The increasing application of solid phase microextraction (SPME) technology has improved the development of various fields. The advantages of this technology are that it facilitates on-site sampling and sample preparation, realizes high-throughput automatic operation, and can be combined with gas chromatography and mass spectrometry for remote monitoring. Further improvements to the speed, efficiency and specificity of this technology are, however, still required. SPME has now been applied to plant and insect volatiles, insect sex pheromones and egg volatiles. Here, we summarize the methods, disadvantages and advantages of solid-phase microextraction, and the optimal extraction materials for different insect semio-chemicals. Glutathione transferases (GSTs) are ubiquitous key enzymes with different activities as transferases or isomerases. As key detoxifying enzymes, GSTs are expressed in the chemosensory organs. They fulfill an essential protective role because the chemosensory organs are located in the main entry paths of exogenous compounds within the body. In addition to this protective function, they modulate the perception process by metabolizing exogenous molecules, including tastants and odorants. Chemosensory detection involves the interaction of chemosensory molecules with receptors. GST contributes to signal termination by metabolizing these molecules. By reducing the concentration of chemosensory molecules before receptor binding, GST modulates receptor activation and, therefore, the perception of these molecules. The balance of chemoperception by GSTs has been shown in insects as well as in mammals, although their chemosensory systems are not evolutionarily connected. This review will provide knowledge supporting the involvement of GSTs in chemoperception, describing their localization in these systems as well as their enzymatic capacity toward odorants, sapid molecules, and pheromones in insects and mammals. Their different roles in chemosensory organs will be discussed in light of the evolutionary advantage of the coupling of the detoxification system and chemosensory system through GSTs. Spodoptera exigua and S. litura are two sympatric species in China and many other countries. Both moths employ a multiple component sex pheromone blend, including a common component Z9,E12-14:OAc, and two specific components Z9-14:OH and Z11-16:OAc for S. exigua , and one specific component Z9,E11-14:OAc for S. litura. For the two species, it has been well documented that males are able to recognize and behaviorally attracted by their species-specific sex pheromone, which functions as a means of reproductive isolation, but whether males could mutually recognize pheromone components of its sympatric species is unknown. In the present study, the electroantennogram (EAG) and field evaluation were conducted to address this topic. The EAG recordings revealed that males of each species could significantly respond to specific components of its sympatric species, although the response values were lower than that to its own major component. In field tests, the specific components Z9-14:OH and Z11-16:OAc of S. exigua strongly inhibited the male catches of S. litura to its conspecific sex pheromone, while specific component Z9,E11-14:OAc of S. litura significantly reduced the male catches of S. exigua to its sex pheromone. Furthermore, the combined lure of the two species completely inhibited male catches of S. litura , and significantly decreased the male catches of S. exigua , compared to the species-specific lure alone. The results demonstrated that males of the two sibling species could perceive the specific components of its counterpart, suggesting that mutual recognition of pheromone components may function to strengthen the behavioral isolation between the two species. Our study has added new knowledge to the reproductive isolation via sex pheromone communication system in sympatric moth species, and provided a base for designing of mating disruption tactics targeting multispecies by using insect sex pheromones. Olfaction is an essential sensory modality for insects and their olfactory environment is mostly made up of plant-emitted volatiles. The terrestrial vegetation produces an amazing diversity of volatile compounds, which are then transported, mixed, and degraded in the atmosphere. Each insect species expresses a set of olfactory receptors that bind part of the volatile compounds present in its habitat. Insect odorscapes are thus defined as species-specific olfactory spaces, dependent on the local habitat, and dynamic in time. Manipulations of pest-insect odorscapes are a promising approach to answer the strong demand for pesticide-free plant-protection strategies. Moreover, understanding their olfactory environment becomes a major concern in the context of global change and environmental stresses to insect populations. A considerable amount of information is available on the identity of volatiles mediating biotic interactions that involve insects. However, in the large body of research devoted to understanding how insects use olfaction to locate resources, an integrative vision of the olfactory environment has rarely been reached. This article aims to better apprehend the nature of the insect odorscape and its importance to insect behavioral ecology by reviewing the literature specific to different disciplines from plant ecophysiology to insect neuroethology. First, we discuss the determinants of odorscape composition, from the production of volatiles by plants (section "Plant Metabolism and Volatile Emissions") to their filtering during detection by the olfactory system of insects (section "Insect Olfaction: How Volatile Plant Compounds Are Encoded and Integrated by the Olfactory System"). We then summarize the physical and chemical processes by which volatile chemicals distribute in space (section "Transportation of Volatile Plant Compounds and Spatial Aspects of the Odorscape") and time (section "Temporal Aspects: The Dynamics of the Odorscape") in the atmosphere. The following sections consider the ecological importance of background odors in odorscapes and how insects adapt to their olfactory environment. Habitat provides an odor background and a sensory context that modulate the responses of insects to pheromones and other olfactory signals (section "Ecological Importance of Odorscapes"). In addition, insects do not respond inflexibly to single elements in their odorscape but integrate several components of their environment (section "Plasticity and Adaptation to Complex and Variable Odorscapes"). We finally discuss existing methods of odorscape manipulation for sustainable pest insect control and potential future developments in the context of agroecology (section "Odorscapes in Plant Protection and Agroecology"). In the present study, scanning electron microscopy was used to investigate the ultrastructure of sensilla present on the antennae of male and female Zygogramma bicolorata. The antennae in this insect consist of the scape, pedicel, and nine flagellomeres. Six types of sensilla were identified: sensilla trichodea type 1 and 2 (ST1, ST2), sensilla cavitae (SCa), sensilla chaetica (SCh), and sensilla basiconica type 1 and 2 (SB1, SB2). Sensilla were characterized on the basis of their size and density to find out whether sexual dimorphism exists in this species or not. We observed that the size of the antennomeres was comparatively larger in males than females. Moreover, sensilla chaetica were found on all antennomers and have the qualification of being the longest type, suggesting that they probably have a role in mechanoreception. There was an abundance of sensilla trichodea in both males and females, indicating its function as a chemoreceptor. Feeding preference is critical for insect adaptation and survival. However, little is known regarding the determination of insect feeding preference, and the genetic basis is poorly understood. As a model lepidopteran insect with economic importance, the domesticated silkworm, Bombyx mori, is a well-known monophagous insect that predominantly feeds on fresh mulberry leaves. This species-specific feeding preference provides an excellent model for investigation of host-plant selection of insects, although the molecular mechanism underlying this phenomenon remains unknown. Here, we describe the gene GR66, which encodes a putative bitter gustatory receptor (GR) that is responsible for the mulberry-specific feeding preference of B. mori. With the aid of a transposon-based, clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein-9 nuclease (Cas9) system, the GR66 locus was genetically mutated, and homozygous mutant silkworm strains with truncated gustatory receptor 66 (GR66) proteins were established. GR66 mutant larvae acquired new feeding activity, exhibiting the ability to feed on a number of plant species in addition to mulberry leaves, including fresh fruits and grain seeds that are not normally consumed by wild-type (WT) silkworms. Furthermore, a feeding choice assay revealed that the mutant larvae lost their specificity for mulberry. Rice is a staple food in Asia and it contributes significantly to the Gross Domestic Product (GDP) of Malaysia and other developing countries. Brown Planthopper (BPH) causes high levels of economic loss in Malaysia. Identification of BPH presence and monitoring of its abundance has been conducted manually by experts and is time-consuming, fatiguing and tedious. Automated detection of BPH has been proposed by many studies to overcome human fallibility. However, all studies regarding automated recognition of BPH are investigated based on intact specimen although most of the specimens are imperfect, with missing parts have distorted shapes. The automated recognition of an imperfect insect image is more difficult than recognition of the intact specimen. This study proposes an automated, deep-learning-based detection pipeline, PENYEK, to identify BPH pest in images taken from a readily available sticky pad, constructed by clipping plastic sheets onto steel plates and spraying with glue. This study explores the effectiveness of a convolutional neural network (CNN) architecture, VGG16, in classifying insects as BPH or benign based on grayscale images constructed from Euclidean Distance Maps (EDM). The pipeline identified imperfect images of BPH with an accuracy of 95% using deep-learning’s hyperparameters: softmax, a mini-batch of 30 and an initial learning rate of 0.0001. In the Central American region, the aggressive, sibling bark beetles Dendroctonus frontalis Zimmermann (Coleoptera: Curculionidae: Scolytinae) and Dendroctonus mesoamericanus Armendáriz-Toledano & Sullivan (Coleoptera: Curculionidae: Scolytinae) commonly colonize pines concurrently, and in nature they avoid heterospecific pairing, although it can be produced in the lab. We performed walking arrestment bioassays in the lab to examine the capacity of both sexes of both species to discriminate odors from frass expelled from gallery entrances of either solitary females or conspecific pairs of either species. Males of both species strongly preferred odors of frass from solitary, conspecific females over those of heterospecific females or pairs of either species. Female D. frontalis did not discriminate among these frass categories, whereas female D. mesoamericanus preferred frass of conspecific females. In gas chromatography–electroantennographic detection and gas chromatography–mass spectrometry analyses, we determined that males of both species could sense a nearly identical spectrum of approximately 16 host- and beetle-produced compounds present in frass of females of one or both species. Only two of these compounds, endo -brevicomin and ipsdienol, which were present in frass of female D. mesoamericanus and pairs of either species but absent in frass of solitary D. frontalis females, qualitatively distinguished these categories. Several known attractants and synergists for either species declined in concentration postpairing. Our results complement earlier research and indicate how semiochemical composition and concentration in frass might mediate male discrimination of attack sites of conspecific, unpaired females. Cyrtorhinus lividipennis Reuter (Hemiptera: Miridae: Orthotylinae), including nymphs and adults, are one of the dominant predators and have a significant role in the biological control of leafhoppers and planthoppers in irrigated rice. In this study, we investigated the antennal morphology, structure and sensilla distribution of C. lividipennis in different instars using scanning electron microscopy. The antennae of both five different nymphal stages and adults were filiform in shape, which consisted of the scape, pedicel and flagellum with two flagellomeres. There were significant differences found in the types of antennal sensilla between nymphs and adults. The multiporous placodea sensilla (MPLA), basiconica sensilla II (BAS II), and sensory pits (SP) only occurred on the antennae of adult C. lividipennis of both sexes. Moreover, there was chaetica sensilla III (CHA III) only observed in males. Sixteen types of antennal sensilla were recorded altogether. They were microtrichia sensilla (MIC), three types of trichoidea sensilla (TRI I-III), three types of chaetica sensilla (CHA I-III), three types of basiconica sensilla (BAS I-III), two types of coeloconica sensilla (COE I and COE II), placodea sensilla (PLA), campaniform sensilla (CAM), MPLA, and SP. In the five different nymphal stages of C. lividipennis, the length of their antennae was significantly increased with the increase of the instar, as well as the number of the TRI II and TRI III. Moreover, sexual dimorphism usually occurred not only in the distribution (CHA III and SP) and the number of antennal sensilla (MIC, BAS II, TRI II, TRI III and MPLA), but also in the length of flagellum (F1 and F2). The possible functions of antennal sensilla are discussed. Those observations could contribute to a better understanding of the development of the olfactory system, and facilitate future studies on the antennal functions in C. lividipennis. The mechanisms of host shift in phytophagous insects are poorly understood. Among the many proposed processes involved, sexual selection via semiochemicals has recently been suggested. This hypothesizes that sexual communication using pheromones is modified as a result of development on a new host, and such plant-induced phenotypic divergence in mate recognition cues can lead to reproductive isolation between host lines. We tested this hypothesis on Acanthoscelides obtectus, an oligophagous bruchid of Phaseolus vulgaris beans worldwide, which also develops in acceptable non-hosts, such as chickpea (Cicer arietinum L.). Male sex pheromone blends of the bean, chickpea and chickpea/bean host lines during artificially induced host shifts showed different composition. Bean-reared females did not distinguish between blends, whereas chickpea and chickpea/bean females preferred the chickpea male pheromone. However, electrophysiological (EAG) responses to male odour of antennae of the three female host lines were similar, all preferring bean-reared males. Egg-laying choice tests revealed a uniform preference for bean seeds across female host lines, even after multiple generations, whereas larvae did not distinguish between bean and chickpea seeds. We conclude that the development of divergent chemical signalling systems during host shifts does not facilitate the evolution of host races in A. obtectus, because oviposition preferences remain unaffected. The efficient coding hypothesis predicts that sensory neurons adjust their coding resources to optimally represent the stimulus statistics of their environment. To test this prediction in the moth olfactory system, we have developed a stimulation protocol that mimics the natural temporal structure within a turbulent pheromone plume. We report that responses of antennal olfactory receptor neurons to pheromone encounters follow the temporal fluctuations in such a way that the most frequent stimulus timescales are encoded with maximum accuracy. We also observe that the average coding precision of the neurons adjusted to the stimulus-timescale statistics at a given distance from the pheromone source is higher than if the same encoding model is applied at a shorter, non-matching, distance. Finally, the coding accuracy profile and the stimulus-timescale distribution are related in the manner predicted by the information theory for the many-to-one convergence scenario of the moth peripheral sensory system. Conserving populations of environmentally vulnerable insect species requires a greater understanding of the factors that determine their abundance and distribution, which requires detailed knowledge of their population and community ecology. Chemical ecological tools such as pheromones can be used for non-destructive monitoring of scarab beetle populations, enabling European countries to detect and, in some cases, map the range of some of these species, proving a valuable technique for monitoring elusive saproxylic beetles. In this paper, we investigated the behavioural and chemical ecology of the noble chafer, Gnorimus nobilis L., a model insect species of conservation concern across a Europe-wide distribution, and a red-listed UK Biodiversity Action Plan priority species. We identified a potential pheromone of adult beetles using electrophysiological recordings, behavioural measurements and field trials in the UK. Gnorimus nobilis is highly unusual in that although both sexes produce, at high metabolic cost, the natural product 2-propyl (E)-3-hexenoate, it only attracts males. This pattern of chemical signalling makes the classification of the compound, based on current semiochemical terminology, somewhat problematic, but in our view, it should be termed an aggregation pheromone as a consequence of the production pattern. Since both sexes emit it, but apparently only males respond positively to it, 2-propyl (E)-3-hexenoate may reflect an intermediate evolutionary stage towards developing into a sex-specific signal. From an applied perspective, our study provides a model for the non-invasive surveillance of cryptic vulnerable insect species, without the need for habitat searching or disturbance, and continuous human monitoring. Three fruit borers Conogethes punctiferalis (Guenée) (Crambidae), Grapholita molesta Busck (Tortricidae), and Spilonota albicana Motschulsky (Tortricidae) are serious pests of fruit trees. In this study, their antennal morphology, types of sensilla, and distributions were observed by using SEM (Scanning Electron Microscope). Nine types of sensilla were found on the antennae of C. punctiferalis, while eight types of sensilla were presented on each of G. molesta and S. albicana. The sensilla trichodea with two subtypes were the most abundant sensilla among three fruit borers. Two subtypes of sensillum coeloconica (type I with spines and type II without spines) were observed on the antennae of C. punctiferalis and G. molesta. However, sensilla coeloconica (type I) were only found in S. albicana. Although the sensilla campaniformia were only found on the antennae of C. punctiferalis, our observations confirm sensilla campaniformia presence in the moths. In addition, the functions of these sensilla were discussed based on previously reported lepidopteran insects. As a result, our study may provide useful information for advanced electrophysiological and behavioral experiments to better understand the mechanisms related to pests control. Abstract: Division of labour in social insect colonies is facilitated in two ways: through temporal sharing of tasks or by morphologically specialised castes. In casteless species, colony defence is maintained by morphologically indistinct workers, who lack the obvious defensive specialisation of polymorphic species. Discrimination of intruders is carried out via antenna, which also detects defensive social cues such as alarm pheromones. Despite their functional importance however, antennal morphology is rarely considered in studies of nestmate recognition. We investigated antennal morphology and the necessity of social cues in mediating defensive behaviour across differentially tasked workers of a casteless social bee, Tetragonula carbonaria. Our results suggest that the current understanding of division of labour in casteless worker species remains poorly understood, with differences in antennal morphology and aggression creating morphologically and behaviourally distinct ‘cryptic castes’. Further, we found that defensive behaviour was only elicited near nest odours, highlighting the importance of mediating aggression among workers. [ABSTRACT FROM AUTHOR]

Abstract: An improved understanding of the biology of the invasive pest, Drosophila suzukii (Matsumura) (Diptera: Drosophilidae), is critical for the development of effective management strategies. Trapping is one technique used for both detection and control; however, the efficacy of trapping can vary depending on the target insect's physiological state, its behavioural priorities and the type of attractant used in the trap. We conducted a series of caged trapping experiments and a greenhouse trapping experiment to investigate the effects of D. suzukii feeding status, age, mating status, ovipositional status and seasonal morph type on the capture rate of traps baited with fermentation odours. Starved flies were trapped at greater rates compared to fed flies; more virgin flies were trapped than mated flies; flies deprived of an oviposition substrate were trapped more frequently than flies given an oviposition substrate. It is still unclear whether age or seasonal morphology affect bait response. Lastly, a caged choice experiment investigated the relationship between female reproductive status and attraction to fermentation or fruit odours. Fermentation‐based traps captured female flies regardless of their reproductive status but, ripe fruit‐based traps were more attractive to flies with more than seven eggs. In summary, studies that use fermentation‐based traps should recognize that capture rates of D. suzukii will depend on the feeding, mating and oviposition experiences of the population; also, fruit‐based traps may better target gravid females. Covering: up to 2018 Pheromones serve as chemical signals between individuals of the same species and play important roles for mate localization and mate choice as well as other social interactions in insects. A growing body of literature indicates that microbial symbionts can modulate their hosts' chemical profiles, mate choice decisions and social behavior. This modulation can occur by the direct biosynthesis of pheromone components or the provisioning of precursors, or through general changes in the metabolite pool of the host and its resource allocation into pheromone production. Here we review and discuss the contexts in which microbial modulation of intraspecific communication in insects occurs and emphasize cases in which microbes are known to affect the involved chemistry. The described examples for a symbiotic influence on mate attraction and mate choice, aggregation, nestmate and kin recognition highlight the context-dependent costs and benefits of these symbiotic interactions and the potential for conflict and manipulation among the interacting partners. However, despite the increasing number of studies reporting on symbiont-mediated effects on insect chemical communication, experimentally validated connections between the presence of specific symbionts, changes in the host's chemistry, and behavioral effects thereof, remain limited to very few systems, highlighting the need for increased collaborative efforts between symbiosis researchers and chemical ecologists to gain more comprehensive insights into the influence of microbial symbionts on insect pheromones. We investigated the morphology and distribution of antennal sensilla in males and females of two tortricid moths, Epiphyas postvittana and Planotortrix octo , by scanning electron microscopy. The number and overall length of flagellomeres were significantly greater in females than in males in both species. The antennae of each species bearing six morphological types of sensilla (trichodea, basiconica, coeloconica, auricillica, chaetica, and styloconica), with different numbers and distributions along the antennae. Among these sensilla, four types (trichodea, basiconica, coeloconica, and auricillica) displayed multi-porous cuticular surfaces, indicating that their primary sensory function is olfactory. Each of these four types of sensilla could be further classified into subtypes according to their size, shape, and surface structure. Both E. postvittana and P. octo exhibited sexual dimorphism of the profiles of antennal olfactory sensilla. Trichoid sensilla were the most abundant type in both species. Subtype I trichoid sensilla were male-specific in both species, indicating that they are responsible for the perception of conspecific female sex pheromone. By contrast, subtype II trichoid sensilla were more abundant in female antennae in both species, suggesting that some subtype II trichoid sensilla are involved in female-specific behaviors, such as oviposition. Chaetic and styloconic sensilla displayed relatively even distributions along the antennae. Our results indicate that the antennae of E. postvittana and P. octo have species-specific and sex-specific profiles of olfactory sensilla. The morphological information obtained in our study provides a basis for electrophysiological and behavioral studies of the olfactory sensory function of each morphological type of sensilla. An accurate quantitative relationship between key characteristics of an insect population, such as season-long and peak abundances, can be very useful in pest management programs. To the best of our knowledge, no such relationship has yet been established. Here we establish a predictive linear relationship between insect catch Mpw during the week of peak abundance, the length of seasonal flight period, F (number of weeks) and season-long cumulative catch (abundance) A = 0.41MpwF. The derivation of the equation is based on several general assumptions and does not involve fitting to experimental data, which implies generality of the result. A quantitative criterion for the validity of the model is presented. The equation was tested using extensive data collected on captures of male gypsy moths Lymantria dispar (L.) (Lepidoptera: Erebidae) in pheromone-baited traps during 15 years. The model was also tested using trap catch data for two species of mosquitoes, Culex pipiens (L.) (Diptera: Culicidae) and Aedes albopictus (Skuse) (Diptera: Culicidae), in Gravid and BG-sentinel mosquito traps, respectively. The simple, parameter-free equation approximates experimental data points with relative error of 13% and R2 = 0.997, across all of the species tested. For gypsy moth, we also related season-long and weekly trap catches to the daily trap catches during peak flight. We describe several usage scenarios, in which the derived relationships are employed to help link results of small-scale field studies to the operational pest management programs. Histamine is used in bronchial and dermal provocation, but it is rarely considered an environmental risk factor in allergic disease. Because bed bugs defecate large amounts of histamine as a component of their aggregation pheromone, we sought to determine if histamine accumulates in household dust in bed bug infested homes, and the effects of bed bug eradication with spatial heat on histamine levels in dust. We collected dust in homes and analyzed for histamine before, and up to three months after bed bug eradication. Histamine levels in bed bug infested homes were remarkably high (mean = 54.6±18.9 μg/100 mg of sieved household dust) and significantly higher than in control homes not infested with bed bugs (mean < 2.5±1.9 μg/100 mg of sieved household dust). Heat treatments that eradicated the bed bug infestations failed to reduce histamine levels, even three months after treatment. We report a clear association between histamine levels in household dust and bed bug infestations. The high concentrations, persistence, and proximity to humans during sleep suggest that bed bug-produced histamine may represent an emergent contaminant and pose a serious health risk in the indoor environment. ABSTRACT: Odorant‐binding proteins (OBPs) and chemosensory proteins (CSPs) are regarded as carriers of pheromones and odorants in insect chemoreception. These proteins are typically located in antennae, mouth organs and other chemosensory structures; however, members of both classes of proteins have been detected recently in other parts of the body and various functions have been proposed. The best studied of these non‐sensory tasks is performed in pheromone glands, where OBPs and CSPs solubilise hydrophobic semiochemicals and assist their controlled release into the environment. In some cases the same proteins are expressed in antennae and pheromone glands, thus performing a dual role in receiving and broadcasting the same chemical message. Several reports have described OBPs and CSPs in reproductive organs. Some of these proteins are male specific and are transferred to females during mating. They likely carry semiochemicals with different proposed roles, from inhibiting other males from approaching mated females, to marking fertilized eggs, but further experimental evidence is still needed. Before being discovered in insects, the presence of binding proteins in pheromone glands and reproductive organs was widely reported in mammals, where vertebrate OBPs, structurally different from OBPs of insects and belonging to the lipocalin superfamily, are abundant in rodent urine, pig saliva and vaginal discharge of the hamster, as well as in the seminal fluid of rabbits. In at least four cases CSPs have been reported to promote development and regeneration: in embryo maturation in the honeybee, limb regeneration in the cockroach, ecdysis in larvae of fire ants and in promoting phase shift in locusts. Both OBPs and CSPs are also important in nutrition as solubilisers of lipids and other essential components of the diet. Particularly interesting is the affinity for carotenoids of CSPs abundantly secreted in the proboscis of moths and butterflies and the occurrence of the same (or very similar CSPs) in the eyes of the same insects. A role as a carrier of visual pigments for these proteins in insects parallels that of retinol‐binding protein in vertebrates, a lipocalin structurally related to OBPs of vertebrates. Other functions of OBPs and CSPs include anti‐inflammatory action in haematophagous insects, resistance to insecticides and eggshell formation. Such multiplicity of roles and the high success of both classes of proteins in being adapted to different situations is likely related to their stable scaffolding determining excellent stability to temperature, proteolysis and denaturing agents. The wide versatility of both OBPs and CSPs in nature has suggested several different uses for these proteins in biotechnological applications, from biosensors for odours to scavengers for pollutants and controlled releasers of chemicals in the environment. Long-distance olfactory search behaviors depend on odor detection dynamics. Due to turbulence, olfactory signals travel as bursts of variable concentration and spacing and are characterized by long-tail distributions of odor/no-odor events, challenging the computing capacities of olfactory systems. How animals encode complex olfactory scenes to track the plume far from the source remains unclear. Here we focus on the coding of the plume temporal dynamics in moths. We compare responses of olfactory receptor neurons (ORNs) and antennal lobe projection neurons (PNs) to sequences of pheromone stimuli either with white-noise patterns or with realistic turbulent temporal structures simulating a large range of distances (8 to 64 m) from the odor source. For the first time, we analyze what information is extracted by the olfactory system at large distances from the source. Neuronal responses are analyzed using linear–nonlinear models fitted with white-noise stimuli and used for predicting responses to turbulent stimuli. We found that neuronal firing rate is less correlated with the dynamic odor time course when distance to the source increases because of improper coding during long odor and no-odor events that characterize large distances. Rapid adaptation during long puffs does not preclude however the detection of puff transitions in PNs. Individual PNs but not individual ORNs encode the onset and offset of odor puffs for any temporal structure of stimuli. A higher spontaneous firing rate coupled to an inhibition phase at the end of PN responses contributes to this coding property. Social insects vigorously defend their nests against con- and heterospecific competitors. Collective defense is also seen at highly profitable food sources. Aggressive responses are elicited or promoted by several means of communication, e.g. alarm pheromones and other chemical markings. In this study, we demonstrate that the social environment and interactions among colony members (nestmates) modulates the propensity to engage in aggressive behavior and therefore plays an important role in allocating workers to a defense task. We kept Formica rufa workers in groups or isolated for different time spans and then tested their aggressiveness in one-on-one encounters with other ants. In groups of more than 20 workers that are freely interacting, individuals are aggressive in one-on-one encounters with non-nestmates, whereas aggressiveness of isolated workers decreases with increasing isolation time. We conclude that ants foraging collectively and interacting frequently, e.g. along foraging trails and at profitable food sources, remain in a social context and thereby maintain high aggressiveness against potential competitors. Pheromones are chemical compounds used to transmit information between individuals of the same species. Pheromone composition is influenced by both genetic and environmental factors. Numerous studies, predominately of insects, have demonstrated a role for diet in pheromone expression. The chemical composition of spider web-silk varies with diet and in many species these chemicals are crucial to mate choice processes. Here, we investigated individual variation in the chemical compounds found on the surface of web-silk of female Argiope keyserlingi , and further explored the degree to which they are influenced by diet, investment in egg sac production and site of collection. We observed variation in the web-based chemical cues both between and within individuals. Additionally, we found that some of this variation could be explained by diet and gravid status but not by collection site. Male moths possess a highly specialized olfactory system comprised of two segregated sub-arrangements dedicated to processing information about plant odors and pheromones, respectively. Communication between these two sub-systems has been described at the peripheral level, but relatively little is known about putative interactions at subsequent synaptic relays. The male moth faces the challenge of seeking out the conspecific female in a highly dynamic odor world. The female-produced pheromone blend, which is a limited resource serving as guidance for the male, will reach his antennae in intermittent pockets of odor filaments mixed with volatiles from various plants. In the present study we performed calcium imaging for measuring odor-evoked responses in the uni-glomerular antennal-lobe projection neurons (analog to mitral cells in the vertebrate olfactory bulb) of Helicoverpa armigera. In order to investigate putative interactions between the two sub-systems tuned to plant volatiles and pheromones, respectively, we performed repeated stimulations with a selection of biologically relevant odors. We found that paired stimulation with a plant odor and the pheromone led to suppressed responses in both sub-systems as compared to those evoked during initial stimulation including application of each odor stimulus alone. The fact that the suppression persisted also after pairing, indicates the existence of a Hebbian-like plasticity in the primary olfactory center established by temporal pairing of the two odor stimulation categories. Background: The mountain pine beetle (MPB, Dendroctonus ponderosae Hopkins) is a highly destructive pest of pine forests in western North America. During flight to a new host tree and initiation of feeding, mountain pine beetles release aggregation pheromones. The biosynthetic pathways of these pheromones are sex-specific and localized in the midgut and fat body, but the enzymes involved have not all been identified or characterized. Results: We used a comparative RNA-Seq analysis between fed and unfed male and female MPB midguts and fat bodies to identify candidate genes involved in pheromone biosynthesis. The 13,407 potentially unique transcripts showed clear separation based on feeding state and gender. Gene co-expression network construction and examination using petal identified gene groups that were tightly connected. This, as well as other co-expression and gene ontology analyses, identified all four known pheromone biosynthetic genes, confirmed the tentative identification of four others from a previous study, and suggested nine novel candidates. One cytochrome P450 monooxygenase, CYP6DE3, identified as a possible exo-brevicomin-biosynthetic enzyme in this study, was functionally characterized and likely is involved in resin detoxification rather than pheromone biosynthesis. Conclusions: Our analysis supported previously characterized pheromone-biosynthetic genes involved in exo-brevicomin and frontalin biosynthesis and identified a number of candidate cytochrome P450 monooxygenases and a putative cyclase for further studies. Functional analyses of CYP6DE3 suggest its role in resin detoxification and underscore the limitation of using high-throughput data to tentatively identify candidate genes. Further functional analyses of candidate genes found in this study should lead to the full characterization of MPB pheromone biosynthetic pathways and the identification of molecular targets for possible pest management strategies. Insects protect themselves from microbial infections through innate immune responses, including pathogen recognition, phagocytosis, the activation of proteolytic cascades, and the synthesis of antimicrobial peptides. Termites, eusocial insects inhabiting microbe-rich wood, live in closely-related family groups that are susceptible to shared pathogen infections. To resist pathogenic infection, termite families have evolved diverse immune adaptations at both individual and societal levels, and a strategy of trade-offs between reproduction and immunity has been suggested. Although termite immune-inducible genes have been identified, few studies have investigated the differential expression of these genes between reproductive and neuter castes, and between sexes in each caste. In this study, we compared the expression levels of immune-related genes among castes, sexes, and ages in a Japanese subterranean termite, Reticulitermes speratus. Using RNA-seq, we found 197 immune-related genes, including 40 pattern recognition proteins, 97 signalling proteins, 60 effectors. Among these genes, 174 showed differential expression among castes. Comparing expression levels between males and females in each caste, we found sexually dimorphic expression of immune-related genes not only in reproductive castes, but also in neuter castes. Moreover, we identified age-related differential expression of 162 genes in male and/or female reproductives. In addition, although R. speratus is known to use the antibacterial peptide C-type lysozyme as an egg recognition pheromone, we determined that R. speratus has not only C-type, but also P-type and I-type lysozymes, as well as other termite species. Our transcriptomic analyses revealed immune response plasticity among all castes, and sex-biased expression of immune genes even in neuter castes, suggesting a sexual division of labor in the immune system of R. speratus. Long-chain cuticular hydrocarbons (CHCs) are common components of the epicuticle of terrestrial arthropods. CHC serve as a protective barrier against environmental influences but also act as semiochemicals in animal communication. Regarding the latter aspect, species- or intra-functional group specific CHCs composition and variation are relatively well studied. However, comparative knowledge about the relationship of CHC quantity and their relation to surface area—volume ratios in the context of water loss and protection is fragmentary. Hence, we aim to study the taxon-specific relationship of the CHC amount and surface-area to volume ratio related to their functional role (e.g. in water loss). We focused on flower visiting insects and analyzed the CHC amounts of three insect orders (Hymenoptera, Lepidoptera and Diptera) using gas chromatography—mass spectrometry (GC-MS). We included 113 species from two grassland plots, quantified their CHCs, and measured their body mass and surface area. We found differences in the surface area, CHCs per body mass and the CHC density (= amount of CHCs per surface area) across the three insect taxa. Especially the Hymenoptera had a higher CHC density compared to Diptera and Lepidoptera. CHC density could be explained by surface area-volume ratios in Hymenoptera but not in Diptera and Lepidoptera. Unexpectedly, CHC density decreased with increasing surface area—volume ratios. As honey bee populations worldwide are declining there is an urgent need for a deeper understanding of stress reactivity in these important insects. Our data indicate that stress responses in bees (Apis mellifera L.) may be mediated by neuropeptides identified, on the basis of sequence similarities, as allatostatins (ASTA, ASTC and ASTCC). Effects of allatostatin injection are compared with stress-related changes in learning performance induced by the honeybee alarm pheromone, isopentylacetate (IPA). We find that bees can exhibit two markedly different responses to IPA, with opposing effects on learning behaviour and memory generalisation, and that strikingly similar responses can be elicited by allatostatins, in particular ASTCC. These findings lend support to the hypothesis that allatostatins mediate stress reactivity in honey bees and suggest responses to stress in these insects are state dependent. [ABSTRACT FROM AUTHOR]

Castniid moths (Lepidoptera: Castniidae) display a butterfly-like reproductive behavior, i.e., they use visual stimuli for mate location and females have apparently lost their pheromone glands in an evolutionary context. In this paper we report for the first time the identification of three new compounds, namely n-octadecyl acetate, (Z)-9-octadecenyl acetate and (E,Z)-2,13-octadecadienyl acetate, in males of the Castniid Palm Borer, Paysandisia archon, which could be involved in its short-range courtship behavior, and also shed light on recent controversies on the sexual behavior of the species. The compounds are produced in a ring-shaped gland of the male terminalia and have occasionally been detected in very minor amounts (ng) in ovipositor extracts of females, but only while mating or just after copulation. We also report that males use the already known (E,Z)-2,13-octadecadienol to mark their territory by rubbing their midlegs against the upper side of nearby leaves, especially palm leaves. This compound, produced in large amounts, is mostly concentrated in the midleg basitarsi and its maximum production is detected on the sexually mature 1-day-old specimens. In addition, analysis of male wings extracts confirms the presence of Z,E and E,E-farnesals, which are mostly produced in the median band of hindwings of 48–53 h-old insects. The biological significance of farnesals in this species is unknown. One major advantage of sexual reproduction over asexual reproduction is its promotion of genetic variation, although it reduces the genetic contribution to offspring. Queens of social insects double their contribution to the gene pool, while overuse of asexual reproduction may reduce the ability of the colony to adapt to environmental stress because of the loss of genetic diversity. Recent studies have revealed that queens of some termite species can solve this tradeoff by using parthenogenesis to produce the next generation of queens and sexual reproduction to produce other colony members. This reproductive system, known as asexual queen succession (AQS), has been identified in the subterranean termites Reticulitermes speratus, Reticulitermes virginicus and Reticulitermes lucifugus and in the Neotropical higher termites Embiratermes neotenicus and Cavitermes tuberosus. The studies presented here have uncovered the unusual modes of reproduction in termites and have aimed to identify their underlying mechanisms. Monema flavescens Walker ( Lepidoptera: Limacodidae) is a multivoltine, generalist moth whose larvae cause serious damage to many types of trees. Pheromone lures prepared according to a study of a Japanese population were found to be ineffective at attracting M. flavescens nettle caterpillars in China, and some studies have shown intraspecific geographical differences in the composition of sex pheromones. We therefore reexamined the sex pheromone composition of M. flavescens in a Chinese population. In this study, the electroantennographically ( EAG) active compounds in an extract from Chinese virgin females of M. flavescens were identified as ( E)-8-decen-1-ol (E8-10:OH), ( Z)-7,9-decadien-1-ol (Z7,9-10:OH), ( Z)-9,11-dodecadien-1-ol (Z9,11-12:OH), and ( Z)-9,11-dodecadienal (Z9,11-12:Ald) via coupled gas chromatographic-electroantennographic detection ( GC- EAD) and coupled GC-mass spectrometry ( MS). Pheromone dimorphism might occur in this species, as this mixture of compounds in Chinese females was different from that of E8-10:OH and E7,9-10:OH extracted from Japanese females in previous research. In wind tunnel and field tests, the males were significantly attracted to a blend of the pheromone components E8-10:OH, Z7,9-10:OH, and Z9,11-12:OH in a 100:5:4 ratio. The addition of Z9,11-12:Ald did not change the male response. The optimized three-component lure blend may provide a useful tool for monitoring and controlling Chinese populations of M. flavescens. Neonicotinoid insecticides are widely used to protect plants against pest insects, and insecticide residues remaining in the environment affect both target and non-target organisms. Whereas low doses of neonicotinoids have been shown to disturb the behaviour of pollinating insects, recent studies have revealed that a low dose of the neonicotinoid clothianidin can improve behavioural and neuronal sex pheromone responses in a pest insect, the male moth Agrotis ipsilon, and thus potentially improve reproduction. As male moth behaviour depends also on its physiological state and previous experience with sensory signals, we wondered if insecticide effects would be dependent on plasticity of olfactory-guided behaviour. We investigated, using wind tunnel experiments, whether a brief pre-exposure to the sex pheromone could enhance the behavioural response to this important signal in the moth A. ipsilon at different ages (sexually immature and mature males) and after different delays (2 h and 24 h), and if the insecticide clothianidin would interfere with age effects or the potential pre-exposure-effects. Brief pre-exposure to the pheromone induced an age-independent significant increase of sex pheromone responses 24 h later, whereas sex pheromone responses did not increase significantly 2 h after exposure. However, response delays were significantly shorter compared to naïve males already two hours after exposure. Oral treatment with clothianidin increased sex pheromone responses in sexually mature males, confirming previous results, but did not influence responses in young immature males. Males treated with clothianidin after pre-exposure at day 4 responded significantly more to the sex pheromone at day 5 than males treated with clothianidin only and than males pre-exposed only, revealing an additive effect of experience and the insecticide. In highly social hymenopteran societies, males mainly serve reproductive purposes. Semiochemicals enable the different hymenopteran sexes and castes to communicate with each other and to coordinate important functions within colonies. I hereby show that sexual dimorphism in meliponine bees incorporates the alarm and defence communication system. I chemically analysed the mandibular glands of Scaptotrigona aff. depilis using GCMS methods and conducted behavioural experiments in both males and female workers using cephalic and mandibular gland extracts. In addition, behaviour studies with male cephalic extracts were also conducted in Scaptotrigona bipunctata and Partamona cupira. Males and female worker bees showed differences in the content of the mandibular glands, which are responsible for alarm communication in meliponines. Males never attacked but usually fled when confronted with the mandibular gland extract content of other conspecific males or females. Interestingly, however, meliponine males were still able to raise alarm and to induce substantial amounts of aggression at nest entrances, which is different from the much better known and studied honey bees. Gregarious larvae that use chemical communication to feed and move together are widespread among folivorous insects, although social behaviour has been studied almost exclusively in a few temperate zone genera. The Menapis (or variable) tigerwing butterfly Mechanitis menapis mantineus Hewitson (Lepidoptera, Nymphalidae, Danainae, Ithomiini) is a neotropical species whose larvae feed gregariously on Solanaceae host plants. In laboratory experiments conducted in the Ecuador cloud forest, M. menapis caterpillars are attracted to silk produced by conspecifics and show no evidence of pheromone production. Indeed, caterpillars consistently choose arenas with silk over bare arenas but do not show a preference for arenas marked with abdominal cuticular surface residues. Mechanitis menapis caterpillars on silk-coated plants are both more mobile and more cohesive than those on control plants. Nonetheless, caterpillars move independently over unmarked surfaces and groups do not make rapid collective choices between two food sources. Collective behaviour in M. menapis thus appears to be based on aggregation on collectively produced silk to facilitate feeding, as well as using this silk to maintain cohesion. Silk production is common in caterpillars, although M. menapis appears to be unique among species studied so far in using silk to maintain group cohesion. [ABSTRACT FROM AUTHOR]

In the past decades, the sex pheromone composition in female moths has been analyzed by different methods, ranging from volatile collections to gland extractions, which all have some disadvantage: volatile collections can generally only be conducted on (small) groups of females to detect the minor pheromone compounds, whereas gland extractions are destructive. Direct-contact SPME overcomes some of these disadvantages, but is expensive, the SPME fiber coating can be damaged due to repeated usage, and samples need to be analyzed relatively quickly after sampling. In this study, we assessed the suitability of cheap and disposable fused silica optical fibers coated with 100 μm polydimethylsiloxane (PDMS) by sampling the pheromone of two noctuid moths, Heliothis virescens and Heliothis subflexa. By rubbing the disposable PDMS fibers over the pheromone glands of females that had called for at least 15 minutes and subsequently extracting the PDMS fibers in hexane, we collected all known pheromone compounds, and we found a strong positive correlation for most pheromone compounds between the disposable PDMS fiber rubs and the corresponding gland extracts of the same females. When comparing this method to volatile collections and the corresponding gland extracts, we generally found comparable percentages between the three techniques, with some differences that likely stem from the chemical properties of the individual pheromone compounds. Hexane extraction of cheap, disposable, PDMS coated fused silica optical fibers allows for sampling large quantities of individual females in a short time, eliminates the need for immediate sample analysis, and enables to use the same sample for multiple chemical analyses. Pest suppression from combinations of tactics is fundamental to pest management and eradication. Interactions may occur among tactical combinations and affect suppression. The best case is synergistic, where suppression from a combination is greater than the sum of effects from single tactics (AB >> A+B). We explored how mating disruption and insecticide interacted at field scale, additively or synergistically. Use of a pheromone delivery formulation (SPLAT™) as either a mating disruption treatment (i.e. a two-component pheromone alone) or as a lure and kill treatment (i.e. the two-component pheromone plus a permethrin insecticide) was compared for efficacy against the lightbrown apple moth Epiphyas postvittana. Next, four point-source densities of the SPLAT™ formulations were compared for communication disruption. Finally, the mating disruption and lure and kill treatments were applied with a broadcast insecticide. Population assessment used virgin female traps and synthetic pheromone in replicated 9-ha vineyard plots compared with untreated controls and insecticide-treated plots, to investigate interactions. Lure and kill and mating disruption provided equivalent suppression; no additional benefit accrued from including permethrin with the pheromone suggesting lack of contact. The highest point-source density tested (625/ha) was most effective. The insect growth regulator methoxyfenoxide applied by broadcast application lowered pest prevalence by 70% for the first ten weeks compared to pre-trial. Pheromone addition suppressed the pest further by an estimated 92.5%, for overall suppression of 97.7% from the treatment combination of insecticide plus mating disruption. This was close to that expected for an additive model of interactivity between insecticide and mating disruption (AB = A+B) estimated from plots with single tactics as 98% suppression in a combination. Chemical signals are involved in the courtship behavior of many invertebrate and vertebrate species. Lady beetles are no exception to this rule; a significant number of published reports highlight the role of cuticular chemicals involved in the reproduction of lady beetles, including gender recognition. Recent data have also demonstrated the presence of a volatile female sex pheromone that facilitates male attraction in some species. Here, we present a synthetic overview of the current knowledge about the sexual behavior of lady beetles and associated chemicals for which the function and identification have been provided. Because lady beetles are often reared as biological control agents, the chemical cues involved in their sexual behavior could be used as components in integrated management approaches against soft-bodied hemipterans. In conclusion, this review provides new perspectives and potential developments for sustainable insect management mainly based on the semiochemicals involved in the sexual behavior of lady beetles. A better understanding of these chemical signals would help maximize their presence in areas where their predatory behavior is requested, as a result of the application of formulations inducing behavioral manipulation. Odorant receptors (ORs) play an important role in insects to monitor and adapt to the external environment, such as host plant location, oviposition-site selection, mate recognition and natural enemy avoidance. In our study, we identified and characterized OR12 from three closely-related species, Helicoverpa armigera, Helicoverpa assulta, Heliothis virescens, sharing between 90 and 98% of their amino acids. The tissue expression pattern analysis in H. armigera showed that HarmOR12 was strongly expressed both in male and female antennae, but not in other tissues. Functional analysis performed in the heterologous Xenopus expression system showed that all three OR12 were tuned to six structurally related plant volatiles. Electroantennogram recordings from male and female antennae of H. armigera closely matched the data of in vitro functional studies. Our results revealed that OR12 has a conserved role in Heliothinae moths and might represent a suitable target for the control of these crop pests. [ABSTRACT FROM AUTHOR]

Olfactory transduction is a process by which olfactory sensory neurons (OSNs) transform odor information into neuronal electrical signals. This process begins with the binding of odor molecules to receptor proteins on olfactory receptor neuron (ORN) dendrites. The major molecular components involved in olfaction include odorant-binding proteins (OBPs), chemosensory proteins (CSPs), odorant receptors (ORs), gustatory receptors (GRs), ionotropic receptors (IRs), sensory neuron membrane proteins (SNMPs) and odorant-degrading enzymes (ODEs). More importantly, as potential molecular targets, chemosensory proteins are used to identify novel attractants or repellants for environmental-friendly pest management. In this study we analyzed the transcriptome of the flea beetle, Phyllotreta striolata (Coleoptera, Chrysomelidae), a serious pest of Brassicaceae crops, to better understand the molecular mechanisms of olfactory recognition in this pest. The analysis of transcriptomes from the antennae and terminal abdomens of specimens of both sexes identified transcripts from several key molecular components of chemoreception including 73 ORs, 36 GRs, 49 IRs, 2 SNMPs, 32 OBPs, 8 CSPs, and four candidate odorant degrading enzymes (ODEs): 143 cytochrome P450s (CYPs), 68 esterases (ESTs), 27 glutathione S-transferases (GSTs) and 8 UDP-glycosyltransferases (UGTs). Bioinformatic analyses indicated that a large number of chemosensory genes were up-regulated in the antennae. This was consistent with a potential role in olfaction. To validate the differential abundance analyses, the expression of 19 genes encoding various ORs, CSPs, and OBPs was assessed via qRT-PCR between non-chemosensory tissue and antennae. Consistent with the bioinformatic analyses, transcripts for all of the genes in the qRT-PCR subset were elevated in antennae. Most web spiders use chemical and vibratory cues for intersexual communication prior to mating. Little is known regarding the behavior of the web spider, Pholcus manueli Gertsch, 1937. In the laboratory, we examined the behavior of males in female webs. Some webs were washed and allowed to air dry. Both washed and unwashed webs were tested with and without females. Overall, male behaviors were similar to courtship reported for other pholcid species and their frequency was related to male size. Web washing delayed the initiation of activity, reduced abdomen flexes, and increased the likelihood of pedipalp expansion and aggression. The presence of females caused males to initiate movement sooner and to increase silk pulling behavior. These results suggest that courtship is multimodal and that the vibratory dialog between males and females works along with chemical communication in this species. Juvenile Hormone (JH) has a prominent role in the regulation of insect development. Much less is known about its roles in adults, although functions in reproductive maturation have been described. In adult females, JH has been shown to regulate egg maturation and mating. To examine a role for JH in male reproductive behavior we created males with reduced levels of Juvenile Hormone Acid O-Methyl Transferase (JHAMT) and tested them for courtship. JHAMT regulates the last step of JH biosynthesis in the Corpora Allata (CA), the organ of JH synthesis. Males with reduced levels of JHAMT showed a reduction in courtship that could be rescued by application of Methoprene, a JH analog, shortly before the courtship assays were performed. In agreement with this, reducing JHAMT conditionally in mature flies led to courtship defects that were rescuable by Methoprene. The same result was also observed when the CA were conditionally ablated by the expression of a cellular toxin. Route learning is key to the survival of many central place foragers, such as bees and many ants. For ants which lay pheromone trails, the presence of a trail may act as an important source of information about whether an error has been made. The presence of trail pheromone has been demonstrated to support route learning, and the effect of pheromones on route choice have been reported to persist even after the pheromones have been removed. This could be explained in two ways: the pheromone may constrain the ants onto the correct route, thus preventing errors and aiding learning. Alternatively, the pheromones may act as a ‘reassurance’, signalling that the learner is on the right path and that learning the path is worthwhile. Here, we disentangle pheromone presence from route confinement in order to test these hypotheses, using the ant Lasius niger as a model. Unexpectedly, we did not find any evidence that pheromones support route learning. Indeed, there was no evidence that ants confined to the correct route learned at all. Thus, while we cannot support the ‘reassurance’ hypothesis, we can rule out the ‘confinement’ hypothesis. Other findings, such as a reduction in pheromone deposition in the presence of trail pheromones, are remarkably consistent with previous experiments. As previously reported, ants which make errors on their outward journey upregulate pheromone deposition on their return. Surprisingly, ants which would go on to make an error down-regulate pheromone deposition on their outward journey, hinting at a capacity for ants to gauge the quality of their own memories. Bark beetles (Coleoptera: Scolytinae) are pests of many forests around the world. The mountain pine beetle (MPB), Dendroctonus ponderosae Hopkins, is a significant pest of western North American pine forests. The MPB is able to overcome the defences of pine trees through pheromone-assisted aggregation that results in a mass attack of host trees. These pheromones, both male and female produced, are believed to be biosynthesized in the midgut and/or fat bodies of these insects. We used metabolite analysis, quantitative proteomics (iTRAQ) and transcriptomics (RNA-seq) to identify proteins and transcripts differentially expressed between sexes and between tissues when treated with juvenile hormone III. Juvenile hormone III induced frontalin biosynthesis in males and trans -verbenol biosynthesis in females, as well as affected the expression of many proteins and transcripts in sex- and tissue-specific ways. Egg-laying decisions are critical for insects, and particularly those competing for limited resources. Sensory information used by females to mediate egg-laying decisions has been reported to be primarily chemical, but the role of vibration has received little attention. We tested the hypothesis that vibrational cues produced by feeding larvae occupying a seed influences egg-laying decisions amongst female cowpea beetles. This hypothesis is supported by three lines of evidence using two strains of the cowpea beetle (Callosobruchus maculatus), an Indian strain with choosy females and aggressively competing larvae and a Brazilian strain with less choosy females and larvae exhibiting an “accommodating” type of competition. First, in free-choice bioassays of seed selection, choosy Indian females selected control seeds (free of eggs, larvae, or egg-laying marker) over seeds with live larvae (free of eggs and egg-laying marker), but did not discriminate between control seeds and those with dead larvae. In contrast, less choosy Brazilian females showed no preference for seeds containing live or dead larvae over controls. Second, laser-doppler vibrometer recordings confirmed that larvae feeding inside seeds generate vibrations that are available to the female during egg-laying decisions. Third, during dichotomous choice experiments where artificial vibrations approximating those produced by feeding larvae were played back during seed selection, Indian females preferred immobile control seeds over vibrating seeds, but Brazilian females showed no preference. These results support the hypothesis that females use larval vibrations in their egg-laying decisions; whether these vibrations are passive cues exploited by the female, or active signals that ‘steer’ the behaviour of the female is unknown. We propose that vibration cues and signals could be important for host selection in insects, particularly those laying on substrates where visual or chemical cues may be unreliable. This seems to be the case with females of the cowpea beetle since visual cues are not important and chemical egg-marking does not last more than two weeks, allowing vibration cues to improve discrimination of egg-laying substrate particularly by choosy females. Olfaction plays a crucial role in insect population survival and reproduction. Identification of the genes associated with the olfactory system, without the doubt will promote studying the insect chemical communication system. In this study, RNA-seq technology was used to sequence the antennae transcriptome of Athetis dissimilis, an emerging crop pest in China with limited genomic information, with the purpose of identifying the gene set involved in olfactory recognition. Analysis of the transcriptome of female and male antennae generated 13.74 Gb clean reads in total from which 98,001 unigenes were assembled, and 25,930 unigenes were annotated. Total of 60 olfactory receptors (ORs), 18 gustatory receptors (GRs), and 12 ionotropic receptors (IRs) were identified by Blast and sequence similarity analyzes. One obligated olfactory receptor co-receptor (Orco) and four conserved sex pheromone receptors (PRs) were annotated in 60 ORs. Among the putative GRs, five genes (AdisGR1, 6, 7, 8 and 94) clustered in the sugar receptor family, and two genes (AdisGR3 and 93) involved in CO2 detection were identified. Finally, AdisIR8a.1 and AdisIR8a.2 co-receptors were identified in the group of candidate IRs. Octopamine is an important neuromodulator of neural function in invertebrates. Octopamine increases male moth sensitivity to female sex pheromones, however, relatively little is known as to the role of octopamine in the female olfactory system, nor its possible effects on the reception of non-pheromone odorants. The purpose of this study was to determine relative effects of octopamine on the sensitivity of the peripheral olfactory system in male and female Heliothis virescens. Single sensillum recording was conducted in both sexes following injection with octopamine or Ringer solution, and during odorant stimulation with conspecific female sex pheromone or host plant volatiles. Results indicate that octopamine plays a significant modulatory role in female sex pheromone detection in female moths; and that male and female pheromone detection neurons share distinct pharmacological and physiological similarities in H. virescens despite sexual dimorphism at the antennal level. Background: The brown marmorated stink bug (BMSB) is an exotic insect pest that was first recognized in the United States in 2001. As of today, it has been found in more than 42 states. BMSB has a very broad host plant range and damage to crops in mid-Atlantic States has reached a critical level. A reliable and accurate tool for infestation detection and population monitoring is urgently needed to provide better and more timely interventions. Pheromones produced by male BMSB have been previously identified and are currently used in BMSB infestation detection. However, the conditions affecting BMSB production of these pheromones were unknown. Methodology/Principal Findings: In this study, we collected headspace volatiles from male BMSB under laboratory conditions, measured the temporal patterns of release of these pheromones, and assayed the attractiveness to conspecifics. In addition to the pheromone components, tridecane (C13) and E-2-decenal (an alarm compound) were observed in headspace collections of males, as well as in females and nymphs. Exposure of pheromone-emitting adult males to synthetic C13 greatly reduced pheromone emission. Conclusions/Significance: This information should lead to a better understanding of the biology, physiology, and chemical ecology of BMSB, which will help scientists and growers develop more efficient strategies based on natural products to manage BMSB population, therefore, reducing pesticide usage and protecting the crops from BMSB damage. Reproductive division of labour characterises eusociality. Currently little is known about the mechanisms that underlie the ‘sterility’ of the worker caste, but queen pheromone plays a major role in regulating the reproductive state. Here we investigate oogenesis in the young adult honey bee worker ovary in the presence of queen pheromone and in its absence. When queen pheromone is absent, workers can activate their ovaries and have well-developed follicles. When queen pheromone is present, even though workers have non-activated ovaries, they continually produce oocytes which are aborted at an early stage. Therefore, irrespective of the presence of the queen, the young adult worker ovary contains oocytes. By this means young workers retain reproductive plasticity. The degeneration of the germ cells in the ovarioles of workers in the presence of queen pheromone has the morphological hallmarks of programmed cell death. Therefore the mechanistic basis of ‘worker sterility’ relies in part on the regulation of oogenesis via programmed cell death. Our results suggest that honey bees have co-opted a highly conserved checkpoint at mid-oogenesis to regulate the fertility of the worker caste. [ABSTRACT FROM AUTHOR]

Male Nicrophorus burying beetles utilize two alternative mate-finding tactics: searching and signalling. The searching tactic involves flying in search of a carcass on which to mate, while the signalling tactic involves emitting pheromone in the absence of a carcass to attract females. All males use both tactics, but the amount of time spent using each tactic differs between individuals. Because all males are phenotypically plastic in the time spent using alternative behaviours, the expression is likely to be condition dependent on some environmental cue, such as the availability of reproductive resources (i.e. females and carcasses). We tested how the expression of alternative tactics is influenced by previous mating experience and the perceived availability of females versus carcasses by using a repeated measures design in which mate-finding behaviour of males was observed before and after exposure to: (1) multiple females but no carcasses, (2) multiple females and multiple carcasses, (3) multiple carcasses but no females, or (4) no females or carcasses (control). Males in the multiple-female group (but lacking a carcass) allocated significantly more time to signalling, as did males in the control group lacking both mating and carcass experience. Thus, males that did not encounter carcasses during the treatment period increased pheromone emission thereafter. In the two groups that were given carcasses, however, investment in signalling was not altered. With respect to time allocated to searching, control, multiple-female and multiple-carcass groups all significantly decreased time spent searching, whereas males in the female-and-carcass group did not alter time allocated to searching. Our results demonstrate that environmental cues do influence which alternative tactic male burying beetles use, and that the chosen tactic is influenced by the availability of both receptive females and carcasses on which to breed.
