Soybean (Glycine max (L.) Merr.) is one of the important crops in Canada and has the potential to expand its production further north into the Canadian Prairies. Such expansion, however, requires the search for adapted soybean germplasm useful for the development of productive cultivars with earlier maturity and increased protein concentration. We initiated several research activities to characterize 848 accessions of the soybean collection conserved at Plant Gene Resources of Canada (PGRC) for maturity, oil and protein concentration, and genetic distinctness. The characterization revealed a wide range of variations present in each assessed trait among the PGRC soybean accessions. The trait variabilities allowed for the identification of four core subsets of 35 PGRC soybean accessions, each specifically targeted for early maturity for growing in Saskatoon and Ottawa, and for high oil and protein concentration. The two early maturity core subsets for Saskatoon and Ottawa displayed days to maturity ranging from 103 to 126 days and 94 to 102 days, respectively. The two core subsets for high oil and protein concentration showed the highest oil and protein concentration from 25.0 to 22.7% and from 52.8 to 46.7%, respectively. However, these core subsets did not differ significantly in genetic distinctness (as measured with 19,898 SNP markers across 20 soybean chromosomes) from the whole PGRC soybean collection. The objective of this article was to investigate the morphological and molecular characterization of Oestromyia leporina (Pallas, 1778) from wild plateau pikas ( Ochotona curzoniae ) in Qinghai province, China. The third-stage larvae of O . leporina were examined by scanning electron microscopy revealing morphology characteristics of the spines on the cephalic, the thoracic segments, the abdominal segments and the spiracular plates. The coding regions of 25 cytochrome oxidase I (COI) genes of O . leporina were investigated. Eighty-one variable sites and 21 haplotypes were identified and the nucleotide and haplotype diversities were 0.04456 and 0.9767, respectively, indicating a rich genetic diversity in O . leporina . Phylogenetic analysis utilizing sequences of COI revealed two distinct lineages. These findings revealed ultrastructure and molecular characterization among the O . leporina from plateau pikas in Qinghai province, China and had implications for studying morphological identification, molecular epidemiology and population genetics of O . leporina . Genetic diversity is essential for predicting plant evolutionary potential and for formulating conservation strategies. However, little is known about the genetic variation of plains rough fescue [ Festuca hallii (Vasey) Piper], a widespread and diagnostic grass in the Fescue Prairie. We used the amplified fragment length polymorphism (AFLP) technique to assess the genetic variation of 30 fragmented populations of F. hallii across the northern Canadian Prairie and its associations with 12 geographical, fragmentation and environmental attributes. Three AFLP primer pairs were used to screen 840 samples, representing approximately 30 samples from each population, and 246 polymorphic bands were scored. The fescue plant was genetically diverse, as revealed by the proportion of polymorphic bands (0.870–0.967), the mean band frequency (0.364–0.457) and the within-population variation (69.4–85.4). The genetic variation was not highly differentiated, with only 6.5% of the total AFLP variation residing among populations. A Mantel test revealed a significant correlation between genetic and geographical distances ( r = 0.39; P < 0.004) and a spatial autocorrelation up to 60 km among populations was detected. The AFLP variation displayed a longitudinal decline and was significantly associated with environmental attributes related to moisture, indicating local adaptation. However, the AFLP variation was not significantly associated with the estimated population size and geographical distance to the nearest neighbor, suggesting that fragmentation has not generated considerable genetic impact on the fescue populations. Characterization of plant germplasm using molecular techniques is playing an increasingly important role in the management and utilization of plant genetic resources, but has its limitations in the screening of large numbers of accessions held in seed genebanks worldwide. Bulking individual plants from one accession or group to form a representative sample is a promising approach to widening the scope of a characterization, but it is not without technical problems in detecting genetic variation. This review was conducted to assess the technical pitfalls of bulking, and to evaluate the effectiveness of various bulking methods in the assessment of genetic variation and genetic relationships, and in the identification of plant germplasm. Clearly, some alleles, particularly those occurring at low frequency, may go undetected in a bulked sample, depending on the bulking methods and the molecular techniques used. As a result, genetic diversity estimates and genetic relationship inferences can be significantly biased. Germplasm identification may not be always reliable. Thus, it is imperative that the detection limit imposed by bulking be assessed for a newly initiated molecular germplasm characterization and bias be considered in interpretation of the resulting characterization data. Equally imperative is the need for continuous efforts of exploring efficient bulking procedures for the screening of large germplasm collections, particularly by the newly developed marker systems. Molecular characterization of unsequenced plant species with complex genomes is now possible by genotyping-by-sequencing (GBS) using recent next generation sequencing technologies. This study represents the first use of GBS application to sample genome-wide variants of crested wheatgrass [Agropyron cristatum (L.) Gaertn.] and assess the genetic diversity present in 192 genotypes from 12 tetraploid lines. Bioinformatic analysis identified 45,507 single nucleotide polymorphism (SNP) markers in this outcrossing grass species. The model-based Bayesian analysis revealed four major clusters of the samples assayed. The diversity analysis revealed 15.8% of SNP variation residing among the 12 lines, and 12.1% SNP variation present among four genetic clusters identified by the Bayesian analysis. The principal coordinates analysis and dendrogram were able to distinguish four lines of Asian origin from Canadian cultivars and breeding lines. These results serve as a valuable resource for understanding genetic variability, and will aid in the genetic improvement of this outcrossing polyploid grass species for forage production. Recent advances in next generation sequencing technologies make genotyping-by-sequencing (GBS) more feasible for the molecular characterization of plant germplasm with complex and unsequenced genomes. This study represents the first preliminary effort using GBS to discover genome-wide genetic variants of northern wheatgrass (Elymus lanceolatus ssp. lanceolatus (Scribn. and J. G. Sm.) Gould) plants and to assess the genetic diversity present in four cultivated and six wild accessions. The effort generated the first novel set of genomic resources and 5659 single nucleotide polymorphism (SNP) markers for this tetraploid grass. The diversity analysis revealed 8.8% of SNP variation residing among the 10 accessions and 1.9% SNP variation present between cultivated and wild accessions. The Bayesian analysis identified three major clusters of the assayed samples, and the principal coordinates analysis revealed the genetic distinctness of the two accessions collected from Nevada and Wyoming. The flow cytometry analysis confirmed the tetraploid nature of some of the assayed samples and estimated the average genome size to be 9.3–9.4 Gb for this species. These findings are useful for the genetic improvement of this native grass species for forage production and rangeland reclamation. The findings are also encouraging for the broad application of genotyping-by-sequencing in the characterization of genome-wide genetic variability in non-model polyploid plants. [ABSTRACT FROM AUTHOR]
