Psoraleeae consist of c. 228 species in ten genera, distributed worldwide but mostly in the temperate biome. Here, we reconstruct the phylogenetic history of Psoraleeae using eight molecular markers (rpl32-trnL , trnQ-rps16 , trnS-trnG , matK , rbcLa , rpoB-trnC , trnL-trnF and ITS) to test the monophyly and previous hypotheses on generic relationships and to reconstruct the biogeographical history (distribution pattern, potential ancestral area and biome). The monophyly of Psoraleeae is supported. The African Otholobium and Psoralea form a strongly supported clade that is sister to the rest of Psoraleeae. However, Otholobium is not monophyletic; the American taxa of the genus are resolved as closer to Bituminaria , whereas Psoralea is a clade derived from within African Otholobium. South African Otholobium is subsumed into Psoralea , the latter having priority, with a synopsis of species level nomenclature provided here. An expanded sampling of Cullen reveals three clades comprising Mediterranean (Cullen americanum), African (Cullen obtusifolium and Cullen biflorum) and Eurasian (Cullen corylifolium and Cullen drupaceum) taxa, whereas Australian Cullen are a derived clade (BS = 95%, PP = 1.00). Biogeographic reconstructions show that Psoraleeae probably originated in the Mediterranean south biome of Africa in the Late Miocene, c. 8.2 Mya, and then dispersed into the succulent biome of the Americas, Mediterranean north biome, temperate grasslands of Africa and Australia to the temperate forest of Eurasia. Climate changes in the Late Miocene, changes induced by ocean currents during the Quaternary and environmental heterogeneity are proposed as the major drivers of speciation in Psoraleeae. Fabaceae plants contribute greatly to a high quality of pasture in rangelands, because of high levels of protein in their tissues. Despite this established importance, only a few southern African legumes have been evaluated for nutritional value and domestication. The Overberg renosterveld vegetation of the Fynbos biome occupies undulating landscapes that have multiple slope aspects, creating variable microclimatic conditions affecting nutrient dynamics, species richness and plant chemical compositions. We assessed the effect of slope aspect on fiber and mineral composition of forage species present to provide information on the importance of slope for species characterization and nutritional value for domestication. Tissue samples from six replicates of the same species occurring on opposing aspects were collected for nutritional assessment. Results on the data showed that fibre and mineral composition differed in species occurring on different slopes, albeit with few exceptions. The concentrations of P, K and Mg in some species were significantly higher on the cooler and wetter south facing slopes relative to north facing slopes. Nutrient elements in soil samples collected from each site varied with slope aspects and elements. In this study, Indigofera wenholdiae, a new species of Fabaceae from the Agulhas Plain Region of the Western Cape Province, South Africa, is described. A composite photographic plate is included along with a distribution map, description of habitat and ecology and proposed IUCN conservation status. Indigofera wenholdiae is unique in the I. brachystachya group by having digitately compound (vs. pinnately compound) leaves, white and unscented flowers (vs. pink and sweetly scented flowers) and grows on sandstone hillsides (vs. coastal limestone plains and outcrops). Aims The Cape Peninsula is a small area (471 km2) situated on the south-westernmost tip of the Core Cape Subregion (CCR) of South Africa. Within the Cape Peninsula, Fabaceae are the third most species-rich plant family (162 species) and they have the second highest number of endemic species after the Ericaceae. However, legumes are not the dominant taxa in the vegetation. They tend to show patchy distributions within the landscape and different species assemblages usually occupy particular niches at any given locality. The present study undertook to establish if edaphic factors influence legume species distribution in the Cape Peninsula and to determine the key indicator species for the different assemblages. Methods Soils from 27 legume sites, spanning all major geological substrates of the Cape Peninsula, were analysed for 31 chemical and physical properties. Legume species present at each site were recorded and a presence/absence matrix was generated. Cluster analysis and discriminant function analysis (DFA) were run to group the sites based on overall similarity in edaphic characteristics and to identify the soil parameters contributing towards discriminating the groups. Canonical correspondence analysis (CCA) was used to test for a correlation between legume species compositions and edaphic factors. The strength of the association between legume species and site groupings based on edaphic properties was assessed using indicator species analysis. Important findings Based on similarity in overall soil characteristics, the sites formed three clusters: one comprising sites of sandstone geology, one with dune sand sites and the third cluster comprising sites of both shale and granite geologies (hereafter referred to as soil types). The DFA confirmed the distinctness of these clusters and the CCA showed a significant correlation between legume species composition and edaphic factors. The key edaphic parameters were clay content, iron (Fe), potassium (K), sulphur (S) and zinc (Zn). These findings reveal that the Cape Peninsula is edaphically heterogeneous and edaphically distinct habitats contain discrete legume species assemblages that can be distinguished by unique indicator species. Furthermore, multiple soil parameters, rather than a single parameter, are involved. Therefore, edaphic factors play a significant role in driving the distribution of legume species in the Cape Peninsula and discrete legume species assemblages occupy distinct habitats. Psoralea forbesiae C.H.Stirt., A.Bello & Muasya is a new species of Psoraleeae, Fabaceae. Psoralea forbesiae is endemic to the Swartberg Mountains and is a tall densely branched re-sprouting shrub up to 2.5 m, with bluish-green stems and with most parts covered in small crater-like glands, leaves pinnately 3-foliolate, linear-oblong, pale bluish-green, semi-conduplicate, somewhat succulent, glabrous, crowded at the end of bare branches on older stems or distributed along short branches on young shoots, petiolate. The South African Fynbos soils are renowned for nitrogen-fixing Burkholderia associated with diverse papilionoid legumes of the tribes Crotalarieae, Hypocalypteae, Indigofereae, Phaseoleae and Podalyrieae. However, despite numerous rhizobial studies in the region, the symbiotic diversity of Burkholderia has not been investigated in relation to a specific host legume and its geographical provenance. This study analyzed the diversity of nodulating strains of Burkholderia from the legume species Podalyria calyptrata . Diverse lineages were detected that proved to be closely related to Burkholderia taxa, originating from hosts in other legume tribes. By analyzing the genetic variation of chromosomal ( recA ) and nodulation ( nodA ) sequence data in relation to the sampling sites we assessed the geographical distribution patterns of the P. calyptrata symbionts. Although we found a degree of genetically differentiated rhizobial populations, a correlation between genetic ( recA and nodA ) and geographic distances among populations was not observed, suggesting high rates of dispersal and rhizobial colonization within Fynbos soils. Podalyria calyptrata is from fynbos soils with low availability of phosphorus (P) and nitrogen (N). We investigated the physiological basis for tolerance of low P supply in nodulated P. calyptrata and examined responses to increased supply of combined-N as Ca(NO 3 ) 2 and P. It was hypothesized that increasing supply of combined-N would stimulate P-acquisition mechanisms and enhance plant growth with high P supply. Biomass, leaf [N] and [P], organic acid and phosphatase root exudates, and phosphoenolpyruvate carboxylase (PEPC) and malate dehydrogenase (MDH) activity in nodules and roots were examined in two N × P experiments. Low P supply decreased leaf [P] and limited growth, decreasing the nodule:root ratio but increasing nodular PEPC and MDH activity for enhanced P-acquisition or P-utilization. At low P supply, a N-induced demand for P increased root exudation of citrate and PEPC and MDH activity in roots. Greater combined-N supply inhibited nodulation more at low P supply than at high P supply. With a P-induced demand for N the plants nodulated prolifically and increased combined-N supply did not enhance plant growth. The physiological basis for N 2 -fixing P. calyptrata tolerating growth at low P supply and responding to greater P supply is through balanced acquisition of P and N for plant demand. A new species of Psoralea is described. Psoralea karooensis C.H. Stirt., Muasya & Vlok is endemic to mountain streams in the Klein Karoo region of the Western Cape Province, South Africa. The new species is characterised by its flexuose habit of many stiff bare stems with the seasonal shoots arising apically in clusters and its greenish cream flowers borne at the apex of 10--12 mm long peduncles each ending in a trifid cupulum. A new species of Psoralea is described. Psoralea margaretiflora C.H. Stirton & V.R. Clark is endemic to the Sneeuberg Centre of Floristic Endemism, Eastern Cape, South Africa. his resprouter is characterised by its small greenish-white flowers with a small trifid purple nectar patch and translucent veins; 5(-7)-pinnate leaflets; multi-branching erect short seasonal flowering shoots; and tall habit of many stiff bare stems with the seasonal shoots massed at the apex. It is most similar to P. oligophylla Eckl. & Zeyh., a widespread species found in the Eastern Cape. The reseeder P. oligophylla differs in its lax virgate spreading habit with numerous long glaucous seasonal shoots; single stem, 1(-3)- glaucous leaflets; more numerous white flowers; and standard petals with a purple ring surrounding a bright yellow nectar patch. Rhizobia of the genus Burkholderia have large-scale distribution ranges and are usually associated with South African papilionoid and South American mimosoid legumes, yet little is known about their genetic structuring at either local or global geographic scales. To understand variation at different spatial scales, from individual legumes in the fynbos (South Africa) to a global context, we analyzed chromosomal (16S rRNA, recA) and symbiosis (nifH, nodA, nodC) gene sequences. We showed that the global diversity of nodulation genes is generally grouped according to the South African papilionoid or South American mimosoid subfamilies, whereas chromosomal sequence data were unrelated to biogeography. While nodulation genes are structured on a continental scale, a geographic or host-specific distribution pattern was not detected in the fynbos region. In host range experiments, symbiotic promiscuity of Burkholderia tuberum STM678T and B. phymatum STM815T was discovered in selected fynbos species. Finally, a greenhouse experiment was undertaken to assess the ability of mimosoid (Mimosa pudica) and papilionoid (Dipogon lignosus, Indigofera filifolia, Macroptilium atropurpureum, and Podalyria calyptrata) species to nodulate in South African (fynbos) and Malawian (savanna) soils. While the Burkholderia-philous fynbos legumes (D. lignosus, I. filifolia, and P. calyptrata) nodulated only in their native soils, the invasive neotropical speciesM. pudica did not develop nodules in the African soils. The fynbos soil, notably rich in Burkholderia, seems to retain nodulation genes compatible with the local papilionoid legume flora but is incapable of nodulating mimosoid legumes that have their center of diversity in South America. Abstract: The tribe Psoraleeae (Leguminosae subfamily Papilionoideae) comprises 185 species in nine genera that have a nearly worldwide distribution, occurring predominantly in Mediterranean regions. About 60% of the species belong to the genera, Otholobium C.H.Stirt. and Psoralea L., which have a centre of diversity in the Cape Floristic Region of South Africa. Since previous molecular studies have sampled only a few species of the tribe from this region, this study sought to determine the phylogenetic position of the southern African genera and to test whether they are monophyletic. Phylogenetic relationships were reconstructed using DNA sequence data (trnL-F, rpoB-trnC and ITS) and seven morphological characters, which diagnose the two southern African genera. The data were analysed using the parsimony method. There was strong support for the Psoraleeae as a clade, but most of the nodes within the large genera were poorly supported. The southern African species of Psoralea and Otholobium together formed a strongly supported clade. This clade was sister to the genus Hoita Rydb., but without support. However, the Psoralea species were nested within the southern African Otholobium. Additionally, some South American species that are currently recognised as Otholobium were resolved in a clade distinct from the southern African species, making Otholobium polyphyletic. Morphological characters that separate Otholobium and Psoralea are discussed. Finally, the southern African genera as currently circumscribed are not monophyletic. However, further investigations using more informative DNA loci are required to validate this observation. Furthermore, the taxonomic placement of the South American species needs to be reviewed. The limited grazeable size of renosterveld vegetation in the Western Cape warranted assessment of chemical and fibre composition of the species to determine their nutritive value. We hypothesised that Fabaceae has chemically superior palatable species relative to Poaceae, Asteraceae and Asparagaceae species in the area. Plant samples were collected in the Overberg region from 11 sites. The species sampled from these four families Fabaceae, Poacea e , Asteraceae, and Asparagaceae were analysed for macro- and micro-elements, fibre and secondary compounds. At each site, replicated soil samples were also collected and analysed for pH, macro- and micro-elements. Fabaceae recorded the greatest concentration of crude protein, but the Poaceae was the most fibrous relative to Fabaceae and Asteraceae. All the species contained adequate levels of nutrients except P, Zn and sometimes Cu. Soil concentration of macronutrients and carbon in the soil varied with location. Based on crude protein, neutral detergent fibre and digestibility, the three most important parameters defining forage quality, the Fabaceae species Aspalathus hispida , A. angustifolia , A. nigra and A. submissa were regarded as superior forage species in the Mediterranean renosterveld. Livestock relying on only the renosterveld species in the Western Cape as forages should receive P and Zn supplements. • Fabaceae recorded greatest concentrations of crude protein and medium to low fibre content. • All the species contained adequate levels macro- and micro-nutrients except P, Zn and sometimes Cu. • Total phenolic and condensed tannins were low in all species irrespective of plant family. • Livestock relying on only the renosterveld species in the Western Cape as forages should receive P and Zn supplements. • The Fabaceae species A. hispida , A. angustifolia , A. nigra and A. submissa were superior forage species in renosterveld.
