Fall armyworm, Spodoptera frugiperda (J. E. Smith), is a widely recognized global agricultural pest that has significantly reduced crop yields all over the world. S. frugiperda has developed resistance to various insecticides. Insect cytochrome P450 monooxygenases (CYPs or P450s) play an important role in detoxifying insecticides, leading to increased resistance in insect populations. However, the function of the specific P450 gene for lambda-cyhalothrin resistance in S. frugiperda was unclear. Herein, the expression patterns of 40 P450 genes in the susceptible and lambda-cyhalothrin-resistant populations were analyzed. Among them, CYP321A7 was found to be overexpressed in the resistant population, specifically LRS (resistance ratio = 25.38-fold) derived from a lambda-cyhalothrin-susceptible (SS) population and FLRS (a population caught from a field, resistance ratio = 63.80-fold). Elevated enzyme activity of cytochrome P450 monooxygenases (P450s) was observed for LRS (2.76-fold) and the FLRS (4.88-fold) as compared to SS, while no significant differences were observed in the activities of glutathione S-transferases and esterases. Furthermore, the knockdown of CYP321A7 gene by RNA interference significantly increased the susceptibility to lambda-cyhalothrin. Remarkably, the knockdown of CYP321A7 reduced the enzymatic activity of P450 by 43.7%, 31.9%, and 22.5% in SS, LRS, and FLRS populations, respectively. Interestingly, fourth-instar larvae treated with lambda-cyhalothrin at the LC 30 dosage had a greater mortality rate due to RNA interference-induced suppression of CYP321A7 (with increases of 61.1%, 50.0%, and 45.6% for SS, LRS, and FLRS populations, respectively). These findings suggest a link between lambda-cyhalothrin resistance and continual overexpression of CYP321A7 in S. frugiperda larvae, emphasizing the possible importance of CYP321A7 in lambda-cyhalothrin detoxification in S. frugiperda., (Copyright © 2024 Elsevier Inc. Spodoptera litura (Fabricius) (Lepidoptera: Noctuidae) is one of the most destructive insect pests owned strong resistance to different insecticides. Indoxacarb as a novel oxadiazine insecticide becomes the main pesticide against S. litura. DIMBOA [2,4-dihydroxy-7-methoxy-2 H-1,4-benz-oxazin-3(4 H)-one] is involved in important chemical defense processes in corn plants. However, the insects' adaptation mechanism to insecticides when exposed to defensive allelochemicals in their host plants remains unclear. Here, we assessed multi-resistance, and resistance mechanisms based on S. litura life history traits. After 18 generations of selection, indoxacarb resistance was increased by 61.95-fold (Ind-Sel) and 86.06-fold (Dim-Sel) as compared to the Lab-Sus. Also, DIMBOA-pretreated larvae developed high resistance to beta-cypermethrin, chlorpyrifos, phoxim, chlorantraniliprole, and emamectin benzoate. Meanwhile, indoxacarb (LC 50 ) was applied to detect its impact on thirty-eight detoxification-related genes expression. The transcripts of SlituCOE073, SlituCOE009, SlituCOE074, and SlituCOE111 as well as SlGSTs5, SlGSTu1, and SlGSTe13 were considerably raised in the Ind-Sel strain. Among the twenty-three P450s, CYP6AE68, CYP321B1, CYP6B50, CYP9A39, CYP4L10, and CYP4S9v1 transcripts denoted significantly higher levels in the Ind-Sel strain, suggesting that CarEs, GSTs and P450s genes may be engaged in indoxacarb resistance. Herbivore-induced plant volatiles (HIPVs) have been associated with plant-plant-herbivorous-natural enemies communication and an enhanced response to the subsequent attack. Spodoptera litura is a serious cosmopolitan pest that has developed a high level of resistance to many insecticides. However, the underlying molecular and biochemical mechanism by which HIPV priming reduces S. litura larval sensitivity to insecticides remains largely unknown. This study was conducted to explore the potential of volatile from undamaged, or artificially damaged, or S. litura-damaged tomato plants on the susceptibility of S. litura to the insecticides beta-cypermethrin indoxacarb and chlorpyrifos. We found that larvae exposed to volatile from S. litura-damaged or artificially damaged tomato plants were significantly less susceptible to the three insecticides than those exposed to volatile from undamaged tomato plants. Elevated activities of detoxifying enzymes [cytochrome P450 monooxygenases (P450s), glutathione S-transferases (GSTs), and esterases (ESTs)], were expressed in S. litura larvae exposed to volatile from S. litura-damaged tomato plants than those exposed to volatile from undamaged tomato plants. Similarly, seven detoxification-related genes [GSTs (SlGSTe1, SlGSTo1, and SlGSTe3) and P450s (CYP6B48, CYP9A40, CYP321A7, and CYP321B1)] in the midgut and fat body of larvae were up-regulated under exposure to volatile from S. litura-damaged tomato plants. Increased volatile organic compounds emissions were detected in the headspace of tomato plants damaged by S. litura compared to the undamaged plants. Collectively, these findings suggest that HIPVs can considerably reduce caterpillar susceptibility to insecticides, possibly through induction-enhanced detoxification mechanisms, and provide valuable information for implementing an effective integrated pest management strategy., (Copyright © 2022. Many species of devastating insect pests have acquired a high degree of resistance to insecticides in the field during the last few decades. Spodoptera exigua, for example, is the most damaging pests of economic crops with a worldwide spread. In a present study, the comparative growth, reproduction, and detoxification enzyme activity were evaluated along with exposure to three insecticides at low lethal doses of lufenuron, indoxacarb, and spinosad as compared to the control. Results indicate that the larval developmental time was significantly extended on lufenuron (21.5 ± 29 days) followed by indoxacarb (20.28 ± 0.24 days) and spinosad (19.74 ± 0.23 days) as compared to that on the control (18.13 ± 0.13 days). Similarly, the lowest number of eggs of S. exigua females were recorded on lufenuron (328.75 ± 50.81 eggs) followed by spinosad (367 ± 36.4 eggs) and indoxacarb (411.58 ± 42.38 eggs) as compared to that on the control (560.2 ± 13.47). Interestingly, the lowest intrinsic rate of increase (r) (0.121 ± 0.009) and highest mean generation time (T) (36.2 ± 0.35 days) were observed when larvae were treated to a low lethal concentration (LC 20 ) of lufenuron as compared to that of indoxacarb, spinosad, and control. In addition, considerably lower activity of all detoxification enzymes in larvae was recorded on lufenuron after control as compared to that on indoxacarb and spinosad. Our study serves as a reference and basis for the toxicity and low lethal evaluation of lufenuron, indoxacarb, and spinosad on life table parameters and enzymatic properties in S. exigua, which may contribute to identifying targets for effective control of S. exigua., (© 2022. The beet army worm, Spodoptera exigua, is a widely distributed polyphagous pest of economically important crops worldwide. The management of this pest insect continues to face many challenges. Despite synthetic chemicals posing a serious threat to the environment, these remain the conventional approach for controlling S. exigua in the field. An over-reliance on chemical control has not only led to selection for resistance to insecticides and to a reduction of natural enemies, but has also polluted various components of ecosystem. Given these increasing pressures on the ecosystem, there is a need to implement integrated pest management (IPM) approaches exploiting a wider range of tools (biotechnological approaches, microbial control, biological control, cultural control, and use of host plant resistance) for an alternative to chemical control. The IPM approach can not only reduce the hazard of chemical residues in the environment and associated health problems, but may also provide best strategies to control insect pests. Paederus fuscipes is a general predator in rice fields and a non-target organism of chlorantraniliprole, an effective insecticide for insect-pest control in paddy fields. Pesticide hazards to non-target organisms have been a growing global problem for decades. This study was designed to evaluate the toxicity of chlorantraniliprole at lethal and sublethal levels against P. fuscipes larvae and adults. The LC 50 of chlorantraniliprole against P. fuscipes adults and larvae were respectively 535.49 and 111.24 mg a.i. L -1 , which is higher than the dosage recommended for use in the field (59.38 mg a.i. L -1 ), but the LC 30 and LC 10 for larvae are lower than the recommended field dose which showed that the sublethal effects on immature stages are inevitable. Treatment at larval stage with LC 30 of chlorantraniliprole significantly elongated the pre-imaginal developmental and pre-oviposition periods. Also, adults exposed directly to chlorantraniliprole oviposited significantly less number of eggs in both LC 10 and LC 30 treatments. Furthermore, the larval predation efficiency and female bodyweight were also reduced due to exposure to sublethal doses. Meanwhile, the activities of antioxidant (SOD, POD and CAT) and detoxification (P450, AChE and GST) enzymes were also significantly affected by the exposure to these sublethal concentrations. These findings showed that sublethal doses of chlorantraniliprole adversely influenced P. fuscipes development and physiology, and therefore its use as part of integrative pest management should be given further considerations., (Copyright © 2021 Elsevier Ltd. Spodoptera litura (Fabricius) is a widely distributed, highly polyphagous pest that can cause severe damage to a variety of economically important crops. Various populations have developed resistance to different classes of insecticides. In this study, we report on two indoxacarb-resistant S. litura populations, namely Ind-R (resistance ratio = 18.37-fold) derived from an indoxacarb-susceptible (Ind-S) population and a population caught from a field (resistance ratio = 46.72-fold). A synergist experiment showed that piperonyl butoxide (PBO) combined with indoxacarb produced higher synergistic effects (synergist ratio = 5.29) in the Ind-R population as compared to Ind-S (synergist ratio = 3.08). Elevated enzyme activity of cytochrome P450 monooxygenases (P450s) was observed for Ind-R (2.15-fold) and the Field-caught population (4.03-fold) as compared to Ind-S, while only minor differences were noticed in the activities of esterases and glutathione S-transferases. Furthermore, expression levels of P450 genes of S. litura were determined by quantitative reverse transcription PCR to explore differences among the three populations. The results showed that the mRNA levels of CYP6AE68, a novel P450 gene belonging to the CYP6 family, were constitutively overexpressed in Ind-R (32.79-fold) and in the Field-caught population (68.11-fold). CYP6AE68 expression in S. litura was further analyzed for different developmental stages and in different tissues. Finally, we report that RNA interference-mediated silencing of CYP6AE68 increased the mortality of fourth-instar larvae exposed to indoxacarb at the LC 50 dose level (increase by 33.89%, 29.44% and 22.78% for Ind-S, Ind-R and the Field-caught population, respectively). Bradysia odoriphaga is a major insect pest that infests Chinese chive in northern China. Clothianidin is a second-generation neonicotinoid insecticide that is commonly used against B. odoriphaga. In this study, the effect of sublethal clothianidin concentrations (LC 5 and LC 10 ) on key biological characteristics of B. odoriphaga was investigated using an age-stage, two-sex life table method. Bioassays results showed that clothianidin exhibited high toxicity against B. odoriphaga with LC 50 of 1.898 mg L -1 following 24 h exposure. The developmental duration of larvae was significantly increased when exposed to the LC 5 (0.209 mg L -1 ) and LC 10 (0.340 mg L -1 ) of clothianidin. No significant effects were observed on the pupal stage, adult pre-oviposition period (APOP), total pre-oviposition period (TPOP), and mean longevities of male and female. The oviposition period and fecundity of B. odoriphaga were reduced in clothianidin-treated groups. Moreover, key demographic parameters, including the intrinsic rate of increase (r), finite rate of increase (λ), and net reproductive rate (R 0 ), were significantly decreased by the LC 5 and LC 10 of clothianidin, while no effects were noted on mean generation time (T). Overall, this study showed that sublethal concentrations of clothianidin have a detrimental effect on B. odoriphaga developmental period, fecundity, and life table parameters. The beet armyworm, Spodoptera exigua, is a highly polyphagous pest originated from Southeast Asia but has spread globally, attacking economically important crops and fruits. Bistrifluron insecticide is one of the highly active insect growth regulators that has been reported to inhibit development and longevity in other lepidopteran species and could be used in the control of S. exigua. In the present study, the age-stage, two-sex life table technique was applied to assess the sublethal effects of bistrifluron on biological traits and vitellogenin gene (SeVg) expression when 2nd instar larvae fed to sublethal concentrations (LC 10 , LC 20 and LC 40 ) of bistrifluron. Mean generation time from eggs to adults was longer at LC 40 (37.79 ± 0.81 d) and LC 20 (37.04 ± 0.72) compared to the LC 10 (36.89 ± 0.63 d) and control groups (36.07 ± 0.38 d). Fecundity of female at LC 40 (279.17 ± 42.8 eggs), LC 20 (347 ± 35.4 eggs) and LC 10 (411.58 ± 42.38 eggs) were significantly lower than the control treatment (532.47 ± 7.13). Furthermore, the lower intrinsic rates of increase (LC 40 ; r = 0.1207 ± 0.009, LC 20 ; r = 0.1329 ± 0.009 and LC 10 ; r = 0.14398 ± 0.009 compared to the control r = 0.164 ± 0.0076), was observed along with significantly extended mean generation times (LC 40 ; T = 34.825 ± 0.317 days, LC 20 ; T = 33.27 ± 0.368 days and LC 10 ; T = 31.899 ± 0.398 days compared to the control 30.927 ± 0.255 days). Furthermore, the contents of energy reserve macronutrients (carbohydrate, lipid and protein) significantly reduced in dose and time dependent manner in treated insects as compared to control. Furthermore, the expression level of SeVg mRNA significantly decreased by 43.8% in the female adults when one-day-old second instar larvae were treated with sublethal concentrations of bistrifluron in comparison with the control. Insect predators, in general, play an important role in regulating pest populations in agricultural systems, but may be negatively affected by pesticides used in pest management. Convergent lady beetle, Hippodamia convergens Guerin-Meneville (Coleoptera: Coccinellidae) is known as an important biological control agent of soft-bodied insect pests. The development of Integrated Pest Management (IPM) program against insect pests requires an assessment of the side effect of insecticides on biological control agents. In the present work, we assessed the sublethal effects of an insect growth regulator, Pyriproxyfen (juvenile hormone mimic) on a common aphidophagous beetle, H. convergens by computing its demographic parameters through age-stage, two-sex life table theory. Present results showed that sublethal (LC 10 and LC 30 ) concentrations of tested insecticide prolonged the pre-adult developmental duration while adult longevity, fecundity and fertility were reduced following treatment compared with control. However, total pre-ovipositional period (TPOP) was declined with the increasing trend of insecticide concentration. In additions, population growth parameters such as intrinsic rate of increase r, finite rate of increase λ and net reproductive rate R 0 were dramatically reduced in H. convergens population when they treated with sublethal concentrations pyriproxyfen. Therefore, the results obtained through this study reflected that pyriproxyfen impairs the population growth parameters and could reduce the biological services provided by H. convergens. Thus, more attention should be paid to the use of this insecticide in IPM program.

The beet armyworm (Spodoptera exigua) is a highly polyphagous agricultural pest that is distributed worldwide. However, the adaptive mechanisms of S. exigua for various insecticides and defensive substances in host plants are unknown. Insect P450 monooxygenases play an important role in the detoxification of plant toxins and insecticides, leading to insecticides resistance. We investigated the induced effects of xanthotoxin exposure on detoxification enzyme activity and larval tolerance to α-cypermethrin in S. exigua. Our results showed that the lethal concentration (LC 50 ) of α-cypermethrin for xanthotoxin-exposed larvae was 2.1-fold higher than in the control. Moreover, cytochrome P450 enzyme activity was significantly elevated by upregulation of P450 genes in treated larvae. RT-qPCR results showed that CYP9A10 expression level was significantly increased in all treatments, while maximal expression level was observed in xanthotoxin+α-cypermethrin-fed larvae. RNAi-mediated silencing of CYP9A10 further increased mortality by 18%, 26% and 35% at 48 h and by 27%, 43% and 55% at 72 h when larvae were exposed to diets containing chemicals as compared to the control. The results show that CYP9A10 might play an important role in xanthotoxin and α-cypermethrin detoxification in S. exigua. Plants employ an intricate and dynamic defense system that includes physiological, biochemical, and molecular mechanisms to counteract the effects of herbivorous attacks. In addition to their tolerance to phytotoxins, beet armyworm has quickly developed resistance to deltamethrin; a widely used pyrethroid insecticide in cotton fields. The lethal concentration (LC 50 ) required to kill 50% of the population of deltamethrin to gossypol-fed Spodoptera exigua larvae was 2.34-fold higher than the control group, suggesting a reduced sensitivity as a consequence of the gossypol diet. Piperonyl butoxide (PBO) treatment was found to synergize with deltamethrin in gossypol-fed S. exigua larvae. To counteract these defensive plant secondary metabolites, beet armyworm elevates their production of detoxification enzymes, including cytochrome P450 monooxygenases (P450s). Gossypol-fed beet armyworm larvae showed higher 7-ethoxycoumarin-O-deethylase (ECOD) activities and exhibited enhanced tolerance to deltamethrin after 48 and 72 h when compared to the control. Moreover, gossypol pretreated S. exigua larvae showed faster weight gain than the control group after transferring to a deltamethrin-supplemented diet. Meanwhile, gossypol-induced P450s exhibited high divergence in the expression level of two P450 genes: CYP6AB14 and CYP9A98 in the midgut and fat bodies contributed to beet armyworm tolerance to deltamethrin. Knocking down of CYP6AB14 and CYP9A98 , via double-stranded RNAs (dsRNA) in a controlled diet, rendered the larvae more sensitive to the insecticide. These data demonstrate that generalist insects can exploit secondary metabolites from host plants to enhance their defense systems against other toxic chemicals. Impairing this defense pathway by RNA interference (RNAi) holds a potential to eliminate the pest's tolerance to insecticides and, therefore, reduce the required dosages of agrochemicals in pest control.

Spotted bollworm, Earias vittella, is one of the most serious and devastating insect pests of vegetables and cotton. Currently, insecticides are necessary for its control in nearly all crop systems. In this paper, we evaluate the sub-lethal effects of lufenuron on biological traits and activity of detoxification enzymes: cytochrome P450 monooxygenases, esterase, and glutathione S-transeferase (GST) in second instar larvae of E. vittella. Results showed that sub-lethal concentrations (LC 15 and LC 40 of lufenuron), prolonged larval period (at LC 40  = 13.86 ± 1.22 day, LC 15  = 13.14 ± 1.15 day, control = 12.28 ± 0.7), pupal duration (LC 40  = 11.1 ± day, LC 15  = 11.8 ± 0.28 day, control = 9.40 ± 0.52), and extended mean generation time (LC 40  = 27.3 ± 0.43 LC 15  = 29.0 ± 1.19 day, control = 26.0 ± 0.65). Sub-lethal exposure significantly prolonged the pre-adult stage, decreased pupal weight, and reduced adult longevity in the parent (F 0 ) and F 1 generation. Moreover, the fecundity and egg viability were significantly lowered in parental and F 1 generations at both sub-lethal concentrations compared to the control. While no significant effects were noted on reproductive parameters such as the intrinsic rate of increase (r), finite rate of increase (λ), and net reproduction rate (R 0 ) of F 1 generation when compared to the control. Only mean generation time (T) in F 1 at LC 15 was significantly longer compared to the LC 40 and control (LC 40  = 3.79 ± 0.37, LC 15  = 32.28 ± 1.55 day, control = 29.79 ± 0.55). Comparatively, the activities of cytochrome P450 monooxygenases and esterase were higher than GST in treated populations. The increase in resistance development against insecticides may possibly because of elevated activity of detoxification enzymes. These results provide useful information for monitoring resistance in integrated pest management (IPM) programs for E. vittella.

The beet armyworm, Spodoptera exigua (Hübner) (Lepidoptera: Noctuidae) is a highly polyphagous pest which causes considerable economic losses to cotton and many vegetable crops. Tannins are among the most important secondary metabolites in cotton plants. We show that tannic acid enhances the toxic effect of chlorantraniliprole on S. exigua when presented in combination. Bioassays using third-instar S. exigua larvae on an artificial diet showed that consumption of tannic acid with chlorantraniliprole at the concentration of (2 mg/g and LC 50 0.018 mg/L) had higher toxicity when compared to either chlorantraniliprole or tannic acid alone (LC 50 0.027 mg/L). The diet containing tannic acid with chlorantraniliprole significantly prolonged larval and pupal developmental time and extended mean generation time and total pre-oviposition period compared to either chemical alone. Moreover, fecundity, survival rate, reproductive value, intrinsic rate of increase, finite rate of increase and net reproduction rate declined significantly when exposed to the combined treatment. No difference was observed between tannic acid and the control. Meanwhile, tannic acid with chlorantraniliprole had markedly antifeedant effects; causing significant decline in the relative growth rate (RGR), the relative consumption rate (RCR), the efficiency of conversion of ingested food (ECI), the efficiency of conversion of digested food and an increase in the approximate digestibility (AD) compared to either chemical alone. Tannic acid with chlorantraniliprole also decreased the insect's carbohydrate, lipid and protein contents significantly. The results showed that the interaction between tannic acid and chlorantraniliprole on the growth inhibition of larvae was additive and tannic acid increased the toxicity of chlorantraniliprole to insects. The results of this study provide information useful in integrated pest management programs for S. exigua and show that tannic acid combined with chlorantraniliprole may be a route to reducing the use of synthetic pesticides., (Copyright © 2019. Insecticide resistance is a major challenge in successful insect pest control as the insects have the ability to develop resistance to various widely used insecticides. Butene-fipronil is a novel compound with high toxicity to insects and less toxicity to the non-target organisms. In the present study, the effect of butene-fipronil alone and in combination with three enzyme inhibitors, piperonyl butoxide (PBO), diethyl maleate (DEM), and triphenyl phosphate (TPP), was carried out on larvae and adults of Drosophilia melanogaster. Our results indicated that the co-toxicity indices of butene-fipronil + PBO, butene-fipronil + TPP, and butene-fipronil + DEM mixtures were 437.3, 335.0, and 210.3, respectively, in the second-instar larvae, while 186.6, 256.2, and 238.5, respectively, in the adults, indicating synergistic effects. Interestingly, butene-fipronil increased the expression of CYP28A5 in the larvae; CYP9F2, CYP304A1, CYP28A5, and CYP318A1 in the female adults; and CYP303A1 and CYP28A5 in the male adults. Furthermore, high-level expression of Est-7 was observed in the female adults compared to larvae and male adults. Our results suggest that there is no difference in butene-fipronil metabolism in larvae and male and female adults of D. melanogaster.

Simple Summary: The beet armyworm Spodoptera exigua (Hübner) is a polyphagous pest of numerous cultivated crops including potatoes, beans, asparagus, cotton, etc., causing considerable economic losses in crop production worldwide. Currently, the use of chemical insecticides is the most commonly used method to control this pest, however, the overuse of them results in insecticide resistance, environmental pollution and toxicity to other non-target organisms. Our results indicated that S. exigua field populations had developed resistance to almost half of the eleven commonly used insecticides with different modes of action. Thus, it is extremely urgent to seek an efficient strategy for insecticide resistance management. We found that the combined application of the specific entomopathogen of S. exigua (Spodoptera exigua multiple nucleopolyhedrovirus, SeMNPV) reduced insecticide resistance or even recovered the susceptibility to insecticides and enhanced the toxicity obviously in both laboratory experiments and field trials. These findings are valuable to provide a promising efficient way for improving insecticide resistance management strategy and an environmentally friendly approach for pest management with the combined application of nucleopolyhedroviruses and insecticides. Spodoptera exigua is a polyphagous pest of diverse crops and causes considerable economic losses. The overuse of chemical insecticides for controlling this pest results in insecticide resistance, environmental pollution and toxicity to other non-target organisms. Therefore, a sustainable and efficient way for pest management is urgently required. In this study, laboratory bioassays of eleven commonly used insecticides, the specific entomopathogen of S. exigua (Spodoptera exigua multiple nucleopolyhedrovirus, SeMNPV), and SeMNPV-insecticide combinations against the S. exigua laboratory population and two field populations were tested. Our results indicated that the two field populations had developed resistance to almost half of the tested insecticides, while SeMNPV had good virulence in all populations. Interestingly, the combined use of SeMNPV enhanced the toxicity of the tested insecticides against all populations to a different extent and considerably reduced the insecticide resistance of S. exigua field populations or even recovered the susceptibility to above insecticides. Furthermore, the field trial showed that the combined application of SeMNPV contributed to promoting the control efficacy of emamectin benzonate and chlorfenapyr. Simple Summary: The fall armyworm (FAW), Spodoptera frugiperda (J.E. Smith), is a voracious insect pest native to the Western Hemisphere, particularly in South America. The polyphagous fall armyworm feeds on more than 350 plants in such families, including Poaceae, Asteraceae, and Fabaceae. Transgenic plants that express Bacillus thuringiensis proteins (Bt plants) and synthetic insecticides are the main tactics to control Spodoptera frugiperda, although widespread usage of synthetic chemicals has resulted in the emergence of resistance. We assessed cross-resistance, resistance mechanism, and fitness costs based on the life history traits of Spodoptera frugiperda. Results indicated that after 24 generations of selection, the resistance to indoxacarb was increased by 472.67-fold as compared to the Ind-UNSEL. Significantly longer developmental time of larvae extended pupal duration, shorter adult longevity, and lower fecundity were observed in Ind-SEL as compared with the Ind-UNSEL population. Butoxide synergist increased susceptibility to indoxacarb, indicating that P450 enzymes may be involved in indoxacarb resistance. Therefore, it is crucial that we comprehend how insecticides work and how resistance develops in order to develop techniques for managing resistance. Insecticide resistance poses many challenges in insect pest control, particularly in the control of destructive pests such as red imported fire ants (Solenopsis invicta). In recent years, beta-cypermethrin and fipronil have been extensively used to manage invasive ants, but their effects on resistance development in S. invicta are still unknown. To investigate resistance development, S. invicta was collected from populations in five different cities in Guangdong, China. The results showed 105.71- and 2.98-fold higher resistance against fipronil and beta-cypermethrin, respectively, in the Guangzhou population. The enzymatic activities of acetylcholinesterase, carboxylases, and glutathione S-transferases significantly increased with increasing beta-cypermethrin and fipronil concentrations. Transcriptomic analysis revealed 117 differentially expressed genes (DEGs) in the BC-ck vs. BC-30 treatments (39 upregulated and 78 downregulated), 109 DEGs in F-ck vs. F-30 (33 upregulated and 76 downregulated), and 499 DEGs in BC-30 vs. F-30 (312 upregulated and 187 downregulated). Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis revealed that DEGs associated with insecticide resistance were significantly enriched in metabolic pathways, the AMPK signaling pathway, the insulin signaling pathway, carbon metabolism, peroxisomes, fatty acid metabolism, drug metabolism enzymes and the metabolism of xenobiotics by cytochrome P450. Furthermore, we found that DEGs important for insecticide detoxification pathways were differentially regulated under both insecticide treatments in S. invicta. Comprehensive transcriptomic data confirmed that detoxification enzymes play a significant role in insecticide detoxification and resistance development in S. invicta in Guangdong Province. Numerous identified insecticide-related genes, GO terms, and KEGG pathways indicated the resistance of S. invicta workers to both insecticides. Importantly, this transcriptome profile variability serves as a starting point for future research on insecticide risk evaluation and the molecular mechanism of insecticide detoxification in invasive red imported fire ants. Abstract Harmonia axyridis (Pallas) is a major biological control agent that is indirectly threatened by sulfoxaflor insecticide applications targeting pests in agro-ecosystems. However, the genome-wide response of H. axyridis to sulfoxaflor remains largely unknown. Here, we used high-throughput RNA-seq technology to characterize the variation in the transcriptomic profile of second-instar H. axyridis larvae after exposure to sulfoxaflor. Two libraries were generated, with 50,702,976 and 47,005,096 clean reads for the sulfoxaflor treatment and control, respectively; these were grouped into 112,497 transcripts and 52,229 unigenes, of which 33,820 and 25,175 unigenes were respectively matched to proteins in the NCBI (Nr) and Swiss-Prot databases. From these unigenes, 794 significant differentially expressed genes (DEGs) consisting of 446 up- and 348 down-regulated genes, namely P450s, GSTs, USTs, ESTs, and others, were identified between the sulfoxaflor-treated and control groups. Furthermore, these DEGs were involved in several KEGG pathways including drug metabolism and metabolism of xenobiotic pathways. For transcriptome validation, 10 specifically expressed genes were selected and verified by quantitative real-time PCR analysis. Together, these results demonstrated that sulfoxaflor induces notable variation in the transcriptome profile and pathways of H. axyridis , which establishes a basis for further molecular investigations. Insecticide resistance is a major challenge in successful insect pest control as the insects have the ability to develop resistance to various widely used insecticides. Butene-fipronil is a novel compound with high toxicity to insects and less toxicity to the non-target organisms. In the present study, the effect of butene-fipronil alone and in combination with three enzyme inhibitors, piperonyl butoxide (PBO), diethyl maleate (DEM), and triphenyl phosphate (TPP), was carried out on larvae and adults of Drosophilia melanogaster. Our results indicated that the co-toxicity indices of butene-fipronil + PBO, butene-fipronil + TPP, and butene-fipronil + DEM mixtures were 437.3, 335.0, and 210.3, respectively, in the second-instar larvae, while 186.6, 256.2, and 238.5, respectively, in the adults, indicating synergistic effects. Interestingly, butene-fipronil increased the expression of CYP28A5 in the larvae; CYP9F2, CYP304A1, CYP28A5, and CYP318A1 in the female adults; and CYP303A1 and CYP28A5 in the male adults. Furthermore, high-level expression of Est-7 was observed in the female adults compared to larvae and male adults. Except of pest control, insecticides have shown adverse effects on natural enemies as well. Thus, risk assessment of pesticides for biological control agents is critical for effective use in integrated pest management (IPM) schemes. In the present study, the lethal and sublethal effects of chlorpyrifos, a commonly used insecticide that may negatively affect biological control agents, were evaluated on a non-target predator, the Asian ladybeetle Harmonia axyridis. Previous studies have reported on lethal concentrations, but the effects of sublethal concentrations remain unclear. Lethal and sublethal concentrations of chlorpyrifos were applied to third instar larvae of H. axyridis, and different growth and developmental parameters were measured. Treatment with LC10 (4.62 mg a.i. L−1) significantly shortened the developmental period of third instar larvae, whereas it significantly prolonged those of fourth instar larvae and pupa. Treatment with LC30 (9.59 mg a.i. L−1) significantly increased the larval and pupal developmental period compared with that of the control, whereas feeding potential, female fecundity, and adult longevity significantly decreased after LC10 and LC30 treatment. The pre-oviposition period significantly increased compared with that of the control. Population growth parameters, the finite (λ) and intrinsic rate of increase (r) and the net reproductive rate (R0), decreased following exposure to sublethal concentrations of chlorpyrifos. According to the results, the use of chlorpyrifos in IPM schemes requires further research because even sublethal concentrations of this insecticide were harmful to H. axyridis population growth.
