Phosphorus is an essential macronutrient for plant growth and development, but phosphate resources are limited and rapidly depleting due to massive global agricultural demand. This study identified two genes in the phosphate transporter 2 (PHT2) family of soybean by bioinformatics. The expression patterns of two genes by qRT-PCR at leaves and all were induced by low-phosphate stress. After low-phosphate stress, GmPHT2;2 expression was significantly higher than GmPHT2;1, and the same trend was observed throughout the reproductive period. The result of heterologous expression of GmPHT2 in Arabidopsis knockout mutants of atpht2;1 shows that chloroplasts and whole-plant phosphorus content were significantly higher in plants complementation of GmPHT2;2 than in plants complementation of GmPHT2;1. This suggests that GmPHT2;2 may play a more important role in plant phosphorus metabolic homeostasis during low-phosphate stress than GmPHT2;1. In the yeast backfill assay, both genes were able to backfill the ability of the defective yeast to utilize phosphorus. GmPHT2 expression was up-regulated by a low-temperature treatment at 4 °C, implying that GmPHT2;1 may play a role in soybean response to low-temperature stress, in addition to being involved in phosphorus transport processes. GmPHT2;1 and GmPHT2;2 exhibit a cyclic pattern of circadian variation in response to light, with the same pattern of gene expression changes under red, blue, and white light conditions. GmPHT2 protein was found in the chloroplast, according to subcellular localization analysis. Bacterium strain PJ3, isolated from wastewater and identified as Arthrobacter sp. bacterium based on its 16S rDNA gene, could use carbazole as the sole carbon, nitrogen and energy source. The genomic library of strain PJ3 was constructed and a positive clone JM109 (pUCW402) was screened out for the expression of dioxygenase by the ability to form yellow ring-fission product. A 2,3-dihydroxybiphenyl dioxygenase (23DHBD) gene of 933 bp was found in the 3360 bp exogenous fragment of pUCW402 by GenSCAN software and BLAST analysis. The phylogenetic analysis showed that 23DHBD from strain PJ3 formed a deep branch separate from a cluster containing most known 23DHBD in GenBank. Southern hybridization confirmed for the first time that the 23DHBD gene was from the genomic DNA of Arthrobacter sp. PJ3. In order to test the gene function, recombinant bacterium BL21 (pETW-8) was constructed to express 23DHBD. The expression level in BL21 (pETW-8) was highest compared with the recombinant bacteria JM1O9 (pUCW402) and strain PJ3. We observed that 23DHBD was not absolute specific. The enzyme activity was higher with 2,3-dihydroxybiphenyl as a substrate than with catechol. The substrate specificity assay suggested that 23DHBD was essential for cleavage of bicyclic aromatic compounds during the course of aromatic compound biodegradation in Arthrobactersp. strain PJ3. CAZymes play a very important role in the biotransformation of corn stalk biomass, which is an important resource for sustainable development. Pleurotus djamor can produce CAZymes related to the decomposition of corn stalk lignocellulose biomass in sole corn stalk substrate; however, little is known about their encoding genes. In order to identify CAZymes encoding genes, RNA high-throughput sequencing of P. djamor was performed in this study. The results showed that a core set of 70 upregulated genes encoding putative CAZymes were revealed. They encode 19 kinds of CAZymes in total, of which there are 4 EGLs, 8 CBHs, 5 BGLs, and 12 LPMOs related to cellulose degradation, 8 XYNs, 1 XYL, 2 AGUs, 3 ABFs, 2 AGLs, and 2 AXEs related to hemicellulose degradation, and 5 LACCs, 2 MnPs, 5 VPs, 3 CDHs, 1 AAO, 1 GOX, 1 AOX, 2 GAOXs, and 3 GLOXs related to lignin degradation. This variety suggests that CAZymes may play a very important role in decomposing the lignocellulose biomass of corn stalk. This is the first study to report the de novo transcriptome sequencing of P. djamor , which will produce a dataset of genes encoding CAZymes, thereby laying the foundation to elucidate the degradation mechanism of corn stalk biomass and boost the biotransformation of corn stalk biomass resources.
