Background Damage to the adult central nervous system often leads to long-term disruptions in function due to the limited capacity for neurological recovery. The central nervous system of the Mediterranean field cricket, Gryllus bimaculatus, shows an unusual capacity for compensatory plasticity, most obviously in the auditory system and the cercal escape system. In both systems, unilateral sensory disruption leads the central circuitry to compensate by forming and/or strengthening connections with the contralateral sensory organ. While this compensatory plasticity in the auditory system relies on robust dendritic sprouting and novel synapse formation, the compensatory plasticity in the cercal escape circuitry shows little obvious dendritic sprouting and instead may rely on shifts in excitatory and inhibitory synaptic strength. Results In order to better understand what types of molecular pathways might underlie this compensatory shift in the cercal system, we used a multiple k-mer approach to assemble a terminal ganglion transcriptome that included ganglia collected one, three, and 7 days after unilateral cercal ablation in adult, male animals. We performed differential expression analysis using EdgeR and DESeq2 and examined Gene Ontologies to identify candidates potentially involved in this plasticity. Enriched GO terms included those related to the ubiquitin-proteosome protein degradation system, chromatin-mediated transcriptional pathways, and the GTPase-related signaling system. Conclusion Further exploration of these GO terms will provide a clearer picture of the processes involved in compensatory recovery of the cercal escape system in the cricket and can be compared and contrasted with the distinct pathways that have been identified upon deafferentation of the auditory system in this same animal.

Many animals produce acoustic signals to mark territories and attract mates. When different species produce acoustic signals simultaneously, the signals create a noisy environment, with potential acoustic interference between species. Theoretical studies suggest that such reproductive interference may have strong effects on species interaction. For example, the inferior resource competitor can survive if its disadvantage is counterbalanced by superiority in reproductive interference. Two field cricket species, Teleogryllus occipitalis (Audinet-Serville) (Orthoptera: Gryllidae) and Loxoblemmus equestris Saussure (Orthoptera: Gryllidae), cooccur in the same habitat. A previous study has shown that L. equestris is an inferior species to T. occipitalis in terms of resource competition. Therefore, we predicted that mate location and choice behavior of female T. occipitalis would be negatively affected by the acoustic signals of L. equestris and tested this with a series of playback experiments. The mate choice behavior of female T. occipitalis was not significantly affected by the calling song of L. equestris. Our results suggest that the acoustic interference does not explain the cooccurrence of the two species in the same habitat.

Male field crickets (Orthoptera: Gryllidae) produce acoustic signals by wing stridulation, attracting females for mating. A plectrum on the left forewing’s (or tegmen) anal margin rapidly strikes along a serrated vein (stridulatory file) on the opposite tegmen as they close, producing vibrations ending in a tonal sound. The tooth strike rate of the plectrum across file teeth is equal to the sound frequency produced by the cricket (i.e., ~5k teeth/s for ~5 kHz in field crickets) and is specific to the forewing’s resonant frequency. It is subsequently amplified using specialised wing cells. Anatomically, the forewings appear to mirror each other: a file and plectrum exist on both tegmina; however, most cricket species stridulate using right-over-left wing overlap making the stridulatory mechanism directionally asymmetrical by default, rendering the left tegmen’s file unused. Therefore, we hypothesise structural differences between the functional and unfunctional files. Three-dimensional images were used to accurately measure the stridulatory file structure in Gryllus bimaculatus wings. It was found that the left file shows significantly greater variation in inter-tooth distance than the right file, both over the entire file and especially the first sixty teeth (the functional part). Advantages of this modern method over scanning electron microscopy are discussed.

Stressed animals often struggle to maintain optimal investment into a number of fitness-related traits, which can result in some traits being more adversely affected than others. Variation in stress-related costs may also depend on the environment-costs can be facultative and only occur when resources are limited, or they may be obligate and occur regardless of resource availability. Dynamics of oxidative stress may be important in life-history evolution given their role in a range of biological processes-from reproduction to immunity to locomotion. Thus, we examined how resource (food) availability influences the costs of oxidative challenge to fitness-related traits spanning several levels of biological organization. We manipulated food availability and oxidative status in females of the wing-dimorphic sand field cricket (Gryllus firmus) during early adulthood. We then determined investment into several traits: reproduction (ovary mass), soma (body mass and flight musculature), and immune function (total phenoloxidase activity). Oxidative challenge (paraquat exposure) obligated costs to somatic tissue and a parameter of immune function regardless of food availability, but it did not affect reproduction. We show that the costs of oxidative challenge are trait-specific, but we did not detect a facultative (food-dependent) cost of oxidative challenge to any trait measured. Although the dynamics of oxidative stress are complex, our study is an important step toward a more complete understanding of the roles that resource availability and redox systems play in mediating life histories.

Sexual signal evolution may present fitness consequences for the non-signaling sex due to shared genes and altered social conditions, but this is rarely studied in natural populations. Male choosiness of prospective female mating partners provides an increasingly recognized pathway through which males can increase their fitness. For example, males may increase their number of offspring by targeting more fecund females as mating partners. If fecundity is heritable, males that mate with more fecund females can also receive the indirect benefit of more fecund daughters. In species where female fecundity is not directly assessable, female ornaments may act as signals of fecundity. However, whether female ornaments reliably signal the indirect benefit of more fecund daughters is not well understood. We investigated this question using the field cricket, Teleogryllus oceanicus. Previous work had identified the cuticular hydrocarbon (CHC) profile as a female sexual display trait in T. oceanicus. To examine whether CHCs can provide a reliable signal of fecundity, we tested whether individual CHC compounds and the first principal axis of CHC variation (PC1) are genetically correlated with ovary mass, a reliable proxy for fecundity in this species. We found significant genetic correlations between ovary mass and three individual CHC compounds, as well as PC1. This result indicates that by targeting females as mating partners based on their CHC profile, males can sire more fecund daughters.

A functioning immune system is crucial for protection against disease and illness, yet increasing evidence suggests that species living in urban areas could be suffering from immune suppression, due to the presence of artificial light at night (ALAN). This study examined the effects of ecologically relevant levels of ALAN on three key measures of immune function (haemocyte concentration, lytic activity, and phenoloxidase activity) using a model invertebrate species, the Australian black field cricket, Teleogryllus commodus. We reared crickets under an ecologically relevant daily light‐cycle consisting of 12 hr bright daylight (2600 lx) followed by either 12 h darkness (0 lx) or dim environmentally relevant ALAN (1, 10, 100 lx), and then assessed immune function at multiple time points throughout adult life using haemolymph samples. We found that the presence of ALAN had a clear negative effect on haemocytes, while the effects on lytic activity and phenoloxidase activity were more complex or largely unaffected by ALAN. Furthermore, the effects of lifelong exposure to ALAN of 1 lx were comparable to those of 10 and 100 lx. Our data suggest that the effects of ALAN could be large and widespread, and such reductions in the core immune response of individuals will likely have greater consequences for fitness and survival under more malign conditions, such as those of the natural environment.

Most of our knowledge of insect genomes comes from Holometabolous species, which undergo complete metamorphosis and have genomes typically under 2 Gb with little signs of DNA methylation. In contrast, Hemimetabolous insects undergo the presumed ancestral process of incomplete metamorphosis, and have larger genomes with high levels of DNA methylation. Hemimetabolous species from the Orthopteran order (grasshoppers and crickets) have some of the largest known insect genomes. What drives the evolution of these unusual insect genome sizes, remains unknown. Here we report the sequencing, assembly and annotation of the 1.66-Gb genome of the Mediterranean field cricket Gryllus bimaculatus, and the annotation of the 1.60-Gb genome of the Hawaiian cricket Laupala kohalensis. We compare these two cricket genomes with those of 14 additional insects and find evidence that hemimetabolous genomes expanded due to transposable element activity. Based on the ratio of observed to expected CpG sites, we find higher conservation and stronger purifying selection of methylated genes than non-methylated genes. Finally, our analysis suggests an expansion of the pickpocket class V gene family in crickets, which we speculate might play a role in the evolution of cricket courtship, including their characteristic chirping., Ylla, Extavour et al. use genomic data from crickets to investigate the evolution of large genome sizes and DNA methylation events in insects. Their findings indicate that transposable element activity drove genome expansion in hemimetabolous insects, such as crickets and grasshoppers, and that DNA methylation is predominant in conserved genes.

Estimating costs of ejaculate production is challenging. Metabolic investment in ejaculates may come at the expense of other physiological functions and may negatively affect future reproduction and/or survival. These trade-offs are especially likely to occur under constrained resource pools (e.g., poor nutrition). Here, we investigated costs of ejaculate production via trade-offs in the field cricket Gryllus bimaculatus. We experimentally increased rates of ejaculate production, while keeping an unmanipulated group, in adult males kept at high and low feeding regimes and tested the effects of our treatments on (i) somatic maintenance (i.e., changes in male body mass), (ii) future reproduction (i.e., the likelihood of producing a spermatophore and the viability of its sperm), and (iii) lifetime survival and longevity. We predicted investment in ejaculates to impinge upon all measured responses, especially in low-fed individuals. Instead, we only found negative effects of food limitation, suggesting low or undetectable costs of spermatophore production. High mating rates may select for males to maximize their capacity of ejaculate production, making ejaculate traits less prone to trade-offs with other fitness-related life history traits. Nevertheless, males were impaired due to nutrient deficiency in producing viable ejaculates, suggesting condition-dependent costs for ejaculate production.

Carbon nanotubes (CNTs) have been increasingly more prevalent due to their use in product technology owing to their exceptional electrical and thermal conductivity and tensile strength because of their nanostructure and strength of the bonds among carbon atoms. The potential increase of CNTs in the environment is a concern, and studies to assess the toxic effects of these nanomaterials (NMs) are needed. However, so far, most of the studies are focused on aquatic species and much less is understood about the effects of NM in terrestrial organisms. This investigation used a functionalized multi-walled carbon nanotube (f-MWCNT) and the Jamaican cricket Gryllus assimilis to assess the effects of this NM. Cricket nymphs were injected with f-MWCNT suspension-at three different concentrations. The insecticide Fipronil was used as a positive control. Survival was monitored, and histological analysis was made in the brains. Pyknotic cells were quantified in two brain regions, a neurosecretory called Pars intercerebralis (PI), and an associative region called mushroom body (MB). No mortality was observed in any f-MWCNT concentration tested. A significant increase in pyknotic cells was observed as sub-lethal effect for the intermediate concentration of f-MWCNT, at PI, while any significant change was observed at the Kenyon cells of the MB. These results are discussed in the context of agglomeration and dispersion of the f-MWCNT at different concentrations, and availability of the f-MWCNT on the circulatory system, as well as the natural decay of pyknotic cells with time and different patterns of adult cricket neurogenesis. Our results showed that f-MWCNT had negative effects in the neurosecretory region of the brain.

Associations between heritable polymorphisms and life-history traits, such as development time or reproductive investment, may play an underappreciated role in maintaining polymorphic systems. This is because selection acting on a particular morph could be bolstered or disrupted by correlated changes in life history or vice versa. In a Hawaiian population of the Pacific field cricket (Teleogryllus oceanicus), a novel mutation (flatwing) on the X-chromosome is responsible for a heritable polymorphism in male wing structure. We used laboratory cricket colonies fixed for male wing morph to investigate whether males and females bearing the flatwing or normal-wing (wild-type) allele differed in their life-history traits. We found that flatwing males developed faster and had heavier testes than normal-wings, whereas flatwing homozygous females developed slower and had lighter reproductive tissues than normal-wing homozygous females. Our results advance our understanding of the evolution of polymorphisms by demonstrating that the genetic change responsible for a reproductive polymorphism can also have consequences for fundamental life-history traits in both males and females.

Inadvertent cues can be refined into signals through coevolution between signalers and receivers, yet the earliest steps in this process remain elusive. In Hawaiian populations of the Pacific field cricket, a new morph producing a novel and incredibly variable song (purring) has spread across islands. Here we characterize the current sexual and natural selection landscape acting on the novel signal by (1) determining fitness advantages of purring through attraction to mates and protection from a prominent deadly natural enemy, and (2) testing alternative hypotheses about the strength and form of selection acting on the novel signal. In field studies, female crickets respond positively to purrs, but eavesdropping parasitoid flies do not, suggesting purring may allow private communication among crickets. Contrary to the sensory bias and preference for novelty hypotheses, preference functions (selective pressure) are nearly flat, driven by extreme inter-individual variation in function shape. Our study offers a rare empirical test of the roles of natural and sexual selection in the earliest stages of signal evolution., Parasitoid flies eavesdrop on the mating songs of male Hawaiian crickets, creating conflict between sexual and natural selection. Here, the authors investigate the selection acting on a recently evolved male mating signal, a “purring” song, which appears to be undetected by parasitoids.

Sexually selected traits, including threat signals, have been shown to scale steeply positively with body size because their exaggeration maximizes honest signalling. However, the functional allometry hypothesis makes the opposite prediction for some weapons: because the biomechanics of force applied in their use may favor relatively smaller size, sexually selected weapons may exhibit negative allometry. Tests of these ideas in insects have largely focused on holometabolous species, whose adult body size is entirely dependent on nutrients acquired during the larval stage. In contrast, hemimetabolous insects may exhibit different patterns of allometry development because they forage throughout development, between successive moults. Here, we tested complementary and competing predictions made by the positive and functional allometry hypotheses, regarding intrasexually selected trait allometry in a hemimetabolous insect, the Jamaican field cricket (Gryllus assimilis). As expected, head width (a dominance and/or combat trait) was more positively allometric than non-sexually selected traits. In contrast, and consistent with the functional allometry hypothesis, mouthparts (weapons) were either isometric or negatively allometric. We also tested whether trait allometry responded to rearing diet by raising males on either a high protein diet or a high carbohydrate diet; we predicted stronger positive allometry under the high protein diet. However, diet did not influence allometry in the predicted manner. Overall, our results support the functional allometry hypothesis regarding sexually selected trait allometry and raise intriguing possibilities for integrating these ideas with recent paradigms for classifying intrasexually selected traits.

Understanding how species can thrive in a range of environments is a central challenge for evolutionary ecology. There is strong evidence for local adaptation along large-scale ecological clines in insects. However, potential adaptation among neighbouring populations differing in their environment has been studied much less. We used RAD sequencing to quantify genetic divergence and clustering of ten populations of the field cricket Gryllus campestris in the Cantabrian Mountains of northern Spain, and an outgroup on the inland plain. Our populations were chosen to represent replicate high and low altitude habitats. We identified genetic clusters that include both high and low altitude populations indicating that the two habitat types do not hold ancestrally distinct lineages. Using common-garden rearing experiments to remove environmental effects, we found evidence for differences between high and low altitude populations in physiological and life-history traits. As predicted by the local adaptation hypothesis, crickets with parents from cooler (high altitude) populations recovered from periods of extreme cooling more rapidly than those with parents from warmer (low altitude) populations. Growth rates also differed between offspring from high and low altitude populations. However, contrary to our prediction that crickets from high altitudes would grow faster, the most striking difference was that at high temperatures, growth was fastest in individuals from low altitudes. Our findings reveal that populations a few tens of kilometres apart have independently evolved adaptations to their environment. This suggests that local adaptation in a range of traits may be commonplace even in mobile invertebrates at scales of a small fraction of species' distributions.

The variation in lipidome of house cricket, banded cricket, Jamaican field cricket and two-spotted cricket was studied using high-throughput screening techniques for fingerprinting (MALDI TOF MS, GC–MS and LC MS-MS) and well-stablished chromatographic techniques for quantification (HPLC-ELSD, GC- FID). Although the four cricket species were reared in identical conditions, two-spotted & banded crickets had a lipid content 1.5 fold higher than house cricket. The lipids were high in UFA (>63%) and unsaturated TAG (>98%) making them liquid at room temperature, thus an oil. Cholesterol and several phytosterols were profiled finding high cholesterol concentration which is a point of concern. Eight phospholipid types (211 species) were identified with no major differences among cricket species. Using high-throughput screening techniques we demonstrate the complexity of cricket lipidome. Information on the lipidome of these crickets with high commercial value is important to estimate its nutritional value and their potential food applications.

Genetic benefits from mating with multiple males are thought to favour the evolution of polyandry. However, recent evidence suggests that non-genetic paternal effects via seminal fluid might contribute to the observed effects of polyandry on offspring performance. Here, we test this hypothesis using the field cricket Teleogryllus oceanicus . Using interference RNA, we first show that at least one seminal fluid protein is essential for embryo survival. We then show that polyandrous females mated to three different males produced embryos with higher pre-hatching viability than did monandrous females mated with the same male three times. Pseudo-polyandrous females that obtained sperm and seminal fluid from a single male and seminal fluid from two additional males had embryos with viabilities intermediate between monandrous and polyandrous females. Our results suggest either that ejaculate mediated paternal effects on embryo viability have both genetic and non-genetic components, or that seminal fluids transferred by castrated males provide only a subset of proteins contained within the normal ejaculate, and are unable to exert their full effect on embryo viability.

The effects of reproduction are variable among insects, as well as the time of mate. In animals, a trade-off is usually observed between reproduction and self-maintenance, mostly in females because of egg producing costs. In the present investigation, manifestations of aging and senescence at population and cellular levels were studied associated with the mating opportunities. The main goal of this study was to evaluate the effect of mating on lifespan and also on the physiological aging in adult Jamaican field cricket females, Gryllus assimilis (Fabricius, 1775). Three experimental groups were set: virgin, normal-mated (14 days old), and late-mated (30 days old) insects. Lifespan, age-specific mortality rate, number of eggs laid, and in situ amount of the age-pigment neurolipofuscin were quantified. Results showed a trade-off between reproductive females and lifespan, with the strongest effects found in late-mated. Age-specific mortality showed a faster increase for late-mated females from 35 to 50 days old, followed by normal-mated females (35 to 60 days old). Virgins had the lowest age-specific mortality of all, increasing just from 65 to 73 days old. Normal-mated had the highest number of eggs laid, followed by late-mated and virgins, respectively. Neurolipofuscin accumulation rate was similar among groups, which was reflected in similar physiological rate of aging. Results indicate that reproductive status did affect the life-history of these field cricket females.

Gigagryllus omayrae n. gen et n. sp., a new genus of giant field cricket, from the middle Magdalena of Colombia, more precisely from the department of Tolima, in the region of the Armero tragedy, for which this species is dedicated to the memory of Omayra Sanchez, a symbol of the most terrible natural tragedy that has occurred in the recent history of Colombia. Gigagryllus n. gen. is one of the largest genera in America, with a size similar to Titanogryllus, Megalogryllus and some species of Gryllita. The classification of the American Gryllinae is discussed, and a key to the American genera is provided. Besides, genera groups are proposed for the subtribe Brachytrupine (Gryllini) taxa, which differs from Old World genera. Geogryllus n. syn. and Rubrogryllus n. syn. are synonymized under Gryllita and Cophogryllus americanus is considered as nomen dubium, ruling out the presence of subtribe Cophogryllina in America.

We describe a pair of labial gland lobes on either side of the retrocerebral complex in the head of the Australian black field cricket, Teleogryllus commodus Walker. As the retrocerebral complex includes the corpora cardiaca and corpora allata, hormones secreted by these glands can be absorbed by these lobes. These lobes of the labial gland are connected to the thoracic lobes via a relatively long duct that enters the main duct draining the thoracic lobes. Measurement of the flow rate of dye from head to thorax in the ducts is rapid, suggesting that these glands may serve as a transport system into the thoracic region. Both serotonin and adipokinetic hormone are shown to be present in the lobes near the retrocerebral complex and the ducts of the thoracic lobes, but whether this connection between the head and thorax acts as a hormone transporter is still unclear., Graphical Abstract Graphical AbstractIn the black field cricket (Teleogryllus commodus), one pair of salivary lobes are located on either side of the corpora cardiaca and corpora allata, connected to the thoracic salivary glands by a long narrow duct. Ectotherms are sensitive to changes in ambient temperature that impact their physiology and development. To compensate for the effects of variation in temperature, ectotherms exhibit short or long-term physiological plasticity. An extensive body of literature exists towards understanding these effects and the solutions ectotherms have evolved. However, to what extent rearing temperature during early life stages impacts the behaviour expressed in adulthood is less clearly understood. In the present study, we aimed to examine the effects of developmental temperature on life-history traits and mating call features in a tropical field cricket, Acanthogryllus asiaticus. We raised A. asiaticus at two different developmental conditions: 25 °C and 30 °C. We found developmental time and adult lifespan of individuals reared at 30 °C to be shorter than those reared at 25 °C. Increased developmental temperature influenced various body size parameters differentially. Males raised at 30 °C were found to be larger and heavier than those raised at 25 °C, making A. asiaticus an exception to the temperature-size rule. We found a significant effect of change in immediate ambient temperature on different call features of both field-caught and lab-bred individuals. Developmental temperature also affected mating call features wherein individuals raised at higher temperature produced faster calls with a higher peak frequency compared to those raised at lower temperature. In addition, an interactive effect of both developmental and immediate temperature was found on mating call features. Our study highlights the importance of understanding how environmental temperature shapes life-history and sexual communication in crickets.

We present the first comprehensive molecular phylogeny of Gryllus field cricket species found in the United States and Canada, select additional named Gryllus species found in Mexico and the Bahamas, plus the European field cricket G. campestris Linnaeus and the Afro-Eurasian cricket G. bimaculatus De Geer. Acheta, Teleogryllus, and Nigrogryllus were used as outgroups. Anchored hybrid enrichment was used to generate 492,531 base pairs of DNA sequence from 563 loci. RAxML analysis of concatenated sequence data and Astral analysis of gene trees gave broadly congruent results, especially for older branches and overall tree structure. The North American Gryllus are monophyletic with respect to the two Old World taxa; certain sub-groups show rapid recent divergence. This is the first Anchored Hybrid Enrichment study of an insect group done for closely related species within a single genus, and the results illustrate the challenges of reconstructing the evolutionary history of young rapidly diverged taxa when both incomplete lineage sorting and probable hybridization are at play. Because Gryllus field crickets have been used extensively as a model system in evolutionary ecology, behavior, neuro-physiology, speciation, and life-history and life-cycle evolution, these results will help inform, interpret, and guide future research in these areas.

A growing body of evidence exists to support a detrimental effect of the presence of artificial light at night (ALAN) on life-history and fitness traits. However, few studies simultaneously investigate multiple traits and the life stages at which changes manifest. We experimentally manipulated ALAN intensities, within those found in the natural environment, to explore the consequences for growth, survival, and reproductive success of the field cricket, Teleogryllus commodus. We reared crickets from egg to adult under a daily light-cycle consisting of 12 hr bright daylight (2,600 lx) followed by either 12 hr darkness (0 lx) or dim-light environments (1, 10, or 100 lx). We found egg hatch, adult survival, and reproductive measures were largely comparable for all treatments. However, juvenile development time (number of days from egg to adult) was on average 10 days (14%) longer and adults were also larger when crickets were exposed to any light at night (1, 10, or 100 lx). Our data demonstrate that chronic lifetime exposure to ALAN can modulate the timing of life-history events and may disrupt phenology to a similar extent as other abiotic factors.

Evidence suggests that brain serotonin (5-HT) is one of the central mediators of different types of animal personality. We tested this assumption in field crickets Gryllus integer using a selective serotonin reuptake inhibitor (SSRI). Crickets were selected for slow and rapid development and tested for their coping styles under non-stressful conditions (time spent exploring a novel object). Resting metabolic rate, maximum metabolic rate and latency to resume activity were measured under stressful conditions (stress reactivity). Measurements were taken (i) before and (ii) during the SSRI treatment. Before the SSRI treatment, a strong negative correlation was observed between coping style and stress reactivity, which suggests the existence of a behavioral syndrome. After the SSRI treatment, the syndrome was no longer evident. The results of this study show that 5-HT may be involved in regulating behavior not only along a stress reactivity gradient but also along a coping styles axis. The relationship between personality and the strength and direction of 5-HT treatment on observed behaviors indicates trait-like individual differences in 5-HT signaling. Overall, these findings do not support recent ideas arising from the pace-of-life syndrome (POLS) hypothesis, which predict higher exploration and metabolic rates in rapidly developing bold animals.

We recorded the wing movements and sound signals during the production of calling, rivalry, and courtship song in the bispotted field cricket Gryllus bimaculatus. Recordings confirm that salient sound pulses during calling and rivalry song are generated during the closing movements of the wings. Wing movements for calling and rivalry song start from an elevated wing position and are performed with a very similar opening-closing movement, indicating that both types of songs may be generated by the same neuronal network. Wing movements for courtship song start from a low wing position; rapid closing movements generate high-frequency ticks and low-amplitude wing oscillations lead to low-amplitude pulses, generated during the opening and closing movements with a carrier frequency corresponding to the calling song. The two types of wing movements underlying courtship song indicate a different motor control as compared to calling song and may represent an early evolutionary phenotype.

No. of bitstreams: 0 Previous issue date: 2020-05-29 The common black field cricket, Gryllus assimilis (Orthoptera: Gryllidae), damages young plants of red cedar, Juniperus virginiana (Cupressaceae); strawberry, Fragaria × ananassa (Rosaceae); sugarcane, Saccharum officinarum (Poaceae); teak, Tectona grandis (Lamiaceae); upland cotton, Gossypium hirsutum (Malvaceae); and, mainly, Eucalyptus spp. (Myrtaceae). The objective of this study was to investigate the biological and behavioral parameters of this insect in the laboratory and in Eucalyptus spp. plantations in Inhambupe, Bahia State, Brazil. The incubation period and the viability of G. assimilis eggs were 11.87 days and approximately 22%, respectively. The duration of the nymphal stage was 62.34 days with approximately 60% of the nymphs obtained in the laboratory being females. The average number of egg batches per female, eggs per female, and eggs per batch per female of this insect were 25.50, 862.17, and 34.65, respectively. G. assimilis females lived for 76.50 days in the adult stage, and 138.34 days in total, from egg through nymph to adult. Males produced three characteristic sounds: one for the marking of territory, one for courtship, and one when alone. G. assimilis fed primarily on weeds but, in their absence, it damaged young Eucalyptus spp. plants. This paper presents important data on the biology and behavior of G. assimilis; this information may encourage additional biological research, laboratory rearing, and integrated management of this pest. Some ectotherms can survive internal ice formation. In temperate regions, freeze tolerance is often induced by decreasing temperature and/or photoperiod during autumn. However, we have limited understanding of how seasonal changes in physiology contribute to freeze tolerance, and how these changes are regulated. During a six week autumn-like acclimation, late-instar juveniles of the spring field cricket Gryllus veletis (Orthoptera: Gryllidae) become freeze tolerant, which is correlated with accumulation of low molecular weight cryoprotectants, elevation of the temperature at which freezing begins, and metabolic rate suppression. We used RNA-Seq to assemble a de novo transcriptome of this emerging laboratory model for freeze tolerance research. We then focused on gene expression during acclimation in fat body tissue due to its role in cryoprotectant production and regulation of energetics. Acclimated G. veletis differentially expressed more than 3,000 transcripts in fat body. This differential expression may contribute to metabolic suppression in acclimated G. veletis, but we did not detect changes in expression that would support cryoprotectant accumulation or enhanced control of ice formation, suggesting that these latter processes are regulated post-transcriptionally. Acclimated G. veletis differentially regulated transcripts that likely coordinate additional freeze tolerance mechanisms, including upregulation of enzymes that may promote membrane and cytoskeletal remodelling, cryoprotectant transporters, cytoprotective proteins, and antioxidants. Thus, while accumulation of cryoprotectants and controlling ice formation are commonly associated with insect freeze tolerance, our results support the hypothesis that many other systems contribute to surviving internal ice formation. Together, this information suggests new avenues for understanding the mechanisms underlying insect freeze tolerance.

Proteins and carbohydrates have profound impacts on the ecology of gut microbiota, but disentangling the single and interactive effects of different dietary constituents is challenging. Here, we used a multidimensional approach, the Geometric Framework, to study the interactions between nutrition and bacterial abundances with respect to protein and carbohydrate intakes in field cricket, Teleogryllus oceanicus. Our study revealed that species richness decreased as crickets consumed more macronutrients, and species evenness peaked at high intake of protein-rich diets. Sex and protein:carbohydrate (P:C) ratios in diets were the primary factors influencing the gut bacterial community, but most of the microbial operational taxonomic units (OTUs) that were significantly different between males and females were present in low abundance. In contrast, protein intake had a greater influence than carbohydrate consumption on the relative abundances of the core bacterial taxa, as an increase in dietary protein availability could remove the growth constraint imposed by limited nitrogen. Taken together, the use of the Geometric Framework provides a deeper insight into how nutritional intakes influence the relative abundances of gut microbes, and could be a useful tool to integrate the study of gut microbiome and fitness traits in a host.

Main thiols and disulfides were determined in the hemolymph of the Jamaican field cricket Gryllus assimilis of various developmental stages. On the basis of these data, redox potentials of the glutathione, cysteine and homocysteine redox systems were calculated. The concentrations of all thiols studied decreased during development (at a stage of 6 molts) with respect to young crickets, and increased again in adult insects. Redox potentials of the glutathione and cysteine systems increased from values of -131.0 ± 5.6 mV and -86.9 ±17.1 mV, respectively in young crickets to -58.0 ± 3.6 mV and -36.1 ± 4.2 mV, respectively, at the stage of 6 molts and decreased to values of -110.4 ± 24.8 mV and -66.3 ± 12.2 mV, respectively, in adult insects. Redox potentials of the glutathione and cysteine systems in the hemolymph of young and adult insects were similar to those reported for human plasma, suggesting similar redox conditions of extracellular fluids in insects and in mammals.

Females often mate more than is necessary to ensure reproductive success even when they incur significant costs from doing so. Direct benefits are hypothesized to be the driving force of high female mating rates, yet species in which females only receive an ejaculate from their mate still realize increased fitness from multiple mating. Using the Texas field cricket, Gryllus texensis, we experimentally test the hypothesis that multiple mating via monandry or polyandry increases female fitness by replenishing ejaculates, thereby allowing females to produce more offspring for a longer period of time. We found that higher rates of female mating significantly increased lifetime fecundity and oviposition independent of whether females mated with one or two males. Further, although interactions with males significantly increased rates of injury or death, females that replenished ejaculates experienced an increased rate and duration of oviposition, demonstrating that the immediate benefits of multiple mating may greatly outweigh the long-term costs that mating poses to female condition and survival. We suggest that ejaculate replenishment is a driving factor of high mating rates in females that do not receive external direct benefits from mating and that a comparative study across taxa will provide additional insight into the role that ejaculate size plays in the evolution of female mating rates.

Chill-susceptible insects enter the reversible state of chill coma at their critical thermal minimum (CTmin). During chill coma, movement of Na+ and water from the hemolymph to the gut lumen disrupt ion and water balance. Recovery from cold exposure requires re-establishment of this balance, and failure to do so results in chilling injury or death. We hypothesized that the passive leak of Na+ and consequently water during cold exposure is driven by the [Na+] differential between the gut and hemolymph. To determine the extent to which this [Na+] differential affects cold tolerance, we used artificial diets to load the guts of fall field crickets (Gryllus pennsylvanicus) with various concentrations of Na+. Manipulating [Na+] differentials had no effect on the CTmin, agreeing with recent studies demonstrating that chill coma onset precedes loss of ion balance in the cold. A high [Na+] diet reversed the direction of the [Na+] differential between the gut and hemolymph. Crickets fed a high [Na+] diet recovered from 12 h of chill coma nearly twice as fast as those fed low [Na+] diets. However, the high [Na+] diet was detrimental to survival after prolonged cold exposure (three days at 0 °C). Therefore, while a reduced [Na+] differential helps crickets recover from short-term cold exposure, an increased gut Na+ load itself appears to carry longer-term costs and promotes irreversible chilling injury.

The Bahama endemic field cricket, Gryllus bryanti Morse, 1905, previously known from only the lost female holotype, is redescribed, including designation of a neotype. Known only from Andros and Eleuthera Islands in The Bahamas, where it occurs with G. assimilis, it can be separated from the latter by song, tegmen file characteristics, head wider than pronotum, and genetics.

Many temperate insects encounter temperatures low enough to freeze their body fluids. Remarkably, some insects are freeze-tolerant, surviving this internal ice formation. However, the mechanisms underlying freeze tolerance are not well-understood, in part due to a lack of tractable model organisms. We describe a novel laboratory model to study insect freeze tolerance, the spring field cricket Gryllus veletis (Orthopera: Gryllidae). Following acclimation to six weeks of decreasing temperature and photoperiod, G. veletis become freeze-tolerant, similar to those exposed to natural autumn conditions in London, Ontario, Canada. Acclimated crickets suppress their metabolic rate by c. 33%, and survive freezing for up to one week at −8 °C, and to temperatures as low as −12 °C. Freeze-tolerant G. veletis protect fat body cells from freeze injury in vivo, and fat body tissue from freeze-tolerant cricket survives brief freeze treatments when frozen ex vivo. Freeze-tolerant crickets freeze at c. −6 °C, which may be initiated by accumulation of ice-nucleating agents in hemolymph or gut tissue. Although we hypothesize that control of ice formation facilitates freeze tolerance, initiating ice formation at high subzero temperatures does not confer freeze tolerance on freeze-intolerant nymphs. Acclimation increases hemolymph osmolality from c. 400 to c. 650 mOsm, which may facilitate freeze tolerance by reducing ice content. Hemolymph ion concentrations do not change with acclimation, and we therefore predict that freeze-tolerant G. veletis elevate hemolymph osmolality by accumulating other molecules. Gryllus veletis is easily reared and manipulated in a controlled laboratory environment, and is therefore a suitable candidate for further investigating the mechanisms underlying freeze tolerance.

Recent theory predicts that increased phenotypic plasticity can facilitate adaptation as traits respond to selection. When genetic adaptation alters the social environment, socially mediated plasticity could cause co-evolutionary feedback dynamics that increase adaptive potential. We tested this by asking whether neural gene expression in a recently arisen, adaptive morph of the field cricket Teleogryllus oceanicus is more responsive to the social environment than the ancestral morph. Silent males (flatwings) rapidly spread in a Hawaiian population subject to acoustically orienting parasitoids, changing the population's acoustic environment. Experimental altering crickets’ acoustic environments during rearing revealed broad, plastic changes in gene expression. However, flatwing genotypes showed increased socially mediated plasticity, whereas normal-wing genotypes exhibited negligible expression plasticity. Increased plasticity in flatwing crickets suggests a coevolutionary process coupling socially flexible gene expression with the abrupt spread of flatwing. Our results support predictions that phenotypic plasticity should rapidly evolve to be more pronounced during early phases of adaptation. Postprint

Because sexual signals can arise from the coupling of specialized motor activity with morphological structures, disruption to a single component could lead to overall loss of function. Opportunities to observe this process and characterize any remaining signal components are rare, but could provide insight into the mechanisms, indirect costs and evolutionary consequences of signal loss. We investigated the recent evolutionary loss of a long-range acoustic sexual signal in the Hawaiian field cricket Teleogryllus oceanicus. Flatwing males carry mutations that remove sound-producing wing structures, eliminating all acoustic signalling and affording protection against an acoustically-orientating parasitoid fly. We show that flatwing males produce wing movement patterns indistinguishable from those that generate sonorous calling song in normal-wing males. Evolutionary song loss caused by the disappearance of structural components of the sound-producing apparatus has left behind the energetically costly motor behaviour underlying normal singing. These results provide a rare example of a vestigial behaviour and raise the possibility that such traits could be co-opted for novel functions. Postprint

Experiments with artificial diets demonstrated that black field cricket (Teleogryllus commodus (Walker)) and Lepidogryllus sp. were highly responsive to presence of lolines in their diet-quantities of diet consumed declined exponentially with increasing loline concentration. Amount consumed by black field cricket and Lepidogryllus sp. on diet containing 5,600 µg/g lolines was only 8 and 2% relative to those on loline-free diet, respectively. Additional experiments with Festulolium seeds demonstrated that both cricket species predated heavily on endophyte-free seed but largely avoided Epichloë uncinata-infected seed. By 12 h, black field cricket had destroyed 98.8% of endophyte-free but only 24.8% of E. uncinata-infected, loline-containing seed. By 36 h, Lepidogryllus sp. crickets had destroyed 40% of endophyte-free but had not fed on E. uncinata-infected, loline-containing seed. Glasshouse experiments demonstrated this aversion to lolines greatly reduces the damage potential of black field cricket in E. uncinata-infected Festulolium. When microswards were sown with E. uncinata-infected Festulolium, seedling numbers were reduced 25-26%, and yields 29-40%, by black field crickets relative to microswards sown without insect infestation. This contrasts with 70-78% reduction in seedling numbers and 67-80% reduction in yields in microswards sown to either endophyte-free Festulolium, endophyte-free perennial ryegrass, or Epichloë festucae var. lolii-infected Festulolium. Yields of mature E. uncinata-infected Festulolium plants were not adversely affected by black field crickets, irrespective of the presence of the endophyte-free standard Festulolium sown as a companion. In contrast, yields of endophyte-free Festulolium, endophyte-free perennial ryegrass, and E. festucae var. lolii-infected Festulolium plants were reduced by 56-61% by crickets.

Changes in feeding behaviour, especially the overconsumption of calories, has led to a rise in the rates of obesity, diabetes, and other associated disorders in humans and a range of animals inhabiting human-influenced environments. However, understanding the relative contribution of genes, the nutritional environment, and their interaction to dietary intake and lipid deposition in the sexes still remains a major challenge. By combining nutritional geometry with quantitative genetics, we determined the effect of genes, the nutritional environment, and their interaction on the total nutritional preference (TP), total diet eaten (TE), and lipid mass (LM) of male and female black field crickets (Teleogryllus commodus) fed one of four diet pairs (DPs) differing in the ratio of protein to carbohydrate and total nutritional content. We found abundant additive genetic variance for TP, TE, and LM in both sexes and across all four DPs, with significant genetic correlations between TE and TP and between TP and LM in males. We also found significant genotype-by-DP and genotype-by-sex-by-DP interactions for each trait and significant genotype-by-sex interactions for TE and LM. Complex interactions between genes, sex, and the nutritional environment, therefore, play an important role in nutrient regulation and lipid deposition in T. commodus. This finding may also help explain the increasing rate of obesity and the maintenance of sex differences in obesity observed across many animal species, including humans.

Inhabitants of the Indonesian island of Sumatra are faced with the problem of insufficient food supplies and the consequent risk of undernourishment and health issues. Edible insects as a traditional and readily available food source could be part of the solution. The nutritional value of insects depends on many factors, e.g., species, developmental stage, sex, diet, and climatic conditions. However, edible insects bred in Sumatra for human consumption have never before been assessed with regard to their nutritional value. Our study involved analyses of crude protein, chitin, fat and selected fatty acid contents of giant mealworm larvae (Zophobas morio), larvae of the common mealworm (Tenebrio molitor) and nymphs of the field cricket (Gryllus assimilis). Crude protein content in the samples ranged from 46% to 56%. Highest (35%) and lowest (31%) amounts of fat were recorded in giant mealworm larvae and larvae of the common mealworm, respectively. Chitin amounts ranged from 6% to 13%. Based on these values, which are comparable to those known from other food insects reared in different regions of the world, the edible species bred in Sumatra could become food sources with a potential to help stave off hunger and undernourishment. Druhu, vývojové fázi, pohlaví, stravě a klimatických podmínkách. Avšak jedlé hmyz chovaný v Sumatře pro lidskou spotřebu nebyl nikdy předtím hodnocen vzhledem k jejich nutriční hodnotě. Naše studie zahrnovala analýzu surového proteinu, chitinu, tuku a vybraných obsahů mastných kyselin obřích larv morušových (Zophobas morio), larvů obyčejných (Tenebrio molitor) a nymfů polní kriket (Gryllus assimilis). Obsah dusíkatých látek ve vzorcích se pohyboval od 46% do 56%. Nejvyšší (35%) a nejnižší (31%) množství tuku byly zaznamenány v larvách obrovských obilních červů a larvách běžných jedů. Množství chitinu se pohybovalo od 6% do 13%. Na základě těchto hodnot, které jsou srovnatelné s hodnotami, které jsou známy z jiných potravinářských hmyzu chovaných v různých oblastech světa, by jedlé druhy chované na Sumatře mohly být zdroji potravin, které by mohly pomoci odstranit hlad a podvýživu.
