Abstract: The impact of flavonoids released by phosphorus-deficient white lupin roots on inorganic P and soil microorganisms is largely unknown. We report that flavonoids isolated from white lupin roots mobilized inorganic phosphorus and decreased soil microbial respiration, citrate mineralization, and soil phosphohydrolase activities, but did not reduce the soil ATP content. The results suggest that white lupin''s release of flavonoids into the rhizosphere plays a significant role in its efficient P-acquisition strategy by solubilizing Fe-bound P and by limiting the microbial mineralization of citrate. White lupin is very often used as a model plant for root exudation studies due to its capability to release huge amounts of organic acids and flavonoids. The complex nature of these organic compounds makes not only their analytical determination difficult but also their extraction from soil samples. For these reasons simplified approaches, as hydroponic-based systems are widely used to study the root exudation. Therefore, the composition of a trap solution is crucial to limit artefacts causing over/underestimation of exudation rates and/or a biased molecular composition of the collected compounds. The present study was aimed at assessing the influence of different trap solutions and collection times on the quali- and quantitative root exudation pattern of white lupin ( Lupinus albus L.) grown under phosphorus (P) and iron (Fe) deficiency. Our results suggest that, in works aimed at studying root exudation processes, water is the most effective trap solution to collect the exudates like organic acids and flavonoids, especially in short time (e.g. 2 h). For longer times, low concentrations of Ca could be helpful to limit osmotic stress and possible passive leakage and/or diffusion. The use of bacteriostatic compounds as NaN and Micropur bias the results, due to interferences either with the metabolism or inhibition of the exudation processes, especially in the case of flavonoids such as quercetin. Also, the use of a pH buffer solution like 2-(N-morpholino)ethanesulfonic acid (MES) should be avoided for its undesired interferences with the release. [ABSTRACT FROM AUTHOR]

The low bioavailability of nutrients, especially nitrogen (N) and phosphorus (P), is one of the most limiting factors for crop production. In this study, under N- and P-free nutrient solution (-N-P), nodulating white lupin plants developed some nodules and analogous cluster root structures characterized by different morphological, physiological, and molecular responses than those observed upon single nutrient deficiency (strong acidification of external media, a better nutritional status than -N+P and +N-P plants). The multi-elemental analysis highlighted that the concentrations of nutrients in white lupin plants were mainly affected by P availability. Gene-expression analyses provided evidence of interconnections between N and P nutritional pathways that are active to promote N and P balance in plants. The root exudome was mainly characterized by N availability in nutrient solution, and, in particular, the absence of N and P in the nutrient solution triggered a high release of phenolic compounds, nucleosides monophosphate and saponines by roots. These morphological, physiological, and molecular responses result from a close interplay between N and P nutritional pathways. They contribute to the good development of nodulating white lupin plants when grown on N- and P-free media. This study provides evidence that limited N and P availability in the nutrient solution can promote white lupin-Bradyrhizobium symbiosis, which is favourable for the sustainability of legume production. White lupin (Lupinus albusL. cv. Amiga) reacts to phosphate deficiency by producing cluster roots which exude large amounts of organic acids. The detailed knowledge of the excretion physiology of the different root parts makes it a good model plant to study plant-bacteria interaction. Since the effect of the organic acid exudation by cluster roots on the rhizosphere microflora is still poorly understood, we investigated the abundance, diversity and functions of bacteria associated with the cluster roots of white lupin, with special emphasis on the influence of root proximity (comparing root, rhizosphere soil and bulk soil fractions) and cluster root growth stages, which are characterized by different excretion activities. Plants were grown for five weeks in microcosms, in the presence of low phosphate concentrations, on acidic sand inoculated with a soil suspension from a lupin field. Plate counts showed that bacterial abundance decreased at the stage where the cluster root excretes high amounts of citrate and protons.In vitrotests on isolates showed that the frequencies of auxin producers were highest in juvenile and mature cluster roots and significantly decreased in senescent cluster roots. However, no significant difference in the frequency of auxin producers was found between cluster and non cluster roots. The diversity and structure of bacterial communities were investigated by DGGE of 16S rDNA and 16S rRNA. The diversity and community structure were mostly influenced by root proximity and, to a lesser extent, by cluster root stage. The richness of bacterial communities decreased with root proximity, whereas the proportion of active populations increased. The high citrate and proton excretion occurring at the mature stage of cluster roots had a strong impact on the structure and richness of the bacterial communities, both in the root and in the rhizosphere soil. White lupins ( Lupinus albus L.) respond to phosphate deficiency by producing special root structures called cluster roots. These cluster roots secrete large amounts of carboxylates into the rhizosphere, mostly citrate and malate, which act as phosphate solubilizers and enable the plant to grow in soils with sparingly available phosphate. The success and efficiency of such a P-acquisition strategy strongly depends on the persistence and stability of the carboxylates in the soil, a parameter that is influenced to a large extent by biodegradation through rhizosphere bacteria and fungi. In this study, we show that white lupin roots use several mechanisms to reduce microbial growth. The abundance of bacteria associated with cluster roots was decreased at the mature state of the cluster roots, where a burst of organic acid excretion and a drastic pH decrease is observed. Excretion of phenolic compounds, mainly isoflavonoids, induced fungal sporulation, indicating that vegetative growth, and thus potential citrate consumption, is reduced. In addition, the activity of two antifungal cell wall-degrading enzymes, chitinase and glucanase, were highest at the stage preceding the citrate excretion. Therefore, our results suggest that white lupin has developed a complex strategy to reduce microbial degradation of the phosphate-solubilizing agents. [ABSTRACT FROM AUTHOR]
