Objective: The study was intended to explore whether Monosodium glutamate (MSG) induces oxidative stress on the liver of Wistar albino rats when fed chronically at three different doses, namely, low, mid, and high doses identical to human consumption doses in growing countries. The use of Monosodium Glutamate (MSG) as a food flavor enhancer is increasing worldwide despite its neurotoxic effects. Fluoxetine (FLX) and Rosemary extract (RE) are known to have beneficial neuroprotective properties. Rats were divided into five groups: control group; MSG group, rats received 2 g/kg/day intraperitoneal (i.p.) injections of MSG for seven days; RE/MSG group, rats received 50 mg/kg/day of oral RE for 28 days starting prior to MSG; FLX/MSG group, rats received 10 mg/kg/day of oral FLX for 28 days beginning before MSG; and RE/FLX/MSG group, received combined treatments as mentioned above. Rats underwent the Barnes maze test, in addition to histopathological, immunohistochemical, morphometric and ultrastructural evaluations for their hippocampi. MSG increased the number of errors and escaped latency in the Barnes maze test that was significantly minimized in the three treatment groups. The MSG group exhibited pyramidal cell (PC) degeneration, shrunken glial cells and massive vascular dilatation that were improved with RE and/or FLX treatment. The number of glial fibrillary acidic protein (GFAP)-immunopositive cells were increased, and the number of PCs was decreased in the MSG group, while these values were significantly reversed with the three treatment groups with the most significant improvement at RE/FLX/MSG one. Ultrastructurally, PCs were shrunken with degenerated nuclei, dilated endoplasmic reticulum, swollen mitochondria, and vacuolations in the MSG group that were improved with RE and/or FLX. In conclusion, the combined RE and FLX treatment can ameliorate the toxic effect of MSG on rat hippocampus probably through its antioxidant and anti-inflammatory effects.

Obesity indices, such as body weight, body length, Lee index, body temperature, food intake, fat weight, and leptin level, were examined. Mice that did not exhibit obesity were defined as the obesity-resistant group. The morphological changes of white adipose tissue were observed by hematoxylin and eosin staining, and expression levels of PR domain containing 16 (Prdm16) and uncoupling protein-1 (Ucp-1) in white adipose tissue were measured by Western blot.Results: Obesity-resistant mice fed a high-fat diet showed resistance beginning at week 5 along with lower weights and lengths than those in the obesity group from weeks 5 to 12. MSG-induced obesity-resistant mice showed features consistent with resistance to obesity from week 1 along with higher body lengths relative to the obesity group; however, the weight difference was not significant until week 10, when body weights decreased significantly in obesity-resistant mice. The Lee index was lower in obesity-resistant mice than in the obesity group and the normal group, further suggesting obesity resistance. Additionally, obesity-resistant mice showed higher levels of leptin, whereas obese mice induced by a high-fat diet showed leptin resistance. Furthermore, Prdm16 and Ucp-1 levels were both downregulated in the obesity group and upregulated in obesity-resistant mice, showing that white fat browning was highest in obesity-resistant mice.Conclusion: The phenotypes of mice with DIO and MSG-induced obesity differed. People in the fast-food era rely on pre-packaged foods and engage in limited physical activity, which leads to a shift in eating patterns. Monosodium glutamate (MSG), a dietary ingredient used in this sort of cuisine, has been found to be hazardous to both experimental animals and humans. The objective of this study was to explore at the unnecessary changes caused by consuming MSG in secret and exceeding the recommended dosage. Hence, we decided to evaluate the impact of MSG by using three different doses (200, 400, and 600 mg/kg body weight orally) for 28 days in rats. We uncovered that all three MSG dosages result in a rise in body weight, dyslipidemia, inflammatory response, and hepato-cardiac marker enzymes, all of which imply hepatic and cardiac toxicity. Furthermore, changes in redox status suggest oxidative stress, which was higher in all three MSG dosages although not as much as in the MSG-600 group when compared to control. Such effects eventually manifested themselves in tissue architecture of the liver and heart, resulting in severe hepato-cardiac derangement, but the degree of tissue damage was greater in the MSG-600 group. As a result, it is possible that MSG has a negative influence on the liver and heart. However, the MSG-600 group showed a substantial effect, indicating that MSG should not be used in food preparation. Therefore, the findings of the study may aid in the formulation of health-care strategies and serve as a warning to the general public regarding the use of MSG in daily diet.

Umami refers to the savoury taste that is mediated by monosodium glutamate (MSG) and enhanced by inosine monophosphate and other nucleotides. Umami foods have been suggested to increase the risk for obesity and metabolic syndrome but the mechanism is not understood. Here we show that MSG induces obesity, hypothalamic inflammation and central leptin resistance in male mice through the induction of AMP deaminase 2 and purine degradation. Mice lacking AMP deaminase 2 in both hepatocytes and neurons are protected from MSG-induced metabolic syndrome. This protection can be overcome by supplementation with inosine monophosphate, most probably owing to its degradation to uric acid as the effect can be blocked with allopurinol. Thus, umami foods induce obesity and metabolic syndrome by engaging the same purine nucleotide degradation pathway that is also activated by fructose and salt consumption. We suggest that the three tastes—sweet, salt and umami—developed to encourage food intake to facilitate energy storage and survival but drive obesity and diabetes in the setting of excess intake through similar mechanisms.

Introduction: The effect of orally consumed monosodium glutamate (MSG), which is a common additive in the food industry, on the cochlea has not been investigated. The present study aimed to investigate the possible cochleotoxic effects of oral MSG in guinea pigs using electrophysiological, biochemical, and histopathological methods. Methods: Thirty guinea pigs were equally divided into control and intervention groups (MSG 100 mg/kg/day; MSG 300 mg/kg/day). At 1 month, 5 guinea pigs from each group were sacrificed; the rest were observed for another month. Electrophysiological measurements (distortion product otoacoustic emission [DPOAE] and auditory brainstem response [ABR]), glutamate levels in the perilymph and blood samples, and histopathological examinations were evaluated at 1 and 2 months. Results: Change in signal-to-noise ratio at 2 months was significantly different in the MSG 300 group at 0.75 kHz and 2 kHz (p = 0.013 and p = 0.044, respectively). There was no statistically significant difference in ABR wave latencies of the guinea pigs given MSG compared to the control group after 1 and 2 months; an increase was noted in ABR thresholds, although the difference was not statistically significant. In the MSG groups, moderate-to-severe degeneration and cell loss in outer hair cells, support cells, and spiral ganglia, lateral surface junction irregularities, adhesions in stereocilia, and partial loss of outer hair cell stereocilia were noted. Conclusion: MSG, administered in guinea pigs at a commonly utilized quantity and route of administration in humans, may be cochleotoxic.

The present study was designed to examine the possible modulatory effect of Fish, Walnuts or Fenugreek Oils against Attention Deficit Hyperactivity Disorder (ADHD)-like Behavior induced by Monosodium Glutamate (MSG) in Rats.lt;bgt;Materials and Methods:lt;/bgt; Fifty weaning rats were divided into five groups, (each group contain 10 rats) as follows: Group 1: Normal control rats were fed on a balanced diet. Groups from 2-5 rats were fed on a balanced diet+MSG (0.4 g kglt;supgt;lt;/supgt;lt;supgt;1lt;/supgt; diet), Group 2 served as a positive control group whereas group 3, 4 and 5 treated with Fish, Walnuts and Fenugreek oil, respectively, (200 mg kglt;supgt;lt;/supgt;lt;supgt;1lt;/supgt; b.wt.) by intra-gastric tube. Biochemical and behavioural parameters were tested as well as microscopic examination of brain tissue was done.lt;bgt;Results:lt;/bgt; MSG ingestion caused marked disruption in locomotors activity, memory function and brain tissue structure along with significant abnormalities in some bio-markers and reduction in the gene expression level of Bcl-2 in brain tissue. However, treatment with the tested oils showed remarkable effect by reversing the condition.lt;bgt;Conclusion:lt;/bgt; Dietary supplementation with walnut; fenugreek or fish oils at the tested dose could modulate the condition of ADHD in rats.

Monosodium glutamate (MSG) is a flavoring food additive that’s found in many foods like chips, and fast foods. The aim of this study was to demonstrate the injurious effects of MSG on the adrenals of the female albino rats and its potentiality to cross the placental barrier carrying its injurious effects to their pups. Twenty-five female albino rats weighing about (200) g were mated then separated into two groups. The control group (10 rats), and MSG-treated Group(15 rats) received 10 g/kg MSG orally, dissolved in water at a concentration of 5 g/100 ml, starting on day 7 to 20 day of gestation.A day after delivery, animals were weighed, blood samples collected then sacrificed, and the adrenals were collected and examined by the light and electron microscope. Statistically, there was a significant increase in the body weight and decrease in adrenal weight of MSG-treated mothers and their pups, as well as a decrease in the corticosterone level in MSG-treated mothers. Histologically, MSG-treated mothers and pups showed many degenerative changes, with thinning of the capsule that was confirmed by Masson trichrome stain, a disorganized structure of Zona Golmeruloza and Zona Fasciculata, and areas of shedding and hemorrhage were noticed. Shrunken nuclei and degenerated mitochondria with disrupted cristae and penetration of the lipid droplets to form “myelin-like” structures were noticed by E/M examination. High intake of monosodium glutamate (MSG) –containing foods was reported as a major health problem in Egypt. Thus, dietary interventions aim to prevent the deleterious effects of this additive were badly needed. This study was carried out to investigate the effect of concurrent consumption of MSG and Spirulina (S. platensis) on body weight as well as hepato-renal functions and histopathology in female rats. Twenty adult female albino rats were divided into four equal groups, including the control group, while groups 2 to 4 were administered 6 mg MSG/kg body weight daily, and kept untreated (group 2), or concurrently fed pelleted balanced diet supplemented with 0.5 and 1 % of Spirulina powder for 6 weeks (groups 3 and 4, respectively). At the end, body weight gain, feed intake and feed efficiency ratio were calculated. Liver and kidney functions were determined in sera, while oxidative stress markers were determined in liver and kidney tissue homogenates. Moreover, specimens from liver and kidney of sacrificed rats were histopathologically examined. On the other hand, phenolic profile of Spirulina powder was identified and quantified by HPLC. Findings indicated that MSG consumption resulted in overweight, liver and kidney dysfunction along with oxidative stress, which was further confirmed by histological staining. Due to the high content of phenolic compounds rather than other antioxidant agents, Spirulina powder exerted anti-obesity properties and alleviated the toxic hepato-renal effects caused by MSG consumption. So, dietary supplementation of MSG –containing food products with Spirulina powder is recommended to prevent its accompanied health abnormalities.

Introduction: Monosodium glutamate (MSG) is one of the most widely employed food enhancers. Although the umami compound, controversy persists regarding the effects of MSG intake on body weight. Chronic MSG intake may result in excessive body weight gain and obesity. Consumption of MSG result in organ damage, cardiovascular disease, oxidative stress, and also risk factors for obesity. This study aims to determine the effect of oral MSG on obesity in adult male Wistar rats (Rattus norvegicus).Methods: This true experimental study used the post-test control group design. Twenty-four adult male Wistar rats were randomly divided into four groups: control (received distilled water), Group 1 (MSG 0.378 mg/gr BW), Group 2 (0.756 mg/gr BW) and Group 3 (1.512 mg/gr BW). The obesity parameter was obtained by the Lee index. Kruskal-Wallis test follows by Mann-Whitney test were used to compare the Lee index between groups.Results: Lee’s index mean for each group was 358.4%, 314.1%, 287.8%, and 320.9%, respectively. The Kruskal Wallis test showed a significant difference in the Lee index between groups (p = 0.043). High-dose Monosodium Glutamate (MSG) expo sure increases the estrogen level in pregnant rats. However, there are limited data available on whether the MSG-related maternal hormonal effects can affect male litters' genitalia phenotype. This study aimed to analyze the impact of MSG on estrogen level in pregnant rats and anogenital distance in male pups. Experiment for this study was performed at the animal facility of Biomedical Laboratory at the Faculty of Medicine, Universitas Andalas, from April 2019 to February 2020. Pregnant Wistar rats were given MSG orally at 2 and 4 mg/g body weight (BW) for 20 days. On day 21, pregnant rats were sacrificed and blood was drawn intracardially. Estradiol serum level was measured by ELISA. Male pups were counted, and the anogenital distance (AGD) was measured. Maternal serum estradiol levels were statistically analyzed by One-Way ANOVA and the AGD of male litters were analyzed by the Kruskal-Wallis test. BackgroundMonosodium glutamate (MSG) is frequently consumed as a flavor enhancer or food additive. Possible damages induced by MSG effects on some organs have been stated in experimental animal models. The aim of the present study was to evaluate the protective effects of L-carnitine (L-ca) on the renal tissue in MSG-Induced Rats.MethodsIn this regard, 60 male rats were randomly divided into six groups (n = 10/each): 1 (Control); 2 (sham); 3 (L-carnitine 200 mg/kg b.w); 4 (MSG 3 g/kg b.w); 5 (MSG + L-carnitine 100 mg/kg); and 6 (MSG + L-carnitine 200 mg/kg). After 6 months, the rats were sacrificed, the blood sample collected and the kidneys harvested for evaluation of biochemical analytes, genes expression, and histopathological changes.ResultsMSG significantly increased the serum level of MDA, BUN, creatinine, uric acid and renal Caspase-9, NGAL and KIM-1 expression, but it decreased the serum activity also renal expression of SOD, catalase, GPX, and Bcl-2 expression compared to the control group. Treatment with L-ca significantly reduced the serum BUN, creatinine, uric acid and MDA level and increased catalase, GPX and SOD compared to the MSG group. However, only administration of L-ca 200 significantly decreased the caspase-9, NGAL and KIM-1; also, it increased the Bcl-2 expression in the kidney compared to the MSG group.ConclusionsOur findings indicated that L-carnitine had a major impact on the cell protection and might be an effective therapy in ameliorating the complications of the kidney induced by MSG via its antioxidant and anti-apoptotic properties.

Herein, Vernonia amygdalina stem ethanolic extract, VASEE, was elucidated for its antibacterial activity in vitro by disk-diffusion technique and, in vivo effect on some hematological parameters of normal and monosodium glutamate, MSG-intoxicated rats. For in vivo study, 20 male albino rats assigned into five groups (A, B, C, D, and E) were, for 14 days, respectively administered MSG (8000 mg/kg body weight, bw), VASEE (200 mg/kg bw), Control (Vital feed and tap water), MSG (8000 mg/kg bw + VASEE, 200 mg/kg bw), and MSG (8000 mg/kg bw + VASEE, 400 mg/kg bw). In vitro, the VASEE-related activity against the tested bacterial pathogens was significant (P < 0.05), dose dependent and comparable to that by the standard antibacterial drug, Ciprofloxacin. In vivo, VASEE compared to control and MSG groups improved (P < 0.05) and, notably at the highest tested dose modulated the monosodium glutamate intoxication-related effect on, the studied hematology of rats. Thus, VASEE exhibited potent activity against the tested bacterial pathogens, improved, and potentially modulated MSG-intoxication-related effect on, the rats’ hematological functions. The study underscored a promising antibacterial application of the extract of hitherto wasted bitter leaf stem that could offer novel therapeutic benefits on the hematology of especially MSG-intoxicated rats, warranting further studies. Monosodium glutamate (MSG) is a common flavor enhancer and stabilizer for ready-made or packaged foods. This research investigated the impact of MSG on the maternal and fetal liver. The present study was carried out on sixteen mature female Albino rats and eight male rats of reproductive age. The control group was dissected on day 20 of gestation. MSG group was administrated MSG daily at a dosage of 1 g/5 mL/kg body weight from day 0 to day 20 of gestation. The liver function and lipid profile of the control and treated mothers were investigated in the blood sera. The levels of nitric oxide (NO), tumor necrosis factor (TNF-α), superoxide dismutase (SOD), and reduced glutathione (GSH) activities in the liver homogenate of maternal and fetal tissue were assayed, in addition to histopathological, histochemical and immunohistochemical studies were done to the liver tissue. The activities of liver functions and lipid profile significantly altered in the treated mothers with MSG. MSG significantly reduced the SOD and reduced GSH activities in addition to the elevated TNF-α and NO in liver tissue of pregnant mothers and their fetuses. Severe histopathological alterations were observed in both maternal and fetal liver tissues of MSG-treated groups. Moreover, histochemical observations showed a reduction of total polysaccharides in the liver of pregnant rats and fetuses. A significant increase in the percentage area of positive immunoreaction for caspase 3 was observed in the liver of treated rats with MSG compared to the liver of the control. The liver of fetuses treated with MSG revealed an alteration like their mother. This study showed that during the gestational period MSG exposure resulted in several biochemical, histological, and histochemical changes in the maternal and fetal liver tissues which emphasize the toxic effect of MSG.

Obesity is considered one of the major global health problems and increases the risk of several medical complications, such as diabetes and mental illnesses. The present study investigated the effect of 7-chloro-4-(phenylselanyl) quinoline (4-PSQ) on obesity parameters, behavioral and neurochemical alterations in hypothalamic obese rats. Methods: Male Wistar rats received subcutaneous neonatal injections of monosodium glutamate (MSG, 4g/kg) or saline. After the Lee Index evaluation, rats were divided into groups and treated with 4-PSQ (5 mg/kg, intragastric route) or canola oil once a day (post-natal days (PND) 60→76). Open-field, elevated plus-maze, forced swim task, object recognition/location memory, and stepdown inhibitory avoidance tasks were conducted from PND 66 to 74. On PND 76, rats were euthanized and epididymal fat, blood, cerebral cortex, andhippocampus were removed. Blood biochemical parameters and cortical/hippocampal acetylcholinesterase (AChE) and Na /K -ATPase activities were assessed. MSG increased the Lee Index characterizing the chemically induced hypothalamic obesity model. 4-PSQ reversed the increases of epididymal fat, blood glucose, and triglyceride levels caused by MSG exposure. 4-PSQ attenuated anxiety-like and depression-like behaviors induced by neonatal administrations of MSG. Memory deficits found in MSG-obese rats were reversed by treatment with 4-PSQ. Neurochemical alterations produced by MSG evidenced by stimulation ofNa+/K+-ATPase and AChE activities in the cerebral cortex and hippocampus of rats were normalized by 4-PSQ treatment. In brief, 4-PSQ therapy improved hypothalamic obesity-related parameters, as well as psychiatric symptoms, cognitive impairment, and neurochemical alterations found in obese rats.

Male infertility is considered one of the most critical health problems that are expected to expand worldwide. Ulva lactuca is a species of green seaweeds which is known to be a rich source of many important nutrients. Accordingly, this study is designated to investigate the therapeutic role of Ulva lactuca water fraction (UL) against monosodium glutamate (MSG)-induced male reproductive system disorders in male rats. Ulva lactuca methanolic crude extract was prepared firstly, and then water-dissolved compounds of this crude methanolic extract were separated. Ulva lactuca water fraction active phenolic compounds were determined using high-performance liquid chromatography (HPLC). Thirty-two male rats were divided equally into four groups; male infertility was induced in sixteen experimental animals by MSG at dose of 15 mg/Kg for 45 days. Eight infertile animals were treated with 100 mg/Kg of Ulva lactuca water fraction for 30 days. The rest of the animals were divided into two control groups; one control group (eight animals) was used to study the effect of UL on healthy rats at dose of 100 mg/Kg for 30 days and healthy control group (eight animals). Semen quality parameters (concentration and motility ratio), serum testosterone, prostate-specific antigen (PSA), and phosphatases were estimated by using standard protocols. Moreover, prooxidants and endogenous antioxidant enzymes were measured in prostate and testis homogenates. In addition, relative expression of pro-inflammatory genes (inducible nitric oxide synthase (i-NOS), cyclooxygenase-2 (COX-2), tumor necrosis factor-α alpha (TNF-α), and tumor protein (P53)) were assessed in testicular and prostatic tissues. Finally, histological alterations were measured by hematoxylin and eosin (H&E) stain. Results revealed that Ulva lactuca water fraction contains active phenolic constituents responsible for its antioxidant bioactivity. Oral administration of MSG significantly induced histological alterations. Oxidative stress was observed with elevated levels of nitric oxide (NO), thiobarbituric acid reactive substances (TBARS), and xanthine oxidase (XO) activity in both testis and prostate tissues. MSG adversely affected prostate function via elevation of PSA, prostatic acid phosphatases (PAPs), and total acid phosphatases (TAPs). In addition, it upregulated pro-inflammatory genes in testis and prostate tissues. Meanwhile, MSG reduced serum testosterone, semen quality, and antioxidant enzyme activities (glutathione peroxidase (GPx), glutathione-s-transferase (GST), and superoxide dismutase (SOD)). Treatment with UL notably ameliorated the state of oxidative stress and downregulated the expression of pro-inflammatory gene markers. This study highlighted the potential efficacy of Ulva lactuca water fraction on MSG-induced male infertility in rats.

Background: Monosodium Glutamate (MSG) is one of the most commonly used flavor-enhancing substances that may lead to neurological disorders. Objectives: The present work aimed to evaluate the role of L-ascorbic acid (AA) and -tocopherol (T) on the MSG-induced memory and neurobehavioral changes in rats. Methods: Thirty male Wistar albino rats were randomized into five equal groups: (1) control group, (2) MSG group received MSG (2mg/g BW) daily, (3) MSG+A group received MSG as in MSG group, and AA (100 mg/kg BW) daily, (4) MSG+T group received MSG as in MSG group, and T (600 mg/kg BW) twice weekly, and (5) MSG+AT group received MSG as in MSG group, AA as in MSG+A group, and T as in MSG+T group. After 3 weeks, neurobehavioral changes were assessed by open field test and Y maze. Oxidative stress markers were estimated, and immunohistochemistry was studied in hippocampal region. Results: MSG resulted in impairment of memory and induction of anxiety, with increased hippocampal malondialdehyde and decreased superoxide dismutase and glutathione peroxidase. Treatment with AA or T improved all the measured biochemical parameters, and the MSG-induced hippocampal degenerative changes, with decreased glial fibrillary acidic protein (GFAP) and synaptophysin expression. Combined administration of both vitamins was more effective in amelioration of MSG-induced impairments rather than taking AA or T alone. Conclusion: Both AA and T exhibit protective effects against neurobehavioral changes, oxidative stress and hippocampal degenerative changes induced by MSG toxicity, with more potent efficacy of their combination.

This study was carried out to investigate the ameliorative effect of L-carnitine on monosodium glutamate (MSG)-induced testicular toxicity in male mice. Sixty adult male mice were randomized into 6 groups (n = 10). In addition, to the control male mice group (Gp1) that orally administered distilled water, Gp2 mice received 150 mg/kg/day L-carnitine for 35 days. Monosodium glutamate (MSG) was orally administered to male mice at doses of 0.3 and 0.6 mg/g body weight individually (Gp3 & Gp4) and in combination with 150 mg/kg body weight of L-carnitine for 35 days (Gp5 & Gp6). The morphometric parameters, histopathological findings and immunohistochemical studies for PCNA, Ki-67 and Claudin-1 of the testis tissue demonstrated that L-carnitineattenuated and ameliorated the alterations in testicular tissues caused by MSG exposure. Conclusions: The findings of the present study indicated that treatment of male mice with L-carnitine banned MSG-induced testicular toxicity by improving testicular structure status.

The consequences of dietary intake of significant amounts of monosodium glutamate (MSG) are the excess body weight; structural and functional disorders of the central nervous system, liver, kidneys. We have not found information about the influence of excessive using of the MSG by a woman during pregnancy and lactation on the fetus and infants. The aim of the study was the experimental evaluation of the MSG consumption consequences during pregnancy and lactation to the offspring health. Material and methods. The offspring of 3-month old pregnant female white Wistar rats, who received 1% MSG solution (200 mg per kg of body weight per day) ad libitum as the source of liquid during the pregnancy and lactation, have been studied (MSG group). The control group included offspring of pregnant female rats that received water as the source of liquid. In 25-day-old offspring histological examination and morphometry of the nucleo-nucleolar apparatus of neurons in the neocortex of the proper parietal lobe, cardiomyocytes of the subendocardial zones of the left and right ventricles have been performed. Gravimetry have been also carried out (body weight and weight of brain, heart, liver, kidney, thymus and spleen); mitotic activity of anterior corneal epithelium has been evaluated, the state of erythrocyte membranes have been analyzed by the method of acid erythrograms; behavioral tests "Open field", "The elevated plus-maze test", "Hanging on a horizontal wire" have been performed. Results. MSG consumption during pregnancy and lactation led to an increase of brain (by 19.1%) and kidneys (by 7.8%) relative masses; masses of thymus and spleen were decreased. Significant decrease of locomotor activity and increase of time of hanging in "horizontal wire test" were registered. A histological study showed an increase in the number of nucleoli in the neurons of the V layer of the neocortex of the proper parietal lobe (control - 1.56±0.09; MSG group - 1.81±0.07, р=0.03); decrease of the nucleolar parameters of cardiomyocytes; increase of mitotic activity of anterior corneal epithelium (control - 4.021±0.612‰; MSG group - 6.985±0.889‰, р=0.019). A decrease of the resistance of erythrocyte membranes to acid hemolysis was also registered. Conclusion. The results obtained indicate the effect of oral consumption of MSG food additive during pregnancy and lactation on the organism of the offspring.

The pineal gland synthesizes melatonin exclusively at night, which gives melatonin the characteristic of a temporal synchronizer of the physiological systems. Melatonin is a regulator of insulin activities centrally and also peripherally and its synthesis is reduced in diabetes. Since monosodium glutamate (MSG) is often used to induce the type 2 diabetic and metabolic syndrome in animal models, the purpose of this work is to evaluate the potential effects of MSG given to neonates on the pineal melatonin synthesis in different aged male and female rats. Wistar rats were subcutaneously injected with MSG (4mg/g/day) or saline solution (0.9%) from the second to eighth post-natal day. The circadian profiles both melatonin levels and AANAT activity were monitored at different ages. Body weight, naso-anal length, adipose tissues weight, GTT, ITT and serum insulin levels were also evaluated. Typical obesity with the neonatal MSG treatment was observed, indicated by a great increase in adipose depots without a concurrent increase in body weight. MSG treatment did not cause hyperglycemia or glucose intolerance, but induced insulin resistance. An increase of melatonin synthesis at ZT 15 with phase advance was observed in in some animals. The AANAT activity was positively parallel to the melatonin circadian profile. It seems that MSG causes hypothalamic obesity which may increase AANAT activity and melatonin production in pineal gland. These effects were not temporally correlated with insulin resistance and hyperinsulinemia indicating the hypothalamic lesions, particularly in arcuate nucleus induced by MSG in early age, as the principal cause of the increase in melatonin production.

Monosodium glutamate (MSG) is one of the most common food additives extensively used as a flavor enhancer. MSG induced lipid peroxidation and inflammation. The present study aimed to assess the neurotoxicity, testicular impairment, inflammation and apoptosis induced by MSG. Thirty adult male Wistar rats, weighing about 180-200 g were assigned equally into five groups, each consists of six rats. Animals of Group I are controls and they received distilled water, whereas animals of Groups II, III, IV and V were given oral daily doses of MSG 0.8, 1, 2 and 3 g/kg body weight, respectively for consecutive 70 days. Administration of different doses of MSG revealed a significant elevation in the levels of malondialdehyde (MDA), nitric oxide (NO), β amyloid 1-42, proinflammatory cytokines (IL-6, TNF-α), cholesterol and sperm abnormality while it showed reduction in the level of GSH and SOD, CAT and GST antioxidant enzymes activities, sperm count and sperm motility. MSG led to disruption in neurotransmitter levels; serotonin, norepinephrine, glutamate and GABA, also disorders in sexual hormones; testosterone, FSH and LH. The present results were confirmed by histological and immunohistochemical observations that obviously designate the neurodegeneration and reproductive toxicity. In conclusion, administration of low and high doses of MSG provoke deleterious effects on oxidant/ antioxidant markers, neurotransmitters, inflammatory cytokines, sexual hormones, brain and testes structures. Prominence hazards of lasting exposure to low and high doses of MSG on neuronal and testicular health. Therefore, its use should be restricted.

Monosodium glutamate (MSG) at high concentration has been reported to alter the physiological and biochemical states of animals and humans. Ganoderma lucidum (G. lucidum) is a polypore mushroom reported to possess many medicinal attributes such as anticholesterolemia and the control of hormonal disorders. The present study investigated the effect of water extract of G. lucidum in the changes of haematology, oestradiol, cholesterol and protein levels of Wistar rats induced by MSG. Haematological analysis was determined from plasma, while oestrogen, serum total protein and cholesterol levels were determined from the serum of the rats. Results showed that MSG significantly raised the level of oestrogen (62.5 ± 0.28 pg/mL) in the rats which was significantly reduced in the rats fed with MSG for 30 days before treating them with the extracts of G. lucidum (30.85 ± 12.94 pg/mL–44.15 ± 0.92 pg/mL) and in rats fed concurrently with MSG and G. lucidum. The cholesterol level was significantly reduced in the rats treated with MSG and G. lucidum (200 mg/kg) concurrently compared to rats fed with MSG alone. The white blood cell (WBC) and red blood cell (RBC) levels were within normal in rats fed with both MSG and G. lucidum as in the control group while the rats fed with MSG only had low WBC, neutrophil (NEU) and RBC. This could imply that G. lucidum ameliorates the effect of MSG on serum oestrogen, serum cholesterol, WBCs, NEU, platelets and lymphocytes.

This study investigated the effect of high doses of monosodium glutamate (MSG), a known food additive on hepatic, memory and locomotor functions in mice, and the ameliorative potentials of Jobelyn® (JB), a unique dietary supplement. Twenty four male Swiss mice divided into 4 groups (n = 6) were given MSG (2, 4 and 8 g/kg) or normal saline (10 mL/kg) orally for 14 days. In the intervention study, another set of 30 male Swiss mice distributed into 5 groups (n = 6) received normal saline, MSG (8 g/kg) alone or in combination with JB (5, 10 and 20 mg/kg) orally, for 14 days. Memory and locomotor functions as well as brain oxido-nitrergic stress biomarkers were then assessed in both studies. The hepatic oxido-nitrergic stress biomarkers, liver enzymes functions and histomorphology of the liver were also assessed. MSG (2, 4 and 8 g/kg) produced memory dysfunction, hyperlocomotion, increased malondialdehyde and nitrite levels accompanied by decreased antioxidant status in the brain and hepatic tissues. MSG-treated mice had increased hepatic enzyme activities (alanine aminotransferase, aspartate aminotransferase and alkaline phosphatase) and distorted cyto-architectural integrity of the liver. These findings further suggest that MSG compromised hepatic functioning, which might also contribute to its neurotoxicity. However, JB (5, 10 and 20 mg/kg, p.o) attenuated the memory deficit, hyperlocomotion, increased oxido-nitrergic stress responses in the brain and hepatic tissues induced by MSG (8 g/kg, p.o). JB also normalized hepatic enzymes activities and histomorphological changes in MSG-treated mice. Taken together, JB mitigated MSG-induced toxicity through mechanisms relating to enhancement of cellular antioxidant-machineries and normalization of hepatic enzymatic functions.

Monosodium glutamate (MSG) toxicity is fast becoming a global health challenge due to the increase in its consumption as a food additive. This study investigated the effect of consumption of MSG and treatment with graded doses of omega 3 fatty acids (ω-3). Forty-eight male Wistar rats (n=8) grouped into six; control, MSG, MSG + Low dose of ω-3 (LD ω-3); MSG + High dose of ω-3 (HD ω-3), LD ω-3, and HD ω-3 were used for this study. MSG was administered at 4 g/L/day in their drinking water for 6 weeks, while ω-3 was administered at low and high doses of 100 and 300 mg/kg BW, p.o. respectively for 4 weeks. Results revealed that administration of MSG induced imbalance in lipid metabolism, oxidative stress and hepatic dysfunction. These were revealed by significant decreases in TG, HDL-C, CAT, GSH, albumin and total protein; but, significant increases in LDL-C, MDA, AST, ALT, ALP, and total bilirubin (TB), compared to control group. Administration of graded doses of ω-3 following treatment with MSG was characterized with significant reductions in ALT, ALP, TB and MDA. The administration of ω-3 showed no effects on the antioxidant indices. Conclusively, LD ω-3 is a potent ameliorative supplement which can be administered after pre-exposure to MSG.

Background: Chlorogenic Acid (CA) has diverse, recognized health effects. Objective: This study aimed to explore the effects of CA on fat reduction and the underlying mechanism of these effects. Materials and Methods: First, we established a Monosodium Glutamate (MSG)-induced obesity mouse model and subjected the mice to 4 weeks of CA gavage. Then, we established an oleic acidinduced model of human fatty liver in HepG2 cells, and administered a CA intervention to the cells for 48 h. Finally, we used Oil red O staining, biochemical detection kits, RT-PCR and Western blot analysis to evaluate the effects of CA on fat reduction and on related pathways. Results: The CA treatment could reduce fat accumulation in the liver and reduce blood lipid levels. In addition, CA decreased the mRNA and protein levels of peroxisome proliferator-activated receptor gamma, coactivator 1 α (PGC-1α) and Uncoupling Protein 1 (UCP1) in the MSG-induced obesity mouse model and the oleic acid-induced HepG2 cells. Conclusion: Based on the above results, we deduced that CA could reduce body weight and fat deposition in vitro and in vivo and that the mechanism may be related to the PGC-1α/UCP-1 pathway. CA can be developed as a drug to lower blood lipids and to treat obesity.

Increased exposure to food additives may exhibit harmful effect on the male reproductive system. Neonatal application of high doses of popular taste enhancer monosodium glutamate (MSG) to different kinds of animals cause lesions of the hypothalamic nuclei and the retina. Later in adulthood, animals exhibit a series of neuroendocrine disorders (stunted growth, obesity and decreased fertility). The mechanism of MSG action is not completely explained yet. We hypothesized that high concentration of MSG could alter permeability of neural membrane for calcium. The objective of our study was to find out whether the pretreatment with diltiazem, a calcium channel blocker, could prevent harmful effect of MSG in the rat testes. Male rat pups were treated with: 0.9% sodium chloride (C group), 4 mg/g BW of MSG (M group), 5 mg/g BW of diltiazem (D group) and diltiazem 5 mg/g BW with MSG (DM group) on 2nd , 4th , 6th , 8th, 10th postnatal day. Animals were sacrificed at the age of six months. MSG treatment resulted in: stunted growth (decreased naso-anal length and tail length; p < 0.05), decreased relative testis weight (p < 0.05), and increased adipose tissue mass (Lee index; p < 0.05), testicular atrophy and decreased histomorphometric parameters (tubular area, tubular perimeter, Feret diameter, tubular diameter, epithelial height; p < 0.001). The rats of C, D and DM groups had normal testicular histology and morphometric parameters. Pretreatment with diltiazem has prevented the development of morphological disorders of testes. Our results suggest that calcium overloading may play an important role among mechanisms of MSG testicular toxicity.

Objectives: Monosodium Glutamate (MSG) is one of the world’s most extensively used food additives which are ingested as part of commercially processed foods. MSG is a slow excitotoxin food additive which can cause generation of numerous amounts of free radicals which affects many organs such as heart. Vitamin C is an antioxidant protects the body against oxidative stress. Methodology: the experiment was carried out on 100 albino rats weighing about (150-200) g. The animals randomly were divided into equal 4 groups (n=25/group).The first group was served as control , the second group was administered oral doses of Vit.C (200 mg/Kg), the third group was administered oral doses of Monosodium Glutamate (MSG) dissolved in distilled water equal to 60 mg/kg daily (60 mg/Kg) the fourth group was administered oral doses of Monosodium Glutamate (MSG) dissolved in distilled water equal to 60 mg/kg daily (60 mg/Kg) and Vitamin C (200 mg/Kg) for 90 days respectively. Heart tissue oxidative stress biomarkers (Glutathione "GSH" and Mucosal Lipid Peroxidation "MDA") were estimated spectrophotometrically. Cardiac dysfunction biomarkers as protein kinase C (PKC) and troponin-I (Tn-I) were estimated using ELISA. Immunohistochemically, tissues caspase-3 and tumor necrosis factor- alpha (TNF-α) were estimated. Histopathology: Samples from heart were fixed in Formalin 10% for 48 h. Routine histological procedures followed by routine hematoxylin and eosin stain and Masson’s trichrome stains for collagen identification. Results: In control negative and vitamin C group, a normal histological structure of cardiac muscles could be found. Vitamin C administration corrected the disturbances caused by MSG. Conclusions: MSG may produce oxidative stress via the generation of highly toxic oxygen and nitrogen free-radicals and the inhibition of several antioxidant enzymes, altering several physiological and functional changes in the heart; this resembles human myocardial infarction, which causes sudden death in humans. Vitamin C administration showed their cardiac protective effects.

Many xenobiotics in the environment affect the human body in various ways. Among those xenobiotics, lithium chloride (Li, LiCl) and monosodium glutamate (L-glutamic acid monosodium salt, MSG) compounds affect the crucial processes of stem cell differentiation, cell proliferation, developmental gene expression, and overall development in animals. In this study, we aimed to examine the developmental effects of exposure to flavor enhancer MSG and LiCI medicament on Xenopus embryos using the frog embryo teratogenesis assay of Xenopus test. To this purpose, Xenopus laevis embryos were exposed to four different concentrations of MSG (120, 500, 750, 1000 mg/dL) and Li (0.02 g/L) alone and in combinations for a period of 96 h, and then normal, abnormal, and death ratios were determined in all exposure groups. Besides, length values of all groups and membrane potentials of fertilized and non-fertilized oocyte groups treated with 120- and 500-mg/dL MSG doses and 0.02-g/L LiCI dose were measured. Treatment with ADI (acceptable daily intake) dose of MSG alone did not lead to a substantial effect on the development of Xenopus laevis embryos. But, exposure to daily doses exceeding the ADI level (500, 750, 1000 mg/dL) caused significant harmful effects. Besides, Li-involving treatments caused dramatic deleterious effects on embryo development. MSG attenuated harmful effects of Li in MSG+Li combined treatments. Membrane potentials of non-fertilized oocytes and fertilized eggs were significantly changed in all groups that their membrane potentials were measured. Extrapolating these results into humans require similarly designed studies conducted on human embryos.

Background Uterine leiomyomas (fibroids), a menace of the reproductive age, is characterized by proliferation of smooth muscle cells (hyperplasia) of the uterus. Alpha Stone Decoction is a poly-herbal formulation that is used for the shrinkage and prevention of uterine fibroids in folkore medicine. Objective We investigated the efficacy and safety of Alpha Stone Decoction (ASD), on monosodium glutamate (MSG) induced uterine hyperplasia. Materials and methods Twenty-eight mature virgin female rats were randomly divided into four study groups: A-control B- MSG (200 mg/kgbw), C- MSG + ASD (100 mg/kgbw) and D- ASD 100 mg/kgbw alone. The administration was carried out by as a single daily dose via intraperitoneal route for 14 days. Total protein, triglycerides, estradiol (estrogen), progesterone, and total cholesterol levels in sera were determined using appropriate kits. Uterine hyperplasia was assessed via histomorphometric method using the mitotic image plus software to compute the fibroblast cell count density while the uteri and ovaries of animals were stained with mason-tricon stain for histological examination. Background It is known that cholecystokinin (CCK) plays an essential role in reducing food intake and driving weight loss. Previous studies demonstrated that amino acids were capable of triggering CCK release through G protein-coupled receptors, but the sensing mechanism remains obscure, especially the intracellular signaling pathway. Results l-Glu, rather than its d-isomer, robustly stimulated CCK secretion in a porcine duodenal model, and the secretory response was augmented by incubation with the allosteric ligand of T1R1, while T1R3 antagonist attenuated it. Upon inhibiting phospholipase C (PLC) or transient receptor potential M5 (TRPM5) activity, l-Glu failed to increase CCK release. Oral administration of monosodium glutamate in rats also suppressed food intake and increased plasma CCK levels, accompanied by elevated expression of T1R1, PLCβ2 and TRPM5 in the duodenum. Conclusion These data demonstrated that l-Glu stimulated CCK secretion through the activation of T1R1/T1R3 in a PLC/TRPM5-dependent manner. © 2020 Society of Chemical Industry.

The results of an experimental study conducted on white male rats and females of reproductive age were analyzed in order to detect submicroscopic changes in the structural components of the spleen under the action of monosodium glutamate in the dynamics. For two, four, six and eight weeks, the animals received monosodium glutamate at a dose of 0.07 g / kg body weight daily with food. Sections of the spleen were made on a UMTP-6M ultramicrotome with a diamond knife (DIATOM) and double contrast was performed according to Reynolds and uranyl acetate. Submicroscopic examinations of the organ were performed using an electron transmission microscope TEM-100. The investigated material was photodocuted using a SONY – H9 digital camera. The first violations of the structural components of the spleen are observed after two weeks, namely the expansion of intercellular spaces in both white and red pulp of the spleen, which contain vacuole-like structures, an increase in plasma cells, the cytoplasm of which is filled with dilated tubules. In dynamics with increase in duration of reception changes deepen, reaching a maximum in 8 weeks of experiment. Electron microscopy revealed that the signs of adaptive-compensatory processes by the end of the experiment lead to a loss of regenerative function.

The World Health Organization considers tobacco smoke as a global health and social problem. The active use of food supplements is ambiguous as regards human health and safety. So, the study of molecular mechanisms of toxicity of isolated and combined action of tobacco smoking and monosodium glutamate is one of the topical issues of modern science. The study aimed to determine changes in the C-reactive protein and tumor necrosis factor-α of blood serum in rats under secondhand tobacco smoking combined with a long-term monosodium glutamate injection in the age aspect. Experiments were carried out on 64 outbred white male rats divided into the following groups: I – control group; II – rats with modeled secondhand tobacco smoking; III - rats, which were injected with monosodium glutamate; IV - rats with modeled secondhand tobacco smoking combined with the monosodium glutamate injection. Background: Monosodium glutamate (MSG), an extensively used food additive, is claimed to cause many health problems, its use is still under debate. Aim: to explore the metabolic and hypertensive effect of MSG dietary administration in pregnant and lactating female rats, and its possible underlying mechanism. Methods: 16 adult female albino Wister rats were divided into 2 groups, control group; were fed standard rat chow, and MSG group; were fed 2% MSG supplemented rat chow throughout gestation and lactation period. Oral glucose tolerance test (OGTT) and arterial blood pressure (ABP) were measured at mid and late gestation, mid and late lactation periods. At mid lactation period, a retroorbital samples were analyzed for fasting glucose, lipid profile; triglycerides (TGs), total cholesterol (TC) and low density lipoprotein-cholesterol (LDL-C), hormonal assays; insulin, glucagon, prolactin, corticosterone and ACTH, as well as pancreatic lipase, amylase and malondialdehyde (MDA). By the end of lactation, adipose tissue was assessed for glucose transporter-4 (GLUT-4) and hormone sensitive lipase (HSL) relative expression. Results: MSG rats showed elevated ABP during pregnancy and lactation, with impaired OGTT at mid and late lactation period. Fasting serum glucose, TGs, TC, LDL-C, glucagon, corticosterone, pancreatic lipase and amylase, MDA as well as adipose tissue HSL expression were significantly elevated, while, fasting serum insulin, ACTH and adipose tissue GLUT-4 expression were significantly declined in MSG fed rats as compared to control group. Conclusion: MSG feeding during pregnancy and lactation induced hypertension and metabolic alterations that could be due to pancreatic and adrenal affection secondary oxidative stress.

The aim of this study was to find the effect of long-term food additive monosodium glutamate (MSG) on the tissue of pancreas. For this purpose, two treated groups of adult wistar rats were administered 15 mg/kg body weight for 30 and 75 days respectively. Immediately after sacrificing, specimens of pancreas were collected and fixed. Routine histological processes were carried out to prepare the specimens for light microscopy. Hematoxylin and Eosin, and Gomori aldehyde fuchsin stains were used. The study confirmed that the pancreas had a histological compensatory defense mechanism. In terms of exocrine units, the size of islets of Langerhans were increased due to increase the size of alpha and beta cells and the engorgement of the surrounding blood vessels. The exocrine acini neighboring to islets of Langerhans were engorged with secretion. After 75 days, fusion of some preexisting islets among the exocrine units were recorded. The study firstly focused the light on the pancreatic adipose tissue differentiation to give rise a newly–formed islets of langerhans. The alpha and beta cells of the islets had a well cytological architecture with apparent network of functional blood vessels. Creation of a new generation of beta cells induced by appetizer, may regarded as an important approach in the treatment of type 1 diabetes mellitus. The study concluded that, in small animals, the endocrine mass of pancreatic islets proportionate with the need of insulin. Moreover, adipose tissue act as endocrine tissue and may secreting the hormone insulin. Aim of the study: The goal of the current study was to evaluate the protective impact of chitosan nanoparticles (COS-NPs) against monosodium glutamate (MSG)-induced oxidative stress in submandibular salivary glands and genotoxicity of bone marrow chromosomes. Material and methods: 30 adult male albino rats were randomly divided into three equal groups and treated orally for 24 days as follow: Group I the (control) group, group II (MSG-treated group): each rat received a daily MSG at a dose of (400mg/Kg. b. w.) and group III: (MSG + COS-NPs treated group): rats treated orally with monosodium glutamate (400 mg/kg. b. w.) then the chitosan nanoparticles (280 mg/kg b. w.) respectively was administered to rats daily by oral gavage. After 24 days, both sides of the submandibular salivary glands were excised, processed and finally inspected histologically and immunohistochemically. Bone marrow was obtained from both femora of rats, 500 well spread metaphases per each animal were checked microscopically for both structural and numerical chromosomal aberrations. Results: The submandibular salivary glands (SMSG) of MSG treated rats revealed severe histopathological changes, moderate to strong granular cytoplasmic reaction of caspase-3 immunoreactivity in the acinar and ductal cells. And increase in the frequency of structural and numerical chromosomal aberrations of bone marrow cells. Chitosan nanoparticles (COSNPs) exhibited significant amelioration in histological and immunohistochemical picture in the submandibular salivary glands of MSG treated rats. Also chitosan nanoparticles could significantly inhibit the chromosomal aberrations and succeeded to neutralize the MSG genotoxic effects in bone marrow cells of rats. Conclusion: It could be concluded that COS-NPs have potential antioxidant effect that protect against cytotoxicity and genotoxicity induced by MSG in rats.

Objective Monosodium glutamate (MSG) is a food additive that has been shown to be toxic to rodents at high concentrations. The available studies in Drosophila melanogaster suggest that MSG toxicity depends on concentration and gender, thus the safety of MSG as a food enhancer still requires further investigation. We have documented impaired locomotor activity and altered oxidative stress markers in cockroaches co-exposed to methylmercury and monosodium glutamate (MSG). We herein examined the susceptibility of Nauphoeta cinerea to high and low concentrations (4% and 1%) of MSG, while monitoring the activities of acetylcholinesterase (AChE), as well as markers of oxidative stress and antioxidant activity over 30 days. Results There was no significant alteration in the parameters assessed at 1% MSG while 4% MSG caused an increase in the activity of reactive oxygen and nitrogen species, with a corresponding reduction in the activities of acetylcholinesterase, glutathione-S-transferase and catalase, suggesting the capacity of MSG to alter redox homeostasis in Nauphoeta cinerea.

The increasing of obesity is one of the main challenges of public health, with an incentive to the development of healthy life habits and practise of physical exercise. Among the physical exercises, the use of whole body vibration is highlighted, which can promote gain of muscle strength. This study aimed atevaluating the effects that vibratory platform causes on the histomophometry and fiber types of the soleus muscle of Wistar rats with induced obesity by monosodium glutamate (MSG). Thirty-two male Wistar rats were used, 16 of which were induced by obesity with intradermal injections of MSG, equally randomized into four groups: control (GC), control with intervention (GCP), obese (GO), and obese with intervention (GOP). Diabetes is one of the most common metabolic diseases. Aside from the genetic factor, previous studies stated that other factors such as environment, lifestyle, and paternal-maternal condition play critical roles in diabetes through DNA methylation in specific areas of the genome. One of diabetic cases is caused by insulin resistance and changing the homeostasis of blood glucose control so glucose concentration stood beyond normal rate (hyperglycemia). High fat diet has been frequently studied and linked to triggering diabetes. However, most Asians consume rice (or food with high carbohydrate) and food with monosodium glutamate (MSG). This habit could lead to pathophysiology of type 2 diabetes mellitus (T2D). Previous studies showed that high-carbohydrate or high-MSG diet could change gene expression or modify protein activity in body metabolism. This imbalanced metabolism can lead to pleiotropic effects of diabetes mellitus. In this study, the authors have attempted to relate various changes in genes expression or protein activity to the high-carbohydrate and high-MSG-induced diabetes. The authors have also tried to relate several genes that contribute to pathophysiology of T2D and proposed several ideas of genes as markers and target for curing people with T2D. These are done by investigating altered activities of various genes that cause or are caused by diabetes. These genes are selected based on their roles in pathophysiology of T2D.

Introduction : Monosodium Glutamate (MSG) increase free radicals and effecting on male reproductive hormone balance. CMCE propolis extract can be used as supplement to protect cells from free radical. Objective : To prove the effect of CMCE propolis extract to increase FSH, LH and Testosterone levels in male Wistar rats induced by MSG. Methods : This research was an experimental with post test only control group design used 18 male Wistar rats and divided into 3 groups. Each group on Control (K), P1 and P2 were induced by MSG of 140mg, then on P1 added 8,3mg CMCE propolis extract and P2 10,8mg for 21 days. Then on 22 th day of blood was taken through the orbitalis sinus and serum was used for the analysis FSH, LH and Testosteron levels with ELISA method. Results : ANOVA analysis test showed significant differences each group p=0,000(p˂0,05). Post hoc LSD test showed FSH, LH and Testosterone levels in the 8,3mg dose CMCE propolis extract and 10,8mg were significant different compared with control group p=0,000(p˂0,05). CMCE propolis extract 10,8mg dose had the significantly highest effect of FSH, LH and Testosterone levels than CMCE propolis extract 8,3 mg. Conclusion : CMCE propolis extract 10,8mg/day for 21 days is proven to increase FSH, LH and testosterone levels in male Wistar rats induced by MSG.

BACKGROUND AND AIMS: сomparative animal study of effectiveness of intermittent administration of lyophilized single-, three- and alive multistrain probiotic in short courses on insulin resistance (IR) in rats with experimental obesity. METHODS: 70 rats were divided into 7 groups (n = 10 in each). Rats of group I were left intact. Newborn rats in groups II-VII were administered monosodium glutamate (MSG) (4 mg/g) by injection. Rats in group II (MSG-obesity group) were left untreated. The rats in groups III-V received lyophilized mono-probiotics B.animalis VKL, B.animalis VKB, L.casei IMVB-7280 respectively. The rats in group VI received all three of these probiotic strains mixed together. Group VII was treated with multi-probiotic “Symbiter”, containing 14 different live probiotic strains (Lactobacillus, Bifidobacterium, Propionibacterium, Acetobacter genera). RESULTS: Treatment of newborn rats with MSG lead to the development of obesity in all MSG-obesity rats and up to 20–70% after probiotic administration. Summary: Leucine (Leu) is an essential branched amino acid that regulates several aspects of metabolism and may have the ability to mitigate obesity and type 2 diabetes. In the present study, we evaluated the effect of chronic Leu supplementation on the obesity and metabolic abnormalities induced by neonatal monosodium l-glutamate (MSG) treatment. From the 2nd to 6th day after birth, male Wistar rats received MSG subcutaneously (4 g/kg/body weight) and Controls received saline. After weaning, rats were subdivided into 4 groups (n = 15/rats by group): Control-NS: non-supplemented lean rats; Control-Leu: lean rats that received Leu supplementation; MSG-NS: non-supplemented obese rats and MSG-Leu: obese rats that received Leu supplementation. Leucine (1.5%) was administered via drinking water for four months. Body weight, food and liquid intake were recorded during the duration of treatment and animals were euthanized at 120 days of life. Fat content, glucose homeostasis and glucose-induced insulin secretion (GIIS) were also evaluated. MSG-treated rats developed obesity, glucose intolerance, insulin resistance (IR), impaired GIIS and presented altered cholinergic responses. Despite promoting changes in the cholinergic response of isolated pancreatic islets, chronic Leu supplementation did not abrogate obesity or metabolic abnormalities, including IR and GIIS, in MSG-treated rats. Background: Monosodium glutamate (MSG) has been recognized as flavor enhancer that adversely affects male reproductive systems. Objective: The study was conducted to explore the conceivable protective effects of vitamin C and/or vitamin E on testicular toxicity induced by MSG in rats. Materials and Methods: Thirty male Wistar albino rats were divided (six per group) into: control, MSG, MSG + Vitamin C, MSG + Vitamin E and MSG + Vitamin C + Vitamin E groups. The duration of the study was three weeks. Assessment of serum testosterone, leuteinizing hormone (LH), malondialdehyde (MDA), glutathione peroxidase (GPX), interleukin-10 (IL-10) ,and tumor necrosis factor (TNF-α) were done. Histopathological examination of the testes of the rats was performed using histological, histochemical (Periodic Acid Schiff reaction (PAS)), and immunohistochemical (Proliferating cell nuclear antigen (PCNA), androgen receptors (ARs), Caspase-3) techniques. Results: MSG-group was associated with significant decrease in serum testosterone, LH, GPX, and IL-10 (P < 0.05) and significant increase in serum MDA and TNF-α (P < 0.05) when compared with control group. MSG-group revealed many histopathological changes in the testis including degeneration of the germinal epithelium, absence of sperms in the lumina of tubules, widened vacuolated interstitium, marked deposition of the collagen fibers, very strong PAS reaction and marked immunohistochemical changes. Administration of vitamin C or vitamin E significantly reduced these changes; however, the combination of vitamin C and vitamin E provided more potent protection against the toxic effect of MSG than using each vitamin alone. Also, there was insignificant difference (P > 0.05) between MSG +Vitamin C and MSG +Vitamin E groups. Conclusion: Vitamin C and Vitamin E act synergistically in reducing MSG-induced testicular toxicity via antioxidant, anti-inflammatory and antiapoptotic effects of both vitamins.

Obesity is a worldwide public health issue, however, efficient strategies to treat obesity without systemic damage is still in serious lack. Herein, we proved that a significant decrease in body weight and abdominal circumference was observed when monosodium glutamate (MSG)-induced obese mice were orally treated with Coicis Semen formula. Consistently, computed tomography and magnetic resonance imaging results showed that both whole-body fat content and subcutaneous fat thickness were lowered, which was further confirmed by hematoxylin–eosin staining. In addition, the weight ratios of adipose tissues and liver to the body weight were reduced, due to the decreased lipid accumulation. The increased triacylglycerol, total cholesterol, and inflammatory cytokines could also be downregulated after the treatment. Furthermore, the neuronal injury marker and inflammatory cytokines in the hypothalamus were obviously reduced after the treatment, revealing that the protective effect of Coicis Semen formula on MSG-induced obesity was achieved by alleviating hypothalamic injury.

Monosodium glutamate (MSG) is a flavour enhancer commonly used in processed food to increase palatability. Several studies have reported that chronic exposure of MSG causes renal fibrosis via oxidative stress mechanism. However, till date, the effects of low dose of MSG on the oxidative stress status and its histopathological observation of renal are still unclear. A total of 18 male Sprague Dawley rats (170 – 200 g) were divided randomly into three groups consisted of the control (received distilled water = 1 ml/kg), MSG60 (received 60 mg/kg MSG) and MSG 120 (received 120 mg/kg MSG) groups. All of the substances were given via force-feed oral for 28 consecutive days. At the end of the study, all rats were sacrificed and the renal were isolated for biochemical and histological evaluation. The superoxide dismutase (SOD) activity and protein carbonyl (PC) level showed significantly increased (p < 0.05) in MSG 60 and MSG 120 group compared to the control group. However, no significant difference was found in glutathione (GSH) and malondialdehyde (MDA) level in all treated groups. The histology observation showed glomerulus shrinkage in MSG60 and MSG120 groups. In conclusion, these findings confirmed low dose of MSG-induced oxidative stress and histopathological changes on the renal of male Sprague-dawley rats. Accordingly, care must be taken on the intake of in our daily basis.

Background Paclitaxel (PTX), a chemotherapeutic agent, and monosodium glutamate (MSG) have oxidative effects on testicular tissue. Objective In this study, the effects of MSG administration on the exacerbation of testicular tissue alterations related to PTX treatment were evaluated. Materials and Methods MSG (30 & 60 mg/kg i.p.) was administrated to six groups (n = 8/each) of adult mice before or after PTX treatment: control, PTX-treated, MSG30 + PTX, MSG60 + PTX, PTX + MSG30, and PTX + MSG60. Following the euthanizing, the body weight measurement, pituitary–testicular axis hormonal analysis and serum lipid peroxidation index assessment was prepared, testicular histomorphometry (tubular diameter and germinal epithelium height), immunohistochemistry of p53 was completed. Microscopic indices of spermatogenesis (tubular differentiation, spermiogenesis and repopulation indices) were studied. Results Body weight was not changed significantly. The levels of testosterone (p = 0.0001), follicle stimulating hormone (p = 0.019), and luteinizing hormone (p = 0.08) were decreased while the level of lipid peroxidation index was increased (p = 0.208) in the treated groups. The histomorphometry indices (p < 0.0001 and p = 0.001, respectively), germ cells population (p < 0.05) and microscopic indices of spermatogenesis (p = 0.001, p = 0.005, p < 0.0001, respectively) were significantly reduced in all treated groups. The administration of MSG before PTX treatment induces more changes. The most positive reaction to p53 was observed in MSG30 or 60 + PTX groups compared to other groups. Conclusion The administration of MSG could intensify testicular tissue alterations related to PTX chemotherapy.

Here, we investigate the effects of exercise training on glucose- and cholinergic-induced insulin secretion in pancreatic islets from obese and lean rats. Male Wistar rats were treated with monosodium glutamate (MSG) for the first 5 days of life, while control (CON) rats received saline. At 21 days, the rats were divided into exercised (EXE) and sedentary (SED) groups. The EXE rats swam for 30 minutes, three times/week, for 10 weeks. After this, MSG-SED rats showed hyperglycaemia, hypertriglyceridaemia and hyperinsulinaemia. Besides, islets from MSG-SED rats exhibited increased glucose-stimulated insulin secretion (GSIS), followed by impaired glucose sensitivity, absence of glucose-amplifying pathway and weak cholinergic response. In contrast, adiposity, hyperinsulinaemia and hypertriglyceridaemia were reduced in MSG-EXE rats. Moreover, islets from MSG-EXE rats exhibited lower GSIS and improved islet glucose sensitivity, without restoration of the glucose-amplifying pathway or alteration in the weak cholinergic effect of these islets. In islets from CON-EXE rats we also observed reduced GSIS and absence of glucose-amplifying effects and an accentuated reduction in cholinergic insulinotropic responses, without effect on glucose sensitivity in pancreatic islets from this group. Neither obesity nor exercise modified Muscarinic Receptor 3 (M3R) immunocontent or its downstream pathways (PKC and PKA). Moreover, only CON-EXE showed increased GSIS in the presence of calcium blocker, Thapsigargin. In conclusion, swimming training reduces GSIS and cholinergic responsiveness in isolated pancreatic islets from lean and hypothalamic obese rats, which could be due to the inhibition of glucose-amplifying pathways.

Background:Monosodium glutamate (MSG) is a flavour enhancer which induces behavioural changes in animals. However the influence of sex on the behavioural response to MSG has not been investigated.Objective:The sex-differential effects of MSG on open-field behaviours, anxiety-related behaviour, behavioural despair, place-preference, and plasma/brain glutamate levels in adult mice were assessed.Methods:Mice were assigned to three groups (1-3), based on the models used to assess behaviours. Animals in group 1 were for the elevated-plus maze and tail-suspension paradigms, group 2 for the open-field and forced-swim paradigms, while mice in group 3 were for observation in the conditioned place preference paradigm. Mice in all groups were further assigned into five subgroups (10 males and 10 females), and administered vehicle (distilled water at 10 ml/kg) or one of four doses of MSG (20, 40, 80 and 160 mg/kg) daily for 6 weeks, following which they were exposed to the behavioural paradigms. At the end of the behavioural tests, the animals were sacrificed, and blood was taken for estimation of glutamate levels. The brains were also homogenised for estimation of glutamate levels.Results:MSG was associated with a reduction in locomotion in males and females (except at 160 mg/kg, male), an anxiolytic response in females, an anxiogenic response in males, and decreased behavioural despair in both sexes (females more responsive). Postconditioning MSG-associated place-preference was significantly higher in females. Plasma/ brain glutamate was not significantly different between sexes.Conclusion:Repeated MSG administration alters a range of behaviours in a sex-dependent manner in mice.
