Background: Oleaginous fungi can accumulate lipids by utilizing a wide range of waste substrates. They are an important source for the industrial production of omega-6 polyunsaturated fatty acids (gamma-linolenic and arachidonic acid) and have been suggested as an alternative route for biodiesel production. Initial research steps for various applications include the screening of fungi in order to find efficient fungal producers with desired fatty acid composition. Traditional cultivation methods (shake flask) and lipid analysis (extraction-gas chromatography) are not applicable for large-scale screening due to their low throughput and time-consuming analysis. Here we present a microcultivation system combined with high-throughput Fourier transform infrared (FTIR) spectroscopy for efficient screening of oleaginous fungi., Results: The microcultivation system enables highly reproducible fungal fermentations throughout 12 days of cultivation. Reproducibility was validated by FTIR and HPLC data. Analysis of FTIR spectral ester carbonyl peaks of fungal biomass offered a reliable high-throughput at-line method to monitor lipid accumulation. Partial least square regression between gas chromatography fatty acid data and corresponding FTIR spectral data was used to set up calibration models for the prediction of saturated fatty acids, monounsaturated fatty acids, polyunsaturated fatty acids, unsaturation index, total lipid content and main individual fatty acids. High coefficients of determination (R 2  = 0.86-0.96) and satisfactory residual predictive deviation of cross-validation (RPD CV  = 2.6-5.1) values demonstrated the goodness of these models., Conclusions: We have demonstrated in this study, that the presented microcultivation system combined with rapid, high-throughput FTIR spectroscopy is a suitable screening platform for oleaginous fungi. Sample preparation for FTIR measurements can be automated to further increase throughput of the system.

Isolation is the main bottleneck in the analysis of fatty acids in biological samples and foods. In the last few decades some methods described direct derivatization procedures bypassing these steps. They involve the utilization of methanolic HCL or BF3 as catalysts, but several evidences from previous works suggest these reagents are unstable, lead to the formation of artifacts and alter the distribution of specific compounds as hydroxy fatty acids or CLA. However, the main issue is that they are excellent esterification reagents but poor in transterification, being not suitable for the analysis of all lipid classes and leading to erroneous composition quantitations. The present research work is a comprehensive comparison of six general methylation protocols using base, acid or base/acid catalysts plus a proposed method in the analysis of total fatty acids in lipid standards mixtures, foodstuff and biological samples. The addition of aprotic solvents to the reaction mixture to avoid alterations was also tested. Results confirmed that procedures solely involving acid catalyst resulted in incomplete derivatizations and alteration of the fatty acid profile, partially corrected by addition of the aprotic solvent. The proposed method combining sodium methoxyde and sulfuric acid showed absence of alteration of the FAME profile and the best values for response factors (short chain fatty acids to PUFA), accuracy in the determination of total cholesterol and derivatization performance, thus showing a high reliability in the determination of the total fatty acid composition in biological samples and foods., (Copyright © 2014 Elsevier B.V. Depleting fossil fuel reserves and the effects of greenhouse gases on climate change have led to the investigation of alternative energy sources such as biofuel, including biodiesel and renewable diesel. Unfortunately, the cost of the feedstock exceeds 70% of the total production cost. The current work investigates the potential of utilizing a waste product (activated sludge biomass) from wastewater treatment, as an alternative feedstock for biodiesel production. The activated sludge biomass was pretreated with subcritical water to enhance the lipid yield before extracting the lipid using solvent extraction. The result showed that lipid yield from the activated sludge was 4.73–8.58%. After subcritical water pretreatment with varying residence time, temperature, and biomass loading, the lipid yield increased by 158.04–400%. Gas chromatography–mass spectrometry (GC–MS) analysis of the fatty acid methyl esters (FAME) from the transesterification of the lipid extract using calcined egg shell as a catalyst identified the following lipids: triglyceride, fatty acids, sphingolipid, phospholipid, glycerolipid, glycerophospholipid, steroid, and cholesterol. The predominant lipid was triglyceride, and the high percentage of unsaturated fatty acids, including arachidonic acid (23.54%), eicosadienoic acid (8.37%), and oleic acid (6.23%) suggest that the lipid extract is a potential feedstock for biodiesel production. Summary: This study focused on the preparation of lipid concentrates with high contents of bioactive molecules. Different one‐step and two‐step extractions with non‐halogenated solvents (hexane, HX; isopropanol, IP) were carried out on Patagonian squid (Doryteuthis gahi) discards. As a first step, HX extractions (one, two or three times) were carried out. The resulting substrates were extracted with IP (two, three or four times). Analysis of lipid extracts included total lipid, phospholipid (PL) and tocopherol values, and fatty acid (FA) profile. HX extractions led to remarkable α‐ and γ‐tocopherol values (ca. 1400 and 23 mg kg−1 lipids, respectively); no effect (P > 0.05) of the number of HX extractions was observed. IP extracts provided high PL values (383–501 mg kg−1 lipids); the highest average values were detected with four IP extractions after three HX extractions. Values obtained for tocopherols and PLs were higher (P < 0.05) than those obtained by employing the conventional (chloroform/methanol, 1:1) lipid extraction (ca. 496 and 11 mg kg−1 lipids and 400 g kg−1 lipids, respectively). Range values detected for eicosapentaenoic and docosahexaenoic acid values and the ω3/ω6 FA ratio were similar to those obtained by the conventional procedure. Increased dietary phosphate consumption intensifies renal phosphate burden. Several mechanisms for phosphate-induced renal tubulointerstitial fibrosis have been reported. Considering the dual nature of phosphate as both a potential renal toxin and an essential nutrient for the body, kidneys may possess inherent protective mechanisms against phosphate overload, rather than succumbing solely to injury. However, there is limited understanding of such mechanisms. To identify these mechanisms, we conducted single-cell RNA sequencing (scRNA-seq) analysis of the kidneys of control and dietary phosphate-loaded (Phos) mice at a time point when the Phos group had not yet developed tubulointerstitial fibrosis. scRNA-seq analysis identified the highest number of differentially expressed genes in the clusters belonging to proximal tubular epithelial cells (PTECs). Based on these differentially expressed genes, in silico analyses suggested that the Phos group activated peroxisome proliferator-activated receptor-α (PPAR-α) and fatty acid β-oxidation (FAO) in the PTECs. This activation was further substantiated through various experiments, including the use of an FAO activity visualization probe. Compared with wild-type mice, Ppara knockout mice exhibited exacerbated tubulointerstitial fibrosis in response to phosphate overload. Experiments conducted with cultured PTECs demonstrated that activation of the PPAR-α/FAO pathway leads to improved cellular viability under high-phosphate conditions. The Phos group mice showed a decreased serum concentration of free fatty acids, which are endogenous PPAR-α agonists. Instead, experiments using cultured PTECs revealed that phosphate directly activates the PPAR-α/FAO pathway. These findings indicate that noncanonical metabolic reprogramming via endogenous activation of the PPAR-α/FAO pathway in PTECs is essential to counteract phosphate toxicity. Activation of pregnane X receptor (PXR) by xenobiotics has been associated with metabolic diseases. This study aimed to reveal the impact of PXR activation on hepatic metabolome and explore novel mechanisms underlying PXR-mediated lipid metabolism disorder in the liver. Wild-type and PXR-deficient male C57BL/6 mice were used as in vivo models, and hepatic steatosis was induced by pregnenolone-16α-carbonitrile, a typical rodent PXR agonist. Metabolomic analysis of liver tissues showed that PXR activation led to significant changes in metabolites involved in multiple metabolic pathways previously reported, including lipid metabolism, energy homeostasis, and amino acid metabolism. Moreover, the level of hepatic all- trans retinoic acid (ATRA), the main active metabolite of vitamin A, was significantly increased by PXR activation, and genes involved in ATRA metabolism exhibited differential expression following PXR activation or deficiency. Consistent with previous research, the expression of downstream target genes of peroxisome proliferator-activated receptor α (PPARα) was decreased. Analysis of fatty acids by Gas Chromatography-Mass Spectrometer further revealed changes in polyunsaturated fatty acid metabolism upon PXR activation, suggesting inhibition of PPARα activity. Background: Extracellular vesicles in human milk are critical in supporting newborn growth and development. Bioavailability of dietary extracellular vesicles may depend on the composition of membrane lipids. Single‐nucleotide polymorphisms (SNPs) in the fatty acid desaturase gene cluster impact the content of long‐chain polyunsaturated fatty acids in human milk phospholipids. This study investigated the relation between variation in FADS1 and FADS2 with the content of polyunsaturated fatty acids in extracellular vesicles from human milk. Methods: Milk was obtained from a cohort of mothers (N = 70) at 2–4 weeks of lactation. SNPs in the FADS gene locus were determined using pyrosequencing for rs174546 in FADS1 and rs174575 in FADS2. Quantitative lipidomic analysis of polyunsaturated fatty acids in human milk and extracellular vesicles from human milk was completed by gas chromatography–mass spectrometry. Results: The rs174546 and rs174575 genotypes were independent predictors of the arachidonic acid content in extracellular vesicles. The rs174546 genotype also predicted eicosapentaenoic acid and docosahexaenoic acid in extracellular vesicles. The reduced content of long‐chain polyunsaturated fatty acids in extracellular vesicles in human milk may be due to lower fatty acid desaturase activity in mothers who are carriers of the A allele in rs174546 or the G allele in rs174575. Conclusion: The polyunsaturated fatty acid composition of milk extracellular vesicles is predicted by the FADS genotype. These findings yield novel insights regarding extracellular vesicle content and composition that can inform the design of future research to explore how lipid metabolites impact the bioavailability of human milk extracellular vesicles. This study conducted a comparative analysis of fatty acids (FAs) concentration derived from polar (PL) and non-polar (NPL) lipids in the liver and muscle tissues of rats and mice. The objective was to elucidate species-specific differences in tissue FA distribution. Employing targeted GC/MS-based methodology, the study aimed to provide insights into conserved and species-specific aspects of lipid metabolism, thereby enhancing future experimental design, linked with comprehension of the interactions between diet, metabolism, and health. Results revealed markedly higher levels of total fatty acids (TFAs) derived from PL in mice skeletal muscle compared to rats and elevated saturated fatty acids (SFAs) levels in mice. Unsaturated fatty acid levels, mainly monounsaturated fatty acids (MUFAs) and polyunsaturated fatty acids (PUFAs), were substantially higher in mice muscle across all lipid classes, resulting in a higher PUFA/TFA ratio in mice muscle. Detailed analysis of specific unsaturated fatty acids (C16:1, C18:1n9c, C18:2n6c, C22:6n3) indicated elevated levels in mice relative to rats. Conversely, rats exhibited higher SFA levels derived from the NPL fraction in the liver, particularly in myristic (C14:0), stearic (C18:0), and tricosanoic (C23:0) acids. Mice liver PL fractions displayed significantly elevated unsaturated FA levels, with notably higher MUFAs and lower PUFAs in NPL fractions compared to rats. Analysis of specific unsaturated FAs revealed higher levels of palmitoleic acid (C16:1) in mice, while rats exhibited increased linoleic (C18:2n6c) and linolenic (C18:3n3) acids. In conclusion, significant differences in FAs tissue distribution between rats and mice underscore the importance of considering species-specific FAs variations when utilizing these animal models and interpreting experimental results related to FA metabolism. In this present study, potential oleaginous Mucorales fungi that have been isolated from fruits from local markets in Sleman, Indonesia, were screened for lipid production. A total of six fungal cultures were isolated and screened using a semisynthetic medium with glucose as a carbon source and a limited nitrogen supply. The highest lipid content was observed in isolate JR 1.1, up to 43.46% and 3.28 g/L lipid yield. Therefore, it was selected for molecular identification and fatty acid analysis. The result showed that JR 1.1 was identified as Mucor irregularis. The fatty acid profile of JR 1.1 showed 16.89% palmitoleic acid, 4.85% oleic acid, 45.22% linolenic acid, 30.79% gamma-linolenic acid, and 2.25% other fatty acids. It can be concluded that M. irregularis JR 1.1 is a potential strain to be used as a lipid producer for biodiesel feedstock. An oleaginous microalga Micractinum inermum isolated from Mariana Lake, AB, Canada was cultured in a 1000 L photobioreactor with an f/2 medium to study its lipid content and neutral lipid profile. Algal biomass was collected at the stationary phase contained a significant amount of lipids (44.2%), as determined by Folch's method. The lipid was fractionated into neutral lipid, glycolipid and phospholipid fractions. The neutral lipid constitutes almost 77.3% of the total lipid species and is mainly composed of triacylglycerols (TAGs) determined by a proton NMR study. UHPLC-HRMS analysis allows us for the first time to identify 81 TAGs in the neutral lipid fraction of M. inermum. The fatty acid acyl side chains were identified based on fragment ions observed in MSMS analysis. TAGs with fatty acid acyl chains 18:1/18:1/18:1, 18:1/18:1/16:0, 18:2/18:1/16:0, and 18:2/18:2/18:0 were the major ones among the identified TAGs. Fatty acid analysis further supports the fact that oleic acid was the major fatty acid present in the neutral lipid fraction of M. inermum constituting 41.7%, followed by linoleic acid at 21.5%, and palmitic acid at 21.2%. The saturated and monounsaturated fatty acids were 67.8% or higher in the lipid fraction. Long-chain fatty acids were only present in a minor quantity. Objective: In this study, the biochemical compositions of fatty acids, amino acids, fat-soluble vitamins (ADEK), cholesterol and phytosterols were examined in raw milk samples of Simmental breed cows fed with different forage combinations. Methods: Milk samples were provided from their farm in Kovancılar district of Elazığ province in Türkiye. The cows were fed in different forage combinations the spring season. These were Group 1: cows fed with Beet pulp+Factory feed+Hay (n: 10); Group 2: cows fed with Maize silage+Barley+Hay (n: 10), and Group 3: cows free-fed in pasture (n: 10). Milk samples were taken on the 15th day of feeding and brought to the laboratory. Biochemical analyses were performed on the milk samples brought to the laboratory in accordance with the cold chain rules. After the milk samples went through a number of biochemical processes, fatty acid, ADEK vitamins, cholesterol and amino acid analyses were performed. The analysis of cholesterol and ADEK vitamins was performed with a high performance liquid chromatography (HPLC) device, and analysis of amino acids and fatty acids was performed with gas chromatography (GC) and FID detector. Results: 18 amino acids were detected in the milk samples examined. Compared to Group 1, it was found that the amounts of arginine (p<0.001), isolosine (p<0.05), methionine (p<0.05), threonine (p<0.05), phenylalanine (p<0.001), lysine (p<0.05) and tryptophan (p<0.001) amino acids increased statistically, while the amounts of alanine (p<0.001), glycine (p<0.05), leucine (p<0.05), aspartic acid (p<0.01) and glutamic acid (p<0.05) decreased statistically in Group 3. Compared to Group 1, it was found that the amounts of arginine (p<0.001), threonine (p<0.05), phenylalanine (p<0.001) and lysine (p<0.05) amino acids increased statistically, while the amounts of alanine (p<0.001), glycine (p<0.001), proline (p<0.01), aspartic acid (p<0.05), glutamic acid (p<0.001), histidine (p<0.001) and tryptophan (p<0.05) decreased statistically in Group 2. Compared to Group 1, the amounts of lauric acid (p<0.05), myristic acid (p<0.01), pentadecanoic acid (p<0.05), palmitic acid (p<0.01), stearic acid (p<0.001), oleic acid (p<0.001) and cholesterol (p<0.05) were statistically decreased, while the amounts of linoleic acid (p<0.001), alpha-linolenic acid (p<0.001), dihomo-gamma-linolenic acid (p<0.05), vitamin K2 (p<0.001), γ-tocopherol (p<0.001), vitamin D2 (p<0.001), a-tocopherol (p<0.001), a-tocopherol acetate (p<0.05), vitamin K1 (p<0.001), stigmasterol (p<0.01), ß-sitosterol (p<0.05) were statistically increased in Group 3. Compared to Group 1, the amounts of stearic acid (p<0.001), oleic acid (p<0.01), a-tocopherol acetate (p<0.001) and ß-sitosterol (p<0.05) were statistically decreased, while the amounts of vitamin K2 (p<0.001), γ-tocopherol (p<0.01), vitamin D2 (p<0.01), a-tocopherol (p<0.01), vitamin K1 (p<0.01) and stigmasterol (p<0.01) were statistically increased in Group 2. It was determined that the amounts of palmitic, oleic, myristic, stearic, lauric, linoleic, palmitoleic and pentadecanoic acids were found to be above 1% in the fatty acid composition of milk among the groups, respectively. It was determined that there were significant changes in the amount of some fatty acids among the groups. In addition, it was determined that there were significant changes in the amounts of cholesterol, phytosterol and some ADEK vitamins among the groups. Conclusion: According to these data, it is seen that the use of different feed combinations in animal nutrition by dairy producers is very important for enriching the biochemical content of milk. It has been determined that the nutrient content of milk changes depending on different feed combinations and feeding in the flora. Oil cakes are pomace that is a by-product of pressing oil from oilseeds. Due to the content of bioactive ingredients, there is more and more talk about their valorization through use in human and animal nutrition as well as microbiological processes aimed at the biosynthesis of biosurfactants, enzymes and fragrance compounds. The aim of this research was a qualitative assessment of selected oil cakes (linseed, rapeseed, hemp, safflower and camelina oil cakes) in the context of their potential use in microbiological cultures aimed at the biosynthesis of green note aroma compounds. The scope of the research included the analysis of the fatty acid composition of the lipid fraction of oil cakes as well as the determination of lipoxygenase activity and the polyphenol content in the extracts of these raw materials. Chromatographic analysis of the lipid fraction showed that hemp and safflower cakes had the highest content of polyunsaturated fatty acids. PUFAs constitute 67.18 ± 2.2% and 73.72 ± 1.8% in their extracts, respectively. Hemp cake extracts were also characterized by the highest lipoxygenase activity—76.4–1.2 U/mL with a low content of phenolic compounds at the level of 0.116 ± 0.005 mg GAE/mL of extract. Chronic kidney disease (CKD) is a global health problem with rising incidence and prevalence. Among several pathogenetic mechanisms responsible for disease progression, lipid accumulation in the kidney parenchyma might drive inflammation and fibrosis, as has been described in fatty liver diseases. Lipids and their metabolites have several important structural and functional roles, as they are constituents of cell and organelle membranes, serve as signalling molecules and are used for energy production. However, although lipids can be stored in lipid droplets to maintain lipid homeostasis, lipid accumulation can become pathogenic. Understanding the mechanisms linking kidney parenchymal lipid accumulation to CKD of metabolic or non-metabolic origin is challenging, owing to the tremendous variety of lipid species and their functional diversity across different parenchymal cells. Nonetheless, multiple research reports have begun to emphasize the effect of dysregulated kidney lipid metabolism in CKD progression. For example, altered cholesterol and fatty acid metabolism contribute to glomerular and tubular cell injury. Newly developed lipid-targeting agents are being tested in clinical trials in CKD, raising expectations for further therapeutic development in this field. Chronic kidney disease is characterized by dyslipidaemia and lipid accumulation in the kidney. In this Review, the authors examine the evidence that links alterations in lipid metabolism to kidney injury and progression of kidney disease, and explore potential lipid-targeted therapeutic approaches. Key points: Lipids and lipid-related enzymes have a major role in modulating the function of glomerular and tubular cells, and can drive chronic kidney disease (CKD) irrespective of circulating lipid levels. The mechanisms that initiate the accumulation of kidney lipids might differ between CKD of different aetiologies. Thus, the accumulation of kidney lipids in diabetic kidney disease is driven by increased glucose and fatty acids levels owing to insulin resistance, whereas in glomerulonephritis, inflammation can disrupt normal kidney lipid metabolism. Several lipid species, such as cholesterol, triglycerides, fatty acids and phospholipids, are dysregulated in podocytes, endothelial and tubular cells, and contribute to CKD progression. Accumulation of kidney parenchymal cholesterol occurs in association with impaired reverse cholesterol transport in diseases of both metabolic and non-metabolic origin, and contributes to CKD progression. Accumulation of fatty acids triggers mitochondrial and kidney cell damage by promoting inflammation, including cellular sterile inflammation, via innate immune system activation and fibrosis; lipophagy has a protective effect. Patients with non-alcoholic steatohepatitis (NASH) show significantly faster progress in the stages of fibrosis compared to those with non-alcoholic fatty liver (NAFL) disease. The non-invasive diagnosis of NASH remains an unmet clinical need. Preliminary data have shown that sphingolipids, especially ceramides, fatty acids, and other lipid classes may be related to the presence of NASH and the histological activity of the disease. The aim of our study was to assess the association of certain plasma lipid classes, such as fatty acids, acylcarnitines, and ceramides, with the histopathological findings in patients with NASH. The study included three groups: patients with NASH (N = 12), NAFL (N = 10), and healthy [non non-alcoholic fatty liver disease (NAFLD)] controls (N = 15). Plasma samples were collected after 12 h of fasting, and targeted analyses for fatty acids, acylcarnitines, and ceramides were performed. Baseline clinical and demographic characteristics were collected. There was no significant difference in baseline characteristics across the three groups or between NAFL and NASH patients. Patients with NASH had increased levels of several fatty acids, including, among others, fatty acid (FA) 14:0, FA 15:0, FA 18:0, FA 18:3n3, as well as Cer(d18:1/16:0), compared to NAFL patients and healthy controls. No significant difference was found between NAFL patients and healthy controls. In conclusion, patients with NASH exhibited a distinctive plasma lipid profile that can differentiate them from NAFL patients and non-NAFLD populations. Nutritional quality, proximate and fatty acid compositions of fish species from the Khabur, Ambar and Tigris Rivers in SE Türkiye were investigated for the first time. The fish with the lowest total lipids were Mastacembelus mastacembelus (0.93%) and Carasobarbus luteus (0.99%), and the fish with the highest total lipid was Chondrostoma regium from the Tigris River (7.47%). The highest cholesterol content was in Barbus lacerta (26.3 mg/100 g) and Capoeta umbla (29.98 mg/100g) of the Ambar Stream. However, the Tigris River Cyprinus carpio (7.9 mg/100g) and C. luteus (7.91 mg/100g) had the lowest cholesterol. The results showed that all species are good sources of SSFA and SMUFA, specifically C14:0, C16:0, C18:0, C16:1-7 and C18:1-9. However, the fish were poor for -6 and -3, particularly C20:4-6, C20:5-3 and C22:6-3, probably due to hot water adaptation in summer. Nevertheless, C. carpio (Tigris River) and A. mossulensis (Khabur River) had relatively high SPUFA. Among all the fish, C. regium and A. mossulensis from the Khabur River were good for protein, and M. mastacembelus, C. luteus, C. carpio and C. trutta from the Tigris River can be recommended as lean fish. Objective: Previous observational studies revealed a potential but partially controversial relation between lipid metabolism and the risk of amyotrophic lateral sclerosis (ALS), potentially prone to bias. Therefore, we aimed to study whether lipid metabolism involves genetically determined risk factors for ALS through Mendelian randomization (MR) analysis. Methods: Using genome‐wide association study summary‐level data for total cholesterol (TC) (n = 188,578), high‐density lipoprotein cholesterol (HDL‐C) (n = 403,943), low‐density lipoprotein cholesterol (LDL‐C) (n = 440,546), apolipoprotein A1 (ApoA1) (n = 391,193), apolipoprotein B (ApoB) (n = 439,214), and ALS (12,577 cases and 23,475 controls), we implemented a bidirectional MR study to evaluate a genetic relation between lipids and ALS risk. We performed a mediation analysis to assess whether LDL‐C is a potential mediator on the pathway from traits of LDL‐C‐related polyunsaturated fatty acids (PUFAs) to ALS risk. Results: We identified genetically predicted increased lipid levels to be associated with the risk of ALS, whereby elevated LDL‐C had the most potent effect (OR 1.028, 95% CI 1.008–1.049, p = 0.006). The effect of increased levels of apolipoproteins on ALS was similar to their corresponding lipoproteins. ALS did not cause any changes in lipid levels. We found no relation between LDL‐C‐modifying lifestyles and ALS. The mediation analysis revealed that LDL‐C could act as an active mediator for linoleic acid, with the mediation effect estimated to be 0.009. Conclusions: We provided high‐level genetic evidence verifying the positive link between preclinically elevated lipid and ALS risk that had been described in previous genetic and observational studies. A whole-grain diet is associated with the prevention of metabolic syndromes, including obesity, diabetes, and cardiovascular diseases. Sprouting improves the nutritional profile and bioactive properties of grains, which are important for use as raw ingredients in the food industry. The aim of this review was to examine the lipid and fatty acid composition of germinated grains. The methods discussed include germination and analytical procedures for determining fat and fatty acid contents of grains. The effects of sprouting on the fat content and storage stability of grains were also assessed. Lipid levels ranged from 1.43% to 6.66% in the sprouted grains. The individual fatty acid content of grains changed depending on the germination conditions (17–37 °C, 1–9 days). Limited findings showed that sprouting grains at higher temperatures (20–25 °C) and longer times generated a healthy balance of omega-6 and omega-3 fatty acids, which is beneficial to humans. Future studies are needed to determine the optimum incubation and germination periods specific to each grain to improve the omega-6/omega-3 ratio. Free fatty acids were produced more slowly and levels of oxidation products were lower in sprouted grains than in the raw ingredients when stored for a year. Krill species are key organisms in the Antarctic food web. Biochemical composition in terms of lipids, proteins, carbohydrates, and fatty acids and its implications for spatial distribution were investigated in specimens of Euphausia superba and Euphausia crystallorophias collected in the Ross Sea and the adjacent Pacific region during an acoustic survey carried out within the framework of the 19th Italian National Program for Research in Antarctica (PNRA) Expedition, to gain insights into their trophic relationships and bioenergetic strategies. In both species, the body biochemical composition (wet) showed a predominance of proteins (62–86%), followed by lipids and carbohydrates, and, among identified lipid classes, the two species did not seem to differ much in fatty acid composition. Results showed the highest dissimilarity in biochemical composition between species relative to differences in latitude (24%) and to inshore/offshore haul (22%). Fatty acid analysis, and particularly PUFA/SFA and 18PUFA/16PUFA ratios, allowed identification of a more pronounced omnivorous kind of diet in E. crystallorophias relative to E. superba. Objective: The types and functions of lipids involved in glioblastoma (GB) are not well known. Lipidomics is a new field that examines cellular lipids on a large scale and novel aplication of lipidomics in the biomedical sciences have emerged. This study aimed to investigate the potential of blood lipids for use as biomarkers for the diagnosis of GB via untargated lipidomic approach. Gaining a deeper understanding of lipid metabolism in patients with GB can contribute to the early diagnosis with GB patiens and also development of novel and better therapeutic options. Methods: This study was performed using blood samples collected from 14 patients (eight females and six males) and 14 controls (eight females and six males). Lipids were extracted from blood samples and quantified using phosphorus assay. Lipid profiles of between patients with GB and controls were compared via an untargeted lipidomics approach using 6530 Accurate-Mass Q-TOF LC/MS mass spectrometer. Results: According to the results obtained using the untargeted lipidomics approach, differentially regulated lipid species, including fatty acid (FA), glycerolipid (GL), glycerophospholipid (PG), saccharolipid (SL), sphingolipid (SP), and sterol lipid (ST) were identified between in patients with GB and controls. Chemical and physical mutagens like sodium azide, H2O2 and UV light have a significant role in the metabolism of living organisms. Effect of these mutagens was recorded during the present work on lipid content of Oscillatoria sp. and Ulothrix sp. The algal species were subjected to the mutagens by adding them in culture media. Lipids were extracted by Microwave-assisted extraction using methanol, ethanol and chloroform. During the present work generally a significant effect of algal species and mutagens was observed. All the tested mutagen significantly affected the lipid content. A significant effect of solvent on lipid content of algal species was observed when the algae were extracted with different solvents after treatment with H2O2 and UV light however this effect was non-significant when algae were subjected to sodium azide. In general, the mutagen resulted in an increase in lipid content. In Oscillatoria sp. the lipid content was generally less as compared to Ulothrix sp. The highest lipid content of 0.5433±0.002 g100g-1 was recorded in Ulothrix sp. after 3 days stress of sodium azide stress followed by H2O2 treated (50mM) samples with a content of 0.5133±0.05g100g-1. In Oscillatoria sp. 15mM treatment for 3 days gave a lipid content of 0.3733±0.0001g100g-1 after 3 days. A reduction in fresh weights of both tested algal species was recorded after UV stress. The oil content also showed a significant reduction after UV exposure. GC-MS analysis of the transesterified lipids showed that different fatty acids show variation when algal samples were treated with mutagens. Simple Summary: Insects are an important source of protein but insect fat increasingly slides into focus due to its high content of unsaturated fatty acids. Tenebrio molitor larvae were reared on several substrates with different nutritional contents to influence the fat and fatty acid contents of the larvae. The fat and fatty acid composition of the mealworm larvae were then analyzed in order to determine if a nutritional change could be detected using near-infrared reflectance spectroscopy. This is a rapid and non-destructive method for the online analysis of chemical composition. In this study, the diet used for rearing had a significant effect on the larval fat content and fatty acid composition as well as the absorbance of the near-infrared spectra and the larval growth rate and weight gain, i.e., a high fat content substrate reduced weight gain and larval growth. The most prevalent fatty acids identified and quantified were palmitic, oleic, and linoleic acid, showing a correlation between larval content and rearing diets containing high fatty acid amounts. A high dietary content of lauric acid, myristic acid, and α-linolenic acid resulted in a high content of these fatty acids in mealworm larvae. The fat and fatty acid content could be predicted accurately using near-infrared reflectance spectroscopy and were highly influenced by several diets having different proximate compositions. Several studies have shown that mealworms (Tenebrio molitor L.) could provide animals and humans with valuable nutrients. Tenebrio molitor larvae were studied to determine whether their rearing diets affected their fat and fatty acid content and to ascertain if it is possible to detect the changes in the larval fat composition using near-infrared reflectance spectroscopy (NIRS). For this reason, a standard control diet (100% wheat bran) and an experimental diet, consisting of wheat bran and the supplementation of a different substrate (coconut flour, flaxseed flour, pea protein flour, rose hip hulls, grape pomace, or hemp protein flour) were used. The results showed lesser weight gain and slower growth rates for larvae raised on diets with a high fat content. A total of eight fatty acids were identified and quantified, where palmitic, oleic, and linoleic acids were the most prevalent and showed a correlation between larval content and their content in the rearing diets. There was a high content of lauric acid (3.2–4.6%), myristic acid (11.4–12.9%), and α-linolenic acid 8.4–13.0%) in mealworm larvae as a result of the high dietary content of these fatty acids. NIR spectra were also influenced by the fat and fatty acid composition, as larval absorbance values differed greatly. The coefficient of the determination of prediction (R2P) was over 0.97, with an RPD value of 8.3 for the fat content, which indicates the high predictive accuracy of the NIR model. Furthermore, it was possible to develop calibration models with great predictive efficiency (R2P = 0.81–0.95, RPD = 2.6–5.6) for all fatty acids, except palmitoleic and stearic acids which had a low predictive power (R2P < 0.5, RPD < 2.0). Tocopherols are natural bioactive compounds with several health benefits. This study evaluated the effect of different ratios of α- and δ- tocopherol homologs to protect sunflower oil (SO) and olive pomace oil (OPO) against oxidation. A synergistic effect was recorded when the two tocopherols were combined at a ratio of 7:1 (α-T/δ-T). The oil samples were exposed to accelerated oxidation conditions using a Rancimat (90 °C and airflow of 15 L/h for 24 h) and protection from tocopherols was compared with that from butylated hydroxytoluene (BHT). Assessment of oil stability was examined using well-known parameters such as peroxide value (PV), thiobarbituric acid reactive substances (TBARS), p-anisidine value (p-AV), conjugated dienes (CD) and trienes (CT), and total oxidation (Totox) value, which were all significantly reduced when tocopherols were added at a ratio of 7:1 α-T/δ-T. Primary oxidative compounds measured according to PV were only reduced in SO samples (6.11%). Off-flavor compounds measured via TBARS assay in SO samples were reduced by above 20%, while p-AV was also reduced. CDvalue was correlated with PV in SO samples, while the 7:1 mixture was more effective than BHT for CTvalue. Total oxidation values in SO samples and OPO samples were reduced by 6.02% and 12.62%, respectively. These values in SO samples also provided a remarkable correlation (R2 > 0.95) with incubation time. Moreover, the synergistic effect was not only effective in reducing the oxidation values of oil samples, but also in lowering the degradation rate of tocopherols. Protective effects from tocopherols were mainly observed in SO samples, as OPO samples were more resistant to oxidation processes. This effect was even observed in fatty acid analysis, where the 7:1 mixture provided better results than BHT-spiked samples. The objective of this research was to determine the effect of the extended dry aging period (0, 20, 40, and 60 days) on lipid oxidation, fatty acid profile, and tenderness in selected dry-aged Hanwoo cow beef cut loins. Dry aging regimes were 40C and 85% relative humidity. Lipid oxidation was determined by using 2- thiobarbituric acid reactive substances. Shear force values of the highly marbled beef decreased first 20d aging and did not significantly change after that, until 60d. The hardness of texture profile analysis gradually decreased, but the chewiness value did not differ. A total of 16 fatty acids were identified in dryaged beef. The most predominant FAs in the aged beef were C18:1, C16:0, and C18:0. The C18:2 was the predominant polyunsaturated fatty acid in the samples. Total PUFA gradually and C18:2n-6 decreased (p<0.05) during aging. C20:1, C18:3n-3, C20:4, and C22:1 acid significantly decreased (p<0.05) until 20d aging, and no changes were observed after 20d to 60d. 2-thiobarbituric acid reactive values of beef increased (p<0.001) during dry-aging days, dry aging for 40d and 60d resulted in excessive oxidation of lipids. Dry-aged meat is so popular because of its unique taste. Improvements in tenderness are associated with the 20d aging of beef subprimal, however, some lipid oxidation may be associated with the development of specific dry-aged beef flavor. Summary: Bile acid: sodium symporter family protein 2 (BASS2) is a sodium‐dependent pyruvate transporter, which transports pyruvate from cytosol into plastid in plants. In this study, we investigated the function of chloroplast envelope membrane‐localized BnaBASS2 in seed metabolism and seed oil accumulation of Brassica napus (B. napus). Four BASS2 genes were identified in the genome of B. napus. BnaA05.BASS2 was overexpressed while BnaA05.BASS2 and BnaC04.BASS2‐1 were mutated by CRISPR in B. napus. Metabolite analysis revealed that the manipulation of BnaBASS2 caused significant changes in glycolysis‐, fatty acid synthesis–, and energy‐related metabolites in the chloroplasts of 31 day‐after‐flowering (DAF) seeds. The analysis of fatty acids and lipids in developing seeds showed that BnaBASS2 could affect lipid metabolism and oil accumulation in developing seeds. Moreover, the overexpression (OE) of BnaA05.BASS2 could promote the expression level of multiple genes involved in the synthesis of oil and the formation of oil body during seed development. Disruption of BnaA05.BASS2 and BnaC04.BASS2‐1 resulted in decreasing the seed oil content (SOC) by 2.8%–5.0%, while OE of BnaA05.BASS2 significantly promoted the SOC by 1.4%–3.4%. The yeast Rhodosporidium babjevae was used to produce carotenoids, lipids, and fatty acids using sugarcane bagasse hydrolysates or glucose. Investigating the impact of glucose concentrations (10–150 g/L), the yeast showed the capability to yield 16.9 g/L biomass and 10.5 g/L lipids at 60 g glucose/L, while the highest pigments concentration (312 μg/g) was observed at 10 g glucose/L. Pre-treatment methods, including organosolv (ethanol 60 %(w/w)), alkali (NaOH 2 %(w/v)) and alkaline-organosolv (ethanol 60 %(w/w) and NaOH 2 %(w/v)) were used on the sugarcane bagasse following enzymatic hydrolysis. Among these, alkaline organosolv pre-treatment exhibited the most effective lignin removal of 81.9 %, releasing 46.7 g/L sugars by hydrolysis. Yeast cultivation on the enzymatic hydrolysates of alkaline organosolv pre-treatment resulted in the highest biomass (9.6 g/L) and lipids (5.0 g/L) yields. However, the highest content of pigments (280 μg/g) was achieved when the yeast was cultivated on the enzymatic hydrolysates of organosolv pre-treatment. Production of 11.3 g/L biomass, 204 μg/g pigments, and 6.7 g/L lipids established that corn steep liquor holds potential as a cheap and viable alternative to yeast extract as nitrogen source. This study investigated the effect of ultrasound assisted irradiation on the carotenoids and lipids extraction from the Chlorella vulgaris and Porphyridium purpureum microalgae in the presence of three solvents with various polarities (water, ethanol, and hexane) allowed in the food industry. The optimization of the ultrasound assisted extraction operational parameters (solvent composition, solvent to sample ratio, power) was realized using Response Surface Methodology based on a face-centered composite design (FCCD). The results of this study showed that the increased solvent polarity led to the maximum extraction yield of 6435.60 µg/g in total carotenoid content. Regarding the distribution of carotenoid between the aqueous ethanolic phase and hexane, the highest yield of 5762.08 µg/g carotenoid was obtained in hexane phase. Regarding the lipid content, the yield increased with the increase of ethanol concentration reaching a maxim value of 29.07 mg/g. Also, it was observed that, when single solvents were used for carotenoid extraction, the yields were much lower compared with mixtures solvents. For example, when hexane was tested the yield was of 226.87 µg/g carotenoid, meanwhile for absolute ethanol the yield was of 2027.72 µg/g The optimal parameters determined was applied for the extraction of carotenoids from Porphyridium purpureum microalgae. The results indicate a total carotenoid content of 947.25 µg/g and 21.59 mg/g lipids. The analysis of fatty acid profile of both microalgae biomass in ethanolic and hexane phase indicated the selective accumulation of saturated and polyunsaturated fatty acids in hexane and respectively in ethanolic phase. Background: Sustainable production of oil for food, feed, fuels and other lipid-based chemicals is essential to meet the demand of the increasing human population. Consequently, novel and sustainable resources such as lignocellulosic hydrolysates and processes involving these must be explored. In this paper we screened for naturally-occurring xylose utilizing oleaginous yeasts as cell factories for lipid production, since pentose sugar catabolism plays a major role in efficient utilization of lignocellulosic feedstocks. Glycerol utilization, which is also beneficial in yeast-based oil production as glycerol is a common by-product of biodiesel production, was investigated as well. Natural yeast isolates were studied for lipid accumulation on a variety of substrates, and the highest lipid accumulating strains were further investigated in shake flask cultivations and fermenter studies on xylose and hydrolysate. Results: By collecting leaves from exotic plants in greenhouses and selective cultivation on xylose, a high frequency of oleaginous yeasts was obtained (> 40%). Different cultivation conditions lead to differences in fatty acid contents and compositions, resulting in a set of strains that can be used to select candidate production strains for different purposes. In this study, the most prominent strains were identified as Pseudozyma hubeiensis BOT-O and Rhodosporidium toruloides BOT-A2. The fatty acid levels per cell dry weight after cultivation in a nitrogen limited medium with either glucose, xylose or glycerol as carbon source, respectively, were 46.8, 43.2 and 38.9% for P. hubeiensis BOT-O, and 40.4, 27.3 and 42.1% for BOT-A2. Furthermore, BOT-A2 accumulated 45.1% fatty acids per cell dry weight in a natural plant hydrolysate, and P. hubeiensis BOT-O showed simultaneous glucose and xylose consumption with similar growth rates on both carbon sources. The fatty acid analysis demonstrated both long chain and poly-unsaturated fatty acids, depending on strain and medium. Conclusions: We found various natural yeast isolates with high lipid production capabilities and the ability to grow not only on glucose, but also xylose, glycerol and natural plant hydrolysate. R. toruloides BOT-A2 and P. hubeiensis BOT-O specifically showed great potential as production strains with high levels of storage lipids and comparable growth to that on glucose on various other substrates, especially compared to currently used lipid production strains. In BOT-O, glucose repression was not detected, making it particularly desirable for utilization of plant waste hydrolysates. Furthermore, the isolated strains were shown to produce oils with fatty acid profiles similar to that of various plant oils, making them interesting for future applications in fuel, food or feed production. Purpose: Human milk (HM) composition is influenced by factors, like maternal diet and body stores, among other factors. For evaluating the influence of maternal fatty acid (FA) status on milk FA composition, the correlation between FA content in HM and in maternal plasma, erythrocytes, and adipose tissue was investigated. Methods: 223 European women who delivered at term, provided HM samples over first four months of lactation. Venous blood and adipose tissue (only from mothers who consented and underwent a C-section delivery) were sampled at delivery. FAs were assessed in plasma, erythrocytes, adipose tissue, and HM. Evolution of HM FAs over lactation and correlations between FA content in milk and tissues and between mother's blood and cord blood were established. Results: During lactation, arachidonic acid (ARA) and docosahexaenoic acid (DHA) significantly decreased, while linoleic acid (LA), alpha-linolenic acid (ALA), and eicosapentaenoic acid (EPA) remained stable. Positive correlations were observed between HM and adipose tissue for palmitic, stearic, oleic, and polyunsaturated fatty acids (PUFAs). Correlations were found between milk and plasma for oleic, LA, ARA, ALA, DHA, monounsaturated fatty acids (MUFAs), and PUFAs. No correlation was observed between erythrocytes and HM FAs. LA and ALA were more concentrated in maternal blood than in infant blood, contrary to ARA and DHA, supporting that biomagnification of LCPUFAs may have occurred during pregnancy. Conclusions: These data show that maternal adipose tissue rather than erythrocytes may serve as reservoir of PUFAs and LCPUFAs for human milk. Plasma also supplies PUFAs and LCPUFAs to maternal milk. If both, adipose tissue and plasma PUFAs, are reflection of dietary intake, it is necessary to provide PUFAs and LCPUFAs during pregnancy or even before conception and lactation to ensure availability for mothers and enough supply for the infant via HM. The present study has examined the influence of various photoperiods, light intensities, and pH on the biomass, total lipids, triacylglycerol (TAGs), biodiesel conversion, and fatty acid composition of Chlorella vulgaris (C. vulgaris) for sustainable fuel production. Of the L/D photoperiods tested, biomass yield was low in the 10:14 h grown culture of about 0.94 g/L, while a high biomass yield of 1.486 g/L was recorded in the 16:08 h grown culture on the 20th day. Of the 1000, 1500, 2000, and 2500 lx light irradiation tested, higher biomass production of 1.613 g/L was achieved in the 2000 lx-grown cells. However, initial pH levels of medium such as 5, 6, 7, 8, and 9 did not have an impact on the biomass yield. Regarding the lipid content, the high total lipid content of 29.85 % and 30.85 % was obtained from the culture grown at 16:08 h L/D photoperiod and 2500 lx, respectively. Concerning the TAG proportion in the total lipids, the maximal TAG of 46.83 and 44.47% (100g total lipids) were from the 16:08 h L/D cycle and 2500 lx condition, respectively. The high biodiesel yield of about 68.58 % and 65.98% was noticed from 16:08 h L/D and 2500 lx, respectively. Further, the fatty acid profile analysis of the biodiesel produced from the optimal conditions indicated that saturated fatty acids (SFAs) and polyunsaturated fatty acids (PUFAs) levels were 54.37 and 45.63% under 16:08 photoperiod, respectively, while they were 52.74 and 47.26%, respectively under 2500 lx light illumination condition. • Quantification by TD-NMR of lipids in organic waste is more accurate than Soxhlet. • TD-NMR analyses should be performed above 66 °C to get a good lipid quantification. • Coupled with chemometrics, TD-NMR quantifies low levels of lipid content (from 2%) Time-Domain Nuclear Magnetic Resonance (TD-NMR) was used to quantify the lipid contents of 48 different organic waste substrates. Results obtained from TD-NMR were compared to those from Soxhlet extraction, currently the prevalent method for organic waste characterization, especially in the field of anaerobic digestion. Two calibration methods were tested. The first was a self-calibration process using pure oils (NMR1) which showed good repeatability compared to Soxhlet extraction with a better coefficient of variation (5%). Analyses of volatile fatty acids (VFA) and long-chain fatty acids (LCFA) by chromatography were carried out to understand why the NMR1 method produced underestimations for some samples. Statistical analysis showed that the presence of saturated fatty acids had a significant effect on differences between the Soxhlet and NMR1 methods. The second calibration method applied chemometrics to TD-NMR raw data (NMR2), taking Soxhlet extraction values as references. It provided a good prediction of lipid content and avoided the lengthy calibration procedure usually required for this type of study. The review considers the principal, as well as less common but promising, methods to determine the fatty acids (FAs) in biological samples. The advantages, disadvantages, and prospects of these methods are described and compared. Special attention is paid to specific features of the preparation of biological samples, such as the extraction of lipids and derivatization process of FAs. Methodological aspects of the determination of FAs via gas chromatography (GC) and high-performance liquid chromatography (HPLC) and features of the applications of various types of stationary phases used for the chromatographic separation of FAs are considered. The potential for the use of capillary electrophoresis (CE) and nuclear magnetic resonance spectroscopy (NMR) to analyze FAs are analyzed. The olive growing in Tunisia has an economic dominance and agricultural importance. However, the huge extraction of olive oil generates a large quantity of olive mill wastewater (OMW), which is discharged to the surroundings. The highly polluting potential (organic load) of OMW threatens the environment and requires an urgent solution. Supercritical fluid extraction (SFE) is a green extraction method that can be applied to purify OMW and, at the same time, to isolate a high quality oil from this wastewater. In order to explore and to valorize the compositions of Olive mill wastewater (OMW), extraction in different solvents (supercritical CO2, hexane) was carried out and chemical composition of the extracted oils were established by GC-FID. The Tunisia OMW were collected from two different zones namely Sousse and Sfax. In this work, we have investigated the effects pressure (P) and temperature (T) on the yield and the quality of oil. The suitable conditions for the extraction of oil from lyophilized OMW by Supercritical carbon dioxide (SC-CO2) were found to be the pressure of 30 MPa and the temperature of 60 °C. In order to simulate the process, the model of broken and intact cells (Sovova's model) was applied. The model well represented the experimental data. Oil yields ranged from 21.3 % to 33.87 % depending on the extraction solvent used. Monounsaturated fatty acids (MUFA) were the major compounds of the oils, based on the fatty acid analysis. Human milk lipids are among the many nutrients delivered to the infant, providing >50% of the infant's calorie intake. These lipids are highly complex and variable, and bioactive, contributing to infant growth, development, and health. The lipid concentration of milk samples is often measured in human cohorts; however, few studies measure infant intake of milk. Intake is important because it considers the variability of both lipid concentration and infants' consumed volume of milk. Measurement of infants' lipid intake in exclusively breastfeeding infants requires 3 main considerations: human milk sampling protocol (ie, the collection of representative samples); measurement of the infant milk intake, because volume varies widely between infants; and appropriate analytical laboratory methods. The purpose of this review was to provide an overview of existing methodology and demonstrate the importance of measuring infants' lipid intake to understand the impact that human milk lipids have on infant outcomes. Parenteral lipid emulsions are a necessary component of nutrition for extremely low gestational age newborns until adequate levels of enteral intake are established. Historically, Intralipid, a 100% soybean oil emulsion, has filled this role. Newer multicomponent lipid emulsions containing a mixture of other oils, including olive oil and fish oil, are now available as options, although the regulatory approval for use in neonates varies worldwide. When dosed at currently published recommendations, each of these lipid emulsions meets total fat and energy requirements without a risk of essential fatty acid deficiency. Thus, when choosing which lipid emulsion to provide, the answer must be based on the metabolic differences induced as a result of these fatty acid-rich emulsions and whether the emulsions provide a health advantage or pose a health risk. The questions of induced fatty acid profiles, health benefit and health risk are discussed sequentially for multicomponent lipid emulsions. Despite the growing acceptance of multicomponent lipid emulsions, there is concern regarding changes in blood fatty acid levels and potential health risk without strong evidence of benefit. There remains no ideal parenteral lipid emulsion option for the preterm infant. Standardising future animal and human studies in lipid delivery with the inclusion of lipid metabolism data will iteratively provide answers to inform the optimal lipid emulsion for the preterm infant. Context: Maternal lipids during pregnancy and placental growth factors are associated with excess fetal growth. However, how these factors interact to increase the risk of delivering large-for-gestational-age (LGA) neonates remains unclear. In this study, we investigated the relationship between maternal plasma triglycerides (TGs) and free fatty acids (FFAs) during pregnancy, cord blood insulin-like growth factors (IGF), and LGA.Objective: In a cell model, we studied the effect of different FAs on placental IGF-1 secretion.Methods: This cohort study included pregnant women with term pregnancy and without diabetes or hypertensive disorders in pregnancy. Maternal fasting plasma TGs and FFAs were measured in the second trimester. Cord blood IGF-1, IGF-2, and IGF binding protein-1 and protein-3 were measured at the time of delivery. A human trophoblast cell line, 3A-sub-E, was used to evaluate the effect of different FFAs on placental IGF-1 secretion.Results: We recruited 598 pregnant women-neonate pairs. Maternal plasma TG (180 mg/dL [152.5-185.5 mg/dL] vs 166 mg/dL [133-206 mg/dL], P = .04) and cord blood IGF-1 concentrations (72.7 ± 23.0 vs 54.1 ± 22.8 ng/mL, P < .001) were higher in the LGA group and were significantly associated with birth weight z score. Maternal plasma free palmitic acid (PA) and stearic acid (SA), but not oleic acid (OA) or linoleic acid (LA), were significantly associated with cord blood IGF-1 concentrations. In 3A-sub-E cells, treatment with PA, SA, and LA, but not OA, induced IGF-1 expression and secretion.Conclusion: Certain FFAs can induce placental IGF-1 secretion, which suggests a potential pathophysiology linking maternal plasma lipids and LGA. Quantitative fatty acid signature analysis (QFASA) as a biochemical tool to study the diet composition of predators is frequently used in marine ecology to infer trophic links in vertebrate consumers. However, the potential and challenges of this method in other ecosystems have only recently been studied. The application in soil ecosystems leads to hurdles not encountered in the marine, such as the low similarity of fatty acid signatures between resource and consumer. So far, diet estimation attempts have been semisuccessful, necessitating to adapt QFASA for use in soil food webs. Dietary fat content may play an important role, as it influences consumer metabolism, and thus calibration coefficients for fatty acid trophic transfer. A series of feeding trials with baker's yeast spiked with five different pure fatty acids at various concentrations was conducted with Collembola, and the changes in calibration coefficients were observed. From there, equations were gained through regression analysis and new sets of calibration coefficients were calculated. QFASA was applied on a range of basal resources and the results compared with previously defined calibration coefficients. Calibration coefficients changed with the proportion of fatty acids in the diet and differed between the three Collembolan species. The re‐estimation of diets showed an improvement of model performance by the new calibration coefficients and indicated several modes of fatty acid assimilation. These greatly influence the outcome of diet estimation, for example, algal and bacterial diets are likely underestimated due to high metabolic turnover rates. The application of QFASA in soil ecosystems remains challenging. The variation in calibration coefficients and the resulting decrease in estimation deviation indicate the merit of calculating calibration coefficients from consumer signatures through linear or exponential equations. Commercial cultivation of eukaryotic microalgae has so far employed a unicellular form of species only (e.g., Chlorella pyrenoidosa, Dunaliella salina, and Haematococcus pluvialis). In this study, we assessed the feasibility of using the filamentous eukaryotic microalga Klebsormidium sp. LGX80 as a new cultivar for biomass and lipid production. The effects of different forms and concentrations of nitrogen on growth and lipid production of Klebsormidium sp. LGX80 were studied by using a glass column (ø4.5 × 60 cm) photobioreactor under laboratory conditions. Growth and lipid production of the new strain were further evaluated in an outdoor pilot‐scale tubular photobioreactor. The results showed that when supplied with urea as a source of nitrogen Klebsormidium sp. LGX80 yielded a final biomass concentration of 8.49 ± 0.10 g · L−1 in which a cellular lipid content was 59.2 ± 0.4% DW. Under such conditions, the biomass and lipid productivities were 471.7 ± 5.9 and 248.1 ± 0.0 mg · L−1 · d−1, respectively. Fatty acid analysis revealed that the main fatty acids of Klebsormidium sp. LGX80 were palmitic acid (C16:0), linoleic acid (C18:2ω6), and linolenic acid (C18:3ω3), of which linoleic acid (C18:2ω6) accounted for up to 67.5 ± 0.1% of total fatty acids. When grown outdoors in a 13,000‐L tubular photobioreactor with an initial nitrogen concentration of 3 mM urea, Klebsormidium sp. LGX80 reached the highest biomass concentration of 2.63 ± 0.09 g · L−1 with the cells containing 38.0 ± 0.5% lipids (% DW), resulting in the volumetric biomass and lipid productivities of 147.2 ± 3.6 and 37.9 ± 0.9 mg · L−1 d−1, respectively. The results of light:dark cycle experiment showed that a durative and prolonged light irradiation hindered the biosynthesis of chlorophyll a and chlorophyll b in the cells, but promoted the carotenoid accumulation. Marine mammals are exposed to O2-limitation and increased N2 gas concentrations as they dive to exploit habitat and food resources. The lipid-rich tissues (blubber, acoustic, neural) are of particular concern as N2 is five times more soluble in lipid than in blood or muscle, creating body compartments that can become N2 saturated, possibly leading to gas emboli upon surfacing. We characterized lipids in the neural tissues of marine mammals to determine whether they have similar lipid profiles compared to terrestrial mammals. Lipid profiles (lipid content, lipid class composition, and fatty acid signatures) were determined in the neural tissues of 12 cetacean species with varying diving regimes, and compared to two species of terrestrial mammals. Neural tissue lipid profile was not significantly different in marine versus terrestrial mammals across tissue types. Within the marine species, average dive depth was not significantly associated with the lipid profile of cervical spinal cord. Across species, tissue type (brain, spinal cord, and spinal nerve) was a significant factor in lipid profile, largely due to the presence of storage lipids (triacylglycerol and wax ester/sterol ester) in spinal nerve tissue only. The stability of lipid signatures within the neural tissue types of terrestrial and marine species, which display markedly different dive behaviors, points to the consistent role of lipids in these tissues. These findings indicate that despite large differences in the level of N2 gas exposure by dive type in the species examined, the lipids of neural tissues likely do not have a neuroprotective role in marine mammals. Lipids are essential components of all living cells. In an oenological context, the supply of unsaturated lipids in grape juice allows the yeasts to grow and ferment, despite very low levels of oxygen. The current study proposes a systematic optimization procedure for the analysis of fatty acids and sterols relevant to the grape fermentation process, including both extracellular and intracellular (i.e., yeast cells) lipids. Even though it was extensive, the sample preparation yielded reproducible results for all compounds of interest. The stability of the analyzed compounds was also tested to offer some implementation flexibility for the extensive procedure. The performance parameters (i.e., selectivity, linearity, limit of detection and quantitation, accuracy, and precision) indicated that the method was suitable for future practical implementation. The proof of concept also suggests that the list of compounds of interest can be expanded if additional peaks are identified. Given the large variation in concentrations, the dilution of the matrix needs to be carefully considered in order to ensure that the lipids of interest are still within the dynamic range and not below the limit of detection and/or quantification. Background: Single-voxel proton cardiovascular magnetic resonance spectroscopy (1H-CMRS) benefits from 3 T to detect metabolic abnormalities with the quantification of intramyocardial fatty acids (FA) and creatine (Cr). Conventional point resolved spectroscopy (PRESS) sequence remains the preferred choice for CMRS, despite its chemical shift displacement error (CSDE) at high field (≥ 3 T). Alternative candidate sequences are the semi-adiabatic Localization by Adiabatic SElective Refocusing (sLASER) recommended for brain and musculoskeletal applications and the localized stimulated echo acquisition mode (STEAM). In this study, we aim to compare these three single-voxel 1H-CMRS techniques: PRESS, sLASER and STEAM for reproducible quantification of myocardial FA and Cr at 3 T. Sequences are compared both using breath-hold (BH) and free-breathing (FB) acquisitions. Methods: CMRS accuracy and theoretical CSDE were verified on a purposely-designed fat–water phantom. FA and Cr CMRS data quality and reliability were evaluated in the interventricular septum of 10 healthy subjects, comparing repeated BH and free-breathing with retrospective gating. Results: Measured FA/W ratio deviated from expected phantom ratio due to CSDE with all sequences. sLASER supplied the lowest bias (10%, vs -28% and 27% for PRESS and STEAM). In vivo, PRESS provided the highest signal-to-noise ratio (SNR) in FB scans (27.5 for Cr and 103.2 for FA). Nevertheless, a linear regression analysis between the two BH showed a better correlation between myocardial Cr content measured with sLASER compared to PRESS (r = 0.46; p = 0.03 vs. r = 0.35; p = 0.07) and similar slopes of regression lines for FA measurements (r = 0.94; p < 0.001 vs. r = 0.87; p < 0.001). STEAM was unable to perform Cr measurement and was the method with the lowest correlation (r = 0.59; p = 0.07) for FA. No difference was found between measurements done either during BH or FB for Cr, FA and triglycerides using PRESS, sLASER and STEAM. Conclusion: When quantifying myocardial lipids and creatine with CMR proton spectroscopy at 3 T, PRESS provided higher SNR, while sLASER was more reproducible both with single BH and FB scans. Seeds of Lithospermum officinale L. from different climatic zones were analyzed for new sources of γ‐linolenic acid (GLA, 18:3n‐6) and stearidonic acid (SDA, 18:4n‐3). Cultured Borago officinalis was also analyzed for comparative purposes. Analyses were conducted for fatty acid (FA) profiles of the glyceride oils from the seeds and in the neutral and polar lipids by gas chromatography (GC); lipid classes by open column chromatography and preparative thin layer chromatography (TLC); and tocopherols, sterols, and phenolic compounds by high‐performance liquid chromatography with diode array detection (HPLC‐DAD), and the later compounds were confirmed by liquid‐chromatography‐mass spectrometry (LC–MS). L. officinale from St. Petersburg Botanical Garden showed the highest percentage of GLA (17.9% of total FA), while wild‐growing L. officinale from the Rostov region contained the highest percentage of SDA (17.2% of total FA). Total FA content ranged from 11.3 to 20.8% of seed weight. Neutral and polar lipids accounted for ~98 and 2.27%, respectively, of total lipids. Five neutral lipid classes were identified (% of NL): triterpene esters (1.3), triacylglycerols (93.1), free FA (1.8), diacylglycerols (1.4), and monoacylglycerols (2.4). The highest tocopherol content was found in samples from Chechen Republic (35.7 mg/100 g), in which the δ isomer was the main component. Samples from the Rostov region had the highest amounts of sterols (83.8 mg/100 g), and Δ5‐avenasterol was the predominant compound in all samples. L. officinale seeds contain high amounts of phenolic compounds (389.9 mg/100 g as upper limit), in which rosmarinic acid is highlighted. Background: Mucor circinelloides WJ11 is a high-lipid producing strain and an excellent producer of γ-linolenic acid (GLA) which is crucial for human health. We have previously identified genes that encode for AMP-activated protein kinase (AMPK) complex in M. circinelloides which is an important regulator for lipid accumulation. Comparative transcriptional analysis between the high and low lipid-producing strains of M. circinelloides showed a direct correlation in the transcriptional level of AMPK genes with lipid metabolism. Thus, the role of Snf-β, which encodes for β subunit of AMPK complex, in lipid accumulation of the WJ11 strain was evaluated in the present study. Results: The results showed that lipid content of cell dry weight in Snf-β knockout strain was increased by 32 % (from 19 to 25 %). However, in Snf-β overexpressing strain, lipid content of cell dry weight was decreased about 25 % (from 19 to 14.2 %) compared to the control strain. Total fatty acid analysis revealed that the expression of the Snf-β gene did not significantly affect the fatty acid composition of the strains. However, GLA content in biomass was increased from 2.5 % in control strain to 3.3 % in Snf-β knockout strain due to increased lipid accumulation and decreased to 1.83 % in Snf-β overexpressing strain. AMPK is known to inactivate acetyl-CoA carboxylase (ACC) which catalyzes the rate-limiting step in lipid synthesis. Snf-β manipulation also altered the expression level of the ACC1 gene which may indicate that Snf-β control lipid metabolism by regulating ACC1 gene. Conclusions: Our results suggested that Snf-β gene plays an important role in regulating lipid accumulation in M. circinelloides WJ11. Moreover, it will be interesting to evaluate the potential of other key subunits of AMPK related to lipid metabolism. Better insight can show us the way to manipulate these subunits effectively for upscaling the lipid production. Adaptive laboratory evolution is a powerful tool for microorganism improvement likely to produce enhanced microalgae better tailored to their industrial uses. In this work, 12 wild-type strains of Tisochrysis lutea were co-cultivated under increasing thermal stress for 6 months. Indeed, temperature was oscillating daily between a high and a low temperature, with increasing amplitude along the experiment. The goal was to enhance the polyunsaturated fatty acid content of the polar lipids. Samples were taken throughout the evolution experiment and cultivated in standardized conditions to analyze the evolution of the lipid profile. Genomic analysis of the final population shows that two strains survived. The lipid content doubled, impacting all lipid classes. The fatty acid analyses show a decrease in SFAs correlated with an increase in monounsaturated fatty acids (MUFAs), while changes in polyunsaturated fatty acid (PUFAs) vary between both photobioreactors. Hence, the proportion of C18-MUFAs (18:1 n-9) and most C18-PUFAs (18:2 n-6, 18:3 n-3, and 18:4 n-3) increased, suggesting their potential role in adjusting membrane fluidity to temperature shifts. Of particular interest, DHA in polar lipids tripled in the final population while the growth rate was not affected. Differences in reproductive strategies of male and female fishes are presumably accompanied by differences in nutrient allocation and predicted to lead to divergence in body composition between the sexes. We compared patterns of variation in fatty acid profiles of lipids extracted from ova, liver, muscle and visceral fat between mature male and female walleye (Sander vitreus) sampled from two wild spawning stocks. Fatty acid profiles differed significantly among body tissues in both males and females, with the strongest contrast between muscle and visceral fat. Significant differences in fatty acid composition between the sexes were found in liver, muscle and visceral fat tissues. Variation among sexes and populations was greater in liver than in the other tissues. Female livers had lower relative abundances of palmitic acid (PA, 16:0) and oleic acid (OA, 18:1(n‐9)), and higher relative abundances of arachidonic acid (ARA, 20:4(n‐6)), eicosapentaenoic acid (EPA, 20:5(n‐3)) and docosahexaenoic acid (DHA, 22:6(n‐3)) compared to male livers. In addition, female muscle had higher relative abundance of OA and lower relative abundance of DHA compared to male muscle. Our results illustrate the differential effects of reproductive demands on the biochemical composition of males and females and have implications for the analysis of fatty acid profiles in studies of wild fish populations. Context: Evidence for an association between sphingolipids and metabolic disorders is increasingly reported. Omega-3 long-chain polyunsaturated fatty acids (n-3 LC-PUFAs) improve apolipoprotein B100 (apoB100)-containing lipoprotein metabolism, but their effects on the sphingolipid content in lipoproteins remain unknown.Objectives: In subjects with hypertriglyceridemia, we analyzed the effect of n-3 LC-PUFAs on the turnover apoB100-containing lipoproteins and on their sphingolipid content and looked for the possible association between these lipid levels and apoB100-containing lipoprotein turnover parameters.Methods: Six subjects underwent a kinetic study before and after n-3 supplementation for 2 months with 1 g of fish oil 3 times day containing 360 mg of eicosapentaenoic acid (EPA) and 240 mg of docosahexaenoic acid (DHA) in the form of triglycerides. We examined apoB100-containing lipoprotein turnover by primed perfusion labeled [5,5,5-2H3]-leucine and determined kinetic parameters using a multicompartmental model. We quantified sphingolipid species content in lipoproteins using mass spectrometry.Results: Supplementation decreased very low-density lipoprotein (VLDL), triglyceride, and apoB100 concentrations. The VLDL neutral and polar lipids showed increased n-3 LC-PUFA and decreased n-6 LC-PUFA content. The conversion rate of VLDL1 to VLDL2 and of VLDL2 to LDL was increased. We measured a decrease in total apoB100 production and VLDL1 production. Supplementation reduced the total ceramide concentration in VLDL while the sphingomyelin content in LDL was increased. We found positive correlations between plasma palmitic acid and VLDL ceramide and between VLDL triglyceride and VLDL ceramide, and inverse correlations between VLDL n-3 LC-PUFA and VLDL production.Conclusion: Based on these results, we hypothesize that the improvement in apoB100 metabolism during n-3 LC-PUFA supplementation is contributed to by changes in sphingolipids.
