Understanding oxidative stress in high-mountain lake fish offers crucial insights into their health, resilience, and adaptation to extreme environmental changes. This study investigates the oxidative stress response of Cottus gobio in a high-mountain lake (Dimon Lake) located in the northeast Italy during the ice-free season, focusing on the relationship between oxidative stress biomarkers and physicochemical water parameters, as well as persistent and emerging contaminants. Significant seasonal variations were observed in water parameters, with lower oxygen, pH, conductivity, and phosphate levels in summer compared to autumn, while temperature, ammonium, and nitrate were higher in summer. Metal concentrations in C. gobio muscle were higher in autumn, with Zn showing the most significant increase. PAHs, NDL-PCBs, and pesticides were all below the limit of quantification in the fish muscle samples. No microplastics items were found in the gastrointestinal tracts of fish. Oxidative stress biomarkers revealed organ-specific and seasonal variations. The liver exhibited the highest activities of catalase (CAT), glutathione peroxidase (GPx), glutathione S-tranferase (GST), and glutathione reductase (GR), highlighting its central role in detoxification and metabolic processes. Superoxide dismutase (SOD) activity was notably higher in muscle tissue during summer, suggesting increased metabolic activity. A strong correlation was found between pH and the activities of SOD, CAT, GPx, GR, and metallothioneins (MTs), emphasizing the importance of water chemistry in influencing oxidative stress responses. This approach not only aids in understanding the local adaptations of these fish but also highlights the impacts of environmental stressors on high-mountain ecosystems. Despite their remote locations, high-mountain lakes located in the Alps are vulnerable to chemical pollution. This discussion explores the important aspects of these lakes as repositories of Persistent Organic Pollutants (POPs) and Contaminants of Emerging Concern (CECs), elucidating their sources and implications for both the environment and human health. In terms of the presence of POPs in high-altitude lakes of the Alps, 14 studies have been identified examining the occurrence of polychlorinated biphenyls, dichlorodiphenyltrichloroethane an its metabolites, polybrominated diphenyl ethers, and polycyclic aromatic hydrocarbons. The bulk of research on POPs in high-mountain lakes is concentrated in the Italian Alps (63%), followed by Switzerland (22%), Austria (12%), and France (3%), respectively. Sediment is predominantly investigated (65%), followed by fish (33%) and water (2%). Similarly, in relation to the presence of CECs in high-mountain lakes of the Alps, six studies have been identified investigating the occurrence of musks, perfluorinated compounds, and microplastics. Investigations into CECs predominantly occur in Switzerland (42%), France (33%), and Italy (25%), with fish samples (muscle and liver) being the primary focus (46%), followed by sediment (17%) and water (17%). Other compartments like zooplankton, frog/tadpoles, and snow remain less explored. The discussion also shed light on various pathways through which pollutants reach these remote landscapes, including atmospheric transport, glacial meltwater, and human activities. High-mountain lakes are pristine ecosystems characterized by extreme environmental conditions. The atmospheric transport of pollutants from lowlands may add further stress to organisms inhabiting these environments. We investigated the environmental stress pressure on brook trout (Salvelinus fontinalis) from a high-mountain lake in the Cottian Alps (Piedmont, northwest Italy). To do this, males and females of brook trout were sampled from Balma Lake in summer (August) and autumn (October) 2017 in order to assess the influence of trace elements accumulation and environmental parameters (physicochemical parameters and nutrient characteristics of water) on oxidative stress biomarkers. Bioaccumulation of Al, As, Cd, Cr, Cu, Fe, Hg, Pb, Ni, Se, and Zn and metallothionein levels were measured in muscle tissue of males and females. Liver, gills, kidney, and spleen tissue samples were analyzed for superoxide dismutase, catalase, total glutathione peroxidase, selenium-dependent glutathione peroxidase, glutathione reductase, and glutathione S-transferase activity. Analysis of environmental parameters showed changes in biomarker levels with seasonal variations. Water temperature was significantly higher in summer than autumn (Wilcoxon test; p = .0078), while pH was significantly higher in autumn than in summer (Wilcoxon test; p = .0078). Sex-related differences in oxidative stress biomarkers in tissues were unremarkable, whereas seasonal variability of oxidative stress biomarkers was observed, with major differences occurred for liver in summer and for gills, kidney, spleen and muscle in autumn. Positive correlations between environmental parameters and biomarkers were noted. Lactococcus petauri has emerged as a pathogen affecting farmed fish. In this case report, L. petauri was isolated from Lepomis gibbosus captured in Lake Candia, located in Piedmont, Northwestern Italy. Initially identified as L. garvieae using conventional methods like RAPID 32 ID STREP and MALDI-TOF, molecular analysis based on sequencing of the 16S-23S rRNA internal transcribed spacer region (ITS) confirmed it as L. petauri. The study also assessed the isolated strain's pathogenicity by examining hemolysin and capsule cluster genes, revealing only hemolysin 2 amplicon; notably, the fish exhibited no lesions or pathological alterations. The L. petauri strain from this study displayed resistance to several antibiotics, being sensitive only to ampicillin, amoxicillin, florfenicol, and G-penicillin. High-altitude lakes have been studied intensively over the last decades both in terms of their biota and general physicochemical patterns and the importance of detailed knowledge of the small scale-regional fauna was underlined. In the present study the littoral macrobenthic invertebrate communities of two alpine lakes placed in the same geographical context (Upper and Lower Balma Lakes, Cottian Alps, Italy) were seasonally characterized, and the observed diversity was investigated in relation to site-specific abiotic features. During the ice-free period, two seasonal sampling campaigns (summer and autumn) were carried out at each lake along the littoral zones: macrobenthic invertebrate were collected and abiotic features (main physicochemical water parameters and main substrates) were monitored. PERMANOVA test disclosed significant differences in the invertebrate assemblages between the lakes, and SIMPER test highlighted that the observed dissimilarity was mainly related to Oligochaeta and Chironomidae taxa. The application of the RDA and VPA showed that differences were mainly due to littoral substrate composition and nutrient concentrations (i.e., phosphorus and nitrates) which could heavily affect both the structures and densities of the macrobenthic invertebrate communities in these environments. Findings from this study improve the knowledge about the macrobenthic invertebrate assemblages in the western area of the European Alps, highlighting the role of site-specific factors in small scales studies, and the importance of detailed knowledge of their local variability. The present study reports seasonal data about chemico-physical trends, macrobenthic invertebrates, and Phragmites australis decomposition rates within a karstic freshwater system in the western classic Karst. Results presented herein were compared with those obtained from the same system five years ago. Chemico-physical data showed higher temperatures and lower levels of pH and conductivity than in the past. The macrobenthic invertebrate community varied through the seasons and through the years, though they are dominated by few taxa, such as Asellus and Gammarus. These shredders were the most abundant functional feeding guild, playing a pivotal role in the decomposition of the vegetal organic matter. Decomposition rates k ranged between 0.009 and 0.016 days−1 and varied seasonally. k values were different from those previously reported for the investigated system. Differences are mainly due to changes in water supply, depending on decreasing rainfall regimes and hydrometric levels of the Soča River, which showed lower levels than in the previous study. Changes in water supply altered the fluctuating pulse that is typical of ephemeral karstic lakes, changing the system dynamics. Potential effects of climate change on local scale are also discussed. Carnobacteria are common bacteria in cold and temperate environments; they are also reported during fish mortality events. In a previous study, carnobacteria were isolated from the eyes of healthy wild salmonids from a high-mountain lake. To better understand these findings, salmonids were captured from three high-mountain lakes (Lower and Upper Balma Lake, Rouen Lake; northwest Italy) during August 2019 and subjected to bacteriological and histological examination. Although all were healthy, 8.7% (Lower Balma Lake), 24% (Upper Balma Lake), and 32.6% (Rouen Lake) were positive for carnobacteria colonization of the eyes. A Trojan-horse effect was hypothesized to explain carnobacteria isolation in the eye. This immune-escaping macrophage-mediated mechanism has been identified in other Gram-positive bacteria. Biochemical, molecular, and phylogenetic analysis were carried out on isolated bacteria (Carnobacterium maltaromaticum and C. divergens). Based on previous references for carnobacteria isolated from fish, C. maltaromaticum strains were tested for the pisA precursor gene of the bacteriocin piscicolin 126. Carnobacterium maltaromaticum strains were found to display genotypic heterogeneity and a low percentage of pisA positive amplification. Features of geomorphology, geographic isolation, and microbiota common to the three lakes are thought to be possibly related to our findings. Moreover, terrestrial insects collected from the lake shoreline and the stomach contents were screened for the presence of carnobacteria. The salmonids in these high-mountain environments feed mainly on terrestrial insects, which are considered possible vectors for carnobacteria that might catabolize the exoskeleton chitin. All insects tested negative for carnobacteria, but as a small number of samples were analyzed, their role as possible vectors of infection cannot be excluded. We investigated the main seasonal drivers of Phragmites australis decomposition dynamics in a freshwater karst lake subject to water level fluctuations. Physical and chemical parameters were monitored every two weeks for one year and P. australis decomposition rates were measured using the leaf bag technique during two seasons (autumn and spring). Decomposition dynamics were analyzed for the contribution of macrobenthic invertebrate colonizers and the influence of intermittent water level variations. Seasonal trends for changes in physical and chemical parameters were observed in relation to the underground origin of the water supply, which also affected the macrobenthic invertebrate communities. Decomposition rates k differed significantly between seasons (k = 0.0131 in autumn and k = 0.0115 in spring) but remained within a narrow range, reflecting the trend for changes in water temperature. Our findings show that the variability in decomposition rates was largely due to water level fluctuations (46.7% relative importance), rainfall (12.4% relative importance) and macrobenthic invertebrate communities dominated by shredders (23.9% relative importance; 40.9–93.7% of the collected samples). These remote and pristine bodies of water have served as invaluable sentinels of global change, repositories of geological history, and sources of freshwater for the surrounding regions. Yet, despite their seemingly imperturbable beauty and seclusion, these high-mountain lakes are not immune to the evolving forces of our changing world. Re-emerging threats, exacerbated by the relentless march of climate change and intensified human activities, cast a shadow over their future, posing profound challenges that demand our attention. The aim of this perspective is to shed light on the main emerging threats, raise awareness, and advocate for proactive actions aimed at preserving and protecting high-mountain lakes. These threats include climate change, retreat of glaciers, UV radiation, long-range transport of contaminants, impact of alien species and water usage for Alpine storage power stations. Additionally, increased tourism and recreational activities in these pristine environments can cause habitat disturbance, further endangering these unique ecosystems. The present perspective article also offers valuable perspectives on the necessity of monitoring and research efforts in high-mountain lakes. Such actions are crucial for comprehending the ecological well-being of these ecosystems, evaluating the consequences of environmental shifts, and devising successful conservation strategies. Alpine lakes are extreme ecosystems located in remote areas and populated by few but well-adapted species. Because of their remote location, they are often considered pristine, unpolluted ecosystems. Since the 1980s, however, they have been affected by global anthropogenic impacts. Macrobenthic invertebrates play a pivotal role in these ecosystems and can be used as bioindicators also for monitoring the accumulation of trace elements. We characterized the macrobenthic invertebrates community of Balma Lake (Cottian Alps, Northwest Italy) and Dimon Lake (Carnic Alps, Northeast Italy) in summer and autumn and measured the levels of nine trace elements (As, Cd, Cr, Cu, Fe, Ni, Pb, Se, Zn) in the most abundant taxa (Chironomidae and Oligochaeta in both lakes and Hirudinea in Dimon Lake) in both seasons. The highest levels of trace elements were recorded for Fe, Cu, and Zn according to their environmental availability and their function as essential elements. The total amount of trace elements was highest for the Chironomidae from both lakes compared to the other two taxa. As, Cd, Pb, and Zn were measured in sediment to calculate bioaccumulation factor (BAF) values. The amount of elements in sediment and macrobenthic invertebrates was higher for Dimon Lake, suggesting a greater flux via precipitation of contaminants from the lowland. The BAF values were decreased with increasing trace elements concentration in sediment, indicating mechanisms of elements excretion in biota where the environment is contaminated. The hypothesis that liver lipid accumulation in fish is an adaptive strategy to survive the winter in the high-altitude environment was assessed in this study. During summer 2019, specimens of Cottus gobio were sampled in 15 watercourses of Friuli Venezia-Giulia Region (Italy) to verify if hepatic steatosis is or not normally present in the species. To do this, hepatic vacuolization was assessed by histology using a semiquantitative score. Furthermore, C. gobio were also captured during the ice-free season at Dimon Lake (1872 m a.s.l.) and But Stream (520 m a.s.l.) to compare the trend in lipid accumulation: water temperature, hepatosomatic index (HSI), gonadosomatic index (GSI), Fulton's condition factor (K), and lipid area percentage (lipid %) were measured monthly. Findings revealed that liver steatosis is rather common in C. gobio. However, the trend in lipid accumulation of this species differed between Dimon Lake and But Stream. Based on the HSI and the GSI, the reproductive cycles differed in fish from the two environments (April–May in But Stream; May–June in Dimon Lake). While K values remained unchanged in the But Stream specimens, significant changes were recorded for Dimon specimens. The increase in lipid % from July to August in the Dimon Lake specimens coincided with greater food availability. With the rapid drop in lake water temperature in October, the lipid % decreased due to slower metabolic rate and lipid utilization from liver stores. There was a slight decrease in lipid % in the But Stream specimens, indicating that lipids were not being accumulated. Though mountain lakes are generally much less influenced by human activities than other habitats, global and local anthropogenic threats can alter their natural condition. The most alarming threats are climate change, water exploitation and abstraction, alien species introduction, and the medium-long range atmospheric transport of contaminates. Moreover, tourism and mountain farming are two other major sources of organic pollutants that can pose a threat to local aquatic biodiversity. Papers submitted to this Special Issue should be original contributions, with a focus on ecological and morphological characterization, environmental pressures (i.e., alien species introduction, environmental contaminates), and the use of bioindicators/tracers to inform adequate management plans. This study aimed to assess the biological and sanitary conditions of alien fish in a high-mountain lake (Balma Lake) located in the Cottian Alps. A single fish sampling session (August 2018) using gillnets collected 90 specimens of brook trout (Salvelinus fontinalis). Sex and age were determined (59 females and 31 males, age class 0+ to 4+). Regression analysis showed no difference in total weight and total length between males and females (ANCOVA: F = 0.453; p = 0.954). The mean condition factor (Kmean) decreased with increasing age for males and females. Terrestrial insects were the main prey found in the fish stomachs. The parasitological exam was negative, and the bacteriological exam was positive for Carnobacterium maltaromaticum and C. divergens in 33% of specimens. The total mercury, cadmium, and lead concentration in muscle tissue was within the maximum limit established by the European Commission for human consumption. The aim of this study was to describe liver alterations observed in a bullhead (Cottus gobio) population from a high-mountain lake (Dimon Lake; 1857 m a.s.l.) located in Carnic Alps (Northeast Italy). Two fish sampling campaigns (summer and autumn) were performed in 2017 to explore the possible causes of this phenomenon. In addition, to investigate the typical liver features of C. gobio, control specimens were captured from Degano Creek in the same seasons. Total length, weight, and liver weight were recorded in fish from both sampling sites. In addition, Fulton's condition factor (K) and hepatosomatic index (HSI) were calculated. Stomach contents were also analyzed. Liver from each specimen was sampled, and histological examination was carried out. Liver steatosis (L) (nuclear displacement and cytoplasm vacuolization) were histologically evaluated by assigning a semiquantitative severity score. A significant difference in L was found between Dimon Lake and Degano Creek in both summer (Mann–Whitney test; p = 0.0001) and autumn (Mann–Whitney test; p < 0.0001). Regarding HSI, a significant difference was also recorded between Dimon Lake and Degano Creek in summer (Mann–Whitney test; p < 0.0001) and also in autumn (Mann–Whitney test; p < 0.0001), but no seasonal change in K values was recorded between the two sites in both summer (Mann–Whitney test; p = 0.8589) and autumn (Mann–Whitney test; p = 0.6415). A significant positive correlation between HSI and L was found (ρS 0.573). The causes of this abnormality might be related to adaptation by the fish to the high-altitude environment, accumulating lipids in the liver to tackle the adverse season, which lasts for 6 months of the year. Alpine lakes are well known as "environmental sentinels", which are sensitive to anthropogenic impacts, and paleoecological studies provide useful information regarding the effects of local and global changes. In the present study we investigate, for the first time, the temporal variations in testate amoeba assemblages in relation to environmental factors and anthropogenic impacts in a high-altitude lake located in the Italian Alps (Balma Lake, Piedmont, Italy). The testate amoebae are well-suited as bioindicators and have been employed to reconstruct past climate and environmental changes from lake sediments. However, these organisms have not been thoroughly studied in alpine lakes, especially in Europe. We detected significant changes in testate amoeba assemblages along an alpine lake sedimentary profile over the last 2000 years. These changes were mainly associated with variations of trace elements, nutrients, and grain size. Moreover, five different time periods were identified and related to pluvial/drought events of the last 2000 years and to the "Late Antique Little Ice Age" (LALIA). Alpine lakes are originally fishless and fish introductions for recreational purposes could cause important changes in these ecosystems. However, the introduction of brook trout in Balma Lake during the 1970s seems to have had little influence on testate amoeba assemblages. The present work provides new insights for the evaluation of temporal changes in testate amoebae through paleoecological investigations. Similarly, the comparison of testate amoeba and chironomid proxies offered an important tool for analyzing recent climatic and environmental changes in alpine lakes (in lacustrine environments), especially for the European Alps.
