Purpose: Age-related macular degeneration is a vision-threatening disorder affecting the posterior segment of the eye. Drug delivery to the posterior segment is challenging owing to the complex anatomical and physiological structure, necessitating monthly injections of antivascular endothelial growth factors. Thermoresponsive hydrogels provide sustained drug delivery and ease of injection, due to their sol-gel transition. Poly (N-isopropyl acrylamide) (PNIPAAm) is a widely researched thermoresponsive hydrogel; however, insufficient wet strength and a wide mesh network make it inept for the entrapment of small molecules. Methods: A novel approach of grafting PNIPAAm with chitosan is exploited. A chitosan concentration altered in 10%, 30%, and 50% compared to PNIPAAm is investigated for entrapment of a small-molecular weight, hydrophilic drug, sunitinib (SUN), a multiple tyrosine kinase receptor inhibitor. Furthermore, these hydrogels were characterized using 1 H-NMR, FTIR, differential scanning calorimetry (DSC), and thermogravimetric analysis for chemical characterization and viscosity, swellability, syringeability, degradation, and In-vitro permeation using Franz-diffusion cell. Results: In-vitro drug release kinetics suggested that the release of SUN could be controlled with the percentage of chitosan grafting; however, gel strength (3%-5% w/v) of 30% Cs-g-PNIPAAm did not significantly affect percentage drug release. Sustained release of SUN was observed for 1 month. In-vitro permeation studies on porcine sclera suggested that a thermoresponsive gel of chitosan grafted PNIPAAm (Cs-g-PNIPAAm) was able to sustain the drug release by 40%, compared to SUN solution. Conclusions: The study indicates that the synthesized Cs-g-NIPAAm hydrogel has the potential to serve as a tailorable injectable platform for intrascleral drug delivery applications.

Contact lenses may act as drug release platforms for the treatment of ocular infections, but there is still the need for extending their typical release periods and enhancing ocular bioavailability. The present study aimed to develop a molecularly imprinted silicone-based hydrogel to be used in the manufacturing of contact lenses that can be loaded efficiently and be able to release the antibiotic moxifloxacin hydrochloride (MXF) in a sustained way. A set of hydrogels was prepared by the molecular imprinting method using acrylic acid (AA) as the functional monomer for the specific recognition of MXF. The modified hydrogels loaded a higher amount of MXF, which was released for a longer time. In vitro experiments, using a microfluidic cell to mimic the ocular surface fluid turnover, showed that the imprinted hydrogel TRIS(300)-I prepared with the highest content in AA led to MXF concentrations in the release medium which were effective against S. aureus and S. epidermidis for about 2 weeks. Furthermore, some important properties such as water uptake, wettability, transmittance, ionic permeability, and Young´s modulus of the modified hydrogel remained within the range of values recommended for contact lenses. No cytotoxicity and no potential ocular irritancy effect were detected. A combined strategy to control the release of two drugs, one anti-inflammatory (diclofenac sodium, DCF) and one antibiotic (moxifloxacin hydrochloride, MXF), from a soft contact lens (SCL) material, was assessed. The material was a silicone-based hydrogel, which was modified by molecular imprinting with MXF and coated by the layer-by-layer (LbL) method using natural polyelectrolytes: alginate (ALG), poly-l-lysine (PLL) and hyaluronate (HA), crosslinked with 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride (EDC). Imprinting was used to increase the amount of MXF loaded and to sustain its release, while the LbL coating acted as a diffusion barrier for DCF and improved the surface properties. The drugs were loaded by soaking in a DCF + MXF dual solution. High hydrostatic pressure (HHP) was successfully applied in the sterilization of the drug-loaded hydrogels. The transmittance, refractive index, wettability and ionic permeability of the hydrogels remained within the required levels for SCLs application. The concentrations of the released DCF and MXF stayed above the IC 50 and the MIC (for S. aureus and S. epidermidis) values, for 9 and 10 days, respectively. No ocular irritancy was detected by the HET-CAM test. A layer-by-layer (LbL) coating was designed using ionic polysaccharides (chitosan, sodium alginate, sodium hyaluronate) and genipin (crosslinker), to sustain the release of diclofenac sodium salt (DCF) from soft contact lens (SCL) materials. The coating was hydrophilic, biocompatible, non-toxic, reduced bacterial growth and had minor effects on the physical properties of the material, such as wettability, ionic permeability, refractive index and transmittance, which remained within the recommended values for SCLs. The coating was applied on a silicone-based hydrogel and on commercial SofLens and Purevision SCLs. The coating attenuated the initial drug burst and extended the therapeutic period for, at least, two weeks. Responsiveness of drug delivery systems (DDS) against internal and external stimuli attracts wide interest as a mechanism that can provide both site-specific release at the target place and feedback regulated release rate. Biological environment is quite complex and the effects that the intricate medium may have on the effectiveness of the stimulus have received certain attention. Differently, the impact that the drug loaded may have itself on the responsiveness of the DDS has been underestimated. Most drugs are not merely trapped in the polymer network, but they effectively interact with some polymer moieties. Nearly all drugs, including therapeutic proteins, are ionizable amphiphilic molecules, and thus ionic, hydrogen bonding and hydrophobic interactions are commonly exploited to increase the loading yield. If the moiety involved in drug binding is also responsible for (or at least partially involved in) the stimuli responsiveness, a strong impact of the drug on the behavior of the DDS can be expected. This review gathers relevant examples of how the drug may modify the sensitiveness (stimulus threshold) and the responsiveness (actuation) of the DDS to therapeutically relevant stimulus, and aims to shed light on the different drug binding modes of the swollen and collapsed states, which in turn modify drug release patterns. The information evidences that drug loading and release may trigger phase transitions in hydrogels non-intended to be drug-responsive (i.e., a priori not analyte-responsive networks). Naltrexone (NTX) is a potent opioid growth factor receptor (OGFR) antagonist proved to be useful for treatment of ocular surface complications. The aim of this work was to explore the feasibility of designing NTX-imprinted 2-hydroxyethyl methacrylate-based hydrogels for sustained drug release on the ocular surface. Acrylic acid (AAc) and benzyl methacrylate (BzMA) were chosen as functional monomers able to form binding cavities mimicking OGFR binding sites for NTX. Imprinted hydrogels containing functional monomers loaded higher amounts of NTX compared to non-imprinted ones by simple soaking in drug aqueous solution. In addition, possibility of carrying out the loading and sterilization processes in one step was investigated. NTX release was evaluated both under agitated sink conditions and in a microfluidic flow chamber mimicking the hydrodynamic conditions of the eye, namely the small volume of lachrymal fluid and its renovation rate. Sustained release profiles together with adequate swelling degree (46 to 57% w/w), light transparency (over 85%) and oxygen permeability may make these hydrogels suitable candidates to NTX-eluting contact lenses. NTX-loaded and non-loaded discs successfully passed the chorioallantoic membrane test for potential ocular irritation and were cytocompatible with human mesenchymal stem cells. Finally, NTX-imprinted hydrogels tested in the bovine corneal permeability assay provided therapeutically relevant amounts of NTX inside the cornea, reaching drug levels similar to those attained with a concentrated aqueous solution in spite the discs showed sustained release., (Copyright © 2019 Elsevier B.V. Chronic wounds are a prevailing cause of decreased quality of life, being microbial burden a factor hindering the normal wound healing process. Aerogels are nanostructured materials with large surface area (>250 m 2 /g) and high porosity (>96%). In this work, vancomycin-loaded chitosan aerogel beads were tested as a potential formulation to treat and prevent infections at the wound site. Processing of chitosan in the form of aerogels endowed this polysaccharide with enhanced water sorption capacity and air permeability. The morphological and textural properties of the particles were studied by image and N 2 adsorption-desorption analysis and scanning electron microscopy. Vancomycin content and release profiles from aerogel carriers showed a fast drug release that permitted to efficiently achieve local therapeutic levels. Cell studies with fibroblasts and antimicrobial tests against S. aureus showed that the vancomycin-loaded aerogel particles were cytocompatible and effective in preventing high bacterial loads at the wound site., (Copyright © 2018 Elsevier Ltd. Efficient ocular drug delivery that can overcome the challenges of topical application has been largely pursued. Contact lenses (CLs) may act as light-transparent cornea/sclera bandages for prolonged drug release towards the post-lens tear fluid, if their composition and inner architecture are fitted to the features of the drug molecules. In this review, first the foundations and advantages of using CLs as ocular drug depots are revisited. Then, pros and cons of common strategies to prepare drug-loaded CLs are analyzed on the basis of recent examples, and finally the main section focuses on bioinspired strategies that can overcome some limitations of current designs. Most bioinspired strategies resemble a reverse engineering process to create artificial receptors for the drug inside the CL network by mimicking the human natural binding site of the drug. Related bioinspired strategies are being also tested for designing CLs that elute comfort ingredients mimicking the blinking-associated renewal of eye mucins. Other bioinspired approaches exploit the natural eye variables as stimuli to trigger drug release or take benefit of bio-glues to specifically bind active components to the CL surface. Overall, biomimicking approaches are being revealed as valuable tools to fit the amounts loaded and the release profiles to the therapeutic demands of each pathology. STATEMENT OF SIGNIFICANCE: Biomimetic and bioinspired strategies are remarkable tools for the optimization of drug delivery systems. Translation of the knowledge about how drugs interact with the natural pharmacological receptor and about components and dynamics of anterior eye segment may shed light on the design criteria for obtaining efficient drug-eluting CLs. Current strategies for endowing CLs with controlled drug release performance still require optimization regarding amount loaded, drug retained in the CL structure during storage, regulation of drug release once applied onto the eye, and maintenance of CL physical properties. All these limitations may be addressed through a variety of recently growing bioinspired approaches, which are expected to pave the way of medicated CLs towards the clinics., (Copyright © 2018 Acta Materialia Inc. Most treatments for diabetic eye conditions rely on systemic (oral) or intravitreal administration, and there is still a demand of efficient and comfortable ocular dosage forms. Our purpose was to design contact lenses (CLs) suitable for local prophylaxis/treatment of diabetes-related ocular pathologies, by means of the incorporation of bioinspired functional groups that can reversibly interact with epalrestat, an aldose reductase inhibitor. Several sets of silicone hydrogels were synthesized varying the contents in 2-hydroxyethyl methacrylate (HEMA), monomethacryloxypropyl-sym-polydimethylsiloxane hydroxypropyl terminated (MCS-MC12), and aminopropyl methacrylamide (APMA). Epalrestat was incorporated before or after polymerization, and loading and release profiles compared. All sets were evaluated regarding optical properties, oxygen permeability, swelling, cytocompatibility, ocular irritation, and corneal drug penetration (using a drug solution as reference). Designed silicone hydrogels showed adequate properties to be used as CLs. Affinity for epalrestat strongly depended on the content in APMA, which endowed the hydrogels with prolonged release in 0.9% NaCl for one week, both after synthesis and after being re-loaded. Bovine corneal permeability tests demonstrated that epalrestat released from the hydrogels can efficiently accumulate into the cornea in spite the concentrations provided on cornea surface were lower than those attained after instillation of concentrated eyedrops. Epalrestat-loaded hydrogels also demonstrated anti-cataract activity in an in vitro model of diabetic eye. Overall, silicone hydrogel CLs functionalized with bioinspired chemical groups represent a first attempt to design CLs adapted to the needs of diabetic eyes, acting as controlled release platforms of epalrestat, promoting drug accumulation and diffusion through cornea., (Copyright © 2017 Elsevier B.V. This paper describes the synthesis of novel molecularly imprinted hydrogels (MIHs) for the natural antioxidant ferulic acid (FA), and their application as packaging materials to prevent lipid oxidation of butter. A library of MIHs was synthesized using a synthetic surrogate of FA, 3-(4-hydroxy-3-methoxyphenyl)propionic acid (HFA), as template molecule, ethyleneglycol dimethacrylate (EDMA) as cross-linker, and 1-allylpiperazine (1-ALPP) or 2-(dimethylamino)ethyl methacrylate (DMAEMA), in combination with 2-hydroxyethyl methacrylate (HEMA) as functional monomers, at different molar concentrations. The DMAEMA/HEMA-based MIHs showed the greatest FA loading capacity, while the 1-ALLP/HEMA-based polymers exhibited the highest imprinting effect. During cold storage, FA-loaded MIHs protected butter from oxidation and led to TBARs values that were approximately half those of butter stored without protection and 25% less than those recorded for butter covered with hydrogels without FA, potentially extending the shelf life of butter. The intrinsic ability of albumin to bind active substances in the physiological fluids has been explored to endow hydrogels with improved capability to regulate drug release. To develop such biomimetic-functional hydrogels, it is critical that albumin conformation is not altered and that the protein remains retained inside the hydrogel keeping its conformational freedom, i.e., it should be not chemically cross-linked. Thus, the hydrogels were prepared with various proportions of albumin by physical cross-linking of anionic polysaccharides (gellan gum and chondroitin sulfate) with the cationic endogen polyamine spermidine under mild conditions in order to prevent albumin denaturation. Texture and swelling properties of hydrogels with various compositions were recorded, and the effect of the preparation variables was evaluated applying neurofuzzy logic tools for hydrogels prepared with and without albumin and associating the antibiotic cloxacillin. Developed hydrogel systems were extensively analyzed by means of nuclear magnetic resonance (NMR) to determine weak-to-medium and strong binding modes and the equilibrium constants of the albumin-cloxacillin association. NMR techniques were also employed to demonstrate the successful modulation of the cloxacillin release from the albumin-containing hydrogels. In vitro microbiological tests carried out with Staphylococcus aureus and Staphylococcus epidermidis confirmed the interest of the albumin-containing hydrogels as efficient platforms for cloxacillin release in its bioactive form.

Controlled drug binding and release stand among top requirements postulated for targeted drug delivery systems of the new generations. "Smart" polymers and gels are highly suitable for the controlled delivery due to their structural sensitivity to minor environmental variations. The aim of this work was to study thermoresponsive polyanionic and polycationic hydrogels of N-isopropylacrylamide copolymers with acrylic acid and N-aminopropylmethacrylamide in terms of their interaction with two widely used drugs, propranolol and ibuprofen. Binding energetics of these drugs by the gels in swollen and collapsed state was estimated by means of high-sensitivity differential scanning calorimetry. Thermodynamic parameters of the gel collapse (transition temperature, enthalpy, heat capacity increment, and width) were determined as a dependence of the drug concentrations. From these data the excess free energy of collapse was calculated as a function of drug concentration. Deconvolution of this function resulted in the evaluation of binding parameters and contributions from interactions of various types to the free energy of binding. The binding mechanism of both drugs to the swollen and collapsed gels was elucidated. Its main features are the cooperative character of the drug binding by the collapsed gel and the predominant role of the hydrophobicity of drugs in their affinity for the swollen gel.

The unique ability of cyclodextrins (CDs) to form inclusion complexes can be transmitted to polymeric networks in which CDs are chemically grafted or cross-linked. Combination of CDs and hydrogels in a single material leads to synergic properties: the hydrophilic network enhances biocompatibility and prevents dilution in the physiological medium increasing the stability of the inclusion complexes, while CDs finely tune the mechanical features and the stimuli-responsiveness and provide affinity-based regulation of drug loading and release. Therefore, CD-functionalized materials are opening new perspectives in pharmacotherapy, emerging as advanced delivery systems (DDS) for hydrophobic and hydrophilic drugs to be administered via almost any route. Medical devices (catheters, prosthesis, vascular grafts, bone implants) can also benefit from surface grafting or thermofixation of CDs. The present review focuses on the approaches tested to synthesize nano- to macro-size covalently cross-linked CD networks: i) direct cross-linking through condensation with di- or multifunctional reagents, ii) copolymerization of CD derivatives with acrylic/vinyl monomers, and iii) grafting of CDs to preformed medical devices. Examples of the advantages of having the CDs chemically bound among themselves and to substrates are provided and their applicability in therapeutics discussed., (Copyright © 2013 Elsevier B.V. Two types of hydrophilic networks with conjugated beta-cyclodextrin (β-CD) were developed with the aim of engineering useful platforms for the localized release of an antimicrobial 5,6-dimethoxy-1-indanone N4-allyl thiosemicarbazone (TSC) in the eye and its potential application in ophthalmic diseases. Poly(2-hydroxyethyl methacrylate) soft contact lenses (SCLs) displaying β-CD, namely pHEMA-co-β-CD, and super-hydrophilic hydrogels (SHHs) of directly cross-linked hydroxypropyl-β-CD were synthesized and characterized regarding their structure (ATR/FT-IR), drug loading capacity, swelling and in vitro release in artificial lacrimal fluid. Incorporation of TSC to the networks was carried out both during polymerization (DP method) and after synthesis (PP method). The first method led to similar drug loads in all the hydrogels, with minor drug loss during the washing steps to remove unreacted monomers, while the second method evidenced the influence of structural parameters on the loading efficiency (proportion of CD units, mesh size, swelling degree). Both systems provided a controlled TSC release for at least two weeks, TSC concentrations (up to 4000μg/g dry hydrogel) being within an optimal therapeutic window for the antimicrobial ocular treatment. Prevention and management of wound infections receive a lot of attention, since the presence of micro-organisms interferes with the wound-healing process. The aim of this work was to use cyclodextrins (CDs) to endow hydrogels and gauzes with the ability to take up antiseptics and sustain their delivery for several hours. Benzalkonium chloride (BzCl) can form inclusion complexes with cross-linked CDs that regulate the release through an affinity-driven mechanism. Grafting of CDs to cotton gauzes using citric acid as the linker, at 190 °C and for 15 min, led to grafting yields of about 148%, much larger than those obtained at 180 °C or with shorter reaction times. Microbiological tests revealed that the BzCl-loaded networks can inhibit the growth of Staphylococcus epidermidis and Escherichia coli both on agar plates and in liquid medium. Furthermore, the antiseptic-loaded gauzes were able to inhibit biofilm formation by Staphylococcus aureus RN1HG pMV158GFP when applied in early stages of biofilm formation and could reduce the number of living cells in preformed biofilms grown in a chronic wound biofilm model. These findings highlight the role of CDs as main components of hydrogels and gauzes for the efficient delivery of antiseptics.

Hydrogels with high affinity for carbonic anhydrase (CA) inhibitor drugs have been designed trying to mimic the active site of the physiological metallo-enzyme receptor. Using hydroxyethyl methacrylate (HEMA) as the backbone component, zinc methacrylate, 1- or 4-vinylimidazole (1VI or 4VI), and N-hydroxyethyl acrylamide (HEAA) were combined at different ratios to reproduce in the hydrogels the cone-shaped cavity of the CA, which contains a Zn(2+) ion coordinated to three histidine residues. 4VI resembles histidine functionality better than 1VI, and, consequently, pHEMA-ZnMA(2) hydrogels bearing 4VI moieties were those with the greatest ability to host acetazolamide or ethoxzolamide (2 to 3 times greater network/water partition coefficient) and to sustain the release of these antiglaucoma drugs (50% lower release rate estimated by fitting to the square root kinetics). The use of acetazolamide as template during polymerization did not enhance the affinity of the network for the drugs. In addition to the remarkable improvement in the performance as controlled release systems, the biomimetic hydrogels were highly cytocompatible and possessed adequate oxygen permeability to be used as medicated soft contact lenses or inserts. The results obtained highlight the benefits of mimicking the structure of the physiological receptors for the design of advanced drug delivery systems.

Poly(hydroxyethyl methacrylate) (pHEMA) hydrogels were functionalized with pendant alpha-, beta- and gamma-cyclodextrins (CD) with the aim of improving the biocompatibility and increasing the ability to host drug molecules. Pendant alpha-, beta- and gamma-CDs did not affect swelling of the hydrogels but slightly decreased the water contact angle. Protein deposition was notably dependent on the nature of the CD, due to their different affinities for hydrophobic moieties of proteins. Lysozyme and albumin sorption was hindered by gamma-CD. Functionalization with beta-CD also reduced protein sorption, although less so, while alpha-CD decreased lysozyme deposition but enhanced albumin sorption compared with control pHEMA hydrogels. Loading of the hydrogels with miconazole was carried out by immersion in drug suspension followed by autoclaving. Functionalization with gamma-CD doubled the affinity of the network for the drug and resulted in the highest amount loaded (up to 170 mgg(-1)). Sustained delivery was observed for several days. The aim of this work was to develop acrylic hydrogels with high proportions of cyclodextrins maintaining the mechanical properties and the biocompatibility of the starting hydrogels, but notably improving their ability to load drugs and to control their release rate. Poly(hydroxyethylmethacrylate) hydrogels were prepared by copolymerization with glycidyl methacrylate (GMA) at various proportions and then beta-cyclodextrin (betaCD) was grafted to the network by reaction with the glycidyl groups under mild conditions. This led to networks in which the betaCDs form no part of the structural chains but they are hanging on 2-3 ether bonds through the hydroxyl groups. The pendant betaCDs did not modify the light transmittance, glass transition temperature, swelling degree, viscoelasticity, oxygen permeability, or surface contact angle of the hydrogels, but decreased their friction coefficient by 50% and improved diclofenac loading by 1300% and enhanced drug affinity 15-fold. The hydrogels were able to prevent drug leakage to a common conservation liquid for soft contact lenses (SCLs) and to sustain drug delivery in lacrimal fluid for two weeks. To summarize, the hydrogels with pendant betaCDs are particularly useful for the development of cytocompatible medicated implants or biomedical devices, such as drug-loaded SCLs.

Advanced drug delivery systems try to adjust the site and/or the rate of the release to the physiological conditions of the patient, to the progression of the illness, or to the circadian rhythms. Being different from classical pre-programmed controlled release dosage forms, the new devices aim to provide the drug release profile best for the needs of each patient. Intelligent drug delivery systems are mostly based on stimuli-responsive polymers which sense a change in a specific variable and activate the delivery; this phenomenon being reversible. This review reports on recent advances in the development of open-loop and closed-loop control systems based on stimuli-responsive polymers and their application in the drug delivery field as pulsatile and self-regulated devices. The aim of this review is to describe the most recent advances in the development of intelligent micelles and hydrogels which are sensitive to pH, specific molecules (with a mention to the molecular imprinting), temperature, irradiation or electric field, and the applications of which these mechanisms are intended.

Copolymerization of hydroxyethyl methacrylate (HEMA) with a methacrylated-derivative of beta-cyclodextrin (beta-CD) was evaluated as a way to obtain hydrogels with tunable mechanical and drug loading and release properties, particularly for preparing medicated soft contact lenses. A fully methacrylated beta-CD monomer was synthesized and added to the HEMA and cross-linker solution at concentrations ranging from 0.042 to 0.333 g ml(-1) (i.e. 0.23-1.82 mol.%). Thermal polymerization led to transparent hydrogels with a degree of conversion above 74%, which showed a high cytocompatibility and did not induce macrophage response. The greater the content in methacrylated beta-CD was, the higher the glass transition temperature, the lower the degree of swelling and free water proportion, and the greater the storage and loss moduli of the swollen disks. These findings are directly related to the increase in the degree of cross-linking caused by the methacrylated beta-CD. Loading studies were carried out with hydrocortisone and acetazolamide, both able to form complexes with CDs in water and in lacrimal fluid. Hydrocortisone loading progressively decreased as the content in methacrylated beta-CD rose due to a decrease in the volume of aqueous phase of the hydrogel. Acetazolamide loading showed a maximum for an intermediate content in beta-CD (0.125-0.167 g ml(-1)) owing to a balance between complexation with beta-CD and hydrogel mesh size. The hydrogels sustained drug delivery for several days, the acetazolamide release rate being dependent on the beta-CD content. An adequate selection of the content in beta-CD enables pHEMA-co-beta-CD hydrogels suitable for specific biomedical applications to be obtained.

Soft contact lenses are receiving an increasing attention not only for correcting mild ametropia but also as drug delivery devices. To provide poly(hydroxyethyl methacrylate), PHEMA, lenses with the ability to load norfloxacin (NRF) and to control its release, functional monomers were carefully chosen and then spatially ordered applying the molecular imprinting technology. Isothermal titration calorimetry (ITC) studies revealed that maximum binding interaction between NRF and acrylic acid (AA) occurs at a 1:1, and that the process saturates at 1:4 molar ratio. Hydrogels were synthesized using different NRF:AA molar ratios (1:2 to 1:16), at two fix AA total concentrations (100 and 200 mM), and using moulds of different thicknesses (0.4 and 0.9 mm). The cross-linker molar concentration was 1.6 times that of AA. Control (non-imprinted) hydrogels were prepared similarly but with the omission of NRF. All hydrogels showed a similar degree of swelling (55%) and, once hydrated, presented adequate optical and viscoelastic properties. After immersion in 0.025, 0.050 and 0.10 mM drug solutions, imprinted hydrogels loaded greater amounts of NRF than the non-imprinted ones. Imprinted hydrogels synthesized using NRF:AA 1:3 and 1:4 molar ratios showed the greatest ability to control the release process, sustaining it for more than 24 h. These results prove that ITC is a useful tool for the optimization of the structure of the imprinted cavities in order to obtain efficient therapeutic soft contact lenses.

The influence of the proportion of acrylamidomethyl-gamma-cyclodextrin (gamma-CD-NMA) on loading and release of the hydrophobic triamcinolone acetonide (TA) and the hydrophilic propranolol (PR) by acrylic acid hydrogels was evaluated. gamma-CD-NMA was synthesized by condensation of gamma-cyclodextrin (gamma-CD) with N-(hydroxymethyl) acrylamide. Hydrogels were prepared with gamma-CD-NMA and sodium acrylate (3 M or 4 M), using N,N'-methylen(bisacrylamide) (BIS) as cross-linker, by free radical polymerization into glass moulds of 2 mm wide and were cut as discs (10 mm diameter). gamma-CD-NMA did not modify the pH-dependent swelling of the hydrogels, but significantly increased the swelling degree in the 40:60 ethanol:water, medium in which TA can be dissolved. Hydrogels prepared with gamma-CD-NMA above 5% (w/w of total monomers) showed a remarkably higher capacity to load TA, e.g., 33 mg/g dry hydrogel versus 0.6 mg/g dry hydrogel without gamma-CD-NMA. This is explained by the formation of 1:1 inclusion complexes of TA with gamma-CD mers that overcomes the lack of interactions with the acrylic groups of the network. The release of TA in water, 0.1 N HCl, or pH 6.8 phosphate buffer was sustained for at least 24 h, whatever the pH and the composition of the medium used. In contrast, loading of PR from the water solutions was greater for hydrogels prepared with 3 M acrylate than with 4 M acrylate, irrespective to their content in gamma-CD-NMA, and in less than 2 h ca. 80% PR was released. The lower affinity of PR for the gamma-CD cavities, compared to the strong intensity of the electrostatic interactions with the acrylic acid groups, explains why the incorporation of gamma-CD-NMA did not increased the loading and control release capacity of the hydrogels of this hydrophilic drug. In summary, the copolymerisation of CD with acrylic monomers can provide highly hydrophilic pH-sensitive networks which load large amounts of hydrophobic drugs and release them in a sustained way.

Purpose: The goal of the study is to develop new hydrogels based on cyclodextrins cross-linked with ethyleneglycol diglycidylether (EGDE) under mild conditions, to be used as carriers of amphiphilic drugs. Also, it aims to characterize the cross-linking and the drug loading and release processes., Methods: The cross-linking of hydroxypropyl-beta-cyclodextrin (HPbetaCD) with EGDE, in the absence or presence of hydroxypropylmethylcellulose (HPMC) Methocel K4M, was optimized applying oscillatory rheometry and Fourier transform infrared. Hydrogels were characterized regarding swelling in water, ability to load diclofenac, and release after different drying treatments., Results: Solutions of HPbetaCD (14.28%), without or with HPMC (0.2-1.0%), provided firm and transparent hydrogels after cross-linking with EGDE (14.28%), in which around two thirds of the OH groups were cross-linked. The incorporation of HPMC progressively reduced the gel time and the swelling degree of hydrogels. HPbetaCD hydrogels efficiently loaded diclofenac and sustained the release for several hours. The presence of HPMC slowed the release from swollen hydrogels, but promoted it from hydrogels dried before the loading and also before the release., Conclusions: HPbetaCD hydrogels with good mechanical properties and tunable loading and release ability can be obtained by direct cross-linking with EGDE.

The aim of this study was to analyse the influence of the template/functional monomer proportion on the achievement of molecularly imprinted hydrogels with cavities with a high enough affinity for the drug to sustain drug release. Imprinted hydrogels were prepared from N,N-dimethylacrylamide and tris(trimethylsiloxy)sililpropyl methacrylate (DMAA and TRIS; main components), methacrylic acid (MAA; functional monomer), ethylene glycol dimethacrylate (EGDMA; cross-linker), and timolol (template drug). Photo-polymerization of the monomer solutions was carried out in poly(propylene) molds (0.3 mm thickness) to obtain contact lens-like devices. Non-imprinted control hydrogels were also prepared in the same way but without the addition of timolol. The imprinted hydrogels showed a higher affinity for timolol and a slower release rate than the non-imprinted hydrogels. The release rate decreased by increasing the MAA/timolol ratio in the gel recipe. Hydrogels prepared with 400 x 10(-3) M MAA, 600 x 10(-3) M EGDMA, and a timolol/MAA mole ratio of 1:16-1:32 had drug diffusion coefficients two orders of magnitude below those of non-imprinted hydrogels. The results obtained clearly indicate that the timolol release rate is critically affected by the conditions under which the hydrogels were synthesized. These effects are discussed on the basis of the influence of drug proportion on the conformation of the imprinted cavities.

The influence of the composition of soft contact lenses on the achievement of a significant increase in drug loading and control release capability was evaluated. Four types of timolol imprinted lenses were prepared by UV irradiation of N,N-diethylacrylamide (DEAA), 2-hydroxyethylmethacrylate (HEMA), 1-(tristrimethyl-siloxysilylpropyl)-methacrylate (SiMA) and N,N-dimethylacrylamide (DMAA) (50:50 v/v), or methylmethacrylate (MMA) and DMAA (50:50 v/v) solutions, to which functional monomer, methacrylic acid (MAA, 100 mM), cross-linker, ethyleneglycol dimethacrylate (EGDMA, 140 mM), and timolol maleate (25 mM) were previously added. Non-imprinted systems were synthesized in the same way but with the omission of timolol. Timolol did not interfere in the polymerization process and all lenses were optically clear and, once wet, showed adequate mechanical properties; nevertheless, the lenses significantly differed in temperature of glass transition (estimated by differential scanning calorimetry and oscillatory rheometry), equilibrium water content and drug loading and release properties. Regarding timolol overall affinity (SK), estimated after fitting the Langmuir equation to the adsorption isotherms obtained in water at 37 degrees C, the lenses ranked in the order HEMA>SiMA-DMAA>MMA-DMAA>DEAA. The highest imprinting effect, i.e. the greatest relative increase in overall affinity (SK) respect to non-imprinted systems, was obtained for the last two systems. These results are explained by interaction capability with timolol, conformational features and hydrophilic character of the lenses. Although all lenses (imprinted and non-imprinted) showed sustained release in 0.9% NaCl solution, the values of diffusion coefficients confirmed that timolol molecules move out easily from hydrophilic networks that present low affinity for the drug; i.e. MMA-DMAA and SiMA-DMAA lenses. The results obtained indicate that modulating the composition of the lenses it is possible to adapt the drug loading and release behavior of the lenses to the treatment requirements of specific pathological processes.

The cross-linking process of two cationic hydroxyethylcelluloses of different hydroxyethyl and ammonium group contents, polyquaternium-4 (PQ-4) and polyquaternium-10 (PQ-10), with ethylenglycol diglycidylether (EGDE) was characterized and optimized through rheometric analysis of the forming network. The influence of NaOH concentration, temperature, and EGDE concentration on the cross-linking rate were studied. The evolution of the elastic (G') and viscous (G") moduli, recorded in time-sweep experiments carried out at a fixed angular frequency, showed that the cross-linker requires a minimum of 0.05 M NaOH and 30 degrees C to be active. The increase in G' and G" followed first order kinetics, the slopes of G' being higher than those corresponding to G". The gel time, i.e. the time at which the crossover of G' and G" occurs, decreases exponentially when temperature increases from 30 to 60 degrees C. Apparent activation energies, estimated from the gel times, ranged between 70 and 90 kJ/mol. The cross-linking rate was greater in PQ-4 than in PQ-10 owing to the initial lower viscosity and higher content in hydroxyethyl groups of the former. However, IR spectra of the final hydrogels suggest the formation of a similar number of cross-linking junctions in both polymer systems. The optimum conditions for hydrogel preparation were 60 degrees C in 0.10 M NaOH medium, and no depolymerization was observed. Such hydrogels were transparent, presented a smooth, continuous surface, and were superabsorbent in water. After drying in an oven, the degree of swelling was lower than that of freshly prepared hydrogels; the behavior of water uptake being Fickian. The hydrogels presented a significant loading capacity of diclofenac sodium, with which they interact through ionic and hydrophobic bonding. The affinity is kept at an acidic pH, preventing drug release. In contrast, at pH 8 the interactions are broken and the release process is sustained for more than 4 h. The results also indicate that the ionic strength as well as the initial pH of the medium, when the release was evaluated switching the pH from acidic to basic, are two critical factors which have to be considered to extract conclusions about the behavior of the hydrogels as site-specific delivery systems under in vivo conditions.

Here, we present the synthesis of a series of chemical homopolymeric and copolymeric injectable hydrogels based on polyethylene glycol methyl ether methacrylate (PEGMEM) alone or with 2-dimethylamino ethyl methacrylate (DMAEM). The objective of this study was to investigate how the modification of hydrogel components influences the swelling, rheological attributes, and in vitro biocompatibility of the hydrogels. The hydrogels' networks were formed via free radical polymerization, as assured by 1H nuclear magnetic resonance spectroscopy (1H NMR). The swelling of the hydrogels directly correlated with the monomer and the catalyst amounts, in addition to the molecular weight of the monomer. Rheological analysis revealed that most of the synthesized hydrogels had viscoelastic and shear-thinning properties. The storage modulus and the viscosity increased by increasing the monomer and the crosslinker fraction but decreased by increasing the catalyst. MTT analysis showed no potential toxicity of the homopolymeric hydrogels, whereas the copolymeric hydrogels were toxic only at high DMEAM concentrations. The crosslinker polyethylene glycol dimethacrylate (PEGDMA) induced inflammation in ATDC5 cells, as detected by the significant increase in nitric oxide synthase type II activity. This work describes for first time how anisotropic gold nanoparticles (AuNPs) can be spontaneously formed inside preformed contact lenses (CLs) avoiding the use of additional reductant agents (reagent-free) through a precise tunning of the monomeric composition, the saline concentration, and the application of steam heat sterilization. Protocols to generate AuNPs in solution using inorganic or small organic reductants are widely available. Differently, gold precursors interactions with polymer networks have been overlooked and, thus, the interest of chemically cross-linked hydrogels as organic reductants is still to be elucidated. In the ocular field, incorporation of AuNPs to CLs may expand their applications in prophylaxis, therapy and diagnosis. To carry out the work, a variety of hydrogels and commercially available CLs were incubated with gold salt solution without any other chemical reagent. AuNPs formation was monitored by changes in localized surface plasmon resonance (LSPR) bands and quantifying the gold sorbed. Only silicone hydrogels induced AuNPs formation at room temperature in few days; methacrylic acid red-shifted the LSPR band (550–600 nm), while monomers bearing F hindered the reduction. Storage of hydrogels in the gold precursor solution allowed a gradual formation of anisotropic AuNPs, which could be stopped at any time by washing the hydrogel with water. The developed CLs behave as efficient filters against highly penetrant light and also exhibit photoresponsiveness as demonstrated as rapid (10 s), focused mild hyperthermia when irradiated with green, red and NIR lasers. Background: Age‐related macular degeneration (AMD) is a potentially blinding posterior segment disease; inflammatory responses and subretinal drusen formation lead to leaky blood vessels. The current treatment method involves intravitreal injections of anti‐VEGF agents, which is highly invasive and requires frequent injections administered by trained personnel. Periocular injections such as trans−/intra‐scleral injections would provide a minimally‐invasive treatment option. The sclera is the outermost protective layer occupying 5/6th of the ocular globe. Owing to its avascular nature and self‐healing ability, sclera could be the potential space for the delivery of depot forming long‐acting formulations. This project focuses on the development of chitosan grafted poly(n‐isopropylacrylamide) (Cs‐g‐PNIPAAm) gel for the sustained intrascleral delivery of small molecular weight, multiple tyrosine kinase inhibitor Sunitinib malate. Methods: Cs‐g‐PNIPAAm hydrogel was prepared using free radical polymerization with varying concentrations of chitosan (varying with 10%, 30%, 50% weight percentages) with respect to PNIPAAm. The hydrogels were characterized for rheology, LCST measurements, swelling studies, degradation, syringeability, drug release and permeation using Franz diffusion studies. Biocompatibility study of hydrogel was performed with ARPE‐19 cells, ocular irritation using HET‐CAM test, Further, choroidal angiogenesis was tested on CAM assay and rat choroidal exoplants. Results: Chitosan grafting was found to have effect on rheological properties of hydrogel and hence on syringeability of formulations. However, chitosan grafting did not significantly affect the LCST of hydrogels, all the formulations exhibited LCST of 32 ± 0.5°C. 20 ul of 30% Cs‐g‐PNIPAAm hydrogel was able to release approximately 10 ug/day sunitinib concentration in‐vitro for 28 days. It was observed that the drug release from the hydrogel was controlled by both the diffusion and erosion mechanism. Further, the ex‐vivo permeation studies on porcine sclera showed that up to 40% sustained release of sunitinib was obtained from hydrogel compared to sunitinib solution (2 mg/ml). The optimized formulation (F8) was found to be biocompatible on ARPE‐19 cells and no ocular irritation was observed on HET‐CAM (Hen's egg test‐ Choriallantoic membrane) assay. Further, the anti‐angiogenic efficacy was conformed using CAM assay and rat‐choroidal angiogenesis assay. Wherein F8 hydrogel was found to prevent formation of new blood capillaries compared to control medium. Conclusions: The F8 hydrogel was found to be injectable using ultra‐thin walled 27G needles. OCT micrographs shows that the F8 hydrogel is able to form depot on‐intrascleral injections. Further, dual control over the drug release i.e. temperature controlled gelation and ionic conjugation of sunitinib with amine group of chitosan gives better control over drug release. Platelet lysate (PL) was encapsulated in collagen (Coll) millimetric gel beads, on biomimetic superhydrophobic surfaces, under mild conditions, with the aim of obtaining easy-to-handle formulations able to provide sustained release of multiple growth factors for skin ulcers treatment. The gel particles were prepared with various concentrations of PL incorporating or not stem cells, and tested as freshly prepared or after being freeze-dried or cryopreserved. Coll + PL particles were evaluated regarding degradation in collagenase-rich environment (simulating the aggressive environment of the chronic ulcers), sustained release of total protein, PDGF-BB and VEGF, cell proliferation (using particles as the only source of growth factors), scratch wound recovery and angiogenic capability. Compared to Coll solely particles, incorporation of PL notably enhanced cell proliferation (inside and outside gels) and favored scratch wound recovery and angiogenesis. Moreover, cell-laden gel particles containing PL notably improved cell proliferation and even migration of cells from one particle towards a neighbor one, which led to cell-cell contacts and the spontaneous formation of tissue layers in which the spherical gels were interconnected by the stem cells. Soft contact lenses (SCLs) constitute a promising vehicle for ocular drug delivery due to their biocompatibility, prolonged contact with the eye and general acceptance. Soaking in the drug solution is the simplest method to load the drug into the SCLs. However, it usually does not ensure a controlled drug release, compatible with the therapeutic needs. Thus, additional approaches, such as the application of coatings on the SCLs that constitute a barrier to the drug release, have been tried to achieve that purpose [1]. The main goal of this work is to investigate the possibility of using a layer-by-layer (LbL) coating strategy, with alginate, chitosan and hyaluronic acid (ALG/CHI/HA) to get a controlled release of diclofenac from silicon based hydrogels. A lab-made silicon-based hydrogel intended for SCLs was loaded by soaking with the anti-inflammatory (diclofenac, DCF) and coated layer-by-layer (LbL) by immersion in solutions of alginate, chitosan and hyaluronic acid. Material properties such as transmittance, wettability, ionic permeability and swelling were studied. Drug release experiments were carried out under sink conditions (3 mL NaCl aqueous solution 130 mM, 36 °C, 180 rpm stirring). The coating stability and lysozyme-coating interaction was evaluated by quartz crystal microbalance with dissipation (QCM-D). A mathematical model was applied to predict the in vivo efficacy of the coated lenses. Chorioallantoic membrane (HET-CAM) tests were carried out to predict potential ocular irritation. The coating did not impair the studied physico-chemical properties, relevant for the application of the material in SCLs. HET-CAM tests did not suggest any potential for ocular irritation of both uncoated and coated samples. QCM-D data revealed the stability of the deposited layers and no adsorption of the protein. DCF release kinetics was controlled by the presence of the coating. The DCF concentration profile in the tear fluid estimated from the mathematical model predicts values above the half maximal inhibitory concentrations (IC50) for cyclooxygenase-1 (COX-1) and cyclooxygenase-2 (COX-2) during more than 2 weeks, for the coated hydrogels, while for the uncoated such levels are only expected in the first day of release. ALG/CHI/HA LBL coated silicon hydrogels present adequate properties to be used in DCF releasing SCLs. They reveal an antifouling behaviour against lysozyme, one of the most abundant protein in lacrimal fluid. Isothermal titration calorimetry (ITC) was used to identify the optimal timolol:functional monomer ratio for preparing soft contact lenses (SCLs) able to sustain drug release. ITC profiles revealed that each timolol molecule required six to eight acrylic acid (AAc) monomers to saturate the binding and that these ratios could be the most suitable for creating imprinted cavities. Various poly(hydroxyethyl methacrylate- co-AAc) hydrogels 0.2 and 0.9 mm thick were prepared with timolol:AAc molar ratios ranging from 1 : 6 to 1 : 32 and also in the absence of timolol. The hydrogels were reloaded with timolol by immersion in 0.04, 0.06, 0.08, and 0.10 m M drug solutions. Both imprinted and nonimprinted hydrogels showed a high affinity for the drug because of the presence of AAc. Nevertheless, the 1 : 6 and 1 : 8 imprinted hydrogels loaded less timolol but sustained the release better than the other hydrogels. These differences were explained in terms of the different arrangement of the functional monomers along the network. The imprinting effect was more noticeable in the case of the thinnest hydrogels, where the contribution of the diffusion path to the release rate was smaller. Abstract: The aim of this work was to develop acrylic hydrogels with high proportions of cyclodextrins maintaining the mechanical properties and the biocompatibility of the starting hydrogels, but notably improving their ability to load drugs and to control their release rate. Poly(hydroxyethylmethacrylate) hydrogels were prepared by copolymerization with glycidyl methacrylate (GMA) at various proportions and then β-cyclodextrin (βCD) was grafted to the network by reaction with the glycidyl groups under mild conditions. This led to networks in which the βCDs form no part of the structural chains but they are hanging on 2–3 ether bonds through the hydroxyl groups. The pendant βCDs did not modify the light transmittance, glass transition temperature, swelling degree, viscoelasticity, oxygen permeability, or surface contact angle of the hydrogels, but decreased their friction coefficient by 50% and improved diclofenac loading by 1300% and enhanced drug affinity 15-fold. The hydrogels were able to prevent drug leakage to a common conservation liquid for soft contact lenses (SCLs) and to sustain drug delivery in lacrimal fluid for two weeks. To summarize, the hydrogels with pendant βCDs are particularly useful for the development of cytocompatible medicated implants or biomedical devices, such as drug-loaded SCLs. Abstract: Copolymerization of hydroxyethyl methacrylate (HEMA) with a methacrylated-derivative of β-cyclodextrin (β-CD) was evaluated as a way to obtain hydrogels with tunable mechanical and drug loading and release properties, particularly for preparing medicated soft contact lenses. A fully methacrylated β-CD monomer was synthesized and added to the HEMA and cross-linker solution at concentrations ranging from 0.042 to 0.333gml−1 (i.e. 0.23–1.82mol.%). Thermal polymerization led to transparent hydrogels with a degree of conversion above 74%, which showed a high cytocompatibility and did not induce macrophage response. The greater the content in methacrylated β-CD was, the higher the glass transition temperature, the lower the degree of swelling and free water proportion, and the greater the storage and loss moduli of the swollen disks. These findings are directly related to the increase in the degree of cross-linking caused by the methacrylated β-CD. The optimum conditions for hydrogel preparation were 60 °C in 0.10 M NaOH medium, and no depolymerization was observed. Such hydrogels were transparent, presented a smooth, continuous surface, and were superabsorbent in water. After drying in an oven, the degree of swelling was lower than that of freshly prepared hydrogels; the behavior of water uptake being Fickian. The hydrogels presented a significant loading capacity of diclofenac sodium, with which they interact through ionic and hydrophobic bonding. The affinity is kept at an acidic pH, preventing drug release. In contrast, at pH 8 the interactions are broken and the release process is sustained for more than 4 h. The results also indicate that the ionic strength as well as the initial pH of the medium, when the release was evaluated switching the pH from acidic to basic, are two critical factors which have to be considered to extract conclusions about the behavior of the hydrogels as site-specific delivery systems under in vivo conditions. The aim of this work was to evaluate the utility of molecular imprinting for improving the loading capability of weakly cross-linked hydrogels, with a view to their use as soft contact lenses for administration of timolol. Imprinted hydrogels were prepared considering preformation of complexes between methacrylic acid (functional monomers) and timolol (target molecules), and polymerization with N,N-diethylacrylamide and ethylenglycol dimethacrylate (EGDMA, cross-linker) after injection in moulds (0.3 mm thickness) and UV irradiation at room temperature. After polymerization, timolol molecules were removed by washing. When the gels were immersed in timolol solutions, the imprinted gels adsorbed much more timolol than the corresponding non-imprinted ones, especially above 60 mM EGDMA. The minimum cross-linker concentration for the imprinting to be effective was 80 mM EGDMA. The imprinted contact lenses improved significantly the affinity of the hydrogels for timolol, estimated after fitting Langmuir equation to the adsorption isotherms. These results indicated that adsorption sites capable of capturing the target molecules were encoded effectively into the polymer network by the molecular imprinting technique and, in consequence, improved the drug loading capacity of the gels. Thermo- and pH-sensitive hydrogels were synthesized using N-isopropylacrylamide (NIPA) and N-aminopropylmethacrylamide, cross-linked with N,N′-methylenebis(acrylamide). The dependence of the degree of swelling on the cross-linking density was analyzed according to the Flory-Huggins theory and a master curve obtained. To optimize the efficiency of these hydrogels in controlled release, we studied the loading and release of a divalent molecule (naphthalenedisulfonic acid, NS-2) in media of different ionic strengths and pH. The uptake process followed the Langmuir adsorption isotherm model. The highest loading occurred when the amino groups in the gel were protonated (acidic pH) and could come close each other to form a binding site for the two sulfonic groups of NS-2, i.e. low degree of cross-linking and collapsed state. Below the phase transition temperature (33 °C), NS-2 loaded hydrogels quickly released a significant amount of adsorbate until a new equilibrium between free NS-2 and adsorbed NS-2 was achieved. Above that temperature, hydrogels not only stopped the release but were even able to take free NS-2 up again from the medium, showing that the loading/release process was reversible and reproducible after several temperature cycles. At 37 °C, the release rate was independent of the degree of cross-linking (NIPA caused the hydrogel to collapse), but was strongly affected by the pH and salt concentrations of the medium, which condition the strength of the interaction between the hydrogel amino groups and the NS-2 sulfonic groups. In an acidic medium, the protonated amino groups bind NS-2 strongly and the amount released is small. In contrast, at pH 7.4 or in the presence of a high salt concentration, the hydrogel loses its affinity for NS-2 and the release rate increases, giving pH- or salt-sensitive delivery systems. Statins are receiving increasing attention in the ophthalmic field. Their activity as 3-hydroxy-3-methylglutaryl–CoA (HMG–CoA) reductase inhibitors is clinically used to regulate cholesterol levels and leads to pleiotropic effects, which may help in the management of diabetes-related ocular pathologies. This work aims to design bioinspired contact lenses (CLs) with an affinity for atorvastatin by mimicking the active site of HMG–CoA reductase. Sets of imprinted and nonimprinted 2-hydroxyethyl methacrylate (HEMA) hydrogels were synthesized, varying the contents in functional monomers that bear chemical groups that resemble those present in HMG–CoA reductase, namely, ethylene glycol phenyl ether methacrylate (EGPEM), 2-aminoethyl methacrylate hydrochloride (AEMA), and N-(3-aminopropyl) methacrylamide hydrochloride (APMA). The hydrogels were characterized in terms of suitability as CLs (solvent uptake, light transmission, mechanical properties, and biocompatibility) and capability to load and release atorvastatin. Three sterilization protocols (steam heat, gamma radiation, and high hydrostatic pressure) were implemented and their effects on hydrogel properties were evaluated. Copolymerization of AEMA and, particularly, APMA endowed the hydrogels with a high affinity for atorvastatin (up to 11 mg/g; KN/W > 200). Only high hydrostatic pressure sterilization preserved atorvastatin stability and hydrogel performance. Permeability studies through the porcine cornea and sclera tissues revealed that the amount of atorvastatin accumulated in the cornea and sclera could be effective to treat ocular surface diseases. Contact lenses (CLs) are prone to biofilm formation, which may cause severe ocular infections. Since the use of antibiotics is associated with resistance concerns, here, two alternative strategies were evaluated to endow CLs with antibiofilm features: copolymerization with the antifouling monomer 2-methacryloyloxyethyl phosphorylcholine (MPC) and loading of the antioxidant resveratrol with known antibacterial activity. MPC has, so far, been used to increase water retention on the CL surface (Proclear® 1 day CLs). Both poly(hydroxyethyl methacrylate) (HEMA) and silicone hydrogels were prepared with MPC covering a wide range of concentrations (from 0 to 101 mM). All hydrogels showed physical properties adequate for CLs and successfully passed the hen's egg-chorioallantoic membrane (HET-CAM) test. Silicone hydrogels had stronger affinity for resveratrol, with higher loading and a slower release rate. Ex vivo cornea and sclera permeability tests revealed that resveratrol released from the hydrogels readily accumulated in both tissues but did not cross through. The antibiofilm tests against Pseudomonas aeruginosa and Staphylococcus aureus evidenced that, in general, resveratrol decreased biofilm formation, which correlated with its concentration-dependent antibacterial capability. Preferential adsorption of lysozyme, compared to albumin, might also contribute to the antimicrobial activity. In addition, importantly, the loading of resveratrol in the hydrogels preserved the antioxidant activity, even against photodegradation. Overall, the designed hydrogels can host therapeutically relevant amounts of resveratrol to be sustainedly released on the eye, providing antibiofilm and antioxidant performance. The aim of this work was to develop drug-soft contact lens combination products suitable for controlled release of antimicrobial peptides on the ocular surface. Incorporation of functional monomers and the application of molecular imprinting techniques were explored to endow 2-hydroxyethyl methacrylate ( HEMA) hydrogels with the ability to load and to sustain the release of polymyxin B and vancomycin. Various HEMA-drug-functional monomer-cross-linker molar ratios were evaluated to prepare polymyxin B imprinted and non-imprinted hydrogels. Acrylic acid-functionalized and imprinted hydrogels loaded greater amounts of polymyxin B and led to more sustained release profiles, in comparison with non-functionalized and non-imprinted networks. Polymyxin B-loaded hydrogels showed good biocompatibility in hen's egg test-chorioallantoic membrane tests. Functionalized hydrogels also loaded vancomycin and sustained its release, but the imprinting effect was only exhibited with polymyxin B, as demonstrated in rebinding tests. Contact lenses (CLs) constitute an advantageous platform for the topical release of corticosteroids due to their prolonged contact with the eye. However, the lipophilic nature of corticosteroids hampers CLs' ability to release therapeutic amounts. Two approaches to improve loading and release of triamcinolone acetonide (TA) from poly(2-hydroxyethyl methacrylate)-based hydrogels were investigated: adding 2-hydroxypropyl-β-cyclodextrin (HP-β-CD) to the monomers solution before polymerization (HEMA/i-CD) and an hydrogels' post-treatment with HP-β-CD (HEMA/p-CD). The effect of HP-β-CD and sterilization by high hydrostatic pressure (HHP) on the hydrogel properties (water content, oxygen and ion permeability, roughness, transmittance, and stiffness) was evaluated. The HEMA/i-CD hydrogels had stronger affinity for TA, sustaining its release for one day. HHP sterilization promoted the formation of cyclodextrin-TA complexes within the hydrogels, improving their drug-loading capacity »60 %. Cytotoxicity and irritability tests confirmed the safety of the therapeutic CLs. TA released from the hydrogels permeated through ocular tissues ex vivo and showed anti-inflammatory activity. Finally, a previously validated mathematical model was used to estimate the ability of the TA-loaded CLs to deliver therapeutic drug concentrations to the posterior part of the eye. Overall, HP-β-CD-containing CLs are promising candidates for the topical ocular application of TA as an alternative delivery system to intraocular injections. Abstract: An in situ photopolymerization-coating technique was applied to wrap the pellets surface with a pH-sensitive hydrogel layer made from acrylic acid and hydrophobic acrylate monomers. Powdered cellulose (Elcema® P100) and poly(vinylpyrrolidone) (Kollidon® 30) pellets containing theophylline were prepared by extrusion–spheronization, sprayed with an ethanol:water 50:50v/v solution of the monomers, the cross-linker (N,N′-methylenebis(acrylamide)) and the initiator (Irgacure® 2959), and immediately irradiated at 366nm. The composition of coating mixture and the time of irradiation were optimized using oscillatory rheometry and analyzing the swelling and the drug release behaviour of the resultant hydrogels. When acrylic acid:lauryl acrylate 88:12 molar ratio was used, the coating did not significantly change the shape, size, or friability of the pellets, but remarkably modified theophylline release profiles. Poly(hydroxyethyl methacrylate), pHEMA, hydrogels are widely used for preparing implants, contact lenses, and other biomedical devices, which in many circumstances should load drugs to deliver them in the adjacent tissues. To enhance the potential of pHEMA hydrogels as nonsteroidal anti-inflammatory drugs (NSAIDs) delivery systems, 4-vinyl-pyridine (VP) and N-(3-aminopropyl) methacrylamide (APMA) were incorporated to the network (25–150 mM). The incorporated monomers did not change the viscoelastic properties neither the state of water, but remarkably increased the amount of ibuprofen (up to 10-fold) and diclofenac (up to 20-fold) loaded. Dried loaded pHEMA-APMA and pHEMA-VP hydrogels quickly swelled in water but ionic/hydrophobic interactions prevented the amount of drug released to be above 10%. By contrast, once the water-swollen hydrogels were transferred to pH 5.8 or 8.0 phosphate buffers or NaCl solutions, the release was prompted by competition with ions of the medium. The remaining of hydrophobic interactions and the high polymeric density of the pHEMA hydrogels contributed to sustain the release process for at least 24 h for ibuprofen and almost 1 week for diclofenac. Local treatment of bladder cancer faces several limitations such as short residence time or low permeation through urothelium tissue. The aim of this work was to develop patient-friendly mucoadhesive gel formulations combining gemcitabine and the enzyme papain for improved intravesical chemotherapy delivery. Hydrogels based on two different polysaccharides, gellan gum and sodium carboxymethylcellulose (CMC), were prepared with either native papain or papain nanoparticles (nanopapain) to explore for the first time their use as permeability enhancers through bladder tissue. Gel formulations were characterized regarding enzyme stability, rheological behavior, retention on bladder tissue and bioadhesion, drug release properties, permeation capacity, and biocompatibility. After 90 days of storage, the enzyme loaded in the CMC gels retained up to 83.5 ± 4.9 % of its activity in the absence of the drug, and up to 78.1 ± 5.3 with gemcitabine. The gels were mucoadhesive and the enzyme papain showed mucolytic action, which resulted in resistance against washing off from the urothelium and enhanced permeability of gemcitabine in the ex vivo tissue diffusion tests. Native papain shortened lag-time tissue penetration to 0.6 h and enhanced 2-fold drug permeability All formulations demonstrated pseudoplastic behavior and no irritability. Abstract: The relationship between the microstructure of poly(2-hydroxyethyl methacrylate) (polyHEMA) hydrogels and the growth of microorganisms on their surface and their penetration through the network was evaluated. The hydrogels were prepared from solutions of HEMA and ethyleneglycol dimethacrylate (EGDMA) with water ranging from 0 to 60%wt. As the content in water upon synthesis increased the glass transition temperature and the degree of swelling of the hydrogels increased. Image analysis of SEM micrographs of the hydrogel surface and BET analysis of nitrogen adsorption isotherms were respectively used to characterize the size distributions of macropores and mesopores. The higher the water proportion in the monomers solution, the greater the size of meso (linear dependence) and macropores (segmental mode dependence), the area occupied by the macropores at the surface of the hydrogels, and the specific surface of the hydrogels. Hydrogels synthesized with 30% or less water showed a quite close distribution of pores with a mean size below 1μm and effectively hindered the penetration of Gram+ Staphylococcus aureus (0.5–1μm cocci) and Gram−Pseudomonas aeruginosa (0.5–0.8μm×1.5–3μm bacilli). Logistic regression analysis points out that the probability of penetration of Staphylococcus aureus or Pseudomonas aeruginosa is above 50% when the mean pore size is greater than 3.67μm or 5.56μm, respectively. Abstract: Cross-linking of hydroxypropyl-β-cyclodextrin (HPβCD) with ethyleneglycol diglycidylether (EGDE) in carbopol dispersions enabled the synthesis of cyclodextrin hydrogels with domains of interpenetrating acrylic microgels (micro-scale-IPNs) in a single step under mild conditions. As carbopol proportion increased, the hardness and compressibility of the ms-IPNs decreased, but their bioadhesion force and pH-responsiveness rose. Control HPβCD hydrogel and ms-IPNs were loaded with estradiol and ketoconazole by immersion in drug suspensions, some of which were autoclaved to enhance (up to a 50%) drug/cyclodextrin affinity. ms-IPNs prepared with 0.8% or 1.0% carbopol showed the highest loading due to their greater swelling degree and, consequently, mesh size. The total loading of the ms-IPNs greatly exceeded (up to 200-fold) the amount dissolved in their aqueous phase, which highlights the main role of drug complexation with the cross-linked cyclodextrins. The affinity of the drug for HPβCD sustained the release for several days; the rate being also dependent on carbopol content and on pH of the medium. Therefore, an adequate design of the HPβCD/carbopol ms-IPNs provides a single material with tunable mechanical properties, in which the complexation ability of cyclodextrins is combined with the bioadhesive and pH-responsive properties of carbopol. The volume phase transition of a hydrogel of the polyampholyte network N -isopropylacrylamide (6000) - N -(3-aminopropylmethacrylamide) (120) - Acrylic acid (120) - N , N′ -Methylenebisacrylamide (40) was investigated by high-sensitivity differential scanning calorimetry in relation to pH, concentrations of NaCl and two oppositely charged drug ligands (ibuprofen and propranolol). The transition temperature, enthalpy, heat capacity increment and width against these thermodynamic variables were determined. The experimental data were used for calculation of the excess free energy functions of the phase transition which are measures of the relative stability of the dense (collapsed) state of the polyampholyte network under different conditions. By analogy with globular proteins, the stability of the network dense state is maximal at the isoelectric point of the polyampholyte and is increased by the kosmotropic salt and specific ligands. We demonstrate that the polyampholyte hydrogel consists of independent cooperative domains. The sizes of these domains in the hydrogel (9–15 kDa) are comparable with those in globular proteins.
