In tropical Africa, wild meat consumption by rural communities who live in or near protected areas can be a major challenge in the conservation of threatened species. Lemurs and the fossa are among the species consumed in Makira Natural Park, Madagascar. Because of their low reproductive rates and the high human population density, these species are known to be hunted unsustainably. We estimated the prevalence of lemur and fossa meat consumption in villages within and around Makira Natural Park using an indirect questioning method, the unmatched count technique (UCT), and compared it with results from direct questioning. The UCT revealed that 53.0% of households had eaten lemur meat over the previous year (95% confidence interval [CI]: 42.0–64.1%) and 24.2% had eaten fossa meat (95% CI: 14.2–34.2%). The percentage of households that ate lemur meat estimated through the UCT was more than 3.3 times higher than that obtained from direct questioning, and 12 times higher for fossa. Our estimates will be used to define the scale needed for a behavior change campaign to be conducted in the study area and will serve as a baseline to assess the impact of conservation activities aimed at reducing the consumption of lemurs and fossas. In wild primates, social behaviour influences exposure to environmentally acquired and directly transmitted microorganisms. Prior studies indicate that gut microbiota reflect pairwise social interactions among chimpanzee and baboon hosts. Here, we demonstrate that higher-order social network structure--beyond just pairwise interactions--drives gut bacterial composition in wild lemurs, which live in smaller and more cohesive groups than previously studied anthropoid species. Using 16S rRNA gene sequencing and social network analysis of grooming contacts, we estimate the relative impacts of hierarchical (i.e. multilevel) social structure, individual demographic traits, diet, scent-marking, and habitat overlap on bacteria acquisition in a wild population of Verreaux's sifaka (Propithecus verreauxi) consisting of seven social groups. We show that social group membership is clearly reflected in the microbiomes of individual sifaka, and that social groups with denser grooming networks have more homogeneous gut microbial compositions. Within social groups, adults, more gregarious individuals, and individuals that scent-mark frequently harbour the greatest microbial diversity. Thus, the community structure of wild lemurs governs symbiotic relationships by constraining transmission between hosts and partitioning environmental exposure to microorganisms. Over the years, we have encountered situations requiring caretakers or veterinarians intervention, to save monkeys abandoned by their parents. There are situations that require helping the female to care for the baby monkey (supplementing the food without removing the baby monkey from the location), but also its takeover to be raised artificially, outside the group. The medical problems commonly encountered in lemur baby are hypoglycemia, hypothermia and dehydration. In such cases, lemur baby are lethargic and anorectic. In the first 72 hours, the mortality rate is very high. Recognition and correction of the perinatal problems can prevent long-term intervention. The milk formula used differs, depending on the species. In our country, in recent years there has been an increase in the number of Callitrichidae kept as pets. Unfortunately, due to improper maintenance, poor nutrition, lack of ultraviolet rays, optimal temperature and humidity gradients, there can be observed numerous conditions that affect the quality of life of these animals. Subspecies were traditionally defined by identifying gaps between phenotypes across the geographic range of a species, and may represent important units in the development of conservation strategies focused on preserving genetic diversity. Previous taxonomic research proposed that phenotypic variation between scattered Indri indri populations warranted the naming of two distinct subspecies, I. i. indri and I. i. variegatus. We tested these subspecific designations using mitochondrial sequence data generated from the control region or D-loop (569 bp) and a large section (2362 bp) of multiple genes and tRNAs known as Pastorini's fragment and nuclear microsatellite markers. This study used 114 samples of I. indri from 12 rainforest sites in eastern Madagascar, encompassing the entire range of the species. These genetic samples represent multiple populations from low- and high-elevation forests from both putative subspecies. Molecular analyses of the mitochondrial sequence data did not support the two proposed subspecies. Furthermore, the microsatellite analyses showed no significant differences across the range beyond population level differentiation. Genetic data can be combined with ecological data to study the demographic history of a species, identify landscape features that influence migration patterns, and guide conservation efforts. Perrier's sifaka ( Propithecus perrieri) is a Critically Endangered, rare, and elusive social lemur living in a very restricted, fragmented landscape. To assess the effect of habitat loss and fragmentation on the genetic diversity of the Perrier's sifaka population we examined 24 microsatellite markers genotyped for 67 samples corresponding to 42 individuals. Perrier's sifaka shows a low current effective population size ( ca. 52-105) and a strong heterozygosity excess, suggesting a historically large but dwindling population. Moreover, we identified a pattern of isolation by distance, typical of continuous habitat, suggesting that sifakas were still able to cross the grasslands between forest fragments in the recent past. Our study calls for a unified conservation plan covering the two protected areas where the species is still present, if not the whole area of its past distribution. ABSTRACT There are at present few comparable studies of lemur locomotion in the wild. This has unfortunately meant we have little knowledge about locomotor variation, and hence flexibility, with regard to differences in support availability and habitat structure. Here we compare the locomotion of Lepilemur edwardsi at Ankarafantsika with that of Lepilemur ruficaudatus at Kirindy-Mitea National Park. While data were collected by two individuals, at different times, both studies used the same data collection protocol and are hence highly comparable. Locomotor mode, support diameter and orientation, heights, and distances traveled were all collected. We find that locomotor specialization, in this case for vertical leaping, has ensured that some support requirements are independent of habitat. For example, both species used vertical supports most often. However, overall support diameter does indicate a certain degree of flexibility, whereby L. ruficaudatus most often used supports ≤5 cm in diameter and L. edwardsi >5 cm in diameter. A functional analysis was conducted to assess the antecedent and reinforcing conditions underlying aggressive behavior in a female lemur in captivity. Results showed that her aggression was primarily the result of human attention. A replacement behavior-training program was introduced, and the lemur's aggression was successfully eliminated. Understanding ecological niche evolution over evolutionary timescales is crucial to elucidating the biogeographic history of organisms. Here, we used, for the first time, climate-based ecological niche models ( ENMs) to test hypotheses about ecological divergence and speciation processes between sister species pairs of lemurs (genus Eulemur) in Madagascar. We produced ENMs for eight species, all of which had significant validation support. Among the four sister species pairs, we found nonequivalent niches between sisters, varying degrees of niche overlap in ecological and geographic space, and support for multiple divergence processes. Specifically, three sister-pair comparisons supported the null model that niches are no more divergent than the available background region. These findings are consistent with an allopatric speciation model, and for two sister pairs ( E. collaris- E. cinereiceps and E. rufus- E. rufifrons), a riverine barrier has been previously proposed for driving allopatric speciation. However, for the fourth sister pair E. flavifrons- E. macaco, we found support for significant niche divergence, and consistent with their parapatric distribution on an ecotone and the lack of obvious geographic barriers, these findings most strongly support a parapatric model of speciation. Abstract: Deforestation and a changing climate threaten the health and survival of lemurs in Madagascar. An important component of lemur health, parasite infection can reduce fitness and survival outcomes. Future lemur parasite richness, abundance and distribution may be highly influenced by climate change. Current knowledge of lemur parasites is narrow in geographic and temporal scope, with sampling at a limited number of sites, and thus far, there have been no attempts to assess the effects of climate change on lemur parasite distributions. We used geospatial tools to predict the distributions of six lemur parasites of high frequency and pathogenic potential. We then assessed how anticipated climate shifts in Madagascar may alter the distributions of these lemur parasites in the future. Under current climate conditions, we found that the focal parasites exhibited widespread potential distributions across Madagascar, covering 12–26% of surface land area and 40–86% of forested area. Our analyses also showed that parasites responded differently to projected climate changes, with shifts ranging from a contraction of current distributions by 7% to an expansion of 60%. A predicted net expansion in parasite distribution may expose naive lemur hosts to new parasites, which could have a profound effect on lemur health. Those parasites with the greatest potential for harmful effects are predicted to experience the largest expansion in range. Predicting these changing distributions will be critical for assessing population health, improving protected area design, preparing for reintroduction efforts and addressing potential parasite risk in lemurs, humans and domestic animals. Metabolic costs of resting and locomotion have been used to gain novel insights into the behavioral ecology and evolution of a wide range of primates; however, most previous studies have not considered gait-specific effects. Here, metabolic costs of ring-tailed lemurs ( Lemur catta) walking, cantering and galloping are used to test for gait-specific effects and a potential correspondence between costs and preferred speeds. Metabolic costs, including the net cost of locomotion (COL) and net cost of transport (COT), change as a curvilinear function of walking speed and (at least provisionally) as a linear function of cantering and galloping speeds. The baseline quantity used to calculate net costs had a significant effect on the magnitude of speed-specific estimates of COL and COT, especially for walking. This is because non-locomotor metabolism constitutes a substantial fraction (41-61%, on average) of gross metabolic rate at slow speeds. The slope-based estimate of the COT was 5.26 J kg−1 m−1 for all gaits and speeds, while the gait-specific estimates differed between walking (0.5 m s−1: 6.69 J kg−1 m−1) and cantering/galloping (2.0 m s−1: 5.61 J kg−1 m−1). During laboratory-based overground locomotion, ring-tailed lemurs preferred to walk at ∼0.5 m s−1 and canter/gallop at ∼2.0 m s−1, with the preferred walking speed corresponding well to the COT minima. Compared with birds and other mammals, ring-tailed lemurs are relatively economical in walking, cantering, and galloping. These results support the view that energetic optima are an important movement criterion for locomotion in ring-tailed lemurs, and other terrestrial animals. The aye-aye is a rare lemur from Madagascar that uses its highly specialized middle digit for percussive foraging. This acoustic behavior, also termed tap-scanning, produces dominant frequencies between 6 and 15 kHz. An enhanced auditory sensitivity to these frequencies raises the possibility that the acoustic and auditory specializations of aye-ayes have imposed constraints on the evolution of their vocal signals, especially their primary long-distance vocalization the screech. Here we explore this concept termed receiver bias, and suggest that the dominant frequency of the screech call (~2.7 kHz) represents an evolutionary compromise between the opposing adaptive advantages of long-distance sound propagation and enhanced detection by conspecific receivers. We present a high-coverage draft genome assembly of the aye-aye (Daubentonia madagascariensis), a highly unusual nocturnal primate from Madagascar. Our assembly totals ∼3.0 billion bp (3.0 Gb), roughly the size of the human genome, comprised of ∼2.6 million scaffolds (N50 scaffold size = 13,597 bp) based on short paired-end sequencing reads. We compared the aye-aye genome sequence data with four other published primate genomes (human, chimpanzee, orangutan, and rhesus macaque) as well as with the mouse and dog genomes as nonprimate outgroups. Unexpectedly, we observed strong evidence for a relatively slow substitution rate in the aye-aye lineage compared with these and other primates. In fact, the aye-aye branch length is estimated to be ∼10% shorter than that of the human lineage, which is known for its low substitution rate. This finding may be explained, in part, by the protracted aye-aye life-history pattern, including late weaning and age of first reproduction relative to other lemurs. Additionally, the availability of this draft lemur genome sequence allowed us to polarize nucleotide and protein sequence changes to the ancestral primate lineage—a critical period in primate evolution, for which the relevant fossil record is sparse. Finally, we identified 293,800 high-confidence single nucleotide polymorphisms in the donor individual for our aye-aye genome sequence, a captive-born individual from two wild-born parents. The resulting heterozygosity estimate of 0.051% is the lowest of any primate studied to date, which is understandable considering the aye-aye's extensive home-range size and relatively low population densities. Yet this level of genetic diversity also suggests that conservation efforts benefiting this unusual species should be prioritized, especially in the face of the accelerating degradation and fragmentation of Madagascar's forests. Abstract: Several recent studies of animals in their natural surroundings found evidence for effects of certain major histocompatibility complex (MHC) immune gene alleles on the parasite load. However, in multi-infected individuals the particular selection pressure exerted by specific parasites has rarely been explored. In this study we took advantage of the parasitological and genetic data of two previously investigated Malagasy lemur species (Cheirogaleus medius and Microcebus murinus). We investigated whether the two sympatric and ecologically similar primates are infected by similar parasite species and explored if certain parasites are associated with particular MHC alleles. Our study revealed that most of the parasite egg morphotypes were found in both hosts. In each lemur species we identified one MHC allele which was positively associated with Ascaris-infection. Interestingly, these MHC alleles were very similar to each other but differed from all other investigated MHC alleles in an amino acid substitution in a putative functional important antigen binding site. Thus, our study gives first intriguing evidence for a direct connection between certain antigen binding sites of MHC molecules with a particular parasite in two wild primate populations. This may indicate that indeed certain parasites exert direct selective pressure on the MHC of wild living hosts. [Copyright &y& Elsevier]

We assessed the consumption and hunting of wild animals by people in urban areas of western Madagascar using structured questionnaires in households and direct observations. Six wild mammal and five wild bird species were reported, or observed, to be sources of bushmeat although fish and domestic animals were the preferred and cheapest sources of animal protein. Bushmeat accounted for 10% of the meat consumed the day before our questionnaires were completed. Common tenrec Tenrec ecaudatus and bush pig Potamochoerus larvatus were the preferred wild meat and the former was also the most expensive type of meat. Taboos and strong dislikes limited the consumption of domestic pigs, bush pigs, goats, lemurs and fruit bats. Game species were hunted according to their availability, which coincided with the legal hunting season for fruit bats but only partly so for the other game species. Illegal hunting of Verreauxi's sifaka Propithecus verreauxi is cause for concern and assessments of primate consumption may have been underestimated because of reluctance of interviewees to admit illegal activities. Three brown lemurs, Eulemur fulvus, and two black lemurs, Eulemur macaco, were trained to select consistently the photograph of the face of a known individual (KI: belonging to the same social group) when it was presented simultaneously with a photograph of the face of an unfamiliar individual (UI: belonging to another social group). After the learning phase, the subjects were presented with new pairs of photographs (KI versus UI: photographs that were not used in the learning phase), the same pairs presented in different associations, and finally pairs of photographs that were completely novel (50 different pairs). All the subjects selected the KI significantly more often than the UI under all conditions. Additional tests showed that the subjects preferentially selected photographs of a UI belonging to their own species versus a UI belonging to the other species. No preference was observed when the subjects were presented with two KI or with their own photograph associated with a UI. These results reveal that the face contains information allowing lemurs to discriminate familiar individuals from unfamiliar ones. [Copyright &y& Elsevier]

Habitat fragmentation may strongly reduce individuals’ dispersal among resource patches and hence influence population distribution and persistence. We studied the impact of landscape heterogeneity on the dispersal of the golden-crowned sifaka ( Propithecus tattersalli), an endangered social lemur species living in a restricted and highly fragmented landscape. We combined spatial analysis and population genetics methods to describe population units and identify the environmental factors which best predict the rates and patterns of genetic differentiation within and between populations. We used non-invasive methods to genotype 230 individuals at 13 microsatellites in all the main forest fragments of its entire distribution area. Our analyses suggest that the Manankolana River and geographical distance are the primary structuring factors, while a national road crossing the region does not seem to impede gene flow. Altogether, our results are in agreement with a limited influence of forest habitat connectivity on gene flow patterns (except for North of the species’ range), suggesting that dispersal is still possible today among most forest patches for this species. Some species have potential for intense mate competition yet exhibit little or no sexual size dimorphism, despite predictions from sexual selection theory. Using a conceptual model, we show the conditions for which passive mate guarding with copulatory plugs can be an alternative and more successful strategy to active (direct) guarding, reducing selection pressure on large male size. The model predicts that copulatory plugs in mammals should be favoured in species for which females have short sexual receptivity periods. Using data on 62 primate species and a phylogenetic regression approach, we show that, as predicted, copulatory plugs are negatively associated with degree of sexual dimorphism and females’ sexual receptivity length. Penile spines are also significantly associated with plug use and short receptivity periods suggesting a possible offensive role in sperm competition. Results highlight that life-history characteristics, such as sexual receptivity lengths, may alter the costs and benefits of alternative male strategies and thus alter the strength of sexual selection. Olfactory signals can communicate a wide variety of information and are very important in many mammalian species. However, little is known about the olfactory communication of primates. This study used gas chromatography to examine the volatile components of the anogenital gland secretions of wild Milne-Edward's sifaka, Propithecus edwardsi (Primates, Indriidae) (n = 17), captured in Ranomafana National Park, Madagascar. We compared scent swabs of animals and examined differences between social group, age class, sex, and season. The only identified differences in the volatiles were between swabs from the different seasons; all other categories were statistically indistinguishable. Am. J. In many primate species, hands and feet are large relative to neonatal body weight, and they subsequently exhibit negative allometric growth during ontogeny. Here, data are presented showing that this pattern holds for a wild population of lemur, Verreaux's sifaka (Propithecus verreauxi verreauxi). Using morphometric data collected on this population, it is shown that younger animals possess relatively large hands and feet. This ontogenetic pattern suggests a simple behavioral test: do juvenile animals with their larger, almost adult-sized hands and feet locomote on similarly sized substrates as adult animals? Using locomotor bout sampling, this question was tested by collecting positional behavior data on this population. Results from this test find no differences in locomotor behaviors or substrate use between yearlings and adult animals. To place these results in a broader evolutionary context, heritabilities and selection gradients of hands, feet, and other limb elements for animals in this population were estimated. Among limb elements, heritabilities range from 0.16- 0.44, with the foot having the lowest value. Positive directional selection acts most strongly on the foot (directional selection gradient = 0.119). The low heritability and positive selection coefficient indicate that selection has acted, and continues to act, on foot size in young animals. These results are interpreted within a functional context with respect to the development of locomotor coordination: larger feet enable young animals to use "adult-sized" substrates when they move through their habitat. It is suggested that the widespread pattern of negative allometry of the extremities in sifaka and other primates is maintained by selection, and does not simply reflect a primitive developmental pathway that has no adaptive basis. Dispersed pair-living primates provide a unique model for illuminating the evolution of mechanisms regulating spacing and cohesiveness in permanently cohesive groups. We present for the first time data on the spatiotemporal distribution and loud-calling behavior of the Milne Edwards' sportive lemur, known to forage solitarily during the night, but to form stable male-female sleeping groups during the day. Data include radio-tracking observations of sleeping associations, and focal follows of pair partners during dispersal in the evenings and reunions in the mornings. Male-female pairs forming stable sleeping associations during the day were pair-bonded. They used sleeping sites and home ranges exclusively, and exchanged loud calls at potentially restricted resources during dispersal in the evenings and during reunion in the mornings. Direct agonistic conflicts between pairs and others were rare. The acoustic analysis of loud calls revealed nine major call types. They carry signatures for sex and pair identity, and provide the substrate for signaling and the potential for recognizing pair ownership. Thus, pairs use loud call exchanges as a vocal display for signaling territory ownership, thus limiting direct aggressive encounters between neighbors and strangers. Altogether, our findings provide the first empirical evidence for the hypothesis that loud calling has evolved as a key mechanism for regulating space use and cohesiveness in dispersed pair-living primates. Although prosimians are greatly olfaction-oriented, little is known about the specifics of how they use scent to communicate. In this preliminary study we attempted to delineate intra- and interspecific differences among the anogenital gland secretions of two lemur species (Lemur catta and Propithecus verreauxi coquereli) using gas chromatography-mass spectrometry (GC-MS). The results indicate that the two species are discernible through scent. Furthermore, we were able to identify reproductive status using this technique. The anogenital secretions of the different sexes in L. catta, though perhaps not P. v. coquereli, are chemically distinguishable. Abstract Gene flow within and between social groups is contingent on behaviourally mediated patterns of mating and dispersal. To understand how these patterns affect the genetic structure of primate populations, long-term data are required. In this study, we analyse 10 years of demographic and genetic data from a wild lemur population (Propithecus verreauxi verreauxi ) at Beza Mahafaly Special Reserve, southwest Madagascar. Our goal is to specify how patterns of mating and dispersal determine kinship and genetic diversity among animals in the population. Specifically, we use microsatellite, parentage, and census data to obtain estimates of genetic subdivision (F [sub ST] ), within group homozygosity (F [sub IS] ), and relatedness (r ) within and among social groups in the population. We analyse different classes of individuals (i.e. adults, offspring, males, females) separately in order to discern which classes most strongly influence aspects of population structure. Microsatellite data reveal that, across years, offspring are consistently more heterozygous than expected within social groups (F [sub IS] mean = -0.068) while adults show both positive and negative deviations from expected genotypic frequencies within groups (F [sub IS] mean = 0.003). Offspring cohorts are more genetically subdivided than adults (F [sub ST] mean = 0.108 vs. 0.052) and adult females are more genetically subdivided than adult males (F [sub ST] mean = 0.098 vs. 0.046). As the proportion of females in social groups increases, the proportion of offspring sired by resident males decreases. Offspring are characterized by a heterozygote excess as resident males (vs. nonresident males) sire the majority of offspring within groups. We link these genetic data to patterns of female philopatry, male dispersal, exogamy, and offspring sex-ratio. Sixteen dinucleotide microsatellite loci were isolated from the genome of Propithecus verreauxi verreauxi . All loci were polymorphic when genotyped on a minimum of 16 animals. The number of alleles across these loci ranges from two to 11. Additionally, seven of these loci were genotyped across a minimum of 200 animals in order to estimate heterozygosity and their potential for parentage assignment in this population. Using these seven loci, the mean heterozygosity in this population is 0.705, and the combined probability of these seven loci to exclude a random individual from parentage, when one parent is known, is 0.996. These data suggest that these loci will be useful for estimating a variety of population genetic and genealogical parameters in P. v. verreauxi populations. Am. J. Tight energetic constraints on reproductively active females are hypothesized to be an important determinant of the phenomenon of female dominance in Propithecus verreauxi, a primate endemic to Madagascar. Five wild sifakas were captured in the Beza Mahafaly Special Reserve in southern Madagascar, and resting metabolic rates (RMR) were measured. A cytogenetical study of the western form of Avahi (Avahi laniger occidentalis) shows that its karyotype differs from that of A. laniger laniger by at least 3 chromosomal rearrangements. Meiotic chromosomal pairing of hypothetical hybrids has been constructed. Its configuration, a tetravalent with an inversion loop, is likely to strongly reduce fertility, especially in the male. This leads us to propose that the two forms correspond to two distinct species, separated by a gametic barrier: Avahi laniger and a species apart, Avahi occidentalis. [ABSTRACT FROM AUTHOR]

In this report we describe the development and characterization of six dinucleotide microsatellite loci for the ring-tailed lemur ( Lemur catta). The new markers were isolated from the genomic DNA of L. catta and all loci were found to be polymorphic when evaluated by genotyping a minimum of 34 individuals. Mendelian inheritance of each locus was verified using four families of captive ring-tailed lemurs.
