Goldfish swimming was analysed quantitatively to determine if it exhibits distinctive individual spatio-temporal patterns. Due to the inherent variability in fish locomotion, this hypothesis was tested using five nonlinear measures, complemented by mean velocity. A library was constructed of 75 trajectories, each of 5 min duration, acquired from five fish swimming in a constant and relatively homogeneous environment. Three nonlinear measures, the 'characteristic fractal dimension' and 'Richardson dimension', both quantifying the degree to which a trajectory departs from a straight line, and 'relative dispersion', characterizing the variance as a function of the duration, have coefficients of variation less than 7%, in contrast to mean velocity (30%). A discriminant analysis, or classification system, based on all six measures revealed that trajectories are indeed highly individualistic, with the probability that any two trajectories generated from different fish are equivalent being less than 1%. That is, the combination of these measures allows a given trajectory to be assigned to its source with a high degree of confidence. The Richardson dimension and the 'Hurst exponent', which quantifies persistence, were the most effective measures.

Long-term potentiation (LTP), the increase in synaptic strength evoked by high-frequency stimulation, is often considered to be a cellular model for learning and memory. The validity of this model depends on the assumptions that physiological stimuli can induce LTP in vivo and that the resulting synaptic modifications correlate with behavioural changes. However, modifiable synapses are generally embedded deep in complex circuits. In contrast, the goldfish Mauthner (M)-cell and its afferent synapses are easily accessible for electrophysiological studies, and firing of this neuron is sufficient to trigger fast escape behaviour in response to sudden stimuli. We have previously shown that tetanic stimulation can induce LTP of the feedforward inhibitory synapses that control the excitability of the M-cell. Here we report that natural sensory stimulation can induce potentiation of this inhibitory connection that resembles the LTP induced by afferent tetanization. Furthermore, comparable acoustic stimulation produced a parallel decrease in the probability of the sound-evoked escape reflex. Thus we demonstrate for the first time, to our knowledge, a behavioural role for the long-term synaptic strengthening of inhibitory synapses.

1. The question of whether active uptake limits the duration of action of the inhibitory transmitter glycine has been addressed in vivo at inhibitory synapses on the goldfish Mauthner (M) cell. The kinetics of inhibitory postsynaptic potentials and inhibitory postsynaptic currents (IPSCs) evoked antidromically and by eighth-nerve stimulation were recorded in control and in conditions expected to block glycine uptake or slow its diffusion. Theoretical considerations, based on simulated quantal currents, predicted that if diffusion was slow, rapid uptake of glycine would be required and its block would prolong the synaptic responses. 2. Temperature coefficient values for IPSC decay time constants (tau S) are in the range of 2.0 for temperatures between 15 and 23 degrees C, suggesting that diffusion is not the rate-limiting step. 3. Li+, Ch+, or N-methyl-D-glucamine were substituted for 80% of the Na+ in the extracellular fluid to analyze the effects of blocking the Na(+)-dependent glycine uptake. These procedures enhanced the maximum inhibitory shunt produced by glycine iontophoresis, leading to the suggestion that uptake may buffer the concentration of the transmitter in the cleft. In contrast, the Na+ substitutes had no effect on the tau of the recurrent collateral IPSC, which involves synchronous activation of a pool of interneurons and has a monoexponential decay (tau approximately 10-11 ms). 4. The decay phase of the disynaptic IPSCs produced by stimulating the contralateral eighth nerve has fast and slow components, with a prolonged tail lasting up to 100 ms, particularly in the case of repetitive nerve stimulation. The tail is inhibitory, as revealed by its shunt of the antidromic action potential, and it is at least partially Cl- dependent. However, it can be accelerated by superfusion with the glutamate receptor antagonists 6-cyano-7-nitroquinoxaline-2,3-dione (CNQX) and DL-2-amino-5-phosphonopentanoic acid (APV). In the presence of these blockers, the IPSC decay remains biexponential (tau fast = 5.2 and 5.9 ms, tau slow = 94 and 130 ms for single and burst stimuli, respectively). Blocking uptake in this condition did not modify tau fast or tau slow. 5. We conclude that an active uptake mechanism does not shape glycinergic IPSCs, including the longer-lasting components that might include a contribution due to persistence of the transmitter. Rather, diffusion alone is sufficient to remove glycine at a rate faster than channel unbinding. 6. To test whether glycine might diffuse to adjacent excitatory synapses and enhance activation of N-methyl-D-aspartate receptors, CNQX and APV were applied locally, by pressure, to the M cell soma, but they had no effect on the prolonged decay of eighth-nerve-evoked responses. Thus the effects of the antagonists when added to the superfusate are exerted at the network level.

Synapses are complex arrangements of pre- and postsynaptic differentiations involved in neural communication. A key element in this synaptic transmission is the presynaptic active zone where the release of neurotransmitter occurs. Active zones can be visualized and analyzed after staining with ethanolic phosphotungstic acid (EPTA) on semithin (0.5 micron) sections. This staining has been used in association with postembedding immunogold labeling for the neurotransmitters glycine or GABA, to investigate the organization of chemically defined inhibitory active zones, viewed in their full extent, on different regions of the goldfish Mauthner (M-) cell. With this approach, a marked variability in size and shape was observed for the release sites contacting the different parts of the postsynaptic neuron. In the axon cap and on the soma, glycinergic afferent terminals have small presynaptic grids (0.066 +/- 0.029 micron2, n = 30 and 0.076 +/- 0.037 micron2, n = 46, respectively). These grids are quite circular and they include 12 to 13 presynaptic dense projections (PDPs). The situation is different on the lateral dendrite, where glycinergic and GABAergic active zones display a greater variability in their surface areas (mean = 0.147 +/- 0.100 micron2, n = 115 and 0.139 +/- 0.080 micron2, n = 125, respectively), and their number of PDPs (mean = 19 +/- 9) per individual grid. Similarly, the shape of the release sites over the dendrite is more complex (annular, horseshoe-shaped) when compared to those on the soma. These differences of dendritic versus somatic release sites could represent a structural basis to maximize the shunting effect of glycinergic and GABAergic inhibitory junctions, i.e., close to excitatory inputs. We also observed that the proportion of endings containing 1 or more active zones also varies. More precisely, 96% and 82% of glycinergic terminals in the axon cap and on the soma, respectively, display only one active zone. On the dendrite, their proportion falls to 65.5% for both glycine- and GABA-containing boutons. The remaining inhibitory terminals contain 2 (30%) and 3 to 4 (4.5%) presynaptic grids. These results reveal a greater variability of morphology and organization of the inhibitory release sites at dendritic versus somatic locations. The functional significance of this observation for the synaptic transmission is discussed.

1. Presynaptic activity in the inhibitory network impinging on the Mauthner (M-) cell was investigated in the goldfish medulla in vivo using extra- and intracellular recordings. The inhibitory presynaptic volley elicited by stimulation of the contralateral vestibular nerve consisted of multiple successive peaks at high frequency (up to 1,000 Hz). Less pronounced multicomponent responses were recorded after antidromic activation of the M-cell. Such high-frequency "oscillatory" field potentials also occurred spontaneously. 2. In intracellular recordings, a subset of inhibitory interneurons showed evoked and spontaneous burst discharge. Burst action potentials were correlated with the peaks in the extracellular volley, suggesting that repetitive firing of these cells is synchronized. Nonbursting cells, on the other hand, fired single action potentials in response to vestibular stimuli and were not activated via the M-cell collateral network. 3. Bursting cells were determined morphologically to be part of the feedback inhibitory circuit. Their responses to stimulation of the contralateral vestibular nerve thus suggest the existence of a crossed excitatory pathway to these interneurons. 4. Vestibular-evoked excitatory postsynaptic potentials (EPSPs) in bursting interneurons had a short latency of 0.781 +/- 0.08 ms (mean +/- SD, n = 18) but reached threshold at 2.25 +/- 1 ms (n = 21). These characteristics are suggestive of a chemically mediated EPSP. Indeed, the evoked synchronous repetitive activity of these cells was prevented by superfusion with excitatory amino-acid receptor antagonists. 5. Bursting neurons showed several characteristics that differentiate them from nonbursting cells, including brief action potentials, plateau responses, and intense spontaneous subthreshold activity. 6. With extracellular recordings, tetanization of contralateral vestibular primary afferents evoked a long-lasting potentiation of oscillatory population responses in 11 of 27 cases. Furthermore in three experiments, the frequency of occurrence of spontaneous bursts was enhanced and a similar facilitation was detected at the intracellular level. 7. We conclude that a subset of interneurons in this inhibitory network is capable of repetitive discharges and that evoked as well as spontaneous firing in this population is synchronized. Although electrical coupling between interneurons may mediate synchronization and intrinsic membrane properties may promote burst activity, our data suggest strongly that repetitive firing requires chemically mediated transmission. Furthermore they indicate that the mechanisms underlying evoked as well as spontaneous bursting in this population show activity-dependent plasticity.

Two neuron types contact the Mauthner cell (M cell) in the axon cap, a specialized region of high electrical resistance surrounding the initial segment of the M cell axon. One type produces a mixed electrical and chemical inhibition of the M cell. The second sends axons into the central core of the axon cap, where they spiral around the initial segment making both conventional synapses and gap junction contacts. The origin and synaptic effects of these spiral fibers have not been studied previously. When goldfish M cells were filled with Lucifer yellow, presynaptic spiral fibers were seen in the axon cap. These fibers could be traced back through the medial longitudinal fasciculus to their somata, near the contralateral fifth nerve motor nucleus. The same somata were labeled by horseradish peroxidase injected extracellularly into the axon cap. Recordings were made in the axon cap and the M cell after stimulation of hindbrain areas near the spiral fiber somata and axons. Extracellularly, a negative potential was observed close to the termination of the spiral fibers and termed the spiral fiber potential (SFP). Intracellularly, a graded, short latency depolarization of the M cell corresponded to the SFP and could cause the M cell to spike. This depolarization did not shunt the membrane, indicating that it may be produced through gap junctions. Intracellular responses to hindbrain stimulation also had a chloride-dependent, second component that shunted the membrane during paired-pulse testing. This inhibitory second component was probably evoked by cells other than the spiral fiber cells themselves.

The effect of partial glycinergic denervation on the cellular distribution of the 93 kDa peripheral polypeptide associated with the glycine receptor was studied at the level of the teleost Mauthner cell, an identified neuron of the goldfish brain (Carassius auratus). Previous studies using monoclonal antibodies raised against purified glycine receptors and immunoperoxidase staining have shown that these proteins are localized in clusters on the entire surface of this neuron. Specifically, the 93 kDa polypeptide was situated only on the cytoplasmic side of the postsynaptic membrane facing active zones. Unilateral electrolytic lesions of the vestibular complex caused the degeneration of some glycinergic afferents to this neuron. When the first signs of this response appeared, 3 d after the surgery, there was also a change in the ultrastructural distribution of the 93 kDa polypeptide in the deafferented cell. The synaptic protein apposed to degenerating axons did not spread onto adjacent extrasynaptic membranes, and it disappeared a few hours after the disruption of its presynaptic element. At the same time, a cytoplasmic immunoreactivity appeared as randomly distributed clusters in the deafferented Mauthner cell; these aggregates, not seen in control preparations, were never found inside membrane-bound organelles. In some preparations these clusters were localized along arrays at a relatively constant distance from the plasma membrane. The intracellular immunoreaction product was found in the soma and the initial part of the dendrites, gradually decreasing in number and intensity toward the extremities of these processes. At later postoperative stages, 10-15 d after surgery, the 93 kDa immunoreactivity remained only at postsynaptic membranes facing intact terminals. Similar alterations following denervation were observed in reticular neurons, at the level at which degenerating presynaptic terminals were also detected. In contrast, continuous 3-d blockade of synaptic transmission by strychnine, an antagonist of the glycine receptor, had no effect on either the distribution of the surface receptor clusters, or the 93 kDa peripheral protein linked to these receptors. Taken together, our results suggest that the ultrastructural distribution of the glycine receptor complex is regulated by "trophic" factors rather than by transmitter-evoked synaptic activity.

The distributions of gamma-aminobutyric acid (GABA)- and serotonin (5-HT)-containing terminals impinging on the surface of the Mauthner (M-) cell were studied at the light microscopic level using double immunofluorescent labeling and were compared with that of the glycine receptor. The latter was visualized indirectly, using a monoclonal mouse antibody which recognizes its 93-kDa associated protein. This neuron has two large principal dendrites: one extending ventrorostrally (ventral dendrite) and the other dorsolaterally (lateral dendrite). There are also two other classes of smaller processes: one that projects ventrally (small ventral dendrites) and one penetrating in the axon cap (cap dendrites), a peculiar neuropil surrounding the initial segment of the M-cell axon. A cellular regionalization of these afferent systems was found: GABA boutons, labeled for glutamic acid decarboxylase (GAD), were localized preferentially on the lateral dendrite while 5-HT-filled endings predominated on the ventral one. The density of these two classes of inputs was comparable in the other areas of the M-cell: less of their terminals were in contact with the soma outside the axon cap, and more numerous boutons, which presented either GABA or 5-HT immunoreactivities, were apposed to the small ventral dendrites. This preferential pattern of innervation differed with the ubiquitous presence of glycine receptor clusters on the M-cell membrane. Finally no evidence of a colocalization of GABA and 5-HT in afferent endings was detected at any portion of the M-cell.(ABSTRACT TRUNCATED AT 250 WORDS)

A monoclonal antibody was raised against dissected Mauthner cells of goldfish, Carassius auratus. The immunoglobulin (mAb 222C2) recognized in this neuron a determinant that was localized on the soma of the Mauthner cell in front of the axon hillock and on the dorsomedial portion of the initial third of its ventral dendrite. When observed with electron microscopy, the staining was associated with polyribosomes and with the reticulum, close to the Golgi cisternae. The antibody also labelled other large neurons (10-40 microns) of the nuclei reticularis superior, medialis and inferior. In these cells, patchy immunolabelled elements could be detected, dispersed within cytoplasm. They did not exhibit the characteristic topological distribution observed in the Mauthner cell. On the basis of their size and location, this group of neurons may send axons to the spinal cord. No staining was observed in other areas of the brainstem, or in other structures such as the cerebellum or the optic tectum. The expression of this antigenic molecule in Mauthner and reticular cells suggests that these two sets of neurons are functionally and/or ontogenetically related. Although the molecular and functional characteristics of the antigenic molecule have not been determined, this antibody should be a useful marker for further developmental studies.

The presence of glycine receptors was examined with a monoclonal antibody and indirect immunofluorescence on reticular neurons of the goldfish (Carassius auratus) brainstem. Images of thin (0.6 microns) optical sections were recorded from 80 microns thick specimen with a confocal microscope thus obviating the need for mechanical slicing. Due to the reduced out-of-focus noise, high resolution was obtained. Lookthrough projections were computer generated. Compared with classical methods involving serial sectioning, this approach allowed the analysis of the subcellular distribution of this receptor with a considerable gain of time and increased resolution. On the Mauthner cell, an identified reticulo-spinal neuron, we found, that the size of glycine receptor microdomains varies depending on the cellular localization, i.e. somatic or dendritic. Furthermore the intensity of fluorescence was uneven within individual clusters, probably reflecting differences in receptor concentration. These heterogeneities may influence the variance of synaptic inhibitory noise in different regions of the Mauthner cell.

Three monoclonal antibodies were used to examine with immunocytochemistry the distribution of the glycine receptors and the related 93kd polypeptide in the adult goldfish brain (Carassius auratus). One immunoglobulin recognizes the 48kd strychnine-binding subunit of the receptor and the two others bind to the peripheral 93kd polypeptide which is coupled to the receptor molecules. Immunofluorescent spots were visualised with all three antibodies on the somatic and dendritic membrane of the cells. A differential intensity of immunofluorescence was detected in the three different brain regions examined: the brainstem, the cerebellum and the telencephalon. For both proteins, the highest fluorescence was observed in the brainstem, particularly on reticular and vestibular neurons. In the cerebellum both the density and the intensity of labelling was low. At the level of the Mauthner cell, an identified bulbar command neuron, the distribution of glycinergic receptors was identical with that of the 93kd polypeptide. Both subunits were visualized on the somatic and dendritic membrane up to their extremities. These proteins appear to be co-localized in neurons other than the Mauthner cell, since they are always co-expressed in the same brain region. Their distributions are comparable with that observed in the mouse and rat nervous system as reported from autoradiographic localization of [3H]strychnine binding.

The morphology and distribution of the serotoninergic (5-HT) input to the Mauthner cell (M cell) of a teleost, Carassius auratus, were analyzed at the light microscopic level. Immunohistochemical methods revealed that 1) most fibers innervating the M cell originate from the ventral and lateroventral regions of the rhombencephalon; 2) two groups of fibers contribute to this innervation, thick ones (type I, 0.4-0.7 microns in diameter) with terminal endings and thin ones (type II, less than 0.2 microns) that issue numerous beaded varicosities 4-10 microns from the target cell and only occasional side endings contacting it; 3) the density of immunoreactive profiles is uneven over the whole cell and predominates on the ventral dendrite; and 4) the two sets of axons, although overlapping, do not have the same distribution. Specifically, both classes are present on the ventral dendrite, whereas type II fibers are the only ones observed on the soma, in the region of the initial segment of the axon, and in the vicinity of the lateral dendrite. Functionally identified inputs on the M cell also have a regionalized distribution, depending, for example, on whether they belong to excitatory or inhibitory networks. Thus we propose that 5-HT inputs have specific influences that are a function of their respective localization.

1. Continuous segments of synaptic noise were recorded, in 12 experiments, from the voltage-clamped goldfish Mauthner (M-)-cell soma during depolarizations beyond the Cl- equilibrium potential so that spontaneous inhibitory postsynaptic currents (IPSCs) were outward going, i.e., opposite in polarity to excitatory transients. Inhibitory components constituting the noise were detected with a program based on their expected waveforms, and their amplitude distributions were analyzed according to a quantal model to determine whether these responses, presumably because of asynchronous impulses in a population of presynaptic cells, were integral multiples of the same minimal unit. 2. Synaptic noise in the M-cell is predominantly inhibitory, as it is abolished by the glycine antagonist strychnine. IPSC amplitudes varied by 20-fold or more and about one-third were grouped in bursts, with one or several components often occurring during the falling phase of a preceding one. To measure individual amplitudes, a base-line correction procedure was designed to extrapolate the decay of the leading IPSC, on the assumption that it was exponential with a time constant, tau, equal to that obtained by fitting averaged waveforms of distinct IPSCs recorded in quiet periods. Currents were expressed in nanoamperes and, for comparison between experiments, as percentages of the full-sized IPSC evoked by activation of the collateral network. 3. The probability distribution functions (PDFs) of inhibitory events generated with such an analysis had from 5 to 12 clear and equally spaced peaks, larger transients not being included because of their sparsity. These PDFs were fit with a sum of Gaussians, on the assumption that all peaks were integral multiples of the smallest unit and had the same standard deviation, sigma. The fits were statistically satisfactory, according to a chi 2 test, and mean quantal size was 0.63 +/- 0.17% (SD; range, 0.42-1.0%) of the collateral IPSC. 4. In eight experiments, tetrodotoxin (TTX) was subsequently applied topically to block presynaptic impulses and isolate "miniature" responses because of single exocytotic events. It had no apparent nonspecific effects, as indicated by the constancy of M-cell input conductance (8.01 +/- 3.11 microS vs. 7.90 +/- 2.89 microS). Amplitude distributions of residual IPSCs were unimodal and Gaussian, with their mean size (0.60 +/- 0.19%) and average rise time (0.548 +/- 0.128 ms) the same as those of the first peaks of control histograms. The latter parameter was also comparable with that of unitary IPSCs studied previously. These results thus confirmed that spontaneous and evoked events are due to transmitter release from the same afferent population.(ABSTRACT TRUNCATED AT 400 WORDS)

Intracellular staining with horseradish peroxidase (HRP) allows the analysis of the extent and diversity of axonal field, as the Golgi techniques did for dendritic fields. In this study, we have used such HRP injections to investigate possible rules of construction that underlie the variability in the observed morphological patterns of axons. The Mauthner cell of the teleost provides a suitable material for such a work since it receives inhibitory inputs from two distinct classes of cells that can be identified physiologically prior to their intracellular staining: those that contribute to a recurrent collateral network and those that are part of the commissural vestibulovestibular pathway. The distribution of their terminal boutons over the M-cell surface was quantified and their axonal arborizations were analyzed topologically with the help of a centripetal method of terminal ordering. The results indicated that both types of interneurons have qualitatively a similar distribution of boutons over this target, encompassing the soma, initial portions of the main dendrites, and the cap-dendrites, and that furthermore, the terminals have a tendency to form clusters, the proportion of which is relatively constant regardless of the total number of boutons established by a given afferent cell. In respect to their projection on the M-cell, the two populations differ mainly in the number of established contacts, which averaged 44.1 +/- 28.2 (n = 10) and 14.4 +/- 11.3 (n = 43) for collateral and commissural interneurons, respectively. These differences reflect the variations in axonal arborizations. A topological analysis has revealed that branching occurs mainly as bifurcations, whereas in contrast three or four segments issuing from a branch point are only occasionally observed at the level of the last segment; for each cell, the distribution of boutons within orders is not random and rather follows an unimodal distribution, centered on the mean order (theta); the terminals of each subset, or class, of neurons have their own pattern of distribution within orders; and finally the mean order (theta) is linked to the number of terminal segments (nT) by the relation theta = 1.28 log2 nT. This parameter is of importance: it characterizes the axonal arborization and it allows one to predict the number of terminal segments of individual neurons. Similar analysis of camera lucida reconstruction of axonal arborizations of various cell types studied by other investigators suggests that this relation can be generalized.(ABSTRACT TRUNCATED AT 400 WORDS)

1. Passive hyperpolarizing potentials (PHPs) have been recorded intracellularly from goldfish medullary neurons in the vicinity of the Mauthner cell (M-cell). They are evoked when this cell is activated antidromically by stimulation of the spinal cord, and orthodromically via the ipsilateral eighth nerve; when appropriately timed they block or delay spikes induced both directly and transsynaptically. 2. Since the PHPs and the M-cell spike have the same latency, time course, and all-or-none character, they cannot be generated by chemically mediated synaptic transmission. This conclusion is further supported by the evidence that PHP amplitude and time course are independent of membrane potential. 3. The analysis of the mechanism underlying PHP generation has been based on the hypothesis that they are brought about by the extracellular currents flowing to the axon cap during an M-cell action potential. Specifically, it was postulated that some of this current is channeled back to the axon cap region intracellularly through processes of PHP-exhibiting neurons, and that these cells are passively hyperpolarized by the associated inward transmembrane current flow. This model would require that PHP-exhibiting neurons send processes into the axon cap. This hypothesis is confirmed by the following: a) When the PHP is timed to occur during the conductance increase associated with a spike after hyperpolarization, it is reduced, as would be expected for a passive current flow across a membrane resistance. b) PHPs are not found in all medullary neurons in the vicinity of the M-cell, but rather in a specific neuronal population. c) PHP-exhibiting neurons have been identified following Procion yellow injections; as predicted, they issue one process, presumably the axon, which projects toward the M-cell axon cap area. d) The PHP can be stimulated by passing a cathodal current from a microelectrode located in the axon cap; it is not mimicked when the cathodal electrode is moved outside this region. The currents necessary to mimic a PHP are comparable to the estimated current flowing back to the axon cap during an M-cell action potential. 4. The input resistance of PHP-exhibiting neurons is in the range of 4 M alpha, and their estimated specific membrane resistance is in the range of 900-2,000 alpha-cm-2, which is not an unusually low value for neuronal membranes. By contrast, the intracellular channeling of current during a PHP can rather be attributed to a high extracellular tissue resistance within the axon cap, which was found to be at least 2.5 times that of the surrounding medullary tissue..

Single and double electrode voltage clamp techniques have been used to analyse the leakage conductance of the goldfish Mauthner cell. The results indicate that the input conductance of this neuron is maximal at the resting potential, having an average value of 4.68 microS. Approximately 50% of this conductance is voltage dependent and could be inactivated by large hyperpolarizing command pulses (30-50 mV magnitude) of 20-35 ms duration. The magnitude and apparent time constant of the inactivation are both functions of membrane potential, such that the relaxation of the inward current increased and occurred more rapidly for greater hyperpolarizations. The presence of instantaneous inward tail currents when the imposed voltage steps were terminated and membrane potential was returned to the resting level indicated that the conductance mechanism inactivated during hyperpolarization had generated a small outward current at the resting level. Thus, we propose that a major fraction of the Mauthner cell's input conductance is a voltage-dependent K+ conductance. Hyperpolarizing inactivation is a feature of an inward K+ rectifier in other cell types, and when the recording microelectrode was located in the Mauthner cell lateral dendrite, it was possible to demonstrate characteristics consistent with rectification, namely a low conductance for outward depolarizing currents and a high conductance for inward currents. The inward rectifier of the Mauthner cell is different from that of other neurons in that it is already maximally activated at the resting potential and therefore is a major determinant of that parameter. This work was carried on to analyze the presynaptic components of synaptic efficacy, which is designated by the term of synaptic strength. To assess the relations between synaptic strength and innervation density, the properties of unitary Cl(-)-dependent inhibitory postsynaptic potentials (IPSPs) evoked in the potentials (IPSPs) evoked in the goldfish Mauthner (M-) cell by single impulses in individual presynaptic cells, including quantal release parameters, were compared with the histological features of the same neurons, determined from their reconstructions after intracellular injection with horseradish peroxidase (HRP). Because the M-cell is a stereotyped target neuron, comparison of the synaptic strength from different experiments was accomplished by using as a quantitative measure of this parameter the mean unitary IPSP amplitude normalized with respect to the reversal potential for Cl- (i.e., the driving force). In 108 experiments at low stimulus frequency, the majority of the normalized responses (63%) were grouped in a rather narrow range, varying about fourfold, or from 1.5 to 6% of the driving force, with results from stained (n = 46) and unstained (n = 62) cells being the same. In contrast, for the same restricted set of responses, the number of presynaptic terminals (histological n) encompassed a larger range, varying from 3 to 52. Impulses in neurons with quite different complements of terminal boutons could evoke similarly sized, normalized IPSPs, and these two parameters were poorly correlated, with there being, at most, a tendency for the responses to increase with histological n for small values of the latter. Quantal fluctuations in IPSP amplitudes were analyzed according to a binomial model having three parameters, p, which is the probability of release, n, which is the number of releasing units and was previously shown to equal the number of presynaptic boutons or active sites, and q, or the quantal size. The normalized quantal size varied randomly, with a mean value of 0.51% (SD = 0.20) and was relatively independent of n. In contrast, the distribution of p, which ranged from 0.17 to 0.74 (mean = 0.40, SD = 0.155), was skewed to the right; this parameter tended to decrease as a function of increasing n. The normalized unitary inhibitory conductance (g'IPSP) underlying an IPSP is equal to the product of npg'q, where g'q is the normalized quantal conductance.(ABSTRACT TRUNCATED AT 400 WORDS)
