The rapid discrimination of bacteria is currently an emerging trend in the fields of food safety, medical detection, and environmental observation. Traditional methods often require lengthy culturing processes, specialized analytical equipment, and bacterial recognition receptors. In response to this need, we have developed a paper-based fluorescence sensor array platform for identifying different bacteria. The sensor array is based on three unique carbon quantum dots (CQDs) as sensing units, each modified with a different antibiotic (polymyxin B, ampicillin, and gentamicin). These antibiotic-modified CQDs can aggregate on the bacterial surface, triggering aggregation-induced fluorescence quenching. The sensor array exhibits varying fluorescent responses to different bacterial species. To achieve low-cost and portable detection, CQDs were formulated into fluorescent ink and used with an inkjet printer to manufacture paper-based sensor arrays. A smartphone was used to collect the responses generated by the bacteria and platform. Diverse machine learning algorithms were utilized to discriminate bacterial types. Our findings showcase the platform's remarkable capability to differentiate among five bacterial strains, within a detection range spanning from 1.0 × 10 3  CFU/mL to 1.0 × 10 7  CFU/mL. Its practicality is further validated through the accurate identification of blind bacterial samples. Antibiotic resistance is a global public health concern. Bacteria have evolved resistance to most antibiotics, which means that for any given bacterial infection, the bacteria may be resistant to one or several antibiotics. It has been suggested that genomic sequencing and machine learning (ML) could make resistance testing more accurate and cost-effective. Given that ML is likely to become an ever more important tool in medicine, we believe that it is important for pre-health students and others in the life sciences to learn to use ML tools. This paper provides a step-by-step tutorial to train 4 different ML models (logistic regression, random forests, extreme gradient-boosted trees, and neural networks) to predict drug resistance for Escherichia coli isolates and to evaluate their performance using different metrics and cross-validation techniques. We also guide the user in how to load and prepare the data used for the ML models. The tutorial is accessible to beginners and does not require any software to be installed as it is based on Google Colab notebooks and provides a basic understanding of the different ML models. The rapid spread of SARS-CoV-2 caused the COVID-19 pandemic and accelerated vaccine development to prevent the spread of the virus and control the disease. Given the sustained high infectivity and evolution of SARS-CoV-2, there is an ongoing interest in developing COVID-19 serology tests to monitor population-level immunity. To address this critical need, we designed a paper-based multiplexed vertical flow assay (xVFA) using five structural proteins of SARS-CoV-2, detecting IgG and IgM antibodies to monitor changes in COVID-19 immunity levels. Our platform not only tracked longitudinal immunity levels but also categorized COVID-19 immunity into three groups: protected, unprotected, and infected, based on the levels of IgG and IgM antibodies. We operated two xVFAs in parallel to detect IgG and IgM antibodies using a total of 40 μL of human serum sample in <20 min per test. After the assay, images of the paper-based sensor panel were captured using a mobile phone-based custom-designed optical reader and then processed by a neural network-based serodiagnostic algorithm. The serodiagnostic algorithm was trained with 120 measurements/tests and 30 serum samples from 7 randomly selected individuals and was blindly tested with 31 serum samples from 8 different individuals, collected before vaccination as well as after vaccination or infection, achieving an accuracy of 89.5%. The competitive performance of the xVFA, along with its portability, cost-effectiveness, and rapid operation, makes it a promising computational point-of-care (POC) serology test for monitoring COVID-19 immunity, aiding in timely decisions on the administration of booster vaccines and general public health policies to protect vulnerable populations.

A new analytical method for chronic kidney disease (CKD) detection utilizing paper spray mass spectrometry (PS-MS) combined with machine learning is presented. The analytical protocol is rapid and simple, based on metabolic profile alterations in urine. Anonymized raw urine samples were deposited (10 μL each) onto pointed PS-MS sample strips. Without waiting for the sample to dry, 75 μL of acetonitrile and high voltage were applied to the strips, using high resolution mass spectrometry measurement (15 s per sample) with polarity switching to detect a wide range of metabolites. Random forest machine learning was used to classify the resulting data. The diagnostic performance for the potential diagnosis of CKD was evaluated for accuracy, sensitivity, and specificity, achieving results >96% for the training data and >91% for validation and test data sets. Metabolites selected by the classification model as up- or down-regulated in healthy or CKD samples were tentatively identified and in agreement with previously reported literature. The potential utilization of this approach to discriminate albuminuria categories (normo, micro, and macroalbuminuria) was also demonstrated. This study indicates that PS-MS combined with machine learning has the potential to be used as a rapid and simple diagnostic tool for CKD.

Background: Accurate real-time eye tracking is crucial in oculomotor system research. While the scleral search coil system is the gold standard, its implantation procedure and bulkiness pose challenges. Camera-based systems are affected by ambient lighting and require high computational and electric power., New Method: This study presents a novel eye tracker using proximity capacitive sensors made of carbon-nanotube-paper-composite (CPC). These sensors detect femtofarad-level capacitance changes caused by primate corneal movement during horizontal and vertical eye rotations. Data processing and machine learning algorithms are evaluated to enhance the accuracy of gaze angle prediction., Results: The system performance is benchmarked against the scleral coil during smooth pursuits, saccades tracking, and fixations. Context.—: Machine learning applications in the pathology clinical domain are emerging rapidly. As decision support systems continue to mature, laboratories will increasingly need guidance to evaluate their performance in clinical practice. Currently there are no formal guidelines to assist pathology laboratories in verification and/or validation of such systems. These recommendations are being proposed for the evaluation of machine learning systems in the clinical practice of pathology., Objective.—: To propose recommendations for performance evaluation of in vitro diagnostic tests on patient samples that incorporate machine learning as part of the preanalytical, analytical, or postanalytical phases of the laboratory workflow. Topics described include considerations for machine learning model evaluation including risk assessment, predeployment requirements, data sourcing and curation, verification and validation, change control management, human-computer interaction, practitioner training, and competency evaluation., Data Sources.—: An expert panel performed a review of the literature, Clinical and Laboratory Standards Institute guidance, and laboratory and government regulatory frameworks., Conclusions.—: Review of the literature and existing documents enabled the development of proposed recommendations. Abstract Manufacturing companies focus on improving productivity, reducing costs, and aligning performance metrics with strategic objectives. In industries like paper manufacturing, minimizing equipment downtime is essential for maintaining high throughput. Leveraging the extensive data generated by these facilities offers opportunities for gaining competitive advantages through data-driven insights, revealing trends, patterns, and predicting future performance indicators like unplanned downtime length, which is essential in optimizing maintenance and minimizing potential losses. This paper explores statistical and machine learning techniques for modeling downtime length probability distributions and correlation with machine vibration measurements. We proposed a novel framework, employing advanced data-driven techniques like artificial neural networks (ANNs) to estimate parameters of probability distributions governing downtime lengths. Our approach specifically focuses on modeling parameters of these distribution, rather than directly modeling probability density function (PDF) values, as is common in other approaches. Experimental results indicate a significant performance boost, with the proposed method achieving up to 30% superior performance in modeling the distribution of downtime lengths compared to alternative methods. Moreover, this method facilitates unsupervised training, making it suitable for big data repositories of unlabelled data. The framework allows for potential expansion by incorporating additional input variables. In this study, machine vibration velocity measurements are selected for further investigation. The study underscores the potential of advanced data-driven techniques to enables companies to make better-informed decisions regarding their current maintenance practices and to direct improvement programs in industrial settings.

With the continuous increase in the number of academic researchers, the volume of scientific papers is also increasing rapidly. The challenge of identifying papers with greater potential academic impact from this large pool has received increasing attention. The citation frequency of a paper is often used as an objective indicator to gauge the academic influence of the paper. The task of citation frequency prediction based on historical citation data in previous studies can achieve high accuracy. However, it can only be executed after the paper has been published for a period. The delay is not conducive to timely discovery of papers with high citation frequency. In this paper, we propose a novel method for predicting cited potential of a paper based on the metadata and semantic information, which can predict the cited potential of academic paper instantly once it has been published. Specifically, the semantic information, such as abstract, semantic span and semantic inflection, is extracted to enhance the ability of the prediction model based on machine learning. To prove the effectiveness and rationality of cited potential prediction model, we conduct two experiments to validate the model and find the most effective combination of input information. Colorimetric paper sensors are used in various fields due to their convenience and intuitive manner. However, these sensors present low accuracy in practical use because it is difficult to distinguish color changes for a minute amount of analyte with the naked eye. Herein, we demonstrate that a machine learning (ML)-based paper sensor platform accurately determines the color changes. We fabricated a colorimetric paper sensor by adsorbing polyaniline nanoparticles (PAni-NPs), whose color changes from blue to green when the ambient pH decreases. Adding glucose oxidase (GOx) to the paper sensor enables colorimetric glucose detection. Target analytes (10 μL) were aliquoted onto the paper sensors, and their images were taken with a smartphone under the same conditions in a darkroom. The red-green-blue (RGB) data from the images were extracted and used to train and test three regression models: support vector regression (SVR), decision tree regression (DTR), and random forest regression (RFR). Of the three regression models, RFR performed the best at estimating pH levels ( R 2 = 0.957) ranging from pH 2 to 10 and glucose concentrations ( R 2 = 0.922) ranging from 0 to 10 mg mL -1 .

Caused by the tick-borne spirochete Borrelia burgdorferi , Lyme disease (LD) is the most common vector-borne infectious disease in North America and Europe. Though timely diagnosis and treatment are effective in preventing disease progression, current tests are insensitive in early stage LD, with a sensitivity of <50%. Additionally, the serological testing currently recommended by the U.S. Center for Disease Control has high costs (>$400/test) and extended sample-to-answer timelines (>24 h). To address these challenges, we created a cost-effective and rapid point-of-care (POC) test for early-stage LD that assays for antibodies specific to seven Borrelia antigens and a synthetic peptide in a paper-based multiplexed vertical flow assay (xVFA). We trained a deep-learning-based diagnostic algorithm to select an optimal subset of antigen/peptide targets and then blindly tested our xVFA using human samples ( N (+) = 42, N (-) = 54), achieving an area-under-the-curve (AUC), sensitivity, and specificity of 0.950, 90.5%, and 87.0%, respectively, outperforming previous LD POC tests. With batch-specific standardization and threshold tuning, the specificity of our blind-testing performance improved to 96.3%, with an AUC and sensitivity of 0.963 and 85.7%, respectively.

Paper spray ionization has been used as a fast sampling/ionization method for the direct mass spectrometric analysis of biological samples at ambient conditions. Here, we demonstrated that by utilizing paper spray ionization-mass spectrometry (PSI-MS) coupled with field asymmetric waveform ion mobility spectrometry (FAIMS), predictive metabolic and lipidomic profiles of routine breast core needle biopsies could be obtained effectively. By the combination of machine learning algorithms and pathological examination reports, we developed a classification model, which has an overall accuracy of 87.5% for an instantaneous differentiation between cancerous and noncancerous breast tissues utilizing metabolic and lipidomic profiles. Our results suggested that paper spray ionization-ion mobility spectrometry-mass spectrometry (PSI-IMS-MS) is a powerful approach for rapid breast cancer diagnosis based on altered metabolic and lipidomic profiles.
