters, a significant decrease was seen in the post-test responses compared to pre-test responses in the imagining group [for instance, the median of the duration of the resistance responses decreased from m1 = 12.6 (SD 4.3) s, and m2 = 7.8 (SD 4.5) s, P0.0001 in imaginers, while no change was observed in non-imaginers: 9.6 (SD 6.0) s vs 9.5 (SD 6.1) s, NS] except in the duration of the heat clearance response where an increase was seen [m1 = 7.3 (SD 5.0) s vs m2 = 7.6 (SD 3.1) s, NS]. Compared to the non-imagining group, the latter result may also have been associated with a response decrease in the imagining group. Thus mental rehearsing induced a specific pattern of autonomic response: decreased amplitude, shorter duration and negative skin potentials compared to the control group. As this pattern was associated with better performance in the tests it can be suggested that in the case of open-ended motor activity, mental rehearsing may help in the construction of schema which can be reproduced, without thinking, in actual practice. Thus a neural information process might develop in the central nervous system changing from a parallel into a serial treatment.

The aim of this paper is to analyse the relationship between self-report hedonic evaluations and the physiological expression of emotion in response to odorants. We try to solve the following questions: (1) Is it possible to find any experimental evidence that the sense of smell is linked with emotion? (2) What kind of odorants can be distinguished by autonomic analysis? (3) Is there a link between hedonics and autonomic information? The effects of odorants on the emotional process were estimated, in terms of autonomic nervous system (ANS) activity. Fifteen subjects inhaled five odorants as olfactory stimuli: lavender (LAV), ethyl acetoacetate (EAA), camphor (CAM), acetic acid (AA) and butyric acid (BA). After inhaling the odorant, subjects were requested to fill out an 11-point hedonic scale to rate its pleasantness versus unpleasantness. ANS parameters were as follows: two electrodermal responses, skin potential (SP) and resistance (SR); two thermovascular parameters, skin blood flow (SBF) and skin temperature (ST); and two cardiorespiratory parameters; instantaneous respiratory frequency (IRF) and instantaneous heart rate (IHR). Simultaneous recording of six parameters showed that specific autonomic patterns were associated with each odorant. An analysis of variance made it possible to differentiate among the five odorants. Two-by-two odorant comparisons for autonomic responses using Tukey's HSD multiple comparison test only permitted differentiation between pleasant odorants (LAV and EAA) and unpleasant (AA and BA) ones, but camphor was differentiated from both pleasant and unpleasant odorants. Each odorant elicited responses in the different parameters, yet subjects responded through their preferential channels; an average of two channels was used by each subject. These results when compared with those obtained with other senses (visual and auditory), did not evidence the postulated preferential link between olfaction and emotion. A strong link between hedonics and ANS response could be demonstrated when considering each subject and mainly through his/her preferential channel(s); conversely a weak correlation (SR duration excepted) was obtained between inter-subjects' hedonic evaluation. It seems that for a given population the autonomic response reflect the odor valence only through some parameters related to the main preferential channel(s) and thus the global autonomic pattern has to be considered.

Co-Drive on-board traffic information system is a complementary tool providing a dynamic management of transportation infrastructure and traffic as well as the diffusion of accurate real-time information about the road environment and motorists’ driving behaviour. The aim of this study was to examine drivers’ acceptability of Co-Drive by investigating the impact of traffic information provided via on-board display devices on motorists’ beliefs and behaviour. 116 drivers (Men = 46.6%), between 22 and 62 years, participated to a driving simulator experiment. They were randomly divided into two experimental groups according to the type of display device (Black-berry vs. iPhone) and a control group. The experimental groups were exposed to fourteen on-board traffic messages: warning (e.g., road crash), recommendation (e.g., the use of seat-belt) and comfort messages (e.g., the location of a gas station). They had to validate each message by pushing the headlight flashing button as soon as they understood it. At the end, all participants had to fill in a questionnaire. Drivers evaluated positively the on-board messages, expressed a high level of confidence in the on-board information and estimated having received it sufficiently in advance for them to adjust their behaviour. Regardless of the type of display device, they took more time to read warning and recommendation messages as compared to comfort messages and complied with them. Finally, those exposed to the messages adapted their behaviour easier to the road events than those who did not receive them. Practical implications of the results are discussed.

Today's increase in the number of cyclists has triggered a change in the interactions to be handled by road users. However, few studies have investigated crash risk perceived by cyclists interacting with other users, and few have compared cyclists' and drivers' perceptions of crash risk in bike-car interactions, the most dangerous situation for cyclists. Our aims here are to study perceived crash risk (no matter the seriousness of the crash) in six common road situations during which cyclist crashes are frequent and also to study cyclists' and drivers' perceived risk in bike-car interactions, in comparison to other interaction types (cyclist vs. cyclist and driver vs. driver). We predicted that perceived risk of being involved in a crash during a particular interaction would be greater when in interaction with a car than with a bike, and that drivers would perceive more risk than cyclists would. We also predicted that perceived risk would decrease with drivers' and cyclists' experience of their transportation mode and their perceived control over the interaction situation. We ran an online survey on two samples, experienced cyclists (N=336) and non-cyclist car drivers (N=92). Participants evaluated their personal risk - as cyclists or as drivers - of being involved in a road crash if they were in an interaction with a bike or a car for each of six risky road situations. Experience was measured in terms of years of vehicle driving and driven km; perceived control was measured in terms of perceived skill and responsibility for the risky behavior. The results validated our hypotheses: perceived risk was higher for car drivers than for cyclists and for interacting with a car than with a bike. The reproduction of the Arabian oryx (Oryx leucoryx) has been studied in a breeding center in Taif (Saudi Arabia). In the first experiment, three groups of females were assigned to groups immediately after calving. In group 1, from the day of calving the females had visual and olfactory but not physical contact with a male. The calves were removed on the day of birth and each female was placed with a male on Day 45. In group 2, females and their calves were placed individually with a male the day after calving. In group 3, females and their calves were kept with neither visual nor olfactory contact with a male and then placed with a male on Day 90. The estrous cycle was deduced from assays of plasma progesterone up to Day 120 after calving. All females exhibited a postpartum estrus a few days after parturition but the occurrence of the estrus was significantly delayed in females of group 3. The length of the estrous cycle was 23.7 +/- 1.3 days and the gestation length was 255.2 +/- 1.5 days. In the second experiment, a group of adult females was formed in 1987 and the intervals between calving recorded over a period of 5 years. The mean interval between calving was 285 +/- 2 days (n = 99 gestations). However, 66.6% of these intervals averaged 276 days whereas 19.2% had averaged 294 and 13% averaged 321 days.

Bioelectronic phenomena related to the activity of the Autonomic Nervous System (ANS) activity mainly take place in the deep part of the brain where they are difficult to record. The hand skin, being richly innervated by sympathetic efferent fibres, offers an interface from which relevant ANS-related signals can be recorded. Two non-invasive bioelectronic measurements (skin resistance and potential) reflecting the activity of the ANS were performed at the skin surface. These measurements were complemented by thermovascular (skin microcirculation and skin temperature) and cardiorespiratory (instantaneous heart rate and instantaneous respiratory frequency) measurements. The bioelectric measurements were performed using non-invasive Ag/AgCl electrodes. Non-metallic NASICON (Na Super Ionic Conductor) electrodes were tested and compared with traditional electrodes for the optimization of bioelectric measurements. Signal analysis and data processing was accomplished by means of original indices on a specially designed PC-based software. The methodology was used to evaluate vigilance level, mental workload, and emotional response during tasks (sporting activity, mental calculation, olfactive stimuli) and in critical situations (car crash avoidance).

The objective of this study was to evaluate the effects of a combination of 6% low-density lipoproteins (LDL) and 20 mm glutamine in comparison with other extenders used for the refrigeration of canine semen: Tris egg yolk (EY) 20% and 6% LDL. The percentages of mobile spermatozoa after 4 days storage in a domestic refrigerator at +4 °C were 53.1%, 44.2% and 52.2% for the 6% LDL + 20 mm glutamine, 20% EY and 6% LDL extenders respectively for 100% of the dogs. After 7 days of storage, these percentages fell to 37.8%, 26.4% and 33.6% in the same extenders for 50% of the dogs. In vitro fertility tests were performed with all of the extenders following the mobility results. These tests were conducted on the day of sampling (D0), and 48 and 96 h after sampling. The results of the hypo-osmotic swelling test were 82.6%, 81.2% and 85.7% on D0, 75.2%, 74.1% and 78.5% on D2, and 70.8%, 71% and 76.1% on D4 for the 6% LDL + 20 mm glutamine, 20% EY and 6% LDL extenders, respectively. For the FITC/pisum sativum agglutinin (PSA) test, the results were 81.5%, 70.2% and 84.8% on D0, 78.9%, 62.3% and 84.2% on D2, and 72.7%, 59.6% and 73.7% on D4 for the 6% LDL + 20 mm glutamine, 20% EY and 6% LDL extenders, respectively. The acridine orange test was positive; in nearly 100% of cases, none of the spermatozoa had been denatured on D0, D2 and D4. The 6% LDL + 20 mm glutamine and the 6% LDL extenders are capable of preserving spermatozoa that have been stored in a domestic refrigerator at +4°C for at least 4 days. This means that the spermatozoa retain good cytoplasmic membrane integrity, had not capacitated and contained intact DNA in comparison with spermatozoa preserved in the egg yolk extender. The duration of storage is a very important consideration when faced with the problem of sending semen over ever-greater distances.

Twenty semen samples taken from 5 dogs were frozen in liquid nitrogen at -196 °C in four different extenders: one control extender based on 20% egg yolk, 6% LDL alone (low density lipoproteins: the active cryoprotective principle in chicken egg yolk), 6% LDL combined with 20 mmol glutamine, and Equex® (a reference extender that we wish to compare with the LDL-glutamine combination). After thawing, spermatozoal motility was evaluated using a HAMILTON THORNE CERROS 12 image analyzer; the percentage of motile spermatozoa was 27.7% in the egg yolk extender (p0.05), 49.9% with 6% LDL alone (p0.05), 54.7% in the 6% LDL+20 mmol glutamine extender, and 47.9% with Equex® (p0.05). The motility parameters (VAP, VCL, VSL and ALH) were also superior in the 6% LDL+20 mmol glutamine extender in comparison with the other extenders. Finally, the spermatozoa were generally better protected during freezing with the 6% LDL+20 mmol glutamine association than with the egg yolk, 6% LDL, or Equex extenders in terms of the flagellar plasma membrane (HOS test), DNA (Acridine orange test), and acrosome integrity (Spermac® test: no significant difference). The Equex® extender obtained the best results for the acrosome, followed by 6% LDL+20 mmol glutamine (FITC-PSA test: p0.05 between each extender).

Objective: The study addressed the role of familiarization on a driving simulator with a forward collision warning (FCW) and investigated its impact on driver behavior. Background: Drivers need a good understanding of how an FCW system functions to trust it and use it properly. Theoretical and empirical data suggest that exploring the capacities and limitations of the FCW during the learning period improves operating knowledge and leads to increased driver trust in the system and better driver-system interactions. The authors tested this hypothesis by comparing groups of drivers differing in FCW familiarity. Method: During the familiarization phase, familiarized drivers were trained on the simulator using the FCW, unfamiliarized drivers simply read an FCW manual, and control drivers had no contact with the FCW. During the test, drivers drove the simulator and had to interact with traffic; both familiarized and unfamiliarized drivers used the FCW, whereas controls did not. Results: Simulator familiarization improved driver understanding of FCW operation. Driver-system interactions were more effective: Familiarized drivers had no collisions, longer time headways, and better reactions in most situations. Familiarization increased trust in the FCW but did not raise system acceptance. Conclusion: Familiarization on the simulator had a positive effect on driver-system interactions and on trust in the system. The limitations of the familiarization method are discussed in relation to the driving simulator methodology. Application: Practicing on a driving simulator with driving-assistance systems could facilitate their use during real driving.

OBJECTIVES: In some driving schools in France, several trainers may successively train a single trainee. This situation can be described as a case of asynchronous collaboration. In this paper, we examine the consequences of this situation in two different studies: the first explores the trainers' activity while the second concerns the trainees. PARTICIPANTS: The participants in the first study were 6 trainers with different levels of experience, each working with 2 different trainees. Four of them met their trainees for the very first time. The second study included 150 trainees from 13 driving schools in Paris. METHODS: In the first study, the driving lessons were recorded and fully transcribed before a thematic analysis was conducted. In the second study, the trainees completed a questionnaire. We collected both socio-demographic and driver training data. RESULTS: In the first study, we observed that changing trainers during a trainee's driving course had an impact on the trainers' activity. We identified difficulties in establishing a diagnosis of the trainee's progress, and the organization of the driving lesson was altered. The second study highlighted the fact that changing trainers at the beginning of a course increased the duration of the training period. CONCLUSION: The impacts of changing trainers during a trainee's driving course are discussed with regard to both the trainers' scaffolding activity and the trainees' skill development. Language: en

We studied a man with distal hypospadias, partial anomalous pulmonary venous return, mild limb-length inequality and a balanced translocation involving chromosomes 9 and 13. To gain insight into the etiology of his birth defects, we mapped the translocation breakpoints by high-resolution comparative genomic hybridization (CGH), using chromosome 9- and 13-specific tiling arrays to analyze genetic material from a spontaneously aborted fetus with unbalanced segregation of the translocation. The chromosome 13 breakpoint was ∼400 kb away from the nearest gene, but the chromosome 9 breakpoint fell within an intron of Basonuclin 2 (BNC2), a gene that encodes an evolutionarily conserved nuclear zinc-finger protein. The BNC2/Bnc2 gene is abundantly expressed in developing mouse and human periurethral tissues. In all, 6 of 48 unrelated subjects with distal hypospadias had nine novel nonsynonymous substitutions in BNC2, five of which were computationally predicted to be deleterious. In comparison, two of 23 controls with normal penile urethra morphology, each had a novel nonsynonymous substitution in BNC2, one of which was predicted to be deleterious. Bnc2(-/-) mice of both sexes displayed a high frequency of distal urethral defects; heterozygotes showed similar defects with reduced penetrance. The association of BNC2 disruption with distal urethral defects and the gene's expression pattern indicate that it functions in urethral development.

Although the prognosis of most glioblastoma patients is poor, 3%-5% patients show long-term survival of 36 months or longer after diagnosis. To study the differences in activation of biochemical pathways, we performed mRNA and protein expression analyses of primary glioblastoma tissues from 11 long-term survivors (LTS; overall survival ≥ 36 months) and 12 short-term survivors (STS; overall survival ≤ 6 months). The mRNA expression ratio of the retinoic acid transporters fatty acid-binding protein 5 (FABP5) and cellular retinoic acid-binding protein 2 (CRABP2), which regulate the differential delivery of retinoic acid to either antioncogenic retinoic acid receptors or prooncogenic nuclear receptor peroxisome proliferator-activated receptor delta, was statistically significantly higher in the tumor tissues of STS than those of LTS (median ratio in STS tumors = 3.64, 10th-90th percentile = 1.43-4.54 vs median ratio in LTS tumors = 1.42, 10th-90th percentile = -0.98 to 2.59; P < .001). High FABP5 protein expression in STS tumors was associated with highly proliferating tumor cells and activation of 3-phosphoinositide-dependent protein kinase-1 and v-akt murine thymoma viral oncogene homolog. The data suggest that retinoic acid signaling activates different targets in glioblastomas from LTS and STS. All statistical tests were two-sided.

Promoting self-assessment accuracy among student drivers could help improve road safety for young novice drivers (Minimum Requirement for Driving Instructor Training, 2005). It is crucial to investigate the development of this ability in student drivers, although few studies have been conducted on this issue to our knowledge. The present study was aimed at examining the development of accuracy in student drivers' self-assessments. Students' self-ratings of their skills were compared to instructors' ratings based on the students' progression through the four steps of the French driver training course. The ratings were collected from two versions of an ad hoc questionnaire about students' ability to turn right, overtake another vehicle, and turn left in driving situations. The questionnaire included a three-part assessment scale (six driving subtasks required to perform the maneuver, the number of driving subtasks managed simultaneously, and the student's autonomy), demographic data, and the students' progress in the training program. The participants were 149 student drivers (58 men and 91 women) with a mean age of 22.2 years, and 38 professional driving instructors (36 men and 2 women) from 13 driving schools in Paris. The psychometric quality of the scale was satisfactory (α(s)>.9) concerning the self-ratings as well as the instructors' ratings in the three parts of the scale. As a whole and as expected, the self-ratings and the instructors' ratings became increasingly positive as the training progressed, on each part of the assessment scale. The students' and the instructors' ratings did not differ significantly between the three driving situations tested, nor between male and female students in each training step. Furthermore, students' overestimation of their driving skills was mainly observed on step 1, less on steps 2 and 3, but never on step 4. Thus, the students became rather aware of their current driving skills as the driver training course progressed. The results on the development of the self-assessment accuracy in driver training are discussed.

Due to the innate complexity of the task drivers have to manage multiple goals while driving and the importance of certain goals may vary over time leading to priority being given to different goals depending on the circumstances. This study aimed to investigate drivers' behavioral regulation while managing multiple goals during driving. To do so participants drove on urban and rural roads in a driving simulator while trying to manage fuel saving and time saving goals, besides the safety goals that are always present during driving. A between-subjects design was used with one group of drivers managing two goals (safety and fuel saving) and another group managing three goals (safety, fuel saving, and time saving) while driving. Participants were provided continuous feedback on the fuel saving goal via a meter on the dashboard. The results indicate that even when a fuel saving or time saving goal is salient, safety goals are still given highest priority when interactions with other road users take place and when interacting with a traffic light. Additionally, performance on the fuel saving goal diminished for the group that had to manage fuel saving and time saving together. The theoretical implications for a goal hierarchy in driving tasks and practical implications for eco-driving are discussed. (C) 2011 Elsevier Ltd. All rights reserved.

International audience; Twenty sperm samples from five dogs were frozen in liquid nitrogen at −196°C in 16 different media, two control media containing 20% egg yolk and 6% low-density lipoproteins (LDL); 10 test media containing 6% LDL (the active cryoprotective ingredient of chicken egg yolk) combined with 10, 20, 30, 40, 50, 60, 70, 80, 90, and 100 mmol of glutamine respectively at 4%, 5%, 7%, and 8% LDL. Following thawing, sperm mobility was assessed using an image analyser, HAMILTON THORN CERROS 12. The percentage of mobile spermatozoa was 62.05% in the 6% LDL + 20 mmol glutamine medium compared with 48.90% in the egg yolk-based medium (p < 0.05) or 57.55% for the 6% LDL medium (p < 0.05). Furthermore, in most cases, the motility parameters (average path velocity, curvilinear velocity, straight line velocity) in the 6% LDL + 20 mmol glutamine medium, were superior, to a statistically significant extent, to those in the control media. Finally, the 6% LDL + 20 mmol glutamine combination provides spermatozoa with better protection during freezing than egg yolk or the 6% LDL medium alone in terms of acrosome integrity (fluorescein isothiocyanate–Pisum sativum agglutinin test: p < 0.05), the flagellar plasma membrane (hypo-osmotic test: p < 0.05 for 6% LDL), the DNA (acridine orange test; no significant difference) and the integrity of the acrosome (Spermac® test: no significant difference)

International audience; Promoting self-assessment accuracy among student drivers could help improve the road safety for young novice drivers (Minimum Requirement for Driving Instructor Training, 2005). However, it is essential to first examine the time course of student drivers’ assessments of their own driving skills. As a result, the present study examined the time course of student drivers’ self-assessments in relation to their general driving abilities during the four steps of French driver training. We used Victoir et al.’s (2005) self-efficacy scale, which we translated into French. We set four goals for the present study: (1) to examine the psychometric qualities of this self-assessment scale, (2) to study the time courses of the students’ self-assessments, (3) to investigate the relationship of these time courses to the number of driving hours that the students estimated that they needed to complete before taking the driving test, and (4) to compare the number of hours estimated by the students to the number of hours estimated by their driving instructors. In total, 150 students (58 men and 92 women) and 38 instructors from 13 driving schools in Paris participated in the present study. The self-assessment scale was composed of 12 items that were rated on a 7-point Likert scale that ranged from 1 (certainly so) to 7 (certainly not). The internal consistency of the scale was satisfactory (α = .88). The self-assessments became increasingly positive as the training progressed (at the beginning of training, M = 3.45 vs. at the completion of the training, M = 4.8). Globally, the men assessed themselves more positively than the women. However, no significant gender difference was observed at each training step. The students’ self-ratings were negatively correlated with the number of driving hours that they estimated they still needed before taking the driving test. This number did not differ significantly from the number of hours that was estimated by the instructors at each training step throughout the training. The results describing the time course of the student drivers’ self-assessments during driver training and this time course's correlation with the estimated number of driving hours still needed to take the driver test were discussed.

International audience; Chicken egg yolk is held as an excellent cryoprotective agent for freezing canine semen. Recent advances have enabled the extraction of low density lipoproteins from egg yolk, which are responsible for the cryoprotective abilities of the latter. The objective of this article was to compare 3 semen extenders for freezing canine semen: 2 containing egg yolk (Tris egg yolk and Equex STAMP) and one containing 6% LDL. After freezing and thawing 20 ejaculates from 5 different dogs, the 6% LDL extender produced 50% mobile spermatozoa, compared with 48% with the Equex® extender and 27.7% with the extender containing egg yolk alone (EY). In vitro functional tests demonstrated that the integrity of the plasma membrane (hypoosmotic test) was respected in 65–66% of spermatozoa as a function of the extender; DNA integrity was respected in more than 97% of the spermatozoa. The Equex® extender provided superior acrosome integrity (FITC/PSA test): 68.4% compared with 55.1% with LDL and 53.3% with egg yolk. However, the 6% LDL extender resulted in fewer spermatozoal anomalies (Spermac® test), with 54.6% normal spermatozoa compared to 53.6% for Equex® and 53.3% with the egg yolk. All six of the bitches inseminated artificially via the intra-uterine route (Scandinavian technique) using semen frozen in the 6% LDL extender became pregnant. The LDL extender resulted in percentages of mobile spermatozoa and movement characteristics that were as good if not better than those obtained with the reference extenders following thawing. The 6% LDL extender appears to have the same cryoprotective qualities as the reference diluent, Equex STAMP

Basonuclin 2 is a recently discovered zinc finger protein of unknown function. Its paralog, basonuclin 1, is associated with the ability of keratinocytes to multiply. The basonuclin zinc fingers are closely related to those of the Drosophila proteins disco and discorelated, but the relation between disco proteins and basonuclins has remained elusive because the function of the disco proteins in larval head development seems to have no relation to that of basonuclin 1 and because the amino acid sequence of disco, apart from the zinc fingers, also has no similarity to that of the basonuclins. We have generated mice lacking basonuclin 2. These mice die within 24 h of birth with a cleft palate and abnormalities of craniofacial bones and tongue. In the embryonic head, expression of the basonuclin 2 gene is restricted to mesenchymal cells in the palate, at the periphery of the tongue, and in the mesenchymal sheaths that surround the brain and the osteocartilagineous structures. In late embryos, the rate of multiplication of these mesenchymal cells is greatly diminished. Therefore, basonuclin 2 is essential for the multiplication of craniofacial mesenchymal cells during embryogenesis. Non- Drosophila insect databases available since 2008 reveal that the basonuclins and the disco proteins share much more extensive sequence and gene structure similarity than noted when only Drosophila sequences were examined. We conclude that basonuclin 2 is both structurally and functionally the vertebrate ortholog of the disco proteins. We also note the possibility that some human craniofacial abnormalities are due to a lack of basonuclin 2.

A mixed foot-in-the-door procedure was set up to lead offenders participating in French driver-rehabilitation training courses to commit to complying with speed limits. To improve this procedure, the reasons that led participants in such courses to commit or not were investigated in this preliminary study of the road safety research programs. The study participants were 89 men ( M= 34 yr., range = 18–64 yr., SD= 13) attending driver-rehabilitation courses. Among them. 55% committed. The commitment was mainly explained by the drivers' will to change their speeding behaviour and their refusal by the perceived positive consequences of speeding. Moreover, some participants stated that they would be unable to follow through with the commitment to comply strictly with speed limits. After talking to the participants, it became clear that complying with speed limits was understood in a more or less strict way. In subsequent studies, some of the reasons given were used to prompt group discussion before asking participants to commit, and the meaning of “compliance with speed limits” was better defined.

The aims of this study were to provide further evidence of validity and reliability for the Driving Anger Expression Inventory (DAX) French adaptation (Villieux & Delhomme, 2008, Le Travail Humain, 71(4), 359-384) and to investigate the relationships between driving anger, how people express their anger while driving, and traffic violations among young drivers in France. Method: The French adaptations of the DAX, of the Driving Anger Scale (DAS), and of the Extended Violations Scale were administered to a sample of 314 drivers. Results: Confirmatory factor analysis of the French DAX items yielded a three factors solution with 11 items, which obtained better goodness-of-fit to the data. Cronbach α reliabilities for DAX factors ranged from .71 to .79. Aggressive forms of anger expression correlated positively with driving anger and traffic violations whereas the 'Adaptive/Constructive Expression' factor correlated negatively with these variables. Discussion: Globally, our results replicated earlier findings and showed that DAX factors are useful predictors of self reported violations and complement established measures like the DAS. Impact on Industry: Implications for driver education and interventions were examined.
