Maleylacetate reductase (EC 1.3.1.32) plays a major role in the degradation of chloroaromatic compounds by channelling maleylacetate and some chlorinated derivatives into the 3-oxoadipate pathway. Several substituted maleylacetates were prepared in situ by alkaline or enzymatic hydrolysis of dienelactones as the precursor. The conversion of these methyl-, chloro-, fluoro-, and bromo-substituted maleylacetates by malelacetate reductase from 3-chlorobenzoate-grown cells of Pseudomonas sp. strain B13 was studied. Two moles of NADH per mole of substrate was consumed for the conversion of maleylacetates which contain a halogen substituent in the 2 position. In contrast, only 1 mol of NADH was necessary to convert 1 mol of substrates without a halogen substituent in the 2 position. The conversion of 2-fluoro-, 2-chloro-, 2,3-dichloro-, 2,5-dichloro-, 2,3,5-trichloro-, 2-bromo-, 2,3-dibromo-, 2,5-dibromo-, 2-bromo-5-chloro-, 2-chloro-3-methyl-, and 2-chloro-5-methylmaleylacetate was accompanied by the elimination of halide from the 2 position and the temporary occurrence of the corresponding dehalogenated maleylacetate as an intermediate consuming the second mole equivalent of NADH. The properties of the halogen substituents influenced the affinity to the enzyme in the following manner. Km values increased with increasing van der Waals radii and with decreasing electronegativity of the halogen substituents (i.e., low steric hindrance and high electronegativity positively influenced the binding). The Km values obtained with 2-methyl-,3-methyl-, and 5-methylmaleylacetate showed that a methyl substituent negatively affected the affinity in the following order: 2 position >/ = 3 position >> 5 position. A reaction mechanism explaining the exclusive elimination of halogen substituents from the 2 position is proposed.

Dioxygenation of substituted benzoic acids by whole cells of 3-chlorobenzoate-utilizing Pseudomonas sp. B 13, benzoate-induced cells of Alcaligenes eutrophus B 9 and toluate-grown cells of Pseudomonas putida mt-2 was examined. Electron-attracting substituents like halogen decreased the reaction rates of benzoate 1,2-dioxygenation. Dioxygenation of substituted benzoic acids by P. putida mt-2 was mostly undisturbed by steric effects of the substituents. Good correlation resulted between the log Vrel values and the Hammett substituent constant sigma. In contrast the reaction rates of dioxygenation by Pseudomonas sp. B 13 and A. eutrophus were decreased predominantly by steric effects of substituents. A non-polar reaction mechanism of benzoate 1,2-dioxygenation is discussed. Results from inhibition studies demonstrate high stereospecificities for the 1,2-dioxygenation by Pseudomonas sp. B 13 of benzoic acids with substituents in ortho- or para-position. In the case of P. putida mt-2 steric handrance by substituents was observed only with orth-substituted benzoic acids. Stereospecificities of the benzoate 1,2-dioxygenation by Pseudomonas sp. B 13 and P. putida mt-2 are illustrated schematically.

DNA fragments containing the xylD and xylL genes of TOL plasmid pWW0 -161 of Pseudomonas putida, which code for the catabolic enzymes toluate 1,2-dioxygenase and dihydrodihydroxybenzoic acid dehydrogenase, respectively, and the nahG gene of the NAH plasmid NAH7 , which codes for salicylate hydroxylase, were cloned in pBR322 vector plasmid. Deletion and insertion mutagenesis were used to localize these genes with respect to crucial endonuclease cleavage sites. The pBR322-based plasmids were ligated to the broad host range cloning vector pKT231 , or derivatives of it, and the hybrid plasmids were introduced into Pseudomonas sp. B13( WR1 ), a bacterium able to degrade 3-chlorobenzoate but not 4-chlorobenzoate, 3,5- dichlorobenzoate , salicylate, or chlorosalicylates . The cloned xylD gene expanded the catabolic range of WR1 to include 4-chlorobenzoate, whereas the cloned xylD - xylL genes enabled the isolation of derivatives of WR1 that degraded 3-chlorobenzoate, 4-chlorobenzoate, and 3,5- dichlorobenzoate . The cloned nahG gene extended the catabolic range of WR1 to include salicylate and 3-, 4-, and 5- chlorosalicylate .

Aromatic compounds with unphysiological substituents like halogen or SO2H-groups are mainly degraded by cometabolism. Investigations with model compounds show, that the negative inductive effect (-I-effect) of the halogen substituents impede the electrophilic attack of the oxygenases, particularly the pyrocatechases. Although the "affinity" of the enzyme for the substrate increases with the number of halogen substituents, the rate of ring cleavage decreases by the presence of halogen substituents. Benzoate oxygenation shows that in certain positions of substitution the -I-effect can be weakened by the + M-effect of the halogen. As soon as aromaticity is lost by the action of dioxygenases, halide can be eliminated with greater ease and total mineralization is possible. When naphthalene-2-sulfonic acid is degraded by a naphthalene utilizing Pseudomonas strain the sulfonic acid group appears to be eliminated oxygenolytically and the carbon skeleton is channelled into the naphthalene pathway.

Biodegradation of most naturally occurring compounds is relatively fast, since microorganisms have evolved appropriate enzyme systems. However, biodegradation is less likely in the case of man-made compounds like haloaromatics, which have structural features rarely or never encountered in natural products. One strategy to develop new metabolic activities for novel compounds by strains of microorganisms is by the alteration of their existing genetic information. The present paper summarizes results of studies where the degradation capabilities of bacteria were expanded by genetic material from external sources either by natural exchange or by cloning procedures.
