In an allozyme electrophoresis survey of 15 hypostomine species from the Itaipu Hydroelectric Reservoir, 25 loci from 14 enzyme systems were scored. Allozyme data allowed recording diagnostic genetic markers for all species analyzed and for some species groups within Hypostomus, a taxon which is taxonomically still unresolved in the Upper Rio Paraná basin. The mean expected heterozygosity of the species was considerably variable and hypotheses to tentatively explain this variation are discussed. A cladogram based upon the allelic frequencies of the species analyzed was produced by the continuous maximum likelihood method: Rhinelepis aspera and M. parananus were separated from the species of Hypostominae by a long branch length. Pterygoplichthys anisitsi was the sister of all the representatives of the genus Hypostomus. Within Hypostomus, two main clades were produced: in the first, H. cochliodon was the sister of the species comprising the H. plecostomus group, and in the second, the tree showed the following relationships: (H. albopunctatus (H. regani + Hypostomus sp. 3) + (H. margaritifer (H. microstomus (Hypostomus sp. 1 (H. ternetzi + Hypostomus sp. 2)). Hypostomus ternetzi and Hypostomus sp. 2 are referred to here as representatives of the H. ternetzi group.

Enzymatic analysis in Anopheles nuneztovari was made using four populations from the Brazilian Amazon and two from Colombia. The enzymes ME and XDH presented a monomorphic locus in all of the studied populations. EST and LAP presented a higher number of loci. In EST, genetic variation was observed in the five loci; LAP presented four loci, with allec variation in two loci. In IDH, three activity regions were stained, with genetic variation for locus Idh-1 in the Brazilian Amazon populations. A locus for MDH was observed, with genetic variation in the six populations. A region was verified for ACON, with four alleles in Sitronela and three in the other populations. PGM constituted one locus, with a high variability in the Brazilian Amazon populations. A locus was observed for 6-PGD with allelic variation in all of the populations with the exception of Tibú. Enzyme PGI presented two loci, both with genetic variability in the Tucuruí population. The enzyme alpha-GPD showed an activity region with polymorphism in the Tucuruí, Tibú and Sitronela populations. The phenotypic variations detected for these enzymes suggest that four (EST, LAP, ACON and PGM) possess monomeric structures and five (IDH, MDH, 6-PGD, PGI and alpha-GPD) dimeric structures in their proteins. These enzymes constitute in important markers to estimate variability and genetic divergence in natural populations of A. nuneztovari.

Isoenzyme electrophoresis was used as a method to determine genetic diversity in various M. esculenta cultivars collected in the Southwestern (SW) and Northwestern (NW) regions of the State of Parana, in the South region of Brazil, and in cultivars produced at the Agronomic Institute of Campinas (IAC), São Paulo State, Southeastern region of Brazil. The cultivars have been maintained by vegetative propagation for 5 years and are useful in production programs. A total of 28 loci in the acid phosphatase (ACP; EC 3.1.3.2), esterases (EST; EC 3.1.1.1), malate dehydrogenase (MDH; EC 1.1.1.37), and shikimate dehydrogenase (SKDH; EC 1.1.1.15) isozymes was analyzed. The proportion of polymorphic loci for NW, SW, and IAC cultivars was 57.14, 50.0, and 53.6% respectively. Genetic diversity calculated by Nei's genetic identity (I) showed high I values for the three M. esculenta subpopulations. The high degree of polymorphism expressed by cassava cultivars is highly relevant to stimulate breeding programs with M. esculenta species.

Three populations of Anopheles darlingi were studied for cuticular hydrocarbons, isoenzymes and patterns of peak biting activity. Differences were found in specimens from Costa Marques, a malaria endemic area; Dourado, a site with a very exophilic population and Juturnaíba, located near the type locality. Twelve hour collections from sunset to sunrise showed that An. darlingi from Costa Marques had a bimodal biting activity profile with a major peak at sunset and a minor peak at sunrise. At Dourado, the pattern was trimodal, with peaks at both morning and evening periods of twilight and near midnight. The Juturnaíba population showed a slight increase in activity near 2000 and 0100 h. Nei's genetic distances, determined by isoenzyme electrophoresis between pairs of populations, were low (D < or = 0.049). Using discriminant analysis for the cuticular hydrocarbons, 92.4% of the specimens from Costa Marques, 91.2% of the specimens from Dourado and 61.3% from Juturnaíba were correctly identified. Cuticular hydrocarbon and isoenzyme results matched very well: the smaller the Nei's distance, the more misidentifications occurred in the jackknife estimator used in the cuticular hydrocarbon analysis. This is the first report of cuticular hydrocarbon analysis in combination with isoenzymes to investigate neotropical anopheline species.

The enzyme profiles of three major Brazilian Trypanosoma cruzi zymodemes (Brazilian Z1, Brazilian Z2, Brazilian Z3) and of two principal Bolivian zymodemes were compared. Relationships were assessed both intuitively and by calculating genetic distances. One of the Bolivian zymodemes, Bolivian Z1, was closely related to the Brazilian Z1. The second Bolivian zymodeme, Bolivian Z2, was related to brazilian Z2 but differed from all the Brazilian zymodemes in the occurrence of typical heterozygous isozyme patterns in five out of 12 enzyme loci. Parental stocks and clones of Bolivian Z2 had the same putative heterozygous patterns. The evidence from enzyme profiles on the ploidy of T. cruzi and the possibility of recombination was considered. The presence of putative heterozygous patterns in Bolivian Z2 supported the hypothesis that T. cruzi is diploid. The definition of T. cruzi as a single polytypic species or as a species complex was considered to be dependent on the presence or absence of genetic recombination between or within the zymodemes, which has not been demonstrated in the ecotopes so far examined.

1. The three structural gene loci of human alcohol dehydrogenase have been studied in liver, jejunum and lung from 300 newborns in a triracially mixed population of Bahia, Brazil. 2. The frequency of the ADH23 allele was 0-1392, suggesting that the ADH23 allele is less frequent in Negroes. 3. A new ADH2 variant was identified. The electrophoretic pattern was interpreted as due to a new allele which is provisionally called ADH2Bahia. 4. By electrophoretic classification the 'atypical' variant was found in 2-8% of the sample. A question is raised regarding the ancestral origin of the 'atypical' variant in the population. Because this variant is common in Japanese it may have reached the present day population of Bahia through their American Indian ancestors. 5. Subjective estimation of the proportions of beta chains by giving scores to the liver isozymes alphaalpha, alphabeta and betabeta showed a clear relationship between the fetal weight and the beta chain activity. 6. The proportion of beta chains in the liver is significantly less when there is no enzyme activity in the lung, indicating some synchronous 'turning on' mechanism for alcohol dehydrogenase synthesis in both tissues.

The aim of this study was to investigate genetic polymorphism in Leishmania braziliensis population previously typed through isoenzyme electrophoresis, isolated from the same patient in two different moments: (A) before the beginning of treatment and (B) after treatment failure to meglumine antimoniate or reactivation after successful initial treatment. Fifteen pairs of isolates were assessed using the polymorphic molecular marker LSSP-PCR and following the phenetic analysis. The genetic profiles of the 30 samples were grouped in four clusters. Only two patients presented total identity in the A and B isolates. Most isolates presented similarity coefficients varying from 0.63 to 0.91. In this group of patients genetic polymorphisms could be observed indicating low similarity between the pairs of isolates. The results demonstrate the existence of genetic polymorphism between the samples isolated before treatment and after reactivation or treatment failure, suggesting a possible differentiation of the structure of the original parasite population which could be involved in the mechanisms of resistance to treatment or reactivation of lesions in the ATL. Abstract: The aim of this study was to investigate the naturally occurring bacteria from Culicidae larvae found in São Paulo state between 2006 and 2008 from the Tiete Ecological Park in the municipality of Caraguatatuba. Bacterial strains were obtained after surface sterilization of larvae followed by thermal treatment and incubation in bacteriological media under laboratory conditions. Identification was determined using cytomorphology, biochemical and physiological tests. Strains were characterized by qualitative evaluation of biological activity against Culicidae larvae, comparing protein profiles obtained by electrophoresis of crystal protoxins, electrophoresis of Lysinibacillus sphaericus isoenzymes, detection of the L2 fraction of Hemolytic BL enterotoxin and amplification of DNA using Multiplex-PCR to detect HBL, NHE, CytK, BceT and EntFM enterotoxins. The results obtained in this study demonstrated that the endosymbiotic bacterial diversity belonging to Bacillus and related genera associated with Culicidae larvae is limited to a number of species that does not vary throughout the seasons. Cold is one of the main problems for rice cultivation in Rio Grande do Sul because the vast majority of cultivars in use are of tropical origin. The occurrence of low temperatures combined with the susceptibility of the materials used can cause serious damage to the establishment of the crop reducing the plant stand. The objective of this study was to evaluate the physiological changes in different rice cultivars, when exposed to low temperature during germination. Seeds from four cultivars, BRS Agrisul, BRS Chuí, Lemont and Oro were used and evaluated regarding low temperature tolerance by the cold test. To evaluate the physiological changes, isoenzyme analyses of alcohol dehydrogenase and esterase were performed at temperatures of 25 °C (ideal for germination) and 13 °C (low temperature). The other characteristics evaluated were germination, germination speed, coleoptile length and dry weight at 25 °C and 13 °C. According to cold test results, seeds of the Lemont and Oro cultivars presented better tolerance to the low temperature. In the isoenzyme analysis, the cold caused an increase in the expression of the isoenzyme alcohol dehydrogenase and reduction in the esterase isoenzyme expression. For the other ratings, the cold caused damage to all the characteristics evaluated, especially in the BRS Agrisul and BRS Chuí cultivars. [ABSTRACT FROM AUTHOR]

Pelagic larvae are highly important for maintaining the gene flow among populations of sessile marine invertebrates. Colonial ascidians consist, exclusively, of brooding species, with lecithotrophic larvae that have a limited dispersal. As a result, there is a marked differentiation among populations. In this work, we used allozyme electrophoresis to access the genetic variation in four populations of Symplegma rubra, a colonial ascidian frequently found in the intertidal zone of Southeastern Brazilian coast. High variability was found at three of the four sites sampled, the exception being Praia Grande in the State of Rio de Janeiro. At this site, there was a great preponderance of clones, which possibly reflected the enclosed nature of the location and its low water circulation that reduce the dispersal capabilities of these animals. S. rubra did not conform to expectations for random mating (Hardy–Weinberg equilibrium): there was a deficit of heterozygotes that was more related to the small population size than to inbreeding processes, since F is analysis per locus revealed a deficiency of heterozygotes at only one locus— MDH*. The greatest variation in allele frequency was found for GPI-2*. Analyses of genetic variability revealed moderate differentiation among the populations ( F ST=0.051), which was unexpected for a species with a low dispersal capability. Rafting, a frequently underestimated means of dispersal, may be the main mode of gene flow in this species over large areas, since colonies of S. rubra are frequently seen growing on drift material and there is no evidence that the larva survive for a long time in the plankton. Thirty-one trypanosotomatid stocks were isolated from various sylvatic hosts and vectors in two different regions from the Parana´ State of Brazil. The stocks were analyzed by multilocus enzyme electrophoresis (MLEE) on cellulose acetate plates (22 genetic loci). All stocks were unambiguously attributed to Trypanosoma cruzi, and were found to be closely related to the formerly described zymodeme I (TC1 subgroup of T. cruzi). By comparison with other sylvatic cycles with similar sample sizes (Southern USA, Colombia, French Guiana), genetic variability among these stocks was very limited, with only two variable loci out of 22, and only three different multilocus genotypes. Population structure of T. cruzi in these cycles appears to correspond to a set of very closely related clonal genotypes. This very limited genetic variability could be due to a recent foundation of these populations (founder effect). Summary: Diffuse cutaneous leishmaniasis (DCL) is characterised by multiple and progressive cutaneous lesions, resistance to chemotherapy and Leishmania-specific T-cell anergy. We report the first autochthonous DCL case and the first human infection with Leishmania amazonensis in Rio de Janeiro State, Brazil, where only L. braziliensis is considered to be the causative agent of cutaneous leishmaniasis. Leishmania amazonensis was identified by multilocus enzyme electrophoresis and PCR-RFLP. Our case was diagnosed as DCL according to clinical, parasitological, histopathological and immunological criteria. These observations indicate that L. amazonensis is increasing its geographical distribution in Brazil, accounting for unusual clinical presentations in new transmission areas. [Copyright &y& Elsevier]
