Hyperpolarization has found many applications in Nuclear Magnetic Resonance (NMR) and Magnetic Resonance Imaging (MRI). However, its usage is still limited to the observation of relatively fast processes because of its short lifetimes. This issue can be circumvented by storing the hyperpolarization in a slowly relaxing singlet state. Symmetrical molecules hyperpolarized by Parahydrogen Induced Hyperpolarization (PHIP) provide straightforward access to hyperpolarized singlet states because the initial parahydrogen singlet state is preserved at almost any magnetic field strength. In these systems, which show a remarkably long (1)H singlet state lifetime of several minutes, the conversion of the NMR silent singlet state to observable magnetization is feasible due to the existence of singlet-triplet level anti-crossings. Here, we demonstrate that scaling the chemical shift Hamiltonian by rf irradiation is sufficient to transform the singlet into an observable triplet state. Moreover, because the application of one long rf pulse is only partially converting the singlet state, we developed a multiconversion sequence consisting of a train of long rf pulses resulting in successive singlet to triplet conversions. This sequence is used to measure the singlet state relaxation time in a simple way at two different magnetic fields. We show that this approach is valid for almost any magnetic field strength and can be performed even in the less homogeneous field of an MRI scanner, allowing for new applications of hyperpolarized NMR and MRI.

The application of parahydrogen for the generation of hyperpolarization has increased continuously during the last years. When the chemical reaction is carried out at the same field as the NMR experiment (PASADENA protocol) an antiphase signal is obtained, with a separation of the resonance lines of a few Hz. This imposes a stringent limit to the homogeneity of the magnetic field in order to avoid signal cancellation. In this work we detect the signal arising from hyperpolarized Hexene by means of a CPMG pulse train. After Fourier transformation the obtained J-spectra not only presents an enhanced spectral resolution but also avoids partial peak cancellation., (Copyright © 2013 Elsevier Inc. Hyperpolarization by means of ParaHydrogen Induced Polarization (PHIP) has found increasing applications since its discovery. However, in the last decade only a few experiments have been reported describing the hydrogenation of symmetric molecules. A general AA'BB' system is studied here. Calculations of the spin dynamics with the density matrix formalism support the experimental findings, providing profound understanding of the experiments in Cs-symmetric molecules. Level anti-crossings between states related to the triplet and the singlet state of one pair of the protons are identified as being responsible for hyperpolarization transfer in a PHIP experiment, when the former p-H(2) protons occupy the sites AA'. The hydrogenation of acetylene dicarboxylic acid dimethylester with parahydrogen is used to illustrate the case. The theoretical treatment applied to this particular reaction explains the signal enhancements in both groups of protons in the spectrum when the sample is placed in the proper magnetic field strength, including the phase inversion of the signal of the methyl group. Two polymorphic forms of a novel pharmaceutical compound, ciprofloxacin-saccharinate (CIP-SAC), are analyzed using one dimensional (1D) and two dimensional (2D) (1)H nuclear magnetic resonance (NMR) at fast magic angle spinning (MAS). Additionally (15)N spectroscopy and (1)H-(13)C correlation experiments were performed to complement our conclusions. The 1D (1)H NMR spectra of CIP and complexes reveal valuable information about the ionic bonding between ciprofloxacin and saccharine. Additionally, these spectra allow us to perform a clear characterization of each solid form, giving the number of molecules per unit cell in one of the polymorphs. From 2D (1)H-(1)H spectra obtained through double quantum correlations we can arrive at important conclusions about the hydrogen bonding, conformation, and intra and inter-molecular interactions present in these compounds. Comparing and contrasting the (1)H-(1)H correlation data obtained for both polymorphic forms and taking into account the single crystal structure data existing for the solid form CIP-SAC (II) was possible to extract some conclusions on the polymorph CIP-SAC (I) where no single crystal information is available. (1)H MAS NMR is shown to be an important tool in the field of polymorphism and for the characterization of multicomponent pharmaceutical compounds.

Diffusion of atoms or molecules in presence of magnetic field gradients not only attenuates the NMR signal but also leads to distortions close to restricting boundaries. This phenomenon is most evident in imaging with laser polarized (LP) noble gases. Diffusion of gases can be manipulated, however, by admixing inert gases of different molecular weight. In this work we analyze the effect of mixing LP-(3)He with SF(6) on the image quality of a phantom consisting of an arrangement of capillaries with different diameters. Admixing buffer gases of higher molecular weight changes the contrast and offers a means to record images with high spatial and time resolution. Additionally we demonstrate how distortions due to edge enhancement can be reduced even for long timed MRI-sequences.

We report on the combined use of fluorescence correlation spectroscopy (FCS) and 1H and 13C NMR spectroscopy to detect the size and type of peptide secondary structures in a series of poly-Z-L-lysine functionalized polyphenylene dendrimers bearing the fluorescent perylenediimide core in solution. In dilute solution, the size of the molecule as detected from FCS and 1H NMR diffusion measurements matches nicely. We show that FCS is a sensitive probe of the core size as well as of the change in the peptide secondary structure. However, FCS is less sensitive to functionality. A change in the peptide secondary conformation from beta-sheets to alpha-helices detected by 13C NMR spectroscopy gives rise to a steep increase in the hydrodynamic radii for number of residues n > or = 16. Nevertheless, helices are objects of low persistence.

The dependence of the individual mean square displacement of rare gases in binary mixtures is studied by a combined experimental and theoretical approach. We show that the diffusion constant can be varied in a considerable range by changing the molar fractions of the mixtures. On the experimental side, NMR diffusion measurements are done on hyperpolarized 3He and 129Xe, mixed with several inert buffer gases, in the presence of a magnetic field gradient. The results are compared to diffusion coefficients obtained from atomistic molecular dynamics simulations based on Lennard-Jones type potentials of the corresponding gas mixtures, and to appropriate analytical expressions, yielding very good mutual agreement. This study is the first quantitative validation of the effects of the mutual interactions between gas particles on the individual diffusion properties. It is shown that the dependency of gas phase diffusion properties on the local chemical environment may not be neglected, e.g. in diffusion-controlled chemical reactions.

With the use of hyperpolarized gases, a great number of experiments have been carried out in order to improve the diagnostics of the lung, both from a structural and a functional point of view. 3He is best suited for structural studies, whereas 129Xe gives more detailed information about the functionality of the lung because it enters the bloodstream. In this work, we propose the use of a gas mixture to perform consecutive analysis of lung structure and functionality upon the delivery of a single bolus of gas. We show images of a helium-xenon gas mixture in the presence of a small amount of liquid toluene in order to demonstrate how both nuclei can be detected independently, extracting the spectroscopic information provided by the 129Xe spectra and obtaining an image with high sensitivity for 3He. A second experiment performed on a dissected mouse lung was used to demonstrate how the mixture of gases can enhance sensitivity in the larger airways of the lung.

In the past few years, solid-state 1H NMR spectroscopy under fast magic-angle spinning (MAS) has developed into a versatile tool for elucidating structure and dynamics. Dipolar multiple-quantum (MQ), in particular double-quantum (DQ), MAS spectroscopy has been applied to a variety of materials and provided unique insight, e.g., into the structure of hydrogen-bonded systems. This review intends to present solid-state 1H DQ and MQ MAS spectroscopy in a systematic fashion with a particular emphasis on methodological aspects, followed by an overview of applications., (Copyright 2001 Academic Press.)

We present a quantum chemical ab initio study which demonstrates a new combined experimental and theoretical approach, whereby a comparison of calculated and experimental (1)H NMR chemical shifts allows the elucidation of structural arrangements in solid-state molecular ensembles, taking advantage of the marked sensitivity of the (1)H chemical shift to intermolecular interactions. Recently, Brown et al. have shown that, under fast magic-angle spinning (MAS) at 35 kHz, the resolution in a (1)H NMR spectrum of the solid phase of an alkyl-substituted hexabenzocoronene (HBC) derivative is sufficient to observe the hitherto unexpected resolution of three distinct aromatic resonances ( J. Am. Chem. Soc. 1999, 121, 6712). Exploiting the additional information about proton proximities provided by (1)H double-quantum (DQ) MAS NMR spectroscopy, it was shown that the results are qualitatively consistent with the aromatic cores packing in a manner similar to that in unsubstituted HBC. Using the HBC-C(12) molecule as an example, we show here that the new combined experimental and theoretical approach allows the observed (1)H chemical shifts to be related in a quantitative manner to the intermolecular structure. In the quantum chemical calculations, a series of model systems of stacked HBC oligomers are used. On account of the marked dependence of the (1)H chemical shift to ring currents arising from nearby aromatic rings, the calculated (1)H chemical shifts are found to be very sensitive to the stacking arrangement of the HBC molecules. Moreover, the ring current effect is found to be particularly long range, with a considerable influence of the second neighbor, at a distance of 700 pm, being observed.

Recent multiple-quantum MAS NMR experiments have shown that a change in the rotor phase (and, hence, in the Hamiltonian) between the excitation and reconversion periods can lead to informative spinning-sideband patterns. However, such "rotor encoding" is not limited to multiple-quantum experiments. Here it is shown that longitudinal magnetization can also be rotor-encoded. Both homonuclear and heteronuclear rotor encoding of longitudinal magnetization (RELM) experiments are performed on dipolar-coupled spin-1/2 systems, and the corresponding sideband patterns in the indirect dimension are analyzed. In both cases, only even-order sidebands are produced, and their intensity distribution depends on the durations of the recoupling periods. In heteronuclear experiments using REDOR-type recoupling, purely dipolar sideband patterns that are entirely free of effects due to the chemical-shielding anisotropy can be generated. Advantages and disadvantages of the heteronuclear RELM experiment are discussed in the context of other methods used to measure heteronuclear dipolar couplings., (Copyright 2000 Academic Press.)

A multidimensional static solid-state NMR experiment is described that combines 13C exchange sequences with 1H spin diffusion. It realizes a spatial correlation of different reorientation rates. By means of this experiment the length scale of dynamic heterogeneities can be measured directly. The pulse sequence and phase cycle as well as the experimental setup procedure and data analysis are described in detail. It complements the previous letter on this subject where a brief report of the main results were presented (U. Tracht et al., 1998, Phys. Rev. Lett. 81, 2727). A new approach for high-resolution solid-state heteronuclear multiple-quantum MAS NMR spectroscopy of dipolar-coupled spin-12 nuclei is introduced. The method is a heteronuclear chemical shift correlation technique of abundant spins, like 1H with rare spins, like 13C in natural abundance. High resolution is provided by ultra-fast MAS and high magnetic fields, high sensitivity being ensured by a direct polarization transfer from the abundant protons to 13C. In a rotor-synchronized variant, the method can be used to probe heteronuclear through-space proximities, while the heteronuclear dipolar coupling constant can quantitatively be determined by measuring multiple-quantum spinning-sideband patterns. By means of recoupling, even weak heteronuclear dipolar interactions are accessible. A new pulse sequence for high-resolution solid-state heteronuclear double-quantum MAS NMR spectroscopy of dipolar-coupled spin-12 nuclei is introduced. It is based on the five-pulse sequence known from solution-state NMR, which is here applied synchronously to both spin species. The heteronuclear double-quantum (HeDQ) spinning-sideband patterns produced by this experiment are shown to be sensitive to the heteronuclear distance, as well as the relative orientations of the chemical-shift and dipolar tensors. In particular, it is shown that the HeDQ patterns exhibit an enhanced sensitivity to the chemical shielding tensors as compared with the single-quantum spinning-sideband patterns. The detection of HeDQ patterns via the I and S spins is discussed. The isolated (13)C-(1)H spin pair in deuterated ammonium formate with (13)C in natural abundance was chosen as a model system, and the perturbing influence of dipolar couplings to surrounding protons on the (13)C-(1)H DQ coherence is discussed. The pulse sequence can also be used as a heteronuclear double-quantum filter, hence providing information about heteronuclear couplings, and thus allowing the differentiation of quaternary and CH(n) bonded carbons. The elucidation of (13)C-(1)H dipolar proximities is presented for a sample of bisphenol A polycarbonate with (13)C in natural abundance, recorded with a broadband version of the synchronized five-pulse sequence., (Copyright 1999 Academic Press.)

A new 2D solid-state CP/MAS 13C NMR exchange experiment for through-space isotropic chemical shift correlation is proposed and demonstrated. Through-space correlation is established via a second cross polarization from 13C to 1H and subsequent 1H spin diffusion. A third cross polarization results in the final 13C-13C isotropic chemical shift correlation. The 1H spin diffusion time is a variable parameter allowing different mean square magnetization displacements to be probed. Experimental results on mixtures of differently 13C-labeled alanine and polyethylene indicate that this site-selective 2D technique can be used to characterize domain sizes and proximities over a wide range of length scales (1-200 nm) in solids such as polymers or biological materials., (Copyright 1998 Academic Press.)

1H double-quantum (DQ) solid-state NMR spectroscopy under fast magic-angle spinning (MAS) is introduced as a new spectroscopic tool for the investigation of the structure and local chain dynamics of elastomers. Dipolar connectivities between the protons of the various functional groups can be directly established from the highly resolved DQ solid-state NMR spectra as is shown for a series of cross-linked poly(styrene-co-butadiene). More quantitatively, residual dipolar couplings within and between the functional groups are evaluated selectively from the build-up curves of the double-quantum signals in the limit of the spin-pair approximation. In particular, the CH-CH and the CH2-CH couplings of butadiene, which both act predominantly along the chain-segment direction, have been measured relative to the CH2 coupling. The total build-up intensity is correlated with the cross-link density.

Two techniques for resolution and sensitivity enhancement are introduced in multiple-quantum (MQ) MAS spectroscopy of rigid solids. The first makes use of ultrafast MAS with spinning frequencies of up to 35 kHz, while the second combines MAS at moderately fast spinning frequencies of about 13 kHz with multiple-pulse (MP) dipolar decoupling. For the latter approach, a semiwindowless WHH-4 sequence is applied during the MQ evolution period (MQ dimension) and/or detection period (single-quantum dimension). In the MQ dimension, the MP sequence has to be supplemented by two bracketing pulses in order to preserve the order and the intensities of the evolving MQ coherences. Double-quantum 1H NMR spectra of l-alanine recorded using both decoupling techniques are shown and compared to each other. Combined rotation and multiple-pulse experiments (CRAMPS) are reported that are performed under the conditions of fast magic-angle spinning. Quasi-static conditions, as are required for CRAMPS experiments, can be fulfilled approximately also for fast sample spinning conditions when windowless or semiwindowless sequences are applied. In order to allow direct detection for these cases also, appropriately timed detection windows are introduced without loss of resolution. In contrast to conventional CRAMPS experiments, high-speed MAS was found to play an important role also in the averaging of residual dipolar contributions. The resolution achieved in these first experiments is comparable to that of conventional CRAMPS experiments and the demands with respect to spectrometer hardware and tuning are much lower.

A comprehensive account of spatially resolved solid-state MAS NMR of 13C is given. A device generating field gradients rotating synchronously with the magic angle spinner is described. Spatial resolution and sensitivity are compared for phase and frequency encoding of spatial information. The suppression of spinning sidebands is demonstrated for both cases. Prior knowledge about the involved materials can be used for the reduction of data from spatially resolved spectra to map chemical structure. Indirect detection via 13C NMR gives access to the information about mobility from proton-wideline spectra. Two-dimensional solid-state spectroscopy with spatial resolution is demonstrated for a rotor synchronized MAS experiment which resolves molecular order as a function of space. By comparison of different experiments the factors affecting the spatial resolution are investigated.

Different procedures based on parameters of the wideline NMR absorption spectrum are presented to obtain localized molecular mobility contrast for imaging of solid polymers. For this purpose a 1H-NMR imaging technique with magic sandwich echoes is used for acquiring localized wideline spectra. With samples composed of polystyrene and high impact strength polystyrene, and polycarbonate and low density polyethylene a spatial difference in NMR absorption spectrum lineshape and linewidth is displayed. Furthermore, the spatial distribution of rigid and mobile domains in a heterogeneous polymer can be derived from the NMR spectral components. It is demonstrated that a van Vleck moment analysis can be performed from spatially resolved magic echo decays. The second (M2) and fourth (M4) moments of the rigid components show considerable variation with the spatial composition of the investigated samples.

The size of micro-separated domains of polyether (PEO), -[CH2CH2O]n-, and polycarbonate (PC), -[(C6H4)-C(CH3)2-(C6H4)-OCO2]m-, in the dialysis membrane 'Gambrane' have been determined using an advanced solid-state NMR technique which exploits differences in 1H spin diffusion. The characteristic diameter of a PEO domain is 4.8 +/- 1.4 nm and that of PC is 5.2 +/- 1.4 nm with a mixed phase region of 0.8 +/- 0.5 nm.

Solid state imaging by 13C MAS imaging is described. The spinning sidebands occurring at moderate spinning speeds, which disturb the images, can be suppressed by TOSS. For rigid solids the spatial resolution that can be achieved in this way is better than that of 1H images at the same spinning speed. Spatially resolved spectra with or without spinning sidebands can likewise be recorded providing information about the isotropic and the anisotropic chemical shifts which can be exploited for the study of structure, order and dynamics. The techniques are demonstrated on a phantom made with 13C-labelled glycine.
