Coordenação de Aperfeiçoamento de Pessoal de Nível Superior As análises histomorfométricas geram parâmetros espermatogênicos que podem ser usados para compreensão da biologia reprodutiva das espécies. Esses dados, aplicados aos componentes testiculares, permitem a descrição da espermatogênese das espécies e, a partir deles, é possível elencar informações precisas dos processos reprodutivos intraespecíficos. O testículo é dividido em compartimento tubular, onde acontece a espermatogênese, e compartimento intertubular onde as células de Leydig produzem a testosterona. Oecomys bicolor é um roedor de pequeno porte encontrado no cerrado brasileiro e Monodelphis americana é um marsupial, também de pequeno porte, presente na Mata Atlântica. Oecomys bicolor tem 89,72% do parênquima testicular alocado em túbulos seminíferos e 10,28% em compartimento intertubular, enquanto Monodelphis americana apresenta 83,16% do parênquima testicular alocado em túbulos seminíferos e 16,84% alocados em compartimento intertubular. Em ambas as espécies, pela distribuição dos componentes testiculares, os machos foram descritos tendo comportamento sexual promíscuo. Pela descrição dos estádios pelo método do desenvolvimento acrossômico foram descritos 12 estádios para Oecomys bicolor e 10 estádios para Monodelphis americana. Palavras-chave: Espermatogênese. Histomorphometric analyzes produces spermatogenic parameters that can be used to understand the reproductive biology of the species. These data applied to testicular components allow the description of the spermatogenesis of the species and from them it is possible to list precise information about the intraspecific reproductive processes. The testis is divided into a tubular compartment where spermatogenesis occurs and an intertubular compartment where Leydig cells produce testosterone. Oecomys bicolor is a small rodent found in the Brazilian cerrado and Monodelphis americana is a small marsupial present in the Atlantic Forest. Oecomys bicolor has 89.72% of the testicular parenchyma allocated in the seminiferous tubules and 10.28% in the intertubular compartment. Monodelphis americana presents 83.16% of the testicular parenchyma allocated in the seminiferous tubules and 16.84% allocated in the intertubular compartment. In both species, by the distribution of testicular components, males were described as having promiscuous sexual behavior. Coordenação de Aperfeiçoamento de Pessoal de Nível Superior Uma tendência significativa para um declínio mundial na fertilidade masculina humana tem sido relatada nos últimos anos. Enquanto isso, a exposição a químicos tem sido proposta para potencialmente contribuir para os distúrbios reprodutivos masculinos, em que químicos ambientais e farmacológicos/recreacionais podem induzir efeitos prejudiciais à saúde reprodutiva masculina. Entre uma variedade complexa de produtos químicos ambientais, o cádmio (Cd) é um dos metais pesados que se classifica entre os metais prioritários com importância para a saúde pública, uma vez que o Cd é considerado como um desregulador endócrino. Os seres humanos estão expostos diariamente ao Cd por meio de fontes ocupacionais (e.g. produção de baterias) e não ocupacionais (e.g. alimentos/água contaminados e fumaça de cigarro). Além disso, o consumo de cannabis para fins terapêuticos e recreativos está em ascensão. Neste contexto de produtos químicos e fertilidade masculina, a exposição aos compostos da cannabis, chamados fitocanabinoides (canabinoides), pode resultar em resultados adversos para a saúde reprodutiva masculina. Portanto, a presente tese teve como objetivo avaliar o impacto dessas duas tendências globais, cádmio e canabinoides presentes na cannabis, no processo espermatogênico. Já se sabe que o cádmio induz danos ao testículo. No entanto, os mecanismos pelos quais o cádmio promove o aparecimento de patologias testiculares e a relação entre dose, via e tempo de exposição e lesões são pouco conhecidos. Nesse sentido, realizamos uma revisão sistemática (Capítulo 1) para responder a essas questões. Depois de recuperar estudos originais em bancos de dados, organizamos os resultados em uma estrutura de Adverse Outcome Pathway (AOP). Além disso, uma análise de viés foi realizada. Nos 37 estudos selecionados, ficou claro que o cádmio induz histopatologias significativas em testículos de modelos murinos independente de via, de maneira dose e tempo-dependente. Os danos podem ser observados nas primeiras horas de exposição, principalmente danos vasculares, sugerindo que a falha na vascularização é o mecanismo primário envolvido. A AOP mostrou que potenciais eventos moleculares iniciais podem ser o mimetismo e a interferência com elementos essenciais desregulando proteínas (estruturais e antioxidantes), alteração na atividade da enzima de fosforilação oxidativa e alteração da expressão gênica, que levam à falha reprodutiva. No entanto, a análise de viés mostrou que as evidências atuais apresentam qualidade metodológica pobre. Em relação ao impacto dos canabinoides na saúde reprodutiva masculina, realizamos dois ensaios biológicos. Em um manuscrito (Capítulo 2), buscamos responder se os dois principais fitocanabinoides presentes na cannabis poderiam representar um perigo para o testículo humano adulto. Para isso, expusemos testículos humanos adultos ao Δ 9- tetrahidrocanabinol (THC), canabidiol (CBD), ou CBD/THC (proporção 1:1) nas concentrações de 10 -9 M a 10 -5 M por 48 horas, utilizando nosso modelo ex vivo único de cultura organotípica. Nossos achados não mostraram alteração na histologia testicular, produção de testosterona, número de células apoptóticas e proliferativas e expressão de genes que codificam importantes proteínas testiculares. Uma vez que alguns problemas reprodutivos masculinos foram hipotetizados como sendo de origem fetal, no último estudo (Capítulo 3) avaliamos se a exposição pré-natal ao CBD/THC poderia levar a distúrbios reprodutivos masculinos na vida adulta. Para isso, expomos camundongos fêmeas prenhas do dia de gestação 6,5 (E6,5) ao E15,5 a 5mg/kg/dia de CBD/THC (proporção 1:1), por gavagem. Observamos diminuição do peso testicular e alterações morfológicas testiculares, estas identificadas por análises morfométricas, na prole adulta. No entanto, essas alterações não foram acompanhadas por mudanças nos números de espermatogônias indiferenciadas e células de Sertoli. A análise da expressão gênica revelou um aumento na expressão de genes pró-apoptótico (Bax), de diferenciação de células germinativas (Sohlh1) e de regulação meiótica (Stra8). Não foram observadas alterações nos níveis séricos de testosterona. Em geral, nossos resultados demonstram que o cádmio, um poluente ambiental, pode afetar a saúde reprodutiva masculina de modelos murinos e conduzir à infertilidade masculina. Entretanto, os dois fitocanabinoides presentes na cannabis, THC e CBD, os quais são também utilizados como químicos farmacológicos, não causam efeitos agudos diretos na histofisiologia testicular usando nossa cultura organotípica bem caracterizada de testículo humano adulto. Porém, exposição pré-natal a uma mistura desses dois canabinoides derivados da canabis pode conduzir ao peso testicular diminuído de camundongos na vida adulta. Ainda, a exposição pré-natal pode acelerar a meiose e a diferenciação celular e induzir apoptose, baseado nos achados moleculares, provavelmente, contribuindo de forma compensatória para a diminuição do peso do testículo. Assim, em camundongos, exposição pré-natal a uma mistura de THC/CBD pode afetar a biologia testicular de proles adultas. No geral, os resultados apresentados por esta tese fornecem novos conhecimentos sobre a toxicologia reprodutiva e a saúde reprodutiva masculina, que podem ser considerados para preservar a fertilidade em toda a população. A significant trend towards a worldwide decline in human male fertility has been reported over the last years. In the meantime, exposure to chemicals has been proposed to potentially contribute to male reproductive disorders, in which environmental and pharmacological/recreational chemicals might induce detrimental effects on male reproductive health. Among a complex variety of environmental chemicals, cadmium (Cd) is one of the heavy metals that rank among the priority metals that are of public health significance, since Cd is considered as an endocrine disruptor. Human beings are daily exposure to Cd through occupational (e.g. production of batteries) and non-occupational (e.g. contaminated food/water, cigarette smoke) sources. Furthermore, cannabis consumption for medical and recreational purposes is on the rise. In this context of chemicals and male fertility, exposure to cannabis compounds, called phytocannabinoids (cannabinoids), could result in adverse outcomes to male reproductive health. So, this present thesis aimed to assess the impact of these two global trends, cadmium and cannabinoids, in the spermatogenic process. It is already known that cadmium induces damage to the testis. However, the mechanisms that cadmium leads to testicular histopathologies and the relationship between dose, route, and time of exposure and injuries are poorly understood. In this sense, we conducted a systematic review (Chapter 1) to answer these questions. After retrieving original studies on databases, we organized the results into an Adverse Outcome Pathway (AOP) framework. Also, a bias analysis was performed. In the 37 studies selected, it was clear that cadmium induces significant histopathologies in the murine model’s testis regarding routes, in a dose- and time-dependent manner. The damages can be observed in the first hours of exposure, mainly vascular damages suggesting that vasculature failure is the primary mechanism involved. The AOP showed that potential molecular initiating events may mimic and interfere with essential elements disrupting proteins (structural and antioxidants), change in the oxidative phosphorylation enzyme activities, and gene expression alteration, which lead to reproductive failure. However, analysis of bias showed that the current evidence presents poor methodological quality. Regarding the cannabinoid impact on male reproductive health, we performed two biological essays. In one manuscript (Chapter 2), we aimed to answer whether the two main phytocannabinoids present in cannabis could represent a hazard for adult human testis. For that, we exposed adult human testis to Δ 9 -tetrahydrocannabinol (THC), cannabidiol (CBD), or CBD/THC (ratio 1:1) at the concentrations from 10 -9 M to 10 -5 M for 48 hours, by using our unique ex vivo model of organotypic culture. Our findings show no alteration in testis histology, testosterone production, number of apoptotic and proliferative cells, and expression of some genes encoding important testicular proteins. Since some male reproductive problems have been hypothesized to have a fetal origin, in the last study (Chapter 3), we assessed whether prenatal exposure to CBD and THC could lead to male reproductive disorders in adult life. For that, we exposed pregnant mice from embryogenic day 6.5 (E6.5) to E15.5 to 5mg/kg/day of CBD/THC (ratio 1:1), by gavage. Then, we observed decreased testicular weight and testicular morphological alterations identified by morphometric analyses in adult offspring. However, these alterations were not followed by changes in the undifferentiated spermatogonia and Sertoli cell numbers. The gene expression analysis revealed an increase in the expression of pro-apoptotic (Bax), germ cell differentiation (Sohlh1), and regulating meiotic (Stra8) genes. Alterations in the serum testosterone levels were not observed. In general, our findings demonstrate that cadmium, an environmental pollutant, can affect the male reproductive health of murine models and lead to male infertility. On the other hand, the two main cannabinoids present in cannabis, THC and CBD, which are also used as pharmacological chemicals, do not cause acute direct effects in the testicular histophysiology using our well-characterized organotypic culture of adult human testis. However, prenatal exposure to a mixture of these cannabis-derived cannabinoids can lead to decreased testis weight of mice in adult life. Also, prenatal exposure can accelerate meiosis and cellular differentiation and induce apoptosis, based on the molecular findings, probably, contributing compensatory to the decreased testis weight. So, in mice, prenatal exposure to a mixture of THC/CBD might affect the testis biology of adult offspring. Overall, the results presented by this thesis provide new insights into male repro-toxicology and reproductive health, which might be considered to preserve fertility across the population. Alguns resíduos estão relacionados a emergências de saúde ambiental por causarem impactos no ecossistema e na saúde humana. Entre esses resíduos se destacam os metais pesados. O termo metal pesado se estende a uma variedade de metais, entre eles o cádmio (Cd), chumbo (Pb), cromo (Cr VI) e níquel (Ni) e semimetais como o arsênio (As), em suas formas oxidadas +5 (arsenato) e +3 (arsenito). Nos testículos, os metais pesados podem prejudicar células em divisão e diferenciação, causando danos morfológicos e funcionais, podendo ainda afetar as células de Leydig e a produção de testosterona. Assim, os objetivos deste estudo foram avaliar o impacto da exposição intraperitoneal crônica aos metais pesados. As +5 , As +3 , Cd, Pb, Cr (VI) e Ni nos testículos de camundongos Swiss adultos e estabelecer uma ordem de toxicidade entre eles. Foram utilizados 42 camundongos Swiss em idade reprodutiva, distribuídos aleatoriamente em sete grupos experimentais (n=6 animais/grupo). A exposição aos metais pesados foi feita em doses semanais, via intraperitoneal, de forma que o grupo 1 recebeu 0,7mL de solução salina 0,9% (controle), o grupo 2 recebeu 1,5 mg/Kg de arsenato, o grupo 3 recebeu 1,5 mg/Kg de arsenito, o grupo 4 recebeu 1,5 mg/kg de cádmio, o grupo 5 recebeu 1,5 mg/Kg de cromo, o grupo 6 recebeu 1,5 mg/Kg de níquel e o grupo 7 recebeu 1,5 mg/Kg de chumbo, durante um período de seis semanas. Os animais foram eutanasiados após 42 dias, sendo os testículos retirados e pesados. Os dados não paramétricos foram comparados por meio do teste de Kruskal-Wallis e os dados paramétricos por meio de análise de variância (ANOVA) seguida pelo teste de Student Newman-Keuls e foram considerados significativos quando p≤0,05. Nossos resultados mostram que Cd e Ni foram capazes de afetar a morfometria do tecido testicular, enquanto As, Pb, Cr (VI) e Ni alteraram o status oxidativo. Além disso, alterações histopatológicas foram identificadas após exposição a todos os metais pesados. Assim, a comparação de toxicidade entre os metais estudados neste trabalho estabelece que Ni>Cd>Pb=As +3 >Cr (VI) > As +5 . Os dados desse trabalho podem contribuir para melhor compreensão da toxicidade dos metais pesados para a saúde reprodutiva e podem guiar futuras terapias para o tratamento de danos por meio de apropriado reparo após exposição aos metais pesados. Among these residues, heavy metals stand out. The term heavy metal extends to a variety of metals, including cadmium (Cd), lead (Pb), chromium (Cr VI) and nickel (Ni) and semimetals like arsenic (As), in their oxidized forms +5 (arsenate) and +3 (arsenite). In the male reproductive system, specifically in the testes, heavy metals can harm cells in division and differentiation, causing morphological and functional damage. In addition, they affect Leydig cells and, consequently, the production of steroid hormones. Thus, the objectives of this study were to identify the impact of chronic intraperitoneal exposure to heavy metals arsenate, arsenite, cadmium, lead, chromium and nickel in the testes of adult Swiss mice and establish an order of toxicity among the metals studied. 42 Swiss mice in reproductive age (140 days) were used, which were randomly divided into seven experimental groups (n = 6 animals / group). The group 1 received 0.7mL 0.9% saline (control), group 2 received 1.5 mg/ kg of arsenic in the form of arsenate, group 3 received 1.5 mg/ kg of arsenic in the form of arsenite, group 4 received 1.5 mg/ kg of cadmium, group 5 received 1.5 mg/ kg of chromium and group 6 received 1.5 mg/ kg of nickel and group 7 received 1.5 mg/ kg of lead. Exposure to heavy metals was made in weekly doses, intraperitoneally, over a period of six weeks. The animals were euthanized after 42 days of experiment and then the testes were removed and weighed. Non-parametric data were compared using the Kruskal-Wallis test and parametric data using analysis of variance (ANOVA) followed by the Student Newman-Keuls test and were considered significant when p≤0.05. Our results indicated that Cd and Ni were able to affect the morphometry of the testicular tissue, while As, Pb, Cr (VI) and Ni altered the oxidative status. In addition, histopathological changes were identified after exposure to all heavy metals. Thus, the toxicity comparison between the metals studied in this work establishes that Ni> Cd> Pb = As +3 > Cr (VI)> As +5 . The data from this work can contribute to a better understanding of the toxicity of heavy metals for reproductive health. Furthermore, they can guide future therapies for treatment of damage through appropriate repair after exposure to heavy metals. Keywords: Heavy metals. Reproductive toxicology. Spermatogenesis.

Metilfenidato é um estimulante que atua no Sistema Nervoso Central, possuindo ações e estrutura semelhantes às anfetaminas. Foi usado inicialmente para tratar fadiga. Hoje é um medicamento muito conhecido no mercado para tratar Transtorno do Déficit de Atenção com Hiperatividade (TDAH). Em indivíduos com TDAH, os neurotransmissores dopamina e noradrenalina apresentam-se deficitários em quantidade e funcionamento. Desse modo, o mecanismo de ação do metilfenidato consiste em bloquear os transportadores de dopamina e noradrenalina. Baseado nestes dados, o objetivo foi avaliar os efeitos de duas doses de metilfenidato, sobre os testículos de camundongos adultos, por meio de avaliações morfológicas, histométricas, hormonais e espermáticas. Vinte e quatro camundongos Balb/c foram divididos aleatoriamente em três grupos experimentais (n=8): Grupo 01: controle (água destilada); Grupo 02: cloridrato de metilfenidato na dosagem de 20mg/kg/dia (MFD20); Grupo 03: cloridrato de metilfenidato na dosagem de 40mg/kg/dia (MFD40). O tratamento com metilfenidato causou alteração do peso corporal que pode indicar efeitos tóxicos causados pelo uso prolongado do medicamento. O diâmetro do túbulo seminífero e o diâmetro do lume não sofreram alterações com o tratamento. Em relação ao percentual e volume dos componentes tubulares, houve aumento na porcentagem de túbulo e epitélio, porém os percentuais de lume e túnica própria diminuíram. Os volumes de túbulo, túnica própria e lume reduziram nos grupos tratados. Neste estudo não foram observadas alterações patológicas significativas, mantendo inalterada a microestrutura tecidual. Houve reduções nos percentuais de espaço linfático, tecido conjuntivo e macrófagos, com consequente redução no percentual de intertúbulo. O tratamento com metilfenidato durante a fase adulta diminuiu as concentrações plasmáticas da testosterona nos grupos tratados e comprometeu o processo espermatogênico levando à redução no número de espermátides e espermatozoides, na produção espermática e no tempo de trânsito espermático no epidídimo. Dessa forma, pudemos comprovar que o metilfenidato pode prejudicar a fertilidade masculina. Methylphenidate is a stimulant that acts in the Central Nervous System, possessing actions and structure similar to amphetamines. It was initially used to treat fatigue. Today is a well-known medicine in the market to treat Attention Deficit Hyperactivity Disorder (ADHD). In individuals with ADHD, the neurotransmitters dopamine and noradrenaline are deficient in both quantity and function. Thus, the mechanism of action of methylphenidate is to block the dopamine and noradrenaline transporters. Based on these data, the objective was to evaluate the effects of two doses of methylphenidate on the testes of adult mice through morphological, histometric and hormonal evaluations. Twenty four Balb/c mice were randomly divided into three experimental groups (n = 8): Group 01: control (distilled water); Group 02: methylphenidate hydrochloride at a dosage of 20 mg/kg/day (MFD20); Group 03: methylphenidate hydrochloride at a dosage of 40 mg/kg/day (MFD40). Treatment with methylphenidate has caused a change in body weight that may indicate toxic effects caused by prolonged use of the drug. The diameter of the seminiferous tubule and lumen diameter did not change with treatment. In relation to the percentage and volume of the tubular components, there was an increase in the tubule and epithelium percentage, but the percentage of lumen and proper tunic decreased. The volume of tubule, proper tunic and lumen reduced in the treated groups. In this study, no significant pathological changes were observed, maintaining the tissue microstructure unchanged. There were reductions in the percentages of lymphatic space, connective tissue and macrophages, with consequent reduction in intertubule percentage. Methylphenidate treatment during adulthood decreased plasma testosterone concentrations in the treated groups and compromised the spermatogenic process leading to a reduction in sperm and sperm count, sperm production and sperm transit time in the epididymis. Thus, we could prove that methylphenidate can impair male fertility.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior Nas últimas décadas a exposição dos seres vivos a tóxicos ambientais, como os metais pesados, vem se tornando uma prática comum. Metais como o arsênio, cádmio, chumbo, cromo VI e níquel são elementos químicos instáveis que podem causar efeitos adversos ao organismo. Nos testículos a exposição a esses metais pode causar redução do peso testicular, estresse oxidativo, alterações histológicas e hormonais, degeneração dos túbulos seminíferos, apoptose e necrose testicular, além de desencadear perda da motilidade, viabilidade e morfologia espermática. Este estudo teve como objetivo avaliar a ação dos metais pesados arsenato, arsenito, cádmio, chumbo, cromo e níquel sobre parâmetros reprodutivos masculinos a fim de estabelecer uma ordem de toxicidade. Os animais foram distribuídos aleatoriamente em 7 grupos experimentais (n=10 animais/grupo), pesados e mantidos em gaiolas coletivas. Assim, o grupo 1 recebeu 0,7mL de solução salina 0,9% (controle), o grupo 2 recebeu arsenato, o grupo 3 arsenito, o grupo 4 cádmio, o grupo 5 chumbo, o grupo 6 cromo VI e o grupo 7 níquel, todos na dose de 1,5 mg/Kg de peso corporal Os metais foram administrados por via intraperitoneal (IP), em dose única e os animais foram eutanasiados sete dias após a aplicação. Os animais tratados com níquel apresentaram maiores danos toxicológicos como histopatologias, dano celular inicial no túbulo seminífero e alterações espermáticas. O Cádmio induziu patologias leves, gerando dano inicial e morte celular no túbulo, além de alterações morfológicas e menor concentração de espermatozoides íntegros. O Cromo VI causou aumento da testosterona plasmática, redução da morfologia espermática normal e diminuição de espermatozoides com membrana íntegra, associado ao aumento de espermatozoides lesados. Os animais tratados com chumbo também apresentaram patologias leves e alterações nas células já diferenciadas ou em final de diferenciação. Adicionalmente, o arsenito também induziu redução no número de espermátides e de espermatozoides, alterações nas células de Leydig e diminuição na integridade de membrana espermática com maior intensidade que o arsenato, sendo assim considerado mais tóxico. As alterações funcionais, celulares e histológicas encontradas neste estudo, podem ser uma resposta inicial ao estresse oxidativo induzido pela intoxicação por metais pesados. In recent decades the exposure of living beings to environmental toxins, such as heavy metals, has become a common practice. Metals such as arsenic, cadmium, lead, chromium VI, and nickel are unstable chemicals that can cause adverse effects to the body. In the testis, the exposure to these metals can cause testicular weight reduction, oxidative stress, histological and hormonal changes, degeneration of seminiferous tubules, testicular apoptosis and necrosis, in addition to triggering loss of motility, viability and sperm morphology. This study aimed to evaluate the action of heavy metals arsenate, arsenite, cadmium, lead, chromium, and nickel on the male reproductive parameters for the purpose of establish an order of toxicity. The animals were randomly divided into 7 experimental groups (n = 10 animals/group), weighed and kept in collective cages. Group 1 received 0.7 ml of 0.9% saline solution (control), group 2 received arsenate, group 3 arsenite, group 4 cadmium, group 5 lead, group 6 chromium VI, and Group 7 nickel, all at the dose of 1.5 mg/Kg of body weight. The metals were administered intraperitoneally (i.p.), single dose, and the animals were euthanized seven days after application. Animals treated with nickel presented greater toxicological damages such as pathologies, initial cell damage in the seminiferous tubule and sperm changes. Cadmium induced pathologies, generating initial damage and cell death in the tubule, in addition to morphological changes and lower concentration of intact sperm. Chromium VI caused an increase in plasma testosterone, reduction in normal sperm morphology, and reduction of sperm with intact membrane, associated with an increase in damaged sperm. Animals treated with lead also showed slight pathologies and alterations in differentiated cells or at the end of differentiation. Additionally, arsenite also induced reduction in the number of spermatids and sperm, changes in Leydig cells and decrease in sperm membrane integrity with a higher intensity than arsenate, and it was considered more toxic. The functional, cellular and histological changes found in this study may be an initial response to oxidative stress induced by heavy metal poisoning.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior O gênero Renealmia pertence à família Zingiberaceae e abrange um conjunto de espécies dadas como raras e ameaçadas de extinção. A espécie Renealmia pycnostachys é representada apenas pelo espécime tipo coletado no Sudeste de Minas Gerais e é utilizada na cultura popular como afrodisíaco. Baseado nessas informações, tivemos como objetivo avaliar o efeito do extrato hidroalcoólico da folha de Renealmia pycnostachys sobre as porções gametogênica e androgênica testicular, nos corpos cavernosos do pênis e nos níveis de testosterona sérica. Camundongos Swiss adultos (n=28) foram divididos em 4 grupos: Controle (água) e 3 grupos recebendo doses diárias do extrato da folha (100, 200 e 400 mg/Kg) por 42 dias. Observamos que o extrato não alterou os parâmetros biométricos, corporal e testicular, percentual e volume dos componentes tubulares e índices tubulossomático e epiteliossomático. Nos grupos tratados com o extrato da folha tivemos aumentos do diâmetro tubular, altura do epitélio seminífero, diâmetro de lume, área de túbulo, epitélio e lume e redução no comprimento de túbulo seminífero total. Verificamos aumento no número corrigido de células de Sertoli por grama de testículo, enquanto que a capacidade suporte diminuiu levando a uma redução na produção espermática diária por testículo e por grama de testículo. Somente a histopatologia leve do túbulo seminífero foi observado. Verificamos no intertúbulo aumento no percentual do citoplasma de Leydig e de células de Leydig, além de redução no percentual de tecido conjuntivo. Houve aumento de gotículas de lipídeos no citoplasma das células de Leydig. Observamos o aumento da testosterona sérica, óxido nítrico peniano e corpos cavernosos no pênis. Podemos comprovar que a folha de Renealmia pycnostachys apresenta efeito afrodisíaco sem causar danos à microestrutura testicular ou desequilíbrio redox. The Renealmia genus belongs to the Zingiberaceae family and covers a number of rare and endangered species. The species Renealmia pycnostachys is represented only by the type of collective specimen in the Southeast of Minas Gerais and is used in the popular culture as an aphrodisiac. Based on these informations, we aimed to evaluate the effect of hydroalcoholic extract of the leaf of Renealmia pycnostachys on the gametogenic and androgenic portions of the testicular, cavernous bodies of the penis and serum testosterone levels. Adult Swiss mice (n = 28) were divided into 4 groups: control (water) and three daily higher doses of leaf extract (100, 200 and 400 mg / kg) for 42 days. We observed that the extract did not alter the biometric parameters, body and testicular, percentage and volume of tubular components and tubulesomatic and epitheliosomatic indexes. The groups treated with the leaf extract had increases in the tubular diameter, height of the seminiferous epithelium, lumen diameter, tubule area, epithelium and lume and reduction in the length of the total seminiferous tubule. We found an increase in the corrected number of Sertoli cells per gram of testis, while the support capacity decreased, leading to a reduction in daily sperm production per testis and gram of testis. Only the mild histopathology of the seminiferous tubule was observed. We verified in the intertubule an increase in the percentage of the Leydig cytoplasm and Leydig cells, in addition to a reduction in the percentage of connective tissue. There was an increase in lipid droplets in the Leydig cell cytoplasm. We observed increased serum testosterone, penile nitric oxide, and cavernous bodies in the penis. We can verify that the leaf of Renealmia pycnostachys presents aphrodisiac effect without causing damage to the testicular microstructure or redox imbalance.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior O Cádmio (Cd) é um metal pesado, cuja contaminação na água e solo é bem conhecida. O Cd interfere no metabolismo de vários órgãos, particularmente nos testículos, onde é capaz de romper a barreira hematotesticular, com apoptose das células germinativas e perda do epitélio. Alteração nas células de Leydig e redução na concentração sérica de testosterona também refletem ação do Cd. O Cd pode competir com minerais essenciais e com isso causar o desbalanço destes elementos no testículo. Além disso, o estresse oxidativo é um dos mecanismos de toxicidade do metal. Dessa forma, vários estudos têm-se voltado para a descoberta de produtos naturais com ação antioxidante frente ao Cd. Dentre as plantas com potencial antioxidante destaca-se o açaí da Amazônia (Euterpe oleracea). Seu fruto é conhecido por sua atividade antioxidante devido a presença de compostos fenólicos, destacando-se as antocianinas, e de ácidos graxos presentes na fração lipídica. Assim, neste trabalho inicialmente foi comparada a toxicidade do Cd por diferentes vias e forma de administração, para posteriormente ser feita a escolha da via na avaliação dos efeitos do óleo e da polpa desengordurada rica em antocianinas (PDRA) de E. oleracea. Camundongos Swiss receberam CdCl 2 nos modos seguintes: 1,5 mg/Kg intraperitoneal (i.p.) dose única; 30 mg/Kg via oral dose única e 4,28 mg/Kg via oral durante 7 dias. No oitavo dia os animais foram eutanasiados. O Cd bioacumulou em todas as vias, sendo maior na oral fracionada. As concentrações de Ca e Cu testicular reduziram em todos os animais expostos ao Cd, enquanto que Zn e Mn reduziram apenas na via i.p.. A atividade de SOD reduziu em ambas formas de administração via oral, CAT aumentou na via i.p., Glutationa aumentou em todos os animais intoxicados. A concentração sérica de testosterona reduziu em ambas administrações via oral. Danos tubulares foram mais intensos naqueles que receberam o metal via oral fracionado. Assim, quanto o Cd é ofertado de forma fracionada via oral, as alterações no epitélio germinativo são mais intensas, sendo esta forma de administração escolhida para se testar os efeitos do fruto de E. oleracea. Camundongos Swiss receberam solução aquosa de CdCl 2 na concentração de 4,28 mg/Kg por via oral durante 7 dias. Após este tratamento, os animais foram separados em grupos e tratados com óleo de E. oleracea nas doses de 50, 100 e 150 mg/Kg e com PDRA nas doses de 100, 200 e 300 mg/Kg por 42 dias. Parâmetros morfométricos tubulares e de Leydig foram recuperados após o tratamento com o óleo associado a aumento na concentração sérica de testosterona e da viabilidade celular. O tratamento com PDRA levou melhoria nestes parâmetros. O tratamento com o óleo preveniu a progressão dos danos na morfologia tubular causados pelo Cd, enquanto que a PDRA apenas de danos leves. A concentração de Mn e Zn testiculares, aumentaram nos animais tratados com o óleo, além da estimulação na atividade de SOD e CAT. A PDRA causou aumento na concentração de Zn e Mg na concentração de 300 mg/Kg. Cu reduziu em todos os tratamentos e Fe naqueles que receberam 100 mg/Kg de PDRA. O tratamento com PDRA aumentou a atividade de SOD e a concentração de 300 mg/Kg reduziu a peroxidação lipídica. Assim, conclui- se que o tratamento com o óleo de E. oleracea apresentou atividade antioxidante curativa contra alterações testiculares induzidas pelo Cd, enquanto a PDRA, nas doses testadas, ameniza os danos testiculares causados pelo Cd, sendo considerada com potencial antioxidante. Cadmium (Cd) is a heavy metal whose contamination in water and soil is well known. Cd interferes in the metabolism of several organs, particularly the testes, where it is able to breakdown the hematotesticular barrier, with apoptosis of germinative cells and loss of the epithelium. Alteration in Leydig cells and reduction in serum testosterone levels also reflect actions of Cd. The Cd can compete with essential minerals and thereby promote the unbalance of these elements in the testicle. In addition, oxidative stress is one of the metal's toxicity mechanisms. Thus, several studies have focused on the discovery of natural products with antioxidant action against Cd. The Amazonian palm (Euterpe oleracea) stands out among the plants with antioxidant potential. Its fruit is known for its antioxidant activity due to the presence of phenolic compounds, especially anthocyanins, and fatty acids present in the lipid fraction. Thus, in this work, the toxicity of Cd was compared by different routes and forms of administration, for the subsequent selection of the route in the evaluation of the effects of the oil and the defatted pulp rich in anthocyanins (DPRA) of E. oleracea. Swiss mice received CdCl 2 in the following ways: 1.5 mg/kg intraperitoneal (i.p.) route single dose; 30 mg/kg oral route single dose; and 4.28 mg/kg oral route for 7 days. On the eighth day the animals were euthanized. The Cd bioaccumulated in all routes, being greater in the fractional oral dose. Ca and Cu testicular concentrations reduced in all animals exposed to Cd, whereas Zn and Mn reduced only in the i.p. route. SOD activity reduced in both forms of oral administration, whereas CAT increased in i.p. route and glutathione increased in all intoxicated animals. Serum testosterone levels decreased in both forms of oral administration. Tubular damages were more intense in the animals that received the fractioned oral metal. Thus, when Cd is fractionally offered orally, changes in the germinal epithelium are more intense, and this form of administration was chosen to test the effects of E. oleracea fruit. Swiss mice received aqueous solution of CdCl 2 at a concentration of 4.28 mg/kg orally for 7 days. After this treatment, the animals were separated into groups and treated with E. oleracea oil at doses of 50, 100 and 150 mg/kg, and with DPRA at doses of 100, 200 and 300 mg/kg for 42 days. Tubular and Leydig morphometric parameters were recovery after the treatment with the oil associated with an increase in serum testosterone level and cell viability. Treatment with DPRA led to improvement in these parameters. The treatment with oil prevented the progression of damage in the tubular morphology caused by Cd, whereas DPRA prevented only slight damages. The concentration of testicular Mn and Zn increased in animals treated with oil, in addition to stimulation in SOD and CAT activity. DPRA promoted an increase in the concentration of Zn and Mg at 300 mg/kg dose. Cu reduced in all treatments and Fe in those who received 100 mg/kg of DPRA. Treatment with DPRA increased SOD activity and the concentration of 300 mg/kg reduced lipid peroxidation. Thus, the treatment with E. oleracea oil presented curative antioxidant activity against Cd-induced testicular alterations, while DPRA, at tested doses, attenuated the testicular damage caused by Cd, being considered with antioxidant potential.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior Plantas medicinais dominam a farmacologia por proverem várias fontes de remédios e assim, a mesma espécie de planta pode ter compostos que melhoram e compostos que prejudicam a espermatogênese. Dianthus caryophyllus apresenta ação antibiótica, antifúngica, antiviral, antioxidativa, sedativa e tranquilizante. Teve ação no controle da pressão arterial em ratos e possível efeito antiurolítico, porém nenhum estudo foi feito para avaliar seus efeitos na reprodução. Assim, esse estudo visou avaliar o efeito do extrato aquoso da flor de Dianthus caryophyllus (EADC) nos testículos de camundongos Balb/c, além da resposta de enzimas antioxidante à planta. Quarenta e oito animais foram distribuídos aleatoriamente em seis grupos experimentais (n=8): G1 (Controle), G2 (Viagra), G3, G4, G5 receberam diariamente 10, 20, 40 mg/kg de EADC, respectivamente, e o G6 recebeu EADC 40mg/kg em dias alternados. Foram realizadas análises morfométricas e estereológicas do parênquima testicular, avaliação de alterações estruturais do tecido, atividade das enzimas catalase (CAT), superóxido dismutase (SOD) e glutationa S-transferase (GST), além da concentração de malondialdeido (MDA) e de óxido nítrico (ON) nos testículos. Houve redução do diâmetro, proporção, volume e área do lúmem tubular, aumento da proporção e área de epitélio seminífero em todos os grupos tratados com a planta, sendo que o diâmetro tubular reduziu apenas em G5. Houve redução da proporção de túbulos seminíferos normais e aumento de vacúolos em todos os grupos. No intertúbulo houve aumento de tecido conjuntivo em G5. A proporção de células de Leydig aumentou apenas em G6, assim como o volume das células de Leydig por grama de testículo no mesmo grupo. O número de células de Leydig por testículo aumentou apenas em G6, enquanto por grama de testículo houve aumento em G5 e G6. Quanto à volumetria dos componentes, houve aumento do volume de tecido conjuntivo em G4, G5 e G6. Houve aumento de CAT em G5 e G6 e de MDA em todos os grupos tratados. Pode-se inferir que houve aumento da produção espermática em todos os grupos, no intertúbulo nota-se mais efeitos em Leydig e tecido conjuntivo principalmente nos grupos G5 e G6 e que a planta gera estresse oxidativo. Medicinal plants dominate pharmacology by providing various sources of medicines and thus, the same plant species may have components that improve and impair spermatogenesis. Dianthus caryophyllus has antibiotic, antifungal, antiviral, antioxidative, sedative and tranquilizing actions. It had action in controlling blood pressure in rats and possible antiurolithic effects, but no study was done to evaluate its effects on reproduction. The aim of this study was evaluate the effect of the aqueous extract of the Dianthus caryophyllus flowers (AEDC) on the testis of Balb/c mice and the antioxidant enzyme response to the plant. 48 animals were randomly distributed in six experimental groups (n = 8): G1 (Control), G2 (Viagra), G3, G4, G5 received daily 10, 20, 40 mg / kg AEDC respectively and G6 40 G / AEDC 40mg / kg every other day. Morphometric and stereological analyzes of the testicular parenchyma, evaluation of tissue structural alterations, activity of catalase (CAT), superoxide dismutase (SOD) and glutathione S-transferase (GST), and malondialdehyde (MDA) and oxide nitric oxide (ON) in the testicles. There was a reduction in the diameter, proportion, volume and area of the tubular lumen, increase in the proportion and area of seminiferous epithelium in all groups treated with the plant, and reduction of tubular diameter only in G5. There was a reduction in the proportion of normal seminiferous tubules and increase of vacuoles in all groups. In the intertubular compartment there was reduction in the proportion of lymphatic space as well as increase of connective tissue in all the treated groups. The proportion of Leydig cells increased only in G6, as did the volume of Leydig cells per gram of testis in the same group. The number of Leydig cells per testis increased only in G5, while per gram of testis there was increase in G5 and G6. In the volumetry of the components, there was an increase of only connective tissue in G4, G5 and G6. There was an increase in CAT in G5 and G6 and in MDA in all treated groups. It can be concluded that there was an increase in sperm production in all groups, in the intertubular compartment there were more effects in Leydig and connective tissue, especially in the G5 and G6 groups, and that the plant generates oxidative stress.

Fundação de Amparo à Pesquisa do Estado de Minas Gerais As folhas e raízes de Pfaffia glomerata são usadas como afrodisíacos, tranquilizantes e antirreumáticos. Devido à ausência de dados científicos experimentais faz-se necessário estudos que identifiquem suas propriedades medicinais. Baseado nisso objetivou-se avaliar o efeito de extrato hidroalcoólico da raiz de P. glomerata (BGE) sobre o parênquima testicular, espermáticos e fertilidade. Camundongos adultos (n=60) foram divididos em controle (água), Citrato de sildenafila (7mg/kg) e 3 grupos recebendo doses diárias (100, 200 e 400 BGE mg/Kg) e uma dose intermitente (200 BGE mg/Kg a cada três dias) por 42 dias. Após a ingestão do extrato, houve redução na proporção de túbulo e epitélio seminíferos, aumento do volume de túnica própria e redução do número de células de Sertoli, sem prejuízo a produção espermática. Entretanto, o BGE promoveu morte celular e alteração na frequência de estádio dos túbulos, além do aumento de óxido nítrico. Verificou-se também aumento do percentual e volume de vasos sanguíneos e espaço linfático, redução da testosterona, aumento do estradiol e morte de células de Leydig. Observou-se aumento de corpos cavernosos, aumento de colágeno, redução do músculo liso e aumento do óxido nítrico peniano. O tratamento causou ainda redução no número e viabilidade de espermatozoides, estes apresentavam altos índices de patologias, como espaços vacuolares, degeneração e descamação. Baixo percentual de espermatozoides com membrana íntegra e alterações na motilidade, leva ainda a perdas embrionárias. O princípio de ação do extrato baseia-se na produção de NO, atuando sobre os vasos sanguíneos aumentando os corpos cavernosos. O tratamento leva ainda redução do músculo liso peniano, danos nas células de Leydig, acumulo de gotículas de lipídios no seu citoplasma levando, inclusive, à morte da célula de Leydig e o aumento do NO. O extrato atua também aumentando ROS, culminando com danos de DNA, que leva a perdas embrionárias. Porém embora o quadro leve a aparente redução da atividade sexual, o aumento de estradiol e colágeno é uma forma de compensação e assim da manutenção da atividade sexual. Assim, o ginseng brasileiro tem ação afrodisíaca, porém com efeitos colaterais na produção de testosterona e na viabilidade das células germinativas. The leaves and roots of Pfaffia glomerata are used as aphrodisiacs, tranquillizers and antirheumatics. Due to the lack of experimental scientific data, studies are necessary to identify its medicinal properties. Based on this the objective was to evaluate the effect of hydroalcoholic extract of the P. glomerata root (BGE) on the testicular parenchyma, spermatozoa and fertility. Adult mice (n = 60) were divided into control (water), sildenafil Citrate (7mg / kg) and 3 groups receiving daily doses (100, 200 and 400 BGE mg / kg) and an intermittent dose (200 BGE mg / kg every three days) for 42 days. After ingestion of the extract, there was a reduction in the proportion of seminiferous tubule and epithelium, increase of the own tunica volume and reduction of the number of Sertoli cells, without prejudice to sperm production. However, BGE promoted cell death and a change in tubule stage frequency, in addition to an increase in nitric oxide. There was also an increase in the percentage and volume of blood vessels and lymphatic space, reduction of testosterone, increase of estradiol and death of Leydig cells. There was an increase of cavernous bodies, increase of collagen, reduction of smooth muscle and increase of penile nitric oxide. The treatment also caused a reduction in the number and viability of spermatozoa, which presented high rates of pathologies, such as vacuolar spaces, degeneration and desquamation. Low percentage of spermatozoa with whole membrane and alterations in the motility, still leads to embryonic losses. The principle of action of the extract is based on the production of NO, acting on the blood vessels increasing the cavernous bodies. The treatment also leads to reduction of the penis smooth muscle, Leydig cell damage, accumulation of lipid droplets in its cytoplasm leading to Leydig cell death and increased NO. The extract also works by increasing ROS, culminating with DNA damage, leading to embryonic losses. However, although the picture leads to the apparent reduction of sexual activity, the increase of estradiol and collagen is a form of compensation and thus the maintenance of sexual activity. Thus, Brazilian ginseng has an aphrodisiac effect, but with side effects on testosterone production and germ cell viability.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior O uso indiscriminado de drogas vegetais ou alopáticas com finalidades afrodisíacas é extremamente temerário, visto que muitas delas apresentam substâncias que podem desencadear reações adversas, seja por seus próprios componentes, seja pela presença de contaminantes ou adulterantes presentes em suas preparações. Tynanthus fasciculatus é uma espécie utilizada popularmente para diversos fins terapêuticos, contra indigestão, dores de estômago, diarreia, controle de vermes e também como afrodisíaco. O presente trabalho aborda aspectos da reprodução animal, com enfoque na histomorfometria testicular, respostas antioxidantes dos testículos, níveis séricos de testosterona e os eventos que ocorrem na espermatogênese sob influência do extrato alcoólico obtido da espécie vegetal T. fasciculatus. Foram utilizados 40 animais distribuídos em quatro grupos (n=10): grupo 1 (G1) controle; grupos 2 (G2), 3 (G3) e 4 (G4) receberam 100, 200 e 300mg de EATF por kg de peso corporal e tratados por 42 dias consecutivos via gavagem. O extrato de T. fasciculatus foi submetido à prospecção fitoquímica e revelou a presença de flavonoides, taninos, triterpenos/esteroides, cumarinas e saponinas. Houve aumento do peso testicular e dos índices gonadossomático, parenquimossomático, tubulossomático e epiteliosomático, assim como nos volúmens de túbulo, epitélio e túnica própria nos grupos EATF100 e EATF200. Nestes grupos também aumentaram o comprimento total dos túbulos seminíferos por testículo e a produção espermática diária. Foi observada redução na capacidade de suporte total de células de Sertoli nos grupos EATF200 e EATF300. Houve aumento nos números corrigidos de espermatócitos primários em preleptóteno/leptóteno e paquíteno, e, proporção volumétrica de célula Leydig no EATF100, ao passo que, espermátide arredondada e os níveis séricos de testosterona aumentaram no EATF100 e 200. Por outro lado, o EATF300 apresentou alterações patológicas na região tubular e peroxidação lipídica, marcada pelo aumento de malondialdeido. Pode-se inferir que o EATF100 e EATF200 promoveram aumento significativo no peso testicular e nos parâmetros tubulares, especialmente aqueles relacionados ao epitélio seminífero, sem comprometimento da espermatogênese. Contudo, o tratamento com EATF300, apresentou alterações no microambiente, evidenciadas pelas alterações patológicas tubulares e aumento de MDA. Diante disso, podemos concluir que as dosagens menores do EATF são mais seguras e eficazes para os parâmetros morfológicos e/ou morfométricos, principalmente sobre o parênquima testicular. The indiscriminate use of vegetable or allopathic drugs for aphrodisiac purposes is extremely reckless, since many of them present substances that can trigger adverse reactions, either by its own components or by the presence of contaminants or adulterants present in its preparations. Tynanthus fasciculatus is a species used popularly for various therapeutic purposes, such as indigestion, stomach pains, diarrhea, control of worms and also as an aphrodisiac. The present work deals with aspects of animal reproduction, focusing on testicular histomorphometry, antioxidant responses of the testis, serum testosterone levels and the events that occur in spermatogenesis under the influence of the alcoholic extract obtained from the plant species T. fasciculatus. Forty animals were distributed into four groups (n = 10): group 1 (G1) control; Groups 2 (G2), 3 (G3) and 4 (G4) received 100, 200 and 300 mg of EATF per kg of body weight and treated for 42 consecutive days via gavage. The extract of T. fasciculatus was submitted to phytochemical prospection and revealed the presence of flavonoids, tannins, triterpenes/steroids, coumarins and saponins. There was an increase in testicular weight and the gonadosomatic, parenchymosomatic, tubulesomatic and epitheliosomatic indices, as well as in the tubule, epithelium and tunica propria volumes in the EATF100 and EATF200 groups. In these groups also increased the total length of the seminiferous tubules by testis and the daily sperm production. Reduction in the total support capacity of Sertoli cells in the EATF200 and EATF300 groups was observed. There was an increase in the corrected numbers of primary spermatocytes in preleptotene/leptotene and pachytene, and the Leydig cell volumetric ratio in EATF100, while, rounded spermatid and serum testosterone levels increased in EATF100 and 200. EATF300 presented pathological alterations in the tubular region and lipid peroxidation, marked by the increase of malondialdehyde. The results obtained allow us to infer that EATF100 and EATF200 promoted a significant increase in testicular weight and tubular parameters, especially those related to the seminiferous epithelium, without compromising spermatogenesis. However, treatment with EATF300 presented alterations in the microenvironment, evidenced by tubular pathological alterations and increased MDA. Therefore, we can conclude that the lower dosages of EATF are safer and more effective for morphological and / or morphometric parameters, especially on the testicular parenchyma.

The rodents are the most diverse order of mammals. They are distributed worldwide, from arctic tundra to deserts. However, little is known about the reproduction of wild species, since rodents, as well as other mammals, are important parts in the dynamics of the forests they inhabit. Thus, the objective of the present study was to describe the process of spermatogenesis of the wild rodents Oxymycterus rufus and Oxymycterus nasutus in order to provide relevant information on testicular activity of these species. Five adult specimens of O. rufus were collected in an Atlantic Forest fragment, whereas 10 adult O. nasutus were collected in a Pampa biome. Euthanasia was performed by anesthetic overdose and testicular fragments were collected, fixed and routinely processed, being analyzed under light and transmission electron microscopy. The body weight of O. rufus and O. nasutus were 82.28 and 58.30 g, respectively, whereas 0.47 and 0.89 % of these were allocated into testes and 0.40 and 0.82 % into seminiferous tubules. The proportions of seminiferous epithelium were 0.35 to 0.56 %, respectively. The length of the seminiferous tubules were 34.93 and 18.62m/g, for O. rufus and O. nasutus, respectively, and the stages of the seminiferous epithelium cycle followed the pattern described for other wild rodents. For O. rufus, the duration of the seminiferous epithelium cycle was determined, being each cycle corresponding to 6.58 days while all the spermatogenic process lasted 29.61 days. The number of Sertoli cells per gram of testis was 232.40 x10 6 and 91.02 x10 6 in O. rufus and O. nasutus respectively, and the daily sperm reserve was 962 x10 6 and 849.64 x10 6 , respectively. The interstitium occupied 9.09 and 4.81 % of the testes of O. rufus and O. nasutus, respectively, and followed the type I organization pattern described by Fawcett. The Leydig cells were the most abundant element in the interstitium (117.23 x10 6 per gram of testis in O. rufus and 40.77 x10 6 per gram of testis in O. nasutus), and the leydigsomatic indexes were 0.018 and 0.019 % in O. rufus and O. nasutus, respectively. Therefore, we concluded that O. rufus and O. nasutus show high efficiency of spermatogenesis with high proportion of seminiferous tubules in the testicular parenchyma, abundant Sertoli cells per gram of xitestis and high sperm reserve, thus demonstrating high energy investment in sperm production, which is compatible with animals displaying promiscuous mating system. Os roedores constituem a ordem mais diversificada dos mamíferos e apresentam ampla distribuição mundial sendo encontrados em todos os habitats, desde a tundra ártica aos desertos. Porém, pouco se sabe ainda sobre a reprodução de espécies silvestres, o que justifica a realização deste estudo, visto que roedores, assim como outros mamíferos são peças importantes na dinâmica das florestas nas quais habitam. Assim, objetivou-se descrever o testículo e o processo espermatogênico dos roedores silvestres Oxymycterus rufus e Oxymycterus nasutus, visando a fornecer informações relevantes sobre a atividade testicular destas espécies. Cinco espécimes de O. rufus foram coletados em fragmento de Mata Atlântica em Minas Gerais, e 10 exemplares de O. nasutus no bioma Pampa, no Rio Grande do Sul, sendo todos os exemplares machos e sexualmente maduros. A eutanásia foi realizada por meio de superdosagem anestésica e os fragmentos testiculares foram coletados, fixados e processados rotineiramente, sendo destinados às análises em microscopia de luz e eletrônica de transmissão. O peso corporal de O. rufus e O. nasutus foi, respectivamente, 82,28 e 58,30 g, sendo que 0,47 e 0,89 % destes foram alocados em testículos, 0,40 e 0,82 % em túbulos seminíferos e 0,35 e 0,56 % alocados em epitélio seminífero. O comprimento dos túbulos foi de 34,93 e 18,62 metros por grama de testículo, respectivamente. Os estádios do ciclo do epitélio seminífero seguiram o padrão descrito para outros roedores silvestres. A duração do ciclo do epitélio seminífero foi determinada em O. rufus, sendo cada ciclo do epitélio seminífero correspondente a 6,58 dias enquanto todo o processo espermatogênico teve duração de 29,61 dias. O número de células de Sertoli por grama de testículo foi de 232,40 x10 6 em O. rufus e 91,02 x10 6 em O. nasutus e a reserva espermática por grama de testículo foi em média 962 x10 6 e 849,64 x10 6 espermátides, respectivamente. O compartimento intertubular ocupou 9,09 e 4,81 % dos testículos de O. rufus e O. nasutus, respectivamente. A organização dos componentes do compartimento intertubular corresponde ao padrão tipo I descrito por Fawcett. As células de Leydig foram o elemento mais abundante no compartimento intertubular totalizando 117,23 x10 6 e 40,77 x10 6 células por grama de testículo em O. rufus e O. nasutus, respectivamente. O índice ixLeydigossomático foi 0,018 e 0,019 % em O. rufus e O. nasutus, respectivamente. Conclui-se que O. rufus e O. nasutus apresentam alta eficiência do processo espermatogênico, com elevada proporção de túbulos seminíferos no parênquima testicular, grande quantidade de células de Sertoli por grama de testículo e elevada reserva espermática testicular, demonstrando assim alto investimento energético no compartimento tubular e, consequentemente, na produção de espermatozoides. Esse padrão é compatível com animais que apresentam sistema de acasalamento promíscuo.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior Hemidactylus mabouia is the gecko species more thoroughly distributed in Brazil, being common in anthropic atmospheres, what turns easy its use as model for study of the reptiles. Likewise, aiming at to describe morphologically the alimentary canal of H. mabouia, 17 adult specimens were used, being collected fragments of the esophagus, of the oesophagogastric transition, of the stomach and of the small and large intestines, that were processed according to routine methods for anatomical, histological, histochemical and ultra-structural analyses. Histological sections were stained with Toluidine Blue or submitted to techniques for identification of glycoconjugates, proteins, alkaline phosphatase activity, mitochondria and argyrophil and argentaffin endocrine cells. The esophagus of H. mabouia is a tubular straight- lined organ, with the cranial portion more dilated, what favours the ingestion of whole preys and contributes to a rapid clearance of the bucco-pharyngeal cavity. The stomach has "J" form, with a long fundic region, which can be subdivided in oral fundic and aboral fundic, and a short pyloric region. The presence of macrophages in the gastric surface constitutes singular fact and reflects the defense need against invader microorganisms. As the one of the other carnivorous reptiles, the intestine of H. mabouia is short, being the small intestine more long and convoluted that the large intestine, that possesses a quite dilated colon followed for a short rectum. The oesophageal covering epithelium is pseudostratified with cells that secrete neutral and acid mucins; at the oesophagogastric transition there is a mixture of this epithelium with mucussecreting prismatic simple epithelium, whose cells secrete neutral mucins predominantly; in the stomach it is prismatic simple secreting neutral mucins; in the small intestine it is prismatic simple with absorptive cells and mucous cells secreting acid and neutral mucins; in the large intestine the absorptive cells are scarce, without brush border or with very short brush border, and the mucus-secreting cells are abundant. In the esophagus, the lamina propria is thin and aglandular; in the esophagus-gastric transition it becomes thick and filled by branched simple acinous glands, with mucous cells secreting neutral mucins and zymogenic cells producing of pepsinogen. Those multicellular glands were not observed in several reptiles, but they were described in many anuran amphibians, what can be related with the food habit of those animals. In the stomach, the oral fundic region presents long tubularacinous ramified simple glands, that become smaller, less ramified and more tubular in the aboral fundic region, and they are short tubular simple in the pyloric region. The fundic glands possess mucous cells secreting neutral mucins and oxynticopeptic cells, which differ of the oral fundic for aboral fundic in relation to the density of zymogen granules and mitochondria, indicating the existence of a secretion gradient of pepsinogen and hydrochloric acid. The pyloric glands just possess mucous cells secreting neutral mucins and argyrophil and argentaffin endocrine cells, which were also observed dispersed in the coating epithelium and in the glands of other segments of the alimentary canal, except in the esophagus. Surprisingly, argentaffin but non argyrophil cells were located in the oesophagogastric transition. In the intestine there are neither villi nor Lieberkühn crypts. Hemidactylus mabouia é a espécie de lagartixa mais amplamente distribuída no Brasil, sendo comum em ambientes antrópicos, o que torna fácil seu uso como modelo para estudo dos répteis. Assim, visando descrever morfologicamente o tubo digestivo de H. mabouia, foram utilizados 17 exemplares adultos, sendo coletados fragmentos do esôfago, da transição esôfago-gástrica, do estômago e dos intestinos delgado e grosso, que foram processados conforme métodos rotineiros para análises anatômica, histológica, histoquímica e ultra-estrutural. Secções histológicas foram coradas com azul de toluidina ou submetidas a técnicas para detecção de glicoconjugados, proteínas, atividade da fosfatase alcalina, mitocôncrias e células endócrinas argirófilas e argentafins. O esôfago de H. mabouia é um órgão tubular retilíneo, com a porção cranial mais dilatada, o que facilita a deglutição de presas inteiras e concorre para a rápida desobstrução da cavidade bucofaringeana. O estômago tem forma de J , com uma extensa região fúndica, que pode ser subdividida em fúndica oral e fúndica aboral, e uma curta região pilórica. A presença de macrófagos na superfície gástrica constitui fato singular e reflete a necessidade de defesa no lúmen. Como o dos demais répteis carnívoros, o intestino de H. mabouia é curto, sendo o intestino delgado mais longo e convoluto que o intestino grosso, que possui um cólon bastante dilatado seguido por um reto curto. O epitélio de revestimento esofágico é pseudo- estratificado ciliado com células secretoras de mucinas ácidas e neutras; na transição esôfagogástrica há mistura deste tipo epitelial com o epitélio simples prismático mucossecretor de mucinas predominantemente neutras; no estômago é simples prismático mucossecretor de mucinas neutras; no intestino delgado é simples prismático com células absortivas e células mucossecretoras de mucinas ácidas e neutras; no intestino grosso as células absortivas são escassas, sem borda estriada ou com borda muito reduzida, e as células mucossecretoras são abundantes. No esôfago, a lâmina própria é delgada e aglandular; na transição esôfago-gástrica se torna espessa e preenchida por glândulas acinosas simples ramificadas, com células mucossecretoras de mucinas neutras e células zimogênicas produtoras de pepsinogênio. Essas glândulas multicelulares não foram observadas em vários répteis, mas foram descritas em muitos anfíbios anuros, o que pode estar relacionado com o hábito alimentar desses animais. No estômago, a região fúndica oral apresenta extensas glândulas túbulo-acinosas simples ramificadas, que se tornam menores, menos ramificadas e mais tubulares na fúndica aboral, e são tubulares simples e curtas na pilórica. As glândulas fúndicas possuem células mucossecretoras de mucinas neutras e células oxintopépticas, que diferem da fúndica oral para aboral em relação à densidade de grânulos de zimogênio e mitocôndrias, indicando a existência de um gradiente de secreção de pepsinogênio e ácido clorídrico. As glândulas pilóricas possuem apenas células mucossecretoras de mucinas neutras e células endócrinas argirófilas e argentafins, que também foram observadas dispersas no epitélio de revestimento e nas glândulas de outros segmentos do tubo digestivo, exceto no esôfago. Curiosamente, células argentafins mas não argirófilas foram localizadas na transição esôfago-gástrica. No intestino não há vilosidades nem criptas de Lieberkühn.
