Some insect species have been proposed as a sustainable alternative to traditional animal‐based food and feed sources. Optimisation of insect production can generally be achieved using two main approaches: optimising environmental conditions and improving traits of interest through selective breeding. These avenues are not inseparable as performance of a genotype might differ between environments due to phenotypic plasticity and because genotypes can respond differently to environmental changes, that is genotype‐by‐environment interactions (G × E). In insect production, diets can be of variable quality and consist of waste‐ and by‐products of low nutritional value, which might result in decreased performance and/or increased trait variability within a population. Therefore, it is of interest to investigate how genotypes perform across various diets. Here, we investigated plasticity and G × E for mean performance and trait variation, which we define as the ability of a genotype to produce consistent phenotypes within and across environments. We did this by rearing 190 full‐sib families of house fly larvae, Musca domestica L. (Diptera: Muscidae), on two diets based on either alfalfa, Medicago sativa L. (Fabales: Fabaceae), or deproteinated grass. Four larval traits were assessed: egg‐to‐larva viability, surface area, dry weight and relative lipid content. Reaction norms were used to investigate the effects of diet on full‐sib family mean and trait variation within and across environments. We found that families reared on the grass‐based diet had higher performance across all investigated traits than families reared on the alfalfa‐based diet. For both mean performance and trait variation, we found G × E for all investigated traits. These findings suggest that there is genetic variation for plasticity (slope of reaction norms) for both trait mean and trait variation, and thus that there is a potential to breed for genotypes with high performance as well as for genotypes with low trait variation within and across diets. Predation exerts a profound influence on the evolution of camouflage and detection abilities in both predators and prey. For instance, flower‐visiting spiders need to ensure their concealment when ambushing on flowers, which compels pollinators to check any spider cues cautiously before landing. Although numerous studies have examined the visual camouflage of spiders deceiving bees, little is known regarding whether spiders also employ olfactory camouflage. Additionally, the detection of spiders by dipteran insects is often overlooked, despite them being major non‐bee pollinators. Here, we explored the detection ability of dipteran pollinators and the crypsis skill of spiders from both olfactory and vision perspectives using the housefly Musca domestica L. (Diptera: Muscidae) as prey, the crab spider Ebrechtella tricuspidata (Fabricius) (Araneae: Thomisidae) as predators and chamomile Matricaria recutita L. (Asteraceae) as substrates. Our olfactory experiments revealed that experienced houseflies could respond to and avoid spider odour. However, this response did not appear to be innate, as naive individuals showed no aversion. Moreover, experienced houseflies did not have any avoidance behaviours to the mixed odours of spider and flower, indicating that spiders achieved olfactory crypsis utilizing floral scent. Our vision experiments and visual modelling demonstrated that houseflies could detect spiders positioned on flower. Surprisingly, instead of avoiding them, houseflies exhibited a preference for flowers occupied by female spiders, suggesting the attractive nature of female spider colouration. This paper provides evidence for the first time that crab spiders use floral scent to achieve olfactory crypsis and proposes a potential yellow‐signalling mechanism for crab spiders to attract insects. Background: Single-domain von Willebrand factor type C (SVWC) constitute a protein family predominantly identified in arthropods, characterized by a SVWC domain and involved in diverse physiological processes such as host defense, stress resistance, and nutrient metabolism. Nevertheless, the physiological mechanisms underlying these functions remain inadequately comprehended. Results: A massive expansion of the SVWC gene family in Musca domestica (MdSVWC) was discovered, with a count of 35. MdSVWC1 was selected as the representative of the SVWC family for functional analysis, which led to the identification of the immune function of MdSVWC1 as a novel pattern recognition receptor. MdSVWC1 is highly expressed in both the fat body and intestines and displays acute induction upon bacterial infection. Recombinant MdSVWC1 binds to surfaces of both bacteria and yeast through the recognition of multiple pathogen-associated molecular patterns and exhibits Ca2+-dependent agglutination activity. MdSVWC1 mutant flies exhibited elevated mortality and hindered bacterial elimination following bacterial infection as a result of reduced hemocyte phagocytic capability and weakened expression of antimicrobial peptide (AMP) genes. In contrast, administration of recombinant MdSVWC1 provided protection to flies from bacterial challenges by promoting phagocytosis and AMP genes expression, thereby preventing bacterial colonization. MdSPN16, a serine protease inhibitor, was identified as a target protein of MdSVWC1. It was postulated that the interaction of MdSVWC1 with MdSPN16 would result in the activation of an extracellular proteolytic cascade, which would then initiate the Toll signaling pathway and facilitate the expression of AMP genes. Conclusions: MdSVWC1 displays activity as a soluble pattern recognition receptor that regulates cellular and humoral immunity by recognizing microbial components and facilitating host defense. Simple Summary: House flies are important carriers of diseases and can cause economic losses in livestock by reducing feeding activity. Overuse of chemical insecticides has led to resistance in many house fly populations worldwide. This study aimed to assess how two house fly populations in Antalya, Türkiye respond to the insecticides cyfluthrin and etofenprox. By exposing adult flies to these chemicals, the researcher found that the Kemer population showed very low resistance to cyfluthrin, while the Serik population exhibited moderate resistance. For etofenprox, Kemer flies showed no resistance, and Serik flies showed very low resistance. The findings suggest that integrated pest control methods and regular resistance monitoring are necessary to manage and prevent further resistance in house fly populations. Presence of this vector causes economic losses due to decreased feeding activity in livestock, resulting in reduced yields of products. The repeated and unconscious application of chemical insecticides has resulted in the development of resistance in the majority of house fly populations worldwide. The aim of this research was to determine the susceptibility levels of two field M. domestica populations to cyfluthrin and etofenprox in Antalya, Türkiye. The LD50 values, resistance ratios, and resistance status were determined by exposing adult house flies to the test chemicals for one hour. The Kemer population exhibited very low resistance to cyfluthrin, with a 5-fold increase, while the Serik population, in contrast, showed moderate resistance with a 29.67-fold increase. The resistance ratios of field populations to etofenprox were 2.33-fold (no resistance) for Kemer and 6.44-fold (very low resistance) for Serik, respectively. This is the first study to determine the resistance levels of house flies against cyfluthrin and etofenprox insecticides in Türkiye. To prevent or reduce the development of resistance to insecticides in house flies, integrated control methods and resistance management programs should be employed. The common housefly, Musca domestica, known for transmitting over 100 infections, was studied using green-synthesized Cadmium Sulfide nanoparticles (CdS NPs) from Agaricus bisporus. These CdS NPs were tested on third-instar larvae under laboratory conditions using dipping and feeding methods with concentrations (75, 100, 125, 150, 175, and 200 µg/mL). The toxicity, measured by LC50, was found to be 138 µg/mL for dipping treatment and 123 µg/mL for feeding treatment. Analysis with an energy-dispersive X-ray microanalyzer confirmed Cd accumulation in the larval midgut, indicating penetration of CdS NPs into the organism, which may potentially increase their toxicity. CdS NPs caused disruptions in Heat Shock Protein 70, cell apoptosis, and various biochemical components. Scanning electron microscopy revealed morphological abnormalities in larvae, pupae, and adults exposed to CdS NPs. Ultrastructural examination showed significant midgut tissue abnormalities in larvae treated with 123 µg/mL of CdS NPs. The plant essential oils are intricate blends of semi-volatile and volatile substances. They primarily consist of monoterpenes and sesquiterpenes; however, very trace amounts of diterpenes, triterpenes, and tetraterpenes are also present. In view of environmental concerns, highly aromatic essential oils, exhibit a variety of bioactivities against the targeted species as a novel pest control alternative to synthetic insecticides. As a result, the study aims to evaluate the impact of essential oils (EO) extracted from various plant species: Azadirachta indica, Citrus limon, Eucalyptus globulus, Laurus nobilis, Mentha piperita , and Ocimum tenuiflorum. Gas chromatography and Mass spectrometry (GC–MS) were used to determine the chemical components of these essential oils. Furthermore, the extracted oils were subjected to individual and combination formulations of bio-insecticides, which were assessed on houseflies and cockroaches in comparison to a blank control and the positive control dichlorvos (dimethyl 2, 2-dichlorovinyl phosphate). The findings showed that the maximum essential oil yields from the leaves of A. indica, E. globulus , and O. tenuiflorum were 11.38, 9.64 and 10.41 %, respectively. Compared to DDVP, which killed all insects within the 12-hour period, the combined form of leaf essential oils at 100 % concentration proved to be more effective in controlling insect mortality. It accounted for 98 % of the deaths in houseflies and 76.4 % of the deaths in cockroaches, according to the contact toxicity evaluation. Background: Flies are acknowledged as vectors of diseases transmitted through mechanical means and represent a significant risk to human health. The study aimed to determine the prevalence of enteropathogens carried by flies in Pudong New Area to inform strategies for preventing and controlling flies. Methods: Samples were collected from various locations in the area using cage trapping techniques between April and November 2021, encompassing various habitats such as parks, residential areas, restaurants, and farmers' markets. The main fly species were identified using cryomicrography and taxonomic enumeration, with 20 samples per tube collected from different habitats. Twenty-five enteropathogens were screened using GI_Trial v3 TaqManTM microbial arrays. Results: A total of 3,875 flies were collected from 6,400 placements, resulting in an average fly density of 0.61 flies per cage. M. domestica were the most common species at 39.85%, followed by L. sericata at 16.57% and B. peregrina at 13.14%. Out of 189 samples, 93 tested positive for enteropathogens, with nine different pathogens being found. 12.70% of samples exclusively had parasites, a higher percentage than those with only bacteria or viruses. The study found that M. domestica had fewer enteropathogens than L. sericata and B. peregrina, which primarily harbored B. hominis instead of bacteria and viruses such as E. coli, Astrovirus, and Sapovirus. During spring testing, all three fly species exhibited low rates of detecting enteropathogens. M. domestica were found in residential areas with the highest number of pathogen species, totaling six. In contrast, L. sericata and B. peregrina were identified in farmers' markets with the highest number of pathogen species, totaling six and seven, respectively. Simple Summary: Enzymes of the detoxification system are known to play a crucial role in insecticide resistance in insects. The role of P450 monooxygenases in providing resistance to chlorfenapyr is ambiguous and of interest due to possible negative cross-resistance to pyrethroids and other insecticides, the resistance to which is ensured by increased activity of these enzymes. This study aimed to perform a biochemical and molecular evaluation of P450 monooxygenases in susceptible and chlorfenapyr-resistant strains of Musca domestica L. depending on sex. The chlorfenapyr-resistant strain of M. domestica L. did not differ from the susceptible strain in terms of the pattern of overall P450-monooxygenase activity: the larvae of both strains had lower enzymatic activity than the adults. However, the development of resistance to chlorfenapyr in house flies was accompanied by an increase in P450-monooxygenase activity without changes in CYP6D1 expression. PCR-RFLP analysis revealed a previously undescribed mutation in the promoter region of the CYP6D1 gene of M. domestica, the effect of which on the gene expression level was not detected. The house fly Musca domestica L. is one of the most common insects of veterinary and medical importance worldwide; its ability to develop resistance to a large number of insecticides is well known. Many studies support the involvement of cytochrome P-450-dependent monooxygenases (P450) in the development of resistance to pyrethroids, neonicotinoids, carbamates, and organophosphates among insects. In this paper, the monooxygenase activity and expression level of CYP6D1 were studied for the first time in a chlorfenapyr-resistant strain of house fly. Our studies demonstrated that P450 activity in adults of the susceptible strain (Lab TY) and chlorfenapyr-resistant strain (ChlA) was 1.56–4.05-fold higher than that in larvae. In females of the Lab TY and ChlA strains, this activity was 1.53- and 1.57-fold higher, respectively (p < 0.05), than that in males, and in contrast, the expression level of CYP6D1 was 21- and 8-fold lower, respectively. The monooxygenase activity did not vary between larvae of the susceptible strain Lab TY and the chlorfenapyr-resistant strain ChlA. Activity in females and males of the ChlA strain exceeded that in the Lab TY strain specimens by 1.54 (p = 0.08) and 1.83 (p < 0.05) times, respectively, with the same level of CYP6D1 expression. PCR-RFLP analysis revealed a previously undescribed mutation in the promoter region of the CYP6D1 gene in adults of the Lab TY and ChlA strains, and it did not affect the gene expression level. The house fly, Musca domestica L. (Diptera: Muscidae), is considered to be a nuisance pest and a mechanical carrier of medical and veterinary pathogenic organisms. Because the fly has developed resistance to a variety of commonly used insecticides, novel alternative pest control tactics are being researched. Five desiccant dust-based formulations, silica gel (CimeXa), perlite (Imergard WP), diatomaceous earth (Celite 610, biologically produced in diatoms), silica gel + pyrethrins + piperonyl butoxide (PBO, a synergist) (Drione), and silica gel + thyme oil (EcoVia WD), were evaluated for effects on house fly egg hatching, larval development to pupation, adult emergence from pupae, and adult survival in the laboratory. While silica gel, perlite, and diatomaceous earth did not reduce egg hatching, pupation, and adult emergence from pupae, the silica gel + pyrethrins + PBO and silica gel + thyme oil formulations provided 100% control of those life stages. Adult house flies exposed to complete bodily coating and limited exposure from tarsal contact were killed by all three desiccant dust products without botanicals and two products formulated with botanicals. The formulations with bioactive botanicals provided more rapid control (3–4 h) than the desiccant dusts without botanicals (<24 h). • GWP of housefly larvae treatment is significantly higher in summer than in winter. • Bioconversion itself contributed very little to GWP. • GWP will increase slightly when larvae are stored by drying rather than freezing. • There is a contradiction between a higher mature larvae production and a lower GWP. The significant increase in the demand for biomass waste treatment after garbage classification has led to housefly larvae treatment becoming an attractive treatment option. It can provide a source of protein while treating biomass waste, which means that nutrients can be returned to the natural food chain. However, the performance of this technology in terms of its environmental impacts is still unclear, particularly with regards to global warming potential (GWP).This study used a life cycle assessment (LCA) approach to assess a housefly larvae treatment plant with a treatment capacity of 50 tons of biomass waste per day. The LCA results showed that the 95% confidence intervals for the GWP in summer and winter were determined to be 24.46–32.81 kg CO 2 equivalent (CO 2 -eq)/ton biomass waste and 5.37–10.08 kg CO 2 -eq/ton biomass waste, respectively. The greater GWP value in summer is due to the longer ventilation time and higher ventilation intensity in summer, which consumes more power. The main GWP contributions are from (1) electricity needs (accounting for 78.6% of emissions in summer and 70.2% in winter) and (2) product substitution by mature housefly larvae and compost (both summer and winter accounting for 96.8% of carbon reduction). The common house fly, Musca domestica L., is a mechanical vector of numerous diseases that may be spread from one living thing to another by sponging their mouthparts on their body and leg hairs after they have vomited. House flies (Musca domestica) have been known as a mechanical vector in spreading infectious diseases such as cholera, shigellosis, salmonellosis and skin infections. The present study aims to investigate the abundance of M. domestica adults in an urban region of Cairo Governorate, Egypt, throughout one year and bacterial diversity on its body surface. Adults of M. domestica were collected monthly by hunting. Collected adults were subjected to molecular identification. Bacteria were isolated from the surface of flies' body pieces and identified by automated identification systems (VITEK). Obtained data revealed that the highest abundance (201.67±15.57) was recorded in May 2022, while the lowest abundance (17.33±0.57) was recorded in April 2023. Also, M. domestica abundance positively correlated significantly (P≤0.001) with temperature (r= 0.895) and relative humidity (r= 0.827). Musca domestica species were molecularly identified, and the sequences were then banked on NCBI under the accession numbers OQ784762-OQ784764. The highest bacterial pathogen presence were Gram-positive Staphylococcus spp. and Bacillus spp. (77.0 %). Gram-negative Escherichia coli, Pseudomonas aeruginosa, and Klebsiella pneumonias had the lowest presence (total of 23.0 %). On the other hand, the results of antibiotic sensitivity revealed that Bacillus spp., Pseudomonas aeruginosa, and Klebsiella pneumonia are more resistant to several antibiotics than Staphylococcus spp. and Escherichia coli. Insect chitinases have been proposed as potential targets for pest control. In this work, a novel group IV chitinase gene, MdCht9, from Musca domestica was found to have multiple functions in the physiological activity, including chitin regulation, development and antifungal immunity. The MdCht9 gene was cloned and sequenced, its phylogeny was analysed and its expression was determined in normal and 20E treated larvae. Subsequently, RNA interference (RNAi)‐mediated MdCht9 knockdown was performed, followed by biochemical assays, morphological observations and transcriptome analysis. Finally, the recombinant protein MdCht9 (rMdCht9) was purified and tested for anti‐microbial activity and enzyme characteristics. The results showed that MdCht9 consists of three domains, highly expressed in a larval salivary gland. RNAi silencing of MdCht9 resulted in significant down‐regulation of chitin content and expression of 15 chitin‐binding protein (CBP) genes, implying a new insight that MdCht9 might regulate chitin content by influencing the expression of CBPs. In addition, more than half of the lethality and partial wing deformity appeared due to the dsMdCht9 treatment. In addition, the rMdCht9 exhibited anti‐microbial activity towards Candida albicans (fungus) but not towards Escherichia coli (G−) or Staphylococcus aureus (G+). Simple Summary: This study investigated the detection and discrimination of the neonicotinoid insecticide imidacloprid by behaviorally resistant and susceptible house flies (Musca domestica L.). Flies were allowed to contact a sucrose solution containing either a low or a high concentration of imidacloprid with their tarsi alone or with both their tarsi and proboscis. The proboscis extension response (PER) for each house fly was recorded at 0, 2, and 10 s following the start of tarsal contact with the test solution. Following proboscis contact with the sucrose solution containing a high concentration of imidacloprid, behaviorally resistant flies had a significant reduction in PER (within 2 s), while imidacloprid-susceptible flies showed no differences in PER associated with the concentration of imidacloprid. When only the tarsi were allowed to contact either solution, there were no significant differences in PER observed for either fly strain (resistant or susceptible). These results suggest that behaviorally resistant house flies detect imidacloprid and can discriminate among low and high concentrations following proboscis contact but not tarsal contact with a sucrose solution containing imidacloprid. Understanding the mechanisms responsible for behavioral resistance to insecticides by the house fly is critical for creating sustainable pest management strategies for this fly. The house fly, Musca domestica L., is a significant human and livestock pest. Experiments used female adult house flies glued onto toothpicks for controlled exposure of their tarsi alone (tarsal assay) or their tarsi and proboscis (proboscis assay) with a sucrose solution containing imidacloprid at either a low (10 µg/mL) or high (4000 µg/mL) concentration. Proboscis extension response (PER) assays were used to characterize the response of imidacloprid-susceptible and behaviorally resistant house fly strains to contact with sucrose solutions containing either a low or high concentration of imidacloprid. In each assay, 150 female flies from each fly strain were individually exposed to sucrose solutions containing either a low or high concentration of imidacloprid by deliberate contact of the fly tarsi to the test solution. The PER for each fly was subsequently recorded at 0, 2, and 10 s following the initial tarsal contact. A significant and rapid reduction in PER was observed only for the behaviorally resistant fly strain and only following contact by the flies' proboscis with the sucrose solution containing the high imidacloprid concentration. The results suggest that chemoreceptors on the fly labellum or internally on the pharyngeal taste organs are involved in the detection of imidacloprid and discrimination of the concentration, resulting in an avoidance behavior (proboscis retraction) only when imidacloprid is at sufficient concentration. Many entomopathogenic fungi cause infections that kill their insect host. Little is understood about changes in the reproductive investment that occurs during an infection by a lethal disease over the waning life of an insect. Life-history theory suggests the host will respond by investing resources into fighting the disease or increasing reproduction. Here, we investigate how the reproductive life of adult house flies, Musca domestica , is impacted by its host-specific fungal pathogen, Entomophthora muscae. Specifically, we test how the week-long infection alters the mating behavior of virgin adult male house flies. We find that the pathogen significantly decreases male libido, an effect which grows stronger over the course of the infection. Furthermore, females were significantly less likely to choose an infected male, reducing male mating success. Additionally, we assessed sperm viability to understand the reproductive costs for monandrous females to mate with infected males. Analyses revealed that sperm quality decreases as early as 3 days post-infection. These results show that E. muscae , which can have a prevalence near 100% in wild populations, causes severe lifetime reproductive costs to male house flies. Cyromazine is a triazine insect growth regulator insecticide that is recommended for control of Musca domestica worldwide. Cyromazine is highly effective in causing mortality of M. domestica; however, some aspects of its lethal and sublethal effects on the biology of M. domestica are still unknown. The present study explored lethal and sublethal effects on several biological traits and population parameters of M. domestica. Concentration–response bioassays of cyromazine against third-instar larvae of M. domestica exhibited sublethal and lethal effects from concentrations of 0.03 (LC10), 0.06 (LC25), and 0.14 (LC50) μg/g of a larval medium. Exposure of M. domestica larvae to these concentrations resulted in reduced fecundity, survival, longevity and oviposition period, and delayed development of immature stages (i.e., egg hatch time and larval and pupal durations) in the upcoming generation of M. domestica. The values of population parameters such as intrinsic rate of increase, finite rate of increase, net reproductive rate, age-specific survival rate and fecundity, and age–stage life expectancy and reproductive value, analyzed using the age–stage and two-sex life table theory, were significantly reduced in a concentration-dependent manner in comparison with the control group. In conclusion, the study highlights the significant effects of cyromazine on the biology of M. domestica that could help suppress its population in cases of severe infestations. The goal of this study, which was carried out in 2022 in the Advanced Entomology Laboratory of the Department of Biology, Girls College of Education, University of Kufa, was to determine how well the fungus filtrates of Alternaria alternata affected the mortality percentages of house fly larvae and pupae, Musca domistica. The results indicated an increase in the mortality percentages of larvae in the first and third instar of the house fly, Musca domistica with increasing concentrations used. In the study, the highest mortality percentages were recorded after three days of treatment with a concentration of 10% (100 and 85%) in the first and third instar larvae, respectively. The effect of the fungus filtrates on some biological aspects when treating the third instar larvae of houseflies, as it led to a decrease in pupation percentages to 15% when treating pupae with a concentration of % 10, and the pupal deformation percentages to 10% and a decrease in adult emergency percentages to 0%. when treating pupae with the fungus filtrates of Alternaria alternata at a concentration of 10 %, which increased the pupal deformation to 80% and decreased adult emergency percentages to 20%. This study assessed the impact of an alcoholic extract of Citrus aurantium leaves on the mortality percentages in the larvae and pupae of house flies, Musca domistica, and was carried out in the Advanced Entomology Laboratory in the Department of Biology at the Girls College of Education at the University of Kufa in 2022. The results indicated an increase in the mortality percentages of larvae in the first and third stars of the house fly, Musca domistica with increasing concentrations used. In this study, the highest mortality percentages were recorded after three days of treatment with a concentration of 50 mg/ml (100 and 61%) in the first and third instar larvae, respectively. The achievement of this research was to study the influence of AgNPs on the life stages of Muscadomestica. The supernatant of Trichodermaharzianum successfully reduces AgNO3 into AgNPs through the duration of incubation, as noticed by the color change from white to red. The absorbance of UV–visible light results in an absorbance peak at 418 nm for nanoparticle solutions of Ag particles. The scanning electron microscopy (SEM) image confirmed the formation of AgNPs with an average particle size of 46.63 nm without aggregation or adhesion. The microscopy of energy-dispersive X-ray (EDX) approved the formation of AgNPs with a high amount of some atoms of carbon and oxygen derivatives from components of fungus extract used to reduce AgNO3. The concentration of AgNPs prepared with serial dilutions of 5%, 10%, 15%, 20% and 25%, also with sterilized distilled water as a control and the influence of AgNPs on phases of M. domestica had been studied. The eggs of the studied insects were affected by 20% of AgNPs, which showed an increase in incubation time of 11 h without any hatched eggs at 25%. The larval stage of the insect was also influenced by AgNPs solution, which at 25% increased the duration of incubation to reach 11 h with 27 larval insects dead, compared with 7 h without any missing membrane in the larval stage of the insect. As well as the pupal phase of insects, we increased the period of incubation to 10 h with 7 h in control, and in addition, the number of pupal insects dead reached 23 in 25% concentration. Finally, the AgNPs also increased the number of adult insects to reach 27 at 25% concentration. Zoophilic Diptera plays a leading role in the epizootic foci formation of many infectious and parasitic diseases and directly affects the quality of livestock products. The current study aimed to analyze the number and species composition of parasitic Diptera in industrial, farm, and homestead agrobiocenoses of large and small cattle, pig, and poultry farms in Eastern Ukraine. The research involved entomological collection during the peak activity daylight hours in early May, July, and early September 2021-2022 per farm. A total of 360 entomological collections were made, and 4310 zoophilous flies were examined. In livestock farms of five districts of the Kharkiv region of Ukraine, 28 species of zoophilic flies were registered, among which Musca domestica, Muscina stabulans, Stomoxys calcitrans, Lucilia sericata, Protophormia terraenovae, and Drosophila species were dominant species. The analysis revealed that cattle biocenoses hosted 27 fly species, pigs had 8 species, and poultry and small cattle each had 7 species. The study indicated an increase in the population of Musca autumnalis, the main species in the pastures, near livestock premises during the summer. Stomoxys calcitrans was also recorded in livestock agrobiocenoses. The species Musca domestica, Musca autumnalis, and Stomoxys calcitrans account for 78.8% to 88.3% of the entire complex of zoophilous flies. The two species of Ortellia caesarion (shiny dung beetle) and Ortellia cornicina (green dung beetle), known for their role as manure mineralizers and deemed non-threatening to animals, were completely absent during the research period. The findings indicated the species of Eristalis tenax in agrobiocenoses in 2021. Therefore, it can be concluded that zoophilic flies are physical irritants to animals and potential carriers of many infectious diseases, especially diseases caused by unicellular organisms. Musca domestica is one of the most important agents in the transmission of pathogenic organisms from contaminated materials to the human body. In various studies, the role of flies in the transmission of many diseases has been well-proven. The purpose of this study was to investigate the bacterial contamination of Musca domestica in different sources. In this study, samples were collected from slaughterhouses, coastal areas, hospital grounds, and urban and rural waste. After collection, the samples were transferred into sterile glass containers and then the isolation and identification of bacterial species was done in the laboratory. Based on the obtained results, Escherichia coli was the dominant species in this study. From all the examined samples, most of the contaminations were related to E. coli, Staphylococcus aureus, Enterobacter aerogenes, and Staphylococcus epidermidis, respectively. Based on the results of this study, it can be said that the presence of these insects in public centers can be carriers of pathogenic agents. Considering the necessity of preventing the spread of infection and pollution in public places and different parts of the environment, it seems necessary to improve and carry out sanitary engineering measures to control these insects. The morphogenetic consequences of exposure to short-term heat stress (STHS) in two housefly strains mass-selected for lifespan were studied based on the assessment of adult wing variability by geometric morphometrics. Significant differences in the size and shape of the wing between the control and impact groups of different genders in the strains were revealed. Shg (short lived) and Lg (long-lived). The STHS effect manifested in an increase in the size of the wing and a directed change in its shape. The between-group hierarchy of gender and stress-induced differences is expressed in the same way in both strains of flies. The range of linear differences is significantly higher than the gender differences, which, in turn, are higher than the level of stress-induced ones. Instability of imago wing development (Vm) in Shg was significantly higher than the Lg strains, and higher in all groups of females, but in most cases significantly lower in impact groups (taking into account the increase in size, the latter may be associated with the effect of hormesis). It is hypothesized that the directed morphogenetic effects of STHS are based on latent species modifications, the appearance of which in the phenotype is due to stress-induced epigenetic genome rearrangements that cause similar morphological changes in the wing in groups of adult males and females of both strains. The phenotypic plasticity of strains during selection for different lifespans and the changes induced by STHS directly indicate the reality of stress-induced rapid morphogenetic rearrangements under a sharp change in environmental conditions. Background: The house fly, Musca domestica, is vector for pathogens and parasites and causes economic damage to livestock by reducing forage conversion efficiency, negatively impacting weight gain and milk production. It has shown resistance to multiple insecticide classes. The aim of this research was to determine the susceptibility levels of seventeen field M. domestica strains to thiamethoxam, a neonicotinoid insecticide, in Türkiye. Methods: Insecticide susceptibility of the house flies to thiamethoxam was determined using the WHO glass jar method. A probit analysis program was used to determine LD50 values, and then the resistance ratios were compared with insecticide-susceptible strain. Results: All strains were ≥18.5-fold resistant to thiamethoxam. The data showed that 10 out of 17 strains had either high or very high resistance levels. Our findings revealed that house flies from solid waste landfills in Samsun, Ankara, and Kocaeli exhibited higher resistance ratios compared to those found in animal shelters. Conversely, in Gaziantep, Antalya, İzmir and Erzurum, the exact opposite trend was observed. Regarding the LD50 values among solid waste storage areas, the lowest rate was obtained from Gaziantep (0.72 gr ai/m²), and the highest rate was obtained from Ankara (9.35 gr ai/m²). Furthermore, regarding the LD50 values among animal shelters, the lowest was obtained from Samsun (0.37 gr ai/m²), and the highest was obtained from Denizli (21800 gr ai/m2). Lipids are an important class of nutrients for insects, as they are the main energy source during metamorphosis and crucial for survival and reproduction. Lipid reserves in insects are mostly accumulated during larval development and can be influenced by environmental factors such as diet and temperature. Genetic factors can also affect larval lipid content and composition. Larvae of the house fly, Musca domestica L. (Diptera: Muscidae), are a valuable emerging resource for the insect feed sector, fueling a growing interest in the factors regulating larval lipid content. We investigated whether strains of three geographical origins vary in ontogeny, quantity, and composition of stored lipids in response to temperature. Italian, Spanish, and Dutch house fly larvae were reared on a similar diet at 25 and 35 °C. Larval dry weight and total lipid content were determined in early‐, mid‐, and late‐third instars by extraction of soluble lipids. The fatty acid composition was analyzed in late‐third instars using an improved extraction and derivatization protocol. We found that strain, larval stage, and temperature affected larval dry weight and absolute and relative lipid content, with significant interaction effects between these three factors. All flies were reared in a common environment, indicating a genetic component to lipid storage. Analysis of lipid composition identified 11 fatty acids, including some rarely reported in the literature. Five fatty acids accounted for 80 and 81% of the total fatty acid methyl esters detected at 25 and 35 °C, respectively. An effect of temperature but not strain was evident on the composition of the fatty acids. As resistance to the limited number of insecticides available for medical and veterinary pests becomes more widespread, there is an urgent need for new insecticides and synergists on the market. To address this need, we conducted a study to assess the toxicity of three monoterpenoids—carvone, menthone, and fenchone—in comparison to permethrin and methomyl against adults of two common pests: the yellow fever mosquito (Aedes aegypti) and the house fly (Musca domestica). We also examined the potential for these monoterpenoids to enhance the effectiveness of permethrin and methomyl when used together. Finally, we evaluated the ability of each monoterpenoid to inhibit acetylcholinesterase, comparing them to methomyl. While all three monoterpenoids performed relatively poorly as topical insecticides (LD50 > 4000 ng/mg on M. domestica; >6000 ng/mg on Ae. aegypti), they synergized both permethrin and methomyl as well as or better than piperonyl butoxide (PBO). Carvone and menthone yielded synergistic co-toxicity factors (23 and 29, respectively), which were each higher than PBO at 24 h. Currently, the mechanism of action is unknown. During preliminary testing, symptoms of acetylcholinesterase inhibition were identified, prompting further testing. Acetylcholinesterase inhibition did not appear to explain the toxic or synergistic effects of the three monoterpenoids, with IC50 values greater than 1 mM for all, compared to the 2.5 and 1.7 µM for methomyl on Aedes aegypti and Musca domestica, respectively. This study provides valuable monoterpenoid toxicity and synergism data on two pestiferous insects and highlights the potential for these chemistries in future pest control formulations. The gastrointestinal tract of all animals, including insects, is colonized by a remarkable array of microorganisms which are referred to collectively as the gut microbiota. The hosts establish mutually beneficial interactions with the gut microbiota. However, the mechanisms shaping these interactions remain to be better understood. Here, we investigated the roles of Musca domestica peptidoglycan recognition protein SC (MdPGRP‐SC), a secreted pattern recognition receptor, in shaping the gut microbial community structure by using biochemical and high‐throughput sequencing approaches. The recombinant MdPGRP‐SC (rMdPGRP‐SC) could strongly bind various pathogen‐associated molecular patterns (PAMPs) including peptidoglycan, lipopolysaccharide and D‐galactose, and exhibited mild affinity to β‐1, 3‐glucan and D‐mannose. Meanwhile, rMdPGRP‐SC could also bind different kinds of microorganisms, including gram‐positive bacteria (Bacillus subtilis and Staphylococcus aureus), gram‐negative bacteria (Escherichia coli and Pseudomonas aeruginosa) and yeast (Pichia pastoris). rMdPGRP‐SC also exhibited weak antibacterial activity against Bacillus subtilis. Knockdown of MdPGRP‐SC by RNAi reduced the persistence of ingested E. coli and a load of indigenous microbiota in the larval gut significantly. In addition, depleted MdPGRP‐SC also altered the gut microbiota composition and led to increased ratios of Gram‐negative bacteria. We hypothesize that MdPGRP‐SC is involved in maintaining gut homeostasis by modulating the immune intensity of the gut through multiple mechanisms, including degrading or neutralizing various PAMPs and selectively suppressing the growth of some bacteria. Considering the functional conservation of the peptidoglycan recognition protein (PGRP) family in insects, the catalytic PGRPs might be promising candidate targets not only for pest and vector control but also for the treatment of bacterial infection in insect farming. Introduction: Although house flies (Musca domestica) do not directly cause disease in humans, they transmit pathogens to them, which provide the basis for many diseases. The main way to deal with this insect is to use insecticides. Due to the resistance from insecticides, the fight against house flies has been hampered. This study aimed to determine the prevalence of knockdown resistance against organochlorine insecticides in house flies worldwide. Methods: This study was conducted via a systematic review and meta-analysis to investigate the prevalence of knockdown resistance against organochlorine insecticides in house flies. Accordingly, by searching the databases of Web of Science, PubMed, Scopus, Proquest, Bioone, and Embase, all published articles were extracted, and reviewed until the end of May 2022. Statistical data analysis was performed using the random-effects model in the meta-analysis, meta-regression, and I2 index. Results: Nine studies entered the meta-analysis process. Based on this, the prevalence of knockdown resistance against organochlorine insecticide in house flies was estimated to be 49.1%. Meta-regression showed that the prevalence of knockdown resistance increased with increasing years of study but decreased with increasing sample size. Conclusion: According to the findings, about 50% of house flies have knockdown resistance against organochlorine insecticide. As a result, it is necessary to adopt effective and combined methods to combat this insect to control it and prevent the transmission of diseases caused by it.

The extensive use of insecticides is the main reason for the development of resistance in many insect species. The development of resistance in house flies was documented for numerous insecticides. Pirimiphos-methyl is used for house fly control in Saudi Arabia. This study was conducted to evaluate the resistance of M. domestica field populations against pirimiphos-methyl. The field-collected populations of M. domestica displayed different levels of resistance. The samples collected from Riyadh city exhibited the highest resistance, followed by populations from Jeddah and Taif. We investigated the genetic mutations of the acetylcholinesterase (Ace) gene in field-collected flies and the survivors after exposure to pirimiphos-methyl. The outcomes of the present study provide valuable information that may help in house fly management in Saudi Arabia. The house fly Musca domestica L. (Diptera: Muscidae) is a worldwide medical and veterinary pest, causing great economic losses. Organophosphate insecticides have been widely used to control house fly populations. The main objectives of the present study were to evaluate the resistance levels of M. domestica slaughterhouse populations, collected from Riyadh, Jeddah, and Taif, against the organophosphate insecticide pirimiphos-methyl and investigate the genetic mutations of the Ace gene associated with pirimiphos-methyl resistance. The obtained data showed that there were significant differences among pirimiphos-methyl LC50 values of the studied populations, where the highest LC50 was recorded for the Riyadh population (8.44 mM), followed by Jeddah and Taif populations (2.45 mM and 1.63 mM, respectively). Seven nonsynonymous SNPs were detected in the studied house flies. The Ile239Val and Glu243Lys mutations are reported for the first time, whereas Val260Leu, Ala316Ser, Gly342Ala, Gly342Val, and Phe407Tyr were previously reported in M. domestica field populations from other countries. Considering three mutations associated with insecticide resistance, at amino acid positions 260, 342, and 407 of acetylcholinesterase polypeptide, 17 combinations were recovered in this study. Three out of these seventeen combinations were frequently found both worldwide and in the three Saudi house fly field populations, as well as their pirimiphos-methyl-surviving flies. Overall, the single and combined Ace mutations are apparently associated with pirimiphos-methyl resistance, and the obtained data can be useful in managing house fly field populations in Saudi Arabia. This study was designed to isolate the bacteria carried by houseflies (Musa domestica), determine the most occurring bacteria associated with houseflies and assess the sanitary conditions of the different sampling locations in Calabar Metropolis. Samples were collected during September and October 2015. Perforated transparent plastic buckets and polyethylene terephthalate (PET) bottles were used to collect houseflies from houses, food spots (restaurants) and drinking spots located in Bogobiri, Goldie and Hawkings in Calabar. The samples were subjected to standard bacteriological analysis to include cultures and biochemical identification. The results of the trap were 137 (51.1%) for the bucket trap and 131 (48.9%) for the Eva bottle trap. Bogobiri location had the highest number of houseflies 95 (35.4%) followed by Hawking 90 (33.6%) and lastly Goldie with 83 (30.9%). and yielded the following results: 13 (61.9%) of the sample had significant bacteria growth while 15 (71.4%) had insignificant bacteria growth. Among bacteria isolated, Coliforms bacteria 5 (23.8%), Salmonella spp. 5 (23.8%), S. aureus 4 (19%) and Bacillus spp. 2 (9.5%). Hawking had the highest number of bacteria isolated. The result of this survey has shown that the association of houseflies with street foods and drinks is a serious public health concern. Essential oils (EOs) have recently attracted more interest due to their insecticidal activities, low harmfulness, and rapid degradation in the environment. Therefore, Lavandula steochas (L. steochas) essential oil was assessed for its chemical constituents, in vitro cytotoxicity, and scolicidal, acaricidal, and insecticidal activities. Using spectrometry and gas chromatography, the components of L. steochas EOs were detected. Additionally, different oil concentrations were tested for their anticancer activities when applied to human embryonic kidney cells (HEK-293 cells) and the human breast cancer cell line MCF-7. The oil's scolicidal activity against protoscolices of hydatid cysts was evaluated at various concentrations and exposure times. The oil's adulticidal, larvicidal, and repelling effects on R. annulatus ticks were also investigated at various concentrations, ranging from 0.625 to 10%. Likewise, the larvicidal and pupicidal activities of L. steochas against Musca domestica were estimated at different concentrations. The analyses of L. steochas oil identified camphor as the predominant compound (58.38%). L. steochas oil showed significant cytotoxicity against cancer cells. All of the tested oil concentrations demonstrated significant scolicidal activities against the protoscoleces of hydatid cysts. L. steochas EO (essential oil) showed 100% adulticidal activity against R. annulatus at a 10% concentration with an LC50 of 2.34%, whereas the larvicidal activity was 86.67% and the LC50 was 9.11%. On the other hand, the oil showed no repellent activity against this tick's larva. Furthermore, L. steochas EO achieved 100% larvicidal and pupicidal effects against M. domestica at a 10% concentration with LC50 values of 1.79% and 1.51%, respectively. In conclusion, the current work suggests that L. steochas EO could serve as a potential source of scolicidal, acaricidal, insecticidal, and anticancer agents. [ABSTRACT FROM AUTHOR]

Background: High frequency of Helicobacter pylori infection and the unknown mode of transmission prompted us to investigate H. pylori–wild housefly relationship. H. pylori causes chronic gastritis, peptic ulcers, and stomach cancer. H. pylori persists in the gut of the experimentally infected houseflies. The existence of H. pylori strains isolated from wild houseflies, on the other hand, has never been documented. Materials and Methods: In this study, 902 wild houseflies from different sites were identified as Musca domestica, then 60 flies were screened by traditional microbiological techniques and H. pylori-specific 16S rRNA gene. The antibiotic resistance (ART) was investigated phenotypically. Wild housefly gut bacterial isolates were further evaluated genotypically to have 23S rRNA gene mutation related to clarithromycin resistance. To find efficient therapeutic alternatives, the potency of three plant extracts (garlic, ginger, and lemon) and the wasp, Vespa orientalis venom was evaluated against H. pylori. The cytotoxic effect of the crude wasp venom, the most potent extract, against Vero and Colon cancer (Caco2) cell lines was investigated using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. Results: All isolates from houseflies were positive. The isolated bacteria have variable resistance to frequently used antibiotics in all isolates. Minimum inhibitory concentration values of 15.625 mg/mL for both ginger and lemon extracts, 7.8125 mg/mL for garlic extract, and 0.0313 mg/mL for wasp venom were recorded. Wasp venom has the most potent antibacterial activity compared with the four antibiotics that are currently used in therapies against H. pylori. Conclusion: We conclude that wild houseflies can play a role in disseminating H. pylori. The housefly gut may be a suitable environment for the horizontal transfer of ART genes among its associated microbiome and H. pylori. Livestock farming is currently reducing the use of synthetic insecticides because of the development of resistance in insect pests. Plant-based bioinsecticides are considered alternatives to synthetic pesticides. Therefore, the present study investigated the chemical composition and discriminating concentrations (DCs) of essential oils from Syzygium aromaticum (L.) Merr. and L. M. Perry (SA), Cinnamomum porrectum (Roxb.) Kosterm (CP), and Litsea cubeba (Loureiro) Persoon (LC) against laboratory-reared Musca domestica (larvae and adults) and field-derived Stomoxys indicus (adults) using larval dip and adult contact bioassays. All essential oils were analyzed using gas chromatography/mass spectroscopy. The most common components in the SA, CP, and LC oils were eugenol, safrole, and terpenoids, respectively. The results of a larval test indicated that CP was most effective against M. domestica with a DC of 6.134% v/v. In adult bioassays, CP was also the most toxic oil against M. domestica (DC = 30.644% v/v), whereas SA displayed the greatest toxicity against S. indicus (DC = 1.434% v/v). Moreover, in the larval bioassay results of oils tested at 1, 5, and 10% v/v in M. domestica , 1% and 5% v/v CP had the shortest median lethal times values of 68.88 and 19.44 min, respectively, whereas, at 10% v/v, SA displayed the shortest median lethal time (0.03 min), followed by CP (1.74 min) and LC (19.02 min). However, additional data are needed to further evaluate the semi-field and field effects of CP and SA on M. domestica and S. indicus under realistic operational conditions. [ABSTRACT FROM AUTHOR]

There are many different traps that have been used to trap insects such as flies and can aide in providing evidence such as time of death of a decedent or narrow down a geographic region. This makes flies very important in the forensic science world. The use of different flytraps is also beneficial to the public when it comes to knowing which ones to use. Musca domestica (Linnaeus) (Diptera: Muscidae) is an extremely common species of fly and there are various types of fly traps available. Genital myiasis is an infestation of genital organs by fly larvae, where they feed and develop as parasites. They can cause severe infection, inflammatory reaction and can be linked to psychiatric disturbances. We report a rare case of genital myiasis in an elderly postmenopausal woman aged 82 years from Udaipur, Rajasthan. She presented with complaints of intense pain in the genital region and was diagnosed as a case of genital myiasis of a prolapsed uterus. Pelvic examination revealed 'Stage-IV' genitourinary prolapse according to Pelvic Organ Prolapse-Quantification (POP-Q classification), with a big excavatory ulcer indwelled with maggots of Musca domestica. About 100 such maggots were subsequently manually removed with forceps. With proper surgical and nonsurgical interventions, the patient healed completely and the prolapse was reduced completely. This work aimed to evaluate the chemical composition, insecticidal and acaricidal potential of the essential oil (EO) obtained from the resurrection plant Myrothamnus moschatus (Baill.) Baill. (Myrothamnaceae) from Madagascar. The EO bioactivity was evaluated against selected arthropod pests and vectors of agricultural and public health relevance. The most abundant volatile compounds were trans-pinocarveol (37.7 ± 4.2%) and pinocarvone (20.8 ± 3.1%), similar to the EO of the chemotype collected from the same region. Lethal concentrations (LC50) or doses (LD50) from acute toxicity tests were estimated for Musca domestica (L.) adults at 22.7 µg adult−1, for Spodoptera littoralis (Boisduval) larvae at 35.6 µg larva−1, for Culex quinquefasciatus (Say) at 43.6 µg mL−1, for adults of Metopolophium dirhodum (Walker) at 2.4 mL L−1, and for adults of Tetranychus urticae (Koch) at 1.2 mL L−1. The good insecticidal and acaricidal activities determined in this work may open a new perspective on the use of this plant as a source of botanical insecticide ingredients. Adult house flies feed and breed in a variety of microbe‐rich habitats and serve as vectors for human and animal pathogens. To better understand their role in harbouring and disseminating bacteria, we characterized the composition and diversity of bacterial communities in the gut of female house flies collected from three different habitats in Kansas: agricultural (dairy farm), urban (business area dumpsters) and mixed (business located between residential and animal agriculture areas). Bacterial community composition and diversity were influenced more by the house flies' habitat than by sampling time. The most abundant taxa were also highly prevalent in the house flies collected from all three habitats, potentially representing a 'core microbiome' attributable to the fly's trophic and reproductive associations with substrates and food sources comprised of decaying matter and/or animal waste. Bacterial taxa associated with vertebrate guts/faeces and potential pathogens were highly abundant in agricultural fly microbial communities. Interestingly, taxa of potential pathogens were highly abundant in flies from the mixed and urban sites. House flies harboured diverse bacterial communities influenced by the habitat in which they reside, including potential human and animal pathogens, further bolstering their role in the dissemination of pathogens, and indicating their utility for pathogen surveillance. Horn flies, Haematobia irritans irritans (L.); house flies, MuscadomesticaL.; and stable flies, Stomoxys calcitrans (L.), are problematic to livestock production and, in the instance of M. domestica, to human health. p-Anisaldehyde was assessed for its bioactivity against the eggs of the three muscid species. The compound, which is found in many edible plants, was lethal to the eggs of H. irritans and S. calcitrans at a relatively low (=0.01%) concentration when eggs were exposed to the treated substrate and when exposed to fumigation (=0.0001%). Although M. domestica eggs were less vulnerable to the treated substrate than the other two muscids, egg hatch was delayed, and fumigation with a low p-anisaldehyde concentration (0.001%) induced substantial egg mortality. In many instances, larval development inside the eggs did not occur, while other larvae developed, swelled, and although they broke the egg casing, they failed to emerge. The possible utility of p-anisaldehyde against the eggs of the three muscid species as a possible integrated pest management tactic is discussed. [ABSTRACT FROM AUTHOR]

The housefly, Musca domestica Linnaeus (Diptera: Muscidae), is an important pest not only causing annoying to humans but is also known for its ability to transmit different infectious microorganisms to humans and animals. Recently, zinc oxide nanoparticles (ZnONPs) synthesized by microbial organisms have gained much attention in pest control because of their extensive antimicrobial activity, eco-friendliness, and simplicity. Therefore, the green synthesis of ZnO nanoparticles was applied in the current study as a new and effective biopesticide against the house fly, Musca domestica by feeding the first instar larvae on different concentrations of ZnONPs synthesized by Bacillus foraminis (0.01, 0.025, 0.05, 0.1, 0.25, and 0.5 mg). Results showed significant mortality values in the larval stage (1.84 ± 0.10, 3.50 ± 0.29, 5.75 ± 0.25, 7 ± 0.41, 8 ± 0.41 and 8.75 ± 0.25) while pupae recorded mortality values of (1.75 ± 0.14, 1 ± 0.00, 1.63 ± 0.13 and 1 ± 0.00) at the higher concentrations (0.05, 0.1, 0.25, 0.5 mg), respectively and some morphological changes appeared in some pupae. Also, the adult emergence had a significant reduction (100 ± 0.00, 82.5 ± 2.50, 62.5 ± 4.79, 37.5 ± 2.50, 28.35 ± 0.95, 17 ± 0.08 and 0 ± 0.00), in comparison with the control group C (10 ± 0.00). Insects have the phenomenon of immune priming by which they can have enhanced protection against reinfection with the same pathogen, and this immune protection can be passed on to their offspring, which is defined as "trans-generational immune priming (TGIP)." But whether housefly possesses TGIP is still unclear. Therefore, we used the housefly as the insect model and Candida albicans as the pathogen to explore whether the housefly is capable of eliciting TGIP, and RNA sequencing (RNA-seq) was performed to explore the molecular mechanism of TGIP of the housefly. We found that the housefly possesses TGIP, and adults pre-exposed to heat-killed C. albicans could confer protection to itself and its offspring upon reinfection with a lethal dose of C. albicans. RNA-seq results showed that 30 and 154 genes were differentially expressed after adults were primed with heat-killed C. albicans (CA-A) and after offspring larvae were challenged with a lethal dose of C. albicans (CA-CA-G), respectively. Among the differentially expressed genes (DEGs), there were 23 immune genes, including 6 pattern recognition receptors (PRRs), 7 immune effectors, and 10 immunoregulatory molecules. More importantly, multiple DEGs were involved in the Toll signaling pathway and phagosome signaling pathway, suggesting that the Toll signaling pathway and phagocytosis might play important roles in the process of TGIP of housefly to C. albicans. House fly Musca domestica L. is a pest of humans, poultry, and livestock across the world. Dependence on chemical insecticides to contain the flies provided varying results and their continued use has led to development of insecticide resistance. Bioactive compounds in plants are an alternative source to manage M. domestica. Lemon grass, Cymbopogon citratus Stapf essential oil caused fumigant toxicity to eggs (LC50 1.299 mg/dm³) and adults (16.56 mg/ dm³). The C. citratus EO caused larval repellence. Polyvinylpyrrolidone when used as polymer matrix to load C. citratus at 1:1, 1:2, and 1:3 caused toxicity to flies for a longer period as compared to use of EO alone. The EO loaded in polymer matrix had a slower dissipation, EO+ PVP polymer mixed at 1:3 retained over 80% of EO after 72 hr when exposed to 60oC. EO, whilst EO alone without a dispenser dissipated in 3 hr. The biological effect of C. citratus EO on M. domestica can be enhanced for a longer period if loaded into a polymer matrix and this would be an effective strategy to manage M. domestica. Patagonia is a geographical area characterized by a wide plant biodiversity. Several native plant species are traditionally used in medicine by the local population and demonstrated to be sources of biologically active compounds. Due to the massive need for green and sustainable pesticides, this study was conducted to evaluate the insecticidal activity of essential oils (EOs) from understudied plants growing in this propitious area. Ciprés (Pilgerodendron uviferum), tepa (Laureliopsis philippiana), canelo (Drimys winteri), and paramela (Adesmia boronioides) EOs were extracted through steam distillation, and their compositions were analyzed through GC–MS analysis. EO contact toxicity against Musca domestica L., Spodoptera littoralis (Boisd.), and Culex quinquefasciatus Say was then evaluated. As a general trend, EOs performed better on housefly males over females. Ciprés EO showed the highest insecticidal efficacy. The LD50(90) values were 68.6 (183.7) and 11.3 (75.1) µg adult−1 on housefly females and males, respectively. All EOs were effective against S. littoralis larvae; LD50 values were 33.2–66.7 µg larva−1, and tepa EO was the most effective in terms of LD90 (i.e., <100 µg larva−1). Canelo, tepa, and paramela EOs were highly effective on C. quinquefasciatus larvae, with LC50 values < 100 µL L−1. Again, tepa EO achieved LD90 < 100 µL L−1. This EO was characterized by safrole (43.1%), linalool (27.9%), and methyl eugenol (6.9%) as major constituents. Overall, Patagonian native plant EOs can represent a valid resource for local stakeholders, to develop effective insecticides for pest and vector management, pending a proper focus on their formulation and nontarget effects. Introduction: Housefly (Musca domestica), because of its body structure, filthy and feeding habits, is reported to be capable of carrying a variety of microbes, including potential pathogens, over its body appendages. Aims: This study aimed to investigate M. domestica in Calabar for pathogenic gut parasites, determine its role in parasitic diseases transmission, make appropriate recommendations toward its management and effective control of the associated parasitic diseases, and hence promote the society health status. Settings and Design: This was a cross-sectional study. Materials and Methods: A total of 300 M. domestica specimens (150 each from sanitary and insanitary areas, pooled in batches of five flies per pool) were used for the study. The parasites were detected from the spun deposits of fly body surface wash and gut contents using direct microscopy and formol-ether technique. Statistical Analysis Used: Data were analyzed using Statistical Package for the Social Sciences (SPSS), version 21.0, and Chi-square test at a significant level of P < 0.05. Results: Overall frequency of parasites detection was 46.7%, with helminths and protozoa showing similar frequency, 23.3% each. External body surface of flies recorded a statistically significant higher parasites frequency 76.7% than their gut contents 16.7% (P = 0.000). A statistically significant higher parasite detection rate was observed in the insanitary (63.3%) than in sanitary areas (30.0%) (P = 0.0114). The parasites detected in this study were Entamoeba histolytica/dispar 39.9%, hookworm (21.4%), Ascaris lumbricoides (17.9%), Trichuris trichiura (14.3%), and Giardia intestinalis (7.2%). Conclusion: This study has confirmed M. domestica in Calabar as a mechanical carrier of potential pathogenic gut parasites, especially in the insanitary areas, with E. histolytica/dispar being the most frequently encountered. The synthesized pyrazolopyrimidine derivatives conjugated with selenium nanoparticles were prepared via a reaction of pyrazolone 1 with aryl-aldehyde and malononitrile or 3-oxo-3-phenylpropanenitrile in the presence ammonium acetate or pipridine using an ultrasonic bath as a modified method in the organic synthesis for such materials. The structure of the synthesized compounds was elucidated through various techniques. All the synthesized pyrazolopyrimidines were used in the synthesis of selenium nanoparticles (SeNPs). These nanoparticles were confirmed using UV-spectra, Dynamic Light scattering and (TEM) techniques. The larvicidal efficiency;of the synthesized;compounds; was investigated against some strains such as Culex pipiens; and Musca domestica larvae. Bioassay test showed pyrazolopyrimide derivatives to exhibit an acceptable larvicidal;bio-efficacy. The derivative (3) exhibited;the highest;efficiency for more than; lab strains of both species. Moreover, C. pipiens larvae were more sensitive towards the examined compounds than M. domestica. The field;strain displayed lower affinity for the 2 folds compounds. Some biochemical changes were tracked through analysis of insect main metabolites (protein, lipid and carbohydrate), in addition to measuring the changes in seven enzymes after treatment. Generally, there was a reduction in the protein, lipids and carbohydrates after treatment with all tested compounds. Moreover, a decrement was noticed for acetylcholine esterase and glutathione;S-transferase; enzymes. There was an increment in the acid;phosphatase; and alkaline phosphatase. In addition, there was elevation in Phenoloxidase level but it noticed the declination in both Cytochrome P450 and Ascorbate peroxidase activity after treatment both flies with derivatives of selenium-nanoparticles in both lab and field strain. Generally, the experiments carried out indicate that antioxidant and detoxification enzymes may play a significant role in mechanism of action of our novel nanocompounds. The cytotoxicity of the synthesized compounds and conjugated with SeNPs showed enhanced compatibility with human normal fibroblast cell line (BJ1) with no toxic effect. Over the past few years, there has been growing interest in the ability of insect larvae to convert various organic side-streams containing mycotoxins into insect biomass that can be used as animal feed. Various studies have examined the effects of exposure to aflatoxin B 1 (AFB 1) on a variety of insect species, including the larvae of the black soldier fly (BSFL; Hermetia illucens L.; Diptera: Stratiomyidae) and the housefly (HFL; Musca domestica L.; Diptera: Muscidae). Most of these studies demonstrated that AFB 1 degradation takes place, either enzymatic and/or non-enzymatic. The possible role of feed substrate microorganisms (MOs) in this process has thus far not been investigated. The main objective of this study was therefore to investigate whether biotransformation of AFB 1 occurred and whether it is caused by insect-enzymes and/or by microbial enzymes of MOs in the feed substrate. In order to investigate this, sterile and non-sterile feed substrates were spiked with AFB 1 and incubated either with or without insect larvae (BSFL or HFL). The AFB 1 concentration was determined via LC-MS/MS analyses and recorded over time. Approximately 50% of the initially present AFB 1 was recovered in the treatment involving BSFL, which was comparable to the treatment without BSFL (60%). Similar patterns were observed for HFL. The molar mass balance of AFB 1 for the sterile feed substrates with BSFL and HFL was 73% and 78%, respectively. We could not establish whether non-enzymatic degradation of AFB 1 in the feed substrates occurred. The results showed that both BSFL and substrate-specific MOs play a role in the biotransformation of AFB 1 as well as in conversion of AFB 1 into aflatoxin P 1 and aflatoxicol, respectively. In contrast, HFL did not seem to contribute to AFB 1 degradation. The obtained results contribute to our understanding of aflatoxin metabolism by different insect species. Antibiotic-resistant bacteria (including MRSA) in the clinic pose a growing threat to public health, and antimicrobial peptides (AMPs) have great potential as efficient treatment alternatives. Houseflies have evolved over long periods in complex, dirty environments, developing a special immune system to overcome challenges in harmful environments. AMPs are key innate immune molecules. Herein, two differentially expressed AMPs, Phormicins A and B, were identified by screening transcriptomic changes in response to microbial stimulation. Structural mimic assays indicated that these AMPs exhibited functional divergence due to their C-terminal features. Expression analysis showed that they had different expression patterns. Phormicin B had higher constitutive expression than Phormicin A. However, Phormicin B was sharply downregulated, whereas Phormicin A was highly upregulated, after microbial stimulation. The MIC, MBC and time-growth curves showed the antibacterial spectrum of these peptides. Crystal violet staining and SEM showed that Phormicin D inhibited MRSA biofilm formation. TEM suggested that Phormicin D disrupted the MRSA cell membrane. Furthermore, Phormicin D inhibited biofilm formation by downregulating the expression of biofilm-related genes, including altE and embp. Therefore, housefly Phormicins were functionally characterized as having differential expression patterns and antibacterial & antibiofilm activities.
