The chirality issues in the essential oils (EOs) of leaves and flowers from Bubonium graveolens were addressed by chiral high-performance liquid chromatography (HPLC) with polarimetric detection and vibrational circular dichroism (VCD). The chemical compositions of the crude oils of three samples were established by gas chromatography / mass spectrometry (GC/MS). The well-known cis-chrysanthenyl acetate ( 1), oxocyclonerolidol ( 2), and the recently disclosed cis-acetyloxychrysanthenyl acetate ( 3), the three major chiral compounds, were isolated by preparative HPLC. The naturally occurring oxocycloneroledol ( 2), mostly found in the leaf oil (49.4-55.6%), presents a (+) sign in the mobile phase during HPLC on a chiral stationary phase (CSP) with a Jasco polarimetric detection. The naturally occurring cis-chrysanthenyl acetate ( 1) and cis-acetyloxychrysanthenyl acetate ( 3), mostly found in the flower EO (35.9-74.9% and 10.0-34.3%, respectively), both present a (−) sign. HPLC on a CSP with polarimetric detection is an unprecedented approach to readily differentiate the flower and leaf EOs according to their chiral signature. The comparison of the experimental and calculated VCD spectra of pure isolated 1, 2, and 3 provided their absolute configuration as being (1 S,5 R,6 S)-(−)-2,7,7-trimethylbicyclo[3.1.1]hept-2-en-6-yl acetate 1, (2 R,6 R)-(+)-6-ethenyl-2,6-dimethyl-2-(4-methylpent-3-en-1-yl)dihydro-2H-pyran-3(4H)-one) 2 and (1 S,5 R,6 R,7 S)-(−)-7-(acetyloxy)-2,6-dimethylbicyclo[3.1.1]hept-2-en-6-yl]methyl acetate 3. Compounds 1, 2, and 3 were already known in B. graveolens but this is the first report of the absolute configuration of (+)- 2 and (−)- 3. The title compound was obtained as a side product during dimerization-oxidation steps of the carbene generated from N-methylbenzothiazolium iodide. Chromatography on ( S, S)-Whelk O1 column showed on cooling a typical plateau shape chromatogram indicating an exchange between two enantiomers on the column. Enantiopure (±)-2,6-di-sec-butyl-4-methylpyridine and its oxidized form (±)-2,6-di-sec-butyl-4-methylpyridine-N-oxide were prepared and then separated by chiral HPLC. Their stereochemical structures and their conformational distribution in solution were investigated using vibrational circular dichroism (VCD) and infrared spectroscopy (IR) combined with density functional theory (DFT). The experimental spectra have been compared with theoretical data. This comparison indicated that (−)-2,6-di-sec-butyl-4-methylpyridine and its oxidized form (+)-2,6-di-sec-butyl-4-methylpyridine-N-oxide have an (S,S)-configuration and exist in several conformations. The good fit confirms the reliability of the conformational analysis. Our results indicated that going from the reduced form to the oxidized one strongly influences the type of conformers in solution. Moreover, DFT calculations showed that for all of the conformers of (S,S)-2,6-di-sec-butyl-4-methylpyridine-N-oxide, the formation of four centered intramolecular hydrogen bonds between the hydrogen of the –CH, –CH2, and –CH3 groups and the nitroxide oxygen is possible. An unprecedented methodology was developed to simultaneously assign the relative percentages of the major chiral compounds and their prevailing enantiomeric form in crude essential oils (EOs). In a first step the infrared (IR) and vibrational circular dichroism (VCD) spectra of the crude essential oils were recorded and in a second step they were modelized as a linear weighted combination of the IR and VCD spectra of the individual spectra of pure enantiomer of the major chiral compounds present in the EOs. The VCD spectra of enantiomer of known enantiomeric excess shall be recorded if they are not yet available in a library of VCD spectra. For IR, the spectra of pure enantiomer or racemic mixture can be used. The full spectra modelizations were performed using a well known and powerful mathematical model (least square estimation: LSE) which resulted in a weighting of each contributing compound. For VCD modelization, the absolute value of each weighting represented the percentage of the associate compound while the attached sign addressed the correctness of the enantiomeric form used to build the model. As an example, a model built with the non-prevailing enantiomer will show a negative sign of the weighting value. For IR spectra modelization, the absolute value of each weighting represented the percentage of the compounds without of course accounting for the chirality of the prevailing enantiomers. Comparison of the weighting values issuing from IR and VCD spectra modelizations is a valuable source of information: if they are identical, the EOs are composed of nearly pure enantiomers, if they are different the chiral compounds of the EOs are not in an optically pure form. The method was applied on four samples of essential oil of Artemisia herba - alba in which the three major compounds namely (−)-α-thujone, (+)-β-thujone and (−)-camphor were found in different proportions as determined by GC–MS and chiral HPLC using polarimetric detector. In order to validate the methodology, the modelization of the VCD spectra was performed on purpose using the individual VCD spectra of (−)-α-thujone, (+)-β-thujone and (+)-camphor instead of (−)-camphor. During this work, the absolute configurations of (−)-α-thujone and (+)-β-thujone were confirmed by comparison of experimental and calculated VCD spectra as being (1 S ,4 R ,5 R ) and (1 S ,4 S ,5 R ) respectively. Abstract: Asymmetric allylic alkylation leading to dimethyl [(2E)-1,3-diphenylprop-2-en-1-yl]propanedioate 1 is a privileged reaction which has been considered in more than 800 references from 1985 to early 2012. This paper thus begins with a thorough review of the literature with a particular focus on the way the ee''s and absolute configuration of the prevailing enantiomer were claimed and reported by the authors. In a large majority of articles chiral chromatography is used for ee''s determination. Unfortunately, in too many cases the data, the column or the eluent are not provided. In a significant proportion (5%) the column name is ambiguous. Furthermore, several discrepancies are detected in the assigned order of elution when chiral chromatography data are provided. We therefore decided to firmly establish the chromatographic behavior of the enantiomers of 1, which were obtained from the corresponding racemate by semi-preparative chiral chromatography and their absolute configuration assigned by ECD and VCD spectroscopies. ORD curves show that optical rotation is very weak at 350nm with indication of inversion of the sign at lower wavelengths. It results in a low sensitivity for on line JASCO polarimeter detector. Chiroptical detection was nicely performed by on line JASCO CD detector set at 254nm: (−)-(S)-1 shows a (+)-CD254nm sign. Pure enantiomers of authenticated absolute configuration allowed a safe assignment of the order of elution during HPLC or SFC on major chiral stationary phases. Quite interestingly for practical application, the order of elution is reversed on Chiralpak AD-H and IA on going from hexane/EtOH to hexane/2-PrOH in HPLC or on going from CO2/EtOH (or MeOH) to CO2/2-PrOH in SFC. Binuclear square planar Ni(II) complexes are described, formed by two tridentate ligands with two imine-nitrogens coordinating two nickel atoms. Such complexes are synthetically readily available with great structural variety and present new types of ridge-tile-like chiral compounds that are reasonably stable in the appropriate "bent" conformation. Enantiomerically pure samples of these compounds have been obtained for the first time using HPLC with a chiral stationary phase. Absolute configurations and chiroptical properties are fully characterized by ECD, VCD, ORD spectroscopy, and theoretical calculations. These new compounds with ridge-tile-like chiral topology are configurationally reasonably stable [ΔG‡ = 121.4 kJ mol-1, t½ = 14.9 h (78 °C, ethanol)], and therefore their chemistry, physical properties, and applications can be systematically studied. Abstract: An inherently chiral ABii diphosphonato cavitand (±)-4 bearing a single quinoxaline bridging moiety was synthesized and resolved by chiral HPLC. Its chiroptical properties were investigated and VCD experiments allowed the determination of its absolute configuration. Distinguishable diastereomeric complexes in solution with l-adrenaline were observed by 1H and 31P NMR together with a noticeable enantio-discrimination at 253K (dr ∼2:1) in favor of the dextrorotatory cavitand (+)-4.
