Receptor interacting serine/threonine kinase 1 (RIPK1) mediates apoptosis by regulating the classic proapoptotic effectors Bcl-2-associated X protein (Bax) and Bcl-2 homologous antagonist/killer (Bak). Although Bcl-2-related ovarian killer (Bok) is structurally similar to Bak and Bax, it is unclear whether it mediates apoptosis in skeletal muscle ischemia reperfusion (IR) injury. We hypothesized that by regulating Bok-mediated apoptosis, inhibiting RIPK1 with necrostatin-1 would reduce skeletal muscle IR injury.Rats were randomized into four groups: sham (SM), IR, IR treated with necrostatin-1 (NI), or vehicle dimethyl sulfoxide (DI). For the IR group, the right femoral artery was clamped for 4 hours and then reperfused for 4 hours, and for the NI and DI groups, necrostatin-1 (1.65 mg/kg) and the equal volume of dimethyl sulfoxide were intraperitoneally administered prior to IR induction. The structural damage of muscle tissue and protein expression of Bok, Bcl-2, and cleaved caspase-3 were investigated, and apoptotic cells were identified with terminal dUTP nick-end labeling (TUNEL) staining. In vitro, human skeletal muscle cells (HSMCs) were exposed to 6 hours of oxygen-glucose deprivation followed by normoxia for 6 hours to establish an oxygen-glucose deprivation/reoxygenation (OGD/R) model. To determine the role of Bok, cell viability, lactate dehydrogenase (LDH) release, and flow cytometry were examined to demonstrate the effects of necrostatin-1 and Bok knockdown on the OGD/R insult of HSMCs.Necrostatin-1 pretreatment markedly reduced IR-induced muscle damage and RIPK1, Bok, and cleaved caspase-3 expression, whereas upregualted Bcl-2 expression (p0.05). Furthermore, necrostatin-1 prevented mitochondrial damage and decreased TUNEL-positive muscle cells (p0.05). In vitro, HSMCs treated with necrostatin-1 showed reduced Bok expression, increased cell viability, and reduced LDH release in response to OGD/R (p0.05), and Bok knockdown significantly blunted the OGD/R insult in HSMCs.Necrostatin-1 prevents skeletal muscle from IR injury by regulating Bok-mediated apoptosis.

Aerobic glycolysis, also known as the Warburg effect, is a hallmark of cancer cell glucose metabolism and plays a crucial role in the activation of various types of immune cells. Glyceraldehyde 3-phosphate dehydrogenase (GAPDH) catalyzes the conversion of D-glyceraldehyde 3-phosphate to D-glycerate 1,3-bisphosphate in the 6th critical step in glycolysis. GAPDH exerts metabolic flux control during aerobic glycolysis and therefore is an attractive therapeutic target for cancer and autoimmune diseases. Recently, GAPDH inhibitors were reported to function through common suicide inactivation by covalent binding to the cysteine catalytic residue of GAPDH. Herein, by developing a high-throughput enzymatic screening assay, we discovered that the natural product 1,2,3,4,6-penta-O-galloyl-β-D-glucopyranose (PGG) is an inhibitor of GAPDH with K(i) = 0.5 μM. PGG blocks GAPDH activity by a reversible and NAD(+) and Pi competitive mechanism, suggesting that it represents a novel class of GAPDH inhibitors. In-depth hydrogen deuterium exchange mass spectrometry (HDX-MS) analysis revealed that PGG binds to a region that disrupts NAD(+) and inorganic phosphate binding, resulting in a distal conformational change at the GAPDH tetramer interface. In addition, structural modeling analysis indicated that PGG probably reversibly binds to the center pocket of GAPDH. Moreover, PGG inhibits LPS-stimulated macrophage activation by specific downregulation of GAPDH-dependent glucose consumption and lactate production. In summary, PGG represents a novel class of GAPDH inhibitors that probably reversibly binds to the center pocket of GAPDH. Our study sheds new light on factors for designing a more potent and specific inhibitor of GAPDH for future therapeutic applications.

Introduction. Preeclampsia (PE) is a major contributor to maternal and foetal morbidity and mortality worldwide. It manifests as high blood pressure and proteinuria in women at more than 20 weeks of gestation. Abnormal levels of anti- and pro-angiogenesis factors are known to be associated with PE. In the present study, we aimed to determine the localisation of angiogenic factor with G patch and FHA domains 1 (AGGF1) in the placenta and to compare the expression levels of AGGF1 in the third-trimester placentas of preeclamptic and normotensive pregnancies. Materials and methods. Placental tissue samples were collected from women with PE (n = 28) and without PE (n = 28). The normotensive controls without PE were matched for gestational age at delivery with the patients with PE. The expression levels of AGGF1 in the placental tissues were evaluated using immunohistochemistry, quantitative reverse transcription polymerase chain reaction and Western blot. Results . The immunoexpression of AGGF1 was localised in the syncytiotrophoblast tissue. Notable, the mRNA and protein expression levels of AGGF1 were decreased in preeclamptic placentas as compared with the normotensive control group (P < 0.05). Discussion. Our results suggest that the decreased AGGF1 in preeclamptic placentas may be related to the pathogenesis of preeclampsia.

Background Vitamin D (VitD) can regulate immune responses and maternal VitD-deficiency can affect immune responses in the offspring. This study aimed at investigating the effects of maternal VitD-deficiency during pregnancy on Treg and Breg responses in offspring mice with house dust mite (HDM)-induced allergic airway inflammation. Methods Female BALB/c mice were randomized and fed with normal chow or VitD-deficient diet until their offspring weaned. The offspring mice were fed with normal chow and injected with vehicle or HDM to induce allergic airway inflammation. The levels of serum 25(OH)D, cytokines and infiltrate numbers as well as percentages of Tregs and Bregs in the bronchoalveolar lavage fluid (BALF) were analyzed. The relative levels of VitD receptor (VDR), VitD-binding protein (VDBP), Cytochromes P450 (CYP) 27b1, and CYP24A1 mRNA transcripts in the lungs of different groups of mice were measured. Results Maternal VitD-deficiency significantly reduced serum 25(OH)D levels in offspring mice. VitD-deficiency significantly increased the relative levels of VDR, VDBP and CYP27B1 mRNA transcripts, but decreased CYP24A1 expression in the lungs of mice. In comparison with the control mice, significantly elevated levels of pro-inflammatory cytokines, increased numbers of lymphocytes and eosinophils, but decreased levels of anti-inflammatory cytokines were detected in the BALF of VitD-deficient mice. VitD-deficiency significantly increased the frequency of Th1, Th2, Th9, Th17 cells, but decreased regulatory T (Tregs) and B cells (Bregs) in the BALF of mice with allergic airway inflammation. Conclusion Maternal VitD-deficiency lowed serum 25(OH)D levels and enhanced HDM-induced allergic airway inflammation in the offspring by impairing Breg and Treg responses.

Methyltransferase-like 3 (METTL3) was originally known to be responsible for N6-methyladenosine (m6A) modification of mRNA. Recent studies have found that METTL3 plays important roles in a variety of tumors by regulating the translation of oncogenes. However, the functional and regulating mechanisms of METTL3 in human gastric cancer have not yet been understood. Here we knocked down METTL3 in human gastric cancer cell lines, AGS and MKN45, by using shRNA transfection. RT-qPCR assay and western blotting verified the effectiveness of RNA interference on mRNA and protein levels, respectively. Then we found that METTL3 knockdown inhibited cell proliferation, migration and invasion in AGS and MKN45 cells. Moreover, METTL3 knockdown decreased Bcl2 and increased Bax and active Caspase-3 in gastric cancer cells, which suggested the apoptotic pathway was activated. Mechanistic investigation suggested that METTL3 led to inactivation of the AKT signaling pathway in human gastric cancer cells, including decreased phosphorylation levels of AKT and expression of down-stream effectors p70S6K and Cyclin D1. In conclusion, our study reveals that down-regulation of METTL3 inhibits the proliferation and mobility of human gastric cancer cells and leads to inactivation of the AKT signaling pathway, suggesting that METTL3 may be a potential target for the treatment of human gastric cancer.

PURPOSE This study investigated the frequency and absolute numbers of different subsets of peripheral blood cells, including CD4+ T cells and CD19+ interleukin (IL) 10+ B regulatory cells (Breg) and their potential association with clinical laboratory measurements in children diagnosed with simple asthma or asthma plus allergic rhinitis (AR). METHOD The frequency and numbers of peripheral blood CD4+ interferon (IFN) gamma+ T-helper (Th) type 1, CD4+ IL-4+ Th2, CD4+ IL-17A+ Th17, CD4+CD25+ Forkhead Box P3+ regulatory T cells (Treg) and CD19+ IL-10+ Bregs in 22 children with asthma, 17 children with asthma and AR, and 25 healthy controls were determined by flow cytometry. The levels of serum cytokines were determined by enzyme-linked immunosorbent assay. RESULTS In comparison with healthy controls, patients with asthma alone had significantly increased numbers of Th1, Th2, and Th17 cells, and their cytokines but decreased numbers of Tregs and Bregs, and the cytokines IL-10 and transforming growth factor beta 1. This imbalance between effector and regulatory cells and their cytokines further increased in patients with asthma and AR. The ratios of percentage effector T cells (Th1 + Th2 + Th17) to regulatory cells (Treg + Breg) were positively correlated with fractional exhaled nitric oxide but negatively correlated with forced expiratory volume in 1 second in patients with asthma or asthma plus AR. CONCLUSION The imbalance of effector T cells and regulatory cells contributed to the pathogenesis of airway inflammation of asthma and AR in children.

Objectives Circular RNAs (circRNAs) exist extensively in the eukaryotic genome. The study aimed to identify the role of hsa_circ_0008365 (Circ‐SERPINE2) in gastric carcinoma (GC) cells and its downstream mechanisms. Materials and methods Gene Expression Omnibus (GEO) database was applied to screen differentially expressed circRNAs. CircInteractome, TargetScan and miRecords websites were used to predict target relationships. qRT‐PCR and RNase R treatment were utilised to detect molecule expression and confirm the existence of circ‐SERPINE2. RNA pull‐down assay and dual‐luciferase reporter assay were performed for interaction between circRNA and miRNA or mRNA. EdU assay, colony formation assay, and flow cytometry for apoptosis and cell cycle detections were utilised to assess cell function. Western blot and immunohistochemistry (IHC) assays were applied for detection of proteins in tissues or cells. Results Circ‐SERPINE2 and YWHAZ were upregulated, and miR‐375 was downregulated in GC tissues and cells. Circ‐SERPINE2 and YWHAZ targetedly bound to miR‐375. Circ‐SERPINE2 promoted cell proliferation and cell cycle progress and inhibited cell apoptosis by sponging miR‐375 and regulating YWHAZ expression in vitro. Circ‐SERPINE2 repressed solid tumour growth through enhancing miR‐375 expression and reducing YWHAZ expression in vivo. Conclusions Circ‐SERPINE2 is a novel proliferative promoter through the regulation of miR‐375/YWHAZ. Circ‐SERPINE2/miR‐375/YWHAZ axis might provide a novel therapeutic target of GC.

Olfactory bulb, as one of sensory organs opening to the outside, is susceptible to toxic environment and easy to deteriorate. Recent studies in Parkinson’s disease (PD) patients and 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-treated monkeys have shown that abnormal α-synuclein is accumulated in the olfactory glomeruli, suggesting that the lesions of PD are not only confined to the substantia nigra (SN) but also located in the olfactory bulb. Thus, olfactory bulb might be the region of onset in PD pathogenesis and a targeted region for diagnosis and treatment of PD. However, the relationship between olfactory bulb and pathogenesis of PD remains unclear. In the present study, we investigated the inflammatory pathological alterations in olfactory bulb and the underlying mechanisms in chronic MPTP mice. Mice were treated with MPTP/P, i.e., MPTP (25 mg/kg, s.c.) plus probenecid (250 mg/kg, i.p.) every 4 days, for ten times. The mice displayed typical parkinsonian syndrome. Then we examined their olfactory function and the pathologic changes in olfactory bulb. The mice showed obvious olfactory dysfunction in a buried pellet test. Immunohistochemical studies revealed that tyrosine hydroxylase (TH) protein levels were significantly decreased, whereas abnormal α-synuclein was significantly increased in the olfactory bulbs. Furthermore, the olfactory bulbs in MPTP/P-treated mice showed significantly increased levels of interleukin-1β (IL-1β), caspase-1, glial fibrillary acidic protein (GFAP), Toll receptor 4 (TLR4), phosphorylation of p65, as well as activated molecules of NOD-like receptor protein 3 (NLRP3) that were associated with neuroinflammation. Our results demonstrate that MPTP/P-caused olfactory bulb damage might be related to NLRP3-mediated inflammation.

The latent expression pattern of Epstein-Barr Virus (EBV) genes in nasopharyngeal carcinoma (NPC) has been extensively investigated, and the expression of several lytic genes in NPC has been reported. However, comprehensive information through EBV transcriptome analysis in NPC is limited. We performed paired-end RNA-seq to systematically and comprehensively characterize the expression of EBV genes in NPC tissue and C666-1 NPC cell line, which consistently carries EBV. In addition to the transcripts restricted to type II latency infection, the type III latency EBNA3s genes and a substantial number of lytic genes, such as BZLF1, BRLF1, and BMRF1, were detected through RNA-seq and were further verified in C666-1 cells and NPC tissue through realtime PCR.We also performed clustering analysis to classify NPC patient groups in terms of EBV gene expression, which presented two subtypes of NPC samples. Results revealed interesting patterns of EBV gene expression in NPC patients. This clustering was correlated with many signaling pathways, such as those related to heterotrimeric G-protein signaling, inflammation mediated by chemokine and cytokine signaling, ribosomes, protein metabolism, influenza infection, and ECM-receptor interaction. Our combined findings suggested that the expression of EBV genes in NPC is restricted not only to type II latency genes but also to type III latency and lytic genes. This study provided further insights into the potential role of EBV in the development of NPC.

This paper introduces a design program for supporting of deep foundation pit through the actual example for Beijing Moisten the world Center project to solve the difficulties of high water content in foundation soil, sensitiveness to surroundings, and difficult control of quick sand and flowing mud in the process of supporting of foundation pit. This program adopts a supporting system with upper soil nailing wall and lower pre-stressed anchor pile and a comprehensive water-proof curtain (Retaining piles + Long spiral high pressure jet grouting & mixing piles) to effectively solve the above difficulties. It is indicated through actual monitoring that displacement and settlement of foundation pit meet the specification and design requirements with excellent water-proof effect. The successful application of this program provides a reference for foundation pit engineering with rich underground water, especially for engineering at hard soil layer and stratum with sand and gravel that are apt to the formation of cement-soil grouting and mixing piles, as well as engineering with limitation on dewatering of foundation pit.

Aiming at the problem that temperature filed changes can lead to edge cracks emerge to Al-Si alloy 4004 in the process of hot rolling, in this paper, finite element model of temperature field variations in the process of rolling have been established by the method of finite element numerical simulation. The relationship between temperature field variations and edge crack defects has been analyzed. The results show that temperature field of rolled piece is gradient variations, low in the center and high in the edge, metal accumulation in the edge, leading to stress concentration and forming cracks source, resulting in edge cracks arising in edge of roller piece. Finally, the reliability and accuracy of the numerical simulation method have been verified by hot-rolled tests and tensile tests. All the studies provide ideas for theoretical analysis of temperature variation during hot rolling and have important significance on improve the quality of products.

The rehabilitation robot which consists of two mechanical legs and an automatic seat was produced according to ergonomics. Each mechanical leg has three DOF. It can train patients combined with sEMG signal and FES. The rehabilitation robot has three train modes: passive mode, assistant mode and impedance mode, and it suits patients with different rehabilitation phases.

Anchoring technique has been widely used in slope engineering because of its reinforcing function to rock and soil mass. Yet the full understanding of the anchoring mechanism still lags behind the engineering practice. This paper introduces the design process and construction technology of the grouting anchor in slope reinforcement engineering. Based on bolt grouting test, it is found that the effective anchorage length of the grouting anchor depends on the size of the anchor solid and relative physical and mechanical properties of surrounding rock and soil. Research shows that the distribution of shear stress is uneven along the anchoring length. It mainly amasses within the front 1/3 part of the anchoring and increases with the development of outside load. The conclusion is helpful to describe the anchoring mechanism and instructive for engineering practice.

In this paper we study some combinatorial properties and inequalities of some classes of Z-matrices. These matrices arise in many problems in the mathematical and physical sciences. We show that all (inverse)N0-matrices are irreducible and some eigenvalue inequalities of (inverse)N0-matrices.

Widely use of composite structures such as fiber-reinforced composites advances the demand for reliable and effective techniques of ultrasonic nondestructive testing. The model to calculate acoustic field around anisotropic materials is introduced, including propagation and penetration model of Gaussian beams in front of and behind the composites. Simulation and experiments on transmitted acoustic field of composites with different thickness were conducted. Some characteristics of ultrasonic filed around composite materials, such as focal distance and beam spread have been researched theoretically and experimentally. The accuracy of the proposed model is verified by the comparison of the model-based predictions with the experimental results. The achievement will be useful for detecting the defects inside of composites nondestructively.

The interworking with the PSTN network is a network integration issues which must be solved. M2PA, M2UA and M3UA were compared and M3UA was selected because of its stronger flexibility. For the convenience of equipment expansion, IP switch was used in the hardware designation of PSTN Gateway. The software designation was based on independent entity. Socket communication mode which is loosely coupled was used between different entities. The software was divided into the SIP Proxy Module, the Signal Adapter Module, the Service Adapter Module, the Code and Decode Resource Control Module, and so on. The conversion of format, code, identification between SIP and No.7 signaling was realized. Multi-channel controller was used in the conversion of voice stream between the E1 interface circuit mode and RTP/RTCP mode. The interworking between PSTN and IMS was realized through the gateway and the old equipments can be used in the new network. This work is helpful to promote the network integration work.

Polyoxymethylene dimethyl ethers (DMMn) were synthesized in moderate yields by the reaction of methylal and paraformaldehyde catalyzed by solid acid. The pure components of DMM2, DMM3, DMM4, and DMM5 obtained by rectification were characterized by 1H NMR and mass spectra. The suitable components for blending into fuel were obtained by distillation separation. Furthermore, the combustion and emission characteristics of a diesel engine fueled with diesel and a blended fuel of DMM3-5 and diesel were investigated respectively.

In this paper, an Improved Ant Colony Algorithm is applied to the identification of the ship motion modelNomotos 1st-order nonlinear model. A robust method based on Ant Colony Algorithm is proposed for the optimization of the continuous function. The transfer criterion of the ants between each layer and global pheromone updating process are described. Some experiments results show that this method has good identification accuracy. The algorithm is feasible and effective, is of enormous significance to the system identification of ship motion.

Objectives Virtual Touch tissue quantification (VTQ) is a quantified ultrasound (US) acoustic radiation force impulse (ARFI) imaging method that provides numerical measurements (wave-velocity values) of tissue stiffness. The purpose of this study was to detect whether VTQ could be applied to differentiate between benign and malignant thyroid lesions. Methods Healthy subjects’ thyroid tissue and thyroid lesions were examined by VTQ to analyze their elasticity after conventional ultrasound. All the thyroid lesions were analyzed pathologically after surgery and correlated the VTQ values with the pathological results. Results The VTQ value of healthy thyroid tissue, the benign lesions, and the malignant lesions were 1.69 ± 0.41 m/s, 2.03 ± 0.42 m/s, and 3.10 ± 1.08 m/s, respectively. Life and ROP of PDC bits are improved greatly when rotational impact drill assemblies (TorkBuster) are used in hard rock formations. The transient drilling processes of PDC bits combined with and without TorkBuster are simulated based on ANSYS software when drilling in densify sandstone formations. Simulated results are compared. It’s found that the torsion angle of the drill pipe is lessened greatly when torsional impact is loaded with same bit pressure and torque as that without torsional impact. Torsional impact reduces accumulated energy stored in the drill pipe, relieves stick-slip vibration and improves working conditions of PDC bits greatly.

In this paper, continuous phase modulation (CPM) with rate-compatible punctured ring convolutional codes is investigated. Some typical schemes with maximum normalized minimum squared euclidean distance (NMSED) are searched and given. The performance of bit error rate for rate-compatible punctured ring convolutional coded CPM on AWGN channel is simulated, and simulation results show that this system can provide good performance of bit error rate and variable-rate capabilities. Furthermore, simulation results also prove that the transmission efficiency increases when code rate is decreasing.

The aims of this study were to investigate the expression of SOX9 (sex determining region Y [SRY]-related high-mobility group box 9) and carcinoembryonic antigen-related cell adhesion molecule 1 (CEACAM1) in benign, premalignant, and malignant gastric lesions and to explore the association between SOX9 and CEACAM1 in gastric carcinogenesis. SOX9 and CEACAM1 expression was detected in normal gastric mucosa, hyperplastic polyp, intestinal metaplasia, gastric intraepithelial neoplasia, and adenocarcinoma by immunohistochemistry. There was low expression of SOX9 and no CEACAM1 expression in normal gastric mucosa and hyperplastic polyps. Intestinal metaplasia began to express CEACAM1 and showed more membranous staining of CEACAM1 than normal mucosa and hyperplastic polyps (P = .000), but SOX9 expression had no significant difference, and the coexpression of SOX9 and CEACAM1 ascended; therefore, the difference was significant (P = .000). Gastric intraepithelial neoplasia showed more SOX9 expression, coexpression of SOX9, and CEACAM1 than in intestinal metaplasia (P = .014 and P = .026, respectively). Carcinoma showed more cytoplasmic CEACAM1 (P = .010), more SOX9 expression (P = .001), and more their coexpression (P = .023) than gastric intraepithelial neoplasia. As to the histologic classification, poorly differentiated carcinoma showed more cytoplasmic CEACAM1 than well and moderately differentiated carcinoma (P = .006 and P = .024, respectively). In the Laurén classification, diffuse carcinoma showed more cytoplasmic CEACAM1 than intestinal carcinoma (P = .0035), but the SOX9 expression and their coexpresison showed no difference (P = .065 and P = .074, respectively). With the elevation of SOX9 expression and the changing of CEACAM1 expression patterns, the coexpressions of SOX9 and CEACAM1 were highly elevated from benign proliferative lesions to malignant lesions. Moreover, the SOX9 expression and the coexpression with CEACAM1 were correlated positively (r = 0.310; P = .015). In addition, SOX9 expression was positively correlated with CEACAM1 expression patterns (r = 0.124; P = .032). In addition, CEACAM1 expression patterns and coexpression of SOX9 and CEACAM1 show significant difference between T1 and T2 and T3 and T4 (P = .021 and P = .011, respectively). Accordingly, compared with N0, N2 and N3 showed significant difference in SOX9 expression (P = .018), CEACAM1 expression patterns (P = .010), and their coexpression (P = .010). SOX9 expression significantly increased from nonneoplastic lesions to neoplastic lesions, and CEACAM1 expression patterns markedly changed; their coexpression also showed signally elevated suggesting that SOX9, as a transcriptional regulator, play important roles in the changing of CEACAM1 expression patterns, which might promote the tumor progression.

In order to achieve the target of controlling SO2emissions in fumes in a short period of time in China, it is necessary to develop a SO2on-line monitoring system (CEMS) that is suitable for China. To monitor total emissions of SO2in fumes, the concentration of SO2in the fumes and fumes flow rate must be monitored simultaneously. To monitor fumes flow rate, first we must monitor its average flow velocity, this is a major technical problem need to solve in the SO2CEMS. By means of study on velocity field constant in flue, An advanced method for monitoring average flow velocity of fume by mean of velocity field constant is presented, meanwhile an innovative device has been developed to detect velocity field constants automatically, which has resolved the technical difficulty in automated monitoring of fume average velocity in the flue. Through experiment and testing, the monitoring method researched is correct, structure of detecting device is reasonable, its monitoring results are stable and reliable.

According to the characteristics of fault vibration of rolling bearing (non-stationary signals), a method of fault diagnosis based on wavelet analysis is put forward. In the method, the signals included faulty information of rolling bearing are decomposed and reconstructed by Matlab software, and by zoom frequency spectrum analysis, the fault information composition of the rolling bearings can be extracted effectively to determine the faulty type of the rolling bearings. Based on the above, an applicable fault diagnosis system for rolling bearing with surface damage in friction pair was developed. It has been proven by practice that the diagnosis method of system is surefire and its results are stable and reliable. Through being improved and expanded，the system can be applied to collect and analyze other vibration signals.

Direct Torque Control (DTC) has an important role in current motor control. This paper focus on its torque ripple of the reference vector made, summarize a 5 levels structure and novel strategy. Pre-tension of filament is critical to the characteristics of solo-sirofil composite yarn. In this paper, solo-sirofil yarns under filament pre-tension of 5-25cN were produced by modified EJM-128K ring spinning frame. The hairiness, breaking strength, breaking work and yarn evenness were tested under standard test conditions. It’s explored that when the pre-tension of filament is 15cN, perform of solo-sirofil achieve the best level with lower hairiness, higher breaking strength and breaking work, and better yarn evenness.

An experimental device capable of producing uniform metal droplet stream and electric charging and deflecting has been developed. Uniform spherical powder of 180μm in diameter was obtained after cooling and solidification. Numerical models of heat transfer and solidification of droplets were established. The thermal history including solid fraction of droplets in uniform droplet spray process were calculated. Under the common processing conditions, the cooling rate of a droplet of 180μm diameter is of the order of 103−104°C/s. To obtain homogeneous and dense preforms, the substrate should be positioned at a distance of 1.1-1.3m from the nozzle.

Aiming at the water quality characteristics of low turbidity and low temperature of the Reservoir Water derived from Yellow River(RWDFYR), the technology of micro-flocculation direct filtration for RWDFYR was studied. The effluent index such as CODMn, UV254, TOC and turbidity was investigated under the different filtration rate conditions of 4m/h, 5m/h, 6m/h, 8m/h and 10m/h. The results show that the value of removal efficiency of turbidity reached at 98%, and the value of removal efficiency of organic matter was 40% also. In this study, the removal effect was good under the filtration rate of 6m/h. Moreover, the removal effect of different filter layers was also carried out. Finally, it concluded that the backwashing cycle of filter layer should not exceed 3days.

Successive point prevalence surveys were conducted in November 2007 and 2008 to monitor the prevalence of healthcare-associated infection (HCAI) in 13 grade III, 1st class hospitals in Hubei Province of China, using the case definition criteria established by the Ministry of Health in the People's Republic of China. In total, of 20 350 patients surveyed, 833 (4.09%) HCAIs were observed in 790 (3.88%) patients. There was no significant difference between the overall prevalence of HCAI in 2007 (4.14%) and 2008 (3.72%). Respiratory tract infection was the most common HCAI (63.15%), followed by surgical site infection (9.60%) and urinary tract infection (8.64%). Only 35.29% (294/833) of HCAI patients had positive microbiology results. Gram-negative bacteria were isolated most frequently and the most frequent organism was Pseudomonas aeruginosa, followed by Escherichia coli, Acinetobacter baumannii and Staphylococcus aureus. Antibiotic use was documented for 10,344 (50.83%) patients, and cephalosporins, penicillins, and quinolones were the most commonly used agents for treatment or prophylaxis.

The aim of this paper is to construct an image recongnition system based on Gabor wavelets for infrared image. An enhanced representation of the Gabor wavelets is proposed, in which the properties of Gaussian mask in Gabor wavelets is developed to enhance the envelope function, and simultaneously the parameters of the filter based on Gabor wavelets is designed depengding on the frequency response of the training images. Some experiments including infrared image recognitions are given. The good performances are verified through using the proposed scheme in this paper.

In order to improve quality and efficiency of the overall five-axis milling for arbitrary surface impellers. In rough milling, the arbitrary surface was replaced by fitted ruled surface, the flank milling was a good method to replace the point milling. In semi-finish milling, the primal surface was replaced by piecewise ruled surfaces, using step flank milling to gain the satisfaction in model’s nicety and machining efficiency. Then , algorithm of the paper is proved to be viable by Model building, NC programming and machining.

A simple and versatile method for highly efficient synthesis of various polysubstituted indoles via a sequential one-pot Rh(III)-catalyzed direct C2–H and C7–H alkylation with diazo compounds has been developed. This process features with mild reaction conditions, wide substrate scope, and good functional group tolerance (24 examples, up to 96% yield).

The treatment of patients with advanced gastric cancer remains a most challenging task in the clinical practice. Recently, targeted therapies have significantly impacted the treatment strategy for many common malignancies. The use of trastuzumab, a monoclonal antibody against human epidermal growth factor receptor 2 (HER2; also known as ERBB2), plus chemotherapy proved to improve median overall survival in patients with advanced gastric cancer, compared with chemotherapy alone in Trastuzumab for Gastric Cancer (ToGA) trial. However, the prognostic value of HER2 status in gastric cancer remains controversial. Therefore, the aim of this study was to investigate the clinical pathology significance of HER2 overexpression in resectable gastric cancer for selecting the right patients with gastric cancer who may benefit from trastuzumab treatment.Publications reported the clinicopathological factors associated with HER2 status in gastric cancer from 2012 to 2017 were collected. The literature databases, such as "Cochrane Library", "Sciencedirect", "Springer", "PubMed", "Embase", were extensively searched to retrieve the clinical studies of HER2 expression in gastric cancer. The major outcomes measures were odds ratios (ORs) and their 95% CIs. Statistical analysis was carried out by Revman software 5.3. The Newcastle-Ottawa scale was used to assess the quality of evidence.Fifteen studies met our inclusion criteria. This study demonstrated that the pooled OR for HER2 positivity was associated with being male (OR: 1.42; 95% CI: 1.23-1.64), well/moderately differentiated tumor (OR: 2.76; 95% CI: 1.72-4.45), and for intestinal-type tumor (OR: 0.31; 95% CI: 0.25-0.38). However, it had no correlation with depth of tumor (P = .07), venous invasion (P = .82), and lymphovascular invasion (P = .24).HER2-positive expression was associated with male gender, intestinal type, and well/moderate cell differentiation. We recommend that those gastric cancer patients who may benefit from trastuzumab treatment should be subjected to targeted therapies. However, detecting HER2 status may contribute to the target therapy for gastric carcinoma using trastuzumab. This would be strengthened by further studies incorporating comorbidity data, and outcomes from centralized programs.

A sensitive and rapid ultra performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS) method was developed to determine kurarinone in rat plasma using chlorzoxazone as the internal standard (IS). Sample preparation was accomplished through a liquid-liquid extraction procedure with ethyl acetate to 0.2 mL plasma sample. The analyte and IS were separated on an Acquity UPLC BEH C18 column (2.1 mm × 50 mm, 1.7 μm) with the mobile phase of acetonitrile and 1% formic acid in water with gradient elution at a flow rate of 0.40 mL/min. The detection was performed on a triple quadrupole tandem mass spectrometer equipped with electrospray ionization (ESI) by multiple reactions monitoring (MRM) of the transitions at m/z 437.0→301.2 for kurarinone and m/z 168.1→132.1 for IS. The linearity of this method was found to be within the concentration range of 20-2000 ng/mL with a lower limit of quantification of 20 ng/mL. Only 3.0 min was needed for an analytical run. The matrix effect was 94.7-107.2% for kurarinone. The intra- and inter-day precision (RSD%) were less than 8.2% and accuracy (RE%) was within ±9.0%. The recovery ranged from 77.3% to 85.6%. Kurarinone was sufficiently stable under all relevant analytical conditions. The method was also successfully applied to the pharmacokinetic study of kurarinone in rats.

Value of viscosity of liquid at an arbitrary velocity is important in many manufacturing processes, method of rotation is used commonly to measure it at a scheduled velocity, but it is not convenient to measure it at an arbitrary velocity. A method based on cylinder rotation is presented in the paper and it is used to measure it at an arbitrary velocity. Derivation of mathematical models is executed and a simulation is carried in Matlab. Results show that the velocity of the cylinder in the liquid attenuates by exponential law and it is in accordance with mathematical models. Error analysis indicates that that the generation of them mainly results from unsuited velocity of cylinder, slippage between the cylinder and the liquid and misalignment between the cylinder and the cup.

Background Longikaurin A is a natural ent-kaurene diterpenoid isolated from Isodon genus. The ent-kaurene diterpenoids isolated from medicinal plants have been shown to have anti-disease effects. The present study was designed to examine the anti-tumour effects of longikaurin A (LK-A) in nasopharyngeal carcinoma in vitro and in vivo. Methods Apoptosis and cell cycle arrest were determined by flow cytometry analysis of the cells treated with Longikaurin A. The proteins of apoptosis signaling pathway were detected by western blotting analysis. Finally, we examined whether LK-A exhibits anti-tumour activity in xenograft models. Results Longikaurin A inhibited the cell growth by inducing apoptosis and cell cycle arrest. At low concentrations, longikaurin A induced S phase arrest and at higher concentrations, longikaurin A induced caspase-dependent apoptosis by regulating apoptotic molecules. Finally, longikaurin A significantly inhibited the tumour growth of CNE2 xenografts in vivo and showed no obvious effect on the body weights of the mice. Conclusion Our results suggest that Longikaurin A exhibited anti-tumour activity in nasopharyngeal carcinoma in vitro and in vivo.

The aim of this study was to investigate the expression patterns of IGF2 and IMP3 in osteosarcoma as well as its relationship with angiogenesis in the tumor. IGF2 and IMP3 expression was detected by immunohistochemical staining in the serial sections of the osteosarcoma. The impacts of IGF2 and IMP3 expression patterns on tumor angiogenesis were evaluated by statistics. The IGF2 and IMP3 staining had different expression patterns in different osteosarcoma. Twelve out of the sixty-four cases of conventional osteosarcoma showed nuclear staining patterns, and twenty-nine showed cytoplasmic staining of IGF2 and IMP3 simultaneously. On the other hand, fourteen cases showed nuclear IGF2 staining but cytoplasmic IMP3 expression, and nine cases showed nuclear IMP3 staining and cytoplasmic IGF2 expression. Twenty-eight out of forty-seven cases of parosteal osteosarcoma showed nuclear IGF2 and IMP3 expression, nine showed cytoplasmic IGF2 and IMP3 expression simultaneously. Seven out of forty-seven cases of parosteal osteosarcoma expressed IGF2 with nuclear staining but expressed IMP3 with cytoplasmic staining. Meanwhile, three cases expressed IGF2 with cytoplasmic staining but expressed IMP3 with nuclear staining. Similar to the parosteal osteosarcoma, the periosteal osteosarcoma expressed IGF2 and IMP3 mainly with nuclear staining simultaneously, forty out of fifty-five cases of periosteal osteosarcoma did that. Five out of fifty-five cases expressed IGF2 and IMP3 with cytoplasmic staining at the same time. Four cases showed nuclear IGF2 staining and cytoplasmic IMP3 staining. In the parosteal and periosteal osteosarcoma, there was no significant difference in IGF and IMP3 expression patterns (P = 0.216). However, compared with conventional osteosarcoma, the parosteal and periosteal osteosarcoma showed significant difference in IMP3 and IGF2 expression (P = 0.016, P = 0.023). IGF2 and IMP3 expression patterns were positive correlation in the different osteosarcoma (r = 0.1021, P = 0.032). The Microvessel density (MVD) in osteosarcoma with IGF2 and IMP3 cytoplasmic staining was more than that with nuclear expression of IGF2 and IMP3, and the difference was significant (P = 0.024). Moreover, the conventional osteosarcoma with cytoplasmic IGF and IMP3 showed more MVD than parosteal and periosteal osteosarcoma with cytoplasmic IGF and IMP3, and the difference was significant (P = 0.035). IGF2 and IMP3 had different expression patterns, which might be associated with angiogenesis. However, cytoplasmic and nuclear expression of IGF2 and IMP3 might play different roles in the angiogenesis of osteosarcoma.

AIM: To investigate the protective effect of target suppression of uncoupling protein-2 (UCP-2) on ischemia/reperfusion (I/R) injury in fatty liver in ob/ob mice. METHODS: Plasmids suppressing UCP-2 expression were constructed, and transfected into fatty liver cells cultured in vitro and the ob/ob mouse I/R injury model. Serum tumor necrosis factor (TNF)-α levels, UCP-2 mRNA expression, alanine aminotransferase (ALT) levels in ob/ob mice were tested, and the pathological changes in fatty liver were observed in experimental and control groups. RESULTS: In ob/ob mouse I/R models, serum TNF-α levels were significantly higher than in normal controls. After the plasmids were transfected into the cultured cells and animal models, expression of UCP-2 mRNA was significantly reduced as compared with that in the control group (21.56 ± 0.15 vs 2-0.45 ± 0.15, P < 0.05). In ob/ob mouse models, in which expression of UCP-2 was suppressed, serum ALT levels were significantly lower than those of other groups, and pathological analysis revealed that injury of liver tissues was significantly alleviated. CONCLUSION: The target suppression of UCP-2 expression in fatty liver can alleviate the I/R injury in the ob/ob mice.

AIM: To determine the role of Fas/Fas ligand (FasL) in the immune escape of colon cancer cells. METHODS: Immunohistochemistry was used to observe the expression of Fas and FasL in the tissues of colon cancer patients.In situ hybridization was used to detect the localization of FasL mRNA expression in cancer tissues. Terminal deoxynucleotide transferase-mediated dUTP nick end labeling (TUNEL) assay and CD45 staining were performed to detect the apoptosis of tumor-infiltrating lymphocytes (TILs). Co-culture assays of colon cancer cells (SW480) and Jurkat cells (Fas-sensitive cells) were performed to observe the counterattack of colon cancer cells to lymphocytes. RESULTS: Of 53 cases of colon carcinomas, 23 cases (43.4%) expressed Fas which was significantly lower as compared to the normal colonic mucosa (73.3%, P
