مقدمه: سرطان‌های بدخیم کولورکتال، چه در صورت تکی و چه در صورت خانوادگی، اگر با افزایش میزان ناپایداری در میکروستلایت همراه باشند، پیش‌آگهی بهتری دارند، راهکار متفاوتی برای جراحی در آن‌ها اتخاذ می‌شود و پاسخ متفاوتی نسبت به داروی شیمی‌درمانی 5FU (Fluorouracil) خواهند داد. بنا بر این، امروزه برای بیماران مبتلا به سرطان کولورکتال ارثی و غیر ارثی، آزمایش سنجش وضعیت ناپایداری میکروستلایت انجام می‌شود. هدف این مطالعه، سنجش کارایی نشانگر تک نوکلئوتیدی MT1XT20 در جمعیت بیماران مبتلا به سرطان کولون ارثی غیر پولیپی (HNPCC یا Hereditary nonpolyposis colorectal cancer) کشور ایران بود. علاوه بر نشانگر پیش‌گفته، از دو روش بررسی وضعیت میکروستلایت با پنل 5 نشانگری و استاندارد Promega و نیز آزمایش ایمنوهیستوشیمیایی استفاده شد. روش‌ها: در این مطالعه، نشانگر تک نوکلئوتیدی MT1XT20 و کیت استاندارد Promega از طریق روش PCR (Polymerase chain reaction) بر روی 20 خانواده‌ی مبتلا به HNPCC ایرانی مورد سنجش قرار گرفت و آزمایش ایمنوهیستوشیمیایی نیز برای بررسی وضعیت 4 پروتئین مهم دخیل در مسیر ترمیم ناجور جفت شدگی مورد استفاده قرار گرفت. یافته‌ها: در روش سنجش ناپایداری میکروستلایت از طریق پنل Promega و نشانگر تک نوکلئوتیدی MT1XT20 به ترتیب 8 (40 درصد) و 5 نمونه (25 درصد) از 20 نمونه وضعیت ناپایداری نشان دادند. در روش ایمنوهیستوشیمیایی نیز، 7 نمونه از 20 نمونه (35 درصد) وضعیت کمبود پروتئین مسیر ترمیم ناجور جفت شدگی را نشان دادند. در میان نشانگرهای مورد استفاده در کیت Promega، نشانگر BAT26 ناپایدارترین نشانگر بود و در هر 8 نمونه، Microsatellite instability-high (MSI-H) تشخیص داده شده توسط کیت Promega ناپایداری نشان داد. نشانگرهای NR21 و NR24 در 7 نمونه (5/87 درصد) ناپایداری نشان دادند و نشانگرهای BAT25 و MONO27 به ترتیب با نشان دادن ناپایداری در 6 (0/75 درصد) و 5 نمونه (5/62 درصد)، پایدارترین نشانگرها بودند. نتیجه‌گیری: MT1XT20 در جمعیت ایتالیایی، به عنوان نشانگر تک نوکلئوتیدی مورد اعتماد ارزیابی شده است، اما این مسأله در رابطه با جمعیت ایرانی صدق نمی‌کند. در عوض، BAT26 با نشان دادن ناپایداری در تمام نمونه‌های MSI-H ارزیابی شده توسط کیت Promega، می‌تواند به عنوان نشانگری موثق و قابل مقایسه با پنل 5 نشانگری Promega در جمعیت ایرانی در نظر گرفته شود.

Background: Integral membrane proteins involve in a variety of valuable cellular processes including signal transduction, transport and cell-to-cell communication. Thus, due to their importance, are targeted to treatment of diseases or disorders. To achieve this goal, it is necessary to obtain adequate membrane proteins. A variety of expression systems have been used to overexpress these proteins but mammalian cells can produce membrane proteins structurally close to natural form of them and are favourable for this purpose. TOSO/FAIM3 (Fas Apoptosis Inhibitory Molecule), highly conserved membrane protein, playing important role in cell survival, immune surveillance, and homeostasis and in this study was overexpressed on HEK-293T membrane. Methods: Eukaryotic expression vector pEZ-M67-TOSO was transformed into Escherichia coli top 10F’ strain to multiply. Then, the plasmid was extracted and digested with restriction enzyme Eco31I to make a linear plasmid. Subsequently, HEK-293T cells were transfected with linear plasmid and treated with hygromycin to select TOSO expressing stable cells. After three weeks, resistant colons were expanded, Chromosomal DNA and total RNA were extracted and the presence of TOSO-cDNA in HEK-293T genome and the expression level of TOSO were evaluated using polymerase chain reaction (PCR) and Real-time PCR methods, respectively. Findings: The integration of TOSO expressing cassette in HEK-293T genome was confirmed. HEK-293 cells expressed 1700 mRNA TOSO molecule/cell which was in the range of protein with abundant expression. Conclusion: Mammalian cells represent membrane proteins in desirable form versus other expression systems. For this reason, we used HEK293T cells to overexpress transmembrane TOSO protein. In this investigation, we successfully constructed the HEK293 cell line with stable TOSO overexpression, which will facilitate further researches to characterize biophysical and biochemical structure and function of TOSO and pharmaceutical biological researches about it.

Background: Cystinuria, one of the first diagnosed inborn errors of metabolism, recognized by hyperexcretion of cystine, lysine, ornithine and arginine into the urine. So far, two genes associated with cystinuria have been identified: SLC3A1 (2p16.3) that encodes the heavy subunit rBAT of the renal b0,+ transporter and SLC7A9 (19q13.1), which encodes the light subunit b0,+AT. Patients with type A cystinuria have two SLC3A1 mutations, whereas patients with type B have two SLC7A9 mutations and finally patients with type AB have one mutation in each gene. Considering the population-specific distribution of mutations in disease, limited studies on the genetic bases of the cystinuria have been done in Middle East. This research presents the results of mutation analysis on patients with cystinuria in Iran. Methods: Thirty unrelated patients with cystinuria operated to remove kidney stones were screened by urologist .The patients were analyzed for mutation using amplification refractory mutation system (ARMS) and polymerase chain reaction-Restriction fragment length polymorphism-polymerase chain reaction (RFLP-PCR) methods. Findings: We found some variations including missense mutations, polymorphism and intron variant, but the most frequent mutations, M467T and also T216M and R333W, were not detected in our patients. Conclusion: Our research may confirm the ethnic distribution of mutations in cystinuria and this study can expand our concept of the genetic basis of cystinuria in Iranian patients.

Background: Micro ribonucleic acids (miRNA) are a type of ribonucleic acids with 20-24 nucleotides. They seem to have an important regulating role in cells. MiR-211 is an miRNA whose expression decreases in cancer cells, especially human melanoma cells. It is of high importance in suppression of tumor growth and invasion. Up to now, no study has evaluated miR-211 expression in murine melanoma cells for in vivo and in vitro conditions. We evaluated and compared the expression of miR-211 in murine melanoma cell line and melanoma tumors using real-time polymerase chain reaction (PCR). Methods: Melanoma B16F10 cell line was cultured. Subcutaneous melanoma tumors were induced by injecting this line to C57 mice. Melanoma cell line and melanoma tumors were lysed and miRNAs were extracted. After complementary deoxyribonucleic acid (cDNA) synthesis, real-time PCR of samples was performed using miR-211 primers. Data was statistically analyzed by Student’s t-test. Findings: Average expression of miR-211 in melanoma cell line was significantly higher than melanoma tumors. Conclusion: Due to the lower expression of miR-211 in murine melanoma tumors in comparison to murine melanoma cells, miRNA may be involved in invasive properties of melanoma cells in the mouse model of melanoma. Soluble forms of nonclassical human leukocyte antigen (HLA)-G have recently been suggested as immunomodulatory factors in multiple sclerosis (MS). HLA-G inhibits the effecter function of T cells and natural killer (NK) cells. Also regulatory T cells (Treg) are considered as pivotal players in MS pathogenesis. Thus, we aimed to evaluate the presence of HLA-G molecules and Treg cells in Relapsing-Remitting Multiple Sclerosis (RRMS) patients and compare it to healthy controls.Patients with RRMS (n=205, mean age=31.32±8.53) and healthy subjects (n=205, mean age=32.2±7.48) were studied. The patients subgrouped to untreated and treated with Interferon beta. Then sHLA-G levels (sHLA-G1 and sHLA-G5) were measured using ELISA method. Treg (CD4+CD25+ Foxp3+) cells in patients who had sHLA-G>10 U/ml were characterized by using flow cytometry.Our data showed that there was no significant differences between RRMS patients and healthy controls in sHLA-G concentration (p>0.05). Background: Severe mitral stenosis is occasionally associated with significant tricuspid regurgitation (TR) and this association has an adverse impact on morbidity and mortality in patients undergoing mitral valve intervention. However, the effect of successful mitral balloon valvotomy (MBV) on significant TR is not fully elucidated. The aim of this study was to investigate the course of TR after MBV in patients with severe mitral stenosis with TR.Methods: The present study was performed in Tabriz Madani heart center from March 2007 to February 2008. Among 110 patients with mitral stenosis who were candidates of MBV, 68 cases with more than mild TR were selected and the fate of TR after MBV and its predictors were evaluated. Results: Among 68 patients who were enrolled in this study, 58 individuals (85.3%) were female with mean age of 36.85± 14.32 years. Before intervention, 48 patients (70.6%) had severe TR and 20 (29.4%) cases had moderate TR. Background Helicobacter pylori (H. pylori) is a gram negative curved bacilli colonized in the human stomach. It causes duodenal ulcer, gastritis and is associated with adenocarinoma. There might be a possible relationship between cagA, vacA and ice a genes and clinical outcomes. The aim of this study was to identify the frequency of vacA genotypes of H. pylori isolated from the upper gastrointestinal. Methods One hundred H. pylori strains were isolated from patients with different gastrointestinal disease who admitted to Isfahan Al-zahra hospital. Vac A alleles were typed using PCR with specific primers. Findings There were four Vac A mosaicsms, including 28 for s1a/m1 (28%), 23 for s1b/m1 (23%), 26 for s1a/m2 (26%) and 23 for s1b/m2 (23%), s2 form was not found. Conclusion The results showed there is no significant relationship between different genotypes of vacA and the related diseases. Background: Diabetes mellitus is the most prevalent metabolic disorder worldwide with many subsequent medical complications. An important complication of diabetes is microalbuminuria which is a major indication for development of diabetic nephropathy. The prevalence of diabetic nephropathy in type 2 diabetic patients has been estimated to be 47%. Since the disorder initiates with microalbuminuria, its early detection and prevention are vital. On the other hand, helicobacter pylori infection is more common in diabetic patients due to their low immune tolerance. The vacA positive genotype is reported to be effective in microalbuminuria development in type 2 diabetic patients. In this study, we investigated the relation between H. pylori vacA and microalbuminuria in type 2 diabetic patients. Methods: This study included 88 type 2 diabetic patients among whom 60 had microalbuminuria. Stool samples were collected from all patients and DNA was extracted using QiaAmp Stool DNA Extraction Minikit. In order to detect H. pylori infection, a nested polymerase chain reaction (PCR) protocol was developed and all H. pylori positive samples were genotyped for cagA positivity. Findings: Overall, 16 patients (18.2%) were detected to be H. pylori positive out of whom 12 had microalbuminuria (75%). VacA positivity was detected in one patient without microalbuminuria. Conclusion: We could not establish a relationship between vacA positivity and microalbuminuria in the studied type 2 diabetic patients. Background: Mouse mammary tumor virus (MMTV), a milk-transmitted retrovirus, is considered to be involved in the development of breast tumors in mice. Although several previous studies have identified MMTV-like sequences in human breast cancer, the presence of human endogenous retrovirus (HERs) makes these results controversial, i.e. the role of MMTV in breast cancer has never been definitely proven. In addition, various results have been reported in this regard from different countries. We selected MMTV envelope (Env) gene sequences to eliminate the effects of HERs. Methods: In this case-control study, DNA was extracted from 59 paraffin-embedded malignant tumor tissues and 59 normal control samples. Nested polymerase chain reaction (PCR) was then used to detect MMTV Env-like sequences. Findings: MMTV genome was detected in 19 malignant breast samples (32%) and three normal control samples (5%) (P < 0.001). Conclusion: Based on our findings, MMTV can play a major role in the development of breast cancer in Iranian women. According to PCR results, the obtained sequences had 99% homology with BR6 strain of MMTV and 100% homology with GR and C3H strains.

Background: A major bottleneck in the production of recombinant proteins in conventional linear method is the heterogeneity in number of transgene copies in the genome of the host cell that lead to variable levels of transgene expression. Aside from the low efficiency of the random integration, other phenomena such as positional effect contribute to low efficiency of transgene expression. PiggyBac is a class of DNA transposons which can transpose through “cut and paste” mechanism in host genome. Some specific characteristic of this transposon makes it promise vector in gene transfer studies. High capacity of transgene transposition and flexibility in molecular engineering of transposase are characteristics of PiggyBac transposons that are important in recombinant protein and gene therapy approaches. The aim of this study was estimation of transgene copy number based on helper-dependent and independent PiggyBac transposition system in human embryonic kidney (HEK) cells. Methods: Plasmid containing interferon gamma inducible protein 10 (IP-10) coding sequence flanked by PiggyBac terminal repeat element and plasmid containing transposase system and a unit plasmid containing both transposae and IP-10 coding sequence were used for generating recombinant cells in helper-dependent and independent system, respectively. Human embryonic kidney cells were transfected by each group. After clonal selection, absolute quantitative real-time polymerase chain reaction was used for estimation of transgene copy number. Findings: Estimation of IP-10 copy number has revealed 5 and 2 copies per cell in helper-independent and dependent system, respectively. Conclusion: Here, we report activity of PiggyBac transposition in human embryonic kidney cell line. PiggyBac helper-independent and dependent system was used for generation of stable cell line producing mouse IP-10 protein and our data confirmed permanent expression of this transgene with the mean of about 5 transgene copies per each cell.

مقدمه: مولتیپل اسکلروزیس (MS یا Multiple sclerosis) یک بیماری خودایمن مزمن در سیستم عصبی مرکزی (CNS یا Central nervous system) است که با پاسخ سلول‌های لنفوسیت T به آنتی‌ژن‌های میلین ایجاد می‌شود. یکی از انواع گیرنده‌های سطح سلولی لنفوسیت‌های T مولکول‌های خانواده‌ی TIM (T-cell immunoglobulin and mucin domain) هستند که SNPهای (Single nucleotide polymorphisms) متعددی در ژن مربوط به آن‌ها شناسایی شده است. این SNPها همواره با بیماری‌های خودایمن مختلفی در ارتباط بوده‌اند. هدف از این مطالعه، مقایسه‌ی فراوانی پلی‌مورفیسم +4259A>C ژن 3-TIM در بیماران دچار مولتیپل اسکلروزیس و افراد سالم در اصفهان بود تا مشخص شود که آیا این پلی‌مورفیسم زمینه‌ساز بیماری فوق در اصفهان می‌باشد؟ روش‌ها: DNA سلول‌ها با استفاده از کیت استخراج DNA، از خون افراد جدا شد. در این مطالعه، از روش PCR-RFLP (Polymerase chain reaction-restriction fragment length polymorphism) استفاده کردیم. ابتدا ژن 3-TIM تکثیر شد؛ سپس، در مجاورت آنزیم خرد کننده قرار گرفت. با تکنیک الکتروفورز، قطعات حاصل از هضم آنزیمی تفکیک شد و مورد بررسی قرار گرفت. یافته‌ها: تفاوت معنی‌داری بین فراوانی ژنوتیپ‌ها و الل‌های پلی‌مورفیسم +4259A>C در بیماران و افراد سالم مشاهده شد (029/0 = P). شانس ابتلا به بیماری در افرادی که حامل الل C بودند، حدود 2 برابر افراد فاقد این الل بود (010/0 = P). نتیجه‌گیری: پلی‌مورفیسم +4259A>C ممکن است در ابتلا به بیماری مولتیپل اسکلروزیس نقش داشته باشد. برای درک بیشتر می‌توان، اثر این پلی‌مورفیسم را بر بیان و نحوه‌ی عمل مولکول 3-TIM بررسی کرد.

In order to detect H. pylori infection, a nested polymerase chain reaction (PCR) protocol was developed and all H. pylori positive samples were genotyped for cagA positivity. Findings: Overall, 16 patients (18.2%) were detected to be H. pylori positive out of whom 12 had microalbuminuria (75%) and 4 (25%) did not. CagA positivity was detected in 3 subjects who belonged to the group with microalbuminuria. Conclusion: In the population studied and the sample size used, no correlation was found between cagA gene and microalbuminuria in patients with type 2 diabetes. Background: Microsatellite instability (MSI) is a mutational signature that is the hallmark of Lynch syndrome, and MSI testing is a cost-effective method to screen the disease. Since there is no enough data about MSI status and associated clinicopathologic features of hereditary nonpolyposis colorectal cancer (HNPCC) in Iran, our study is a new trial to describe them in center of Iran (Isfahan). Materials and Methods: It is a descriptive retrospective study to screen HNPCC families using Amsterdam II criteria in Central Iran within 2000-2013. For MSI testing, we used a commercially available kit evaluating mononucleotide markers (BAT-25, BAT-26, MON0-27, NR-21 and NR-24). After a fluorescent multiplex polymerase chain reaction amplification of the markers, samples were sequenced to fragment analysis. Data analysis was performed using SPSS 16 software (SPSS Inc., Chicago, IL, USA). Results: Overall, 31 of 45 screened HNPCC families were eventually included to MSI testing. Totally, 9/31 patients (29.0%) showed MSI in their tumor tissues. BAT-26 was the most instable marker with instability in 7/24 MSI tumors (29.2%). The mean age at diagnosis in microsatellite stable (MSS), MSI-Low (MSI-L), and MSI-High (MSI-H) probands was respectively 44.7 (standard deviation [SD] = 11.83), 51.7 (SD = 16.17), and 36.0 (SD = 3.41) years. The most common tumor sites in MSS, MSI-L, and MSI-H probands were rectosigmoid (∼72.8%), rectum (66.7%) and right colon (50.0%), respectively. Of 186 cancer patients among 31 HNPCC families, 86 patients (46.2%) had colorectal cancer (CRC) and 100 patients (53.8%) had extracolonic cancers. The average of CRC affected members among MSS, MSI-L, and MSI-H groups of our HNPCC families was 2.2 (SD = 1.30), 3.3 (SD = 3.21), and 4.7 (SD = 2.42) patients per family, respectively. Stomach with 18.3% and 26.7% of all extracolonic cancers were most common involved organ in MSS and MSI-H families, respectively. Conclusion: Our different molecular results could be suggested to describe HNPCC families based on some new molecular mechanisms leading to MSS HNPCC phenotypes. Meanwhile, more evaluations within our population are recommended.

Background: Dental caries is a worldwide health problem involving almost all age groups in underdeveloped as well as developed countries. So for no considerable progress achieved in the disease prevention, so new preventive/thrapeutique measures are urgently needed. Probiotic bacteria and their biproducts, like biosurfactants, are cost-effective with no considerable side effects for dental caries prevention. The aim of the present study was to evaluate the effects of biosurfactants purified from Lactobacillus casei (ATCC39392) and Lactobacillus reuteri (DSM20016) on ftf and gtf B/C gene expression level of the standard strains of streptococcus mutans (ATCC35668). Methods: Lactobacillus species purchased from the bacterial collection center of Iran. For biofilm production, microtiter plate assay in presence of 1% glucose was used. For biosurfactant production, lactobacillus species were cultured in MRS broth medium, dialyzed and lyophilized using freeze-dryer. To evaluate the effects of biosurfactant on gene expression, bacterial cells were cultured in presence of the appropriate biosurfactant (2.5 mg/ml); after four hours incubation, total cellular RNA was then extracted and used for cDNA synthesis. Findings: Expression of ftf and gtf B/C genes of the standard strains of Streptococcus mutans (ATCC35668) was considerably down regulated by the application of the biosurfactants. The reduction in gene expression was statistically significant (P < 0.05). Conclusion: Considerable down regulation of all genes in presence of biosurfactants was observed. Therefore, it is concluded that the biosurfactants prepared in this study is very successful in dental caries prevention.

Background: Analysis of venous blood gas (VBG) can represent arterial blood gas (ABG) analysis in patients with various diseases. The effects of hypotension on differences between the results of simultaneous venous and arterial blood gas analyses were reviewed. Methods: This observational, cross-sectional study was conducted from March to October 2010 in emergency depart-ments of two university hospitals in Tehran (Iran) on consecutive adult patients for whom ABG had been indicated for diagnosis/treatment. Arterial and peripheral venous bloods were simultaneously sampled with blood pressure measure-ment. The VBG-ABG amount of difference regarding pH, HCO 3 , PCO 2 , PO 2 , SO 2 , and Base Excess (BE) was com-pared between those with and without hypotension. Results: During the study, 192 patients (51.6 ± 23.6 years, 67.7% males) were entered into the hypotension (n = 78) and normotensive groups (n = 114). The average VBG-ABG amount of difference (95% limits of agreement) in the hypotension versus normotensive group were -0.030 (-0.09 to 0.03) vs. -0.016 (-0.1 to 0.068) for pH (p = 0.01), 1.79 (-1.91 to 5.49) vs. 1.32 (-1.94 to 4.58) mEq/L for HCO 3 (p = 0.032), 2.69 (-20.43 to 25.81) vs. 2.03 (-7.75 to 11.81) mmHg for PCO 2 (p = 0.295), -35.97 (-130.17 to 58.23) vs. -32.65 (-104.79 to 39.49) mmHg for PO 2 (p = 0.293), -18.58 (-14.66 to 51.82) vs. -9.06 (-31.28 to 13.16) percent (p < 0.001) for SO 2 , and 0.25 (-3.73 to 4.23) vs. 0.79 (-2.51 to 4.09) for BE (p = 0.036). Conclusions: Hypotensive status is associated with an increase in the amount of difference between VBG and ABG analysis regarding pH, HCO 3 , and BE, though the amount of increase does not seem to be clinically important. Studying the precise effects of replacing ABG with VBG on the clinical decision-making and the following outcomes is worth-while.

Background: Leishmania is an obligate interacellular protozoa and sand fly, as a vector, transmits infectious forms of the parasite to vertebrate host. In this way it is important to find candidate antigens which could tend to prevent the disease. Methods: The gene coding mannose 1 phosphate guanyl transferase enzyme was amplified from genomic DNA isolated from the Iranian strain of L. major (MRHO/IR/75/ER) as a template. The Polymerase Chain Reaction (PCR) product was ligated into the pTZ57R plasmid and the recombinant gene was digested using restriction enzymes, BamHI and EcoRI. The fragment was ligated into the pET32a plasmid, as an expression vector. The cloned pET32a-GDP mannose was confirmed using restriction enzyme digestion method and the DNA fragment was sequenced. Findings: Electrophoresis method confirmed the PCR product is related to the enzyme mannose 1 phosphate guanyl transferase. After the ligation of the product into the pTZ57R and pET32a, and the restriction enzyme digestion by BamHI and EcoRI, the correct frame of cloned gene in vectors was confirmed. Conclusion: There was 92 percent homology between the cloned gene coding enzyme mannose 1 phosphate guanyl transferase in this study and the ones present in gene bank. It is suggested that the gene encoding mannose 1 phosphate guanyl transferase enzyme is conserved among different genera of Leishmania. Background: Bone is a tissue with high repair capacity; but, this ability dramatically is limited during irrecoverable bone and skeletal defects. Stem cell therapy is a promising approach for construction of bone tissue. Mesenchymal stem cells (MSCs) have been introduced as basic tools for bone tissue generation in tissue engineering. Core binding factor alpha 1 (Cbfa1) is one of the main genes involved in osteogenesis. Since relative expression of Cbfa1 in differentiated osteoblasts from adipose derived stem cells is unknown, this study aimed to evaluate the gene expression pattern of Cbfa1 in adipose derived differentiated osteoblasts in 4 different time intervals. Methods: Real-time PCR was used for studying the gene expression of Cbfa1 in human normal osteoblasts and adipose derived osteogenic osteoblasts at the days 7, 14, 21 and 28. Findings: There was a progressive increase in Cbfa1 expression over the differentiation period of adipose derived mesenchymal stem cells (ADSCs) from day 7 to day 28. Differentiated osteoblasts expressed Cbfa1 lower than native osteoblasts (P < 0.05). Conclusion: ADSCs are an attractive source for engineering a new bone tissue. According to our results, adipose derived differentiated osteoblasts express candidate gene at least on day 7 of culture. Our study indicates that longer treatment of ADSCs in osteogenic medium confer better osteoblastic properties to these cells.
