Summary: The aim of the study was to evaluate the influence of different dietary protein and lipid levels and their ratios on larval growth, survival and the incidence of cannibalism in Pseudoplatystoma punctifer. Larvae were raised in a recirculation system from 3 to 26 days post‐fertilization (dpf) (2–25 days post hatching, dph) at an initial density of 40 larvae L
−1 , 27.8 ± 0.65°C and 0L : 24D photoperiod. Larvae were fed from 4 to 12 dpf with Artemia nauplii and weaned onto four different compound diets from 13 dpf within 3 days, then fed exclusively with these diets until 26 dpf. These diets contained 30 : 15, 30 : 10, 45 : 15 or 45 : 10 protein : lipid (P : L) (in % of dry matter) levels. A control group was fed Artemia nauplii until 17 dpf and weaned thereafter with the 45P : 10L compound diet. The experiment was carried out in triplicate. Results showed higher growth and survival rates and lower incidence of cannibalism in the group fed the 45P : 15L diet than in the other treatments. Differences in larval survival and growth performance were associated with the higher protein and lipid content rather than the protein : lipid ratio of this diet. When comparing diets with the same protein level, the increase in dietary lipid led to an improvement in growth, suggesting that energy from lipids spares protein for growth in P. punctifer fingerlings. 5. Histological development of the digestive system of the Amazonian pimelodid catfish Pseudoplatystoma punctifer.

The organogenesis of the digestive system was described in the Amazonian pimelodid catfish species Pseudoplatystoma punctifer from hatching (3.5 mm total length, TL) to 41 days post-fertilization (dpf) (58.1 mm TL) reared at 28°C. Newly hatched larvae showed a simple digestive tract, which appeared as a straight undifferentiated and unfolded tube lined by a single layer of columnar epithelial cells (future enterocytes). During the endogenous feeding period, comprised between 20 and 96 h post-fertilization (3.5 to 6.1 mm TL), the larval digestive system experienced a fast transformation with the almost complete development and differentiation of most of digestive organs (buccopahrynx, oesophagus, intestine, liver and exocrine pancreas). Yolk reserves were not completely depleted at the onset of exogenous feeding (4 dpf, 6.1 mm TL), and a period of mixed nutrition was observed up to 6 to 7 dpf (6.8 to 7.3 mm TL) when yolk was definitively exhausted. The stomach was the organ that latest achieved its complete differentiation, characterized by the development of abundant gastric glands in the fundic stomach between 10 and 15 dpf (10.9 to 15.8 mm TL) and the formation of the pyloric sphincter at the junction of the pyloric stomach and the anterior intestine at 15 dpf (15.8 mm TL). The above-mentioned morphological and histological features observed suggested the achievement of a digestive system characteristic of P. punctifer juveniles and adults. The ontogeny of the digestive system in P. punctifer followed the same general pattern as in most Siluriform species so far, although some species-specific differences in the timing of differentiation of several digestive structures were noted, which might be related to different reproductive guilds, egg and larval size or even different larval rearing practices. According to present findings on the histological development of the digestive system in P. punctifer, some recommendations regarding the rearing practices of this species are also provided in order to improve the actual larval rearing techniques of this fast-growing Neotropical catfish species. Dicentrarchus labrax were fed from 10 to 40 days posthatching with six microdiets differing in the inclusion level (60 g kg
-1 and 120 g kg-1 ) and type of protein hydrolysate ( PH; yeast, YPH; pig blood, PBPH; pig red blood cells, PRBCPH). A microdiet containing 120 g kg-1 fish PH ( FPH) was used as a control. PH differed in their amino acid ( AA) profile and molecular weight distribution and therefore the tested microdiets too. The estimated content in FAA and di- and tripeptides in the FPH microdiet was 2 g kg-1 and 44 g kg-1 , respectively. FAA estimated levels in YPH and PBPH microdiets were 26 g kg-1 and 53 g kg-1 , whereas levels of di- and tripeptides were 30 g kg-1 and 60 g kg-1 , respectively. The estimated levels of FAA in PRBCPH microdiets were 8 g kg-1 and 17 g kg-1 , whereas estimated levels of di- and tripeptides were 11 g kg-1 and 22 g kg-1 , respectively. Results revealed that FPH may be replaced by alternative PH from yeast and pig blood products, as fish fed those diets performed, in terms of growth, survival, digestive function and incidence of skeletal deformities, as well as those fed the FPH microdiet. To improve the understanding of the incidence of skeletal deformities in Senegalese sole ( Solea senegalensis), a feeding trial was carried out to evaluate the effect of increasing dietary arachidonic acid (ARA) levels during the Artemia feeding period on larval growth and skeletogenesis. Larvae were fed from 8 to 50 days post hatching (dph) with Artemia nauplii enriched with three different levels of ARA: ARA-Low, ARA-Medium and ARA-High (1.0, 4.5 and 7.0% ARA of total fatty acids, respectively). Increasing levels of dietary ARA did not affect significantly the survival. However, it influenced growth; larvae from the ARA-M group presented the highest values of final standard length and dry weight (11.36 ± 0.47 mm and 5.86 ± 1.06 mg at 50 dph, respectively) and larvae from the ARA-H group the lowest ones (9.53 ± 0.27 mm and 2.47 ± 0.26 mg at 50 dph). The skeleton of larvae fed the ARA-M diet tended to be more calcified at 15 dph than that of larvae fed ARA-L and ARA-H diets. Larvae from the ARA-M group tended to show a higher incidence of fusion of hypurals 3 and 4 (34.4 ± 3.1%) at 50 dph than the larvae from the other groups (18.7 ± 1.6%, in average). The latter results reflected a trend to a higher degree of skeletal development rather than a deformity, this being in agreement with the higher larval growth of this group. Besides, ARA levels did not affect significantly the incidence of total skeletal deformities (41.4 ± 1.5% in average). Larvae from all dietary treatments mostly displayed fusions of the vertebra 43 and 44 (32.2 ± 1.3% in average). The amount of 4.5% of total fatty acid of dietary ARA during Artemia feeding period promoted the best growth and proper skeletogenesis in Senegalese sole larvae. [ABSTRACT FROM AUTHOR]

Several studies have evaluated the effects of dietary vitamin A (VA) on the incidence of skeletal deformities during early ontogeny of fish, but little is known about its effects on bones depending on their process of ossification (dermal or chondral). We examined the incidence of skeletal deformities along development (30 and 48 dph) by double staining technique, in dermal (haemal and caudal vertebral bodies) and chondral (neural and haemal spines, epural, parahypural and hypurals) bones in Senegal sole post metamorphosed larvae fed with different dietary VA levels (37 000, 44 666, 82 666 and 203 000 UI total VA kg−1 DW) during Artemia feeding phase (6–37 dph, at 18°C). Results obtained in this study showed that dietary VA disrupted the skeletogenesis in Senegalese sole post metamorphosed larvae by increasing the incidence of skeletal deformities in the axial skeleton and caudal fin complex, which were dependent on both bone morphogenesis and ossification processes. Fish fed with the highest dietary VA content showed the highest incidence of skeletal deformities and its value increased along ontogeny. However, when we compared the incidence of deformities in skeletal structures considering their ossification process, most skeletal structures derived from chondral ossification showed a significant higher increase in deformity incidences in fish fed an excess of VA (44 666, 82 666 and 203 000 UI kg−1 DW), however within chondral bones, hypurals deformity incidence only increased in sole larvae fed Artemia highest VA content. In contrast, this dietary dose-response effect was only noted in dermal bones from fish fed the highest dose of VA (203 000 UI kg−1 DW). In addition, the incidence of deformities in chondral bones increased even when the dietary imbalance of VA was corrected, whereas dermal bones were not affected at later ages. These results indicated that depending on the ossification process from which different skeletal structures are derived, bones might be differentially affected by high dietary VA content. Those directly originated from the connective tissue with a preliminary cartilage stage were more sensitive to dietary VA excess than those formed by intramembranous ossification. Abstract: Two fish protein hydrolysates (FPH) were incorporated into four diets prepared for start-feeding sea bass larvae, at two different levels (10% and 19% of total ingredients): a commercial FPH, CPSP, in which the molecular mass of the main fraction of soluble peptides (51%) was between 500–2500 Da, and an experimental FPH obtained by acidic silage of sardine offal, SH, with a main portion of soluble peptides (54%) ranging from 200 to 500 Da. The diet with 10% of the commercial FPH gave the best results in terms of growth, survival and intestinal development, as evaluated by the early activity of digestive enzymes in the brush border membrane (alkaline phosphatase and aminopeptidase N). This was related to the low level of Vibrio spp. counted in the larvae of group C10. The high dose of FPH, especially in the experimental preparation rich in short peptides, seemed to favour the dominance of Vibrio sp. TYH3, which behaved opportunistically. The effect of the experimental FPH was ambiguous, since early larvae challenged with Vibrio anguillarum were more resistant to the pathogen, especially at high FPH dose (group S19). This might be due either to direct antagonism between V. anguillarum and Vibrio sp. TYH3, or to the stimulation of the immune response in the larvae. These results indicate that different molecular weight fractions and concentrations of feed-soluble peptides may affect the growth performance and immunological status of sea bass larvae. Consequently, a low dose of commercial FPH seems advisable, both for larval development and for the bacterial environment, although further research is required to determine and characterize peptide fractions that may have a beneficial effect on growth and immune response, and to determine their optimal inclusion levels in diets for sea bass larvae. The ontogenetic development of the gut and accessory organs in large yellow croaker Pseudosciaena crocea was investigated using light microscopy from hatching up to the juvenile stage (40 days post hatch, dph). At 3 dph (mean ± s. d., 4·1 ± 0·1 mm total length, L
T ), coinciding with the buccopharynx opening, larvae started to feed exogenously, and the gut consisted of a well-developed buccopharynx, a partially-differentiated oesophagus and an intestine divided in three regions (anterior intestine, intermediate intestine and rectum). Yolk reserves were not completely depleted at the onset of exogenous feeding, and a period of mixed nutrition was observed up to 6 dph (4·3 ± 0·1 mm LT ), when yolk was definitively exhausted. Important morphological changes occurred at the end of the larval period, coinciding with metamorphosis. At 17 dph (6·8 ± 0·6 mm LT ), pyloric caeca differentiated at the junction of the pyloric stomach and the anterior intestine. Gastric glands were first observed at 21 dph (9·2 ± 1·2 mm LT ), coinciding with the morphological development of the stomach in three different regions (cardiac, fundic and pyloric) according to the histological characteristics of their mucosa. At this age, large longitudinal folds appeared in the median and posterior oesophageal mucosa. California halibut Paralichthys californicus is an important commercial species with high aquaculture potential in Baja California Sur, México. To optimize the feeding process using live prey and/or inert diets, we evaluated alkaline proteases, pepsin, trypsin, chymotrypsin, leucine aminopeptidase, lipase, α-amylase, and acid and alkaline phosphatase activities on starved larvae and larvae fed live prey. Highest activities were observed for alkaline protease, trypsin, chymotrypsin, leucine aminopeptidase, and alkaline phosphatase in feeding larvae than starved larvae on day 4 after hatching. At day 5, a sizeable increase in all enzymatic activities was detected in feeding larvae. Alkaline protease, trypsin, chymotrypsin, and alkaline phosphatase decreases progressively from day 5 until day 18. At day 18, a slight pepsin activity was observed. This was considered an indicator of the start of digestive system maturation. We concluded that total enzymatic equipment for this species is complete between day 18 and 30 after hatching. Based on this evidence, early weaning from live prey to inert feed would be possible at this time. [ABSTRACT FROM AUTHOR]

The effect of the timing of first feeding (3, 4, 5, 6, 7 and 8 days post-hatch, dph) on laboratory-reared California halibut Paralichthys californicus larvae was evaluated by means of morphologic, morphometric and histological criteria. Larvae began to feed exogenously at 3 dph (2·7 ± 0·01 mm standard length, LS) at 18° C. Eye pigmentation, rather than mouth opening was the most distinctive trait of California halibut larvae at first feeding. Larval growth was significantly affected by the time of first exogenous feeding. At notochord flexion (21 dph), the LS of larvae fed for the first time at 3 dph was significantly larger (5·1 ± 0·1 mm) than that of those fed at 4 and 5 dph (4·9 ± 0·1 mm), although the latter fish had a more uniform size distribution. The point of no return was reached at 7 dph. Survival of larvae initially fed at 3, 4 and 5 dph was similar (58·4–60%), while no larvae were able to survive when food was offered for the first time between 6 and 8 dph. Food deprivation resulted in a progressive deterioration of the larval digestive system and atrophy of skeletal muscle fibres. Significant changes in the anterior and posterior enterocyte height were detected after 2 days of food deprivation. Similarly, tail height: LS and trunk length: LS ratios were the most sensitive morphometric indices to detect the effect of fasting on larval condition. Present results show that a combination of morphometric and histological variables can be used to evaluate the nutritional condition of California halibut larvae. [ABSTRACT FROM AUTHOR]

Since first large-scale attempts to culture sturgeon from the larval stage were carried out in the former U.S.S.R. at the end of the nineteenth century, rearing technology has advanced considerably during the last 20 years and noticeable improvements in incubation and larval rearing techniques have been implemented in normal hatchery procedures. Siberian sturgeon eggs are incubated in MacDonald jars at 13–14° C to prevent fungal infections. Mass hatching takes place 7 days after fertilization and hatching rate can be predicted as a function of the percentage of eggs fertilized. Survival at the end of the endogenous feeding stage is correlated with hatching rate. Egg size has no direct implications for larval growth and survival of Siberian sturgeon. Experimental studies have demonstrated that behavioural observations are useful criteria to assess the quality of larvae and to synchronize the physiological state of fish with the appropriate rearing procedures. Special attention should be given to the transition to exogenous feeding, where cannibalism, difficulties in adaptation to a new diet, overfeeding and resulting bacterial infections dramatically reduces survival to the fingerling stage. Although a commercial artificial diet specifically formulated for larvae of Siberian sturgeon and other acipenserids is still lacking, commercial non-purified rainbow trout diets and starter marine fish diets are currently used and their results are reasonably acceptable in terms of larval growth and survival. Further research must be focused on the determination of egg quality indicators in order to provide the producer with the tools to estimate the viability and performance of the progeny.
