HIV-1 Vpu is a small transmembrane phosphoprotein of 16 kDa which performs critical roles in CD4 proteolysis and virus release. Previous studies have demonstrated that Vpu-induced degradation of CD4 occurs in the endoplasmic reticulum (ER), and that the proteolytic process is sequence specific requiring both the transmembrane and cytoplasmic domains of CD4. In the present study, we investigated the effects of Vpu expression on the intracellular membrane trafficking pathway of mammalian cells. In singly transfected cells, the HIV envelope glycoproteins and vesicular stomatitis virus glycoprotein (VSV G) were properly transported to the cell surface undergoing oligosaccharide modifications characteristic of their movement through the Golgi complex. In contrast, the cell surface delivery of glycoproteins was severely impeded in cells expressing Vpu. Biochemical analyses revealed that Vpu expression blocked the transfer of proteins from the ER-Golgi complex to the plasma membrane in a dose- and protein-dependent manner. Soluble gp120 exhibited extreme transport defects in the presence of Vpu, whereas transmembrane proteins (e.g., gp160, VSV) responded only moderately to wild-type Vpu. To gain insight into Vpu-mediated transport inhibition, we performed mutational analysis of the CK-2 phosphorylation sites (serines at 52 and 56) in the Vpu protein. CK-2 phosphorylation of Vpu has been shown to regulate the activity of the protein in reactions that involve the proteolysis of CD4 in the ER. We demonstrate here that the phosphorylation mutant is defective in both sequence-specific degradation of VRE-containing substrates and the transport inhibition of gp120 and VSV-G in the secretory pathway. Thus, these experiments have revealed that Vpu-mediated proteolysis and transport inhibition are mechanistically coupled requiring the same structural elements of the Vpu protein in both processes. We propose that the primary effect of Vpu expression is to impede the secretion process and then access glycoproteins bearing the VRE for Vpu-mediated proteolysis in the ER of mammalian cells.

The Vpu protein of HIV-1 induces degradation of CD4 in the endoplasmic reticulum. Previous studies have elucidated the role of the CD4 cytoplasmic domain in the Vpu-induced degradation process, and the minimal Vpu responsive element mapped to a small region in the CD4 tail. In the present study, we have carried out both biochemical and biological experiments to analyze the role of the CD4 anchor domain in the Vpu-induced degradation process. We generated chimeric proteins that possessed the ecto-anchor domains of gp160 and the cytoplasmic domain of CD4. The chimeric envelope glycoproteins were functionally active in the fusion of HeLa CD4+ cells, with the exception of those having the arginine to isoleucine (R to I) substitution in the gp160 anchor domain. Coexpression studies revealed that these chimeric glycoproteins were stable and functionally active in the presence of Vpu, as opposed to those having the anchor-cytoplasmic domains of CD4. The half-life of Vpu-sensitive chimeric glycoproteins was calculated to be approximately 60-90 min, whereas Vpu-resistant envelope glycoproteins exhibited relatively longer half-lives in the presence of Vpu. Taken together, these studies strongly suggest that the CD4 anchor domain appears to provide critical sequence or structural elements through which the Vpu protein could access CD4 or glycoproteins bearing the Vpu responsive element for degradation in the endoplasmic reticulum.

We investigated endoproteolytic processing of the human immunodeficiency virus (HIV) envelope glycoprotein precursor, gp160, as well as envelope-mediated membrane fusion in the presence of CD4 molecules that were either partially or fully retained in the endoplasmic reticulum (ER). Pulse-chase analyses revealed that gp160 formed complexes with CD4 molecules, and gp160 in the complex was endoproteolytically cleaved to gp120 and gp41 in the secretory pathway. The gp120/gp41 complex thus generated was properly targeted to the plasma membrane in cells expressing gp160 and wild-type CD4 or mutant CD4 molecules that were partially retained in the ER. Additionally, membrane fusion (syncytium) assays were performed to monitor the presence or absence of gp120/gp41 complexes at the cell surface of cotransfected cells and demonstrated that the HIV-1 envelope glycoprotein-mediated membrane fusion was appreciably reduced in the presence of wild-type CD4 or either one of the mutant CD4 molecules. Reduction in the formation of syncytia appears to be due predominantly to saturation of the CD4 binding site on the gp120/gp41 complex at the cell surface of cotransfected cells, but partial retention of the complex in the ER could also partly account for the reduction. However, the intracellular gp120/gp41 complex generated in cells expressing gp160 and CD4 mutant having the transmembrane ER retention signal (KKTC) was completely retained in the ER and hence could not participate in membrane fusion events at the plasma membrane. Taken together, these data suggest that the endoproteolytic cleavage of gp160 occurs in the ER or cis-Golgi network, and ER retention strategies can potentially be used in preventing the spread of HIV-1 infection in permissive cells.

The human immunodeficiency virus type 1 (HIV-1) Vpu protein is a transmembrane phosphoprotein which induces rapid degradation of CD4 in the endoplasmic reticulum (ER). To identify sequences in CD4 for Vpu-induced degradation, we generated four chimeric envelope glycoproteins having the ectodomain of HIV-1 gp160, the anchor domain of CD4, and 38, 25, 24, and 18 amino acids (aa) of the CD4 cytoplasmic domain. Using the vaccinia virus-T7 RNA polymerase expression system, we analyzed the expression of chimeric proteins in the presence and absence of Vpu. In singly transfected cells, the chimeric envelope glycoproteins having 38, 24, and 18 aa of the CD4 cytoplasmic domain were endoproteolytically cleaved and biologically active in the fusion of HeLa CD4+ cells. However, one of the chimeras having 25 aa of the CD4 cytoplasmic tail was retained in the ER using the transmembrane ER retention signal and was defective in membrane fusion. Furthermore, biochemical analyses of the coexpressing cells revealed that the Vpu protein induced degradation of the envelope glycoproteins having 38, 25, and 24 aa of the CD4 cytoplasmic tail and degradation occurred in the ER. Consequently, the fusion-competent glycoproteins did not induce the formation of syncytia in HeLa CD4+ cells expressing Vpu. However, the HIV-1 gp160 and chimeric envelope glycoprotein having the membrane-proximal 18 aa of the CD4 cytoplasmic tail were stable and fusion competent in cells expressing Vpu. In addition, we examined the stability of CD4 molecules in the presence of Vpu. Coexpression analyses revealed that the Vpu protein induced degradation of CD4 whereas mutant CD4 having the membrane-proximal 18 aa of the cytoplasmic domain was relatively stable in the presence of Vpu. Taken together, these studies have elucidated that the Vpu protein requires sequences or sequence determinants in the cytoplasmic domain of CD4 to induce degradation of the glycoproteins in the cell.
