Production of metal debris from implant wear and corrosion processes is now a well understood occurrence following hip arthroplasty. Evidence has shown that metal ions can enter the bloodstream and travel to distant organs including the brain, and in extreme cases, can induce sensorial and neurological diseases. Our objective was tosimultaneously analyze brain anatomy and physiology in patients with long-term and well-functioning implants. Included were subjects who had received total hip or hip resurfacing arthroplastywith an implantation time of a minimum of 7 years (n = 28) and age- and sex-matched controls (n = 32). Blood samples were obtained to measure ion concentrations of cobalt and chromium, and the Montreal Cognitive Assessment was performed. 3T MRI brain scans were completed with an MPRAGE sequence for ROI segmentation and multiecho gradient echo sequences to generate QSM and R2* maps. Mean QSM and R2* values were recorded for five deep brain and four middle and cortical brain structures on both hemispheres: pallidum, putamen, caudate, amygdala, hippocampus, anterior cingulate, inferior temporal, and cerebellum. No differences in QSM or R2* or cognition scores were found between both groups (p > 0.6654). No correlation was found between susceptibility and blood ion levels for cobalt or chromium in any region of the brain. Direct imaging of neuronal activity (DIANA) by functional magnetic resonance imaging (fMRI) could be a revolutionary approach for advancing systems neuroscience research. To independently replicate this observation, we performed fMRI experiments in anesthetized mice. The blood oxygenation level-dependent (BOLD) response to whisker stimulation was reliably detected in the primary barrel cortex before and after DIANA experiments; however, no DIANA-like fMRI peak was observed in individual animals' data with the 50 to 300 trials. Extensively averaged data involving 1050 trials in six mice showed a flat baseline and no detectable neuronal activity-like fMRI peak. However, spurious, nonreplicable peaks were found when using a small number of trials, and artifactual peaks were detected when some outlier-like trials were excluded. Further, no detectable DIANA peak was observed in the BOLD-responding thalamus from the selected trials with the neuronal activity-like reference function in the barrel cortex. Thus, we were unable to replicate the previously reported results without data preselection.

Background: The small common marmoset (Callithrix jacchus) is an ideal nonhuman primate for awake fMRI in ultra-high field small animal MRI scanners. However, it can often be challenging in task-based fMRI experiments to provide a robust stimulus within the MRI environment while using hardware (an RF coil and restraint system) that is compatible with awake imaging., New Method: Here we present an RF coil and restraint system that permits unimpeded access to an awake marmoset's head subsequent to immobilization, thereby permitting the setup of peripheral devices and stimuli proximal to the head., Results: As an example application, an fMRI experiment probing whole-brain activation in response to marmoset vocalizations was conducted-this paradigm showed significant bilateral activation in the inferior colliculus, medial lateral geniculate nucleus, and auditory cortex., Comparison With Existing Method(s): The coil performance was evaluated and compared to a previously published restraint system with integrated RF coil. The image and temporal SNR were improved by up to 58 % and 27 %, respectively, in the peripheral cortex and by 30 % and 3 % in the centre of the brain. Purpose: To develop an RF coil with an integrated commercial field camera for ultrahigh field (7T) neuroimaging. The RF coil would operate within a head-only gradient coil and be subject to the corresponding design constraints. The RF coil can thereafter be used for subject-specific correction of k-space trajectories-notably in gradient-sensitive sequences such as single-shot spiral imaging., Methods: The transmit and receive performance was evaluated before and after the integration of field probes, whereas field probes were evaluated when in an optimal configuration external to the coil and after their integration. Diffusion-weighted EPI and single-shot spiral acquisitions were employed to evaluate the efficacy of correcting higher order field perturbations and the consequent effect on image quality., Results: Field probes had a negligible effect on RF-coil performance, including the transmit efficiency, transmit uniformity, and mean SNR over the brain. Modest reductions in field-probe signal lifetimes were observed, caused primarily by nonidealities in the gradient and shim fields of the head-only gradient coil at the probe positions. Frontoparietal networks contribute to complex cognitive functions in humans and macaques, such as working memory, attention, task-switching, response suppression, grasping, reaching, and eye movement control. However, there has been no comprehensive examination of the functional organization of frontoparietal networks using functional magnetic resonance imaging in the New World common marmoset monkey (Callithrix jacchus), which is now widely recognized as a powerful nonhuman primate experimental animal. In this study, we employed hierarchical clustering of interareal blood oxygen level-dependent signals to investigate the hypothesis that the organization of the frontoparietal cortex in the marmoset follows the organizational principles of the macaque frontoparietal system. We found that the posterior part of the lateral frontal cortex (premotor regions) was functionally connected to the anterior parietal areas, while more anterior frontal regions (frontal eye field [FEF]) were connected to more posterior parietal areas (the region around the lateral intraparietal area [LIP]). These overarching patterns of interareal organization are consistent with a recent macaque study. Social cognition is a dynamic process that requires the perception and integration of a complex set of idiosyncratic features between interacting conspecifics. Here we present a method for simultaneously measuring the whole-brain activation of two socially interacting marmoset monkeys using functional magnetic resonance imaging. MRI hardware (a radiofrequency coil and peripheral devices) and image-processing pipelines were developed to assess brain responses to socialization, both on an intra-brain and inter-brain level. Notably, the brain activation of a marmoset when viewing a second marmoset in-person versus when viewing a pre-recorded video of the same marmoset-i.e., when either capable or incapable of socially interacting with a visible conspecific-demonstrates increased activation in the face-patch network. The common marmoset has enormous promise as a nonhuman primate model of human brain functions. While resting-state functional MRI (fMRI) has provided evidence for a similar organization of marmoset and human cortices, the technique cannot be used to map the functional correspondences of brain regions between species. This limitation can be overcome by movie-driven fMRI (md-fMRI), which has become a popular tool for noninvasively mapping the neural patterns generated by rich and naturalistic stimulation. Here, we used md-fMRI in marmosets and humans to identify whole-brain functional correspondences between the two primate species. In particular, we describe functional correlates for the well-known human face, body, and scene patches in marmosets. We find that these networks have a similar organization in both species, suggesting a largely conserved organization of higher-order visual areas between New World marmoset monkeys and humans. However, while face patches in humans and marmosets were activated by marmoset faces, only human face patches responded to the faces of other animals. Together, the results demonstrate that higher-order visual processing might be a conserved feature between humans and New World marmoset monkeys but that small, potentially important functional differences exist., Competing Interests: The authors declare no competing interest.

Magnetic resonance imaging radio frequency arrays are composed of multiple receive coils that have their signals combined to form an image. Combination requires an estimate of the radio frequency coil sensitivities to align signal phases and prevent destructive interference. At lower fields this can be accomplished using a uniform physical reference coil. However, at higher fields, uniform volume coils are lacking and, when available, suffer from regions of low receive sensitivity that result in poor sensitivity estimation and combination. Several approaches exist that do not require a physical reference coil but require manual intervention, specific prescans, or must be completed post-acquisition. This makes these methods impractical for large multi-volume datasets such as those collected for novel types of functional MRI or quantitative susceptibility mapping, where magnitude and phase are important. This pilot study proposes a fitted SVD method which utilizes existing combination methods to create a phase sensitive combination method targeted at large multi-volume datasets. This method uses any multi-image prescan to calculate the relative receive sensitivities using voxel-wise singular value decomposition. These relative sensitivities are fitted to the solid harmonics using an iterative least squares fitting algorithm. Fits of the relative sensitivities are used to align the phases of the receive coils and improve combination in subsequent acquisitions during the imaging session. This method is compared against existing approaches in the human brain at 7 Tesla by examining the combined data for the presence of singularities and changes in phase signal-to-noise ratio. Two additional applications of the method are also explored, using the fitted SVD method in an asymmetrical coil and in a case with subject motion. The fitted SVD method produces singularity-free images and recovers between 95-100% of the phase signal-to-noise ratio depending on the prescan data resolution. Using solid harmonic fitting to interpolate singular value decomposition derived receive sensitivities from existing prescans allows the fitted SVD method to be used on all acquisitions within a session without increasing exam duration. Recent methodological advances in MRI have enabled substantial growth in neuroimaging studies of non-human primates (NHPs), while open data-sharing through the PRIME-DE initiative has increased the availability of NHP MRI data and the need for robust multi-subject multi-center analyses. Streamlined acquisition and analysis protocols would accelerate and improve these efforts. However, consensus on minimal standards for data acquisition protocols and analysis pipelines for NHP imaging remains to be established, particularly for multi-center studies. Here, we draw parallels between NHP and human neuroimaging and provide minimal guidelines for harmonizing and standardizing data acquisition. We advocate robust translation of widely used open-access toolkits that are well established for analyzing human data. We also encourage the use of validated, automated pre-processing tools for analyzing NHP data sets. These guidelines aim to refine methodological and analytical strategies for small and large-scale NHP neuroimaging data. This will improve reproducibility of results, and accelerate the convergence between NHP and human neuroimaging strategies which will ultimately benefit fundamental and translational brain science., (Copyright © 2021. A crucial component of social cognition is to observe and understand the social interactions of other individuals. A promising nonhuman primate model for investigating the neural basis of social interaction observation is the common marmoset ( Callithrix jacchus ), a small New World primate that shares a rich social repertoire with humans. Here, we used functional magnetic resonance imaging acquired at 9.4 T to map the brain areas activated by social interaction observation in awake marmosets. We discovered a network of subcortical and cortical areas, predominately in the anterior lateral frontal and medial frontal cortex, that was specifically activated by social interaction observation. This network resembled that recently identified in Old World macaque monkeys. High-resolution functional MRI studies have become a powerful tool to non-invasively probe the sub-millimeter functional organization of the human cortex. Advances in MR hardware, imaging techniques and sophisticated post-processing methods have allowed high resolution fMRI to be used in both the clinical and academic neurosciences. However, consensus within the community regarding the use of gradient echo (GE) or spin echo (SE) based acquisition remains largely divided. On one hand, GE provides a high temporal signal-to-noise ratio (tSNR) technique sensitive to both the macro- and micro-vascular signal while SE based methods are more specific to microvasculature but suffer from lower tSNR and specific absorption rate limitations, especially at high field and with short repetition times. Fortunately, the phase of the GE-EPI signal is sensitive to vessel size and this provides a potential avenue to reduce the macrovascular weighting of the signal (phase regression, Menon 2002). In order to determine the efficacy of this technique at high-resolution, phase regression was applied to GE-EPI timeseries and compared to SE-EPI to determine if GE-EPI's specificity to the microvascular compartment improved. To do this, functional data was collected from seven subjects on a neuro-optimized 7 T system at 800 μm isotropic resolution with both GE-EPI and SE-EPI while observing an 8 Hz contrast reversing checkerboard. Phase data from the GE-EPI was used to create a microvasculature-weighted time series (GE-EPI-PR). Anatomical imaging (MP2RAGE) was also collected to allow for surface segmentation so that the functional results could be projected onto a surface. A multi-echo gradient echo sequence was collected and used to identify venous vasculature. The GE-EPI-PR surface activation maps showed a high qualitative similarity with SE-EPI and also produced laminar activity profiles similar to SE-EPI. When the GE-EPI and GE-EPI-PR distributions were compared to SE-EPI it was shown that GE-EPI-PR had similar distribution characteristics to SE-EPI (p < 0.05) across the top 60% of cortex. Furthermore, it was shown that GE-EPI-PR has a higher contrast-to-noise ratio (0.5 ± 0.2, mean ± std. dev. across layers) than SE-EPI (0.27 ± 0.07) demonstrating the technique has higher sensitivity than SE-EPI. In EEG data acquired in the presence of fMRI, gradient-related spike artifacts contaminate the signal following the common preprocessing step of average artifact subtraction. Spike artifacts compromise EEG data quality since they overlap with the EEG signal in frequency, thereby confounding frequency-based inferences on activity. As well, spike artifacts can inflate or deflate correlations among time series, thereby confounding inferences on functional connectivity. We present Schrödinger filtering, which uses the Schrödinger equation to decompose the spike-containing input. The basis functions of the decomposition are localized and pulse-shaped, and selectively capture the various input peaks, with the spike components clustered at the beginning of the spectrum. Schrödinger filtering automatically subtracts the spike components from the data. On real and simulated data, we show that Schrödinger filtering (1) simultaneously accomplishes high spike removal and high signal preservation without affecting evoked activity, and (2) reduces spurious pairwise correlations in spontaneous activity. In these regards, Schrödinger filtering was significantly better than three other despiking techniques: median filtering, amplitude thresholding, and wavelet denoising. Most neuroanatomical studies are based on T 1 -weighted MR images, whose intensity profiles are not solely determined by the tissue's longitudinal relaxation times (T 1 ), but also affected by varying non-T 1 contributions, hampering data reproducibility. In contrast, quantitative imaging using the MP2RAGE sequence, for example, allows direct characterization of the brain based on the tissue property of interest. Combined with 7 Tesla (7T) MRI, this offers unique opportunities to obtain robust high-resolution brain data characterized by a high reproducibility, sensitivity and specificity. However, specific MP2RAGE parameter choices - e.g., to emphasize intracortical myelin-dependent contrast variations - can substantially impact image quality and cortical analyses through remnants of B 1 + -related intensity variations, as illustrated in our previous work. To follow up on this: we (1) validate this protocol effect using a dataset acquired with a particularly B 1 + insensitive set of MP2RAGE parameters combined with parallel transmission excitation; and (2) extend our analyses to evaluate the effects on hippocampal morphometry. The latter remained unexplored initially, but can provide important insights related to generalizability and reproducibility of neurodegenerative research using 7T MRI. We confirm that B 1 + inhomogeneities have a considerably variable effect on cortical T 1 estimates, as well as on hippocampal morphometry depending on the MP2RAGE setup. While T 1 differed substantially across datasets initially, we show the inter-site T 1 comparability improves after correcting for the spatially varying B 1 + field using a separately acquired Sa2RAGE B 1 + map. Finally, removal of B 1 + residuals affects hippocampal volumetry and boundary definitions, particularly near structures characterized by strong intensity changes (e.g. cerebral spinal fluid). Taken together, we show that the choice of MP2RAGE parameters can impact T 1 comparability across sites and present evidence that hippocampal segmentation results are modulated by B 1 + inhomogeneities. The zona incerta (ZI) is a small gray matter region of the deep brain first identified in the 19th century, yet direct in vivo visualization and characterization has remained elusive. Noninvasive detection of the ZI and surrounding region could be critical to further our understanding of this widely connected but poorly understood deep brain region and could contribute to the development and optimization of neuromodulatory therapies. We demonstrate that high resolution (submillimetric) longitudinal (T1) relaxometry measurements at high magnetic field strength (7 T) can be used to delineate the ZI from surrounding white matter structures, specifically the fasciculus cerebellothalamicus, fields of Forel (fasciculus lenticularis, fasciculus thalamicus, and field H), and medial lemniscus. Using this approach, we successfully derived in vivo estimates of the size, shape, location, and tissue characteristics of substructures in the ZI region, confirming observations only previously possible through histological evaluation that this region is not just a space between structures but contains distinct morphological entities that should be considered separately. The magnetization-prepared two-rapid-gradient-echo (MP2RAGE) sequence is used for structural T 1 -weighted imaging and T 1 mapping of the human brain. In this sequence, adiabatic inversion RF pulses are commonly used, which require the B 1 + magnitude to be above a certain threshold. Achieving this threshold in the whole brain may not be possible at ultra-high fields because of the short RF wavelength. This results in low-inversion regions especially in the inferior brain (eg cerebellum and temporal lobes), which is reflected as regions of bright signal in MP2RAGE images. This study aims at eliminating the low-inversion-efficiency induced artifacts in MP2RAGE images at 7 T. The proposed technique takes advantage of parallel RF transmission systems by splitting the brain into two overlapping slabs and calculating the complex weights of transmit channels (ie RF shims) on these slabs for excitation and inversion independently. RF shims were calculated using fast methods implemented in the standard workflow. The excitation RF pulse was designed to obtain slabs with flat plateaus and sharp edges. These slabs were joined into a single volume during the online image reconstruction. The two-slab strategy naturally results in a signal-to-noise ratio loss; however, it allowed the use of independent shims to make the B 1 + field exceed the adiabatic threshold in the inferior brain, eliminating regions of low inversion efficiency. Accordingly, the normalized root-mean-square errors in the inversion were reduced to below 2%. The two-slab strategy was found to outperform subject-specific k T -point inversion RF pulses in terms of inversion error. An object that is looming toward a subject or receding away contains important information for determining if this object is dangerous, beneficial or harmless. This information (motion, direction, identity, time-to-collision, size, velocity) is analyzed by the brain in order to execute the appropriate behavioral responses depending on the context: fleeing, freezing, grasping, eating, exploring. In the current study, we performed ultra-high-field functional MRI (fMRI) at 9.4T in awake marmosets to explore the patterns of brain activation elicited by visual stimuli looming toward or receding away from the monkey. We found that looming and receding visual stimuli activated a large cortical network in frontal, parietal, temporal and occipital cortex in areas involved in the analysis of motion, shape, identity and features of the objects. Looming stimuli strongly activated a network composed of portions of the pulvinar, superior colliculus, putamen, parietal, prefrontal and temporal cortical areas. These activations suggest the existence of a network that processes visual stimuli looming toward peripersonal space to predict the consequence of these stimuli. Purpose: Arterial spin labeling (ASL) is an established noninvasive MRI technique used for cerebral blood flow measurement, which generally suffers from a low signal-to-noise ratio (SNR). The use of ultra-high fields to enhance sensitivity inevitably results in an increase in TR because of specific absorption rate (SAR) constraints, causing inadequate sampling of hemodynamic response in functional MRI, and adversely affecting concurrent measurement such as blood oxygen level dependent. To address this problem, variable-rate selective excitation (VERSE) radiofrequency (RF) pulses were used., Methods: The conventional (sinc) selective RF pulses of the Q2TIPS block in the PICORE pulsed ASL (PASL) sequence used for blood saturation were replaced by their equivalent VERSE RF waveforms. Nine healthy volunteers were scanned using the conventional and VERSE PASL sequences on a head-only 7T scanner operating in parallel transmit mode., Results: VERSE PASL sequence provides perfusion images similar to the conventional version, with comparable perfusion SNR (conventional, 3.33 ± 0.48; VERSE, 3.26 ± 0.55) and temporal SNR (conventional, 1.02 ± 0.20; VERSE, 1.05 ± 0.12) for TR = 3.5 seconds and 70 measurements. With shorter acquisition time (TR = 2.5 seconds), VERSE PASL sequence still provides similar results to those acquired using the conventional PASL sequence with longer TR = 3.5 seconds., Conclusion: The use of VERSE RF pulses in the Q2TIPS block of a PASL sequence allowed for the reduction of RF power deposition and, consequently, an increase in the temporal resolution and/or perfusion SNR., (© 2019 International Society for Magnetic Resonance in Medicine.)

Radio-frequency (RF) arrays constructed using electric dipoles have potential benefits for transmit and receive applications using the ultra-high field (UHF) MRI. This paper examines some of the implementation barriers regarding dipole RF arrays for human head imaging at 7 T. The dipole array was constructed with conformal, meandered dipoles with dimensions selected utilizing an evolutionary-based optimization routine to shape-optimize the dipole structure. Coupling matrix synthesis (CMS) was utilized to decouple the dipole array. Mean and worst-case transmission between nearest-neighbour dipoles was -17.2 and -15.5 dB, respectively (±2.4 dB). Transmit efficiencies of 24.6 nT/V for the entire brain and 26.0 nT/V across the axial slice were observed. The total and peak 10-g SAR, normalized to 1 Watt accepted input power per channel, was 0.163 and 0.601 W/kg, respectively. Maximum and mean noise correlations were -17 dB and -32 dB, respectively. The use of both CMS and a novel shape optimization routine to design a dipole array translated into sufficient transmit uniformity with a simultaneous reduction in 10-g SAR in comparison to a non-optimized dipole array of the same geometry. As a receiver, the dipole array maintained high orthogonality between elements, resulting in strong parallel imaging performance.

Marmosets are small New World primates that are posited to become an important preclinical animal model for studying intractable human brain diseases. A critical step in the development of marmosets as a viable model for human brain dysfunction is to characterize brain networks that are homologous with human network topologies. In this regard, the use of functional magnetic resonance imaging (fMRI) holds tremendous potential for functional brain mapping in marmosets. Although possible, implementation of hardware for fMRI in awake marmosets (free of the confounding effects of anesthesia) is not trivial due to the technical challenges associated with developing specialized imaging hardware. Here, we describe the design and implementation of a marmoset holder and head-fixation system with an integrated receive coil for awake marmoset fMRI. This design minimized head motion, with less than 100  μm of translation and 0.5 degrees of rotation over 15 consecutive resting state fMRI runs (at 15 min each) across 3 different marmosets. The fMRI data was of sufficient quality to reliably extract 8 resting state networks from each animal with only 60-90 min of resting state fMRI acquisition per animal. The restraint system proved to be an efficient and practical solution for securing an awake marmoset and positioning a receive array within minutes, limiting stress to the animal. This design is also amenable for multimodal imaging, allowing for electrode or lens placement above the skull via the open chamber design. Background: Small-animal MRI is an important investigative tool for basic and preclinical research. High-resolution anatomical and functional studies of the brain require artifact-free images that are acquired with a highly sensitive radiofrequency (RF) coil., New Method: The animal holder plays an important role in mitigating image artifacts: motion artifacts are reduced by immobilizing the animal and geometric-distortion artifacts are reduced by accurately positioning the animal to improve static-field shimming. The RF coil, in turn, must provide high sensitivity over the whole brain and not physically interfere with the animal holder. To accomplish these tasks, the animal holder and RF coil should be designed in tandem. In this manuscript, animal holders and RF coils for mice, rats, and marmoset monkeys are described. Each animal holder includes components for anesthesia delivery and animal immobilization, as well as a compatible receive coil., Results/comparison With Existing Method(s): Animal holders were capable of accurate and reproducible positioning (for the marmoset, this was in the stereotactic plane), consequently reducing geometric distortion in echo-planar images. Ear bars were designed in conjunction with receive-coil formers, thereby maximizing the sensitive region of coils, while concurrently reducing motion to less than a pixel over EPI time series. Motion and SNR were quantified to facilitate direct comparison to existing animal holders and RF coils. The close homology of monkeys and humans has increased the prevalence of non-human-primate models in functional MRI studies of brain connectivity. To improve upon the attainable resolution in functional MRI studies, a commensurate increase in the sensitivity of the radiofrequency receiver coil is required to avoid a reduction in the statistical power of the analysis. Most receive coils are comprised of multiple loops distributed equidistantly over a surface to produce spatially independent sensitivity profiles. A larger number of smaller elements will in turn provide a higher signal-to-noise ratio (SNR) over the same field of view. As the loops become physically smaller, noise originating from the sample is reduced relative to noise originating from the coil. In this coil-noise-dominated regime, coil elements can have overlapping sensitivity profiles, yet still possess only mildly correlated noise. In this manuscript, we demonstrate that inductively decoupled, concentric coil arrays can improve temporal SNR when operating in the coil-noise-dominated regime-in contrast to what is expected for the more ubiquitous sample-noise-dominated array. A small, thin, 7-channel flexible coil is developed and operated in conjunction with an existing whole-head monkey coil. The mean and maximum noise correlation between the two arrays was 5% and 23%, respectively. When the flex coil was placed over the sensorimotor cortex, the temporal SNR improved by up to 2.3-fold in the peripheral cortex and up to 1.3-fold at a 2- to 3-cm depth within the brain. When the flex coil was placed over the frontal eye fields, resting-state maps showed substantially elevated sensitivity to correlations in the prefrontal cortex (54%), supplementary eye fields (39%), and anterior cingulate cortex (41%). Background and Purpose: Radiation necrosis remains an irreversible long-term side-effect following radiotherapy to the brain. The ability to predict areas that could ultimately develop into necrosis could lead to prevention and management of radiation necrosis., Materials and Methods: Fischer 344 rats were irradiated using two platforms (micro-CT irradiator and x-Rad 225 IGRT) with radiation up to 30 Gy for the micro-CT and 40 Gy for the xRAD-224 to half the brain. Animals were subsequently imaged using a 9.4 T MRI scanner every 2-4 weeks for up to 28 weeks using a 7-echo gradient echo sequence. The apparent transverse relaxation constant ([Formula: see text]) was calculated and retrospectively analyzed., Results: Animals irradiated with the low-dose rate micro-CT did not exhibit any symptoms or imaging changes associated with RN. Animals irradiated with the xRAD-225 exhibited imaging changes consistent with RN at week 24. Analysis of the [Formula: see text] coefficient within the lesion and hippocampus shows the potential for detection of RN up to 10 weeks prior to morphological changes., Conclusions: The ability to predict areas of RN and increases of [Formula: see text] within the hippocampus provides a method for long-term monitoring and prediction of RN.

Objective: To determine whether multiparametric MRI data can provide insight into the acute and long-lasting neuronal sequelae after a concussion in adolescent athletes., Methods: Players were recruited from Bantam hockey leagues in which body checking is first introduced (male, age 11-14 years). Clinical measures, diffusion metrics, resting-state network and region-to-region functional connectivity patterns, and magnetic resonance spectroscopy absolute metabolite concentrations were analyzed from an independent, age-matched control group of hockey players (n = 26) and longitudinally in concussed athletes within 24 to 72 hours (n = 17) and 3 months (n = 14) after a diagnosed concussion., Results: There were diffusion abnormalities within multiple white matter tracts, functional hyperconnectivity, and decreases in choline 3 months after concussion. Tract-specific spatial statistics revealed a large region along the superior longitudinal fasciculus with the largest decreases in diffusivity measures, which significantly correlated with clinical deficits. This region also spatially intersected with probabilistic tracts connecting cortical regions where we found acute functional connectivity changes. Hyperconnectivity patterns at 3 months after concussion were present only in players with relatively less severe clinical outcomes, higher choline concentrations, and diffusivity indicative of relatively less axonal disruption., Conclusions: Changes persisted well after players' clinical scores had returned to normal and they had been cleared to return to play. Ongoing white matter maturation may make adolescent athletes particularly vulnerable to brain injury, and they may require extended recovery periods. The consequences of early brain injury for ongoing brain development and risk of more serious conditions such as second impact syndrome or neural degenerative processes need to be elucidated., (Copyright © 2017 The Author(s). The present study sought to elucidate the functional contributions of sub-regions of the swallowing neural network in swallowing preparation and swallowing motor execution. Seven healthy volunteers participated in a delayed-response, go, no-go functional magnetic resonance imaging study involving four semi-randomly ordered activation tasks: (i) "prepare to swallow," (ii) "voluntary saliva swallow," (iii) "do not prepare to swallow," and (iv) "do not swallow." Results indicated that brain activation was significantly greater during swallowing preparation, than during swallowing execution, within the rostral and intermediate anterior cingulate cortex bilaterally, premotor cortex (left > right hemisphere), pericentral cortex (left > right hemisphere), and within several subcortical nuclei including the bilateral thalamus, caudate, and putamen. In contrast, activation within the bilateral insula and the left dorsolateral pericentral cortex was significantly greater in relation to swallowing execution, compared with swallowing preparation. Still other regions, including a more inferior ventrolateral pericentral area, and adjoining Brodmann area 43 bilaterally, and the supplementary motor area, were activated in relation to both swallowing preparation and execution. These findings support the view that the preparation, and subsequent execution, of swallowing are mediated by a cascading pattern of activity within the sub-regions of the bilateral swallowing neural network.

The common marmoset (Callithrix jacchus) is an increasingly popular animal model for translational neuroscience studies, during which anatomical and functional MRI can be useful investigative tools. To attain the requisite SNR for high-resolution acquisitions, the radiofrequency coil must be optimized for the marmoset; however, relatively few custom coils have been developed that maximize SNR and are compatible with accelerated acquisitions. For the study of large populations of animals, the heterogeneity in animal size reduces the effectiveness of a "one size fits all" approach to coil sizing and makes coils tailored to individual animals cost and time prohibitive. The approach taken in this study was to create an 8-channel phased-array receive coil that was adjustable to the width of the marmoset head, thereby negating the need for tailored coils while still maintaining high SNR. Two marmosets of different size were imaged on a 9.4-T small-animal scanner. Consistent SNR was achieved in the periphery of the brain between head sizes. When compared to a 15-channel, "one size fits all" receive coil, the adjustable coil achieved 57% higher SNR in the superior frontal and parietal cortices and 29% higher SNR in the centre of the brain. The mean geometry factor of the adjustable coil was less than 1.2 for a 2-fold reduction factor in the left-right and anterior-posterior directions. Geometry factors were compared to the 15-channel coil to guide future designs. Previous studies have demonstrated altered brain activity in Alzheimer's disease using task based functional MRI (fMRI), network based resting-state fMRI, and glucose metabolism from 18F fluorodeoxyglucose-PET (FDG-PET). Our goal was to define a novel indicator of neuronal activity based on a first-order textural feature of the resting state functional MRI (RS-fMRI) signal. Furthermore, we examined the association between this neuronal activity metric and glucose metabolism from 18F FDG-PET. We studied 15 normal elderly controls (NEC) and 15 probable Alzheimer disease (AD) subjects from the AD Neuroimaging Initiative. An independent component analysis was applied to the RS-fMRI, followed by template matching to identify neuronal components (NC). A regional brain activity measurement was constructed based on the variation of the RS-fMRI signal of these NC. The standardized glucose uptake values of several brain regions relative to the cerebellum (SUVR) were measured from partial volume corrected FDG-PET images. Comparing the AD and NEC groups, the mean brain activity metric was significantly lower in the accumbens, while the glucose SUVR was significantly lower in the amygdala and hippocampus. The RS-fMRI brain activity metric was positively correlated with cognitive measures and amyloid β1-42 cerebral spinal fluid levels; however, these did not remain significant following Bonferroni correction. There was a significant linear correlation between the brain activity metric and the glucose SUVR measurements. This proof of concept study demonstrates that this novel and easy to implement RS-fMRI brain activity metric can differentiate a group of healthy elderly controls from a group of people with AD.

Multi-channel radio-frequency (RF) arrays, composed of multiple resonant coils, provide significant benefits for MRI during both signal reception (receive) and excitation (transmit). Demonstration of increased signal-to-noise ratio (SNR) and acceleration factors during parallel acquisitions has lead to the development of receive arrays. Conversely, transmit arrays have demonstrated considerable potential for mitigating excitation inhomogeneity arising at ultra-high magnetic field strengths ( ≥ 7 T) , present due to wave-like interactions inside the sample. Due to geometric constraints, the design of both receive and transmit arrays requires the resonating coils to be closely spaced. Significant overlap in the near-field distributions from each coil results in coupling. Without an adequate decoupling strategy applied between individual elements in an RF array, the MRI performance of the array can be significantly degraded. This work presents a method to design decoupling networks for arbitrarily large RF arrays based on direct synthesis of a coupling matrix. Reflection coefficients are fitted to transfer polynomials with transmission coefficients simultaneously minimized through a nonlinear optimization. The method demonstrates the design of n th -order distributed filters and lumped element networks that compensate for all first-order and cross-coupling terms arising in an RF array suitable for MRI. The synthesis results are computed for 4-, 8-, and 32-channel RF arrays. Monte Carlo analyses and experimental results for two RF array constructions demonstrate the robustness of this approach.

The aim was to identify neuroimaging predictors of clinical improvements following constraint-induced movement therapy. Resting state functional magnetic resonance and diffusion tensor imaging data was acquired in 7 children with hemiplegic cerebral palsy. Clinical and magnetic resonance imaging (MRI) data were acquired at baseline and 1 month later following a 3-week constraint therapy regimen. A more negative baseline laterality index characterizing an atypical unilateral sensorimotor resting state network significantly correlated with an improvement in the Canadian Occupational Performance Measure score (r = -0.81, P = .03). A more unilateral network with decreased activity in the affected hemisphere was associated with greater improvements in clinical scores. Higher mean diffusivity in the posterior limb of the internal capsule of the affect tract correlated significantly with improvements in the Jebsen-Taylor score (r = -0.83, P = .02). Ultra-high field magnetic resonance imaging (MRI) scanners ( ≥ 7T) require radio-frequency (RF) coils to operate in the range of the electromagnetic spectrum where the effective wavelength in the tissue approaches the patient dimensions. Multi-channel transmit arrays, driven in parallel, have been developed to increase the transmit field (B1(+)) uniformity in this wavelength regime. However, the closely packed array elements interact through mutual coupling. This paper expands on the ability of a distributed planar filter (the "magnetic wall") to decouple individual elements in an entire array. A transmit RF coil suitable for neuroimaging at 7T was constructed. The transmit coil, composed of 10 individual surface coil elements, was decoupled with magnetic walls. A separate receive coil array was used for signal reception. The hardware and imaging performance of the transmit coil was validated with electromagnetic simulation, bench-top measurements, and in vivo MRI experiments. Analysis and measurements confirmed that the magnetic wall decoupling method provides high isolation between transmit channels, while minimally affecting the B1(+) field profiles. Electromagnetic simulations confirmed that the decoupling method did not correlate to local specific absorption rate (SAR) "hot spots" or increase local-to-global SAR fractions in comparison to previously reported 7T multi-channel transmit arrays employing different decoupling methods.

Multi-channel radio-frequency (RF) transmit coil arrays have been developed to mitigate many of the RF challenges associated with ultra-high field ( ≥ 7T) magnetic resonance imaging (MRI). These arrays can be used for parallel RF transmission to produce spatially tailored RF excitation over the field of view. However, the realization of such arrays remains a challenge due to significant reactive interaction between the array coils, i.e., mutual coupling. In this paper, a novel bandstop filter ("magnetic wall") is used in an MRI RF transmit array to decouple individual coils. The proposed decoupling method is inspired by periodic resonator designs commonly used in frequency selective surfaces and is used as a distributed RF filter to suppress the transmission of RF energy between coils in an array. The decoupling of the magnetic wall (MW) is analyzed in terms of equivalent circuits that include terms for both magnetic and electric coupling for an arbitrary number of MW resonant conductors. Both frequency-and time-domain full-wave simulations were performed to analyze a specific MW structure. The performance of the proposed method is experimentally validated for both first-order coupling and higher-order coupling with a three-coil 7T array setup. Analysis and measurements confirm that the rejection band of the MW can be tuned to provide high isolation in the presence of cross coupling between RF array coils.

Purpose: To evaluate the potential of quantitative susceptibility (QS) and R2* mapping as surrogate biomarkers of clinically relevant, age-adjusted demyelination and iron deposition in multiple sclerosis (MS)., Materials and Methods: All study participants gave written informed consent, and the study was approved by the institutional review board. Quantitative maps of the magnetic resonance imaging susceptibility parameters (R2* and QS) were computed for 25 patients with either clinically isolated syndrome (CIS) or relapsing-remitting MS, as well as for 15 age- and sex-matched control subjects imaged at 7 T. The candidate MR imaging biomarkers were correlated with Extended Disability Status Scale (EDSS), time since CIS diagnosis, time since MS diagnosis, and age., Results: QS maps aided identification of significant, voxel-level increases in iron deposition in subcortical gray matter (GM) of patients with MS compared with control subjects. These voxel-level increases were not observed on R2* maps. Region-of-interest analysis of mean R2* and QS in subcortical GM demonstrated that R2* (R ≥ 0.39, P < .01) and QS (R ≥ 0.44, P < .01) were strongly correlated with EDSS. In white matter (WM), the volume of total WM damage (defined by a z score of less than -2.0 criterion, indicating demyelination) on QS maps correlated significantly with EDSS (R = 0.46, P = .02). Voxelwise QS also supported a significant contribution of age to demyelination in patients with MS, suggesting that age-adjusted clinical scores may provide more robust measures of MS disease severity compared with non-age-adjusted scores., Conclusion: Using QS and R2* mapping, evidence of both significant increases in iron deposition in subcortical GM and myelin degeneration along the WM skeleton of patients with MS was identified. Both effects correlated strongly with EDSS.

When conducting auditory investigations using functional magnetic resonance imaging (fMRI), there are inherent potential confounds that need to be considered. Traditional continuous fMRI acquisition methods produce sounds >90 dB which compete with stimuli or produce neural activation masking evoked activity. Sparse scanning methods insert a period of reduced MRI-related noise, between image acquisitions, in which a stimulus can be presented without competition. In this study, we compared sparse and continuous scanning methods to identify the optimal approach to investigate acoustically evoked cortical, thalamic and midbrain activity in the cat. Using a 7 T magnet, we presented broadband noise, 10 kHz tones, or 0.5 kHz tones in a block design, interleaved with blocks in which no stimulus was presented. Continuous scanning resulted in larger clusters of activation and more peak voxels within the auditory cortex. However, no significant activation was observed within the thalamus. Also, there was no significant difference found, between continuous or sparse scanning, in activations of midbrain structures. Higher magnitude activations were identified in auditory cortex compared to the midbrain using both continuous and sparse scanning. These results indicate that continuous scanning is the preferred method for investigations of auditory cortex in the cat using fMRI. Also, choice of method for future investigations of midbrain activity should be driven by other experimental factors, such as stimulus intensity and task performance during scanning., (Copyright © 2014 Elsevier B.V. Current knowledge of sensory processing in the mammalian auditory system is mainly derived from electrophysiological studies in a variety of animal models, including monkeys, ferrets, bats, rodents, and cats. In order to draw suitable parallels between human and animal models of auditory function, it is important to establish a bridge between human functional imaging studies and animal electrophysiological studies. Functional magnetic resonance imaging (fMRI) is an established, minimally invasive method of measuring broad patterns of hemodynamic activity across different regions of the cerebral cortex. This technique is widely used to probe sensory function in the human brain, is a useful tool in linking studies of auditory processing in both humans and animals and has been successfully used to investigate auditory function in monkeys and rodents. The following protocol describes an experimental procedure for investigating auditory function in anesthetized adult cats by measuring stimulus-evoked hemodynamic changes in auditory cortex using fMRI. This method facilitates comparison of the hemodynamic responses across different models of auditory function thus leading to a better understanding of species-independent features of the mammalian auditory cortex.

The fidelity of gradient waveforms in MRI pulse sequences is essential to the acquisition of images and spectra with minimal distortion artefacts. Gradient waveforms can become nonideal when eddy currents are created in nearby conducting structures; however, the resultant magnetic fields can be characterised and compensated for by measuring the spatial and temporal field response following a gradient impulse. This can be accomplished using a grid of radiofrequency (RF) coils. The RF coils must adhere to strict performance requirements: they must achieve a high sensitivity and signal-to-noise ratio (SNR), have minimal susceptibility field gradients between the sample and surrounding material interfaces and be highly decoupled from each other. In this study, an apparatus is presented that accomplishes these tasks with a low-cost, mechanically simple solution. The coil system consists of six transmit/receive RF coils immersed in a high-molarity saline solution. The sensitivity and SNR following an excitation pulse are sufficiently high to allow accurate phase measurements during free-induction decays; the intrinsic susceptibility matching of the materials, because of the unique design of the coil system, results in sufficiently narrow spectral line widths (mean of 19 Hz), and adjacent RF coils are highly decoupled (mean S12 of -47 dB). The temporal and spatial distributions of eddy currents following a gradient pulse are measured to validate the efficacy of the design, and the resultant amplitudes and time constants required for zeroth- and first-order compensation are provided., (Copyright © 2013 John Wiley & Sons, Ltd.)

Altered tissue temperature and/or pH is a common feature in pathological conditions, where metabolic demand exceeds oxygen supply such as in tumors and following stroke. Therefore, in vivo tissue temperature and pH may become valuable biomarkers for disease detection and the monitoring of disease progression or treatment response in conditions with altered metabolic demand. In this study, pH is measured using the amide protons of a thulium (Tm(3+)) complex with a DOTAM-Glycine-Lysine (ligand: Tm(3+)-DOTAM-Gly-Lys). The pH was uniquely determined from the linewidth of the asymmetry curve of the chemical exchange saturation transfer spectrum, independent of contrast agent concentration, or temperature for a given saturation pulse. pH maps with an inter-pixel standard deviation of less than 0.1 pH units were obtained in 10 mM Tm(3+)-DOTAM-Gly-Lys solutions with pH ranging from 6.0 to 8.0 pH units at 37°C. Temperature maps were simultaneously obtained using the chemical shift of the chemical exchange saturation transfer peak. Much of what is known about the cortical organization for audition in humans draws from studies of auditory cortex in the cat. However, these data build largely on electrophysiological recordings that are both highly invasive and provide less evidence concerning macroscopic patterns of brain activation. Optical imaging, using intrinsic signals or dyes, allows visualization of surface-based activity but is also quite invasive. Functional magnetic resonance imaging (fMRI) overcomes these limitations by providing a large-scale perspective of distributed activity across the brain in a non-invasive manner. The present study used fMRI to characterize stimulus-evoked activity in auditory cortex of an anesthetized (ketamine/isoflurane) cat, focusing specifically on the blood-oxygen-level-dependent (BOLD) signal time course. Functional images were acquired for adult cats in a 7 T MRI scanner. To determine the BOLD signal time course, we presented 1s broadband noise bursts between widely spaced scan acquisitions at randomized delays (1-12 s in 1s increments) prior to each scan. Baseline trials in which no stimulus was presented were also acquired. Our results indicate that the BOLD response peaks at about 3.5s in primary auditory cortex (AI) and at about 4.5 s in non-primary areas (AII, PAF) of cat auditory cortex. The observed peak latency is within the range reported for humans and non-human primates (3-4 s). The time course of hemodynamic activity in cat auditory cortex also occurs on a comparatively shorter scale than in cat visual cortex. The results of this study will provide a foundation for future auditory fMRI studies in the cat to incorporate these hemodynamic response properties into appropriate analyses of cat auditory cortex., (Copyright © 2012 Elsevier Inc. Parallel transmission has been used to reduce the inevitable inhomogeneous radiofrequency fields produced in human high-field MRI greater than 3 T. Further improvements in the transmit homogeneity and efficiency are possible by leveraging the additional degree of freedom permitted by multislice acquisitions. Compared to simple scaling of the flip angle to compensate for B1+ falloff along the radiofrequency coil, calculation of B1+ shim solutions on a slice-by-slice basis can markedly improve homogeneity and/or reduce transmitted power and global SAR. Performance measures were acquired at 7 T with a 15-channel head-only transceive array featuring elements distributed over all three logical axes, facilitating B1+ shimming over arbitrary orientations. Compared to a circularly polarized volume mode of the same coil, shimming to maximize excitation efficiency on a slice-by-slice basis yielded improvements in mean B1+ by 12.8±2.4% and a reduction in standard deviation of B1+ of 16.3±6.8%, while reducing relative SAR by 6.2±3.1%. When shimming for greater uniformity, the mean and standard deviation of B1+ were further improved by 15.9±2.6% and 26.2±10.4%, respectively, at the expense of a 135±8% increase in global SAR. The first few years of fMRI were dominated by block design tasks that were essentially adapted from the PET literature. In 1995 the concept of acquiring single trials that took better advantage of fMRI's strengths was introduced. Single trials opened up the possibility of regressing fMRI timing extracted from hemodynamic responses with behavioral measures that were also acquired on a trial-by-trial basis. Utilizing the enhanced sensitivity of higher field MRI scanners, a number of us started to exploit this paradigm design to explore the temporal nature of processing in the human brain using fMRI of one to a few slices. I try to trace back the various events by which these studies came about and outline why the field stalled for almost a decade. From the earliest fMRI experiments, it was quickly appreciated by those working with BOLD at high field that the signal change originated from visible veins whose spatial localization was relatively coarse ("the macrovasculature"), and smaller vessels ("the microvasculature") that were not individually visible in BOLD images. It was expected that a functional brain imaging technique that was predominantly sensitive to the macrovasculature would not have the same effective resolution as one sensitive to the microvasculature. Elimination of the venous signal and enhancement of the microvascular one offered the tantalizing ability to image columnar and lamellar structures in the brain and distinguished fMRI from its predecessor techniques. Subregions of the cingulate cortex represent prominent intersections in the structural networks of the primate brain. The relevance of the cingulate to the structure and dynamics of large-scale networks ultimately requires a link to functional connectivity. Here, we map fine-grained functional connectivity across the complete extent of the macaque (Macaca fascicularis) cingulate cortex and delineate subdivisions pertaining to distinct identifiable networks. In particular, we identified 4 primary networks representing the functional spectrum of the cingulate: somatomotor, attention-orienting, executive, and limbic. The cingulate nodes of these networks originated from separable subfields along the rostral-to-caudal axis and were characterized by positive and negative correlations of spontaneous blood oxygen level-dependent activity. These findings represent a critical component for understanding how the anterior and midcingulate cortices integrate and shape information processing during task performance. The connectivity patterns also suggest future electrophysiological targets that may reveal new functional representations including those involved in conflict monitoring.

Although the frontal eye field (FEF) has been identified in macaque monkeys and humans, practical constraints related to invasiveness and task demands have limited a direct cross-species comparison of its functional connectivity. In this study, we used resting-state functional MRI data collected from both awake humans and anesthetized macaque monkeys to examine and compare the functional connectivity of the FEF. A seed region analysis revealed consistent ipsilateral functional connections of the FEF with fronto-parietal cortical areas across both species. These included the intraparietal sulcus, dorsolateral prefrontal cortex, anterior cingulate cortex, and supplementary eye fields. The analysis also revealed greater lateralization of connectivity with the FEF in both hemispheres in humans than in monkeys. Cortical surface-based transformation of connectivity maps between species further corroborated the remarkably similar organization of the FEF functional connectivity. The results support an evolutionarily preserved fronto-parietal system and provide a bridge for linking data from monkey and human studies.

The first 16-channel transceive surface-coil array that conforms to the human head and operates at 298 MHz (7 T) is described. Individual coil elements were decoupled using circumferential shields around each element that extended orthogonally from the former. This decoupling method allowed elements to be constructed with arbitrary shape, size, and location to create a three-dimensional array. Radiofrequency shimming achieved a transmit-field uniformity of 20% over the whole brain and 14% over a single axial slice. During radiofrequency transmission, coil elements couple tightly to the head and reduce the amount of power necessary to achieve a mean 90° flip angle (660-μs and 480-μs pulse lengths were required for a 1-kW hard pulse when shimming over the whole brain and a single axial slice, respectively). During reception, the close proximity of coil elements to the head increases the signal-to-noise ratio in the periphery of the brain, most notably at the superior aspect of the head. The sensitivity profile of each element is localized beneath the respective shield. When combined with the achieved isolation between elements, this results in the capacity for low geometry factors during both transmit and receive: 1.04/1.06 (mean) and 1.25/1.54 (maximum) for 3-by-3 acceleration in the axial/sagittal plane. High cortical signal-to-noise ratio and parallel imaging performance make the conformal coil ideal for the study of high temporal and/or spatial cortical architecture and function., (Copyright © 2011 Wiley Periodicals, Inc.)

A 15-channel transmit-receive (transceive) radiofrequency (RF) coil was developed to image the human brain at 7 T. A hybrid decoupling scheme was implemented that used both capacitive decoupling and the partial geometric overlapping of adjacent coil elements. The decoupling scheme allowed coil elements to be arrayed along all three Cartesian axes; this facilitated shimming of the transmit field, B₁⁺, and parallel imaging acceleration along the longitudinal direction in addition to the standard transverse directions. Each channel was independently controlled during imaging using a 16-channel console and a 16 × 1-kW RF amplifier-matrix. The mean isolation between all combinations of coil elements was 18 ± 7 dB. After B₁⁺ shimming, the standard deviation of the transmit field uniformity was 11% in an axial plane and 32% over the entire brain superior to the mid-cerebellum. Transmit uniformity was sufficient to acquire fast spin echo images of this region of the brain with a single B₁⁺ shim solution. Signal-to-noise ratio (SNR) maps showed higher SNR in the periphery vs center of the brain, and higher SNR in the occipital and temporal lobes vs the frontal lobe. Parallel imaging acceleration in a rostral-caudal oblique plane was demonstrated. Paramagnetic chemical exchange saturation transfer (PARACEST) contrast agents are under development for biological target identification by magnetic resonance imaging. Image contrast associated with PARACEST agents can be generated by radiofrequency irradiation of the chemically shifted protons bound to a PARACEST contrast agent molecule or by direct irradiation of the on-resonance bulk water protons. The observed signal change in a magnetic resonance image after the administration of a PARACEST contrast agent is due to both altered relaxation time constants and the CEST effect. Despite high sensitivity in vitro, PARACEST agents have had limited success in vivo where sensitivity is reduced by the magnetization transfer effect from endogenous macromolecules. The purpose of this study was to demonstrate the in vivo detection of a PARACEST contrast agent using the on-resonance paramagnetic chemical exchange effect (OPARACHEE) in a mouse glioblastoma multiforme tumor model and to isolate the OPARACHEE effect from the changes in relaxation induced by the PARACEST agent. Three mice with tumors were imaged on a 9.4 T MRI scanner following tail vein injection of 150 μL 50 mM Tm(3+)-DOTAM-glycine-lysine. A fast low angle shot pulse sequence with a low power radiofrequency pulse train (WALTZ-16) as the preparation pulse was used to generate OPARACHEE contrast. To study the dynamics of agent uptake, reference images (without the preparation pulse) and OPARACHEE images were acquired continuously in an alternating fashion before, during and after agent injection. Signal intensity decreased by more than 10% in tumor in the control images after agent administration. Despite these changes, a clear OPARACHEE contrast of 1-5% was also observed in brain tumors after contrast agent injection and maintained in the hour following injection. A novel method for decoupling coil elements of transmit/receive (transceive) arrays is reported. Each element of a coil array is shielded both concentrically and radially to reduce the magnetic flux linkage between neighboring coils; this substantially reduces the mutual inductance between coil elements and allows them to behave independently. A six-channel transceive coil was developed using this decoupling scheme and compared with two conventional decoupling schemes: the partial overlapping of adjacent elements and capacitive decoupling. The radiofrequency coils were designed to image the human head and were tested on a 7-T Varian scanner. The decoupling, transmit uniformity, transmit efficiency, signal-to-noise ratio, and geometry factors were compared between coils. The individually shielded coil achieved higher minimum isolation between elements (2.7-4.0 dB) and lower geometry factors (2-14%) than the overlapped and capacitively decoupled coils, while showing a reduction in transmit efficiency (2.8-5.9 dB) and signal-to-noise ratio (up to 34%). No difference was found in the power absorbed by the sample during a 90° radiofrequency pulse. The inset distance of coil elements within their shields was then reduced, resulting in significant improvement of the transmit efficiency (1.3 dB) and signal-to-noise ratio (28%). Several pulse sequences have been used to detect paramagnetic chemical exchange saturation transfer (PARACEST) contrast agents in animals to quantify the uptake over time following a bolus injection. The observed signal change is a combination of relaxation effects and PARACEST contrast. The purpose of the current study was to isolate the PARACEST effect from the changes in bulk water relaxation induced by the PARACEST agent in vivo for the fast low-angle shot pulse sequence. A fast low-angle shot-based pulse sequence was used to acquire continuous images on a 9.4-T MRI of phantoms and the kidneys of mice following PARACEST agent (Tm(3+)-DOTAM-Gly-Lys) injection. A WALTZ-16 pulse was applied before every second image to generate on-resonance paramagnetic chemical exchange effects. Signal intensity changes of up to 50% were observed in the mouse kidney in the control images (without a WALTZ-16 preparation pulse) due to altered bulk water relaxation induced by the PARACEST agent. Despite these changes, a clear on-resonance paramagnetic chemical exchange effect of 4-7% was also observed. A four-pool exchange model was used to describe image signal intensity. This study demonstrates that in vivo on-resonance paramagnetic chemical exchange effect contrast can be isolated from tissue relaxation time constant changes induced by a PARACEST agent that dominate the signal change., ((c) 2010 Wiley-Liss, Inc.)

Blood oxygenation level-dependent (BOLD) functional magnetic resonance imaging (fMRI) is currently the dominant technique for non-invasive investigation of brain functions. One of the challenges with BOLD fMRI, particularly at high fields, is compensation for the effects of spatiotemporally varying magnetic field inhomogeneities (DeltaB(0)) caused by normal subject respiration and, in some studies, movement of the subject during the scan to perform tasks related to the functional paradigm. The presence of DeltaB(0) during data acquisition distorts reconstructed images and introduces extraneous fluctuations in the fMRI time series that decrease the BOLD contrast-to-noise ratio. Optimization of the fMRI data-processing pipeline to compensate for geometric distortions is of paramount importance to ensure high quality of fMRI data. To investigate DeltaB(0) caused by subject movement, echo-planar imaging scans were collected with and without concurrent motion of a phantom arm. The phantom arm was constructed and moved by the experimenter to emulate forearm motions while subjects remained still and observed a visual stimulation paradigm. These data were then subjected to eight different combinations of preprocessing steps. The best preprocessing pipeline included navigator correction, a complex phase regressor and spatial smoothing. The synergy between navigator correction and phase regression reduced geometric distortions better than either step in isolation and preconditioned the data to make them more amenable to the benefits of spatial smoothing. The combination of these steps provided a 10% increase in t-statistics compared to only navigator correction and spatial smoothing and reduced the noise and false activations in regions where no legitimate effects would occur., (Copyright 2010 Elsevier Inc. Image contrast associated with paramagnetic chemical exchange saturation transfer agents can be generated by off-resonance irradiation of agent-bound water or amide protons or on-resonance irradiation of bulk water. Previously, a four-pool model was developed to describe an in vivo system. The model incorporated the magnetization transfer effect from macromolecules when using off-resonance irradiation. In the current study, this four-pool model is modified to describe the in vivo system when using on-resonance irradiation. The influences of pulse power, pulse duration, the chemical shift of bound water, the proton exchange rate between bulk water and bound water, and agent concentration on the on-resonance paramagnetic agent chemical exchange effects were simulated using a WALTZ-16 pulse train in the absence and presence of the macromolecule pool. The results demonstrated that while contrast increases with pulse duration in aqueous solution, there is an optimal pulse duration that maximizes on-resonance paramagnetic agent chemical exchange effects contrast in vivo. This predication was verified by experimental spectroscopic and imaging results from aqueous solution, bovine serum albumin phantoms, and a tissue phantom containing thulium-DOTAM (1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetamide)-glycine-lysine. Most of our knowledge about the functional organization of the nonhuman primate brain has come from single neuron recordings, whereas functional magnetic resonance imaging (fMRI) has rapidly become the method of choice for the study of the human brain. In some cases these two methods have resulted in conflicting models of frontal lobe function. Based on the finding that the frontal eye fields (FEF) exhibit a higher blood-oxygenation-level dependent (BOLD) activation for anti-saccades compared with pro-saccades, it has been proposed that this area is more involved in voluntary than automatic saccade generation. This model has been questioned by the finding of decreased single neuron activity in FEF for anti-compared with pro-saccades in monkeys. To reconcile these findings, we employed fMRI to compare BOLD activation between anti-saccades and pro-saccades in monkeys. FEF and a number of other cortical and subcortical areas showed an increased activation for anti-saccades. The results indicate that previous contrary findings between single neuron recordings and fMRI were due to differences between these techniques and were not related to differences between the two primate species.
