Background: The success of malaria control using long-lasting insecticidal nets (LLINs) is threatened by pyrethroid resistance developed by the malaria vectors, worldwide. To combat the resistance, synergist piperonyl butoxide (PBO) incorporated LLINs is one of the available options. In the current phase II hut trial, the efficacy of Veeralin ® LN (an alpha-cypermethrin and PBO-incorporated net) was evaluated against Anopheles culicifacies, a pyrethroid resistant malaria vector., Methods: The performance of Veeralin ® LN was compared with MAGNet ® LN and untreated net in reducing the entry, induced exit, mortality and blood feeding inhibition of target vector species., Results: The performance of Veeralin was equal to MAGNet in terms of reducing hut entry, inhibiting blood feeding and inducing exophily, and with regard to causing mortality Veeralin was better than MAGNet. When compared to untreated net, a significant reduction in hut entry and blood feeding and an increase in exophily and mortality were observed with Veeralin. In cone bioassays, unwashed Veeralin caused > 80% mortality of An. culicifacies., Conclusions: Veeralin performed equal to (entry, exit, feeding) or better than (mortality in huts and cone bioassays) MAGNet and could be an effective tool against pyrethroid resistant malaria vectors.

Background: The development of resistance in vectors is one of the major impediments for malaria control. Adding synergists to insecticides has proven to be an alternative choice for controlling resistant mosquitoes. DawaPlus 3.0 and DawaPlus 4.0 are new long-lasting insecticidal nets (LLINs) in which deltamethrin and a synergist, piperonyl butoxide (PBO) are added into filaments and their efficacy was tested against resistant malaria vector, Anopheles culicifacies in experimental huts in India., Methods: The performance of two trial nets in terms of deterrence induced exiting, blood-feeding inhibition and mortality of An. culicifacies was compared with DawaPlus 2.0 and untreated net., Results: There was a significant reduction in entry, blood feeding and mortality (p < 0.05) and increase in exit rates of An. culicifacies in the treatment arms compared to untreated arm. But, both candidate LNs washed 20 times could not perform better than the washed reference net (DawaPlus 2.0). Cone bioassay results showed that all the treatment arms (both washed and unwashed) produced < 80% mortality of An. culicifacies before and after hut evaluation., Conclusions: DawaPlus 3.0 and DawaPlus 4.0 with their current specification may not be as effective as required to control the resistant vector, An. culicifacies, in east-central India.

Japanese encephalitis (JE) has been reported in different districts of Odisha state (east central India) since 1992. During 2016, a major outbreak of JE and acute encephalitis syndrome (AES) occurred in the Malkangiri district of Odisha, causing 103 deaths in children, of which 37 were caused by JE and 66 by AES. Information on insecticide resistance in JE vectors is important for the selection of appropriate insecticides for use in vector control. The present study was designed to determine the resistance status of three important vectors of JE, Culex vishnui, Culex tritaeniorhynchus and Culex bitaeniorhynchus (Diptera: Culicidae), against dichlorodiphenyltrichloroethane (DDT), malathion and deltamethrin in three districts of Odisha state affected by JE. Female adult mosquitoes were collected using mouth aspirators both indoors and outdoors in JE-affected villages and used in susceptibility bioassays following World Health Organization guidelines. Knock-downs were recorded every 10 min up to 1 h and mortality rates were recorded at 24 h post-exposure. Culex vishnui and Cx. tritaeniorhynchus showed resistance to DDT, malathion and deltamethrin, whereas Cx. bitaeniorhynchus was susceptible in all study districts. LifeNet, a deltamethrin incorporated long-lasting insecticidal (polypropylene) net (LLIN), was qualified by the World Health Organization Pesticide Evaluation Scheme (WHOPES) for Phase-II trial in India. The purpose of this trial was to assess the bio-efficacy of unwashed and 20 and 30 times washed LifeNet in comparison to the nets conventionally treated with deltamethrin against the natural population of Anopheles fluviatilis, a major malaria vector, in terms of deterring hut entry, inhibiting blood feeding, inducing exophily and causing mortality. The trial was carried out in six experimental huts constructed at Kandhaguda village in Malkangiri district, Odisha State. The efficacy of unwashed and washed (20 or 30 times) LifeNet was compared with untreated polypropylene and conventionally treated (with deltamethrin) polyester net washed to just before exhaustion or washed 20 times. The study showed a significant reduction of entry (treatment: 1.61-4.78; control: 7.61 per hut) and an increase in exit (50.7-64.4% and 39.1%) of An. fluviatilis in the treated arms compared to the control arm (untreated net) (P < 0.05). Blood feeding rates reduced in treated arms (20.7-68.0%) compared to the control (80.3%) (P < 0.05). Total mortality was significantly higher in LifeNet arms (73.8-98.3%) than the control (2.2%) (P < 0.05). After 30 washes, the active ingredient (AI) retention in LifeNet was 62%. Background & Objectives: Biolarvicides may offer alternatives to chemical larvicides as these are known to be safe to environment and selective against the target species. However, only a limited number of biolarvicides have been approved for mosquito larval control. In the current study, a new formulation of spinosad, 20 per cent emulsifiable concentrate (EC) was tested for its efficacy against Culex quinquefasciatus, in comparison to its 12 per cent suspension concentrate (SC)., Methods: Spinosad 20 per cent EC was tested against Cx. quinquefasciatus immature at 25, 50, 100 and 150 mg active ingredient (ai)/m [2] in cesspits, drains and abandoned wells in comparison with spinosad 12 per cent SC at the optimum field application dosage of 50 mg ai/m [2] ., Results: The 20 per cent EC caused 90-100 per cent reduction of pupal density for 7-14 days in cesspits, 10-17 days in drains and 14-30 days in abandoned wells at all dosages tested. At lower dosages of 25 and 50 mg ai/m [2] , >90 per cent reduction of pupal density was observed for one week in cesspits and street drains and for two weeks in abandoned wells. The effective duration of control provided by the higher dosages, 100 and 150 mg ai/m [2] was 1.4 to 2 times greater than the lower dosages, 25 and 50 mg ai/m [2] ., Interpretation & Conclusions: The findings showed that the spinosad 20 per cent EC can be used for larval control against Cx. quinquefasciatus, at the dosage of 25 mg ai/m [2] at weekly interval in cesspits and drains and at fortnightly interval in abandoned wells. Background: New brands of potential long lasting insecticide nets (LLINs) and LLIN treatment kits require field evaluation before they are used in a vector control programme., Objectives: The aim of this study was to evaluate the bio-efficacy, usage, washing practice and physical integrity of nets treated with LLIN treatment kit, ICON MAXX in a phase III field trial in Odisha state, India., Methods: A total of 300 polyester nets treated with ICON MAXX and 140 polyester nets treated conventionally with lambda-cyhalothrin CS 2.5% ITNs were distributed. The bio-efficacy was evaluated with WHO cone bioassay. The chemical analysis of netting pieces was done at the beginning, after 12 and 36 months of the trial., Findings: After one year of distribution of nets, the bioassay showed 100% mortality on both ITNs and ICON MAXX treated nets. At 36 months, the overall pass rate was 58.8% and the mean lambda-cyhalothrin content of LLINs was 34.5 mg ai/m2, showing a loss of 44.4% of the original concentration., Conclusion: ICON MAXX treated LLIN was found to retain bio-efficacy causing 97% knockdown of Anopheles stephensi up to 30 months and met the WHOPES criteria. However, the desired bio-efficacy was not sustained up to 36 months.

Background: Fast development of pyrethroid resistance in malaria vectors prompted the development of new vector control tools including combination of insecticides with different modes of action as part of resistance management strategies. Olyset Plus® is a new long-lasting insecticidal net, in which, permethrin and a synergist, piperonyl butoxide (PBO), are incorporated into filaments. Mixture nets such as this may have application against resistant mosquitoes, particularly those whose resistance is based on oxidative metabolism. There may also be enhanced activity against susceptible mosquitoes since mixed function oxidases are involved in a many metabolic activities including activation to form bioactive compounds., Methods: Bio-efficacy of Olyset Plus was evaluated against susceptible malaria vector, Anopheles fluviatilis in experimental huts. Deterrence, blood feeding inhibition, induced exophily and killing effect were measured to assess the bio-efficacy. The results were compared with Olyset Net®, a polyethylene permethrin-incorporated LLIN and a conventionally treated polyester net (with permethrin) washed to just before exhaustion., Results: Results showed significant reduction in entry (treatment: 0.4-0.8; control: 4.2 per trap-night) and increase in exit (56.3-82.9 % and 44.2 %) rates of Anopheles fluviatilis in the treatment arms compared to control (P < 0.05). While blood feeding rates declined in treatment arms (18.8-30.6 %), it increased in control (77.6 %) (P < 0.05). This was further evident from the blood-feeding inhibition rates in treatment arms (60.6-90.6 %). Total mortality was significantly higher in all treatment arms (96.3-100 %) compared to control arm (2 %) (P < 0.05). Chemical analysis for active ingredient (AI) showed retention of 75 and 88 % in Olyset plus and Olyset net respectively after 20 washes. Performance of Olyset Plus washed 20 times was equal to the CTN and Olyset Net against the susceptible malaria vector An. fluviatilis, fulfilling the WHO efficacy criteria of Phase II evaluation for LLIN. However, the benefit of incorporating PBO and permethrin together in a long-lasting treatment could not be demonstrated in the current study as the target vector species was fully susceptible to pyrethroids., Conclusion: Olyset Plus, with its intrinsic bio-efficacy could be an effective vector control tool to prevent transmission of malaria by susceptible vectors like An. fluviatilis. However, the results of the current study need to be further supported by testing the net at village level (Phase III) for community acceptability. Before taking the net to village level, it needs to be verified whether the net is better than pyrethroid nets in terms of bio-efficacy against resistant An. culicifacies, another malaria vector that has developed resistance to synthetic pyrethroids in India.

Background & Objectives: Selection of an insecticide and its appropriate formulation is a prerequisite of formulating any chemical control strategy against vectors. A hut scale field trial was carried out to study the effectiveness of house spraying with capsule suspension (CS) formulation of lambdacyhalothrin in comparison with its wettable powder (WP) formulation on mortality, density and behaviour of malaria vector in Malkangiri district, Odisha, India., Methods: The two formulations were tested at 20 and 30 mg (a.i.)/m 2 for their effectiveness in terms of deterrence, excito-repellency, blood-feeding success, mortality and residual activity against Anopheles fluviatilis, the major malaria vector, in experimental huts in Malkangiri district, Odisha State, India., Results: Both CS and WP formulations prevented the entry of An. fluviatilis in to the sprayed huts by >90 per cent for >6 months, the entire peak malaria transmission season in the area. The exit rate increased (90-99%) with different treatments and the feeding rate was reduced (91-97%). There was no significant difference between WP 30, CS 20 and CS 30 mg/m 2 in these respects. However, WP 20 mg/m 2 caused a lesser effect than the other three groups. The formulations or the dosages differed only in causing vector mortalities. Overall, the total mortality rate of An. fluviatilis was higher in the huts sprayed with CS 30 (58%) than the huts sprayed with CS 20 (48%), WP 20 (37%) and WP 30 mg/m 2 (47%)., Interpretation & Conclusions: Considering the duration of residual action and the effect on entry, exit, feeding and mortality of An. fluviatilis, the dosage 20 mg/m 2 of CS formulation of lambdacyhalothrin could be considered for further use.

Five types of long-lasting insecticidal nets (LNs), namely, Olyset, Netprotect, PermaNet, DuraNet, and Interceptor, were tested after 20 washes for efficacy in terms of mortality, deterrence effect, blood-feeding inhibition, and induced exophily of the malaria vector Anopheles fluviatilis in experimental huts in Malkangiri district of Odisha State, India. Efficacy of the three synthetic pyrethroids (SPs) used in the LNs was also analyzed. Use of LNs reduced the entry of An. fluviatilis into the huts by 73.3-83.2%, and the five LNs were comparable in terms of deterrence. The exit rate of An. fluviatilis from the huts with untreated net was 56.3%, and relative to this, Olyset followed by DuraNet induced significantly a higher exophily. In contrast, the exit rate was significantly lower with Interceptor. Among the three SPs, permethrin induced significantly greater exophily relative to the untreated control, and as a result of this, permethrin-treated Olyset produced a lower mortality. Blood-feeding rate of An. fluviatilis was significantly lower with all the five LNs than the control. Similarly, all the three SPs significantly inhibited blood feeding compared with the control. Interceptor and DuraNet, both alphacypermethrin-treated LNs, caused relatively a higher mortality of An. fluviatilis than the other LNs. The five brands of LNs and three SPs tested in the current study were equally effective in terms of deterrence and blood-feeding inhibition; only exiting and killing effect differed among them. Permethrin-treated LNs induced greater exophily, while, overall, alphacypermethrin-treated LNs killed more An. fluviatilis that entered the huts. Advantage of deterrence, excito-repellent, and killing effects of LNs and appropriate selection of SP for net treatment are discussed in this paper.

Background & Objectives: Updating information on response (susceptible / resistant status) of vectors to the insecticides in use is essential to formulate and introduce appropriate resistance management strategy. Therefore, a study was undertaken in the 10 southern districts of Odisha State, which are endemic for Plasmodium falciparum malaria, to determine the insecticide susceptibility/ resistance status of Anopheles fluviatilis and An. culicifacies, the vectors of malaria., Methods: Mosquitoes were collected during September 2010 - February 2012 from 60 randomly selected villages in the 10 districts and blood-fed females were exposed to the diagnostic dosage of DDT (4.0%), malathion (5.0%) and deltamethrin (0.05%) for one hour. Mortality was recorded at 24 h after the exposure. The test mortality was corrected to the control mortality., Results: An. fluviatilis was susceptible to the three insecticides tested while, An. culicifacies was resistant to DDT and malathion in all the 10 districts except in two, where its response against malathion was under 'verification required' category. Against deltamethrin, An. culicifacies was susceptible in two districts; while in the other eight districts its response was under 'verification required' category., Interpretation & Conclusions: Since An. fluviatilis the vector species primarily associated with transmission of malaria, was still susceptible to DDT, indoor residual spraying with DDT could be continued in the 10 districts. Also, in view of the large scale implementation of long lasting insecticidal nets and the signs of development of resistance in An. culicifacies to deltamethrin, response of the vectors to synthetic pyrethroids needs to be periodically monitored.

Background & Objective: Pirimiphos-methyl, an organophosphorus insecticide is known to overcome resistance mechanisms of mosquitoes to other organophosphates. Information on the duration of efficacy of pirimiphos-methyl (50% emulsifiable concentrate) for the control of immatures of Culex quinquefasciatus, the vector of bancroftian filariasis, is scanty and hence site specific field trials are necessary to determine the field dosage. Pirimiphos-methyl (50% EC) was tested for its efficacy in cesspits and drains against the immatures of Cx. quinquefasciatus at Puducherry, an endemic area for filariasis, south India., Methods: In cesspits, Pirimiphos-methyl (50% EC) was tested at the dosage of 25, 50, 100 and 200 g(ai)/ ha and in drains at 50, 100 and 200 g(ai)/ha. Immature density during pre-treatment and post-treatment periods was recorded and the efficacy was determined by calculating percentage reduction in larval and pupal densities for a period of about one month., Results: In cesspits, application of the insecticide at 25 and 50 g(ai)/ha reduced the density of larvae by >80 per cent for 2-4 days only, whereas at 100 and 200 g(ai)/ha, the efficacy lasted for 7-8 days. More than 80 per cent reduction was observed in pupal density for 4 days at 50 g(ai)/ha and for 10-12 days at 100 and 200 g(ai)/ha respectively. In drains treated with 50 g (ai)/ha, >or=80 per cent reduction in larval density was recorded up to 3(rd) day post-treatment. The efficacy showing >or=80 per cent reduction in larval density lasted for 12-13 days and the same in pupal density lasted for 10-12 days at 100 and 200 g(ai)/ha., Interpretation & Conclusion: At the dosages of 100 and 200 g(ai)/ha, pirimiphos-methyl (50% EC) showed no difference in the control of larvae of Cx. quinquefasciatus in cesspits and drains. Pirimiphosmethyl may be applied at the field dosage of 100 g (ai)/ha at 10 day-intervals in the larval habitats of Cx. quinquefasciatus to achieve >80 per cent reduction in immature density.

Novaluron 10% emulsifiable concentrate (EC), an insect growth regulator (IGR), was tested against Culex quinquefasciatus immatures at 1, 5, and 10 mg active ingredient (AI)/m2 applied to cesspits, drains, and abandoned wells. The IGR produced 80-100% inhibition of adult emergence (IE) for > 1 week (10-14 days) in cesspits, 1-2 weeks in street drains, and 5-10 weeks in abandoned wells at the 3 application rates. The efficacy increased with the increasing dose in street drains and abandoned wells. The residual activity of the IGR was longer in abandoned wells compared to drains and cesspits. Based on the small-scale trial, doses of 1, 5, and 10 mg AI/m2 were used in the medium-scale trial for cesspits, abandoned wells, and street drains, respectively. The efficacy observed in medium-scale trial was similar to that in the small-scale trial. The results indicate that novaluron 10% EC can be used for larval control against Cx. quinquefisciatus at the application rate of 1 mg AI/m2 at 10-day intervals in cesspits, at 5 mg AI/m2 at 2-month intervals in abandoned wells, and at 10 mg AI/m2 applied at fortnightly intervals to drains. Even at the lowest rate of 1 mg AI/m2, the IGR prevented adult emergence for at least a week in cesspits and street drains and for a month in abandoned wells. Novaluron 10% EC could be one of the options for larval control operations in integrated vector management.

Spinosad 12% suspension concentrate (SC) and 0.5% granular (G) formulations were tested against Culex quinquefasciatus immatures at 3 dosages--50, 100, and 150 mg active ingredient (AI)/m2--in cesspits, street drains, and disused wells in comparison with temephos 1% G at 1 part per million. The 2 formulations reduced the density of pupae of Cx. quinquefasciatus by 80-100% for 7-14 days in cesspits, 7-18 days in drains, and 33-69 days in disused wells at the 3 dosages tested. The 12% SC and 0.5% G formulations were equally effective in suppressing the larval and pupal populations of Cx. quinquefasciatus in all the habitats treated. At 100 mg AI/m2 in drains and cesspits, the efficacy of spinosad 0.5% G was greater than that of temephos (1% G) and that of spinosad 12% SC was comparable to that of temephos (1% G) applied to the 2 habitats. In disused wells, both the formulations lasted longer than temephos. Based on the small-scale trial, the rate of 150 mg AI/m2 of spinosad was selected for field application to cesspits and drains and 50 mg AI/m2 for application to disused wells. The efficacy observed in medium-scale trials was in agreement with that of small-scale trials. The trial showed that the 2 formulations of spinosad can be used for larval control of Cx. quinquefasciatus, at 50 mg AI/m2, applied at weekly intervals to cesspits and street drains and at monthly intervals to disused wells. At the higher dose of 150 mg AI/m2, the 2 formulations were effective in suppressing the pupal population for 2 wk in cesspits and street drains and for 2 months in disused wells.

Background & Objective: Insecticide treated mosquito nets are increasingly being used in malaria control programmes. One of the problems with the treatment of bed nets with conventional formulations of insecticides was that regular washing of treated nets diminish insecticidal effect. Lambdacyhalothrin 2.5 capsule suspension (CS) (2.5% a.i., w/v), a new water-based microencapsulated formulation is reported to have wash-resistant property and longer persistence on the netting material than other formulations. We evaluated the impact of the use of nylon bed nets treated with lambdacyhalothrin 2.5 CS at 10 mg (a.i.)/m(2) in comparison to untreated nets and no nets on malaria in tribal villages in Orissa., Methods: Nine foothill villages, highly endemic for falciparum malaria, from the Primary Health Centre (PHC) areas of Khairput and Kudumulugumma of Malkangiri district, Orissa, were divided into three groups, each with a population of about 500 and allocated randomly for treated (TN) and untreated nets (UN) and no nets (NN). Bed nets were distributed in September 2001 and retreatment was done in June 2002. The impact was assessed based on the changes in vector density, parous rate, malaria incidence and parasite rates. Indoor-resting collections of Anopheles fluviatilis and An. culicifacies were made at fortnightly intervals from fixed human dwellings. Mass blood surveys before and after distribution of nets and fortnightly active surveillance were carried out to assess the change in parasite rates and malaria incidence. Bioassays were conducted at fortnightly intervals on the bed nets supplied to the villagers., Results: The reductions in indoor resting catches of An. fluviatilis and An. culicifacies were 96 and 38 per cent in villages with treated nets and 2.6 and 23 per cent in villages with untreated nets respectively compared to no net villages. For six months following treatment, 100 per cent mortality of An. fluviatilis was observed on the unwashed nets and on the nets washed once or twice. After re-treatment, 100 per cent mortality of An. fluviatilis or An. culicifacies was observed for nine months even after two washes. Usage rates of treated and untreated nets varied seasonally; 58.9 and 46.3 per cent in rainy season, 48.6 and 37.1 per cent in winter season and 38.1 and 31.6 per cent in summer season respectively. Reductions in malaria parasite rates were about 65 per cent in the treated net villages and 39 per cent in the untreated net villages compared to no net villages. About 75 per cent of treated nets and 60 per cent of untreated nets were in usable condition 19 months after distribution., Interpretation & Conclusion: The estimated protection factor based on malaria incidence was 86 per cent for the treated nets during both post-treatment and post-retreatment periods and 34 and 51 per cent for untreated nets for the corresponding periods compared to no nets. The results of the study showed that the use of bed nets treated nets with CS formulation of lambdacyhalothrin at 10 mg (a.i.)/m(2) was acceptable to the community and re-treatment of nets at nine-monthly intervals can significantly reduce density and survival of An. fluviatilis and incidence of falciparum malaria.

The study assessed the operational feasibility and acceptability of insecticide-treated mosquito nets (ITNs) in one Primary Health Centre (PHC) in a falciparum malaria endemic district in the state of Orissa, India, where 74% of the people are tribes and DDT indoor residual spraying had been withdrawn and ITNs introduced by the National Vector Borne Disease Control Programme. To a population of 63,920, 24,442 ITNs were distributed free of charge through 101 treatment centers during July-August 2002. Interview of 1,130, 1,012 and 126 respondents showed that the net use rates were 80%, 74% and 55% in the cold, rainy and summer seasons, respectively. Since using ITNs, 74.5-76.6% of the respondents observed reduction of mosquito bites and 7.2-32.1% reduction of malaria incidence; 37% expressed willingness to buy ITNs if the cost was lower and they were affordable. Up to ten months post-treatment, almost 100% mortality of vector mosquitoes was recorded on unwashed and washed nets (once or twice). Health workers re-treated the nets at the treatment centers eight months after distribution on a cost-recovery basis. The coverage reported by the PHC was only 4.2%, mainly because of unwillingness of the people to pay for re-treatment and to go to the treatment centers from their villages. When the re-treatment was continued at the villages involving personnel from several departments, the coverage improved to about 90%. Interview of 126 respondents showed that among those who got their nets re-treated, 81.4% paid cash for the re-treatment and the remainder were reluctant to pay. Majority of those who paid said that they did so due to the fear that if they did not do so they would lose benefits from other government welfare schemes. The 2nd re-treatment was therefore carried out free of charge nine months after the 1st re-treatment and thus achieved coverage of 70.4%. The study showed community acceptance to use ITNs as they perceived the benefit. Distribution and re-treatment of nets was thus possible through the PHC system, if done free of charge and when personnel from different departments, especially those at village level, were involved.

The Directorate of Public Health (DPH), Tamil Nadu, in southern India employed spraying of dichlorvos (76% EC) for quick elimination of fly concentrations in the tsunami-hit coastal villages at the concentration of 304g (a.i.)/10,000m(2). However, nuisance of house flies remained high particularly in temporary shelters and centralized relief kitchens. Susceptibility of house fly, Musca domestica to dichlorvos was determined in the laboratory to provide information for an effective management of this pest. Various concentrations of dichlorvos (76% EC) viz., 0.1, 0.2, 0.4, 0.6 and 0.8microg (a.i.) per fly, were tested using topical application against F(1) progenies of house flies collected 12 months after insecticide applications from different habitats in the tsunami-hit coastal villages. Fly mortality was recorded at 24h post treatment. Parallel controls were maintained for comparison. Mortality of the house flies varied between 17.5% and 100% and increased with an increase in dosage of the insecticide. Mortality was >80% at 0.6 and 0.8microg (a.i.) per fly. The LD(50) of dichlorvos tested against flies collected from different villages varied from 0.218microg (a.i.) to 0.235microg (a.i.) per fly and the LD(90) varied from 0.574microg (a.i.) to 0.639microg (a.i.) per fly. House flies collected from a rural village, Thirukanur that had never been exposed for insecticide treatment in the past one decade, when tested, the mortality varied between 92.5% and 100% and increased with concentration of dichlorvos. Mortality was >90% from 0.2microg (a.i.) per fly and the LD(50) was 0.0399microg (a.i.)/fly, while the LD(90) was 0.1604microg (a.i.)/fly. The LD(90) values of the flies collected from the tsunami-hit villages were 3.5-3.9 times higher than that of the flies collected from Thirukanur. Fly abundance remained high in tsunami-hit villages with no marked reduction, suggesting that the flies had developed tolerance to dichlorvos. It is suggested that for an effective management of these resistant populations changing insecticides, application of unrelated insecticide, together with an appropriate environmental sanitation measure is necessary to keep the population under check.

Background & Objective: In cockroach control, bait formulations are extensively used. Fipronil is one of the broad spectrum insecticides from phenyl pyrazole family available in bait formulation. The compound has been tested under field conditions. However, information on its effectiveness on different surfaces and at various density levels is not known. Therefore, the efficacy of a new formulation of fipronil 0.03 per cent (Goliath gel) was tested under laboratory conditions., Methods: The bait formulation was tested against Periplaneta americana at the dosages of 0.025, 0.05, 0.1, 0.15 and 0.2 g/m(2), and Blatella germanica at 0.025, 0.05, 0.075, 0.1 and 0.125 g/m(2) on wood, cement, mud and thatch surface with three density levels of cockroaches viz., 5, 10 and 20 numbers/m(2). Mortality after 24 h of exposure was observed daily for five days., Results: Rate of mortality varied between 16.0 and 96.0 per cent in P. americana and 51.0 and 100.0 per cent in B. germanica on different surfaces tested. The mortality rate of P. americana increased line rarly from the dosage 0.025g to 0.15 g/m(2) and at 0.2 g/m(2), it was independent. The LD(80) values of the gel against P. americana at high density were 0.208, 0.246, 0.361 and 0.466 g/m(2) on wood, cement, mud and thatch surfaces, whereas against B. germanica the values were 0.079, 0.081, 0.089 and 0.055 g/m(2) respectively, indicating a significantly (P<0.001) higher efficacy of Goliath gel against B. germanica than P. americana. The results of fitting logistic regression model to the observed percentage mortality with log dose and cockroach density as explanatory variables satisfactorily described the observations at all densities on each surface. The pattern of response to increasing dosages was similar for all the three density levels on each of the surfaces in P. americana and B. germanica., Interpretation & Conclusion: The effectiveness assessed against P. americana and B. germanica using logistic regression model suggested that the gel when applied at the appropriate dosages on wood, cement, mud and thatch surfaces could cause >80 per cent mortality of these species in dwellings having these types of surfaces.

The impact of use of bed-nets treated with alphacypermethrin, at 20 mg (ai)/m2, in comparison to untreated nets or no nets on malaria vectors and malaria incidence was studied in tribal villages of Malkangiri district, Orissa state, India, which are highly endemic for falciparum malaria. Treated or untreated nets were supplied to the villagers in June 1999 and the nets were re-treated in September 1999, just before the rise in vector abundance and malaria incidence. The seasonal pattern of indoor resting Anopheles fluviatilis females was similar in all the three groups of villages before the start of intervention and the indoor resting catches were not significantly different between treatment-groups (two-way ANOVA, F = 1.53; d.f. = (2, 78); P = 0.2). During intervention, the indoor resting catches differed significantly among treatment groups (two-way ANOVA, F = 38.9; d.f.= (2, 66); P < 0.005). There was a 99% reduction in the indoor resting catches of An. fluviatilis in villages with treated nets and 61% reduction in villages with untreated nets compared with no-net villages. Comparison between villages with and without treated nets showed that there was 97% reduction in indoor resting catches in villages with treated nets. Pair-wise comparison showed that the reductions between villages with and without nets as well as between villages with treated and untreated nets were significant (Dunnett's C-test, P < 0.05). The indoor resting catches of Anopheles culicifacies did not differ significantly among the three groups of villages either before (F = 0.99; d.f. = (2, 121); P = 0.4) or during intervention (F = 0.21; d.f. = (2, 66); P = 0.8). Bioassay with 3 min exposure to treated bed nets showed 100% mortality of An. culicifacies for 2 months and with An. fluviatilis for 4.5 months after which tests were not carried out. In villages with treated nets, the Annual Parasite Incidence (API) significantly declined (P < 0.05) by about 65.7% and prevalence of infection among children (< 15 years) declined by 57.1%, whereas in villages with untreated nets, there was only 34% reduction in API and 13% in the prevalence of infection. In villages with treated nets, there was 48% reduction in API and 64% in prevalence of infection compared with villages with untreated nets. The impact of use of treated bed nets on other arthropod pests lasted for at least 1.5 months. After 1.5 months, observations on arthropod pests were not continued. The use rate of treated nets varied from 49.8 to 93.7% in three seasons and about 68.3% of treated bed nets and 60% of untreated nets were in good condition 1 year after distribution. Out of 489 users of treated net, five people complained of a burning sensation on the face (skin irritation) for 5 days following the distribution of treated nets. There were no other complaints of any discomfort in using the treated nets. The use of alphacypermethrin treated bed nets at 20 mg (ai)/m2 can be one of the options for reducing the vector abundance and incidence of malaria in this area.

Background & Objectives: The resistance in vectors to conventional insecticides warrants development of newer insecticides for mosquito control besides use of other counter-measures. The present study was undertaken to test the efficacy of a new and toxicologically safe organophosphorus compound, Reldan (chlorpyrifos-methyl) and compare its efficacy with Dursban (chlorphyrifos-ethyl) against larvae of different mosquito vectors under laboratory conditions and against Culex quinquefasciatus in three larval habitats., Methods: The susceptibility of larvae of mosquito vector species to Reldan and Dursban was determined by following the standard procedure and the LC50 in mg/l values were calculated by probit analysis. The insecticides were evaluated in the larval habitats of C. quinquefasciatus in and around Pondicherry. The percentage reduction in larval density was calculated using Mulla's formula and duration showing more than 80 per cent reduction was calculated by annotating the graph obtained by plotting percentage reduction in larval density against days of post-treatment., Results: Dursban and Reldan were effective against Anopheles fluviatilis larvae at lower LC50 of 5.90 x 10(-7) and 1.07 x 10(-9) mg/l respectively compared to other species of mosquitoes. Dursban and Reldan were equally effective against Aedes aegypti, An. stephensi and An. culicifacies. Reldan was about nine times more effective than Dursban against C. quinquefasciatus with respective LC50 of 1.17 x 10(-4) and 1.34 x 10(-3) mg/l. Field evaluation of these insecticides against C. quinquefasciatus showed that Reldan and Dursban at the application rate of 500 g (ai)/ha reduced immature density by more than 80 per cent for a mean duration of 4.0 and 4.5 days and 4.6 and 3.7 days in cesspits and cement tanks respectively. In U-drains, at 500 g (ai)/ha, Reldan was effective for a period of 3.6 days whereas, Dursban was not effective. It was effective for 4.0 days only at 1000 g(ai)/ha. There was no significant increase in the residual activity of both Dursban and Reldan with increase in application rates., Interpretation & Conclusion: In view of the lack of sustained control with the applications, it is inferred that these insecticides will be of limited use as larvicides against C. quinquefasciatus breeding in polluted habitats.

Spicbiomoss, an aqueous suspension formulation of Bacillus sphaericus was evaluated for its efficacy against immatures of Culex quinquefasciatus at the application rate of 120 l/ha in cement tanks, cesspits and drains in Pondicherry and in drains in Kochi, Kerala. The formulation was also tested against anophelines (Anopheles fluviatilis and An. culicifacies) breeding in stream pools in Malkangiri district, Orissa. In cement tanks and cesspits more than 80 per cent reduction in immature density was observed for a period of 6-13 days (mean 9.8 +/- 1.25 days) and 3-8 days (mean 5.2 +/- 0.7 days) respectively. The same level of reduction was found to last for 1-4 days (mean 2.2 +/- 0.52 days) in drains in Pondicherry and 2-9 days (mean 4.8 +/- 1.17 days) in Kochi. In bunded stream pools 40-79 per cent reduction in immature density of Anopheles was obtained for an average period of 1-7 (mean 3.83 +/- 0.98) days. There was no improvement in the efficacy of the formulation against anophelines even at the higher application rate (240 l/ha). Thus, Spicbiomoss can be used against Cx. quinquefasciatus in an integrated vector management programme.

A village scale trial was carried out to evaluate the efficacy of bed-nets impregnated with lambdacyhalothrin, at the dose of 0.025 g/m2, in reducing malaria transmission in villages of Koraput District of Orissa, India, inhabited by tribals. The nets were distributed before peak transmission season. There was an overall decline in the parasite rate in all the age groups, six months after the supply of impregnated nets while the same increased in control village and in a village where untreated nets were supplied. The vector densities (resting and man landing) were lower in the treated village as compared to untreated and control villages throughout the study period. The reduction in the parasite rate was consistent when the reimpregnation was done at six monthly interval and the same tend to increase when the gap between the two impregnations was increased to one year. Though malaria incidence was reduced, transmission was not completely interrupted during the study period, due to outdoor transmission. The insecticidal effect of bednets was retained upto six months. Washing of bednets by the community did not affect the efficacy. The acceptance and usage was better with impregnated nets as compared to ordinary nets.

The role of permethrin impregnated bednets at the application rate of 0.5 g (ai)/m2 in reducing A. culicifacies population in a tribal village of Orissa, highly endemic for Plasmodium falciparum was studied. The per man-hour density was significantly reduced in the Kenduguda village for about three months where impregnated bednets were supplied to the villagers, while in the control village Deulaguda where unimpregnated nets were supplied, vector density remained high during the study period. Bioassay carried out on the treated nets showed that there was 100 per cent mortality in A. culicifacies for over three months with the unwashed nets as well as with bednets washed once or twice.
