A fourth of the global seabed sediment volume is buried at depths where temperatures exceed 80 °C, a previously proposed thermal barrier for life in the subsurface. Here, we demonstrate, utilizing an extensive suite of radiotracer experiments, the prevalence of active methanogenic and sulfate-reducing populations in deeply buried marine sediment from the Nankai Trough subduction zone, heated to extreme temperature (up to ~120 °C). The small microbial community subsisted with high potential cell-specific rates of energy metabolism, which approach the rates of active surface sediments and laboratory cultures. Our discovery is in stark contrast to the extremely low metabolic rates otherwise observed in the deep subseafloor. As cells appear to invest most of their energy to repair thermal cell damage in the hot sediment, they are forced to balance delicately between subsistence near the upper temperature limit for life and a rich supply of substrates and energy from thermally driven reactions of the sedimentary organic matter.

Carbon cycling in Peruvian margin sediments (11°S and 12°S) was examined at 16 stations from 74 m on the inner shelf down to 1024 m water depth by means of in situ flux measurements, sedimentary geochemistry and modeling. Bottom water oxygen was below detection limit down to ca. 400 m and increased to 53 μM at the deepest station. Sediment accumulation rates and benthic dissolved inorganic carbon fluxes decreased rapidly with water depth. Particulate organic carbon (POC) content was lowest on the inner shelf and at the deep oxygenated stations (<5%) and highest between 200 and 400 m in the oxygen minimum zone (OMZ, 15-20%). The organic carbon burial efficiency (CBE) was unexpectedly low on the inner shelf (< 20%) when compared to a global database, for reasons which may be linked to the frequent ventilation of the shelf by oceanographic anomalies. CBE at the deeper oxygenated sites was much higher than expected (max. 81 %). Elsewhere, CBEs were mostly above the range expected for sediments underlying normal oxic bottom waters, with an average of 51 and 58% for the 11° Sand 12° S transects, respectively. Organic carbon rain rates calculated from the benthic fluxes alluded to a very efficient mineralization of organic matter in the water column, with a Martin curve exponent typical of normal oxic waters (0.88 ±0.09). Yet, mean POC burial rates were 2-5 times higher than the global average for continental margins. The observations at the Peruvian margin suggest that a lack of oxygen does not affect the degradation of organic matter in the water column but promotes the preservation of organic matter in marine sediments. Arkona Basin (southwestern Baltic Sea) is a seasonally-hypoxic basin characterized by the presence of free methane gas in its youngest organic-rich muddy stratum. Through the use of reactive transport models, this study tracks the development of the methane geochemistry in Arkona Basin as this muddy sediment became deposited during the last 8 kyr. Four cores are modeled each pertaining to a unique geochemical scenario according to their respective contemporary geochemical profiles. Ultimately the thickness of the muddy sediment and the flux of particulate organic carbon are crucial in determining the advent of both methanogenesis and free methane gas, the timescales over which methanogenesis takes over as a dominant reaction pathway for organic matter degradation, and the timescales required for free methane gas to form. Anaerobic methane-oxidizing microbial communities in sediments at cold methane seeps are important factors in controlling methane emission to the ocean and atmosphere. Here, we investigated the distribution and carbon isotopic signature of specific biomarkers derived from anaerobic methanotrophic archaea (ANME groups) and sulphate-reducing bacteria (SRB) responsible for the anaerobic oxidation of methane (AOM) at different cold seep provinces of Hydrate Ridge, Cascadia margin. The special focus was on their relation to in situ cell abundances and methane turnover. In general, maxima in biomarker abundances and minima in carbon isotope signatures correlated with maxima in AOM and sulphate reduction as well as with consortium biomass. We found ANME-2a/DSS aggregates associated with high abundances of sn-2,3-di- O-isoprenoidal glycerol ethers (archaeol, sn-2-hydroxyarchaeol) and specific bacterial fatty acids (C16:1ω5c, cyC17:0ω5,6) as well as with high methane fluxes ( Beggiatoa site). The low to medium flux site ( Calyptogena field) was dominated by ANME-2c/DSS aggregates and contained less of both compound classes but more of AOM-related glycerol dialkyl glycerol tetraethers (GDGTs). ANME-1 archaea dominated deeper sediment horizons at the Calyptogena field where sn-1,2-di- O-alkyl glycerol ethers (DAGEs), archaeol, methyl-branched fatty acids ( ai-C15:0, i-C16:0, ai-C17:0), and diagnostic GDGTs were prevailing. AOM-specific bacterial and archaeal biomarkers in these sediment strata generally revealed very similar δ13C-values of around −100. In ANME-2-dominated sediment sections, archaeal biomarkers were even more 13C-depleted (down to −120), whereas bacterial biomarkers were found to be likewise 13C-depleted as in ANME-1-dominated sediment layers (δ13C: −100). The zero flux site ( Acharax field), containing only a few numbers of ANME-2/DSS aggregates, however, provided no specific biomarker pattern. Deeper sediment sections (below 20 cm sediment depth) from Beggiatoa covered areas which included solid layers of methane gas hydrates contained ANME-2/DSS typical biomarkers showing subsurface peaks combined with negative shifts in carbon isotopic compositions. The maxima were detected just above the hydrate layers, indicating that methane stored in the hydrates may be available for the microbial community. The observed variations in biomarker abundances and 13C-depletions are indicative of multiple environmental and physiological factors selecting for different AOM consortia (ANME-2a/DSS, ANME-2c/DSS, ANME-1) along horizontal and vertical gradients of cold seep settings. Oxygen minimum zones (OMZ) are key regions for fixed nitrogen loss in both the sediments and the water column. During this study, the benthic contribution to N cycling was investigated at ten sites along a depth transect (74–989 m) across the Peruvian OMZ at 12°S. O 2 levels were below detection limit down to ~500 m. Benthic fluxes of N 2 , NO 3 – , NO 2 – , NH 4 + , H 2 S and O 2 were measured using benthic landers. Flux measurements on the shelf were made under extreme geochemical conditions consisting of a lack of O 2 , NO 3 – and NO 2 – in the bottom water and elevated seafloor sulphide release. These particular conditions were associated with a large imbalance in the benthic nitrogen cycle. The sediments on the shelf were densely covered by filamentous sulphur bacteria Thioploca , and were identified as major recycling sites for DIN releasing high amounts of NH 4 + up to 21.2 mmol m −2 d −1 that were far in excess of NH 4 + release by ammonification. This difference was attributed to dissimilatory nitrate (or nitrite) reduction to ammonium (DNRA) that was partly being sustained by NO 3 – stored within the sulphur oxidizing bacteria. Sediments within the core of the OMZ (ca. 200–400 m) also displayed an excess flux of N of 3.5 mmol m −2 d −1 mainly as N 2 . Benthic nitrogen and sulphur cycling in the Peruvian OMZ appears to be particularly susceptible to bottom water fluctuations in O 2 , NO 3 − and NO 2 − , and may accelerate the onset of pelagic euxinia when NO 3 − and NO 2 − become depleted. Abstract: Many biological seep studies focused on the distribution, structure, nutrition and food web architecture of seep communities as well as on their interaction with the seep geochemistry. However, overall respiration at cold seeps received only little attention. We conducted in-situ oxygen flux measurements in combination with ex-situ oxygen micro-profiles, respiration measurements, as well as rate determinations of microbial methane and sulfate turnover to assess respiration pathways as well as carbon turnover at a seep habitat that was recently discovered alongside the Hikurangi Margin offshore northern New Zealand. This habitat is dominated by dense beds of tube-building, heterotrophic ampharetid polychaetes. Average total oxygen uptake (TOU) from this habitat was very high (83.7 mmol m−2 day−1). TOU at a non-seep reference site ranged between 2.7 and 5.8 mmol m−2 day−1. About 37% (30.8 mmol m−2 day−1) of the average TOU was consumed by ampharetids. Considering mean diffusive oxygen uptake (8.5 mmol m−2 day−1) the remaining fraction of ~53% of the TOU (44.4 mmol m−2 day−1) might be explained by respiration of epibenthic organisms as well as aerobic methane and sulfide oxidation at the sediment–water interface. The strongly negative carbon isotopic signatures (−52.9±5‰ VPDB) of the ampharetid tissues indicate a methane derived diet. However, carbon production via anaerobic oxidation of methane (AOM) was too low (0.1 mmol C m−2 day−1) to cover the mean carbon demand of the ampharetid communities (21 mmol C m−2 day−1). Likely, organic carbon generated via aerobic methane oxidation represents their major carbon source. This is in contrast to other seep habitats, where energy bound in methane is partly transferred to sulfide via AOM and finally consumed by sulfide-oxidizing chemoautotrophs providing carbon that subsequently enters the benthic food web.
