Lipopolysaccharides (LPSs) of 11 bacterial strains from the type species of the genera Bacteroides (B. fragilis), Prevotella (Pr. melaninogenica), Porphyromonas (Po. gingivalis), Campylobacter (C. fetus subsp. fetus), and Wolinella (W. succinogenes), and from the type strains of B. distasonis, B. forsythus, B. ureolyticus, Po. levii, Po. macacae, and C. gracilis, were extracted with hot water-phenol (Westphal method). S-form LPSs, obtained from all organisms, were well resolved with tricine-sodium-dodecyl-sulphate polyacrylamide gel electrophoresis and visualized by silver staining. Lipid A was not stained. Also profiles from LPS of Escherichia coli, serotypes 0111:B4 and 055:B5, could be distinguished. While W. succinogenes showed a relatively short S-form LPS on electrophoregrams, the other bacteria, including B. fragilis, exhibited long-ladder LPSs. Po. gingivalis displayed the largest number of bands and the longest O-chain. The long O-chain of this bacterium may be important for its virulence.

Objective: We examined the influence of in vitro aging on Campylobacter rectus (C. rectus) lipopolysaccharide (LPS)-stimulated prostaglandin (PG), E2 release frOm human gingival fibroblasts (HGFs)., Materials and Methods: LPS was prepared from C. rectus ATCC33238. HGFs were established from healing gingival tissue removed from three patients (donors A, B and C), aged 10-12 years. Aging of the cells in culture was determined with increasing population doubling. The cells were cultured until confluence, then stimulated with LPS (1.0 micrograms ml-1), and the levels of PGE2 in the medium were measured after 24 h by radioimmunoassay., Results: The LPS-stimulated PGE2 production in each old cell (passage 17-20) was significantly increased to about 1.6-2.6 times than that in the corresponding young cells (passage 5-6). The gene expression of cyclooxygenase-2 mRNA in the old cells was higher than that in the young cells in response to LPS. In the absence of LPS, PGE2 production levels in both the young and old cells were very low, and also at the same level. However, there was a higher level of LPS-stimulated PGE2 production in the young cells from donor C compared to that in the old cells from donor B. The LPS-stimulated PGE2 production in each young cell from donors A and C was almost equal to that in each old cell from donors B and A, respectively., Conclusions: The results suggested that aging in HGFs may be one of the factors that take part in the stimulation of C. rectus LPS-stimulated PGE2 production in old cells.

Structural investigations were performed on the liberated oligosaccharide and the extracellular polysaccharide, variously using 1H, 13C, and 31P NMR spectroscopy, linkage analysis, and fast atom bombardment-mass spectrometry of permethylated derivatives of the glycans and their products of chemical and enzymic degradation. Previously, we reported the antigranulocytic activity of Campylobacter rectus media supernatants containing lipopolysaccharide (LPS) and a 104 kDa protein. Here, we monitored the release of protein and LPS through the growth cycle of C. rectus ATCC 33238 and identified the 104 kDa protein as the cytotoxin. LPS in media supernatants was quantitated by a KDO assay; the 104 kDa protein was detected on immunoblots with specific antibody (A104) and quantitated by amino acid analysis of membrane immobilized protein bands. C. rectus cell product release was independent of cell lysis. Over 24 h, the 104 kDa protein was released linearly while LPS was released in two plateaus; both increased in C. rectus culture supernatants 3 h after inoculation achieving maximum concentrations at 21 h of 3.1 micrograms/ml and 14.6 micrograms/ml, respectively. In 2 h, trypan blue viability assays, 37-47 micrograms of 12, 18 and 24 h supernatant protein killed 33-43% of HL-60 cells. Supernatant toxicity was heat sensitive and inhibited by A104. Sequencing the 16 N-terminal amino acids of the cytotoxin distinguished it from described C. rectus proteins. Similarities between epitopes and amino acid compositions of the Actinobacillus actinomycetemcomitans leukotoxin and C. rectus cytotoxin were observed. These data indicate that C. rectus secretes a 104 kDa cytotoxin.

Outer membranes were isolated, by sodium lauryl sulphate extraction, from the American type strain, five Australian, and four English isolates of Campylobacter hyointestinalis. On SDS-PAGE examination, the protein profiles of seven strains (including the type strain) were similar, and were dominated by two major proteins of 47 and 50 kDa. Three other isolates had unique major protein profiles. The largest of these proteins was heat-modifiable in these isolates, and in the type strain. The flagellin of three isolates screened was of similar M(r) to that of Campylobacter jejuni/Campylobacter coli. The lipopolysaccharides of C. hyointestinalis isolates were heterogeneous in structure; 5/10 isolates synthesised material of M(r) value greater than that of the low M(r) C. jejuni/C. coli lipopolysaccharide. By gel excision and re-electrophoresis, it was shown that the higher M(r) materials of one isolate were not artifactual aggregates of lower M(r) species.

Lipopolysaccharides (LPS) were extracted from cells of Wolinella recta ATCC 33238, W. curva ATCC 33224, W. succinogenes ATCC 29543 and Campylobacter sputorum ssp. sputorum A 3563 by a hot phenol-water method and purified by nuclease treatment and by repeated ultracentrifugation. Chemical compositions of the purified LPS including fatty acid and sugar composition were examined and sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) was performed. All LPS preparations contained a monosaccharide identified as L-glycero-D-mannoheptose, and another heptose isomer identified as D-glycero-D-mannoheptose was a typical constituent of the LPS from all three Wolinella species.
