Beside the well-documented involvement of secondary somatosensory area, the cortical network underlying late somatosensory evoked potentials (P60/N60 and P100/N100) is still unknown. Electroencephalogram and magnetoencephalogram source imaging were performed to further investigate the origin of the brain cortical areas involved in late somatosensory evoked potentials, using sensory inputs of different strengths and by testing the correlation between cortical sources. Simultaneous high-density electroencephalograms and magnetoencephalograms were performed in 19 participants, and electrical stimulation was applied to the median nerve (wrist level) at intensity between 1.5 and 9 times the perceptual threshold. Source imaging was undertaken to map the stimulus-induced brain cortical activity according to each individual brain magnetic resonance imaging, during three windows of analysis covering early and late somatosensory evoked potentials. Results for P60/N60 and P100/N100 were compared with those for P20/N20 (early response). According to literature, maximal activity during P20/N20 was found in central sulcus contralateral to stimulation site. During P60/N60 and P100/N100, activity was observed in contralateral primary sensorimotor area, secondary somatosensory area (on both hemispheres) and premotor and multisensory associative cortices. Late responses exhibited similar characteristics but different from P20/N20, and no significant correlation was found between early and late generated activities. Specific clusters of cortical activities were activated with specific input/output relationships underlying early and late somatosensory evoked potentials. Cortical networks, partly common to and distinct from early somatosensory responses, contribute to late responses, all participating in the complex somatosensory brain processing., (© 2024 The Authors. Statistical power is key for robust, replicable science. Here, we systematically explored how numbers of trials and subjects affect statistical power in MEG sensor-level data. More specifically, we simulated "experiments" using the MEG resting-state dataset of the Human Connectome Project (HCP). We divided the data in two conditions, injected a dipolar source at a known anatomical location in the "signal condition", but not in the "noise condition", and detected significant differences at sensor level with classical paired t-tests across subjects, using amplitude, squared amplitude, and global field power (GFP) measures. Group-level detectability of these simulated effects varied drastically with anatomical origin. We thus examined in detail which spatial properties of the sources affected detectability, looking specifically at the distance from closest sensor and orientation of the source, and at the variability of these parameters across subjects. In line with previous single-subject studies, we found that the most detectable effects originate from source locations that are closest to the sensors and oriented tangentially with respect to the head surface. In addition, cross-subject variability in orientation also affected group-level detectability, boosting detection in regions where this variability was small and hindering detection in regions where it was large. Incidentally, we observed a considerable covariation of source position, orientation, and their cross-subject variability in individual brain anatomical space, making it difficult to assess the impact of each of these variables independently of one another. We thus also performed simulations where we controlled spatial properties independently of individual anatomy. These additional simulations confirmed the strong impact of distance and orientation and further showed that orientation variability across subjects affects detectability, whereas position variability does not. Importantly, our study indicates that strict unequivocal recommendations as to the ideal number of trials and subjects for any experiment cannot be realistically provided for neurophysiological studies and should be adapted according to the brain regions under study., (Copyright © 2021 The Author(s). Background: Amygdala is a key brain region for face perception. While the role of amygdala in the perception of facial emotion and gaze has been extensively highlighted with fMRI, the unfolding in time of amydgala responses to emotional versus neutral faces with different gaze directions is scarcely known., Methodology/principal Findings: Here we addressed this question in healthy subjects using MEG combined with an original source imaging method based on individual amygdala volume segmentation and the localization of sources in the amygdala volume. We found an early peak of amygdala activity that was enhanced for fearful relative to neutral faces between 130 and 170 ms. The effect of emotion was again significant in a later time range (310-350 ms). Moreover, the amygdala response was greater for direct relative averted gaze between 190 and 350 ms, and this effect was selective of fearful faces in the right amygdala., Conclusion: Altogether, our results show that the amygdala is involved in the processing and integration of emotion and gaze cues from faces in different time ranges, thus underlining its role in multiple stages of face perception.

The relationship between episodic and semantic memory systems has long been debated. Some authors argue that episodic memory is contingent on semantic memory (Tulving 1984), while others postulate that both systems are independent since they can be selectively damaged (Squire 1987). The interaction between these memory systems is particularly important in the elderly, since the dissociation of episodic and semantic memory defects characterize different aging-related pathologies. Here, we investigated the interaction between semantic knowledge and episodic memory processes associated with faces in elderly subjects using an experimental paradigm where the semantic encoding of famous and unknown faces was compared to their episodic recognition. Results showed that the level of semantic awareness of items affected the recognition of those items in the episodic memory task. Event-related magnetic fields confirmed this interaction between episodic and semantic memory: ERFs related to the old/new effect during the episodic task were markedly different for famous and unknown faces. The old/new effect for famous faces involved sustained activities maximal over right temporal sensors, showing a spatio-temporal pattern partly similar to that found for famous versus unknown faces during the semantic task. By contrast, an old/new effect for unknown faces was observed on left parieto-occipital sensors. These findings suggest that the episodic memory for famous faces activated the retrieval of stored semantic information, whereas it was based on items' perceptual features for unknown faces. Overall, our results show that semantic information interfered markedly with episodic memory processes and suggested that the neural substrates of these two memory systems overlap.

Others' gaze and emotional facial expression are important cues for the process of attention orienting. Here, we investigated with magnetoencephalography (MEG) whether the combination of averted gaze and fearful expression may elicit a selectively early effect of attention orienting on the brain responses to targets. We used the direction of gaze of centrally presented fearful and happy faces as the spatial attention orienting cue in a Posner-like paradigm where the subjects had to detect a target checkerboard presented at gazed-at (valid trials) or non gazed-at (invalid trials) locations of the screen. We showed that the combination of averted gaze and fearful expression resulted in a very early attention orienting effect in the form of additional parietal activity between 55 and 70 ms for the valid versus invalid targets following fearful gaze cues. No such effect was obtained for the targets following happy gaze cues. This early cue-target validity effect selective of fearful gaze cues involved the left superior parietal region and the left lateral middle occipital region. These findings provide the first evidence for an effect of attention orienting induced by fearful gaze in the time range of C1. In doing so, they demonstrate the selective impact of combined gaze and fearful expression cues in the process of attention orienting.

Abstract: Inferring someone''s personality from his or her photograph is a pervasive and automatic behavior that takes place even if no reliable information about one''s character can be derived solely from facial features. This illustrates nicely the idea that perception is not a passive process, but rather an active combination of current sensory inputs with endogenous knowledge derived from prior experience. To understand how and when neural responses to faces can be modulated by prior experience, we recorded magneto-encephalographic (MEG) responses to new faces, before and after subjects were exposed for a short period of 15–20min to an experimentally induced association between a facial feature (inter-eye distance) and a response (personality judgment). In spite of the absence of any observable response bias following such a short reinforcement phase, our experimental manipulation influenced neural responses to faces as early as 60–85ms. Source localization of magneto-encephalographic signals, confirmed by intracranial recordings, suggests that prior experience modulates early neural processing along two initially independent neural routes, one initiated in an anterior system that includes the orbitofrontal cortex and the temporal poles, and the second one involving face-sensitive regions in the ventral visual pathway. The two routes are both active as early as 60ms but engage in reciprocal interactions only later, between 135 and 160ms. These experimental findings support recent models assuming the existence of a fast anterior pathway activated in parallel with the ventral visual system which would link prior experience with current sensory inputs. Gaze direction is an important social signal for human beings. Beside the role of gaze in attention orienting, direct gaze (that is, gaze directed toward an observer) is a highly relevant biological stimulus that elicits attention capture and increases face encoding. Brain imaging studies have emphasized the role of the superior temporal sulcus (STS) in the coding of gaze direction and in the integration of gaze and head cues of social attention. The dynamics of the processing and integration of these cues remains, however, unclear. In order to address this question, we used deviated and frontal faces with averted and direct gaze in a combined electro- and magneto- encephalography (EEG–MEG) study. We showed distinct effects of gaze direction on the N170 and M170 responses. There was an interaction between gaze direction and head orientation between 134 and 162 ms in MEG and a main effect of gaze direction between 171 and 186 ms in EEG. These effects involved the posterior and anterior regions of the STS respectively. Both effects also emphasized the sensitivity to direct gaze. Abstract: The processing of upright, inverted and contrast-reversed faces was investigated using MEG. Peak and global field power analyses revealed that the M100, M170 and M220 components were delayed for inverted and contrast-reversed compared to normal upright faces but no amplitude modulations were found. Source analyses using an event-related SAM beamformer technique revealed bilateral occipital sources for the M100 and M220 components. For the M170, two distinct sources simultaneously active were found, a bilateral and posterior source (M170A) and a right lateralized ventral and more anterior source (M170B) around the fusiform gyrus. None of the sources varied in location or intensity between face types. However, although different from the M100, the location of the M170A was not significantly different from that of the M220, suggesting the latter could be a reactivation of the former. Confirming previous ERP results on the processing of inverted faces, the present study extends the findings to contrast-reversed face stimuli and suggests that deviations from the standard upright face format do not activate extra areas but simply result in the delayed activation of the sources generating the M100, M170 and M220 components. The data confirm the sensitivity of the M100 to face manipulations and further suggest that the M170 is generated by two distinct sources, one of which situated in occipital extrastriate areas (M170A) could be reactivated around 220 ms to generate the M220 component. Other's eye gaze is a powerful attention orienting cue that can change our perception of objects in the environment. Here, we seek to characterize the influence of attention orienting by eye gaze on the neural processing of visual targets. We used a Posner-like cueing paradigm to investigate with magnetoencephalography the brain responses associated with target processing. We analyzed the cerebral sources of the evoked responses to visual targets that were validly or invalidly cued by eye gaze. The effect of attention orienting was reflected in faster reaction times to valid than invalid targets. At the brain level, we showed an early influence of attention orienting by gaze with enhanced brain responses for invalid relative to valid targets. This influence was maximum contra-laterally to the target, with a right hemisphere dominance. Responses to targets presented in the left visual field were modulated between 91 and 400 ms in the right posterior parietal and occipital cortices. Responses to targets presented in the right visual field were modulated between 174 and 218 ms in the left superior parietal cortex. Our results confirm previous EEG studies that demonstrated early influence of attention orienting by gaze on target processing and provide evidence for the sources of this effect in occipito-parietal regions. Abstract: Recent electrophysiological studies have demonstrated modulations of the very first stages of visual processing (<100ms) due to prior experience. This indicates an influence of a memory trace on the earliest stages of stimulus processing. Here we investigated if emotional audio-verbal information associated with faces on first encounter can affect the very early responses to those faces on subsequent exposure. We recorded magneto-encephalographic (MEG) responses to neutral faces that had been previously associated with positive (happy), negative (angry) or neutral auditory verbal emotional contexts. Our results revealed a very early (30–60ms) difference in the brain responses to the neutral faces according to the type of previously associated emotional context, with a clear dissociation between the faces previously associated to positive vs. negative or neutral contexts. Source localization showed that two main regions were involved in this very early association effect: the bilateral ventral occipito-temporal regions and the right anterior medial temporal region. These results provide evidence that the memory trace of a face integrates positive emotional cues present in the context of prior encounter and that this emotional memory can influence the very first stages of face processing. These experimental findings support the idea that face perception can be shaped by experience from its earliest stages and in particular through emotional association effects. Abstract: To determine how emotional information modulates subsequent traces for repeated stimuli, we combined simultaneous electro-encephalography (EEG) and magneto-encephalography (MEG) measures during long-lag incidental repetition of fearful, happy, and neutral faces. Repetition effects were modulated by facial expression in three different time windows, starting as early as 40–50 ms in both EEG and MEG, then arising at the time of the N170/M170, and finally between 280–320 ms in MEG only. The very early repetition effect, observed at 40–50 ms over occipito-temporo-parietal regions, showed a different MEG topography according to the facial expression. This differential response to fearful, happy and neutral faces suggests the existence of very early discriminative visual processing of expressive faces, possibly based on the low-level physical features typical of different emotions. The N170 and M170 face-selective components both showed repetition enhancement selective to neutral faces, with greater amplitude for emotional than neutral faces on the first but not the second presentation. These differential repetition effects may reflect valence acquisition for the neutral faces due to repetition, and suggest a combined influence of emotion- and experience-related factors on the early stage of face encoding. Finally, later repetition effects consisted in enhanced M300 (MEG) between 280 and 320 ms for fearful relative to happy and neutral faces that occurred on the first presentation, but levelled out on the second presentation. This effect may correspond to the higher arousing value of fearful stimuli that might habituate with repetition.
